The most interesting activities associated with bread components such as phenolic compounds, fibre, tocols, or newly formed compounds in the Maillard reaction, are their reducing properties responsible for the formation of the overall reducing capacity of bread. Among the electrochemical methods, the cyclic voltammetry (CV) technique has been recently adapted for this purpose. In this study, the application of the CV assay for the determination of the total reducing capacity of flours, doughs, and breads as well as their crumbs and crusts, originated from wheat, spelt, and rye formulated on white flours (extraction rate of 70%) and dark flours (extraction rate of 100%) and baked at 200 °C for 35 min and at 240 °C for 30 min was addressed. The reducing capacity of hydrophilic extracts from white flours and breads as well as their crumbs and crusts showed double values when compared to that of lipophilic ones whilst hydrophilic and lipophilic extracts from dark breads and their parts revealed comparable levels. The dark wheat, spelt, and rye breads showed an approximately threefold higher total reducing capacity than white breads. Baking at higher temperature slightly increased the total reducing capacity of breads and the highest value was found for dark rye bread as well as its crust baked at 240 °C for 30 min. The cyclic voltammetry methodology showed to be especially suitable for screening the bread technology and allows for obtaining rapid electrochemical profiles of bread samples.

BACKGROUND: Okra pods contain heat-sensitive substances, such as phenolic compounds and other phytochemicals that can be degraded when okra pods are subjected to heat treatment. The understanding of the impact of high humidity hot air impingement blanching (HHAIB) on the changes in physicochemical properties of polysaccharides and phytochemicals of okra pods is of great importance because over-blanching may result in cell membrane disruption and changes in biologically active compounds under prolonged exposure to the thermal treatment. Therefore, the present study aimed to investigate the effect of HHAIB on the changes in physicochemical properties of pectins and phytochemicals extracted from okra pods. RESULTS: Both the HHAIB time and method of extraction influenced their physicochemical characteristics and biological activity. Pectin fractions subjected to HHAIB were composed of polygalacturonic acid, rhamnogalacturonan, glucomannan, galactan, mannose, arabinose, rhamnose, calcium pectate and arabinogalactan. The contents of total phenolics, total flavonoids and antioxidant activity of extracts mostly increased during HHAIB (i.e. up to 19.0%, 13.2% and 35.3%, respectively). However, HHAIB reduced the chlorophyll-a (up to 55.7%) and lycopene (up to 52.6%) contents of okra pods. CONCLUSION: The acquired knowledge may be useful for better understanding and optimization of technologies based on HHAIB treatment. The HHAIB treated okra can be a promising natural alternative in different applications, including its use as a replacement of some ingredients in food or non-food systems as a result of richness in polysaccharides and polyphenols, as well as high antioxidant properties. © 2022 Society of Chemical Industry.

Caffeic acid is one of the most abundant hydroxycinnamic acids in fruits, vegetables, and beverages. This phenolic compound reaches relevant concentrations in the colon (up to 126 µM) where it could come into contact with the intestinal cells and exert its anti-inflammatory effects. The aim of this investigation was to study the capacity of caffeic acid, at plausible concentrations from an in vivo point of view, to modulate mechanisms related to intestinal inflammation. Consequently, we tested the effects of caffeic acid (50-10 µM) on cyclooxygenase (COX)-2 expression and prostaglandin (PG)E2, cytokines, and chemokines (IL-8, monocyte chemoattractant protein-1 -MCP-1-, and IL-6) biosynthesis in IL-1ß-treated human myofibroblasts of the colon, CCD-18Co. Furthermore, the capacity of caffeic acid to inhibit the angiotensin-converting enzyme (ACE) activity, to hinder advanced glycation end product (AGE) formation, as well as its antioxidant, reducing, and chelating activity were also investigated. Our results showed that (i) caffeic acid targets COX-2 and its product PGE2 as well as the biosynthesis of IL-8 in the IL-1ß-treated cells and (ii) inhibits AGE formation, which could be related to (iii) the high chelating activity exerted. Low anti-ACE, antioxidant, and reducing capacity of caffeic acid was also observed. These effects of caffeic acid expands our knowledge on anti-inflammatory mechanisms against intestinal inflammation.

