RESUMO

Intraperitoneal (i.p.) experimental models in mice can recapitulate the process of i.p. dissemination in abdominal cancers and may help uncover critical information about future successful clinical treatments. i.p. cellular composition is studied in preclinical models addressing a wide spectrum of other pathophysiological states such as liver cirrhosis, infectious disease, autoimmunity, and aging. The peritoneal cavity is a multifaceted microenvironment that contains various immune cell populations, including T, B, NK, and various myeloid cells, such as macrophages. Analysis of the peritoneal cavity is often obtained by euthanizing mice and performing terminal peritoneal lavage. This procedure inhibits continuous monitoring of the peritoneal cavity in a single mouse and necessitates the usage of more mice to assess the cavity at multiple timepoints, increasing the cost, time, and variability of i.p. studies. Here, we present a simple, novel method termed in vivo intraperitoneal lavage (IVIPL) for the minimally invasive monitoring of cells in the peritoneal cavity of mice. In this proof-of-concept, IVIPL provided real-time insights into the i.p. tumor microenvironment for the development and study of ovarian cancer therapies. Specifically, we studied CAR-T cell therapy in a human high-grade serous ovarian cancer (HGSOC) xenograft mouse model, and we studied the immune composition of the i.p. tumor microenvironment (TME) in a mouse HGSOC syngeneic model.

This study aimed at understanding the differences in traits of functional twigs and leaves of a typical alpine shrub species, Rhododendron przewalskii, at Kaka Mountain in the headwater region of Minjiang River. Leaf and twig traits were compared for shrubs at different growth stages (flower bud stage and flowering stage) and altitude (3600 m and 3800 m). The effects of spatial heterogeneity on their correlations and trade-offs were evaluated at leaf and twig levels, respectively. Our results showed that twig length was significantly longer at low altitude than high altitude for the shubs at the same growth stage. The number and mass of flowers at flowering stage were significantly higher at high altitude than those at low altitude. At the same altitude, twig mass, number of leaves, total leaf mass, total leaf area and total petiole mass were all significantly greater at the flower bud stage than those at the flowering stage, while the individual leaf mass and individual petiole mass at flower bud stage were significantly smaller than those at flowering stage. Compared with the flower bud stage, the proportion of leaf mass decreased by 13% at the flowering stage, while biomass proportion of twig significantly increased. At the flower bud stage, twig mass had a higher contribution to total twig mass. In contrast, the contribution of total leaf mass to total twig mass was higher at flowering stage. More biomass of leaf was allocated to individual leaf mass at flower bud stage. More biomass of leaf was allocated to individual petiole mass and individual leaf mass at flowering stage at low altitude and high altitude, respectively. At low altitude, allometric growth patterns presented between twig mass and total leaf area, total leaf mass. Similarly, individual petiole mass and individual leaf area had allometric growth. Our results indicated that the functional traits of twigs and leaves varied across both altitude and plant growth stage.

The water logging has become the environmental problem of major cities with the sharp increase of impermeable urban pavement as the contributing cause. Abroad, the green roof has been widely used as a practical measure to intercept rainwater, yet the capacity of green roof to retain rainwater varies with climate conditions. As the hot and humid climate zone features high temperature, humidity and precipitation, it is meaningful to study the capacity of green roof to retain rainwater under such climatic condition. In this research, 3 plat forms were set up in Guangzhou in rainy and hot summer to test the capability of simple green roof to retain rainwater runoff, and the efficiency of green roof to retain rainwater under local climate conditions was worked out based on the meteorological observation and data measurement during the 13-month test period. The results showed that the simple green roof with a substrate thickness of 30, 50 and 70 mm could retain 27.2%, 30.9% and 32.1% of precipitation and reduce the average peak value by 18.9%, 26.2% and 27.7%, respectively. Given an urban built-up area of 1035.01 km2 in Guangzhou and a roof area percentage of approximately 37.3% and assuming the green roofs with 30 mm-thick substrate were applied within the area, the light, medium and heavy rain could be delayed at 72.8%, 22.6% and 17.4%, respectively. Accordingly, the rainwater retained could reach up to 14317×104 m3. It suggested the great potential of the simple green roof in retaining rainwater. The research could serve as reference for the hot and humid climate zone to alleviate water logging and visualize sponge city construction.

OBJECTIVE: To investigate the inhibitory effect of nanoparticle-mediated antisense oligodeoxynucleotide (ASODN) of human telomerase reverse transcriptase (hTERT) on telomerase in the esophageal cancer EC9706 cells. METHODS: Line-polyethylenimine (L-PEI) was used to condense ASODN into nanoparticle and to couple NGR peptides into targeting nanoparticle, and the prepared L-PEI/ASODN complexes were transfected into the EC9706 cells. Cellular uptake of L-PEI/ASODN complexes was detected by laser confocal scanning microscopy. MTT assay was used to detect the inhibitory rate of EC9706 cell growth. The level of hTERT mRNA and its protein expression were measured by RT-PCR and immunohistochemistry, respectively. Annexin V FITC/PI double labeling was used to detect cell apoptosis. The distribution of drug in nude mice was observed by laser confocal scanning microscopy, and the growth and morphology of the tumor was examined. RESULTS: The L-PEI-mediated ASODN uptake was enhanced. After transfection, the inhibitory rate of EC9706 cells was time-dependant and there was a significant difference between control cell group and L-PEI/ASODN group (P < 0.05). At 48 h after transfection, the level of hTERT mRNA was decreased significantly compared with that of control cell group (P < 0.05), and the expression of hTERT protein was negative. There was apparent apoptosis in EC9706 cells after transfection with L-PEI/ASODN complexes. For the two NGR/L-PEI/ASODN groups, fluorescence was observed in the liver, kidney, lung and tumor tissues of nude mice, and their uptake intensity was time-dependent. The mean volume of tumors in the two NGR/L-PEI/ASODN groups was significantly smaller than those in blank control group and SODN group (P < 0.05). Apoptotic bodies were detected in the tumors of L-PEI/ASODN group. CONCLUSION: The NGR/L-PEI/ASODN nanoparticles can effectively reach into the human esophageal cancer xenograft and inhibit the tumor growth in nude mice, and this may provide a theoretical and experimental basis for gene therapy for human esophageal squamous cell carcinoma.
