The objective of this study was to assess the effects of prepartum administration of anti-inflammatory therapies on body condition score (BCS), ß-hydroxybutyrate (BHB) concentration, haptoglobin (HP) concentration, milk yield, milk components, rumination time, clinical health events and reproductive performance in Holstein dairy cows. At 14 d before the expected calving date, cows (PAR; n = 170) and heifers (nulliparous [NUL]; n = 63) were blocked by BCS group (optimal = 3-3.5 [OPT]; over-conditioned cows [OVERC; BCS ≥ 3.75 pts.]) and parity (NUL; PAR) and randomly allocated to one of 3 treatment groups: 1) ASA (n = 78): receive one oral administration of acetylsalicylic acid (4 boluses; 480 grain/bolus); 2) MEL (n = 76): receive one oral administration with meloxicam (1mg/kg of BW), or 3) PLC (n = 77): receive one oral treatment with gelatin capsules filled with water. Body condition score was assessed, and blood samples were collected, weekly starting one week before treatment until 3 weeks after calving. Daily milk yields and daily rumination times were collected from on-farm computer records. Dairy Herd Improvement Association (DHIA) monthly test data were collected to assess milk yield, somatic cell counts, and milk components. Furthermore, health events, culling rate, and reproductive performance data were collected from on-farm computer records. The data were analyzed using MIXED, GLIMMIX, and LIFETEST procedures of SAS as a randomized complete block design. On average, MEL-NUL cows produced 4.77 ± 0.93 kg/d and 4.81 ± 0.92 kg/d more milk from wk 6 to wk 21 of lactation compared with ASA-NUL and PLC-NUL cows, respectively. Similarly, there was a week by treatment by body condition group interaction (P = 0.01), where OVERC cows treated with MEL produced more milk from wk 10 to wk 15 of lactation compared with ASA- OVERC and PLC-OVERC cows. Parous cows treated with ASA had lower BCS compared with PAR cows treated with MEL or PLC. A lower percentage of OVERC cows treated with ASA became sick in the first 60 DIM compared with MEL- OVERC and PLC- OVERC cows (ASA = 23.88 ± 7.26%, MEL = 46.36 ± 8.57%; PLC = 46.74 ± 8.53%; P = 0.04). Parous cows treated with ASA had (P = 0.03) a higher hazard ratio to become pregnant by 300 DIM compared with PAR MEL cows. Although the study was not sized for finding treatment differences in blocking criteria groups, these results suggest that treatment with prepartum anti-inflammatory therapies may have positive effects on milk yield and postpartum health in specific groups of cows, such as NUL and OVERC cows, while it may not be recommended for other animal categories, such as parous cows and cows with optimal BCS. Larger studies are needed to strengthen the associations observed in this study.

The objectives of this study were to identify factors associated with the relative change in prepartum dry matter intake (RCDMI) of 273 cows fed individually, evaluate the performance of linear models to estimate RCDMI using different classes of predictors, and characterize the implications of RCDMI to transition metabolism, health, and subsequent milk production. Two periods of interest were established. Period 1 comprised d -21 to -12 relative to calving, when DMI was stable. Period 2 comprised d -4 to -1, when average DMI was distinctly declined. The RCDMI from period 1 to 2 was calculated as a percentage value, which ranged from -75 to 15% and averaged -18.1% (±15.0). Season, parity, body fatness, body weight, milk production in the previous lactation and at dry-off, and length of dry period were associated with RCDMI and explained 11% of the variation in all cows, and 19% when only parous cows were considered. Performance of linear models to predict RCDMI was improved when data on rumination and physical activities and target blood metabolites were added. The adjusted R2 increased to values between 0.45 and 0.55, and selected models performed consistently in cross-validation analyses. To evaluate the implications of RCDMI, cows were ranked within parity according to RCDMI and classified into terciles as large decline (LD), moderate decline (MD), or small decline (SD). By design, DMI did not differ between tercile groups in period 1 (13.3 ± 0.2 kg/d), but differed substantially in period 2 (LD = 8.8; MD = 11.2; SD = 12.7 ± 0.2 kg/d), creating important differences in RCDMI among groups (LD = -33.8; MD = -16.2; SD = -3.4% ± 0.8%). At enrollment, cows in the LD and MD groups were heavier (LD = 788; MD = 775; SD = 750 ± 7 kg), and the proportion of cows with BCS >3.5 was higher in LD (LD = 63; MD = 47; SD = 38%). An interaction of group and time was observed for postpartum DMI, which started lower in LD than in SD cows, but equaled by the end of transition, and inverted at wk 13 and 14 after calving. Yields of energy-corrected milk were greater in LD than in SD cows, and both groups did not differ from MD (LD = 41.0; MD = 40.3; SD = 39.0 ± 0.5 kg/d). LD cows had decreased energy balance and greater concentrations of nonesterified fatty acids, ß-hydroxybutyrate, and aspartate aminotransferase in serum, and greater glutathione peroxidase activity in plasma than SD cows. Larger declines in prepartum DMI were also associated with increased risk for postpartum disease, although the associations were only weak to moderate. In conclusion, a large decline in prepartum DMI was associated with important adjustments in the energy metabolism and antioxidants activities, and greater milk production in the subsequent lactation. These findings indicate that feed intake decline close to parturition is likely a normal response to physiological adaptations at the onset of lactation when cows are fed at libitum.

