Anastrepha fraterculus (Diptera: Tephritidae) is considered an important pest in Neotropical countries. The laboratory rearing of this species should reproduce conditions in nature; thus, special attention is required to the nutritional quality of diets for larval development. Protein components (wheat germ) are costly and account for most production costs in lab insect rearing. In this sense, this work aimed to identify ingredients to replace wheat germ, without compromising diet quality for the lab rearing of A. fraterculus. We tested diets composed of whole rice flour, corn flour, and a mixture of whole wheat flour + soybean flour as substitutes for wheat germ as well as a raw wheat germ diet, considered the control. The protein sources used in the larval diet influenced the biological performance of both the larval and adult stages of A. fraterculus during six generations. The diet containing corn flour and wheat germ showed similar results in the different developmental parameters. The diet with rice flour also provided adequate biological development for A. fraterculus throughout its life cycle and was nutritionally similar to the control. As it is local product, rice flour can replace wheat germ (costly imported product) in artificial diets for A. fraterculus, reducing production costs by roughly 30% without compromising the biological and nutritional parameters of the insects. Faced with this, the rice flour can be considered suitable for the mass rearing of A. fraterculus in the laboratory.

Phytoseiid predatory mites are one of the most important groups of biocontrol agents, commonly used in biological control. The ability to produce these predatory mites economically, at high density on cheap factitious food sources, is a major contributor to their success. Astigmatid mites are the most widely used factitious food for this purpose. In this study, we investigated the potential application of the leaf-dwelling astigmatid mite Czenspinskia transversostriata (Oudemans) (Acari: Winterschmidtiidae) as a prey mite in biological control. We tested whether C. transversostriata is a suitable food source for the predatory mite Amblyseius swirskii Athias-Henriot (Acari: Phytoseiidae), both in the laboratory and on cucumber plants. Based on a reproduction trial, C. transversostriata proved to be an equally good food source compared to both pollen of Typha angustifolia L. (Poales: Typhaceae) and a frequently used prey mite Carpoglyphus lactis L. (Acari: Carpoglyphidae). In a pre-establishment trial on cucumber plants, populations of A. swirskii reached equally high densities when supplemented with C. transversostriata, compared to C. lactis. Lastly, we show that C. transversostriata is capable of feeding and reproducing on powdery mildew growing on cucumber plants, thereby slowing down the development of the pathogenic fungus. Results derived from this study show that C. transversostriata may have multiple potential applications in biological control programs.

Understanding the biology and ecology of parasitoids can have direct implications for their evaluation as biological control agents, as well as for the development and implementation of mass-rearing techniques. Nonetheless, our current knowledge of the possible influence of lateralized displays (i.e., the asymmetric expression of cognitive functions) on their reproductive behavior is scarce. Herein, we characterized the behavioral elements involved in courtship, and quantified the durations of 2 important aphid parasitoids, Aphidius ervi Haliday and Aphidius matricariae Haliday (Hymenoptera: Braconidae: Aphidiinae). We quantified the main indicators of copulation and examined the occurrence of lateralized traits at population level. Results indicated that A. matricariae exhibited longer durations of wing fanning, antennal tapping, pre-copula and copula phases compared to A. ervi. Postcopulatory behavior was observed only in A. matricariae. Unlike other parasitoid species, the duration of wing fanning, chasing, and antennal tapping did not affect the success of the mating of male A. ervi and A. matricariae. Both species exhibited a right-biased female kicking behavior at the population level during the pre-copula. Our study provides insights into the fundamental biology of aphidiine parasitoids and reports the presence of population-level lateralized mating displays, which can serve as useful benchmarks to evaluate the quality of mass-rearing systems.

