Anaerobic digestion (AD) is widely employed for sludge stabilization and waste reduction. However, the slow hydrolysis process hinders methane production and leads to prolonged sludge issues. In this study, an efficient and eco-friendly lysozyme pre-treatment method was utilized to address these challenges. By optimizing lysozyme dosage, hydrolysis and cell lysis were maximized. Furthermore, lysozyme combined with hydrothermal pretreatment enhanced overall efficiency. Results indicate that: (1) When lysozyme dosage reached 90 mg/g TS after 240 min of pretreatment, SCOD, soluble polysaccharides, and protein content reached their maxima at 855.00, 44.09, and 204.86 mg/L, respectively. This represented an increase of 85.87%, 365.58%, and 259.21% compared to the untreated sludge. Three-dimensional fluorescence spectroscopy revealed the highest fluorescence intensity in the IV region (soluble microbial product), promoting microbial metabolic activity. (2) Lysozyme combined with hydrothermal pretreatment significantly increased SCOD, soluble proteins, and polysaccharide release from sludge, reducing SCOD release time. Orthogonal experiments identified Group 3 as the most effective for SCOD and soluble polysaccharide release, while Group 9 released the most soluble proteins. The significance order of factors influencing SCOD, soluble proteins, and polysaccharide release is hydrothermal temperature > hydrothermal time > enzymatic digestion time.(3) The lysozyme-assisted hydrothermal pretreatment group exhibited the fastest release and the highest SCOD concentration of 8,135.00 mg/L during anaerobic digestion. Maximum SCOD consumption and cumulative gas production increased by 95.89% and 130.58%, respectively, compared to the control group, allowing gas production to conclude 3 days earlier.

This study explores the potential of Cucumaria frondosa (C. frondosa) viscera as a natural source of omega-3 FAs using supercritical carbon dioxide (scCO2) extraction. The extraction conditions were optimized using a response surface design, and the optimal parameters were identified as 75 °C and 45 MPa, with a 20 min static and a 30 min dynamic extraction, and a 2:1 ethanol to feedstock mass ratio. Under these conditions, the scCO2 extraction yielded higher FAs than the solvent-based Bligh and Dyer method. The comparative analysis demonstrated that scCO2 extraction (16.30 g of FAs/100 g of dried samples) yielded more fatty acids than the conventional Bligh and Dyer method (9.02 g, or 13.59 g of FAs/100 g of dried samples with ultrasonic assistance), indicating that scCO2 extraction is a viable, green alternative to traditional solvent-based techniques for recovering fatty acids. The pre-treatment effects, including drying methods and ethanol-soaking, were investigated. Freeze-drying significantly enhanced FA yields to almost 100% recovery, while ethanol-soaked viscera tripled the FA yields compared to fresh samples, achieving similar EPA and DHA levels to hot-air-dried samples. These findings highlight the potential of sea cucumber viscera as an efficient source of omega-3 FA extraction and offer an alternative to traditional extraction procedures.

This study optimized the anaerobic digestion (AD) of separated collected organic fractions of municipal solid waste (OFMSW) to produce energy and digestate as biofertilizer. Due to OFMSW's partial recalcitrance to degradation, enzymatic (UPP2, MCPS, USC4, USE2, A. niger) and physical (mechanical blending, heating, hydrodynamic cavitation) pre-treatments were tested. Experimental and modeling approaches were used to compare AD performance regarding energy sustainability and digestate quality. Digestate was separated into solid and liquid fractions, and then chemically and physically characterized by investigating the nutrient release mechanisms. Principal Component Analysis was applied, equally weighing energy and digestate productions. Unlike previous studies focusing only on biogas, this study evaluated the effects of pre-treatments on both biogas and digestate production, viewing AD as a biorefinery process for urban waste valorization. Results showed that all pre-treatments were energetically sustainable, but enzymatic pre-treatments yielded digestates richer in nutrients (increase of 80% N, 200% P and 150% K as compared to OFMSW) and with greater organic matter degradation compared to physical pre-treatments. The liquid fraction of digestate from enzymatic pre-treatments had higher nutrient concentrations, while those from physical pre-treatments had more balanced nutrient content, making them more suitable for fertigation.

