RESUMO

Phycocyanin (PC) is a naturally occurring green pigment in Spirulina. It was extracted by ultrasonic extraction using green technology, and its structure was studied using IR- and NMR-spectroscopy. Spectral data confirmed the PC structure. This study also involves an in silico assessment of the diverse applications of green pigment PC. Utilizing QSAR, PreADME/T, SwissADME, and Pro-Tox, this study explores the safety profile, pharmacokinetics, and potential targets of PC. QSAR analysis reveals a favorable safety profile, with the parent structure and most metabolites showing no binding to DNA or proteins. PreADME/T indicates low skin permeability, excellent intestinal absorption, and medium permeability, supporting oral administration. Distribution analysis suggests moderate plasma protein binding and cautious blood-brain barrier permeability, guiding formulation strategies. Metabolism assessments highlight interactions with key cytochrome P450 enzymes, influencing drug interactions. Target prediction analysis unveils potential targets, suggesting diverse therapeutic effects, including cardiovascular benefits, anti-inflammatory activities, neuroprotection, and immune modulation. Based on the in silico analysis, PC holds promise for various applications due to its safety, bioavailability, and potential therapeutic benefits. Experimental validation is crucial to elucidate precise molecular mechanisms, ensuring safe and effective utilization in therapeutic and dietary contexts. DFT calculations, including geometry optimization, MEP analysis, HOMO-LUMO energy surface, and quantum reactivity parameters of the PC compound, were obtained using the B3LYP/6-311G(d,p) level. This integrated approach contributes to a comprehensive understanding of PC's pharmacological profile and informs future research directions.

Improving production efficiency and minimizing the environmental impact of dairy farming are 2 important goals of the dairy industry. Achieving these objectives requires improving the feed-to-milk conversion efficiency. One way to achieve this goal is through genetic selection. However, measuring feed efficiency in commercial herds is currently not feasible. As such, we conducted a study to evaluate the genetic accuracy of various selection indices derived from Fourier transform mid-infrared (FTIR)-spectra or milk composition. We use 7,793 weekly records on 537 genotyped cows (78,964 SNPs), with information on residual feed intake (RFI), and FTIR-spectra. We fitted various types of selection indexes using the complete FTIR-spectra of milk samples. The estimated heritability of RFI was 0.12 ± 0.02. The accuracy of indirect selection using the FTIR-spectra was maximized using a principal components selection index (0.16 ± 0.07), followed by a Lasso-type penalized selection index (0.14 ± 0.06). We determined that an index based on milk spectral data recorded on ~25 daughters produced a progeny average with an accuracy comparable to direct phenotypic selection for RFI. We conclude that indirect selection for RFI using FTIR-spectra data can be effective for sires with progeny; however, future studies with a larger sample size are needed to validate these results.

Pesticide transport in the environment is impacted by the kinetics of its adsorption onto soil. The adsorption kinetics of pyrimethanil was investigated in ten soil samples of varying physicochemical properties. The highest adsorption was in the soil having the maximum silt and CaCO3 contents, pH and electrical conductance but the lowest amorphous Fe oxides and CaCl2 extractable Mn. Pseudo-second order kinetics and intra-particle diffusion model best accounted the adsorption kinetics of pyrimethanil. The equilibrium adsorption estimated by pseudo-second order kinetics (q02) was significantly and positively correlated with CaCl2 extractable Cu content (r = 0.709) while rate coefficient (k02) had a negative correlation with crystalline iron oxides content (r = -0.675). The intra-particle diffusion coefficient (ki.d.) had inverse relationship with CaCl2 extractable Mn content in soils (r = -0.689). FTIR spectra showed a significant interaction of pyrimethanil with micronutrient cations. Adsorption kinetic parameters of pyrimethanil could be successfully predicted by soil properties. The findings may help to evolve fungicide management decisions.