Cadra cautella (Walker) (Lepidoptera: Pyralidae) is a pest in barn, and its eggs are often utilized as an alternative prey or as hosts for mass production of insect predators and parasitoids. To aid in developing a mass-rearing system for C. castella, the suitability of using an artificial diet based on brown rice flour and whole brown rice was compared using the age-stage, 2-sex life table. Compared with those reared on brown rice, the insects reared on an artificial diet had a shorter preadult period (32.08 vs 37.38 d), higher fecundity (468.14 vs 356.20 eggs/female), greater intrinsic rate of increase (0.1509 vs 0.1145 d-1), and higher net reproductive rate (199.28 vs 103.52 offspring). Small populations were required to rear the moth on an artificial diet to achieve the same daily production of C. cautella. Still, the food expense was only 60.2% of that of C. cautella reared on brown rice. Approximately 99.44% of the eggs in each rearing procedure could be supplied as alternative prey for predators, with the remainder served to maintain the colonies for subsequent batches. Only eggs laid within 6 d would be utilized to ensure the high quality of alternative prey for the mass production of predacious bugs. Additionally, a multifunctional device was designed for moth rearing and egg collection, reducing labor input and minimizing health risks for workers coping with inhaled scales. To encourage the production of natural enemies, a cost-effective diet for maintaining a sustainable colony, and a system for daily egg-harvesting of alternative prey were proposed.

The black soldier fly (BSF), Hermetia illucens, has the ability to efficiently bioremediate organic waste into usable bio-compounds. Understanding the impact of domestication and mass rearing on fitness and production traits is therefore important for sustainable production. This study aimed to assess patterns of genomic diversity and its association to phenotypic development across early generations of mass rearing under two selection strategies: selection for greater larval mass (SEL lines) and no direct artificial selection (NS lines). Genome-wide single nucleotide polymorphism (SNP) data were generated using 2bRAD sequencing, while phenotypic traits relating to production and population fitness were measured. Declining patterns of genomic diversity were observed across three generations of captive breeding, with the lowest diversity recorded for the F3 generation of both selection lines, most likely due to founder effects. The SEL cohort displayed statistically significantly greater larval weight com the NS lines with pronounced genetic and phenotypic directional changes across generations. Furthermore, lower genetic and phenotypic diversity, particularly for fitness traits, were evident for SEL lines, illustrating the trade-off between selecting for mass and the resulting decline in population fitness. SNP-based heritability was significant for growth, but was low or non-significant for fitness traits. Genotype-phenotype correlations were observed for traits, but individual locus effect sizes where small and very few of these loci demonstrated a signature for selection. Pronounced genetic drift, due to small effective population sizes, is likely overshadowing the impacts of selection on genomic diversity and consequently phenotypic development. The results hold particular relevance for genetic management and selective breeding for BSF in future.

Feeding on mixed, alternating, or changing diets often favor insect development. With the aim to optimize mass rearing and use for the biological control of insect pests, we investigated the effects of various combinations of high-quality (the green peach aphid Myzus persicae) and low-quality (eggs of the grain moth Sitotroga cerealella) foods on the larval development of a predatory ladybird Cheilomenes propinqua. In the first experiment, eggs and aphids were mixed in different proportions; in the second experiment, larvae switched from feeding on aphids to feeding on eggs. Although the beneficial additive effect of mixed foods was detected in some treatments with limited diets, feeding on various combinations of eggs with aphids never resulted in higher survival, faster development, or a larger size of emerging adults than those observed for feeding on unlimited amounts of aphids. For the practice of biological control, we conclude that, if necessary (for example, in the case of temporary shortage or a lack of aphids in mass rearing facilities or in the case of preventing release of C. propinqua adults in greenhouses), C. propinqua larvae can be fed with grain moth eggs by replacing, mixing, or alternating with aphids, although this will inevitably result in a proportional decrease in pre-adult survival, rate of development, weight, and size of the emerging adults. On the other hand, even a minimal addition of aphids can have a substantial positive effect on larvae fed with grain moth eggs.

