ABSTRACT: This study aimed to evaluate the diagnostic performance of diffusion kurtosis imaging (DKI) and prostate-specific antigen (PSA) biomarkers in differentiating prostate cancer (PCa) and benign prostatic hyperplasia (BPH).A total of 43 cases of prostate diseases verified by pathology were enrolled in the present study. These cases were assigned to the BPH group (n = 20, 68.85±10.81 years old) and PCa group (n = 23, 74.13 ±â€Š7.37 years old). All patients underwent routine prostate magnetic resonance imaging and DKI examinations, and the mean diffusivity (MD), mean kurtosis (MK), and fractional anisotropy (FA) values were calculated. Three serum indicators (PSA, free PSA [fPSA], and f/t PSA) were collected. We used univariate logistic regression to analyze the above quantitative parameters between the 2 groups, and the independent factors were further incorporated into the multivariate logistic regression model. The area under the receiver operating characteristic curve (AUC) was used to evaluate the diagnostic efficacy of the single indicator and combined model.The difference in PSA, f/t PSA, MK, and FA between PCa and BPH was statistically significant (P < .05). The AUC for the combined model (f/t PSA, MK, and FA) of 0.972 (95% confidence interval [CI]: 0.928, 1.000) was higher than the AUC of 0.902 (95% CI: 0.801, 1.000) for f/t PSA, 0.833 (95% CI: 0.707, 0.958) for MK, and 0.807 (95% CI: 0.679, 0.934) for FA.The MK and FA values for DKI and f/t PSA effectively identify PCa and BPH, compared to the PSA indicators. Combining DKI and PSA derivatives can further improve the diagnosis efficiency and might help in the clinical setting.

OBJECTIVE: To investigate the diagnostic value of quantitative parameters of intravoxel incoherent motion diffusion-weighted imaging (IVIM-DWI) in prostate cancer. METHODS: From January 2019 to June 2020, 96 patients with prostatic tumor admitted in the department of urological surgery of our hospital were selected as subjects. Magnetic resonance imaging data of 48 cases of benign prostatic hyperplasia and 48 cases of prostate cancer were retrospectively analyzed. The patients included in this study received conventional MRI and IVIM-DWI examinations. Quantitative parameters of IVIM-DWI including D value, D* value, apparent diffusion coefficient (ADC) value and f value in lesions of prostatic tumor were calculated through the double exponential model fitting algorithm. D value, D* value, ADC value and f value were compared between benign prostatic hyperplasia (BPH) group and prostate cancer group. Quantitative parameters of IVIM-DWI were also compared among patients from different Gleason scores groups. The correlation of quantitative parameters of IVIM-DWI with Gleason scores and PSA concentration was analyzed. Diagnostic efficiency of quantitative parameters of IVIM-DWI for prostate cancer was evaluated by ROC curve. RESULTS: Compared with those in BPH group, D value, ADC value and f value in prostate cancer group were significantly lower, but D* value was obviously higher. With the Gleason score increased, D value, ADC value and f value gradually decreased, while D* value gradually increased. The diagnostic efficiency of parameters ADC and D was higher among other parameters. D value, ADC value and f value of prostate cancer were negatively correlated with Gleason score and PSA concentration, respectively (all P<0.05), while D* value was positively correlated with Gleason score and PSA concentration. CONCLUSIONS: Quantitative parameters of IVIM-DWI could be used for the diagnosis and evaluation of prostate cancer, and quantitative parameters of IVIM-DWI were associated with Gleason score and PSA concentration.

AIMS: Parenting interventions in this review refer to supportive parenting training provided for parents or primary caregivers of children and adolescents with type 1 diabetes mellitus (T1DM). The review aimed to synthesize evidence about parenting interventions in parents or caregivers of children and adolescents with T1DM, and to evaluate the effect of interventions in reducing parents' or caregivers' psychological distress, helping them share diabetes management responsibility, seek social support, and improve their quality of life. METHODS: We searched PubMed, MEDLINE, EMBASE, CINAHL, Cochrane, and Web of Science from January 1978 to October 2018. Randomized controlled trials (RCTs) comparing an intervention group of parenting programs with a control group of usual care were included. The primary outcomes were stress, family responsibility and conflict, and social support. Secondary outcomes included other psychological index and quality of life. Pooled effect sizes of weighted mean difference (WMD) were calculated. RESULTS: A total of 17 RCTs with 962 participants met the inclusion criteria. Findings of the meta-analysis showed parenting interventions could significantly reduce parents' depression (WMD = -5.78, 95% CI: -6.23 to -5.33, I2  = 0%) and distress (WMD = -5.28, 95% CI: -10.31 to -.25, I2  = 0%), and help them ask for positive social support (WMD = .83, 95% CI: .03 to 1.64, I2  = 0%). No beneficial changes of other outcomes were found. LINKING EVIDENCE TO ACTION: Parents of children and adolescents with T1DM need support from the multidisciplinary team in health care, especially in mental health, family management of childhood diabetes, and social support. Parenting interventions may help parents reduce psychological distress and depression and assist them to ask for social support. Future research should include well-designed RCTs with large samples, appropriate measures with clear definitions, objective assessment, and separation of effects on mothers and fathers.

