Background: Spontaneous remission (SR) is defined as the complete or partial disappearance of a diagnosed malignant disease in the absence of known active medical treatment. The role of the immune system is thought to be important, but has not yet been elucidated. On this matter, there are studies that suggest that the abscopal effect (AE), which is defined as the remission of untreated lesions beyond the irradiated area, may be explained by the activation of a systemic immune response against the tumor. Sclerosing epithelioid fibrosarcoma (SEF) is a rare variant of soft tissue sarcoma that is characterized by a slow evolution, with local recurrences and late metastases. The treatment is based on surgery, leaving a minimal role to chemotherapy (ChT) and radiotherapy (RT) for metastatic unresectable disease, and no cases of SR have been reported in the literature so far. Case Description: We present the case of a patient with a lung metastatic recurrence of SEF, diagnosed and treated with surgery 8 years before. After progression to pazopanib and other ChT drugs, because of the chest pain associated with a pleural mass invading the second costal arch, the patient received antalgic local RT treatment. Months later, and without any further treatment, a partial remission of all the tumoral lesions was presented, and she is alive 25 years after the first diagnosis. Conclusions: As far as reported in the literature, this is the first case of SR in SEF. Among the possible causes of this SR, we think that the most plausible is that palliative treatment with RT of the pleural mass induced an AE, leading to a reduction of all tumoral lesions, even those outside the irradiated region.

The present paper introduces an innovative strain energy function (SEF) for incompressible anisotropic fiber-reinforced materials. This SEF is specifically designed to understand the mechanical behavior of carbon fiber-woven fabric. The considered model combines polyconvex invariants forming an integrity basisin polynomial form, which is inspired by the application of Noether's theorem. A single solution can be obtained during the identification because of the relationship between the SEF we have constructed and the material parameters, which are linearly dependent. The six material parameters were precisely determined through a comparison between the closed-form solutions from our model and the corresponding tensile experimental data with different stretching ratios, with determination coefficients consistently reaching a remarkable value of 0.99. When considering only uniaxial tensile tests, our model can be simplified from a quadratic polynomial to a linear polynomial, thereby reducing the number of material parameters required from six to four, while the fidelity of the model's predictive accuracy remains unaltered. The comparison between the results of numerical calculations and experiments proves the efficiency and accuracy of the method.

Submarine mud poses a risk to channel navigation safety. Traditional detection methods lack efficiency and accuracy. As a result, this paper proposed an enhanced shallow submarine mud detection algorithm, leveraging an improved YOLOv5s model to increase accuracy and effectiveness in identifying such hazards in marine channels. Firstly, the sub-bottom profiler was employed to assess the submarine channel of Lianyungang Port to acquire the image data of the shallow mud sound print. Concurrently, the analysis incorporated the characteristics of changes in sound intensity peaks to precisely identify the shallow mud's location. Furthermore, the incorporation of C2F feature module into the backbone module enhances the gradient flow of the algorithm, augments the feature extraction information, and improves the algorithm's detection performance. Subsequently, Efficient Multi-Scale Attention (EMA) mechanism is incorporated into the neck module, aiming to optimize the algorithm's channel dimensions, minimize computational overhead, and enhance its detection efficiency. Finally, the study introduced Normalized Wasserstein Distance (NWD) loss function into bounding box regression loss function. This integration effectively addresses the issue of multi-scale defects, emphasizes the transformation of target planar position deviation, and improves the accuracy of the algorithm's detection capabilities. The results indicate that the improved YOLOv5s-EF algorithm outperforms the original YOLOv5s algorithm and other widely used detection algorithms. It achieved a validation set precision rate of 97.8%, recall rate of 97.6%, F1 value of 97.7%, mean Average Precision (mAP)@0.5 of 98.2%, mAP@0.95 of 69.6%, and Frames Per Second (FPS) of 51.8. YOLOv5s-EF algorithm proposed in this study offers a novel technical approach for detecting mud in submarine channels, which is importance for ensuring the safe operation and maintenance of dredging in such channels.