The objective of this study was to develop and evaluate a model for predicting post-treatment survival in hepatocellular carcinoma (HCC) patients using their CT images and clinical information, including various treatment information. We collected pre-treatment contrast-enhanced CT images and clinical information including patient-related factors, initial treatment options, and survival status from 692 patients. The patient cohort was divided into a training cohort (n = 507), a testing cohort (n = 146), and an external CT cohort (n = 39), which included patients who underwent CT scans at other institutions. After model training using fivefold cross-validation, model validation was performed on both the testing cohort and the external CT cohort. Our cascaded model employed a 3D convolutional neural network (CNN) to extract features from CT images and derive final survival probabilities. These probabilities were obtained by concatenating previously predicted probabilities for each interval with the patient-related factors and treatment options. We utilized two consecutive fully connected layers for this process, resulting in a number of final outputs corresponding to the number of time intervals, with values representing conditional survival probabilities for each interval. Performance was assessed using the concordance index (C-index), the mean cumulative/dynamic area under the receiver operating characteristics curve (mC/D AUC), and the mean Brier score (mBS), calculated every 3 months. Through an ablation study, we found that using DenseNet-121 as the backbone network and setting the prediction interval to 6 months optimized the model's performance. The integration of multimodal data resulted in superior predictive capabilities compared to models using only CT images or clinical information (C index 0.824 [95% CI 0.822-0.826], mC/D AUC 0.893 [95% CI 0.891-0.895], and mBS 0.121 [95% CI 0.120-0.123] for internal test cohort; C index 0.750 [95% CI 0.747-0.753], mC/D AUC 0.819 [95% CI 0.816-0.823], and mBS 0.159 [95% CI 0.158-0.161] for external CT cohort, respectively). Our CNN-based discrete-time survival prediction model with CT images and clinical information demonstrated promising results in predicting post-treatment survival of patients with HCC.

RESUMO

Sewage sludge (SS) is a potential source of bioenergy, yet its management is a global concern. Anaerobic digestion (AD) is applied to effectively valorize SS by reclaiming energy in the form of methane. However, the complex floc structure of SS hinders hydrolysis during AD process, thus resulting in lower process efficiency. To overcome the rate-limiting hydrolysis, various pre-treatment methods have been developed to enhance AD efficiency. This review aims to provide insights into recent advancements in pre-treatment technologies, including mechanical, chemical, thermal, and biological methods. Each technology was critically evaluated and compared, and its relative worth was summarized based on full-scale applicability, along with economic benefits, AD performance improvements, and impact on digested sludge. The paper illuminates the readers about existing research gaps, and the future research needed for successful implementation of these approaches at full scale.

The objective of this work was to optimize the application of an enzymatic blend produced by Aspergillus niger ATCC 1004 on the Pimenta dioica fruits for essential oil extraction. The enzyme blend was obtained from the fermentation of cocoa bean shells, an agro-industrial residue. The effects of the enzymatic pre-treatment on the extraction yield, the chemical composition of the oil through gas chromatography, and the fruit structure through scanning electron microscopy (SEM) were assessed. A Doehlert design was used to optimize the process conditions, resulting in an extraction with 117 mL of enzyme during 77 min, which increased the extraction yield by 387.5%. The chemical composition was not altered, which proves that the enzyme blend preserves the quality of the essential oil extracted. The content of eugenol (70%), the major compound in the P. dioica essential oil, had a great increase in its concentration (560%). The enzyme activity analyses showed the presence of endoglucanase (0.4 U/mL), exoglucanase (0.25 U/mL), ß-glucosidase (0.19 U/mL), and invertase (135.08 U/mL). The microscopy analyses revealed changes in the morphology of fruit surface due to the enzymatic action. These results demonstrate the great potential of using enzyme blends produced by filamentous fungi from agro-industrial residues for the essential oils extraction of interest for the pharmaceutical and food industries.

