RESUMO

The orienting reaction (OR) towards a new stimulus is subject to habituation, i.e., progressively attenuates with stimulus repetition. The skin conductance responses (SCRs) are known to represent a reliable measure of OR at the peripheral level. Yet, it is still a matter of debate which of the P3 subcomponents is the most likely to represent the central counterpart of the OR. The aim of the present work was to study habituation, recovery, and dishabituation phenomena intrinsic to a two-stimulus auditory oddball paradigm, one of the most-used paradigms both in research and clinic, by simultaneously recording SCRs and P3 in twenty healthy volunteers. Our findings show that the target stimulus was capable of triggering a more marked OR, as indexed by both SCRs and P3, compared to the standard stimulus, that could be due to its affective saliency and relevance for task completion; the application of temporal principal components analysis (PCA) to the P3 complex allowed us to identify several subcomponents including both early and late P3a (eP3a; lP3a), P3b, novelty P3 (nP3), and both a positive and a negative Slow Wave (+SW; -SW). Particularly, lP3a and P3b subcomponents showed a similar behavior to that observed for SCRs , suggesting them as central counterparts of OR. Finally, the P3 evoked by the first standard stimulus after the target showed a significant dishabituation phenomenon which could represent a sign of the local stimulus change. However, it did not reach a sufficient level to trigger an SCR/OR since it did not represent a salient event in the context of the task.

Sokolov described both phasic and tonic aspects of the Orienting Reflex (OR), but subsequent research and theory development has focussed primarily on the phasic OR at the expense of the tonic OR. The present study used prestimulus skin conductance level (SCL) during a dishabituation paradigm to model the tonic OR, examining its amplitude patterning over repeated standard stimulus presentations and a change stimulus. We expected sensitisation (increased amplitude) following the initial and change trials, and habituation (decrement) over the intervening trials. Prestimulus EEG alpha level was explored as a potential central measure of the tonic OR (as an inverse correlate), examining its pattern over stimulus repetition and change in relation to the SCL model. We presented a habituation series of innocuous auditory stimuli to two groups (each N = 20) at different ISIs (Long 13-15 s and Short 5-7 s) and recorded electrodermal and EEG data during two counterbalanced conditions; Indifferent: no task requirements; Significant: silent counting. Across groups and conditions, prestimulus SCLs and alpha amplitudes generally showed the expected trials patterns, confirming our main hypotheses. Findings have important implications for including the assessment of Sokolov's tonic OR in modelling central and autonomic nervous system interactions of fundamental attention and learning processes.

Habituation, the process by which animals learn to ignore insignificant stimuli, facilitates engagement with salient features of the environment. However, neural mechanisms underlying habituation also allow responses to familiar stimuli to be reinstated when such stimuli become potentially significant. Thus, the habituated state must allow a mechanism for habituation override. The remarkably precise knowledge of cell identity, connectivity, and information coding in Drosophila sensory circuits, as well as the availability of tools to genetically target these cells, makes Drosophila a valuable and important organism for analysis of habituation and habituation-override mechanisms. Studies of olfactory and gustatory habituation in Drosophila suggest that potentiation of GABAergic neurons underlies certain timescales of habituation and have specified some elements of a gustatory habituation-override pathway. More detailed understanding of gustatory habituation and habituation-override mechanisms will benefit from access to robust behavioral assays for (a) the proboscis extension reflex (PER) elicited by a sweet stimulus, (b) exposure paradigms that result in PER habituation, and, most critically, (c) manipulations that result in PER-habituation override. Here, we describe simple protocols for persistent sucrose exposure of tarsal hairs that lead to habituation of proboscis extension and for presentation of a novel appetitive stimuli that reinstate robust PER to habituated flies. This detailed protocol of gustatory habituation provides (a) a simple method to induce habituation by continuous exposure of the flies to sucrose for 10 min without leading to ingestion and (b) a novel method to override habituation by presenting yeast to the proboscis. Key features • A protocol for stimulation of Drosophila's taste (sugar) sensory neurons that induces gustatory habituation without satiation due to ingestion. • A chemical (yeast) stimulation protocol that rapidly induces habituation override/dishabituation in sugar-habituated Drosophila.