BACKGROUND: The rumen is a crucial digestive organ for dairy cows. The rumen microbiota assists in the digestion of plant feed through microbe-mediated fermentation, during which the plant feed is transformed into nutrients for the cow's use. Variations in the composition and function of the rumen microbiome affect the energy utilization efficiency of dairy cows, which is one of the reasons for the varying body condition scores (BCSs). This study focused on prepartum Holstein dairy cows to analyze differences in rumen microbiota and metabolites among cows with different BCSs. Twelve prepartum dairy cows were divided into two groups, low BCS (LBCS, BCS = 2.75, n = 6) and high BCS (HBCS, BCS = 3.5, n = 6), to explore differences in microbial composition and metabolites. RESULTS: In the HBCS group, the genera within the phylum Firmicutes exhibited stronger correlations and greater abundances. Phyla such as Firmicutes, Patescibacteria, Acidobacteriota, Euryarchaeota, and Desulfobacterota, in addition to most of their constituent microbial groups, were significantly more abundant in the HBCS group than in the LBCS group. At the genus level, the abundances of Anaerovibrio, Veillonellaceae_UCG_001, Ruminococcus_gauvreauii_group, Blautia, Eubacterium, Prevotellaceae_YAB2003_group, Schwartzia, and Halomonas significantly increased in the HBCS group. The citrate cycle, involved in carbohydrate metabolism, exhibited a significant enrichment trend, with a notable increase in the abundance of its key substrate, citrate, in the HBCS group. This increase was significantly positively correlated with the differential bacterial genera. CONCLUSION: In this study, prepartum dairy cows with higher BCS exhibited greater abundance of Firmicutes. This study provides theoretical support for microbiological research on dairy cows with different BCSs and suggests that regulating the rumen microbiome could help maintain prepartum dairy cows within an optimal BCS range.

The placenta secretes a prolactin (PRL)-like hormone PRL3B1 (placental lactogen II), a luteotropic hormone essential for maintaining pregnancy until labor in mice. A report from 1984 examined the secretion pattern of PRL3B1 in prepartum mice. In the current study, we found contradictory findings in the secretion pattern that invalidate the previous report. By measuring maternal plasma PRL3B1 and PRL every 4 hrs from gestational day 17 (G17), we newly discovered that maternal plasma PRL3B1 levels decrease rapidly in prepartum C57BL/6 mice. Interestingly, the onset of this decline coincided with the PRL surge at G18, demonstrating a plasma prolactin axis shift from placental to pituitary origin. We also found that maternal plasma progesterone regression precedes the onset of the PRL shift. The level of Prl3b1 mRNA was determined by RT-qPCR in the placenta and remained stable until parturition, implying that PRL3B1 peptide production or secretion was suppressed. We hypothesized that production of the PRL family, the 25 paralogous PRL proteins exclusively expressed in mice placenta, would decrease alongside PRL3B1 during this period. To investigate this hypothesis and to seek proteomic changes, we performed a shotgun proteome analysis of the placental tissue using data-independent acquisition mass spectrometry (DIA-MS). Up to 5,891 proteins were identified, including 17 PRL family members. Relative quantitative analysis between embryonic day 17 (E17) and E18 placentas showed no significant difference in the expression of PRL3B1 and most PRL family members except PRL7C1. These results suggest that PRL3B1 secretion from the placenta is suppressed at G18 (E18).

The negative energy balance occurring in the periparturient period of cows will impede their health and postpartum performance. To target this issue, L-tryptophan was supplied to the prepartum cows. The results showed that L-tryptophan supplementation significantly increased the serum melatonin level and was accompanied with increases in SOD activity, IL-10 and colostrum IgA levels as well as decreases in MDA and IL-6 levels compared to the control cows. The incidence of postpartum diseases was significantly lower and the pregnancy rate was significantly higher in cows fed L-tryptophan than in the control group. A striking observation was that prepartum L-tryptophan supplementation not only improved the milk production but also the quality compared to the control cows. In general, supplementation with L-tryptophan in the prepartum period can improve the postpartum reproduction and lactation performance of cows to some extent.