The study investigates the structural and chemical properties of brown rice flour (WRF), black rice flour (BRF) and their mixtures in ratios of 25%, 50% and 75% to provide reference information for the gluten-free bakery industry. BRF contains higher concentrations of proteins, lipids, total minerals, crude fiber, total polyphenols, proanthocyanidins and flavonoids than WRF. A higher amylose content in BRF than in WRF resulted in flour mixtures with slower starch digestion and a lower glycemic response depending on the BRF ratio added. Differences in the chemical composition of WRF and BRF led to improved composition of the flour mixtures depending on the BRF ratio. The presence of anthocyanidins and phenolic acids in higher concentrations in the BRF resulted in a red-blue color shift within the flour mixtures. The deconvoluted FTIR spectra showed a higher proportion of α-helixes in the amide I band of BRF proteins, indicating their tighter folding. An analysis of the FTIR spectra revealed a more compact starch structure in BRF than in WRF. By processing reflection spectra, nine optically active compound groups were distinguished in rice flour, the proportion in BRF being 83.02% higher than in WRF. Due to co-pigmentation, the bathochromic shift to higher wavelengths was expressed by the proanthocyanins and phenolic acids associated with the wavelengths 380 nm to 590 nm and at 695 nm. Anthocyanins, protein-tannin complexes, methylated anthocyanins and acylated anthocyanins, associated with wavelengths 619, 644 and 668 nm, exhibited a hypsochromic effect by shifting the wavelengths to lower values. This research represents a first step in the development of rice-based products with increased nutritional value and a lower glycemic index.

Cangjie Temple was built to commemorate Cangjie, the legendary inventor of Chinese characters. It stands as one of the few remaining temples in China dedicated to the invention and creation of writing. In this study, the material properties of wooden paintings from the Cangjie temple were characterized using Polarized Light Microscopy (PLM), Scanning Electron Microscopy coupled with Energy Dispersive X-ray Spectroscopy (SEM-EDS), Micro-confocal Raman Spectroscopy, X-ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), and Pyrolysis-Gas Chromatography-Mass Spectrometry (Py-GC/MS). It was confirmed that the pigments of the paintings included cinnabar, lapis lazuli, lead white, Paris green, and carbon black. The proteinaceous glue was used as an adhesive in the pigment samples, with tung oil likely being utilized as a primer for the wooden structures before painting. This study not only provides valuable data support for the conservation and restoration of the architectural features of Cangjie Temple but also provides useful reference for the maintenance and inheritance of similar ancient buildings.

Investigation on accumulation of cell wall components over critical growth stages will surely provide a new insight into dry matter accumulation studies in rice. An elevated biomass production provides an alternative strategy of yield improvement, which in turn maneuvers the species concerned as potential dual-purpose crop. On that note, present study was carried on 33 early and 39 medium duration rice genotypes. The average cellulose accumulation was 6.51% and 8.17% in early and medium duration genotypes, respectively, at flowering stage, which later on dipped to 1.43% and 3.46%, respectively, at physiological maturity. The gene specific marker MDgsp-5.a exhibited highest estimate of polymorphic information content (PIC), i.e., 0.685, closely followed by MDgsp-6.a with polymorphic information content (PIC) of 0.683. The control genotypes, i.e., Pratap and Mandakini, are grouped under the same cluster, i.e., Cluster-I.A, indicating their inherent genetic divergence from that of potential accumulators pertaining to cellulose accumulation. Pratap and Mandakini failed to produce peaks of conspicuous form at 3342 cm-1 and 1635 cm-1, bearing out by their low performance pertaining to cellulose and lignin accumulation at the later stages of development, respectively. From histochemistry studies, it was observed that the cell walls of sclerenchyma, peripheral vascular bundles, and parenchyma of the culm transections in control genotypes stained lightly than that of prolific accumulator cell walls, thus corroborating the findings of compositional analysis. The variation in cell wall thickening is primarily accounted due to altered carbohydrate accumulation across the growth stages as explored under scanning electron micrograph.

In a context where learning databases of microplastic FTIR spectra are often incomplete, the objective of our work was to test whether a synthetic data generation method could be relevant to fill the gaps. To this end, synthetic spectra were generated to create new databases. The effectiveness of machine learning from these databases was then tested and compared with previous results. The results showed that the creation of synthetic learning databases could avoid, to a certain extent, the need for learning databases of environmental microplastics FTIR spectra. However, some limitations were encountered, for example, when two different chemical classes had very similar reference spectra or when the intensities of the bands associated with fouling became too intense. The FTIR study of the ageing and fouling of microplastics in the natural environment is one of the identified ways that could further improve this approach.

