The objective of the present study was to determine the effects of muramidase (MUR) supplemented to diets formulated with different fiber sources (inert or fermentable) on the growth performance and intestinal parameters of broiler chickens raised under different coccidiosis management programs. A total of 2,208 male Ross 308 broilers were housed in 96 floor pens and distributed into a 2 × 3 × 2 factorial arrangement in a completely randomized block design with 2 sources of fiber (inert or fermentable fiber), 3 coccidiosis management programs (none, vaccine, or Salinomycin), and with or without supplementation of MUR at 35,000 LSU(F)/kg of diet. Body weight gain (BWG), feed intake (FI), and feed conversion ratio (FCR) were calculated for each feeding phase (d 0-14, d 14-28, d 28-36) and from d 0 to 36. On d 17 and d 31, samples were taken to analyze several parameters. The experimental data were analyzed with 3-way ANOVA considering the main effect of fiber source, coccidiosis program, inclusion of MUR, and their interactions using JMP 16.2. 16S rDNA sequencing of the ileal and cecal content was carried out to analyze the diversity, composition, and predictive function of the microbiota. From d 0 to 36, BWG increased (P = 0.05) by 2.5% in birds supplemented with Salinomycin (P = 0.04), and by 2.2% with MUR supplementation (P = 0.02). Salinomycin and MUR improved FCR (P < 0.0001) when compared to nonsupplemented birds. The supplementation of MUR, regardless of the coccidiosis management program, reduced the intestinal viscosity (P = 0.03). On d 31, the highest blood concentration of carotenoids was observed in chickens fed diets supplemented with Salinomycin. MUR led to significant changes in the diversity, composition, and predictive function of the ileal microbiota, mainly on d 31. The results observed herein further explain the positive effects of MUR on the growth performance of broiler chickens.

Several factors predisposing to necrotic enteritis (NE) have been identified, including diet and Eimeria spp. infestations. Coccidiosis vaccines are indicated to decrease the intestinal lesions caused by specific Eimeria species that are a known predisposing factor to NE and, consequently, these vaccines could be a holistic approach to the control of NE disease and an alternative solution to coccidiostats. Besides, feed additives have also gained special attention from the poultry industry as an alternative solution to antibiotics to prevent NE as well as other bacterial enteritis. Then, the combination of vaccination against coccidiosis and the supplementation of the diet with feed additives could be a composite approach to the control of NE problems triggered by Eimeria spp. infestation. The objective of this study was to test the efficacy of an attenuated coccidiosis vaccine (EVANT) in combination with different feed additives to prevent the loss of production performance and intestinal lesions in broilers challenged with NE. Healthy day-old broilers (n = 960) were randomly allocated to 6 groups (8 cages/group). Groups 1-2 were left unvaccinated. Groups 3-6 were vaccinated following the manufacturer's instructions. Chickens were grown using a diet favoring the intestinal proliferation of Clostridium perfringens. Moreover, the diets of groups 4-6 were supplemented with medium chain fatty acids (MCFA), butyric acid or phytogenic feed additives (PFA), respectively. A NE infection model was used to challenge groups 2-6; chickens were orally infected with Eimeria maxima (4,500 oocysts) and then C. perfringens (108 CFU) at 15 and 20 d, respectively. Birds were monitored and productive parameters recorded until 42 d; intestinal lesions were scored. Results showed that coccidiosis vaccination, with or without the addition of feed additives, decreased intestinal lesions associated with NE and improved the performance of the birds. Besides, the addition of MCFA to the diet decreased intestinal lesions associated to NE in vaccinated animals compared to all treatment groups. Moreover, the same additive improved the feed conversion rate. Therefore, vaccination with a live attenuated coccidiosis vaccine together with in-feed inclusion of MCFA might be a solution to reduce NE in broilers raised antimicrobial- and coccidiostat-free.

Mycotoxin exposure is common in the poultry industry. Deoxynivalenol (DON) is usually detected at levels below the maximum threshold (5000 ppb), but depending on diet and age, broiler performance can be affected. We evaluated the effects of 900 ppb and 2300 ppb DON on the performance, intestinal morphometry, and lesion scores of broiler chickens. One-day-old male Ross broilers (n = 736) were divided into 4 treatments with 8 replicates each, and a pen containing 23 birds was the experimental unit. The animals were fed diets naturally contaminated with two levels of DON: 900 (Low DON-LD) or 2300 (Moderate DON-MD) ppb, with or without activated charcoal, over 28 days. After this, all birds were fed a marginally DON-contaminated diet without charcoal. During the first 28 days, body weight gain (BWG) and feed conversion ratio (FCR) were significantly impaired when broilers were fed a MD diet without activated charcoal. Even after feeding a marginally contaminated diet from D28-35, birds previously fed the MD diet presented a significantly lower performance. The villus height:crypt depth (VH:CD) ratio was significantly higher in the ileum from 14-day-old broilers fed the MD when compared with the LD diet. At D28, the MD diet caused decreased villus height (VH) and increased crypt depth (CD), affecting VH:CD ratio in both intestinal segments, with higher levels in the jejunum from 28-day-old broilers fed a non-supplemented LD diet. Broiler production was negatively affected by DON, even at moderate levels (2300 ppb).

A surface plasmon resonance (SPR) biosensor assay was developed on the basis of a lipopolysaccharide antigen of Salmonella enterica serovar enteritidis (S. enterica serovar enteritidis) to detect egg yolk antibodies against S. enterica serovar enteritidis. This biosensor assay was compared to two commercial ELISA kits based on LPS antigen and flagellar antigen. A number of 163 egg yolk and combined egg white and yolk samples from chickens experimentally infected with S. enterica serovar enteritidis and 90 egg yolk and combined egg white and yolk samples from uninfected chickens were analyzed. Receiver operating characteristic analysis of the data calculated a diagnostic sensitivity of 82% and a diagnostic specificity of 100%. The within-day coefficient of variation of a positive internal-control egg yolk was 1%. The SPR biosensor assay was able to detect antibodies in a significantly higher percentage of known positive samples than the commercial ELISA's. The anticipated use of the SPR biosensor assay is to determine the S. enterica serovar enteritidis serostatus of non-vaccinated layer hens.