This study evaluated the effects of maternal supplementation of a Bacillus-based direct-fed microbial (DFM) on the physiology and growth performance of Bos indicus-influenced cow-calf pairs. On day 0 (~139 d before expected calving date), 72 fall-calving, Brangus crossbred beef heifers (20 to 22 mo of age) pregnant with first offspring were stratified by their initial body weight (BW; 431 ±â€…31 kg) and body condition score (BCS; 6.0 ±â€…0.36; scale 1 to 9), and randomly allocated into 1 of 12 bahiagrass pastures (1 ha and six heifers per pasture). Treatments were randomly assigned to pastures (six pastures per treatment) and consisted of heifers supplemented with 1 kg/d of soybean hulls (dry matter, DM) that was added (BAC) or not (CON) with DFM containing Bacillus subtilis and B. licheniformis (Bovacillus; Chr. Hansen A/S, Hørsholm, Denmark). Treatments were provided from days 0 to 242 (139 ±â€…4 d prepartum to 104 ±â€…4 d postpartum). Calves were weaned on day 242 (96 ±â€…30 d of age) and then allocated into 1 of 16 drylot pens and fed the same concentrate at 3.25% of BW (DM) until day 319. Maternal treatment effects were not detected (P ≥ 0.29) for herbage allowance and forage chemical composition. Heifer BCS on days 39 and 63 tended (P ≤ 0.09) to be greater for BAC vs. CON heifers, whereas heifer BCS on day 91 was greater (P = 0.01) for BAC vs. CON heifers. Heifer BCS did not differ (P ≥ 0.20) between treatments on days 179 and 242. Plasma glucose concentration did not differ from days 0 to 63 (P ≥ 0.14) but were greater (P < 0.01) on day 179 and tended (P = 0.09) to be greater on day 242 for BAC vs. CON heifers. Calf BW at birth, ADG from birth to weaning, and BW at weaning did not differ (P ≥ 0.19) between treatments, but calf BW at drylot exit (day 319) was greater (P = 0.05) for BAC vs. CON calves. Maternal treatment effects were not detected (P ≥ 0.42) for calf serum concentration of IgG at birth and postvaccination plasma concentrations of glucose, cortisol, and haptoglobin. Serum titers against bovine respiratory syncytial virus (BRSV) were greater (P = 0.04) for BAC vs. CON calves on day 287, whereas seroconversion against parainfluenza-3 virus (PI-3) was greater (P < 0.01) for BAC vs. CON calves on day 271. Thus, maternal supplementation of a Bacillus-based DFM increased prepartum BCS gain and postpartum plasma glucose concentration of heifers and led to positive carryover effects on postweaning BW gain and humoral immune response in their offspring.


Direct-fed microbials (DFM), such as Bacillus spp., have been shown to produce a wide variety of enzymes related to nutrient digestion and to support gastrointestinal tract immune function and integrity, leading to increased nutrient digestibility and cattle performance. Nutritional management of beef cows during gestation and early lactation has been associated with enhanced future offspring growth performance and immune response following birth. The present study combined the use of Bacillus-based DFM for pregnant heifers during critical production stages (late gestation and early lactation) to promote the overall performance of heifers and their offspring. Heifers offered Bacillus-based DFM had greater body condition score at calving and postpartum plasma concentration of glucose, whereas their offspring had similar body weight at birth, but greater growth performance when fed relatively high amounts of protein and energy in drylot compared to cohorts born from heifers that did not receive Bacillus-based DFM supplementation.

BACKGROUND: Prepartum and postpartum maternal symptoms of stress, anxiety and depression are likely to influence the child's sensory processing through hormonal alterations and an influence on mother-child interactions. OBJECTIVE: We investigated the associations between maternal prepartum and postpartum symptoms of depression, anxiety and stress related to the COVID-19 pandemic and childhood sensory avoidance at 18 months. METHODS: Longitudinal data from 409 participants followed during the COVID-19 pandemic were used. They completed questionnaires during pregnancy and up to 18 months after delivery. Maternal distress symptoms were assessed prenatally and at 18 months postnatally using the Edinburgh Postnatal Depression Scale, the Generalized Anxiety Disorders 7-item Scale and a 10-point scale assessing the level of stress felt related to the COVID-19 pandemic. Child sensory avoidance was assessed at 18 months postpartum using the Infant/Toddler Sensory Profile-Second Edition. Pearson correlations and multiple regressions measured the associations between maternal distress symptoms and child sensory avoidance. RESULTS: Prepartum and 18-month postpartum maternal depression and anxiety were significantly correlated with childhood sensory avoidance (p < 0.05). Together, these variables explained 7.18% (F = 2.12, p < 0.05) of the variance of childhood sensory avoidance. CONCLUSIONS: These results support the contributory effect of prepartum and postpartum maternal distress on childhood sensory development.

