RESUMO

Due to the negative impact of Haemonchus contortus in the tropics and subtropics, the detection of serum protein profiles that occur in infected sheep is of high relevance for targeted selective treatment strategies (TST). Herein, we integrated proteomics with phenotypic traits to elucidate physiological mechanisms associated to H. contortus infection in susceptible (Dorper - D) and resistant (Santa Inês - S) sheep breeds. Naïve female lambs were infected with H. contortus third-stage larvae on day zero (D0), and samples were collected weekly, for 28 days. Feces were used for individual fecal egg counts (FEC) blood for packed cell volume (PCV) and serum for specific antibody quantification through ELISA. Sera was collected on D0 (-) and D21 (+), and analyzed using a LC-MS/MS based proteomics approach. FEC, PCV, and anti-H. contortus antibody levels confirmed the absence of infection on D0. On D28 there was a significant difference between the two breeds for logFEC means (D = 3774 and S = 3141, p=0.033) and PCV means (D = 16.3 % and S = 24.3 %, p=0.038). From a total of 754 proteins identified, 68 differentially abundant proteins (DAPs) were noted. Phosphopyruvate hydratase (ENO3) was a DAP in all comparisons, while S+ vs D+ and S- vs D- shared the highest number of DAPs (8). Each of the four experimental groups clustered separately in a principal component analysis (PCA) of protein profile. Among the DAPs, proteins associated with the innate and adaptive immune system were detected when comparing S- vs D- and S+ vs D+. In D-, some proteins were linked to stress response to handling, sampling and heat. Focusing on the consequences of infection in each breed, in the D+ vs D- comparison, upregulated proteins were associated with inflammation control and immune response, where downregulated proteins pointed to a negative impact of infection on tissue anabolism, compromising muscle growth and fat deposition. In the S+ vs S- comparison, upregulated proteins were related to immune response, while the downregulated proteins were possibly linked to muscular development and growth, impaired by infection. Collectively, it can be concluded that ENO3 regulation emerges as a potential factor underlying the differential immune response observed between Santa Inês and Dorper sheep infected with H. contortus. In turn, detected acute phase proteins (APPs) reinforce their relation with infection, inflammation and stress conditions, whereas THEMIS-like may contribute to the immune system in Dorper. GSDMD, Guanylate-binding protein and ACAN warrant further investigation as possible biomarkers for TST strategy development.

Objective: The study was conducted to identify the sequence variation of Toll-like receptor 4 (TLR4) in exon 2 of South African Dorper sheep. Materials and Methods: Blood samples were collected from fifty (n = 50) South African Dorper sheep aged between 3 and 4 years. The Deoxyribonucleic acid (DNA) was extracted, amplified, and sequenced for the TLR4 gene. DNA sequencing was used to identify the sequence variations of the TLR4 gene in South African Dorper sheep. Results: The results showed that one synonymous single nucleotide polymorphism (SNP) of the TLR4 gene in exon 2 position T2249C was identified. Two genotypes (TT and TC) were discovered from the identified SNP. The dominant genotype was TT (0.60) over TC (0.40), with the dominant allele T (0.80) over C (0.20). The results also indicated that the used population was in the Hady-Weinberg Equilibrium. Polymorphism genetic analysis findings suggest that the identified sequence variation of TLR4 in exon 2 of South African Dorper sheep was moderate polymorphism. Conclusion: TLR4 gene at exon 2 of South African Dorper sheep had the SNP (T>C) at position 2249 bp with two genotypes (TT and TC).