Infrared (IR) spectroscopy is a powerful technique for detecting and identifying Microplastics (MPs) in the environment. However, the aging of MPs presents a challenge in accurately identification and classification. To address this challenge, a classification model based on deep convolutional neural networks (CNNs) was developed using infrared spectra results. Particularly, original infrared (IR) spectra were used as the sample dataset, therefore, relevant spectral details were preserved and additional noise or distortions were not introduced. The Adam (Adaptive moment estimation) algorithm was employed to accelerate gradient descent and weight update, the Dropout function was implemented to prevent overfitting and enhance the generalization performance of the network. An activation function ReLu (Rectified Linear Unit) was also utilized to simplify the co-adaptation relationship among neurons and prevent gradient disappearance. The performance of the CNN model in MPs classification was evaluated based on accuracy and robustness, and compared with other machine learning techniques. CNN model demonstrated superior capabilities in feature extraction and recognition, and greatly simplified the pre-processing procedure. The identification results of aged commercial microplastic samples showed accuracies of 40 % for Artificial Neural Network, 60 % for Random Forest, 80 % for Deep Neural Network, and 100 % for CNN, respectively. The CNN architecture developed in this work also demonstrates versatility by being suitable for both limited data cases and potential expansion to include more discrete data in the future.

Data sets derived from practical experiments often pose challenges for (robust) statistical methods. In high-dimensional data sets, more variables than observations are recorded and often, there are also data present that do not follow the structure of the data majority. In order to handle such data with outlying observations, a variety of robust regression and classification methods have been developed for low-dimensional data. The high-dimensional case, however, is more challenging, and the variety of robust methods is much more limited. The choice of the method depends on the specific data structure, and numerical problems are more likely to occur. We give an overview of selected robust methods as well as implementations and demonstrate the application with two high-dimensional data sets from tribology. We show that robust statistical methods combined with appropriate pre-processing and sampling strategies yield increased prediction performance and insight into data differing from the majority.

Although the electron-withdrawing effect of gold (Au) is highlighted in catalytic reactions, its enhancement mechanism for electron transport, especially in the electrochemical process, is still unclear. Herein, Au-decorated Bi2 O3 (Au-Bi2 O3 ) is proposed as a proof-of-concept to investigate the electron-withdrawing effect in the electrocatalytic CO2 reduction reaction (eCO2 RR) process. Evidence from in situ Raman spectra and in situ XRD tests reveals that, compared to Bi2 O3 , Bi species in Au-Bi2 O3 can be reduced to metallic Bi more rapidly and more easily driven by the electron-withdrawing effect of Au. The XPS tests after eCO2 RR further validates the transformation from Bi3+ to Bi0 in Au-Bi2 O3 is more complete. Meanwhile, in the in situ Fourier transform infrared (FTIR) spectra, the key intermediates (CO2 *- and OCHO*- ) appear at the more positive potential, indicating that metallic Bi is favorable for eCO2 RR due to the lower energy barrier as corroborated by density function theory (DFT) calculations. Au don't directly participate in the conversion from CO2 to formate as the reaction sites, but utilize the electron-withdrawing effect to motivate Bi-sites to deliver higher catalytic activity and higher selectivity to formate at a lower applied potential. This study not only has an insight into the electron-withdrawing effect of Au on the eCO2 RR process, but also develops a new perspective for engineering electron-withdrawing effect in electrocatalysts for high-efficient CO2 -to-formate conversion.

