RESUMO

A brand-new class of interstitial cells, called telocytes, has been detected in the heart. Telocytes can connect and transmit signals to almost all cardiomyocytes; this is highly interrelated with the occurrence and development of heart diseases. Modern studies have shown that berberine has a therapeutic effect on cardiovascular health. However, berberine's mechanism of action on the cardiovascular system through cardiac telocytes is unclear. Interestingly, 5 µm of berberine remarkably decreased the concentration of intracellular calcium and membrane depolarization in cultured telocytes, upregulated the expression of CX43 and ß-catenin, and downregulated the expressions of TRPV4 and TRPV1. Here, telocytes were identified in the vascular adventitia and intima, endocardium, myocardium, adventitia, and heart valves. Moreover, telocytes were broadly dispersed around cardiac vessels and interacted directly through gap junctions and indirectly through extracellular vesicles. Together, cardiac telocytes interact with berberine and then deliver drug information to the heart. Telocytes may be an essential cellular target for drug therapy of the cardiovascular system.

Early hearing loss could cause abnormal brain development, which has been linked to the complex process known as cross-modal neuroplasticity. However, previous studies investigating the brain structure of infants with congenital severe sensorineural hearing loss (CSSHL) are scarce and have yielded inconsistent results. This study aimed to further explore the gray matter (GM) density changes in children with CSSHL. Fifteen children aged 0-5 years with CSSHL and 11 healthy children as controls (aged 0-5 years) were recruited. Each participant underwent a structural MRI scan. The voxel-based morphometry method was performed to evaluate GM density for each participant and analyze their characteristics. It was discovered that: (1) GM density of the right superior temporal gyrus and caudate in the CSSHL group was smaller than that of healthy controls (HC). However, GM density was larger in the left posterior central gyrus, superior frontal gyrus, inferior parietal lobule and right cerebellum in the CSSHL group compared with HC. (2) The GM density value of the left superior frontal gyrus and inferior parietal lobule was negatively correlated with age. However, the GM density value of the right superior temporal gyrus in the CSSHL group was positively correlated with age. Compared with HC, the GM density of CSSHL children was larger in somatosensory areas (including left superior frontal gyrus, posterior central gyrus, inferior parietal lobule and right cerebellum), whereas GM density was smaller in auditory-related areas (such as the right superior temporal gyrus and caudate). Moreover, GM density change was influenced by the duration of hearing deprivation.

Meige's syndrome and hemifacial spasm (HFS) are two different forms of dystonic movement disorder, but their difference in terms of resting state functional connectivity (rsFC) remains unclear. The present study applied resting state fMRI on the patients and quantified their functional connectivity with graph theoretical measures, including the degree centrality and the betweenness centrality. Fifteen Meige's syndrome patients and 19 HFS patients matched in age and gender were recruited and their MRI data were collected. To analyze the rsFC, we adopted the Anatomical Automatic Labeling (AAL) template, a brain atlas system including 90 regions of interest (ROIs) covering all the brain regions of cerebral cortex. For each participant, the time-course of each ROI was extracted, and the corresponding degree centrality and betweenness centrality of each ROI were computed. These measures were then compared between the Meige's syndrome patients and the HFS patients. Meige's syndrome patients showed higher betweenness centrality and degree centrality of bilateral superior medial frontal cortex, the left cerebellum cortex, etc. than the HFS patients. Our results suggest that the rsFC pattern in Meige's syndrome patients might become more centralized toward the prefrontal and vestibular cerebellar systems, indicating less flexibility in their functional connections. These results preliminarily revealed the characteristic abnormality in the functional connection of Meige's patients and may help to explore better treatment.

The rate of stigma exsertion (SE) is an important trait in rice breeding because the efficiency of hybrid rice seed production can be improved by increasing the percentage of stigmas that exsert. In this study, we developed a near isogenic line (NIL) from two parents, XieqingzaoB (XQZB) and Zhonghoi9308 (ZH9308), which have high and low SE rates in that order. In our previous study, we employed 75 chromosome segment substitution lines (CSSLs) and analyzed quantitative trait loci (QTLs) for their influence on SE rate. The single gene QTL (qSE11), which is located on chromosome 11, was responsible for this trait. In this study, we focused on one of the CSSLs (C65), namely, the NIL (qSE11XB). It contains an introgression segment of XQZB in the genetic background of ZH9308, and exhibits a significantly higher SE rate than that of the parents. We demonstrated that qSE11 regulated both the single and the dual SE rates. They both contribute to the total SE rate. Genetic analysis revealed that qSE11 acted as a single Mendelian factor and that the allele from XQZB increased the SE rate. The validity of our conclusions was established when C65 was used to develop secondary F2 (BC5F2) and F2:3 (BC5F2:3) populations by backcrossing to ZH9308, with subsequent selfing. We entered 3600 plants from the F2 population and 3200 from the F2:3 populations into a genetic dissection program and dissected the major QTL qSE11 to a 350.7-kb region located on chromosome 11. This study will contribute to the future isolation of candidate genes of SE and will play a vital role in future hybrid rice seed production programs.

Two novel dinuclear complexes [Co(2)(L)(2)(phen)(2)·2H(2)O](n) (1), [Co(2)(L)(2)(bipy)(2)](n) (2) (where phen=1,10-phenanthroline, bipy=2,2'-bipyridine, and L=biphenyl-2,4'-dicarboxylic acid) have been synthesized and characterized using IR, (1)H NMR, element analysis and single-crystal X-ray diffractometry. The binding of the complexes with HC-DNA (HeLa cells DNA, which was extracted by ourselves) was investigated by fluorescence spectrum. The experimental results show that the two complexes have the ability to bind with extracted HC-DNA. Gel electrophoresis assay demonstrates the ability of the complexes to cleave the extracted HC-DNA. The complexes exhibit a higher cytotoxicity against tumor cells as against normal cells in vitro. Further more, the apoptotic tests indicate that the complexes have an apoptotic effect on HeLa cells.