The study aimed to estimate genetic and phenotypic parameters for growth traits and Kleiber ratio in Dorper sheep breed. Data used in this study were collected over 12 years (2012-2023) at Debre Birhan Agricultural Research Center sheep research station in Ethiopia. Studied traits were body weights at birth (WT0), weaning (WT3), six month (WT6), and yearling (WT12) age; average daily gains from birth to weaning (ADG0-3), from weaning to six months (ADG3-6), from six months to yearling (ADG6-12); and Kleiber ratios from birth to weaning (KR1) and from weaning to six months (KR2). The (co)variance components were estimated with different animal models using Average Information Restricted Maximum Likelihood (AI-REML) procedure. The best-fitted model for each trait was determined using likelihood ratio tests. Phenotypic performance for WT3, WT6, WT12, ADG0-3 and ADG3-6 showed a decline trend at a rate of 0.216 kg, 0.794 kg, 0.671 kg, 2.601 g and 4.865 g over years respectively. However, WT3, WT6, WT12, ADG0-3 showed a positive genetic improvement trend at a rate of 0.029 kg, 0.043 kg, 0.049 kg and 0.257 g over years respectively. Year of birth had a significant effect (P < 0.001) on all studied traits. Model including direct genetic as well as permanent environmental effect (Model 2) was chosen as the most appropriate model for WT0. Model which included only direct genetic effect (Model 1) was the best-fit model for all other studied traits. Direct heritability estimates based on suitable models were 0.07 ± 0.06, 0.11 ± 0.06, 0.09 ± 0.07, 0.11 ± 0.09 0.11 ± 0.06, 0.00 ± 0.04, 0.15 ± 0.07 and 0.00 ± 0.04 for WT0, WT3, WT6, WT12, ADG0-3, ADG3-6, KR1 and KR2 respectively. The variance ratio for the permanent environmental effect was 0.13 ± 0.04 for WT0. Genetic correlations among the traits ranged from negative (-0.39) for WT0-KR1 to high (0.99) for WT3-ADG0-3 and phenotypic correlations ranged from negative (-0.31) for WT0-KR1 to high (0.98) for WT3-ADG0-3. The low direct heritability estimates for the studied traits indicated that genetic improvement by direct selection might be difficult. Further investigation for the unexpected declined trend of phenotypic performance over years need to be required.

BACKGROUND: Dorper and Tan sheep are renowned for their rapid growth and exceptional meat quality, respectively. Previous research has provided evidence of the impact of gut microbiota on breed characteristics. The precise correlation between the gastrointestinal tract and peripheral organs in each breed is still unclear. Investigating the metabolic network of the intestinal organ has the potential to improve animal growth performance and enhance economic benefits through the regulation of intestinal metabolites. RESULTS: In this study, we identified the growth advantage of Dorper sheep and the high fat content of Tan sheep. A transcriptome study of the brain, liver, skeletal muscle, and intestinal tissues of both breeds revealed 3,750 differentially expressed genes (DEGs). The genes PPARGC1A, LPL, and PHGDH were found to be highly expressed in Doper, resulting in the up-regulation of pathways related to lipid oxidation, glycerophospholipid metabolism, and amino acid anabolism. Tan sheep highly express the BSEP, LDLR, and ACHE genes, which up-regulate the pathways involved in bile transport and cholesterol homeostasis. Hindgut content analysis identified 200 differentially accumulated metabolites (DAMs). Purines, pyrimidines, bile acids, and fatty acid substances were more abundant in Dorper sheep. Based on combined gene and metabolite analyses, we have identified glycine, serine, and threonine metabolism, tryptophan metabolism, bile secretion, cholesterol metabolism, and neuroactive ligand-receptor interaction as key factors contributing to the differences among the breeds. CONCLUSIONS: This study indicates that different breeds of sheep exhibit unique breed characteristics through various physiological regulatory methods. Dorper sheep upregulate metabolic signals related to glycine, serine, and threonine, resulting in an increase in purine and pyrimidine substances. This, in turn, promotes the synthesis of amino acids and facilitates body development, resulting in a faster rate of weight gain. Tan sheep accelerate bile transport, reduce bile accumulation in the intestine, and upregulate cholesterol homeostasis signals in skeletal muscles. This promotes the accumulation of peripheral and intramuscular fat, resulting in improved meat quality. This work adopts a joint analysis method of multi-tissue transcriptome and gut metabolome, providing a successful case for analyzing the mechanisms underlying the formation of various traits.