Previous research has suggested that operant response decrements within experimental sessions are due in part to habituation to the repeated presentation of reinforcers. One way to assess the role of habituation in within-session response decrements is to conduct a test for dishabituation, a phenomenon in which a habituated response to a given stimulus recovers following the presentation of some strong or novel stimulus other than the habituated stimulus. Dishabituation of operant responding has been demonstrated on several occasions in the literature, but studies with non-human subjects have thus far been limited to those using rats and pigeons. Two experiments attempting to replicate these findings with mice were conducted. Two groups of mice nose-poked for a sweetened condensed milk/water reinforcer on either a fixed-ratio 4 or variable-interval 15 s schedule of reinforcement. During testing, baseline sessions were then alternated with two test conditions and a control condition. Test conditions included a 5 s auditory stimulus or flashing of the house light presented mid-session. Control conditions were identical to baseline. Dishabituation was not observed for either group in Experiment 1. In Experiment 2, dishabituation was not observed for the fixed-ratio 4 group but was observed for the variable interval 15 s group. Considerations for further study of operant dishabituation in mice are discussed.

Animal vocalisations encode a wide range of biological information about the age, sex, body size, and social status of the emitter. Moreover, vocalisations play a significant role in signalling the identity of the emitter to conspecifics. Recent studies have shown that, in the African penguin (Spheniscus demersus), acoustic cues to individual identity are encoded in the fundamental frequency (F0) and resonance frequencies (formants) of the vocal tract. However, although penguins are known to produce vocalisations where F0 and formants vary among individuals, it remains to be tested whether the receivers can perceive and use such information in the individual recognition process. In this study, using the Habituation-Dishabituation (HD) paradigm, we tested the hypothesis that penguins perceive and respond to a shift of ± 20% (corresponding to the natural inter-individual variation observed in ex-situ colonies) of F0 and formant dispersion (ΔF) of species-specific calls. We found that penguins were more likely to look rapidly and for longer at the source of the sound when F0 and formants of the calls were manipulated, indicating that they could perceive variations of these parameters in the vocal signals. Our findings provide the first experimental evidence that, in the African penguin, listeners can perceive changes in F0 and formants, which can be used by the receiver as potential cues for the individual discrimination of the emitter.

RESUMO

In this review, we take a critical look at the methods used to document habituation and the theoretical assumptions that have been made about it. We point out problems associated with measuring habituation merely as a change over the course of repeated presentations of a stimulus. We argue that a common test procedure is essential to assess the relative magnitudes of habituation learning especially when different training procedures are examined. We further suggest that this would be required in order to draw meaningful conclusions about the conditions for optimizing habituation. We also challenge the view that habituation is nonassociative and consider the implications of various associative learning perspectives not only for context-specific habituation but for encoding a representation of the stimulus. We conclude with our recommendations for future research on habituation and we highlight the need to integrate behavioral and neurobiological studies.

The habituation-dishabituation (H-D) paradigm is an established measure of sensory perception in animals. However, it has rarely been applied to canine olfaction. It proposes that animals will lose interest in, or habituate to, a stimulus after successive exposures but will regain interest in, or dishabituate to, a novel stimulus if they can perceive it. This study assessed an H-D test's practicability to determine dogs' olfactory detection thresholds (ODTs) for a neutral odorant. A random selection of mixed-breed pet dogs (n = 26) participated in two H-D tests in a repeated-measures crossover design. They were first habituated to a carrier odor and then presented with either ascending concentrations of n-amyl acetate in the known ODT range (experimental condition) or repeated carrier odor presentations (control condition). No single odor concentration elicited dishabituation in the majority of the dogs. However, individual dogs dishabituated at differing experimental concentrations significantly more often than in the control condition (p = .012). These findings provide some tentative support for using this method in studying canine olfaction. However, further assessment and refinement are needed before it can be a viable alternative to traditional ODT measurement.