Building Information Modelling (BIM), a new concept and methodology, has received much attention lately. Various Structural Engineering Firms (SEFs) have observed substantial competitive benefits after its deployment. BIM offers a wide range of advantages, but its capacity has not even been completely exploited. The challenges of firm implementation, a procedure that necessitates significant alterations in firm business structure, are a major factor in this. However, there hasn't been much in-depth research on the assessment and integration of the research around the application of BIM in firms. To address the planning phase's complexity, which makes implementing BIM in these workplaces challenging, this article provides a framework for BIM deployment in the SEF. Additionally, a brand-new hybrid African Buffalo and African Vulture Optimization (AB-AVO) has been created to assess the state of technology. The technique suggested for BIM execution within SEF obviously and effectively recognises the firm's expectations and resources, sets out the needs required to create the BIM technique, and offers technical and clinical suggestions for monitoring and planning the execution. It is categorised by resource utilization, versatility, and adaptability.•This paper introduces a new concept and methodology of using BIM•The technique suggested for BIM execution within SEF obviously and effectively recognises the firm's expectations and resources•The method establishes guidelines for structural firm to adopt BIM in their monitoring, and planning the execution.

Valeriana jatamansi Jones is a commonly used traditional Chinese medicine, boasting rich effective compositions with versatile chemical structures and wide polarity, including iridoids, chlorogenic acid, and flavonoids. Previous reports indicate that conventional high-performance liquid chromatography (HPLC) analytical methods have proven inefficient performance in comprehensively characterizing components in Valeriana jatamansi. In the present study, a hybrid online analytical platform combining supercritical fluid extraction with both conventional HPLC separation (reverse phase) and supercritical fluid chromatography (normal phase) has been established and validated. This system can provide online extraction with two different chromatographic separation modes to increase separation ability and has been connected to a mass spectrometer to acquire high-resolution mass spectrometry data. Then, the online platform was applied to screening components in Valeriana jatamansi. A total of 117 compounds were identified, including five lignans, 18 organic acids, six flavonoids, and 88 iridoids. Thirty-three compounds were reported from Valeriana jatamansi for the first time. These results enrich our understanding of the components of Valeriana jatamansi and prove that the developed online platform in this study is a robust approach for accelerating working efficiency in comprehensively analyzing complicated samples.

Background Low-grade fibromyxoid sarcoma (LGFMS) and sclerosing epithelioid fibrosarcoma (SEF) are two rare but aggressive soft tissue sarcomas that can be difficult to distinguish due to histopathological similarities. The present study examines the diagnostic capacities of mucin-4 (MUC4), a transmembrane mucin, in identifying different types of sarcomas and broadens its evaluation to include a wide range of sarcomas. Methods Immunohistochemical (IHC) examination of tissue samples from various sarcomas was performed using a mouse anti-MUC4 monoclonal antibody. IHC was conducted on 4-mm thick formalin-fixed paraffin-embedded tissue sections after pressure cooker antigen retrieval with a mouse anti-MUC4 monoclonal antibody. Results MUC4 was shown to be highly expressed in SEF (n=13) and LGFMS (n=10), while focal positivity in synovial sarcoma (n=1). Other sarcomas, such as malignant peripheral nerve sheath tumors, fibrosarcoma, leiomyosarcoma, liposarcoma, and myxofibrosarcoma, exhibited no expression (n=0). These findings are consistent with previous research and support MUC4 specificity as a SEF and LGFMS marker. This study provides information on the diagnostic efficacy of MUC4, particularly in the context of certain subtypes. It not only helps our understanding of these unique instances, but it also provides context for histopathological and IHC findings in soft tissue sarcoma. Furthermore, this study investigates the influence of age and gender on MUC4 expression in a range of sarcomas, which was typically understudied in the literature and found no relation with expression of MUC4. Conclusion In conclusion, this study adds to our understanding of soft tissue sarcomas by emphasizing the crucial role of MUC4 in certain sarcoma subtypes while acknowledging the complex variety of the sarcoma landscape. Further research is needed to understand the molecular mechanism that governs marker expression patterns, as well as the therapeutic implications.