This study evaluated the reproductive and productivity of local and Dorper crossbred ewes in a community-based management system. We analyzed data collected from 2013 to 2021, taking into account different factors such as dam-breed, location, type of birth, season, and year of lambing. Lambing was observed all year-round, but the majority (35%) occurred in September, October, and December. This suggests that pasture availability, which is influenced by climatic-factors, may play a role in the seasonality of lambing. Litter-size at birth and weaning did not show any significant difference. Age at first lambing varied between breeds (P < 0.001), while lambing-interval and annual reproductive rates were unaffected by dam-breed (P > 0.05). The annual number of lambing per year significantly varied based on location and lambing seasons (P < 0.001), with a higher frequency during the major rainy-season compared to the dry-season (1.58vs1.42), highlighting the influence of feed availability. Productivity indices of ewes were calculated. Location and season of lambing had a significant impact on annual ewe productivity, while the ewe genotype showed no significant influence on productivity indices, except for the weight of lambs produced per kilogram of metabolic weight (0.84vs0.72 lambs per kg ewe and year; P < 0.01: 2.02vs1.77 kg lamb per kg0.75 ewe and year), where local ewes outperformed Dorper crossbred ewes. The difference in annual-productivity indices between local and Dorper crossbred ewes was more evident when considering the postpartum weight, as the ewes exhibited higher postpartum weights. However, both ewe genotypes produced comparable lamb weights per year (20.91vs20.16 kg lamb weaned per ewe and year for local and Dorper crossbred ewes, respectively). In summary, under low-input conditions, Dorper crossbred ewes demonstrated comparable reproductive performances and productivity traits to local ewes. Nevertheless, breed and environmental factors identified in this study should be taken into account to enhance sheep productivity in both local and Dorper crossbred ewes.

The increase of meat production in dairy sheep farms, has been evaluated by the extension of the suckling period from the traditional 28 days to 75 days to obtain a new product, the heavy suckling lamb. Nineteen single-born Sarda (S) lambs (10 male, 9 female) and 20 single-born Dorper x Sarda (DS) lambs (9 male, 11 female), randomly selected from autumn lambing season, were fed exclusively with maternal milk until slaughtering at 20 ± 0.28 kg of body weight (BW, mean ± standard deviation, SD) and approximately 11 weeks of age. Body weight were recorded at birth and every 15 days until slaughter to estimate average daily gain (ADG). At slaughter, carcass measurements, pH and colour traits were recorded from the left side of the carcass. Proximate composition, fatty acid (FA) profile, cooking and drip losses were evaluated using the Longissimus thoracis et lumborum (LTL) muscle. In addition, Visual Panel Test (VPT) and Taste Panel Test (TPT) were performed. Experimental results evidenced that ADG did not differ between pure and crossbreed lambs and between sexes. The S lamb carcasses showed a higher fat content and rib fat thickness compared to that of crossbreed. No significant differences were found between genetic types and sex for colour and pH determinations, cooking and drip losses, whereas LTL fat of DS displayed a higher nutritional FA profile with higher content of 22:5n-3, 22:6n-3, branched-chain FA, and odd- and branched chain FA. No differences emerged during VPT and TPT, demonstrating that both DS and S lamb meats present no distinguishable visual and eating quality characteristics. The production of heavy suckling lambs from Sarda and Dorper crossbreed by the extension of suckling period appeared a promising strategy for producing meat of high quality, well appreciated by the consumers.

Limited data are available regarding the effects of Brachiaria decumbens on sheep's growth performance at different times. Therefore, this current study focused on sheep's nutrient apparent digestibility, feed efficiency, body index, and growth hormone when they are fed with low and high levels of B. decumbens diets. A total of 30 six-month-old male Dorper cross sheep were divided randomly into three treatment groups with 10 sheep per treatment. Treatment 1 (control) sheep were fed with Pennisetum purpureum and pellets as the basal diet, whereas Treatment 2 and 3 sheep were fed with feed mixed with low (10%) and high (60%) levels of B. decumbens, respectively. The study was conducted in two phases consisting of short-term feeding (seven days) and long-term feeding (90 days). Throughout the experiment, daily fecal voided were collected in the morning for seven days continuous before the end of each feeding phases for the determination of nutrient apparent digestibility. The amount of feed offered and refusals plus body weight gain were recorded daily to determine the feed efficiency (FE). Besides, the body measurements of each sheep from every treatment were measured weekly and blood samples were collected for the analysis of growth hormone (GH) concentration. There were significant differences (p < 0.05) in the nutrient apparent digestibility, growth performance, body measurement, and GH concentration among treatment sheep throughout the study period. Treatment 3 sheep fed with 60% of B. decumbens diet revealed the lowest dry matter (DM), crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) digestibility during the long-term feeding. Likewise, Treatment 3 (T3) sheep had the lowest total bodyweight gain, average daily gain, total feed intake, and daily feed intake among treatment sheep. The heart girth index (HGI) of T3 sheep was also significantly lower during the short-term feeding. Moreover, the GH concentration of T3 sheep was significantly lower as compared to the control that decreases steadily throughout the study period. In conclusion, high levels of B. decumbens showed the most significant results out of all three treatments indicating the presence of saponins, which produce negative effects on the sheep's overall performance.