Films and fibers of syndiotactic polystyrene (sPS), being amorphous or exhibiting nanoporous crystalline (NC) or dense crystalline phases, were loaded with salicylic acid (SA), a relevant non-volatile antimicrobial molecule. In the first section of the paper, sPS/SA co-crystalline (CC) δ form is characterized, mainly by wide angle X-ray diffraction (WAXD) patterns and polarized Fourier transform infrared (FTIR) spectra. The formation of sPS/SA δ CC phases allows the preparation of sPS fibers even with a high content of the antibacterial guest, which is also retained after repeated washing procedures at 65 °C. A preparation procedure starting from amorphous fibers is particularly appropriate because involves a direct formation of the CC δ form and a simultaneous axial orientation. The possibility of tuning drug amount and release kinetics, by simply selecting suitable crystalline phases of a commercially available polymer, makes sPS fibers possibly useful for many applications. In particular, fibers with δ CC forms, which retain SA molecules in their crystalline phases, could be useful for antimicrobial textiles and fabrics. Fibers with the dense γ form which easily release SA molecules, because they are only included in their amorphous phases, could be used for promising SA-based preparations for antibacterial purposes in food processing and preservation and public health. Finally, using a cell-based assay system and antibacterial tests, we investigated the cellular activity, toxicity and antimicrobial properties of amorphous, δ CC forms and dense γ form of sPS fibers loaded with different contents of SA.

Red raspberries, which contain a variety of nutrients and phytochemicals that are beneficial for human health, can be utilized as a raw material in the creation of several supplements. This research suggests micronized powder of raspberry pomace production. The molecular characteristics (FTIR), sugar, and biological potential (phenolic compounds and antioxidant activity) of micronized raspberry powders were investigated. FTIR spectroscopy results revealed spectral changes in the ranges with maxima at ~1720, 1635, and 1326, as well as intensity changes in practically the entire spectral range analyzed. The discrepancies clearly indicate that the micronization of the raspberry byproduct samples cleaved the intramolecular hydrogen bonds in the polysaccharides present in the samples, thus increasing the respective content of simple saccharides. In comparison to the control powders, more glucose and fructose were recovered from the micronized samples of the raspberry powders. The study's micronized powders were found to contain nine different types of phenolic compounds, including rutin, elagic acid derivatives, cyanidin-3-sophoroside, cyanidin-3-(2-glucosylrutinoside), cyanidin-3-rutinoside, pelargonidin-3-rutinoside, and elagic acid derivatives. Significantly higher concentrations of ellagic acid and ellagic acid derivatives and rutin were found in the micronized samples than in the control sample. The antioxidant potential assessed by ABTS and FRAP significantly increased following the micronization procedure.

The power transformer is vital to the reliability of the power grid which is most commonly insulated with Kraft paper and immersed in mineral oil, among which the aged state of the paper is mainly correlated to the operating life of the transformer. Degree of polymerization (DP) is a direct parameter to assess the aged condition of insulating paper, but existing DP measurement by viscosity methods are destructive and complicated. In this paper, terahertz time-domain spectroscopy (THz-TDS) was introduced to reach rapid, non-destructive detection of the DP of insulating paper. The absorption spectra of insulating paper show that characteristic peak regions at 1.8 and 2.23 THz both exhibit a log-linear quantitative relationship with DP, and their universalities are confirmed by conducting the above relationship on different types of insulating paper. Fourier transform infrared spectroscopy (FTIR) analysis and molecular dynamics modeling further revealed that 1.8 and 2.23 THz were favorably associated with the growth of water-cellulose hydrogen bond strength and amorphous cellulose, respectively. This paper demonstrates the viability of applying THz-TDS to the non-destructive detection of DP in insulating paper and assigned the vibration modes of the characteristic absorption peaks.

Polycyclic aromatic hydrocarbons (PAHs) tend to accumulate in the sediment due to their high hydrophobicity. Despite PAHs have been the subject of several reviews, PAH sorption processes in bottom sediments has not been comprehensively discucorrelation coefficients between sorption parameters and contessed. Understanding the dependencies governing PAH sorption processes will allow to predict, monitor, and mitigate the ecological effects of PAH contamination and the associated risks to humans or wildlife. The objectives of the study were to assess the relationship between the sorption properties and the content of PAHs in bottom sediments and mussels. The PAH profile was dominated by higher-molecular hydrocarbons, which accounted for 73% of the total concentration of PAHs. Potentiometric studies revealed the steric-based PAH sorption mechanism that strongly depended on the presence of negatively dissociating structures such as carboxylic or phenolic functional groups. Based on the changes in Q8 values, the size-exclusion effect was more likely for 5- and 6-ring compounds. Pores < 5 µm, which had the largest share in the specific surface area, were the preferred sites for PAH sequestration and stabilization in bottom sediments. The availability of PAHs was reduced in sediments with high organic matter content. The PAH bioaccumulation factor significantly decreased with increasing TOC content in sediments. Higher mortality and growth inhibition of H. incongruens were observed in samples with high and medium TOC contents than in those with low TOC content.