In early vertebrate development, organizer regions-groups of cells that signal to and thereby influence neighboring cells by secreted morphogens-play pivotal roles in the establishment and maintenance of cell identities within defined tissue territories. The midbrain-hindbrain organizer drives regionalization of neural tissue into midbrain and hindbrain territories with fibroblast growth factor 8 (FGF8) acting as a key morphogen. This organizer has been extensively studied in chicken, mouse, and zebrafish. Here, we demonstrate the enrichment of FGF8-expressing cells from human pluripotent stem cells (hPSCs), cultured as attached embryoid bodies using antibodies that recognize "Similar Expression to Fgf" (SEF) and Frizzled proteins. The arrangement of cells in embryoid body subsets of these cultures and the gene expression profile of the FGF8-expressing population show certain similarities to the midbrain-hindbrain organizer in animal models. In the embryonic chick brain, the enriched cell population induces formation of midbrain structures, consistent with FGF8-organizing capability.

With the rapid development of ultra-high voltage direct current (UHV DC) transmission technology, the intensity of electric fields in the surrounding environment of UHV DC transmission lines significantly increased, which raised public concerns about the potential health effects of electric fields. Previous studies have shown that the exposure of electromagnetic field was associated with cancer. B lymphocytes can produce autoantibodies and tumor growth factors through proliferation, which contributes to the development of cancer. Therefore, this study explored the effect and mechanism of static electric field (SEF) generated by DC transmission lines on the proliferation levels of B lymphocytes. Male mice were exposed to SEF. After the exposure of 7 and 14 days, the proliferation levels of B lymphocytes in the spleens of mice were measured, respectively. To validate biological effect discovered in animal experiments and elucidate the mechanism of the effect from the perspective of signaling pathways, lymphocytes were exposed to SEF. After the exposure of 24, 48 or 72 h, the proliferation levels of B lymphocytes, the expression levels of key proteins and cell cycle were determined. This study found that SEF exposure activated NF-κB pathway by stimulating ERK1/2 pathway and promoted B lymphocytes to enter S phase from G0/G1 phase. Meanwhile, SEF exposure also promoted B lymphocytes to enter G2 phase. Namely, SEF exposure significantly promoted the proliferation of B lymphocytes. This discovery provided theoretical and practical support for the prevention or application of negative or positive effects caused by SEF exposure and provided directions for future research.

The EFSA Panel on Food Additives and Flavourings (FAF) was requested to evaluate the safety of the smoke flavouring Primary Product Scansmoke SEF7525 (SF-004), for which a renewal application was submitted in accordance with Article 12(1) of Regulation (EC) No 2065/2003. This opinion refers to the assessment of data submitted on chemical characterisation, dietary exposure and genotoxicity of the Primary Product. Scansmoke SEF7525 is obtained from a tar produced from a mixture of red oak, white oak, maple, beech and hickory. Based on the compositional data, the Panel noted that the identified and quantified proportion of the solvent-free fraction amounts to 32.6 weight (wt)%, thus the applied method does not meet the legal quality criterion that at least 50% of the solvent-free fraction shall be identified and quantified. At the maximum proposed use levels, dietary exposure estimates calculated with Food Additive Intake Model (FAIM) ranged from 0.6 to 3.8 mg/kg body weight (bw) per day at the mean and from 1.1 to 10.1 mg/kg bw per day at the 95th percentile. Based on the available information on genotoxicity on 44 identified components, the Panel concluded that two substances in the Primary Product, styrene and benzofuran, raise a potential concern for genotoxicity. In addition, a potential concern for genotoxicity was identified for the unidentified part of the mixture. Considering that the exposure estimates for styrene and benzofuran are above the threshold of toxicological concern (TTC) value of 0.0025 kg/kg bw per day for DNA-reactive mutagens and/or carcinogens and since further data are needed to clarify their potential genotoxicity, the Panel concluded that the potential safety concern for genotoxicity of the Primary Product cannot be ruled out.

