RESUMO

Estudou-se o efeito do hipotireoidismo materno na expressão espaço-temporal de mediadores imunológicos e na população de células natural killers (NK) na decídua e na glândula metrial de ratas durante a gestação. Avaliou-se a detecção imunoistoquímica de interferon γ (IFNγ), do fator inibidor de migração (MIF), da interleucina 15 (IL15), do óxido nítrico sintase induzível (iNOS), a marcação com lectina DBA para evidenciação das células NK uterinas DBA+ e a expressão gênica de Ifnγ e Nos2. O hipotireoidismo aumentou o iNOS aos sete dias, a IL15 e o MIF aos 10 e 12 dias, o IFNγ e o MIF aos 14 DG e a expressão dos transcritos gênicos para iNos aos 12 e 19 dias e para Ifnγ aos 14 DG. O hipotireoidismo reduziu a imunomarcação de MIF e lectina DBA aos sete dias, lectina DBA aos 10 e 14 DG, IFNγ aos 12 dias, e a expressão de Ifnγ aos 10 e 19 DG e de iNOS aos 12, 14 e 19 DG, bem como reduziu seus transcritos gênicos aos 10 e 14 DG. Conclui-se que o hipotireoidismo compromete o perfil imunológico na interface materno-fetal ao longo da gestação, particularmente por reduzir o fator anti-inflamatório iNOS e a população de células uNK DBA+.(AU)


The goal of this study was to evaluate the effect of maternal hypothyroidism on the spatiotemporal expression of immunological mediators and population of Natural Killers cells in decidua and metrial gland of rats. Interferon gamma (IFNγ), migration inhibitory factor (MIF), interleukin 15 (IL15), inducible nitric oxide synthase (iNOS), and DBA-Lectin labeling for evidence of uNK DBA+ cells in decidua and genetic expression of Ifnγ and iNos by real-time RT-PCR were evaluated. Hypothyroidism increased protein expression of iNOS at 7 days, IL15 and MIF at 10 and 12 days, IFNγ and MIF at 14 DG in the decidua and/or metrial gland and the gene transcripts for iNOS at 12 and 19 days and for Inf at 14 DG. In addition, hypothyroidism reduced the protein expression of MIF and DBA-Lectin at 7 days, DBA-Lectin at 10 and 14 DG, IFNγ at 12 days, and the gene transcript to Ifnγ at 10 and 19 DGs. Hypothyroidism also reduced the protein expression of iNOS at 12, 14 and 19 DG and reduced its gene transcripts at 10 and 14 DGs. It is concluded that hypothyroidism compromises the immunology profile at the maternal-fetal interface throughout pregnancy, particularly by reducing the anti-inflammatory factor iNOS and population of uNK DBA+ cells.(AU)

Vomiting and diarrhea are two important clinical signs that can cause significant electrolytic and acid-base imbalances. The purposes of this study were to characterize hydric, electrolytic and acid-base disorders presented by puppies with hemorrhagic gastroenteritis and to compare the traditional and quantitative approaches to acid-base status interpretation. Sixty-one animals with a history of vomiting and/or diarrhea were used in this study and the following tests were performed: complete blood count, total plasma protein concentration and hemogasometry. Mean, standard deviation and Kappa values were calculated. The imbalances characterized by both approaches were: 42 (69%) animals without imbalance, 17 (28%) with metabolic alkalosis and 2 (3%) with metabolic acidosis by the traditional approach and 17 (28%) dogs without imbalance, 26 (43%) with metabolic alkalosis and 18 (29%) with metabolic acidosis by the quantitative approach. The agreement calculated between two approaches coincide in 28 cases, with a moderate Kappa value equivalent to 0.459. The most found imbalances were metabolic alkalosis, hypokalemia, and mild dehydration. Most of acid-base disturbances were not identified by the traditional approach, whereas by the quantitative approach, they were easily determined. Thus quantitative approach proved to be superior in identification of possible acid-base imbalances.(AU)