Body size is an important morphological characteristic that covaries with the quality of parasitoids and predators. Data show that the larger the organism is, the better the biological parameters and the host location by natural enemies in the field. The standard way of evaluating the size of parasitoids of the genus Trichogramma (Hymenoptera: Trichogrammatidae) is by measuring the tibia, but using only one body part to estimate the size of organisms can lead to miscalculations. In this paper, commercial Trichogramma pretiosum Riley, 1879 (Hymenoptera: Trichogrammatidae) and Trichogramma galloi Zucchi, 1988 (Hymenoptera: Trichogrammatidae) were mounted on slides for microscopy and photographed, and the photographs were used to measure their antennae, scutellum, ovipositor, tibia, and wing. Principal component analysis (PCA) and linear discriminant analysis (LDA) were performed to select the body part that best represents their size. PCA showed that all body parts represented size in a similar way, and LDA showed that the ovipositor was the most representative. We conclude that the best body parts for representing the size of the Trichogramma species studied are the wing and ovipositor, and at least two body parts are needed to detect two size groups.

Long-term storage is an important component of insect mass-rearing systems, prolonging the shelf life of biocontrol agents during a low-demand period or a temporary lack of suitable food. Macrolophus pygmaeus is a predatory heteropteran, mass-reared and widely used for the biological control of arthropod pests in greenhouses. With the aim to determine the optimal conditions and acceptable duration of cold storage, we evaluated the impact of different periods of storage of fed and starved third instar nymphs of M. pygmaeus at different temperatures on nymphal survival, adult emergence, and female fecundity. Four storage temperatures (3, 6, 9, and 12 °C) were tested. The longevity of starved nymphs decreased with an increase in the storage temperature, with a maximum of about 40 days at 3 °C, whereas the longest lifetime of nymphs fed on eggs of the grain moth Sitotroga cerealella (about 150 days) was observed at 9 °C. Further experiments demonstrated that the third instar nymphs of M. pygmaeus fed with eggs of the grain moth can be stored at 9 °C for 30 days, with a moderate (10-20%) decrease in survival and fecundity, whereas the survival of starved nymphs decreased by half after 10 days of storage at 3 °C.

The rearing of saproxylic insects in laboratory conditions is an important task for studying the biology of insects. Through understanding nutritional needs, it is possible to optimize beetle rearing in laboratory conditions. In this study, an artificial fungi-based diet (FD) was developed for the cultivation of the darkling beetle Zophobas atratus (Fabricius, 1775) (Coleoptera: Tenebrionidae) in laboratory conditions as a model object for studying the biology of saproxylophagous beetles. To assess the influence of the diet, a number of physiological parameters were measured, including development time, body size, and weight of all stages of the beetle's life cycle, as well as its immune status. The immune status of Z. atratus was assessed on the basis of larval hemolymph antibacterial activity against six different bacterial strains assessed using disk-diffusion and photometric tests. Our findings show that the FD reduces development time and boosts the immune status as compared to beetles reared on a standard diet (SD). Samples from FD-reared larvae had pronounced antibacterial activity as compared to samples from SD-reared larvae. This work is of fundamental importance for understanding the correlations between nutrition and development of saproxylic Coleoptera and is the first report on immune status regulation in this group of insects.

Control of African animal trypanosomosis is implemented through an integrated control strategy, with the sterile insect technique (SIT) as one of its components. The SIT requires mass rearing of tsetse fly colonies using an in vitro feeding system. The exposure of blood at 37 °C on heating plates over time can have an impact on the quality of fly productivity. In this study, we investigated the survival and fecundity of adult tsetse flies fed at 37 °C on 8 blood exposure times ranging from 30 min to 4 h with increments of 30 min (treatment 1, flies were fed 30 min after exposure to blood at 37 °C; treatment 2, 1 h and so on until treatment 8 [4 h after]) in order to determine the optimal exposure time. In addition, bacterial growth in blood from each treatment was assessed by agar culture at 37 °C for 72 h. The results showed that the adult female survival rates were similar regardless of the treatment. For males, only those of treatment 1 (30 min) showed a marginal lower survival than those of treatments 7 and 8 fed after 3 h 30 min and 4 h of blood exposure, respectively. Over the 4-h interval of blood exposure at 37 °C, the results showed that the number of pupae produced per initial female and pupal weight tended to increase with exposure time, but the differences were not significant. We discuss the implications of these results on tsetse mass rearing for the SIT program.

