Accurate prediction of energy requirement is important in formulating diets, but an energy model for Yellow Broiler breeder hens is publicly unavailable. The objective of this study was to establish energy prediction models for the nitrogen-corrected apparent metabolisable energy (AMEn) requirement of different categories of Yellow Broiler breeder hens during the egg-laying period. Data for modelling were collected from research papers, public databases and production data from companies. Breeder hens were generally categorised into three BW types: heavy, medium and light (HBWT, MBWT and LBWT). Published articles were cited for providing coefficients of AMEn maintenance requirement (AMEnm, 101 kcal/kg BW0.75, 423 KJ/kg BW0.75) and growth requirement (AMEng, 5.33 kcal/g, 22.3 KJ/g), respectively. Models of AMEn for egg production (AMEnp) were established from the known daily intake of AMEn (AMEni) and those of maintenance and growth by the factorial approach: AMEnp = AMEni - AMEnm - AMEng. For the three types of hens, AMEnp HBWT (kcal, KJ) = 2.55 kcal (10.7 KJ) × egg mass (EM, g); AMEnp MBWT (kcal, KJ) = 2.70 kcal (11.3 KJ) × EM (g), and AMEnp LBWT (kcal, KJ) = 2.94 kcal (12.3 KJ) × EM (g) were determined. The total AMEni requirements, depending on Gompertz models, were HBWT: BW (g) = 3 144 × e-EXP(-0.162×(week of age (wk)-15.6)); MBWT: BW (g) = 2 526 × e-EXP(-0.333×(wk-19.1)); LBWT: BW (g) = 1 612 × e-EXP(-0.242×(wk-16.5)). Models of egg production, HBWT: egg production (%) = 124 × e-0.017×wk/(1 + e-0.870×(wk-26.2)); MBWT: egg production (%) = 144 × e-0.020×wk/(1 + e-0.751×(wk-24.9)); LBWT: egg production (%) = 163 × e-0.024×wk/(1 + e-0.476×(wk-26.5))) and egg weight for each wk of the three types of hens during the egg-laying period were all established. These models showed good applicability in simulating and predicting the literature or production data.

Six assays were conducted to investigate male and female broiler responses to standardized ileal digestible (SID) lysine (Lys) and arginine (Arg). Response data were modeled to estimate the efficiency of utilization (k) of both amino acids and adjust factorial models to predict bird intake for SID Lys and SID Arg. In each assay, 1280 Cobb 500® broilers (640 male and 640 female) were randomly assigned to one of sixteen dietary treatments with four replicates of 20 birds. Dietary treatments consisted of crescent levels of SID Lys or SID Arg based on the dilution technique. The values of k determined for each phase (1- to 14-, 15- to 28-, and 29- to 42-d-old) and sex were contrasted using linear regression with groups (sex and phase). The estimated efficiencies were 0.79 for Lys and 0.62 for Arg, which were unaffected by phase or sex. Factorial models based on body weight and weight gain (M1) and on body and feather protein weight and deposition (M2) were applied to estimate the SID Lys and Arg intake for growth. The amino acid intake based on M2 had a lower error of prediction. Broiler chickens require 94.9 and 92.9 mg/d of SID Lys and SID Arg to deposit 1 g of body protein.

The requirement of net protein (NP) and metabolizable protein (MP) by Dorper crossbred ewe lambs grown from 35 to 50 kg of body weight (BW) was assessed by comparative slaughter experiment. Thirty-five ewe lambs (33.5 ± 0.6 kg BW) of F1 crosses of Dorper × thin-tailed Han sheep were used: 7 lambs were slaughtered as reference animals at the start of the trial, and the remaining 28 lambs were randomly divided into 4 groups of 7 lambs each. Three of the 4 groups were fed a pelleted mixed diet (concentrate/roughage = 44:56, dry matter basis) for ad libitum intake or 65% or 45% of ad libitum intake, and they were all slaughtered when the lambs that were fed ad libitum reached 50 kg BW. The lambs from the fourth group were also fed ad libitum and slaughtered at 43 kg BW as the intermediate group. The intake of MP by the animals of these 4 groups was estimated, and their total body protein and protein retention were measured. The daily requirements of NP and MP for maintenance were 1.52 and 3.98 g/kg BW0.75 , respectively, with a partial efficiency of MP utilization for maintenance of 0.38. The MP requirement for growth ranged from 77.4 to 124.5 g/day for average daily gains from 100 to 250 g BW, and the partial efficiency of MP utilization for growth was 0.66. The Dorper crossbred ewe lambs required more MP for both maintenance and growth in comparison with the recommendations of the US nutritional system.