Retrospective analyses were performed on a dataset of 1188 fall-calving, Brangus cow-calf pairs. Analyses 1 sorted cows according to their initial body condition score (BCS < 5 vs. ≥ 5) and whether they received (SUP) or not (NOSUP) prepartum supplementation of protein and energy. Analyses 2 sorted cows according to their calving BCS (BCS < 5 or ≥ 5) and BCS change from calving until the start of the breeding season (lost, maintained, or gained). Cows were not estrus synchronized and were assigned to natural breeding for 90 days. Prepartum supplementation increased (P = 0.04) pregnancy percentage in cows with initial BCS < 5 but not (P = 0.20) with initial BCS ≥ 5. Calf weaning weight was greatest (P ≤ 0.04) for calves born from SUP cows with an initial BCS ≥ 5 and did not differ (P ≥ 0.56) among all remaining groups. Among cows with calving BCS < 5, pregnancy percentage were less (P = 0.05) for cows that lost vs. maintained/gained BCS. Postpartum BCS change did not (P ≥ 0.16) impact pregnancy percentage of cows calving at BCS ≥ 5. Calf weaning weight increased (P < 0.01) for cows calving with BCS ≥ 5 vs. < 5 and was not impacted (P = 0.47) by postpartum BCS change. Therefore, precalving supplementation improved reproduction of cows with BCS below optimal and weaning weight of calves born from cows with BCS above optimal, whereas calving BCS was the major factor affecting postpartum BCS change and cow reproductive performance.

OBJECTIVE: Parturition is crucial for dams, their calves, and cow managers. The prediction of calving time, which assists cow managers to decide on the relocation of cows to maternity pens and necessity of human supervision, is a pivotal aspect of livestock farming. However, existing methods of predicting calving time in dairy cows based on hormonal changes and clinical symptoms are time-consuming and yield unreliable predictions. Accordingly, we investigated whether heart rate variability (HRV) which is a non-invasive assessment of autonomic nervous system (ANS) activity and behavior during the prepartum period would be useful for predicting calving time in dairy cows. METHODS: Eight pregnant cows were surveilled under electrocardiogram and video recordings for HRV and behavioral analyses, respectively. HRV parameters in time and frequency domains were evaluated. A 24-h time budget was calculated for each of six types of behavior (standing and lying with or without rumination, sleeping, and eating). RESULTS: Heart rate on calving day is considerably higher than those recorded on the days preceding calving. Low frequency power declined, whereas high frequency power escalated on the calving day compared to the period between 24 and 48 h before calving. The time budget for ruminating while lying decreased and that while standing increased markedly on the calving day compared to those allocated on the preceding days; nonetheless, the total time budget for ruminating did not differ during the prepartum period. CONCLUSION: We elucidated the ANS activity and behavioral profiles during prepartum period. Our results confirm that HRV parameters and behavior are useful for predicting calving time, and interestingly indicate that the time budget for ruminating while standing (or lying) may serve as a valuable predictor of calving. Collectively, our findings lay the foundation for future investigations to determine other potential predictors and formulate an algorithm for predicting calving time.

OBJECTIVE: To evaluate the association between maternal fasting time before delivery and the occurrence of hypoglycemia in neonates immediately after birth. METHODS: This prospective single-center cohort study included pregnant women who delivered at the study institution between October 2021 and January 2023 and their neonates. The primary outcome was the incidence of neonatal hypoglycemia after birth, defined as a blood glucose level less than 47 mg/dL. Fasting time was categorized into quartiles, and the association between maternal fasting time and neonatal hypoglycemia was investigated. The crude or adjusted odds ratios of maternal fasting time for neonatal hypoglycemia were calculated using logistic regression analysis. RESULTS: The study included 663 pregnant women and 696 neonates. Compared with the reference group with a short fasting time of 4.3 h or less, the adjusted odds ratios for neonatal hypoglycemia were 1.47 (95% confidence interval [CI] 0.70-3.20) for middle fasting time (4.3-9.8 h), 4.05 (95% CI 2.02-8.56) for long fasting time (9.8-14.6 h), and 4.99 (95% CI 2.59-10.25) for very long fasting time (>14.6 h). In the subgroup analysis, the association between maternal fasting time and neonatal hypoglycemia showed different trends according to the mode of delivery. CONCLUSION: Maternal fasting time over 9-10 h before delivery was associated with the occurrence of neonatal hypoglycemia. Obstetrical management, considering not only maternal safety but also neonatal hypoglycemia prevention, is required.