In this study, 11 apple cultivars were characterized by their total phenolic content (TPC) and total flavonoid content (TFC) and antioxidant, reducing, and chelating capacity by 2,2-diphenyl-1-picrylhydrazyl (DPPH) test, cyclic voltammetry (CV), and ferric reducing antioxidant power (FRAP) assays; and ferrous ion chelating capacity. The phenolic compounds in flesh and peel were determined by liquid chromatography coupled to mass spectrometry and diode array detector (HPLC-DAD-MS/MS) and their electroactivity by CV. The results showed higher TPC, TFC, and antioxidant capacity by DPPH test in the peels of all apple cultivars as compared to the respective flesh. The peel extracts also showed two-fold higher FRAP values as compared to the flesh extracts. The reducing capacity of the peel and flesh determined by CV measurements confirmed the results achieved by spectrophotometric methods of evaluating antioxidant capacity. There was no significant difference in chelating capacity in the peel and flesh. The HPLC-DAD-MS/MS analysis showed the presence of 11 phenolic compounds in the peel and flesh which varied in antioxidant, reducing, and chelating activity. The order of the phenolic compound content in flesh and peel in Quinte cultivar, which showed the highest antioxidant capacity, was as follows: epicatechin > chlorogenic acid > quercetin 3-arabinoside > quercetin 3-glucoside > cyanidin 3-galactoside > quercetin 3-rhamnoside > catechin > phloridzin > rutin > phloretin = quercetin. CV results were highly correlated with those obtained by spectrophotometry and HPLC-DAD-MS/MS, providing evidence to support the use of cyclic voltammetry as a rapid method to determine the phenolic profile and reducing the power of apple flesh and peel. The association between antioxidant assays and phenolic compound content showed that the highest contribution to the antioxidant capacity of apple peel and flesh was provided by catechin, epicatechin, and cyadinin-3-galactoside, while phloretin, phloridzin, and chlorogenic acid were the main contributors to chelating activity. Results from this study clearly indicate that removing the peel from apples may induce a significant loss of antioxidants.

Plant-derived food consumption has gained attention as potential intervention for the improvement of intestinal inflammatory diseases. Apple consumption has been shown to be effective at ameliorating intestinal inflammation symptoms. These beneficial effects have been related to (poly)phenols, including phloretin (Phlor) and its glycoside named phloridzin (Phldz). To deepen the modulatory effects of these molecules we studied: i) their influence on the synthesis of proinflammatory molecules (PGE2, IL-8, IL-6, MCP-1, and ICAM-1) in IL-1ß-treated myofibroblasts of the colon CCD-18Co cell line, and ii) the inhibitory potential of the formation of advanced glycation end products (AGEs). The results showed that Phlor (10-50 µM) decreased the synthesis of PGE2 and IL-8 and the formation of AGEs by different mechanisms. It is concluded that Phlor and Phldz, compounds found exclusively in apples, are positively associated with potential beneficial effects of apple consumption.

The effect of high hydrostatic pressure (HHP) (450-550-650 MPa/5-10-15 min) on polyphenols profile of purée obtained from commercial tomato varieties (Maliniak, Cerise, Black Prince and Lima) was investigated. Individual polyphenols, total phenolic index (TPI) were quantified using a mass spectrometer (HPLC-MS/MS). Photochemiluminescence (PCLACW), cyclic voltammetry (CV) and ferric reducing antioxidant power (FRAP) assays were used to determine the antioxidant capacity of the hydrophilic extract. The results demonstrated that at certain processing conditions, HHP may enhance or decrease the nutritional quality of tomato purée. However, the tomato variety was a key factor influencing the polyphenols profile and the antioxidant capacity. A significant positive correlation was found among TPI, FRAP or CV parameters and the concentration of caffeic, ferulic, sinapic and p-coumaric acids, and epicatechin. On the other hand, significant positive correlation was observed among antioxidant capacity (PCLACW), TPC, rutin and naringenin concentration as well as chlorogenic and isochlorogenic acids.