Introduction: Several studies have found changes in the organization of the primary somatosensory cortex (SI) after amputation. This SI reorganization was mainly investigated by stimulating neighboring areas to amputation. Unexpectedly, the somatosensory representation of the deafferented limb has rarely been directly tested. Methods: We stimulated the truncated peroneal nerve in 24 unilateral transtibial amputees and 15 healthy controls. The stimulation intensity was adjusted to make the elicited percept comparable between both stimulation sides. Neural sources of the somatosensory-evoked magnetic fields (SEFs) to peroneal stimulation were localized in the contralateral foot/leg areas of SI in 19 patients and 14 healthy controls. Results: We demonstrated the activation of functionally preserved cortical representations of amputated lower limbs. None of the patients reported evoked phantom limb pain (PLP) during stimulation. Stimulation that evoked perceptions in the foot required stronger intensities on the amputated side than on the intact side. In addition to this, stronger stimulation intensities were required for amputees than for healthy controls. Exploratorily, PLP intensity was neither associated with stimulation intensity nor dipole strength nor with differences in Euclidean distances (between SEF sources of the healthy peroneus and mirrored SEF sources of the truncated peroneus). Discussion: Our results provide hope that the truncated nerve may be used to establish both motor control and somatosensory feedback via the nerve trunk when a permanently functional connection between the nerve trunk and the prosthesis becomes available.

Accurate prediction of molecular properties is essential in the screening and development of drug molecules and other functional materials. Traditionally, property-specific molecular descriptors are used in machine learning models. This in turn requires the identification and development of target or problem-specific descriptors. Additionally, an increase in the prediction accuracy of the model is not always feasible from the standpoint of targeted descriptor usage. We explored the accuracy and generalizability issues using a framework of Shannon entropies, based on SMILES, SMARTS and/or InChiKey strings of respective molecules. Using various public databases of molecules, we showed that the accuracy of the prediction of machine learning models could be significantly enhanced simply by using Shannon entropy-based descriptors evaluated directly from SMILES. Analogous to partial pressures and total pressure of gases in a mixture, we used atom-wise fractional Shannon entropy in combination with total Shannon entropy from respective tokens of the string representation to model the molecule efficiently. The proposed descriptor was competitive in performance with standard descriptors such as Morgan fingerprints and SHED in regression models. Additionally, we found that either a hybrid descriptor set containing the Shannon entropy-based descriptors or an optimized, ensemble architecture of multilayer perceptrons and graph neural networks using the Shannon entropies was synergistic to improve the prediction accuracy. This simple approach of coupling the Shannon entropy framework to other standard descriptors and/or using it in ensemble models could find applications in boosting the performance of molecular property predictions in chemistry and material science.

We built a portable microchip electrophoresis heavy metal ion detection system and proposed a pH-mediated field amplified sample stacking (pH-mediated FASS) online preconcentration method. The pH-mediated FASS focuses and stacks heavy metal cations by controlling electrophoretic mobilities with a pH change between the analyte and the background electrolyte (BGE) in solution to improve the detection sensitivity of the system. We optimized and adjusted sample matrix solution (SMS) ratios and pH to create concentration and pH gradients for SMS and BGE. Furthermore, we optimize the microchannel width to improve the preconcentration effect further. The system and method analyzed soil leachates polluted with heavy metals and separated Pb2+ and Cd2+ within 90 s, obtaining their levels at 58.01 mg/L and 4.91 mg/L with sensitivity enhancement factors (SEF) of 26.40 and 43.73. Compared with inductively coupled plasma atomic emission spectrometry (ICP-AES), the detection error of the system was less than 8.80%.