Introduction: This study aimed to evaluate the implications of supplementary nutrition during the mid-to-late pregnancy on various parameters in Hanwoo cows and their subsequent neonatal calves. Materials and methods: Eight Hanwoo cows in their first parity were divided into two groups. The control group (C, 100%) received 3kg of concentrate and 5kg of rice straw throughout the pregnancy period, while the treatment group (T, 150%) increased their diet during mid-to-late pregnancy. Both performance assessments and blood metabolite analyses were performed for the pregnant cows. Neonatal calves were subjected to morphometric evaluations, blood sampling, and detailed morphometric analyses of carcasses and gastrointestinal components. Results: Performance indices of the cows showed that both Pregnancy Period (PregP) and Body Condition Score (BCS) were significantly improved with supplemental feeding (p <0.05). Improvements in Body Weight (BW) and Feed Conversion Ratio (FCR) were not statistically significant. Blood metabolite analysis for the cows revealed decreased levels of triglycerides (TGLate), non-esterified fatty acids (NEFALate), and progesterone (P4Late), with a notable increase in glucose (GluLate) levels (p <0.01). In the neonatal calves, anatomical metrics of the gastrointestinal tissues showed increased Omasum Width (OmasWdth) values in the supplemented group (p =0.053). There was significant increase of papillae and villus lengths in the rumen and small intestine (p <0.01 and p <0.05, respectively). Morphometric evaluations displayed longer body lengths (BLnth) and larger chest width (ChestWdth) in the treated calves (p <0.05 and p <0.01, respectively). Carcass characteristics showed no substantial variations between the groups, while blood analysis in the calves revealed decreased GPT levels in the nutritionally supplemented group (p<0.05). Discussion: The findings indicate that supplementing the diets of Hanwoo cows during mid-to-late pregnancy leads to significant changes in select maternal blood metabolites and influences specific anatomical and morphometric features in neonatal calves, all without significant shifts in carcass attributes.

Postpartum hemorrhage (PPH) is responsible for 30% to 50% of maternal deaths. There is conflicting evidence if prepartum anemia facilitates PPH. A comprehensive analysis of studies describing their relation is missing. An extensive database search was conducted applying the terms "anemia" OR "hemoglobin" AND "postpartum hemorrhage." We used a random-effects meta-analysis model to estimate an overall odds ratio (OR) for PPH and prepartum anemia, separating studies that were conformant and non-conformant with the World Health Organization (WHO) definitions for anemia. The search yielded 2519 studies, and 46 were appropriate for analysis. The meta-analyses of WHO-conformant (n = 22) and non-conformant (n = 24) studies showed that the risk of PPH was increased when anemia was present. The ORs were 1.45 (CL: 1.23-1.71) for WHO-conformant studies, 2.88 (CL: 1.38-6.02) for studies applying lower thresholds for anemia, and 3.28 (CL: 2.08-5.19) for undefined anemia thresholds. PPH risk appeared to increase with lower anemia thresholds. Prepartum anemia is associated with an increased risk of PPH, an observation that is important regarding improved anemia correction strategies such as iron supplementation.

RESUMO

This study evaluated the growth and immune response of beef calves born from Bos indicus-influenced beef heifers provided pre- and postpartum heat abatement on pasture. On 83 ±â€…4 d prepartum (day 0), 64 Brangus crossbred beef heifers (~» B. indicus) were stratified by body weight (BW; 454 ±â€…37 kg) and body condition score (BCS; 6.3 ±â€…0.28; scale 1 to 9), and then allocated into 1 of 16 bahiagrass pastures (1 ha and 4 heifers per pasture). Treatments were randomly assigned to pastures (8 pastures per treatment) and consisted of heifers provided (SH) or not (NSH) access to artificial shade (4.5 m2 of shade area per heifer) from 83 d prepartum to 50 d postpartum (days 0 to 133). Heifers and calves were managed similarly from day 133 until the start of the breeding season (day 203). Calves were weaned on day 203 (at 119 ±â€…19 d of age), limit-fed the same drylot diet at 3.5% of BW (DM basis) days 209 to 268 (3 to 4 calves per pen; 8 pens per treatment) and vaccinated against respiratory disease pathogens on days 222 and 236. Heifer intravaginal temperatures from days 35 to 42 were lower (P ≤ 0.03) for NSH vs. SH heifers from 0000 to 0800 hours but greater (P ≤ 0.05) for NSH vs. SH heifers from 1100 to 1800 hours. Heifer intravaginal temperature from days 126 to 132 did not differ (P = 0.99) between NSH and SH heifers. Heifers assigned to NSH had greater respiration rates from days 20 to 96 (P ≤ 0.0007), greater plasma concentration of cortisol on days 35 (P = 0.07) and 55 (P = 0.02), less plasma concentration of insulin-like growth factor 1 (IGF-1) on days 35 (P = 0.10), 55, and 133 (P ≤ 0.05), and less BCS from days 55 to 203 (P ≤ 0.01) compared to SH heifers. Calves born from NSH heifers had less birth BW (P = 0.05), greater overall plasma haptoglobin concentrations (P = 0.05), greater seroconversion against bovine respiratory syncytial virus on day 222 (P = 0.02), tended to have greater ADG from days 209 to 268 (P = 0.07), and had greater BW on day 268 (P = 0.05) compared to SH offspring. Plasma concentrations of cortisol and serum titers against other respiratory disease pathogens did not differ (P ≥ 0.15) between NSH and SH offspring. Hence, removing maternal access to artificial shade: (1) increased prepartum intravaginal temperature and plasma concentrations of cortisol but reduced prepartum BCS and plasma concentrations of IGF-1 in grazing B. indicus-influenced beef heifers; and (2) increased post-weaning BW gain and had positive effects on humoral immune response of their offspring.