Selected functional properties of four types of gluten-free muffins made of unfermented and fermented (by Lactobacillus plantarum) buckwheat flour in comparison with control muffins made using commercial gluten-free corn flour were evaluated in this study. The proximate chemical composition, antioxidant capacity analysed by ABTS, photochemiluminescence and cyclic voltammetry assays, and inhibitory activity against protein glycation in vitro in BSA/Glu systems were investigated. The content of the total phenolic compounds, available lysine, furosine, free and total FIC, browning index and antioxidant capacity of buckwheat-enhanced gluten-free muffins were higher compared to the control samples. Gluten-free muffins made of the fermented buckwheat flour showed a significantly higher antioxidant capacity, an increased activity against AGEs formation and an increased available lysine content, as well as a higher FAST index and browning index as compared to the muffins obtained with unfermented buckwheat flour. The study showed that buckwheat flour fermented by L. plantarum could be used as an ingredient for improving the functional properties of gluten-free muffins.

SCOPE: Buckwheat (BW) consumption has been associated with a broad range of health benefits: antioxidant, anti-inflammatory and anticancer. These beneficial effects have been partially related to the presence of flavonoids. However, some of these compounds (i.e., rutin and quercetin) are metabolized in the gastrointestinal tract generating derived phenolic metabolites. In this study, we investigated the biological activity of rutin (Ru), quercetin (Q) an their derived phenolic metabolites 3,4-dihydroxyphenylacetic acid (3,4-DHPAA), 3-hydroxyphenylacetic acid (3-HPAA), and 4-hydroxy-3-methoxyphenylacetic acid (homovanillic acid, HVA). METHODS AND RESULTS: Q showed the highest antioxidant and reducing activity, and Ru the maximum chelating activity (85.33%). Antioxidant activity of 3,4-DHPAA was 5-fold higher than that of HVA, whereas their reducing activity was similar. The formation of methylglyoxal (MGO)-BSA and glucose-BSA (advanced glycation end products) was inhibited by Ru (98.5 and 92.7%), Q (95.6 and 89.1%) and 3,4-DHPPA (84.4.6 and 77.5%). Furthermore, Q (10-50 µM) and Ru (1-50 µM) downregulated the release of PGE2 , IL-8 and MCP-1, molecules involved in the inflammatory response, in IL1ß-inflamed myofibroblasts of colon CCD-18Co. CONCLUSION: This study suggests that BW phytochemicals and their phenolic metabolites may be responsible for the beneficial effects against chronic diseases attributed to BW consumption.

Pinto bean protein concentrate was hydrolyzed by subtilisins at 0.1, 100, and 200 MPa and 50 °C for 15 min. Alcalase hydrolysis at 100 MPa led to higher ACE inhibition, reducing power, and free radical scavenging activity of hydrolysates. However, hydrolysate obtained by Savinase at 200 MPa showed the best ACE-inhibitory and radical scavenging activities. Proteolysis by Savinase at 200 MPa was considered the most effective treatment to increase small peptides (<3 kDa), flavonoids, total phenolic compounds, and oxygen radical absorbance capacity in hydrolysates. In this hydrolysate, small phaseolin fragments with reported ACE-inhibitory and antioxidant sequences were identified. Catechin, pelargonidin 3-glucoside, and ferulic acid were the main phenolic compounds. Hihg-pressure-assisted hydrolysis of common bean protein concentrates would provide benefits in the production of functional hydrolysates providing higher functionality and added value to the resulting hydrolysate due to synergistic effects of bioactive peptides and soluble phenolics.

ABSTRACT: Coffee being the beverage consumed worldwide is also a very competitive commodity. Consequently, producers seek ways of attracting consumers by proposing e.g. novel ingredient combinations usually without evaluating their health quality. In this study, variations in health-promoting determinants for five special purpose coffee brews were characterized. The major bioactive components - chlorogenic acids (CAs) - detected by HPLC-DAD-MS included caffeoyl-, feruloyl- and dicaffeoylquinic acids. Roasting declined CAs content by 50 %, while caffeine content increased by about 30 % during this process. In roasted coffee brews studied, CAs content varied from 14.1 to 26.6 mg/g; smaller variations were seen in antioxidant potential (determined by spectrophotometric and cyclic voltammetry assays). Profiles generated by HPLC with post-column on-line antioxidant detection revealed that caffeoylquinic acids were the most abundant antioxidants in samples studied. The highest antioxidative potential exhibited coffee enriched with green coffee extract confirming the soundness of such an approach to obtain healthier products. HIGHLIGHTS: • Special purpose coffees are equivalent to regular ones as regards antioxidant potential. • Caffeoylquinic acids are the most abundant antioxidants in coffee samples studied. • Roasting process causes some degradation of chlorogenic acids but not caffeine. • Special purpose coffees retain health benefits while being more attractive to consumers.