BACKGROUND: The World Health Organization (WHO) recommends that HIV treatment scale-up is accompanied by a robust assessment of drug resistance emergence and transmission. The WHO HIV Drug Resistance (HIVDR) monitoring and surveillance strategy includes HIVDR testing in adults both initiating and receiving antiretroviral therapy (ART). Due to limited information about HIVDR in Mozambique, we conducted two nationally representative surveys of adults initiating and receiving first-line ART regimes to better inform the HIV program. METHODS: We carried out a cross-sectional study between March 2017 and December 2019. Adults (older than 15 years) living with HIV (PLHIV) initiating ART or receiving first-line ART for between 9-15 months at 25 health facilities across all eleven provinces in Mozambique were included. Genotypic HIVDR was assessed on dried blood spots (DBS) when viral loads were ≥ 1000 copies/ml. Genotypic resistance for non-nucleoside reverse transcriptase inhibitors (NNRTIs), nucleoside reverse transcriptase inhibitors (NRTIs), and protease inhibitors (PIs) was determined using the Stanford HIV database algorithm 9.5 and calibrated population resistance tool 8.1. RESULTS: Of 828 participants -enrolled, viral load (VL) testing was performed on 408 initiators and 409 ART experienced. Unsuppressed VL was found in 68.1% 419 initiators and 18.8% (77/409) of the ART experienced. Of the 278 initiators and 70 ART experienced who underwent sequencing, 51.7% (144/278) and 75.7% (53/70) were sequenced successfully. Among the new initiators, pretreatment drug resistance (PDR) for NNRTI and PI was found in 16.0% (23/144) and 1.4% (2/144) of the participants, respectively. Acquired drug resistance (ADR) was found in 56.5% (30/53) of the ART-experienced participants of whom 24.5% (13/53) were resistant to both NRTI and NNRTI. CONCLUSION: High rates of PDR and ADR for NNRTI and ADR for NRTI were observed in our study. These findings support the replacement of NNRTIs with dolutegravir (DTG) but high levels of NRTI resistance in highly treatment-experienced individuals still require attention when transitioning to new regimens. Moreover, the study underlines the need for routine VL testing and HIVDR surveillance to improve treatment management strategies.

Peptidomics strategies with high throughput, sensitivity, and reproducibility are key tools for comprehensively analyzing peptide composition and potential functional activities in foods. Nevertheless, complex signal interference, limited ionization efficiency, and low abundance have impeded food-derived peptides' progress in food detection and analysis. As a result, novel functional materials have been born at the right moment that could eliminate interference and perform efficient enrichment. Of note, few studies have focused on developing peptide enrichment materials for food sample analysis. This work summarizes the development of endogenous peptide, phosphopeptide, and glycopeptide enrichment utilizing materials that have been employed extensively recently: organic framework materials, carbon-based nanomaterials, bio-based materials, magnetic materials, and molecularly imprinted polymers. It focuses on the limitations, potential solutions, and future prospects for application in food peptidomics of various advanced functional materials. The size-exclusion effect of adjustable aperture and the modification of magnetic material enhanced the sensitivity and selectivity of endogenous peptide enrichment and aided in streamlining the enrichment process and cutting down on enrichment time. Not only that, the immobilization of metal ions such as Ti4+ and Nb5+ enhanced the capture of phosphopeptides, and the introduction of hydrophilic groups such as arginine, L-cysteine, and glutathione into bio-based materials effectively optimized the hydrophilic enrichment of glycopeptides. Although a portion of the carefully constructed functional materials currently only exhibit promising applications in the field of peptide enrichment for analytical chemistry, there is reason to believe that they will further advance the field of food peptidomics through improved pre-treatment steps.