The properties of biochar (BC) from crustacean chitin are relatively well understood, while there are few studies on BC from insect chitin. This study presents the characterization and phytotoxic assessment of BC produced from crickets and cricket chitin. Cricket powder (BCCR) and cricket chitin (BCCH) were pyrolyzed at 500 °C and 700 °C. Physicochemical characteristics, N ad-/desorption, FTIR, were examined. SEM images were also performed. Regardless of the pyrolysis temperature, biochars were characterized by a densely "packed" solid surface/monolithic type with a non-porous structure (0.05-0.22 m2/g) and high content of N (9.4-11.8%). BCCHs showed a higher pH (12.2-12.4) compared to BCCR (8.7-10.8). Based on the XRD analysis, BCs were characterized by an amorphous carbon turbostratic structure and a randomly oriented graphitic-like micro-crystallite structure. FTIR spectra of BCs confirmed the presence of various O2 and N-functional groups on the BC surface. BCCHs added to soil at rates from 0.5 to 1.5% significantly reduced the germination of Lepidium sativum. Stimulation of root elongation was also observed in the case of BCCR500 1.0% and BCCR700 1.5%. Thermal degradation of cricket powder and cricket chitin promotes the formation of organic N-containing heterocyclic rings, which lead to the production of N-doped carbons with potential uses in energy storage and the contaminations sorption.

The massive accumulation of plastics over the decades in the aquatic environment has led to the dispersion of plastic components in aquatic ecosystems, invading the food webs. Plastics fragmented into microplastics can be bioaccumulated by fishes via different exposure routes, causing several adverse effects. In the present study, the dose-dependent cytotoxicity of 8−10 µm polypropylene microplastics (PP-MPs), at concentrations of 1 mg/g (low dose) and 10 mg/g dry food (high dose), was evaluated in the liver and gill tissues of two fish species, the zebrafish (Danio rerio) and the freshwater perch (Perca fluviatilis). According to our results, the inclusion of PP-MPs in the feed of D. rerio and P. fluviatilis hampered the cellular function of the gills and hepatic cells by lipid peroxidation, DNA damage, protein ubiquitination, apoptosis, autophagy, and changes in metabolite concentration, providing evidence that the toxicity of PP-MPs is dose dependent. With regard to the individual assays tested in the present study, the biggest impact was observed in DNA damage, which exhibited a maximum increase of 18.34-fold in the liver of D. rerio. The sensitivity of the two fish species studied differed, while no clear tissue specificity in both fish species was observed. The metabolome of both tissues was altered in both treatments, while tryptophan and nicotinic acid exhibited the greatest decrease among all metabolites in all treatments in comparison to the control. The battery of biomarkers used in the present study as well as metabolomic changes could be suggested as early-warning signals for the assessment of the aquatic environment quality against MPs. In addition, our results contribute to the elucidation of the mechanism induced by nanomaterials on tissues of aquatic organisms, since comprehending the magnitude of their impact on aquatic ecosystems is of great importance.

The marine weathering of microplastics is spectrally characterized by the appearance of new bands that disturb our understanding of the information carried by the spectra. Yet, no explanation has been provided on the chemical origin of these new bands. Thus, the main objective of this work was to identify the origins of these additional bands. To this end, 4042 spectra of poly (styrene), poly(ethylene) and poly(propylene) microplastics collected in the Mediterranean Sea, were analysed using principal component analysis. The results showed that the spectral variability was mainly related to only three processes: chemical ageing, organic and inorganic fouling. These processes probably differ from one polymer family to another due to surface affinities. This work has also led to the proposal of two new polymer indices that could be used to monitor the intensity of (bio)fouling. Finally, the development of advanced analyses could also provide information on the nature of the plastisphere.