BACKGROUND: Large discrepancies exist among the dietary zinc recommendations set by expert groups. OBJECTIVE: To describe the basis for the differences in the dietary zinc recommendations set by the World Health Organization, the US Institute of Medicine, the International Zinc Nutrition Consultative Group, and the European Food Safety Agency. METHODS: We compared the sources of the data, the concepts, and methods used by the 4 expert groups to set the physiological requirements for absorbed zinc, the dietary zinc requirements (termed estimated and/or average requirements), recommended dietary allowances (or recommended nutrient intakes or population reference intakes), and tolerable upper intake levels for selected age, sex, and life-stage groups. RESULTS: All 4 expert groups used the factorial approach to estimate the physiological requirements for zinc. These are based on the estimates of absorbed zinc required to offset all obligatory zinc losses plus any additional requirements for absorbed zinc for growth, pregnancy, or lactation. However, discrepancies exist in the reference body weights used, studies selected, approaches to estimate endogenous fecal zinc (EFZ) losses, the adjustments applied to derive dietary zinc requirements that take into account zinc bioavailability in the habitual diets, number of dietary zinc recommendations set, and the nomenclature used to describe them. CONCLUSIONS: Estimates for the physiological and dietary requirements varied across the 4 expert groups. The European Food Safety Agency was the only expert group that set dietary zinc recommendations at 4 different levels of dietary phytate for adults (but not for children) and as of yet no tolerable upper intake level for any life-stage group.

During a discovery project of potential inhibitors for three proteins, TNF-α, RANKL and HO-1, implicated in the pathogenesis of rheumatoid arthritis, significant amounts of purified proteins were required. The application of statistically designed experiments for screening and optimization of induction conditions allows rapid identification of the important factors and interactions between them. We have previously used response surface methodology (RSM) for the optimization of soluble expression of TNF-α and RANKL. In this work, we initially applied RSM for the optimization of recombinant HO-1 and a 91% increase of protein production was achieved. Subsequently, we slightly modified a published incomplete factorial approach (called IF1) in order to evaluate the effect of three expression variables (bacterial strains, induction temperatures and culture media) on soluble expression levels of the three tested proteins. However, soluble expression yields of TNF-α and RANKL obtained by the IF1 method were significantly lower (<50%) than those obtained by RSM. We further modified the IF1 approach by replacing the culture media with induction times and the resulted method called IF-STT (Incomplete Factorial-Stain/Temperature/Time) was validated using the three proteins. Interestingly, soluble expression levels of the three proteins obtained by IF-STT were only 1.2-fold lower than those obtained by RSM. Although RSM is probably the best approach for optimization of biological processes, the IF-STT is faster, it examines the most important factors (bacterial strain, temperature and time) influencing protein soluble expression in a single experiment, and can be used in any recombinant protein expression project as a starting point.

Titanium dioxide nanoparticles (TiO2 -NPs) adsorb co-occurring heavy metals in surface waters, modulating their toxicity for freshwater invertebrates. The processes triggering this interaction may be influenced by several environmental parameters; however, their relative importance remains unclear. The present study assessed the implications of aging on the joint acute toxicity of copper (Cu) and TiO2 -NPs for Daphnia magna over a duration of up to 72 h. The influences of aging duration as well as ionic strength, pH, and presence of different qualities of organic matter during aging were assessed. The results indicated that the presence of TiO2 -NPs often reduced the Cu-induced toxicity for daphnids after aging (albeit with varying extent), which was displayed by up to 3-fold higher EC50 (50% effective concentration) values compared to the absence of TiO2 -NPs. Moreover, the Cu speciation, influenced by the ionic composition and the pH as well as the presence of organic additives in the medium, strongly modulated the processes during aging, with partly limited implications of the aging duration on the ecotoxicological response of D. magna. Nonetheless, the present study underpins the potential of TiO2 -NPs to modify toxicity induced by heavy metals in freshwater ecosystems under various environmental conditions. This pattern, however, needs further verification using heavy metal ions with differing properties in combination with further environmental factors, such as ultraviolet irradiation. Filamentous fungi are the predominant source of lignocellulolytic enzymes used in industry for the transformation of plant biomass into high-value molecules and biofuels. The rapidity with which new fungal genomic and post-genomic data are being produced is vastly outpacing functional studies. This underscores the critical need for developing platforms dedicated to the recombinant expression of enzymes lacking confident functional annotation, a prerequisite to their functional and structural study. In the last decade, the yeast Pichia pastoris has become increasingly popular as a host for the production of fungal biomass-degrading enzymes, and particularly carbohydrate-active enzymes (CAZymes). This study aimed at setting-up a platform to easily and quickly screen the extracellular expression of biomass-degrading enzymes in P. pastoris. We first used three fungal glycoside hydrolases (GHs) that we previously expressed using the protocol devised by Invitrogen to try different modifications of the original protocol. Considering the gain in time and convenience provided by the new protocol, we used it as basis to set-up the facility and produce a suite of fungal CAZymes (GHs, carbohydrate esterases and auxiliary activity enzyme families) out of which more than 70% were successfully expressed. The platform tasks range from gene cloning to automated protein purifications and activity tests, and is open to the CAZyme users' community.