HIV drug resistance compromises the ability of anti-retroviral therapy (ART) to suppress viral replication, resulting in treatment failure. This study investigates the prevalence of pre-treatment drug resistance (PDR) in newly diagnosed individuals in a prosperous city (Wenzhou) in Southeastern China. A cross-sectional investigation was carried out among 473 newly diagnosed ART-naive HIV-1-infected individuals between January and December 2022. The protease-reverse transcriptase (PR-RT) region and integrase (IN) region of HIV-1 were amplified by two separately nested PCRs, followed by sequencing. Drug resistance mutations (DRMs) and drug resistance to nucleoside reverse transcriptase inhibitors (NRTIs), non-nucleoside reverse transcriptase inhibitors (NNRTIs), protease inhibitors (PIs) and integrase strand transfer inhibitors (INSTIs) were analyzed. The PDR prevalence was 6.5% [95% CI: 4.4-9.1] for any anti-retroviral drug, 0.9% [95% CI: 0.3-2.3] for NRTIs, 4.1% [95% CI: 2.5-6.5] for NNRTIs, 1.8% [95% CI: 0.8-3.6] for PIs and 0.5% [95% CI: 0.1-1.8] for INSTIs. According to the subtyping results of the PR-RT region, 11 different subtypes and 31 unique recombinant forms (URFs) were found. CRF07_BC was the dominant subtype (53.7%, 233/434), followed by CRF01_AE (25.3%, 110/434). V179D (1.6%) and K103N (1.4%) were the most predominant types of NNRTI DRMs. Q58E (1.2%) and M184V (0.7%) were the most frequent PI DRMs and NRTI DRMs, respectively. The INSTI-related DRMs Y143S (causes high-level resistance to RAL) and G163K (causes low-level resistance to EVG and RAL) were found in one patient each. Given the relatively high PDR prevalence of NNRTI (4.1%), non-NNRTI-based ART may be preferred in the future. It is recommended to include genotypic resistance testing before starting ART in regions where feasible.

Understanding sludge rheology and optimizing equipment performance is crucial for energy efficiency in wastewater treatment plants (WWTPs). This study examined sludge rheology after thermal hydrolysis pre-treatment (THP) at 60, 80, and 120 °C for 2 h, followed by anaerobic digestion (AD) at 37 °C for 20 days, and assessed impacts on pump and agitator performance. Post-treatment, sludge showed reduced viscosity and improved flowability, indicated by changes in Herschel-Bulkley parameters, enhancing pump and agitator efficiency, particularly at 120 °C. These rheological improvements were correlated to the solubilization of sludge components after THP and solids reduction after AD, highlighting the interconnectedness of rheology and treatment outcomes. Despite high heat demands, an energy balance showed that THP scenarios, especially at 120 °C, had lower energy requirements for pumps and agitators, leading to energy savings without increased heat consumption. These findings underscore the influence of rheological changes in improving energy efficiency in WWTPs.

Introduction: Patients with platinum resistant epithelial ovarian cancer have limited treatment options which are further limited by hypersensitivity reactions to first line medications such as paclitaxel. Paclitaxel is a taxane that inhibits microtubules and has a high incidence of hypersensitivity reactions. Mirvetuximab soravtansine-gynx (MIRV) is a folate receptor alpha (FRα) directed antibody and microtubule inhibitor that is approved for patients with FRα positive platinum resistant recurrent epithelial ovarian cancer. Both medications are microtubule-targeting agents with similar binding sites, therefore a theoretical risk of cross reactivity between paclitaxel and MIRV may exist. Additionally, phase II clinical trial, SORAYA, did not include data on patients with prior hypersensitivity to paclitaxel. Case: This is the case of a 33-year-old female with recurrent stage IIIC epithelial ovarian cancer with a history of severe anaphylaxis to paclitaxel. She was deemed eligible for MIRV after progression on multiple regimens, but MIRV was given with caution given her severe reaction history. With proper pre-treatment and monitoring, she was treated with MIRV without a reaction. Discussion: It is suspected that most paclitaxel reactions are due to the cremophor solvent rather than paclitaxel itself; however, cross reactivity with docetaxel which is suspended in a polysorbate solution can also occur. Therefore, there is no clear way to determine the risk of cross reactivity between paclitaxel and similar medications. MIRV is also suspended in polysorbate and has a similar mechanism to taxanes, therefore it was unknown if a patient with a prior grade 5 reaction to paclitaxel would also have a reaction to MIRV. Though this is one case, patients with a history of severe hypersensitivity to paclitaxel and meet the criteria for MIRV could be treated with MIRV with careful monitoring.

The stem cell pre-treatment approaches at cellular and sub-cellular levels encompass physical manipulation of stem cells to growth factor treatment, genetic manipulation, and chemical and pharmacological treatment, each strategy having advantages and limitations. Most of these pre-treatment protocols are non-combinative. This editorial is a continuum of Li et al's published article and Wan et al's editorial focusing on the significance of pre-treatment strategies to enhance their stemness, immunoregulatory, and immunosuppressive properties. They have elaborated on the intricacies of the combinative pre-treatment protocol using pro-inflammatory cytokines and hypoxia. Applying a well-defined multi-pronged combinatorial strategy of mesenchymal stem cells (MSCs), pre-treatment based on the mechanistic understanding is expected to develop "Super MSCs", which will create a transformative shift in MSC-based therapies in clinical settings, potentially revolutionizing the field. Once optimized, the standardized protocols may be used with slight modifications to pre-treat different stem cells to develop "super stem cells" with augmented stemness, functionality, and reparability for diverse clinical applications with better outcomes.