Spodoptera frugiperda is the preferred host of the parasitoid Telenomus remus. Cold storage can preserve a sufficient quantity of parasitoids and their hosts in a laboratory colony for mass release. First, the effects of cold storage on the reproductive capacity of T. remus reared on non-stored S. frugiperda eggs and the hatching rate of unparasitized S. frugiperda eggs were investigated. Further, the dual effects of cold storage and stored S. frugiperda eggs on the reproductive capacity of T. remus were studied to determine the optimal storage conditions and the maximal shelf life for both the host and the parasitoid. The emergence rate, the number of adults produced, and the female proportion of T. remus were affected by cold storage factors. Pupae stored at 13 °C for 15 days is optimum for T. remus reared on non-stored S. frugiperda eggs. Spodoptera frugiperda eggs could only be stored at 10 °C for five days to be suitable for rearing T. remus. The optimum cold storage conditions for T. remus parasitizing stored eggs were 7 °C for 5 days in the larval stage. The maximal shelf lives of T. remus parasitizing cold-stored S. frugiperda eggs were 10 days. Cold storage affected the hatching rate of S. frugiperda eggs, thereby further affecting the reproductive capacity of T. remus. The findings suggest that different storage conditions should be used when mass-rearing T. remus on stored and non-stored eggs. Telenomus remus should be reproduced using fresh laid S. frugiperda eggs for maximum shelf life.

Spider mites Tetranychus urticae, Tetranychus macfarlanei and Oligonychus biharensis are considered to be highly polyphagous in nature and causes severe damage to a wide range of plants around the world. Amblyseius paraaerialis is an efficient phytoseiid predator of spider mites with a potential to survive on both natural and alternative diets. Evaluation of predatory potential and prey stage preference provides valuable information on the efficacy of the predatory species in controlling mite population. Feeding experiments were conducted on mulberry leaf discs under the laboratory conditions of 30 ± 2 °C and 70 ± 5% relative humidity (RH). After 24 h of feeding experiment, the adult female predator exhibited a significant preference in feeding towards the eggs of T. macfarlanei (42.6%) and the larval stages of T. urticae (46%) and O. biharensis (25.3%). The mass rearing possibilities of A. paraaerialis was tested by tracking and comparing the developmental duration of individual life stages on varied food sources like, honey, castor (Ricinus communis) pollen, honey-pollen mixture and mixed life stages of T. urticae. The predator was failed to complete its development on honey and pollen when supplied separately. However it was successfully developed on honey-pollen mixture and mixed life stages of T. urticae. The developmental studies unravelled a shortest developmental duration and an extended adult longevity and lifespan of A. paraaerialis when reared on the alternative diet, thus opened up the mass rearing possibility of the predatory species under laboratory conditions.

Anthocoris minki Dohrn (Hemiptera: Anthocoridae) is used as a biological control agent of various agricultural pests. This study determined the effect of different egg laying materials, i.e., faba bean, common bean, and pea on population parameters of A. minki using age-stage, two-sex life table. The longest (34.45 d) and the shortest (21.32 d) adult longevity was noted on common bean and pea, respectively. Likewise, the highest (92%) and the lowest (69%) preadult survival rate was recorded on faba bean and pea, respectively. The highest fecundity (93.74 eggs/female) was noted on faba bean followed by common bean (43.95 eggs/female) and pea (48.69 eggs/female). Oviposition period remained unaffected, while higher oviposition days (22 d) were noted on faba bean compared with common bean (10 d) and pea (14 d). The shortest and the longest adult pre-oviposition period and total pre-ovipositional period were calculated for common bean and pea, respectively. The highest intrinsic rate of increase (r) (0.1159 d-1) and finite rate of increase (λ) (1.1229 d-1) were noted on common bean, while the lowest (r = 0.0939 d-1; λ = 1.0985 d-1) were noted on pea. Paired bootstrap analyses indicated that the highest net reproductive rate (R0) (43.12 offspring) was recorded on faba bean. Egg hatching rate significantly differed among plants (P < 0.01) and it was 80.50%, 71.10%, and 38.90% on common bean, faba bean and pea, respectively. It is concluded that faba bean would be the most suitable host for mass rearing of A. minki, while pea proved unsuitable.

