RESUMO

Lead-zinc (Pb-Zn) tailings pose a significant environmental threat from heavy metals (HMs) contamination. Revegetation is considered as a green path for HM remediation. However, the interplay between HM transport processes and soil microbial community in Pb-Zn tailings (especially those in production) remain unclear. This study investigated the spatial distribution of HMs as well as the crucial roles of the soil microbial community (i.e., structure, richness, and diversity) during a three-year revegetation of production Pb-Zn tailings in northern Guangdong province, China. Prolonged tailings stockpiling exacerbated Pb contamination, elevating concentrations (from 10.11 to 11.53 g/kg) in long-term weathering. However, revegetation effectively alleviated Pb, reducing its concentrations of 9.81 g/kg. Through 16 S rRNA gene amplicon sequencing, the dominant genera shifted from Weissella (44%) to Thiobacillus (17%) and then to Pseudomonas (comprising 44% of the sequences) during the revegetation process. The structural equation model suggested that Pseudomonas, with its potential to transform bioavailable Pb into a more stable form, emerged as a potential Pb remediator. This study provides essential evidence of HMs contamination and microbial community dynamics during Pb-Zn tailings revegetation, contributing to the development of sustainable microbial technologies for tailings management.

Acorus tatarinowii (A. tatarinowii) is a medicinal plant of the Araceae family. Currently, pharmacology focuses on the study of volatile oils, but there are few reports of another important secondary metabolite, lignan. Dirigent protein is thought to play an important role in plant secondary metabolism and responds to a variety of biotic and abiotic stresses. However, the DIR gene family of A. tatarinowii has not been systematically analyzed, and it is unknown whether it affects lignan synthesis. In this study, a total of 27 AtsDIRs were identified by comprehensive analysis of the genome of the medicinal plant A. tatarinowii, and the candidate gene AtsDIR23 that may be involved in lignan synthesis was screened through bioinformatics and transcriptome analysis. It is worth noting that AtsDIR23 is significantly expressed in rhizomes and is a member of the DIR-a subfamily. Subsequently, subcellular localization revealed that AtsDIR23 was localized in chloroplasts. The functional verification of AtsDIR23 b y the transient transformation of A. tatarinowii and the stable transformation of Arabidopsis thaliana showed that the content of lignans in overexpressed plants increased. Co-expression analysis screening revealed the MYB transcription factor (AtsMYB91) that is highly correlated with AtsDIR23 expression, while yeast one-hybrid assays and double luciferase experiments showed that AtsMYB91 negatively regulated the expression of AtsDIR23 b y binding to the AtsDIR23 promoter. In conclusion, AtsDIR23 can promote the accumulation of lignans, which provides a reference for further research on the regulation of lignans by DIR genes.

In this work, spherical Ni-based metal-organic frameworks (Ni-MOFs) were used as precursors for preparing nanorod-like nickel phosphate (r-NiPO) with enhanced electro-catalytic properties in detection of glucose. It was found that the addition of nickel ion has a profound effect on construction of nickel phosphate with flower-like, rod-like, and ribbon-like structure in presence of H2PO2-. The r-NiPO consisted of multiple phases of nickel phosphate showed porous, interconnected structure and abundant active Ni(II)/Ni(III) pairs, which was beneficial to facilitate glucose oxidation. In comparison with Ni-MOF, r-NiPO exhibited the superior electrocatalytic property on oxidation of glucose because of the exposing more active Ni sites in r-NiPO than that of Ni-MOF precursor. r-NiPO modified glassy carbon electrode (r-NiPO/GCE) showed a good linear response to glucose from 1.0 µM to 3.0 mM with a high sensitivity of 3169 µA mM-1 cm-2. In addition, r-NiPO/GCE can be used to detect glucose in human serum with satisfactory recoveries (90-97%).

In order to optimize the extraction of gigantol from Dendrobium officinale, the influence of methanol concentration, ultrasonic temperature, and liquid ratio on extraction efficiency was analysed by the response surface analysis method. The results show that the extraction rate reached a maximum when the methanol concentration was 92.98%, the solid-liquid ratio was 27.2 mL/g, and the extraction temperature was 41.41 °C. The content of gigantol of Dendrobium officinale in leaves was significantly higher than that in stems, reaching 4.7942 µg/g. The content of gigantol in Dendrobium huoshanensis Fengdou was significantly higher than that of other species of Fengdou. This experiment has practical significance for improving the utilization rate of Dendrobium officinale, and provides a reference for the study of the pharmacological and biological activity of gigantol.

Alkaloids are the main active ingredients in the medicinal plant Dendrobium officinale. Based on the published genomic and transcriptomic data, a proposed terpenoid indole alkaloid (TIA) biosynthesis pathway may be present in D. officinale. In this study, protocorm-like bodies (PLBs) with a high-yielding production of alkaloids were obtained by the optimization of tryptophan, secologanin and methyl jasmonate (MeJA) treatment. The results showed that the total alkaloid content was 2.05 times greater than that of the control group when the PLBs were fed with 9 µM tryptophan, 6 µM secologanin and 100 µM MeJA after 36 days. HPLC analysis showed that strictosidine synthase (STR) activity also increased in the treated plants. A total of 78 metabolites were identified using gas chromatography-mass spectrometry (GC-MS) in combination with liquid chromatography-mass spectrometry (LC-MS) methods; 29 differential metabolites were identified according to the multivariate statistical analysis. Among them, carapanaubine, a kind of TIA, exhibited dramatically increased levels. In addition, a possible underlying process of the metabolic flux from related metabolism to the TIA biosynthetic pathway was enhanced. These results provide a comprehensive view of the metabolic changes related to alkaloid biosynthesis, especially TIA biosynthesis, in response to tryptophan, secologanin and MeJA treatment.