BACKGROUND: Individuals with fetal alcohol spectrum disorders (FASD) often show processing deficits in all sensory modalities. Using an operant light reinforcement model, we tested whether prenatal ethanol exposure (PE) alters operant responding to elicit a contingent sensory stimulus-light onset (turning on the light) and habituation to this behavior in rats. We also explored whether postnatal environmental enrichment could ameliorate PE-induced deficits. METHODS: Pregnant Sprague Dawley rats were gavaged twice/day with 0 or 3 g/kg/treatment ethanol (15% w/v) during gestational days 8-20, mimicking second-trimester heavy PE in humans. The offspring were reared in a standard housing condition or an enriched condition. Adult male and female offspring underwent an operant light reinforcement experiment with either a short-access or a long-access procedure. A dishabituation test was also conducted to characterize the habituation process. RESULTS: In the short-access procedure, PE led to increased operant responding to the contingent light onset in both sexes reared in the standard housing condition. Such an effect was not observed in rats reared in enriched conditions due to an overall decrease in responding. Moreover, rats reared in enriched conditions showed greater short-term habituation. In the long access procedure, PE rats showed increased responding and impaired long-term habituation. The long-access procedure facilitated both short-term and long-term habituation in control and PE rats. CONCLUSION: Prenatal ethanol exposure increases responding to contingent light onset and impairs the long-term habituation process. The PE-induced deficits were ameliorated by rearing in the enriched environment and increasing the duration and frequency of exposure to light onset. The PE-induced effects are like increased sensation-seeking, a subtype of sensory-processing deficit that is often observed in individuals with FASD. Our findings could inform a suitable animal model for investigating the underlying mechanisms and possible intervention strategies for sensory deficits in FASD.

Habituated animals retain a latent capacity for robust engagement with familiar stimuli. In most instances, the ability to override habituation is best explained by postulating that habituation arises from the potentiation of inhibitory inputs onto stimulus-encoding assemblies and that habituation override occurs through disinhibition. Previous work has shown that inhibitory plasticity contributes to specific forms of olfactory and gustatory habituation in Drosophila Here, we analyze how exposure to a novel stimulus causes override of gustatory (proboscis extension reflex; PER) habituation. While brief sucrose contact with tarsal hairs causes naive Drosophila to extend their proboscis, persistent exposure reduces PER to subsequent sucrose stimuli. We show that in so habituated animals, either brief exposure of the proboscis to yeast or direct thermogenetic activation of sensory neurons restores PER response to tarsal sucrose stimulation. Similar override of PER habituation can also be induced by brief thermogenetic activation of a population of tyrosine hydroxylase (TH)-positive neurons, a subset of which send projections to the subesophageal zone (SEZ). Significantly, sensory-neuron induced habituation override requires transmitter release from these TH-positive cells. Treatments that cause override specifically influence the habituated state, with no effect on the naive sucrose response across a range of concentrations. Taken together with other findings, these observations in female flies are consistent with a model in which novel taste stimuli trigger activity in dopaminergic neurons which, directly or indirectly, inhibit GABAergic cells that drive PER habituation. The implications of these findings for general mechanisms of attentional and sensory override of habituation are discussed.SIGNIFICANCE STATEMENT Habituation can be overcome when a new context requires an enhanced response to a familiar stimulus. However, the underlying mechanisms remain incompletely understood. Previous studies have provided evidence that habituation of the sucrose-induced proboscis extension reflex (PER) in Drosophila occurs through potentiation of inhibition onto the PER pathway. This work defines controlled protocols for override of PER habituation and uses them to outline the underlying circuit mechanisms. The results presented support a model in which novel taste stimuli cause dishabituation by activating a subset of tyrosine hydroxylase (TH)-expressing neurons that inhibit GABAergic neurons whose potentiation underlies PER habituation. At a general level, these findings further highlight a central role for inhibition and disinhibition in the control of behavioral flexibility.

Previous studies were not able to show that presentation of change stimuli leads to dishabituation of the auditory evoked potential (AEP) component N1 for repeated stimuli. However, these change stimuli were usually themselves repeatedly presented. Here, we tested whether the presentation of non-repeating distractor stimuli ('novels') would lead to N1 dishabituation. The study sample consisted of 18 healthy participants who had to identify auditory target stimuli (´targets´) among repeated standard stimuli and rare novels. AEPs to standards were separately averaged, depending on the preceding stimulus (standards after standards, standards after targets, and standards after novels) and were compared by F statistics and Bayesian t-test. Moreover, N1 repetition effects within recording blocks were analyzed in single trial analyses. The analyses showed that targets elicited significantly larger N1 amplitudes than standards and standards elicited larger N1 amplitudes than novels. In contrast, the N1 amplitude to standards did not vary with the preceding stimulus. The single trial analyses revealed significant, but similar N1 amplitude decreases within the recording blocks for all standards. The current study revealed no evidence for N1 dishabituation, as the N1 amplitude for standards after novels was not increased as compared to the N1 for standards after standards. Thus, stimulus variation had no impact on the N1 of repeated standards, as also suggested by the single trial analyses. The lack of N1 dishabituation is at odds with the assumption that the N1 amplitude decrease after repeated stimulation results from habituation.