Changes in chemical composition and antioxidative properties of rye ginger cakes during their shelf-life were investigated in this study. In particular, the changes in antioxidants content, antioxidative and reducing capacity, and Maillard reaction development in rye ginger cakes after long-term storage were addressed. Ginger cakes produced according to the traditional and current recipe were stored for 5 years at room temperature in a dark place. The total phenolic compounds (TPC), inositol hexaphosphate (IP6), reduced (GSH) and oxidised glutathione (GSSG) contents, antioxidant and reducing capacity and Maillard reaction products (MRPs) were determined in ginger cakes after storage and then compared to those measured after baking. After long-term storage a decrease in TPC and IP6 contents in cakes was noted. In contrast, an increase in antioxidative and reducing capacity of stored cakes was observed. Long-term storage induced formation of furosine, advanced and final Maillard reaction products and caused changes in both reduced and oxidised forms of glutathione. After long-term storage the modest changes in furosine, FAST index and browning in ginger cake formulated with dark rye flour may suggest that this product is the healthiest among others. Therefore, traditional rye ginger cakes can be considered as an example of a healthy food that is also relatively stable during long term storage as noted by the small chemical changes observed in its composition.

The analysis of major and minor flavonoids, and antioxidant capacity of stems, leaves, flowers, unripe seeds and ripe seeds of common and tartary buckwheat plants collected during different growth periods was addressed in this study. The highest rutin contents were observed in flowers and leaves collected from common and tartary buckwheat at early flowering as well as flowering and seed formation states. A low quercetin contents were found in all studied aerial part of buckwheat plants. Quercitrin (quercetin-3-rhamnoside) was only found in flowers collected at different growth periods while flavone C-glucosides were accumulated preferentially only in unripe seeds collected from common buckwheat at an early flowering state. The rank of antioxidant capacity provided for aerial parts of common and tartary buckwheat at early flowering state was as follows: flowers > leaves > stems. The highest contribution of rutin to the antioxidant capacity of the aerial parts of common and tartary buckwheat was found for stems followed by leaves, flowers and unripe seeds. The results demonstrate that flowers from common and tartary buckwheat collected at early flowering as well as flowering and seed formation states have the future potential to be a useful food ingredient.

This paper describes the use of cyclic voltammetry (CV), spectrophotometric methods [Trolox equivalent antioxidant capacity (TEAC), peroxyl radical trapping capacity (PRTC), DPPH radical scavenging activity (RSA), and Folin-Ciocalteu reagent (FCR) reducing capacity], and photochemiluminescence (PCL) for the measurement of the antioxidant capacity of onion var. Sochaczewska and var. Szalotka. The antioxidant and reducing activity of the dominant onion flavonoids quercetin (Q), quercetin-3- O-beta-glucoside (Q3G), quercetin-4'- O-beta-glucoside (Q4'G), and quercetin-3,4'-di- O-beta-glucoside (Q3,4'G) were determined by spectrophotometric (TEAC and PRTC) and CV methods, respectively. The contribution of quercetin and its glucosides to the antioxidant capacity of onion was calculated in consequence of the qualitative and quantitative analysis of onion flavonoids by high-performance liquid chromatography-ultraviolet-mass spectrometry. The dominant forms of quercetin in the onion var. Sochaczewska and Szalotka included Q4'G (61 and 54%), Q3,4'G (37 and 44%), Q3G (1.4 and 1.1%), and free quercetin (1.1 and 0.7%), respectively. The CV experiment showed the highest reducing activity of Q while Q3G, Q4'G, and Q3,4'G exhibited about 68, 51, and 30% of the reducing power noted for Q. The order of the reducing activity of onion flavonoids was confirmed by their free radical scavenging activity and evaluated by TEAC and PRTC assays as follows: Q > Q3G > Q4'G > Q3,4'G. The Q4'G and Q3,4'G showed poor antioxidant activity under both applied spectrophotometric assays but still exhibited reducing activity based on CV experiments. The reducing capacity of onions determined by CV method was twice higher than the antioxidant capacity formed by water-soluble compounds (ACW) evaluated by PCL, and it was about 50% higher than PRTC and DPPH RSA results and the converted FCR reducing capacity. In contrast, the reducing capacity of onions determined by the CV method was 3-fold and about four times lower when compared to the antioxidant capacity evaluated by the TEAC method and that formed by lipid-soluble compounds (ACL) provided by PCL, respectively. The highest antioxidant capacity of onion was found under cumulative consideration of PCL (ACW + ACL) and TEAC assays. The relative contribution of Q and its glucosides to the antioxidant capacity of onions showed a low contribution of Q, Q3G, and Q3,4'G derived from CV, TEAC, and PRTC assays while the highest contribution to the antioxidant capacity of onions was provided by Q4'G.