The discrete steps of transcriptional rewiring have been proposed to occur neutrally to ensure steady gene expression under stabilizing selection. A conflict-free switch of a regulon between regulators may require an immediate compensatory evolution to minimize deleterious effects. Here, we perform an evolutionary repair experiment on the Lachancea kluyveri yeast sef1Δ mutant using a suppressor development strategy. Complete loss of SEF1 forces cells to initiate a compensatory process for the pleiotropic defects arising from misexpression of TCA cycle genes. Using different selective conditions, we identify two adaptive loss-of-function mutations of IRA1 and AZF1. Subsequent analyses show that Azf1 is a weak transcriptional activator regulated by the Ras1-PKA pathway. Azf1 loss-of-function triggers extensive gene expression changes responsible for compensatory, beneficial, and trade-off phenotypes. The trade-offs can be alleviated by higher cell density. Our results not only indicate that secondary transcriptional perturbation provides rapid and adaptive mechanisms potentially stabilizing the initial stage of transcriptional rewiring but also suggest how genetic polymorphisms of pleiotropic mutations could be maintained in the population.

The present study investigated the effects of hypocapnia and hypercapnia on human somatosensory processing by utilizing somatosensory evoked magnetic fields (SEFs) with magnetoencephalography (MEG). Thirteen volunteers participated in two experiments separately to measure respiratory and cardiovascular data and SEFs. Both experiments consisted of a combination of normal and rapid respiratory rhythms and two inspiratory gas conditions (air and a hypercapnic gas); normal breathing with air (NB), rapid breathing with air (RB), normal breathing with the hypercapnic gas (NB+Gas), and rapid breathing with gas (RB+Gas). Partial pressures of end-tidal CO2 (PETCO2) increased during inhaling the hypercapnic gas and decreased during RB, but the RB+Gas condition continued to cause elevated PETCO2 compared with the baseline. Subsequently, middle cerebral artery blood (MCA) velocity using transcranial Doppler changed as well, while mean MCA velocity increased under the RB+Gas condition. The peak amplitude of the M60 component in SEFs was also significantly larger under with-gas than without-gas conditions, irrespective of the respiratory frequency. These results suggest that there is a close relationship between cerebral blood flow and neural activity of the M60 component in SEFs. This study provides evidence to further understanding on one of the neural mechanisms of hypercapnia.

Gold nanoparticles (AuNPs) are attractive structures for biosensing, most due to different properties at nanoscale and biocompatibility. Localized surface plasmon resonance (LSPR) is one of these properties; LSPR enable the electromagnetic field enhancement closer to metallic surface, which allows surface-enhanced spectroscopies, like surface enhanced fluorescence (SEF). In this study, an immuno-biosensor based on gold nanorods (AuNRs) and SEF was constructed for simple and fast analysis to detect albumin antibody (anti-BSA) using antigen-antibody (anti-BSA/BSA) interaction as the biorecognition model. AuNRs were presented in two distinct configurations, in suspension (S-AuNRs) and adsorbed on glass slides (AuNRs-chip), and the detection was performed through an extrinsic method, wherein the SEF signal of a reporter molecule (IR-820 cyanine-type dye) was monitored. The analyte detection was evidenced by SEF mapping, where the average signal in the presence of anti-BSA was three times more intense than for the assay in the absence of analyte. A digital protocol was proposed to simplify the spectroscopic data analysis and reduce the intensity variability; in this protocol the number of positive events in the presence of anti-BSA is much larger (around two times) compared to the absence of analyte. The AuNRs based SEF immuno-biosensor allowed an efficient and simple analysis with specific biorecognition and may contribute as an efficient spectroscopy platform for immuno-biosensing.

Background: Sclerosing epithelioid fibrosarcoma (SEF) is an extremely rare subtype of soft tissue sarcoma and the data from India is sparse. It is an unusual variant of fibrosarcoma that commonly arises in the soft tissues of the limb, head and neck, trunk and occasionally in the visceral organs and bones. This entity is commonly reported in the middle age group, men and women alike. Pathological clinchers include MUC 4 (Mucin 4, cell surface associated) positivity by immunohistochemistry, FUS-CREB3L1 fusion and EWSR1 rearrangement. This disease is notoriously known for its local recurrence and metastatic spread. Response to systemic therapy is poor and relapses are frequent. The role of targeted and immunotherapy is not well defined. Case presentation: Here we report a 46-year-old gentleman who presented to the Sarcoma Medical Oncology Clinic in our centre. He had primary involvement of right pubic bone with metastasis to liver, lung and diffuse lytic bony lesions. His diagnosis was reviewed multiple times before coming to final diagnosis of SEF. His molecular test for EWSR1 rearrangement was positive by fluorescence in-situ hybridisation. He did not respond to palliative doxorubicin, pazopanib and gemcitabine and docetaxel. Conclusion: Through this case report, we would like to highlight the rarity of this sarcoma, its classical pathological features, its close relationship to low-grade fibromyxoid sarcoma and the limited therapeutic options available. Hence, there is a need for further research in this entity.