C57BL/6 is the most widely used mouse strain in the laboratories. Two substrains of C57BL/6, C57BL/6J (B6J), and C57BL/6N (B6N) are well-known backgrounds for genetic modification and have been shown difference in quite a few tests, including open field test, rotarod test, and Morris water maze. However, difference between these two substrains in olfaction-dependent behaviors remains unknown. Here, we used olfactory two-alternative choice task, which is modified to have two training stages, to evaluate animals' ability in instrumental learning and olfactory association. In the first (rule learning) stage, the mice were trained to use the operant chamber to collect water rewards. An odor cue was provided in the procedure, with no indication about reward locations. In the following (discrimination learning) stage, two odor cues were provided, with each indicating a specific water port. The animals were rewarded upon correct port choices following cue deliveries. We found that during young adulthood (7-10 weeks old), proportionally more B6J than B6N mice were able to pass rule learning (58.3% vs. 29.2%) and ultimately acquire this task (54.2% vs. 25%), with the two substrains showing similar pass rates in discrimination learning (92.9% vs. 85.7%). Surprisingly, at a more mature age (17 weeks old), this substrain difference disappeared. Mature B6N mice had a significant improvement in pass percentages of rule learning and overall task, whereas similar improvement was not observed in the B6J counterparts. Instead, mature B6J mice had an improved speed in rule learning and overall task. We further examined behavioral patterns of 8-week-old B6J and B6N mice in the olfactory habituation or dishabituation test. We observed normal olfactory habituation from subjects of both substrains, with the B6J mice exhibiting stronger investigative responses to newly presented odorants. These results reveal for the first time that B6J and B6N mice are different in acquisition processes of a behavioral task that requires instrumental learning and olfactory association, and that maturation appears to employ different effects on these two substrains during these processes. Furthermore, young adult B6J and B6N mice might be similar in olfactory habituation but different in the olfactory aspects of novelty seeking.

Sensory specific satiety refers to a decline in the hedonic value of the sensory properties of a particular food as it is consumed. This phenomenon is characterized by a decrement in responding as a consequence of repeated exposure, is stimulus specific, and recovers after time. All these characteristics are shared with the habituation phenomenon and for this reason, habituation has been proposed as the underlying mechanism that explains this eating regulatory system. However, several studies conducted with human models have yielded mixed results. Using rats as experimental subjects, the present study tested the following three characteristics of habituation within a Sensory Specific Satiety (SSS) framework: spontaneous recovery, dishabituation and the distractor effect. Experiment 1 demonstrated the basic effect of SSS and its spontaneous recovery over time. In Experiment 2 we found that the presentation of a dishabituator after a pre-feeding procedure had no impact on the SSS effect. Finally, in Experiment 3 the presence of a distractor during a pre-feeding procedure did not alter the expression of SSS. These results challenge the idea that SSS constitutes a typical case of habituation, at least with the procedure used here.

The decline of response as a consequence of repeated stimulation is known as habituation. The goal of the present experiments was extending the knowledge about habituation of abdominal contractions in the pupa of Tenebrio molitor. Both experiments consisted of two phases. During Phase 1, all groups were exposed to a continuous stimulus (light in Experiment 1 and vibration in Experiment 2). At the beginning of this phase, pupae showed a high number of abdominal contractions. However, during the last minute of Phase 1, the number of abdominal contractions was lower. In the next phase, the pupae were divided in different groups to test for response recovery. We found an increase in the abdominal contractions when subjects were exposed to a different stimulus, be it within the same or in a distinct sensory modality. In addition, we also reported response recovery when the pupae were re-exposed to the original stimuli after a resting period. Results indicate that the increase in responding cannot be explained by either sensory adaptation or fatigue. The findings are consistent with the perspective that suggests that habituation plays a major role in the survival of the species, even in non-feeding developmental stages.