BACKGROUND: Extensive use of antibiotics leads to widespread environmental pollution, endangering ecosystems, and human health. It is particularly concerning, posing global threats requiring urgent attention and action. In this regard, the shift to mass spectrometry in determining antibiotics is highly desirable. Significant progress has been made in analyzing and optimizing the sensitivity of high-salt samples. However, the persistence of cumbersome operational procedures presents a significant challenge to this shift. Thus, the persistence of complex operational procedures needs to be addressed. RESULTS: In this study, a rapid and direct method for determining antibiotics in highly saline environmental water samples using microsyringe-based slug-flow microextraction (MSFME)-droplet spray ionization (DSI) mass spectrometry (MS) has been described. The proposed method successfully detected clarithromycin, ofloxacin, and sulfadimidine in seawater within a linear range of 1-1200 ng mL-1, with low limits of detection of 0.19 ng mL-1, 0.17 ng mL-1, and 0.20 ng mL-1, respectively (Signal/Noise = 3). Additionally, spiked real seawater samples of all three antibiotics demonstrated satisfactory recoveries (95.1-107.5%) and precision (RSD≤8.8%). The MSFME-treated high-salt sample (3.5 wt%) showed a mass spectral response intensity 4-5 orders of magnitude higher than the untreated medium-salt sample (0.35 wt%). Furthermore, exploration of the applicability of MSFME showed that it is suitable not only for high-salinity (3.5 wt%) samples but also for salt-free or low-salt and hard water samples rich in calcium and magnesium ions. SIGNIFICANCE: Comparisons with other methods, complex laboratory setups for sample processing are now simplified to a single step, completing the entire process, including desalination and detection, MSFME-DSI-MS provides faster results in less than 1 min while maintaining sensitivity comparable to that of other detection methods. In conclusion, this advancement provides an exceptionally simplified protocol for the rapid, highly sensitive, and quantitative determination of antibiotics in environmental water samples.

BACKGROUND: Patient expectations, including both positive (placebo) and negative (nocebo) effects, influence treatment outcomes, yet their impact on acute repetitive transcranial magnetic stimulation (rTMS) for treatment-resistant depression (TRD) is unclear. METHODS: In this single-center retrospective chart review, 208 TRD patients completed the Stanford Expectation of Treatment Scale (SETS) before starting open-label rTMS treatment. Patients were offered two excitatory rTMS protocols (deep TMS or intermittent theta-burst stimulation), which stimulated the left dorsolateral prefrontal cortex. A minimum of 20 once daily treatments were provided, delivered over 4-6 weeks. Primary outcomes were 1) remission, measured by a post-treatment score of <8 on the Hamilton Depression Rating Scale (HAMD-17), and 2) premature discontinuation. The change in HAMD-17 scores over time was used as a secondary outcome. Physicians were blinded to SETS scores. Logistic and linear regression, adjusting for covariates, assessed SETS and HAMD-17 relationships. RESULTS: Of 208 patients, 177 had baseline and covariate data available. The mean positivity bias score (positive expectancy minus negative expectancy subscale averages) was 0.48 ± 2.21, indicating the cohort was neutral regarding the expectations of their treatment on average. Higher positive expectancy scores were significantly associated with greater odds of remission (OR = 1.90, p = 0.003) and greater reduction in HAMD-17 scores (ß = 1.30, p = 0.005) at the end of acute treatment, after adjusting for covariates. Negative expectancy was not associated with decreased odds of remission (p = 0.2) or treatment discontinuation (p = 0.8). CONCLUSIONS: Higher pre-treatment positive expectations were associated with greater remission rates with open-label rTMS in a naturalistic cohort of patients with TRD.