Oleh itu, kajian semasa ini memberi tumpuan kepada kebolehcernaan ketara nutrien, kecekapan makanan, indeks badan, dan hormon pertumbuhan bebiri yang diberi makan dengan diet B. decumbens pada tahap rendah dan tinggi. Sebanyak 30 ekor bebiri Dorper jantan berumur 6 bulan dibahagikan secara rawak kepada 3 kumpulan rawatan dengan 10 ekor bebiri setiap kumpulan. Rawatan 1 (kawalan) bebiri diberi makan dengan Pennisetum purpureum dan pelet sebagai diet asas, manakala bebiri Rawatan 2 dan 3 masing-masing diberi makan dengan campuran B. decumbens pada tahap rendah (10%) dan tinggi (60%). Kajian ini dijalankan dalam dua fasa yang terdiri daripada pemakanan jangka pendek (7 hari) dan panjang (90 hari). Sepanjang eksperimen, najis harian yang dibuang dikumpulkan pada waktu pagi selama 7 hari berterusan sebelum tamat setiap fasa pemakanan untuk menentukan kebolehcernaan ketara nutrien. Jumlah makanan yang diberi, sisa makanan, dan penambahan berat badan direkodkan setiap hari untuk menentukan kecekapan makanan (FE). Selain itu, ukuran badan setiap bebiri daripada setiap rawatan diukur setiap minggu serta sampel darah diambil untuk analisis kepekatan hormon pertumbuhan (GH). Terdapat perbezaan yang signifikan (p < 0.05) dalam kebolehcernaan ketara nutrien, prestasi pertumbuhan, ukuran badan, dan kepekatan GH di kalangan bebiri rawatan sepanjang tempoh kajian. Bebiri rawatan 3 (T3) yang diberi makan dengan 60% diet B. decumbens menunjukkan kebolehcernaan bahan kering (DM), protein kasar (CP), gentian detergen neutral (NDF), dan gentian detergen asid (ADF) yang paling rendah semasa pemberian makanan jangka panjang. Bebiri T3 mempunyai jumlah penambahan berat badan, purata kenaikan harian, jumlah pengambilan makanan, dan pengambilan makanan harian yang paling rendah di kalangan bebiri rawatan. Dorper sheep was developed for meat production in arid and semi-arid regions under extensive production systems in South Africa. Two variants with distinct head and neck colors were bred during their development process. White Dorper have a white coat while Dorper have a black head and neck. Both variants have grown in popularity around the world. Therefore, understanding the genomic architecture between South African Dorpers and Dorper populations adapted to other climatic regions, as well as genomic differences between Dorper and White Dorper variants is vital for their molecular management. Using the ovine 50K SNP chip, this study compared the genetic architecture of Dorper variants between populations from South Africa and Hungary. The Dorper populations in both countries had high genetic diversity levels, although Dorper in Hungary showed high levels of inbreeding. White Dorpers from both countries were genetically closely related, while Dorpers were distantly related according to principal component analysis and neighbor-joining tree. Additionally, whereas all groups displayed unique selection signatures for local adaptation, Dorpers from Hungary had a similar linkage disequilibrium decay. Environmental differences and color may have influenced the genetic differentiation between the Dorpers. For their molecular management and prospective genomic selection, it is crucial to understand the Dorper sheep's genomic architecture, and the results of this study can be interpreted as a step in this direction.