In dairy cattle, heat stress during late gestation negatively impacted offspring postnatal growth and immune function. The use of artificial shade is a management strategy that alleviates heat stress in environments with high temperatures and humidity. The effects of maternal access to artificial shade and its impacts on offspring performance have not been reported for Bos indicus influenced-beef cattle and were the main objective of the present study. From 83 d prepartum until 50 d postpartum, heifers had access or not to an artificial shade structure located centrally on their pastures. Overall, removing maternal access to artificial shade increased maternal internal body temperature and respiration rates, and reduced maternal body condition score from calving until the start of the breeding season and calf body weight at birth. However, calves born from heifers with no access to shade were remarkably heavier at the end of the drylot period and had lower plasma indicators of inflammatory response and positive effects to humoral immune response to vaccination.

This study evaluated the growth and immune function of beef calves born to cows supplemented with bakery waste containing two concentrations of crude fat. On day 0 (~90 d before calving), 108 multiparous Brangus crossbred cows were stratified by body weight (BW; 551 ±â€…65 kg) and body condition score (BCS, 5.5 ±â€…0.9) and randomly allocated into 1 of 18 bahiagrass (Paspalum notatum) pastures (6 cows and 4.3 ha per pasture). Treatments were randomly assigned to pastures (6 pastures per treatment) and consisted of no prepartum supplementation (NOSUP) and isocaloric and isonitrogenous supplementation of low-fat (LFAT; 6.4% crude fat) or high-fat (HFAT; 10.7% crude fat) bakery waste from days 0 to 70 (1 kg DM per cow per day). Calves were weaned on day 292 (201 ±â€…17 d of age). Then, 15 heifers per treatment were randomly selected and assigned to drylot pens from days 300 to 345 and vaccinated against respiratory pathogens on days 300 and 315. Cow BCS near calving (day 70) was the least (P ≤ 0.05) for NOSUP cows and did not differ (P = 0.12) between LFAT and HFAT cows. Cow BCS at the start of the breeding season (day 140) was greater (P = 0.05) for HFAT vs. NOSUP cows and intermediate (P ≥ 0.35) for LFAT cows. Plasma concentrations of total polyunsaturated fatty acids in HFAT cows did not differ (P ≥ 0.76) compared with LFAT cows but were greater (P ≤ 0.05) compared to NOSUP cows on day 70. Final pregnancy percentage did not differ (P ≥ 0.26) among treatments, but a greater percentage of HFAT cows calved (P ≤ 0.05) their second offspring during the first 21 d of the calving season compared to NOSUP and LFAT cows (bred by natural service). Weaning BW was the greatest (P ≤ 0.05) for LFAT and least for NOSUP calves. Maternal treatments did not impact (P ≥ 0.11) postweaning growth and total DM intake of calves. Average plasma cortisol concentrations were greater (P = 0.03) for NOSUP vs. HFAT calves and intermediate for LFAT calves (P ≥ 0.26). Serum titers against infectious bovine rhinotracheitis and bovine respiratory syncytial virus were greater or tended to be greater (P ≤ 0.08) for HFAT vs. LFAT calves and intermediate (P ≥ 0.27) for NOSUP calves at the end of preconditioning. Thus, supplemental fat concentration fed to late-gestating beef cows had variable effects on calf performance. Low-fat bakery waste led to the greatest calf preweaning growth, whereas high-fat bakery waste enhanced maternal reproduction and had minor benefits to calf humoral immune function.


This study evaluated the effect of bakery waste supplementation during the last trimester of gestation in Bos indicus-influenced beef cows and the subsequent impact on their offspring. Brangus cows were allocated to one of three prepartum treatments consisting of no prepartum supplementation, 1 kg/d of bakery waste with low or high concentration of crude fat. Prepartum supplementation of bakery waste, regardless of crude fat concentration, increased maternal plasma concentrations of ω-6 fatty acids during gestation and body condition score at calving. Offspring birth weights were not affected but offspring born to cows that received prepartum supplementation of bakery waste, regardless of crude fat concentration, were heavier at weaning compared to no prepartum supplementation. However, the greatest improvements to weaning weights were observed for offspring born to cows fed low-fat vs. high-fat bakery waste. In contrast, high-fat bakery waste supplementation during late gestation alleviated physiological stress and improved humoral immune response to vaccination during preconditioning compared to low-fat bakery waste supplementation. Thus, altering the crude fat concentration of bakery waste provided to Bos indicus-influenced beef cows during the third trimester of gestation can be implemented to modulate offspring preweaning growth or postweaning immune function.