The application of the sterile insect technique (SIT) requires the adaptation of insects to mass-rearing conditions. It is generally accepted that this adaptation may include a reduction in genetic diversity and an associated loss of desirable characteristics for the effective performance of sterile insects in the field. Here, we compare the genetic diversity of two mass-reared strains of the Mexican fruit fly, Anastrepha ludens, and a wild (WIL) population collected near Tapachula, Mexico, using seven DNA microsatellites as molecular genetic markers. The mass-reared strains were a bisexual laboratory strain (LAB) with approximately 130 generations under mass-rearing and a genetic sexing strain, Tapachula-7 (TA7), also under mass-rearing for 100 generations. Our results revealed an overall low level of genetic differentiation (approximately 15%) among the three strains, with the LAB and WIL populations being genetically most similar and TA7 most genetically differentiated. Although there were some differences in allele frequencies between strains, our results show that overall, the adaptation to mass-rearing conditions did not reduce genetic variability compared to the wild sample in terms of heterozygosity or allelic richness, nor did it appear to alter the level of inbreeding with respect to the wild populations. These results are contrary to the general idea that mass-rearing always results in a reduction in genetic diversity. Overall, our findings can contribute to a better understanding of the impact that adaptation to mass-rearing conditions may have on the genetic make-up of strains.

The discovery of new and improved factitious and artificial diets is necessary for cost-effective rearing of predatory arthropods. This study evaluated Hermetia illucens black soldier fly (BSF) as a suitable alternative food source for rearing the predatory coccinellid Coleomegilla maculata (Cmac). The hypothesis that BSF larval powder was suitable food to support the growth, development, and reproduction of Cmac was tested in the laboratory. When compared to a standard in-house diet containing brine shrimp egg powder plus Chlorella vulgaris green algae and myristic acid (BSE+CM), the BSF and BSF+CM diets reduced immature growth and development. Immatures successfully reared to teneral adults were smaller when fed BSF or BSF+CM rather than BSE+CM. Combining BSF with a powdered artificial diet (AD), i.e., BSF+AD, did not improve predator growth or development, compared to Cmac reared on BSE+CM. Cmac oviposition responses, i.e., egg clutch production, to BSF vs. BSE+CM or BSF+AD vs. BSE+CM did not differ significantly. In conclusion, BSF has the potential to be food that supports Cmac oviposition behavior. Future research is necessary to discover an ideal mixture of BSF, BSE+CM, or AD that supports Cmac growth, development, and reproduction over multiple generations.

Research on larval rearing and nutrition of tephritid flies on artificial diets is key for the sterile insect technique. Here, we examined the effects of the type of gel (calcium alginate, agar, or carrageenan), at varying percentages in artificial diets for the polyphagous pest Anastrepha ludens, on the physicochemical and nutritional traits of the diets, and the effects of the type of gel, the gel content and the larval density (larvae/g of diet) used in production, quality parameters for mass-reared tephritids, diet removal (an indirect estimation of diet consumption), and nutritional traits of flies. Regardless of the gel content, calcium alginate diets were firmer and more resistant to penetration than the agar and carrageenan diets. The larval recovery, pupation, pupal weight, and flight ability of A. ludens were lower in calcium alginate diets than in agar and carrageenan diets. Diet removal was higher in calcium alginate diets; however, low levels of ammonium and high levels of uric acid in excretions from larvae on these diets suggest an alteration in protein metabolism. The firmness and penetration resistance characteristics of calcium alginate diets may have limited movement and feeding of larvae, but this could be overcome by the collective feeding of large groups of larvae. Our findings provide insights into the mechanism governing gel-diet rearing systems for A. ludens.