Os objetivos deste trabalho foram caracterizar os desequilíbrios hídrico, eletrolítico e ácido-base apresentados por filhotes de cães com gastroenterite hemorrágica e comparar as abordagens de interpretação do equilíbrio ácido-base tradicional e quantitativa. Sessenta e um animais com histórico de vômito e/ou diarreia foram utilizados neste estudo, e os seguintes testes foram realizados: hemograma, concentração de proteína total plasmática e hemogasometria. Média, desvio-padrão e valor de Kappa foram calculados. Os desequilíbrios caracterizados pelas duas abordagens foram: 42 (69%) animais sem desequilíbrio, 17 (28%) com alcalose metabólica e dois (3%) com acidose metabólica, pela abordagem tradicional, e 17 (28%) cães sem desequilíbrio, 26 (43%) com alcalose metabólica e 18 (29%) com acidose metabólica, pela abordagem quantitativa. A concordância calculada entre as duas abordagens coincidiu em 28 casos, com um valor de Kappa moderado, equivalente a 0,459. Os desequilíbrios mais encontrados foram alcalose metabólica, hipocalemia e desidratação leve. A maioria dos distúrbios não foram identificados pela abordagem tradicional, enquanto, pela abordagem quantitativa, foram facilmente determinados. Portanto, a abordagem quantitativa provou-se superior na identificação de possíveis desequilíbrios ácido-base.(AU)

Common bean (Phaseolus vulgaris L., Fabaceae) is a globally important staple crop, which is an important source of calories, protein and essential micronutrients. At the genomic level little is known regarding the small non-coding RNAs within the common bean genome. One of the most important classes of such small non-coding RNAs is microRNAs (miRNAs), which control mRNA and protein expression levels in many eukaryotes. Computational methods have been applied to identify putative miRNAs in the genomes of different organisms. In this study, our objective was to comprehensively identify and characterise miRNAs from the genome and transcriptome of P. vulgaris, including both mature and precursor miRNA forms. We also sought to identify the putative proteins involved in miRNA processing and the likely target genes of common bean miRNAs. We identified 221 mature miRNAs and 136 precursor miRNAs distributed across 52 different miRNA families in the P. vulgaris genome. Amongst these, we distinguished 129 novel mature miRNAs and 123 miRNA precursors belonging to 24 different miRNA families. We also identified 31 proteins predicted to participate in the miRNA-processing pathway in P. vulgaris. Finally, we also identified 483 predicted miRNA targets, including many which corroborate results from other species, suggesting that miRNA regulatory systems are evolutionarily conserved and important for plant development. Our results expand the study of miRNAs and their target genes in common bean, and provide new opportunities to understand their roles in the biology of this important staple crop.

Seed germination and dormancy are complex phenomena that are controlled by many genes and environmental factors. Such genes are indicated by phytohormones that interact with each other, and may cause dormancy or promote seed germination. The objective of this study was to investigate gene expression associated with the biosynthetic pathways of abscisic acid (ABA), gibberellic acid (GA), and ethylene (ET) in dormant and germinated lettuce seeds. The expressions of LsNCED, LsGA3ox1, and ACO-B were evaluated in germinating and dormant seeds from the cultivars Everglades, Babá de Verão, Verônica, Salinas, Colorado, and Regina 71. The expressions of LsNCED, LsGA3ox1, and ACO-B were related to the biosynthesis of ABA, GA, and ET, respectively; therefore, the presence of these substances depends on genotype. LsNCED expression only occurred in dormant seeds, and was connected to dormancy. LsGA3ox1expression only occurred in germinated seeds, and was connected to germination. The ACO-B gene was involved in ET biosynthesis, and was expressed differently in germinated and dormant seeds, depending on the genotype, indicating different functions for different characteristics. Furthermore, sensitivity to phytohormones appeared to be more important than the expression levels of LsNCED, LsGA3ox1, or ACO-B.

O objetivo deste estudo foi analisar taxas de prevalências dos tipos de aumentos de volume em pele, glândulas sebáceas, sudoríparas e mamárias de cães atendidos no Hospital Veterinário da Universidade Estadual de Londrina/PR, em relação à raça, gênero e idade, no período de janeiro de 2008 a janeiro de 2013. Os dados foram obtidos a partir de registros do Laboratório de Anatomia Patológica desse Hospital. Foram estudados 1906 cães, observando-se 712 (37,4%) cães sem raça definida (SRD) e 1394 (73%) fêmeas. Notou-se que em 29 (65%) das 45 raças, a idade média de acometimento foi maior que 8 anos. Foram analisados 2296 aumentos de volume, sendo diagnosticados 1978 (86,2%) tumores neoplásicos e 318 (13,8%) não neoplásicos. Dentre os neoplásicos, 1259 (63,7%) eram de origem epitelial, 592 (29,9%) mesenquimal e 73 (3,7%) melanocítica; foram encontradas 54 (2,7%) neoplasias de origem inconclusiva. RESUMO