This work investigated the effectiveness of free nitrous acid (FNA) in enhancing organic waste solubilization to improve biogas production in anaerobic digestion (AD). The results indicated that FNA pretreatment can enhance soluble organic content and control H2S odor in tested organic wastes, including food waste, sewage sludge, and their combination. However, a significant decrease (>50 %) in FNA concentration was found in the reactors, possibly due to denitrifier-driven NO2- consumption. Biochemical methane potential (BMP) tests showed a 25 ± 8 % enhancement in CH4 production in the reactors fed with mixed substrate pretreated with 2.9 mg FNA-N/L. However, the presence of NO2- (325.6-2368.0 mg N/L) in some BMP reactors, due to carryover from FNA pretreatment, adversely affected CH4 production (>55 %) and prolonged lag time (>4.2 times). These findings are valuable for researchers and practitioners in waste management, offering insights for implementing FNA pretreatment to enhance the biodegradability of organic wastes in AD.

(1) Background: The objective of this in vitro study was to evaluate the impact of different etching times and ethanol pre-treatments on the immediate bond strength of a hydrophilic multi-mode universal adhesive (Clearfil Universal Bond Quick, Kuraray, UBQ) and on the consequent gelatinolytic activity of metalloproteinases (MMPs) on radicular dentin. (2) Methods: Sixty single-root teeth were selected and divided into four groups according to the adhesive protocol applied for fiber post cementation: (G1) 15 s H3PO4 application + UBQ; (G2) 30 s H3PO4 application + UBQ; (G3) 15 s H3PO4 application + ethanol pre-treatment + UBQ; (G4) 30 s H3PO4 + ethanol pre-treatment + UBQ. After adhesive procedures, fiber posts were luted into the post space with a dual-curing cement (DC Core, Kuraray) and light-cured for 40 s. To perform the push-out test and nanoleakage analyses for both coronal end apical areas, 1 mm slices were prepared, following a 24 h storage period in artificial saliva. Additionally, an in situ zymographic assay was conducted to explore endogenous MMP activity within the radicular layer. Results were statistically analyzed with ANOVA and Tukey post hoc tests. Statistical significance was set at p < 0.05. (3) Result: ANOVA revealed a statistically significant difference in push-out bond strength related to the pre-treatment variable but did not highlight any significance of etching time. Specimens pre-treated with ethanol wet bond application showed higher bond strength (p < 0.01). In situ zymography quantification analyses revealed that all tested groups, independently of etching time end ethanol pre-treatment, activated MMP gelatinolytic activity. A significant increase in MMP activity was detected for the 30 s etching time. However, ETOH pre-treatment significantly reduced MMP activity within the adhesive interface (p < 0.01). (4) Conclusions: The tested adhesive showed similar results regardless of the etching time protocol. The gelatinolytic activity of MMPs was observed in all the groups. Further investigations and extended follow-ups are required to validate the results of the present study in vivo.

Polylactic Acid (PLA) based compostable bioplastic films degrade under thermophilic composting conditions. The purpose of our study was to understand whether sample pre-treatment along with bioaugmentation of the degradation matrix could reduce the biodegradation time under a simulated composting environment. Sepcifically, we also explored whether the commercial composts could be replaced by landfill-mined soil-like fraction (LMSF) for the said application. The effect of pre-treatment on the material was analysed by tests like tensile strength analysis, hydrophobicity analysis, morphological analysis, thermal degradation profiling, etc. Subsequently, the degradation experiment was performed in a simulated composting environment following the ASTM D5338 standard, along with bioaugmentation in selected experimental setups. When the novel approach of material pre-treatment and bioaugmentation were applied in combination, the time necessary for 90% degradation was reduced by 27% using compost and by 23% using LMSF. Beyond the improvement in degradation rate, the water holding capacity increased significantly for the degradation matrices. With pH, C: N ratio and microbial diversity tested to be favourable through 16s metabarcoding studies, material pre-treatment and bioaugmentation allow LMSF to not only replace commercial compost in polymer degradation but also find immense application in the agricultural sector of drought-affected areas (for better water retention) after it has been used for PLA degradation.