Heat stress is considered as a limiting factor for sheep production; it is necessary to screen for sheep breeds with heat tolerance. This study was to compare the serum metabolomes of Hu sheep and Dorper sheep and identify potential biomarkers related to heat stress. The results revealed that the respiratory rate, heart rate, and rectal temperature of Dorper sheep were significantly higher than those of Hu sheep. Compared to Dorper sheep, the serum activities of total antioxidant capacity and glutathione peroxidase in Hu sheep were significantly higher, while the concentration of malondialdehyde was lower. Metabolomics analysis identified 107 differential serum metabolites. The pathways enriched from the altered serum metabolites between the two breeds were mainly involved in protein metabolism, carbohydrate metabolism, and lipid metabolism. The levels of antioxidant- and energy-related metabolites were higher in the serum of Hu sheep than that of Dorper sheep; however, the levels of lipid catabolism- and inflammation-related were higher in the serum of Dorper sheep. The results indicate that Hu sheep had better heat stress resistance capability than Dorper sheep. Moreover, high levels of metabolites in the serum of Hu sheep are potential biomarkers for heat stress tolerance, including l-methionine, s-adenosylmethionine, and nicotinuric acid.

Background: Livestock is an excellent source of high nutritional value protein for humans; breeding livestock is focused on improving meat productivity and quality. Dorper sheep is a distinguished breed with an excellent growth performance, while Tan sheep is a Chinese local breed famous for its delicious meat. Several studies have demonstrated that the composition of gut microbiome and metabolome modulate host phenotype. Methods: In the present study, we performed 16S amplicon sequencing and metabolomic analyses of the rumen and hindgut microbiome of 8-month-old Dorper and Tan sheep, raised under identical feeding and management conditions, to explore the potential effects of gut microbiome and its metabolites on growth performance and meat quality. Results: Our study identified Lactobacillus, a marker genus in the rumen, to be significantly associated with the levels of fumaric acid, nicotinic acid, and 2-deoxyadenosine (P-value < 0.05). Statistical analysis showed that nicotinic acid was significantly negatively correlated with body weight (P-value < 0.01), while 2-deoxyadenosine was significantly positively correlated with fatty acids content (P-value < 0.05). There was a biologically significant negative correlation between Phascolarctobacterium and deoxycytidine levels in the hindgut. Deoxycytidine was significantly positively correlated with body weight, protein, and amino acid content. Differences in rumen fermentation patterns that are distinctive among breeds were identified. Tan sheep mainly used Lactobacillus and fumaric acid-mediated pyruvic acid for energy supply, while Dorper sheep utilize glycogenic amino acids. The difference of iron metabolism in the hindgut of Dorper sheep affects lipid production, while Phascolarctobacterium in Tan sheep is related to roughage tolerance. The accumulation of nucleosides promotes the growth performance of Dorper sheep. Conclusion: These findings provide insights into how the microbiome-metabolome-dependent mechanisms contribute to growth rate and fat contents in different breeds. This fundamental research is vital to identifying the dominant traits of breeds, improving growth rate and meat quality, and establishing principles for precision feeding.

This research was conducted to evaluate the replacement potential of Acacia nilotica dried leaf meal for Noug Seed Cake (NSC) to supplement low quality grass hay in the diet of crossbred sheep (25% Dorper). In doing so, four treatments were set up in such a way that 0%, 33%, 67%, and 100% NSC is replaced with dried leaf of Acacia nilotica from a conventional supplement while the treatments were kept isonitrogenous. The experiment was set up in a Randomized Complete Block Design, with initials weight used as the blocking factor. Twenty animals were allocated to the four treatments. The experiment consisted of hundred days of feeding trial followed by evaluation of carcass components at the end. In addition, the experimental feed ingredient was studied in vitro for gas production, methane (CH4) production, fractional rate of degradation (Kd), and in vitro dry matter digestibility (IVDMD). In view of chemical composition, grass hay contained lower crude protein (CP = 3.2% DM) and high cell wall contents that makes it lower quality feed. On the other hand, Acacia nilotica leaf meal was moderate in quality (CP = 14.3% DM) that can support moderate level of ruminant production. In an in vitro study, Acacia nilotica was found to have lower (P < 0.01) levels of CH4, total gas, kd, and IVDMD compared to the other feed ingredients. This indicates that tannin have an effect on Acacia nilotica feed. Partial and complete replacement of NSC with Acacia nilotica leaf meal significantly increased total dry matter intake (TDMI) (P < 0.001). Average daily gain (ADG) and dressing percentage was also higher for leaf meal supplemented groups. Hot carcass weight was in the range of 14.8-17.8 kg, which is higher than the national average carcass weight for Ethiopian sheep, and it was also found to be higher when NSC was partially (33%) as well as completely replaced by the leaf meal. In general, NSC can be replaced either partially or completely with Acacia nilotica leaf meal in the diet of 25% Dorper crosses while weight gain as well as carcass yield is promoted. The maximum inclusion level of Acacia nilotica was 61.3% or 319.2 g per day for 25% Dorper cross sheep fed natural pasture hay as a basal diet.