FERMT3, also known as kindlin-3, is one of three kindlin family members expressed in mammals. Kindlins are cytosolic, adaptor proteins that are important activators and regulators of integrin function. They have also been shown to play critical roles in the development and progression of various cancers. In the present study, we hypothesized that FERMT3 would enhance glioblastoma multiforme (GBM) cell survival. Indeed, expression level analyses showed significant FERMT3 upregulation in human glioma tissues as compared to normal brain tissues. The effect was particularly pronounced in high-grade gliomas. We then demonstrated that FERMT3 knockdown suppresses glioma cell proliferation and chemoresistance to temozolomide (TMZ). To determine the mechanism by which FERMT3 enhances glioma cell proliferation and chemoresistance, we examined the effects of FERMT3 on integrin activation and Wnt/ß-catenin signaling. Through the use of western blot assays and TOPflash and FOPflash plasmid transfection into glioma cells lines, we demonstrated that FERMT3 regulates glioma cell activity through integrin-mediated Wnt/ß-catenin signaling. These results suggest that FERMT3 activates integrin activity in high-grade gliomas to enhance glioma cell survival and chemoresistance. The present study thus indicates a potential role for FERMT3 as a genetic target in the treatment of GBM.

AIM: To evaluate the effect of intensive lifestyle intervention on patients with isolated impaired fasting glucose. BACKGROUND: The application of lifestyle intervention to clinical nursing can help improve outcomes of patients with impaired fasting glucose. DESIGN: We performed a meta-analysis of randomized controlled trials using methods outlined in the Cochrane Handbook. METHODS: We performed keyword searches of the literature databases up to September 2014 and performed manual searches of selected article reference lists. Data were extracted by the lead reviewer, cross-checked by the second reviewer, entered into RevMan5·3, and pooled and analysed using the fixed effect inverse-variance model or random effects model to determine the mean difference and calculate the weighted mean difference. The heterogeneity statistic (I2 ), overall effect (Z) and P-value were calculated. RESULTS: A total of five studies were included in our analysis. Following the heterogeneity test statistic, overall effect Z score with P-value was calculated. The targets of fasting plasma glucose, weight, body mass index, triglycerides, high-density lipoprotein cholesterol and total cholesterol all showed significant differences between the intervention and control groups. CONCLUSION: Lifestyle intervention can improve glycaemic control in patients with isolated impaired fasting glucose. It also reduces blood-lipid levels and promotes weight loss. Larger patient cohorts and adequate follow-up should be included in future trials.

The wheat haploid plantlets from anther culture were of theoretical and applied interest. Induced haploid spirophytes may play a key role, not only in increasing the efficiency of breeding programs by producing pure lines rapidly, but also in the elucidation of fundamental genetics. However large number of experiments turned out that there are many problems with low rate of green plantlet from anther culture, difficulties of saving plantlets though summer, low rate of double chromosome, extrusive contradiction between technique of wheat anther culture and variety improvement. So wheat anther culture has not played a role in wheat breeding. Studies were carried out on the relationship between the breeding efficiency of anther culture and the use of different hybrid generations as anther-donor plants in wheat. It was shown that previously the low breeding efficiency of anther culture in wheat cross breeding was mainly due to the use of F1 as anther donors. Experimental results demonstrated that the breeding efficiency of anther culture in F1 generation was very low, especially when the doubled haploid population was very small. Based on the experimental results as well as the theoretical analysis this paper proposes that the use of F2 or F3 (especially F3) as anther donors in combination with the conventional selection in the fields will increase the breeding efficiency of anther culture. Under the guidance of this proposal two wheat varieties have been developed within 5-7 years through anther culture.