Biologically active substances from microalgae can exhibit antioxidant, immunostimulating, antibacterial, antiviral, antitumor, antihypertensive, regenerative, and neuroprotective effects. Lipid complexes of microalgae Chlorella vulgaris and Arthrospira platensis exhibit antibacterial activity and inhibit the growth of the Gram-positive strain Bacillus subtilis; the maximum zone of inhibition is 0.7 ± 0.03 cm at all concentrations. The carbohydrate-containing complex of C. vulgaris exhibits antibacterial activity, inhibits the growth of the Gram-positive strain B. subtilis, Bacillus pumilus; the maximum zone of inhibition is 3.5 ± 0.17 cm at all concentrations considered. The carbohydrate complex of A. platensis has antimicrobial activity against the Gram-negative strain of Escherichia coli at all concentrations, and the zone of inhibition is 2.0-3.0 cm. The presence of mythelenic, carbonyl groups, ester bonds between fatty acids and glycerol in lipid molecules, the stretching vibration of the phosphate group PO2, neutral lipids, glyco- and phospholipids, and unsaturated fatty acids, such as γ-linolenic, was revealed using FTIR spectra. Spectral peaks characteristic of saccharides were found, and there were cellulose and starch absorption bands, pyranose rings, and phenolic compounds. Both algae in this study had phenolic and alcohol components, which had high antibacterial activity. Microalgae can be used as biologically active food additives and/or as an alternative to antibiotic feed in animal husbandry due to their antibacterial properties.

Four complexes of Cd(II) with benzimidazole derivatives were synthesized and named C1, C2, C3, and C4. All coordination compounds were characterized through elemental analysis (EA), flame atomic absorption spectrometry (FAAS), Fourier-transform infrared spectroscopy (FTIR), thermogravimetric analysis coupled with mass spectrometry) (TG-MS), a cytotoxicity assay (MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide)), and computational chemical analysis for absorption, distribution, metabolism, and excretion (ADME). All of the obtained results are compatible and are consistent with the respective structures of the obtained compounds and their properties. The various techniques used allowed the determination of the composition, proposed structure of the compounds, their thermal stability and thermal properties, and the method of coordination between the metal (II) ion and the ligand. The ADME technique was also used to estimate the physicochemical and biological properties. The antitumor activity of the compounds was determined with an MTT assay on the glioblastoma (T98G), neuroblastoma (SK-N-AS), and lung adenocarcinoma (A549) cell lines, as well as normal human skin fibroblasts (CCD-1059Sk). Compound C2 was found to have potential antitumor properties and to be effective in inhibiting the growth of neuroblastoma cells. The antimicrobial activity of Cd complexes, free ligands, and reference drugs was tested against six strains of Gram-positive bacteria, five strains of Gram-negative rods, and three strains of yeasts. Compound C3 significantly increased activity against Gram-positive bacteria in comparison to the ligand.

Results of efficiency of obtaining humic substances (HSs) from peat in traditional alkaline extraction (TAE) and ultrasound-assisted alkaline extraction (UAAE) are presented. The influence of the duration of the process and ultrasound intensity on the efficiency of extraction of humic acids (HAs) and fulvic acids (FAs) extraction was determined. The composition of the fulvic acid fraction was examined depending on the type of eluent used. Fulvic acids were divided into fractions using columns packed with DAX-8 resin. For this process, 0.1 M NaOH and 0.5 M NH3∙H2O were used as eluents. For the quality assessment of specific fulvic acids fractions, spectroscopic methods (UV-Vis and FTIR) were used. Ultrasound had a positive effect on HS extraction efficiency, especially in increasing the amount of a desired hydrophobic fraction of fulvic acids (HPO). However, a negative effect of the excessive prolongation and ultrasound intensity (approximately 400 mW∙cm-2) on the extraction efficiency of HPO eluted with 0.1 M NaOH solution was observed. Using peat as a raw carbon material for the HS extraction process can be used as an alternative industrial application of peat. UAAE may be considered as an alternative method to TAE, which provides a higher efficiency in HS isolation from peat.