Tsetse flies (Glossina spp.; Diptera: Glossinidae) are viviparous flies that feed on blood and are found exclusively in sub-Saharan Africa. They are the only cyclic vectors of African trypanosomes, responsible for human African trypanosomiasis (HAT) and animal African trypanosomiasis (AAT). In this study, we employed high throughput sequencing of the 16S rRNA gene to unravel the diversity of symbiotic bacteria in five wild and three laboratory populations of tsetse species (Glossina pallidipes, G. morsitans, G. swynnertoni, and G. austeni). The aim was to assess the dynamics of bacterial diversity both within each laboratory and wild population in relation to the developmental stage, insect age, gender, and location. Our results indicated that the bacterial communities associated with the four studied Glossina species were significantly influenced by their region of origin, with wild samples being more diverse compared to the laboratory samples. We also observed that the larval microbiota was significantly different than the adults. Furthermore, the sex and the species did not significantly influence the formation of the bacterial profile of the laboratory colonies once these populations were kept under the same rearing conditions. In addition, Wigglesworthia, Acinetobacter, and Sodalis were the most abundant bacterial genera in all the samples, while Wolbachia was significantly abundant in G. morsitans compared to the other studied species. The operational taxonomic unit (OTU) co-occurrence network for each location (VVBD insectary, Doma, Makao, and Msubugwe) indicated a high variability between G. pallidipes and the other species in terms of the number of mutual exclusion and copresence interactions. Improvements are needed in mosquito mass-rearing to effectively implement the sterile insect technique (SIT). However, managing this technique is challenging and resource intensive. SIT relies on mass rearing, sterilization, and release of adult males to reduce field populations. Maintaining an acceptable level of female presence, who can transmit viruses through biting, is crucial. Females are also essential for facility sustainability. Sex sorting plays a vital role in the production process, and our current mechanical sorting approach aims to obtain a high number of adult males with minimal female contamination within 24 h of pupation. Utilizing protandry helps control female contamination. While the 24-h sorting period achieves desired contamination levels, it may not yield enough females to sustain breeding lines, leading to increased labor costs that impact project sustainability. By delaying the sorting procedure to 48 h, we obtained sufficient females to sustain breeding lines, achieving a balance between male production and female contamination using the automatic version of the Fay-Morlan device as the sorting tool.

Wasps in the genus Sclerodermus are ectoparasitoids that typically attack the larvae of woodboring coleopterans. Interest in these species is increasing as they are used in programs to control longhorn beetle pests of economic importance in China and have invasive pest control potential in Europe. Wasps may be mass reared for field release, but using the target host species can be time consuming and physically demanding. There is thus a need for factitious hosts with lower production costs and that are easier to rear. The present research focuses on Sclerodermus brevicornis, which was found in Italy in association with the invasive longhorn beetle, Psacothea hilaris hilaris, and can be laboratory reared on this longhorn beetle and on a factitious lepidopteran host, Corcyra cephalonica. As it is known that the biology of natural enemies can be influenced by the host they emerge from and that the behavior of S. brevicornis is relatively complex due to its degree of sociality (multiple foundress females cooperate to paralyze the host and produce offspring communally), we explored whether, and how, performance and behavioral traits of adult females are influenced by the host species on which they were reared, both when no choice or a choice of current host species was offered. We evaluated the survival of foundresses and their movements between offered hosts and their tendency to form groups with other foundresses according to kinship and host characteristics. We also evaluated the production of offspring and the timing of their development. We found that S. brevicornis reared from C. cephalonica do have some disadvantages compared with those that have developed on P. h. hilaris but also that they recognize, prefer, and can reproduce on P. h. hilaris. We conclude that the use of the more convenient factitious host for mass-rearing is unlikely to greatly compromise the potential of S. brevicornis to suppress longhorn beetle pests in the field.

In recent years, Aedes albopictus (Skuse, 1984) has expanded its distribution globally due to its high ecological plasticity. This expansion has increased the population's susceptibility to contracting diseases such as dengue, Zika, and chikungunya, among others, which are transmitted by this mosquito species. In the absence of effective control methods, the application of the sterile insect technique (SIT) is proposed as part of an integrated vector management (IVM) program. From 2007 to 2020, this strategy has been tested in a non-isolated mosquito population urban area of 45 ha, representative of the municipalities of the Valencian region (Spain). The population levels of adult females and eggs collected in the traps have been reduced by 70-80% compared to the control area, demonstrating its efficacy in reducing mosquito populations. This work analyzes the impact of the migration of the wild mosquito population from the peri-urban area to the urban core.