The objectives of this study were to determine the dry matter intake (DMI), urine pH, Ca concentration in blood, Ca output in urine, and frequency of hypocalcemia in pregnant and nonlactating dairy cows consuming diets containing different hays and acidogenic products during the prepartum period. Eighty pregnant and nonlactating Holstein cows approaching their second or greater calving were fed 1 of 4 experimental diets according to a 2 × 2 factorial arrangement of treatments during the prepartum period (21 d before calving). Diets included either grass hay (GH) or alfalfa hay (AH) and calcium chloride (CL) or polyhalite (PO) as the acidogenic products. All diets had a dietary cation-anion difference (DCAD) below -190 mEq/kg of dry matter (DM). Grass hay contained 75 g/kg crude protein, 749 g/kg neutral detergent fiber, 3.6 g/kg Ca, 0.9 g/kg Na, 18.8 g/kg K, 3.8 g/kg Cl, 1.5 g/kg S, and a cation-anion difference equal to 290 mEq/kg of DM. Alfalfa hay contained 196 g/kg crude protein, 456 g/kg neutral detergent fiber, 15.2 g/kg Ca, 1.6 g/kg Na, 25 g/kg K, 7.7 g/kg Cl, 3.2 g/kg S, and a cation-anion difference equal to 292 mEq/kg of DM. Cows consuming GH tended to consume more DM than cows consuming AH (11.6 vs. 10.8 kg/d) but DMI did not differ between acidogenic products. Urine pH decreased below 6.5 for all diets, although cows consuming the GHPO diet had the highest urine pH. The concentration of Ca in plasma decreased substantially around calving but neither hay type nor acidogenic product affected it. Urinary Ca output was lowest for cows consuming the GHPO diet. No associations existed between dietary treatments and the frequencies of normocalcemia and hypocalcemia. Under the conditions of this study, in which alfalfa and grass hays had similar cation-anion differences, we concluded that the inclusion of alfalfa hay in prepartum diets does not necessarily increase the frequency of hypocalcemia. The cation-anion difference of the alfalfa hay, more than the concentration of potassium alone, may be a key determinant of whether alfalfa hay fits in a prepartum feeding program for prepartum dairy cows. Further research should explore this relationship.

This experiment investigated the effects of feeding low and high supplies of vitamin A (VA) during the transition period on plasma metabolites, prevalence of ketosis, and early milk production. In a randomized complete block design, 42 prefresh Holstein cows and 21 heifers were blocked by parity and calving date and assigned to 1 of 3 dietary treatments (n = 21 per treatment unless noted): CON, a transition diet with supplemental VA (75,000 IU/d) to meet the requirement; LVA, a transition diet with no supplemental VA; or HVA, a transition diet receiving supplemental VA (187,500 IU/d) 2.5 times greater than the requirement. Experimental periods were prepartum (-14 d prepartum), postpartum (1 to 30 d in milk), and carryover period (31 to 58 d in milk; common lactating diet with adequate VA was fed). Differences in dry matter intake in the pre- and postpartum periods and milk yield were not detected among treatment. Milk fat, protein, and lactose yields were similar among treatments and not affected by VA. Somatic cell count increased linearly with increasing VA. Body weight and body condition score decreased postpartum, but no VA effect was observed. Plasma retinol concentrations (n = 10 per treatment) decreased at d 2 postpartum and increased as lactation progressed, but the concentrations were unaffected by treatment. Plasma ß-carotene (n = 10 per treatment) had a treatment by time interaction and its concentration decreased after parturition and remained low for 2 wk. Plasma fatty acids and ß-hydroxybutyrate did not differ among treatments. Milk retinol concentration and yield (n = 10 per treatment) increased as VA supply increased. Segmented neutrophils (%) decreased, and lymphocytes (%) increased in blood with increasing VA supply. In conclusion, providing different supplies of VA did not affect production, mobilization of body fat, and risk of ketosis; however, excessive VA supply may have negatively affected the immune response, in part contributing to increased milk somatic cell counts during early lactation.

The objective of the present study was to quantify the relationships between prepartum nonesterified fatty acid (NEFA) concentrations and the development of subsequent diseases or culling and to identify the optimal thresholds allowing identification of animals at high risk of developing postpartum diseases or being culled. A total of 1,299 Holstein cows from 50 commercial herds located around Saint-Hyacinthe (QC, Canada) were enrolled in this observational study. Blood samples were collected from enrolled cows between 1 and 14 d before calving for serum NEFA quantification. Data concerning postpartum diseases and culling were collected from computerized record systems. The association between prepartum NEFA concentrations and postpartum diseases and culling was quantified using generalized linear mixed models, accounting for parity, season, week of sampling, and herd. Optimal NEFA thresholds were evaluated with receiver operator characteristic curves analysis for all diseases and then confirmed with generalized linear mixed models, considering NEFA as a categorical variable (high or low). Prepartum serum NEFA concentrations were associated with diseases diagnosed during the first 30 d in milk (DIM) and culling within the first 50 DIM. The optimal NEFA threshold associated with diseases was ≥290 µmol/L for retained placenta, ≥300 µmol/L for metritis and abomasal displacement, and ≥280 µmol/L for clinical mastitis and hyperketonemia. The level associated with the occurrence of at least one of these diseases in the first 30 DIM was ≥280 µmol/L, but it was ≥260 µmol/L for culling in the first 50 DIM. No relationship was found between NEFA concentrations and reproductive tract diseases (purulent vaginal discharge or cytological endometritis) or subclinical intramammary infection. Despite the strong relationship between prepartum NEFA concentrations and many diseases, the NEFA optimal threshold accuracy found in our study was low. In conclusion, our results demonstrate a relationship between NEFA concentrations in the 14-d period before calving and the subsequent development of diseases and culling. Prepartum NEFA concentrations thresholds between ≥260 and 300 µmol/L appear to be a strategic choice. However, considering the low accuracy, their use at the cow level should be performed with caution.