Rumen microbes play an important role in the growth and development of ruminants. Differences in variety will affect the rumen community structure. The three excellent sheep breeds were selected for this study (Hu sheep, Tan sheep, and Dorper sheep) have different uses and origins. The sheep were raised on the same diet to 180 d of age in a consistent environment. 16S rDNA V3 to V4 region sequencing was used to assess the rumen microbes of 180 individuals (60 per breed). There were differences in microbial diversity among different sheep breeds (P < 0.05). Principal coordinate analysis showed that the three varieties were separated, but also partially overlapped. Linear discriminant analysis effect size identified a total of 19 biomarkers in three breeds. Of these biomarkers, five in Hu sheep were significantly negatively correlated with average feed conversion rate (P < 0.05). Six biomarkers were identified in the rumen of Dorper sheep, among which Ruminococcus was significantly positively correlated with body weight at 80 d (P < 0.05). In Tan sheep, Rikenellaceae_RC9_gut_group was significantly positively correlated with meat fat, and significantly positively correlated with volatile fatty acids (VFAs), such as butyric acid and isobutyric acid (P < 0.05). The Rikenellaceae_RC9_gut_group may regulate Tan mutton fat deposition by affecting the concentration of VFAs. Functional prediction revealed enrichment differences of functional pathways among different sheep breeds were small. All were enriched in functions, such as fermentation and chemoheterotrophy. The results show that there are differences in the rumen microorganisms of the different sheep breeds, and that the microorganisms influence the host.


The rumen is the most important digestive organ of ruminants, and the rumen microflora plays an important role in the process of digestion. This study compared the differences in rumen microbes of different breeds of sheep, identified key biomarkers of each breed, and analyzed their correlation with important economic traits of sheep. The results showed that the biomarkers of various breeds were significantly correlated with key traits. Therefore, we believe that there is a link between sheep rumen microbes and the differential traits of different breeds of sheep. This provides a new idea for sheep trait improvement.

It is well known that Dorper (DP) is a full-bodied, fast-growing and high dressing percentage breed, while the production performance of Small-tailed Han sheep (STH) is not so excellent, in contrast to DP. Therefore, in this study, a comparative transcriptomic analysis of liver and muscle tissues from DP and STH breeds was carried out to find differentially expressed genes (DEGs) that affect their growth and meat quality traits. The results showed that the total number of DEGs was 2,188 in the two tissues. There were 950, 160 up-regulated and 1,007, 71 down-regulated genes in the liver and muscle, respectively. Several DEGs such as TGFB1, TGFB3, FABP3, LPL may be associated with growth and development in DP. Also, several GO terms were found to be associated with muscle growth and development, such as developmental growth (GO:0048589), and myofibril (GO:0030016). Further validation of eight genes (6 up-regulated, and 2 down-regulated) was performed using quantitative RT-PCR. These findings will provide valuable information for studying growth and development as well as meat quality traits in sheep.