A central primitive neuroectodermal tumour (PNET) with ependymal differentiation was identified in an 18-month-old female Australian terrier dog that died following signs of central nervous system disease. A large space-occupying haemorrhagic mass replaced the thalamus and part of the hypothalamus of the brain. Microscopical examination revealed a tumour formed by a compact sheet of small cells that contained ependymal and perivascular pseudorosettes. Immunohistochemically, the neoplastic cells variably expressed glial fibrillary acidic protein, neuron-specific enolase, vimentin and cytokeratin, consistent with divergent differentiation of the tumour. This case is thought to represent the first description of PNET with ependymal differentiation in a dog.

RESUMO

A study was undertaken during November 2005 to October 2006 to assess about utilization aspects of ICTC at a tertiary care teaching hospital and a government district hospital in Karnataka. The average monthly numbers of persons visiting ICTC per month at the tertiary care teaching hospital (509) were higher than that of Government district hospital (222). However, the average monthly number of direct walk-in persons and follow up, attending the centre at district hospital were higher than that of teaching hospital. Thus optimal use of information, education and communication (IEC) techniques needs to be strengthened to improve the utilization of ICTC services at the tertiary care teaching hospital.

The inheritance of the tendency to set parthenocarpic fruit in the summer squash (Cucurbita pepo L.) line Whitaker was studied. Two parental lines, Whitaker (parthenocarpic) and Caserta (non-parthenocarpic), and the F1 and F2 generations and backcrosses to both parents were tested. The parthenocarpic tendency of individual plants was scored on a scale from 1 (non-parthenocarpic fruit) to 5 (parthenocarpic fruit). The Whitaker line produced parthenocarpic fruit and had a mean score of 4.2, whereas Caserta did not set parthenocarpic fruit and had a score of 1.55. The heritability estimates indicated that genetic gains from selection were feasible. The additive-dominant model showed a good fit, with epistasis being negligible or nonexistent. The hypothesis of monogenic inheritance with incomplete dominance was not rejected within the degree of dominance range from 0.2 to 0.5. These results indicate that parthenocarpy is controlled by a single locus, with incomplete dominance in the direction of parthenocarpic expression

The effects of the fruit ripening mutant gene alcobaça (alc) and color development mutants, old gold-crimson (ogc) and high pigment (hp), on yield and post-harvest quality of tomato fruits were investigated. Five tomato hybrids were obtained by crossing near isogenic lines with Flora-Dade background [Flora-Dade (alc+/alc+ ogc+/ogc+ hp+/hp+), TOM-559 (alc/alc ogc+/ogc+ hp+/hp+), TOM-591 (alc/alc ogc/ogc hp+/hp+), TOM-593 (alc/alc ogc+/ogc+ hp/hp), and TOM-589 (alc/alc ogc/ogc hp/hp)] with the pollen parent line Mospomorist (alc+/alc+ ogc+/ogc+ hp+/hp+). Hybrid fruit was harvested at the breaker stage and stored on shelves at 15oC and 60 relative humidity for 16 days, and then evaluated for firmness, development of red color, and carotenoid contents. The different genotypic combinations at the loci alc, ogc and hp had no effect on fruit yield. The alc+/alc hybrid genotype significantly increased fruit firmness and significantly delayed the development of red color in maturing fruit. Simultaneous usage of ogc+/ogc and hp+/hp promoted an increase in the red color and lycopene content of alc+/alc hybrids, but did not have any additional effect on fruit firmness

RESUMO

The effect of an original drug Kagocel on the reproduction of Herpes simplex virus including its mutant strains resistant to basic antiherpetic medicine Acyclovir was evaluated. Kagocel was shown to have a low cytotoxic effect on Vero cell and inhibited reproduction of Herpes virus type 1 and Herpes virus type 2 in noncytotoxic concentrations. Kagocel was also demonstrated to inhibit the reproduction of Herpes virus type 1, resistant to combination of Acyclovir and phosphonoacetic acid. Detection of direct antiherpetic activity of Kagocel in vitro opens good perspectives to its clinical application, especially in combination with Acyclovir, the medication with other antiherpetic mechanism of action.