Due to the highly stable structure of keratin, the extraction and dissolution steps of animal medicines rich in keratin are complex, which seriously restricts the detection efficiency and flux. Therefore, this study simplified the pre-treatment steps of horn samples and optimized the detection methods of characteristic peptides to improve the efficiency of identifying the specificity of horn-derived animal medicines. For detection of the characteristic peptides in horn-derived animal medicines treated with/without iodoace-tamide(IAA), the ion pair conditions of the characteristic peptides were optimized, and the retention time, intensity and other data of the specific peptides were compared between the samples treated with/without IAA. Two pre-treatment methods, direct enzymatic hydrolysis and total protein extraction followed by enzymatic hydrolysis, were used to prepare horn-derived animal medicine samples. The effects of different methods on the detection of specific peptides in the samples of Saiga antelope horn, water buffalo horn, goat horn, and yak horn were compared regarding the retention time of specific peptides and ion intensity. The results indicated that after direct enzymatic hydrolysis, the specific peptides in the samples without IAA treatment can be detected. Compared with the characteristic peptides in the samples treated with IAA, their retention time shifted back and the mass spectrometry response slightly decreased. The specific peptides of the samples without IAA treatment had good specificity and did not affect the specificity identification of horn-derived animal medicines. Overall, the process of direct enzymatic hydrolysis can be used to treat horn samples, omitting the steps of protein extraction and dithiothreitol and IAA treatment, significantly improving the pre-treatment efficiency without affecting the specificity identification of horn-derived animal medicines. This study provides ideas for quality research and standard improvement of horn-derived animal medicines.

Replacing petroleum-based plastics with biodegradable polymers is a major challenge for modern society especially for food packaging applications. To date, poly(lactic acid) represents 25 % of the total biodegradable plastics and it is estimated that, in the future, it could become the main contributor to the biodegradable plastics industry. Anaerobic digestion is an interesting way for the poly(lactic acid) end of life, even if its biodegradability is limited in mesophilic conditions. The aims of this study were to identify the best pre-treatment for maximizing the methane yield, minimizing the anaerobic digestion duration and limiting residual plastic fragments in the digestate. A systematic comparison was carried out between thermal, chemical, and thermo-chemical pre-treatment. Pre-treatment with 4 M KOH for 48 h at 35°C was effective in improving the mesophilic anaerobic digestion of the poly(lactic acid). Such pre-treatment allows obtaining 90 % of the theoretical methane potential, in 24 - 30 days. Importantly, such pre-treatment completely solubilized the poly(lactic acid), leaving no solid residues in the digestate. In addition, using KOH permits to avoid the sodication of the soil due to the digestate application as fertilizer.

BACKGROUND: Isolated microspore culture is a useful biotechnological technique applied in modern plant breeding programs as it can produce doubled haploid (DH) plants and accelerate the development of new varieties. Furthermore, as a single-cell culture technique, the isolated microspore culture provides an excellent platform for studying microspore embryogenesis. However, the reports on isolated microspore culture are rather limited in rice due to the low callus induction rate, poor regeneration capability, and high genotypic dependency. The present study developed an effective isolated microspore culture protocol for high-frequency androgenesis in four japonica rice genotypes. Several factors affecting the isolated microspore culture were studied to evaluate their effects on callus induction and plantlet regeneration. RESULTS: Low-temperature pre-treatment at 4 â„ƒ for 10-15 days could effectively promote microspore embryogenesis in japonica rice. A simple and efficient method was proposed for identifying the microspore developmental stage. The anthers in yellow-green florets located on the second type of primary branch on the rice panicle were found to be the optimal stage for isolated microspore culture. The most effective induction media for callus induction were IM2 and IM3, depending on the genotype. The optimal concentration of 2, 4-D in the medium for callus induction was 1 mg/L. Callus induction was negatively affected by a high concentration of KT over 1.5 mg/L. The differentiation medium suitable for japonica rice microspore callus comprised 1/2 MS, 2 mg/L 6-BA, 0.5 mg/L NAA, 30 g/L sucrose, and 6 g/L agar. The regeneration frequency of the four genotypes ranged from 61-211 green plantlets per 100 mg calli, with Chongxiangjing showing the highest regeneration frequency. CONCLUSIONS: This study presented an efficient protocol for improved callus induction and green plantlet regeneration in japonica rice via isolated microspore culture, which could provide valuable support for rice breeding and genetic research.