Dorper sheep (Ovis aries) (DPS), developed in the 1930s by crossing Dorset Horn and Blackhead Persian sheep in South Africa, is a world-famous composite breed for mutton production. The genetic basis underlying this breed is yet to be elucidated. Here, we report the sequencing and assembly of a highly contiguous Dorper sheep genome via integration of Oxford Nanopore Technology (ONT) sequencing and Hi-C (chromatin conformation capture) approaches. The assembled genome was around 2.64 Gb with a contig N50 of 73.33 Mb and 140 contigs in total. More than 99.5% of the assembled sequences could be anchored to 27 chromosomes and they were annotated with 20,450 protein-coding genes. Allele-specific expression (ASE) genes of Dorper sheep were revealed through ASE analysis and they were involved in the immune system, lipid metabolism, and environmental adaptation. A total of 5,701 and 456 allelic sites were observed in the SNP and indels loci identified from relevant whole-genome resequencing data. These allelic SNP and INDEL sites were annotated in 1,002 and 294 genes, respectively. Moreover, we calculated the number of variant sites and related genes derived from the maternal and paternal ancestors, revealing the genetic basis of outstanding phenotypic performance of Dorper sheep. In conclusion, this study reports the first reference genome of Dorper sheep and reveals its genetic basis through ASE. This study also provides a pipeline for mining genetic information of composite breeds, which has an implication for future hybrid-breeding practices.

The supplementation of rumen bypass fat (RBF) has remained one of the preferred approaches used to decrease undesirable saturated fatty acids (FA) and increase beneficial unsaturated FA in the meat. This study was planned to evaluate the influences of rumen bypass fats on meat quality, fatty acid and metabolic profiles in male Dorper sheep (n = 36) with 24.66 ± 0.76 kg (mean ± standard error) initial body weight. Treatment comprised a basal diet (30:70 rice straw to concentrate) with no added RBF as a control (CON), basal diet with prilled fat (PF), basal diet with prilled fat plus lecithin (PFL) and basal diet with calcium soap of palm fatty acids (CaS). The findings revealed that cooking loss, drip loss and shear force in longissimus dorsi (LD) muscle were not affected by RBF supplementation, while meat pH was significantly higher in the CaS on aging day 1. However, the diet supplemented with prilled fat and lecithin modified the meat's fatty acid profile significantly by increasing unsaturated fatty acids and decreasing saturated fats. The relative quantification of the major differentiating metabolites found in LD muscle of sheep showed that total cholesterol, esterified cholesterol, choline, glycerophosphocholine and glycerophospholipids were significantly lower in CaS and PFL diets, while glycerol and sphingomyelin were significantly higher in CaS and PFL diets. Most of the metabolites in the liver did not show any significant difference. Based on our results, the supplementation of protected fats did not have a negative influence on meat quality and the meat from Dorper sheep fed prilled fat with lecithin contained more healthy fatty acids compared to other diets.

Cytokines are immune response components important in innate immunity and inflammatory response. They are harnessed as part of local immunological responses by animals to combat local infections and/or infestations. This study investigated expression of four selected cytokine genes, namely, interleukin 1 beta (IL-1ß), chemokine C-C ligand 2 (CCL2), chemokine C-C ligand 26 (CCL26) and interleukin 8 (IL-8), at tick attachment and control sites in a South African indigenous sheep breed the Namaqua Afrikaner (NA) and two commercial breeds, the Dorper and South African Mutton Merino (SAMM). The NA was previously shown to be more resistant to infestation by ticks than the two commercial breeds. NA ewes expressed IL-1ß more at tick attachment sites compared to Dorpers. The NA breed was also more likely to upregulate the expression of the CCL2, CCL26 and IL-8 genes at tick attachment sites compared to control sites than the other breeds. The results of this study gave an indication that cytokines are involved in immune responses to tick challenge and laid a foundation for further studies under controlled challenge conditions.