This paper describes the use of cyclic voltammetry, photochemiluminescence (PCL), and spectrophotometric methods (TEAC and FCR reducing capacity) for the measurement of the antioxidant capacity of dark- and light-grown buckwheat sprouts. Moreover, the flavonoid profile of studied material is provided. Methanol extracts (80%) from ungerminated buckwheat grain and 6 and 8 DAS (days after seeding) sprouts were used. The 8 day germination period was sufficient to obtain good quality sprouts with completely removed pericarps. The ungerminated buckwheat grain contained only rutin, whereas in sprouts produced in dark or light, a high level of isoorientin, orientin, vitexin, rutin, and isovitexin was found. The flavonoid content in sprouts produced under light was almost 2 times higher than those of sprouts produced in the dark. The antioxidant capacity of light-grown sprouts was higher than that of dark-grown ones. The results from voltammetric experiments obtained for buckwheat seeds and 6 and 8 DAS sprouts harvested under dark or light conditions highly correlated with those obtained by PCL antioxidant capacity of water-soluble substances (PCL ACW) (r = 0.99), PCL antioxidant capacity of lipid-soluble substances (r = 0.99), TEAC (r = 0.99), and FCR reducing capacity (r = 0.99). The use of cyclic voltammetry, PCL ACW, and TEAC was fully applicable for the evaluation of the antioxidant capacity of buckwheat sprouts.

This paper reports the use of spectrophotometric and voltammetric methods for the determination of the antioxidant capacity of buckwheat and its products originated from a technological line of a buckwheat roasted groats producer. 80% methanol extracts from raw and roasted buckwheat and groats and hulls obtained from roasted buckwheat were used. The spectrophotometric methods included (1) free radical scavenging activities of the extracts against ABTS*+ radical cation (TEAC) and 2,2-diphenyl-1-picrylhydrazyl radical (DPPH RSA) and (2) determination of reducing capacity by the means of Folin-Ciocalteu reagent (FCR) application. The radical scavenging activities of the extracts were also investigated using a voltammetric assay. Moreover, the flavonoids profiles of the studied materials were provided. Buckwheat roasting caused a decrease in TEAC, DPPH RSA, and FCR reducing capacity by 70%. The lowest TEAC, DPPH RSA, and FCR reducing capacities were noted for roasted groats. Both DPPH RSA and TEAC methods were highly positively correlated with the FCR reducing capacity assay (r = 0.98 and r = 0.99). Cyclic voltammograms of analyzed buckwheat extracts were useful for evaluation of the antioxidant capacity. The total charge below the anodic current waveform was correlated with the data obtained by TEAC (r = 0.770), DPPH RSA (r = 0.88), and FCR reducing capacity (r = 0.81). The changes in the antioxidant capacity of buckwheat and its products followed the changes in flavonoids composition. In particular, the concentration of flavonoids was related to measurements by cyclic voltammetry.