Olfactory dysfunction is commonly observed in patients with obstructive sleep apnea (OSA), which is related to chronic intermittent hypoxia (CIH). OSA patients exhibit alterations in discrimination, identification and odor detection threshold. These olfactory functions strongly rely on neuronal processing within the main olfactory bulb (MOB). However, a direct evaluation of the effects of controlled CIH on olfaction and MOB network activity has not been performed. Here, we used electrophysiological field recordings in vivo to evaluate the effects of 21-day-long CIH on MOB network activity and its response to odors. In addition, we assessed animals´ olfaction with the buried food and habituation/dishabituation tests. We found that mice exposed to CIH show alterations in MOB spontaneous activity in vivo, consisting of a reduction in beta and gamma frequency bands power along with an increase in the theta band power. Likewise, the MOB was less responsive to odor stimulation, since the proportional increase of the power of its population activity in response to four different odorants was smaller than the one observed in control animals. These CIH-induced MOB functional alterations correlate with a reduction in the ability to detect, habituate and discriminate olfactory stimuli. Our findings indicate that CIH generates alterations in the MOB neural network, which could be involved in the olfactory deterioration in patients with OSA.

N1 of event-related potentials (ERPs) is augmented in amplitude in ∼50-150 ms by occasional changes (deviants) in the physical features of a sound repeated at intervals of from ∼400 ms to seconds (standard). The release-from-refractoriness hypothesis links the N1 augmentation to a deviant-feature-specific neural population that is fresh to fully respond as opposed to a standard-feature-specific neural population that is unresponsive due to its post-response refractoriness. The present work explored this hypothesis in the context of ERP studies, behavioral habituation studies and studies on stimulus-specific adaptation (SSA). The idea of hundreds of milliseconds neural population-level refractoriness was observed to be founded upon negative N1 evidence (no observable effect of dishabituating stimuli on N1 to standards - the null hypothesis retained) and merely supported by positive N1 evidence (null hypotheses rejected). This idea was also found to be directly challenged by positive N1 evidence. No conclusive network- or single-neuron-level evidence was found for the refractoriness. Therefore, the validity of the release-from-refractoriness hypothesis of N1 to guide psychophysiological research needs reassessment.

Similar to other event-related potential (ERP) components, the amplitude of the auditory evoked N1 depends on the interstimulus interval (ISI). At ISIs > 0.4 s, the amplitude of the N1 increases with longer ISIs, until it saturates at ISIs around 10 s. This amplitude increase with increasing ISI has been conceptualized as a function of N1 recovery or N1 refractoriness. Habituation (as a simple form of learning) represents an elaborated, opposing account for such stimulus repetition effects. For passive oddball experiments (stimulation protocols with frequent standards and rare deviants), the two accounts make different predictions. According to the habituation account, the presentation of small deviants should lead to an increased N1 for subsequent standards (= dishabituation); according to the N1 refractoriness account, there should be no or just minor effects on the N1. In the current study, we tested these predictions and compared the ERPs to standards after small deviants and to standards preceded by other standards. We observed that the ERPs to standards after small deviants were characterized by a small mismatch negativity with an onset latency > 150 ms, but the N1 to standards after deviants did not differ from the N1 to standards preceded by other standards. This negative finding is in line with other previous studies that were also not able to reveal evidence for N1 dishabituation. Aside from this repeated lack of evidence for dishabituation, the N1 habituation account is challenged by the finding that the N1 decrease is stronger for more intense stimuli. Overall, the current and previous findings are more compatible with the N1 refractoriness account, although the mechanisms underlying N1 refractoriness remain to be elucidated. Knowledge about these mechanisms would also help to understand why N1 deficits in schizophrenia are more pronounced at longer ISIs.

Migraine and epilepsy share in many ways with regard to their clinical presentation and pathophysiological mechanisms. A state of central hyper-excitability or hyper-responsivity is fundamental in their pathophysiology. Antiepileptic drugs (AEDs) have long been used for migraine prevention, other than for seizure control. Evidence from numerous clinical trials has well demonstrated the therapeutic effects of some of the AEDs in this regard. In this paper, we will first discuss the clinical similarity between migraine and epilepsy. Then, to address the rationale of AEDs usage in migraine prevention, we focus on those electrophysiological studies conducted in patients with migraine demonstrating evidence of central hyper-excitability or hyper-responsivity. Although controversies remain, three common findings are derived from the literature: (1) cortical hyper-excitability, (2) impaired intra-cortical inhibition or increased intra-cortical facilitation, and (3) dis-habituation (hyper-responsivity). Each single feature may actually represent one of the different manifestations derived from an overall central excitation/inhibition imbalance. Finally, we review those studies addressing the therapeutic effects of AEDs and the associated alteration in central excitability. These studies provided direct evidence that effective prevention by AEDs is associated with the restoration of excitation/inhibition imbalance.