Resistance to the root-knot nematodes Meloidogyne spp. would be a valuable attribute of lettuce Lactuca sativa L. cultivars grown in tropical regions. The looseleaf lettuce 'Grand Rapids' is resistant to both M. incognita and M. javanica. Resistance to M. incognita has a high heritability, under the control of a single gene locus, in which the 'Grand Rapids' allele, responsible for resistance (Me), has predominantly additive gene action, and has incomplete penetrance and variable expressivity. We studied the inheritance of the resistance of 'Grand Rapids' (P(2)) to M. javanica in a cross with a standard nematode-susceptible cultivar Regina-71 (P(1)). F(1)(Regina-71 x Grand Rapids) and F(2) seed were obtained, and the F(2) inoculated, along with the parental cultivars, with a known isolate of M. javanica to evaluate nematode resistance. A high broad sense heritability estimate (0.798) was obtained for gall indices. Class distributions of gall indices for generations P(1), P(2), and F(2) were in agreement with theoretical distributions based on a monogenic inheritance model for the range of assumed degrees of dominance between approximately -0.20 and 0.20. M. javanica resistance appears to be under control of a single gene locus, with predominantly additive gene action. Whether or not the Grand Rapids allele imparting resistance to M. javanica is the same Me allele imparting resistance to M. incognita remains to be determined

The identification of arthropod bloodmeals is important in many epidemiological studies, as, the understanding of the life cycle of vectors and the pathogens they transmit, as well as helping to define arthropods' control strategies. The precipitin test has been used for decades, but ELISA is slowly becoming more popular. To compare the two tests for sensitivity, specificity and accuracy to detect small insect bloodmeals, Aedes aegypti or Ae. fluviatilis mosquitoes were fed either on feline, canine or human hosts. Mosquitoes were frozen at 6, 12, 24, 48 or 72 h after feeding. Precipitin test showed better specificity and accuracy and ELISA test showed higher sensitivity. Better results with both tests were achieved when mosquitoes were frozen within 48 h from feeding.

RESUMO

The results of the study of the phagocytic element of immunity in cases of primary immunodeficiency, made with the use of a complex of functional tests, are presented. As shown in this study, in patients with definitely characterized phagocytic defect (chronic granulomatous disease) a decrease in all parameters under study, with the exception of phagocyte index, is observed. In patients with immunodeficient states of nonphagocytic nature moderate mosaic deviations of phagocytosis characteristics are noted. For further and more profound study of phagocytosis disturbances in such patients, it is expedient, in particular, to carry out studies in the activity of myeloperoxidaze and the intensity of oxygen-dependent metabolism. The use of the complex of methods used in this investigation has shown their high diagnostic value. It may be recommended for clinical practice under the condition of strict standardization of experimental techniques.

There were 3 groups of children with recurrent purulent infections, who were entered into randomized clinical studies. Group 1 received sodium nucleinate, group 2 diucifon and group 3 placebo (calcium gluconate). After 6 months a well-defined clinical effect manifested by the lowering of the rate and gravity of exacerbations was recorded in 68% of the patients belonging to groups 1 and 2 and in 39% of group 3 patients, which is much more seldom (p less than 0.05). During immunological studies according to the level I tests, no significant changes in the indicators were recorded either before or after the treatment both among the patients who responded to immunotherapy and among those who did not respond to it. The conclusion is drawn that if indicated clinically, sodium nucleinate of diucifon can be administered in spite of the normal immune status in accordance with the level I tests.

Altogether 79 children with primary immunodeficiency conditions (PIC) and 21 children suffering from frequently occurring bacterial infections were examined for the level of rosette-forming cells and lymphocytes reacting positively with monoclonal antibodies to different phenotypic membranous markers (SD-antigens and others). The patients with combined forms of immunodeficiency, with T deficiency playing the key role manifested a significant lowering of the content of the subpopulations of SD3+, SD5+, SD7+ and SD4+ lymphocytes and the decrease of the SD4/SD8 ratio. The the reduction of the number of B lymphocytes (smIg + K1, SD19 +), the low levels of Ia+ -cells, and the decrease of the SD4/SD8 at the expense of the content of SD8+ -cells. In the patients with selective immunodeficiency and recurrent bacterial infections without verified PIC, these parameters did not undergo any essential changes. As compared with immunofluorescent assays, the measurement of E-RFL and M-RFL amounts was found to have insufficient information content for the diagnosis of PIC.