We determined the effect of prepartum plane of energy intake on liver function and metabolism pre- and postpartum by combining in vivo and in vitro data with mRNA expression data. A subset of multiparous prepartal Holsteins (n = 18) from a previously conducted experiment consumed 1 of 3 amounts of dietary energy intake, relative to their requirements. A diet formulated to allow consumption of ≥150% of net energy requirements during the far-off dry period and the close-up dry period was fed for ad libitum intake (150E) or in restricted amounts so that cows consumed 80% of requirements for energy (80E). A second diet was formulated to include wheat straw (26.1% of dry matter) to limit energy intake to 100% of NRC (2001) requirements for energy when fed ad libitum during the far-off period (100E). In the close-up period, 100E was fed the 150E diet for ad libitum intake. Expression of mRNA for genes related to fatty acid oxidation (PPARA, CPT1A, ACOX1) was greater for 100E cows than 150E cows on d 14 postpartum. These expression patterns were related to in vitro data for conversion of palmitate to CO2, acid-soluble products, and esterified products by liver slices. Abundance of mRNA for PC displayed a sharp peak for all groups on d 1 postpartum, but serum glucose did not reflect this peak. The mRNA expression of SREBF1 was greater for 150E and 100E cows prepartum compared with 80E, and was positively related to rate of palmitate esterification postpartum. Expression of NR1H3 (LXRA) mRNA was greater for 100E cows on d 14 postpartum compared with 150E cows, which corresponded to expression of PPARA. An inflammatory response occurred in the liver around the time of parturition for 150E cows, as expression of IL1B was elevated both pre- and postpartum compared with 100E cows. The spike in IL1B expression for 150E cows on d 14 postpartum corresponded to the peak concentration of total lipids in liver tissue for all groups in this experiment. Overconsumption of energy prepartum was detrimental to the expression of important genes related to PPAR and liver function, especially postpartum. Furthermore, results provide evidence for inflammation related to accumulation of lipids in liver and overnutrition prepartum.

Introduction: Negative effects of impaired postpartum mother-infant-bonding on mental health of mothers, their newborn children and subsequent child development are well documented. Previous research demonstrated an association between a negative birth experience and postpartum mental health affecting postpartum mother-infant bonding. This study investigates the extent to which prepartum depression and birth experience influence the postpartum mental health of mothers and their bonding toward their newborns, and whether these influences differ according to parity and self-reported prior mental health problems. Method: Three hundred and fifty-four women (18-43 years; M = 30.13, SD = 5.10) filled in the Edinburgh Postnatal Depression Scale (EPDS), the Maternal-Fetal Attachment Scale (MFAS), Salmon's Item List (SIL) assessing the birth experience, and the Postpartum Bonding Questionnaire (PBQ) at pre- and postpartum; they were also asked about birth complications and parity status. Results: Primipara reported significantly more birth complications (p = 0.048), with path analysis confirming this result (p < 0.001). Birth complications were associated with a more negative rating of the overall birth experience (p < 0.001). Mothers with self-reported prior mental health problems had higher prepartum depression scores (p < 0.001) but did not differ in other variables from mothers without prior self-reported mental health problems. Differences in depression scores between mothers with self-reported prior mental health problems and those without vanished at postpartum assessment (p > 0.05). Path-analysis highlighted the key role of postpartum depression, which was the only significant predictor of postpartum impairment in maternal-child bonding (p < 0.001). Birth experience and prepartum depression scores exerted an indirect effect on postpartum maternal-child bonding, mediated by postpartum depression. Discussion: The present study demonstrates the relevance of prepartum mental health of expectant mothers, especially of those who self-report prior mental health problems. The results support that reducing mental health problems of pregnant mothers might contribute to a more positive birth experience and potentially reduce postpartum depressive symptoms. As postpartum depression is associated with impaired parent-child bonding, such targeted interventions could promote child development. Group differences between primiparous and multiparous mothers suggest that the birth experience may be an influential factor for postpartum mental health.