The ability to recognize and interact with members of the same species is essential for social communication. Investigating the neural substrates of social interest and recognition may offer insights into the behavioral differences present in disorders affecting social behavior. Assays used to study social interest in rodents include the 3-chamber test, a partition test, and a social interaction test. Here, we present a single protocol that can be used to quantify the level of social interest displayed by mice, the ability to distinguish between different individual mice (social recognition), and the level of repetitive self-grooming displayed. In the first part of the protocol, a social habituation/dishabituation test, the time spent by a test mouse sniffing a stimulus mouse is quantified over 9 trials. In the first 8 interactions, the same stimulus mouse is used repeatedly; on the ninth trial, a novel stimulus mouse is presented. Intact social recognition is indicated by a progressive decrease in the investigation time over trials 1-8, and an increase in trial 9. The interval between each social trial is used to quantify self-grooming, a stereotyped repetitive behavior in mice. We also present a method for randomized, blinded analysis of these behaviors to increase rigor and reproducibility of results. Therefore, this single behavioral test enables ready assessment of phenotypes of both social and repetitive behaviors in an integrated manner in the same animals. This feature can be advantageous in understanding interactions between these behaviors and phenotypes in mouse models with genetic variants associated with autism and other neurodevelopmental or neuropsychiatric disorders, which are often characterized by these behavioral differences.

Data from three published studies examining autonomic and ERP measures in variants of a dishabituation paradigm were re-analysed to clarify ambiguous novelty results. The three studies manipulated 1. Novelty, 2. Novelty and Intensity, and 3. Novelty and Significance, in auditory dishabituation paradigms at very long interstimulus intervals (ISIs). The question of whether any single ERP matches SCR as the benchmark for the phasic Orienting Reflex (OR) was also addressed. Finally, we aimed to align the re-analysed measures of this and the previous analyses with processes of Preliminary Process Theory (PPT). The SCR demonstrated decrement, recovery, and dishabituation. A summary temporal PCA extracted PN (Processing Negativity), P3a, P3b, Novelty P3, and classic SW for detailed analysis. P3b and SW showed decrement but no recovery at the change trial, while Respiratory pause (RP) and Novelty P3 demonstrated decrement and recovery, but no dishabituation. Post hoc exploration of observed power versus sample size for each of these findings confirmed their robustness. No decrement for PN was also confirmed. Five autonomic and ERP groupings emerged and aligned with modules of processing in PPT: ECR1 (cardiac deceleration), P1, N1-3, and PN - stimulus registration; RP and Novelty P3 - Novelty registration; Peripheral Vasoconstriction (PVC) and P3b - Intensity registration; and ECR2 (cardiac acceleration) and classic SW - Response system. The SCR was confirmed as the sole index of the phasic OR. The pattern of results for the Late Positive Complex (LPC) components (P3a, P3b, Novelty P3, and SW) suggests each is differentially sensitivity to selective determinants of the phasic OR, and consequently the summary LPC is presented as the most appropriate central index of the phasic OR.

Within a week of fertilization, a zebrafish larva has developed a robust behavioral repertoire that includes the ability to learn about noncontingent stimuli. I begin this paper with a brief review of the t1-t2 framework in which groups receive different experiences at the first time point (t1) followed by a common assessment at the second time point (t2) and the strengths of this framework for studying stimulus learning. I then describe assays that have been implemented within the t1-t2 framework to demonstrate stimulus learning in the developing zebrafish. I discuss how these assays have been used to address three fundamental questions about stimulus learning: What are the conditions for stimulus learning? What is the content of stimulus learning? How is stimulus learning reflected in behavior? For each of these three questions, I also identify those issues regarding stimulus learning in the developing zebrafish that warrant further analysis at the behavioral level.