The objective of this study was to compare the proximate, quality, and sensory attributes of Dorper sheep meat (Dorper), domestic commercial crossbred (DCC) and Australian commercial crossbred (ACC). A total of 60 untrimmed loins from the three sheep sources were purchased (20 sheep loins/source) and processed. The objective color, objective tenderness [Warner-Bratzler Shear Force (WBSF)], and proximate composition of the sheep meat were evaluated. A consumer panel and a trained sensory panel were also conducted to evaluate the sensory attributes. Dorper had greater (P = 0.04) carbohydrate content compared to DCC, but was not (P = 0.86) different from ACC. In addition, Dorper had the greatest WBSF value, followed by DCC, with ACC having the least WBSF out of the three (P < 0.0001). For the consumer panel, Dorper was rated to be less tender than ACC (P = 0.01), but was not different from DCC (P = 0.76). Dorper was also rated with lower flavor acceptability compared to DCC (P = 0.02), but was not different from ACC (P = 0.86). In addition, Dorper had the lowest overall acceptance rating by the consumers (P = 0.01). Trained sensory panel results followed the same trend as the consumer panel results which rated Dorper to be less tender than ACC (P = 0.002), but was not different from DCC (P = 0.10). Dorper was also rated with greater off-flavor intensity compared to DCC (P = 0.009), but was not different from ACC (P = 0.53). Finally, no differences were found for all other attributes evaluated among the sheep sources. The results indicated that consumers did not prefer Dorper over ACC and DCC. However, additional research with a more controlled environment is needed to shed light on the true palatability traits of Dorper.

Ticks can compromise productivity and welfare in free-ranging sheep. Chemical tick control may not be sustainable in the long term. Alternative control measures must be sought for an integrated control programme. Birth and weaning weights as well as log transformed overall tick count of indigenous fat-tailed Namaqua Afrikaner (NA), commercial Dorper and NA x Dorper cross lambs were studied under extensive conditions. Relative to NA lambs, Dorper lambs were 22.2% heavier at weaning (P < 0.05). Geometric means for total tick count on Dorper lambs exceeded those of their Namaqua Afrikaner contemporaries by more than twofold (P < 0.05). Relative to the pure-breed midparent value, the mean performance of NA x Dorper lambs was 7.9% more for birth weight, 11.2% more for weaning weight and 26.2% less for the back transformed means for total tick count (P < 0.05). Heterosis for total tick count was slightly greater at -29.3% when data were adjusted for the larger size of NA x Dorper lambs. Crossing commercial Dorper sheep with a hardy, indigenous breed therefore resulted in lower levels of tick infestation without compromising live weight in progeny so derived. Hardy, indigenous genetic resources like the NA should be conserved and used in further studies of ovine genetics of resistance to ticks in South Africa.

The objective of this study was to examine sex and breed effects on heat shock protein 70 (HSPA1A), blood stress indicators and meat quality attributes of lambs. A hundred male and female lambs from the Dorper (n = 50) and Merino (n = 50) breeds were used in this study. Breed and sex had a significant (p < 0.05) effect on the levels of plasma HSPA1A and lactate; where the Merino lambs had higher levels than Dorper. The female lambs had higher levels of plasma HSPA1A than male lambs. Significant sex and breed interactions (p < 0.05) on the levels of plasma HSPA1A were seen. Females had higher (p < 0.05) pHu than males. Dorper lambs had higher (p < 0.05) pH45, meat lightness, thawing loss and tougher meat the Merino breed. Significant correlations were found amongst plasma stress indicators and meat quality attributes. The results indicate that female lambs were more stressed by the pre-slaughter period than males, while the Merino had a higher physiological stress response compared to the Dorper. However, the Dorper breed produced tougher meat.

Twenty-seven Dorper lambs were used to determine the effect of supplementing corn as a source of energy into the palm kernel cake (PKC) urea-treated rice straw basal diet on the blood metabolic profile and metals in lambs. The lambs were randomly allotted to three experimental treatments according to their initial body weight for a 120 day trial. Dietary treatments were: T1 (control diet) = 75.3% of PKC + 0% corn, T2 = 70.3% of PKC + 5% corn, and T3 = 65.3% of PKC + 10% corn. The results of this study indicated that copper (Cu), selenium (Se), zinc (Zn), and iron (Fe) concentration intake, retention, and its absorption from the gut and apparent mineral digestibility were highly significant for the levels of corn supplementation. The biochemical and hematological parameters remained within normal levels with the treatments, but the white blood cell, eosinophil count, cholesterol, and low-density lipoprotein cholesterol (LDL) were significantly higher in T3. Treatment 3 significantly increased the concentration of Se and Fe, while Zn was reduced in the blood serum of lambs on day 120. The result shows that the inclusion of corn has no effect on the hematological and biochemical parameters of lambs after incorporating corn into the PKC-based diet at 5% and 10%.