Background: Emergence delirium (ED) usually occurs in children after surgery with an incidence of 10−80%. Though ED is mostly self-limited, its potential injuries cannot be ignored. Whether electroencephalography (EEG)-parameter-guided anesthesia could reduce the incidence of ED in pediatric surgery has not been fully discussed to date. Methods: Fifty-four boys aged 2−12 years undergoing elective hypospadias surgery under sevoflurane anesthesia were selected. In the EEG-parameter-guided group (E group), sevoflurane was used for anesthesia induction and was maintained by titrating the spectral edge frequency (SEF) to 10−15 and combining the monitoring of density spectral array (DSA) power spectra and raw EEG. While in the control group (C group), anesthesiologists were blinded to the SedLine screen (including SEF, DSA, and raw EEG) and adjusted the intraoperative drug usage according to their experience. Patients with a Pediatric Anesthesia Emergence Delirium (PAED) score > 10 were diagnosed with ED, while patients with a PAED score > 2 were diagnosed with emergence agitation (EA). Results: Finally, a total of 37 patients were included in this trial. The incidence of ED in the E group was lower than in the C group (5.6% vs. 36.8%; p = 0.04), while the incidence of EA was similar in the two groups (61% vs. 78.9%; p = 0.48). Intraoperative parameters including remifentanil dosage and the decrease in mean arterial pressure (MAP) were not different between the two groups (p > 0.05), but the mean end-tidal sevoflurane concentration (EtSevo) was lower in the E group than in the C group (p > 0.05). Moreover, during PACU stay, the extubation time and discharge time of the groups were similar, while the PAED scores within 5 min from extubation and the Face, Legs, Activity, Cry, and Consolability (FLACC) scores within 30 min from extubation were lower in the E group than in the C group. Conclusion: EEG-parameter-guided anesthesia management reduced the incidence of ED in children. Studies with larger sample sizes are needed to obtain more convincing results.

This study shows a new SERS (Surface-enhanced Raman Scattering) and SEF (Surface-enhanced Fluorescence) platform approach, in which substrates were constructed from the silver nanoparticles stabilized by alginate polymer (AgALG) and encapsulated in hydrogel calcium alginate beads (AgALGbead). In this regard, the electrostatic repulsion or attraction concerning the charged dyes and the carboxylate groups of the alginate could define the distances between the probe molecules and metallic nanoparticles to determine the SERS or SEF effect. In this sense, the anionic dye named New Indocyanine Green (IR-820) and the cationic dye Rhodamine 6G (Rh6G) were selected to discuss the alginate's ability to quench or enhance the fluorescence and the Raman dyes signals. Furthermore, the SEF effect using the IR-820 dye can be detected for the near-infrared emission (S1 â†’ S0) using the 532 and 633 nm laser lines as well at the visible region (S2 â†’ S0) applying the excitation at 532 nm in the AgALGbead substrates. Nevertheless, the cationic dye provides the Surface-enhanced Resonance Raman Scattering (SERRS) effect and quenching of the fluorescence for the same AgALGbeads substrate at 532 nm laser line.

The preparation of metal@(dielectric or semiconductor) core@shell hybrid materials have been shown promising for both SERS and SEF applications due to improved stability in the presence of ions and the adsorbate compared to non-covered metallic nanoparticles. However, fine control over the thickness of the covering layer is essential to maximize the intrinsic trade-off between the plasmonic enhancement and the chemical stability improvement. Here, the preparation of manganese dioxide ultrathin layers covered gold nanorods (AuNR@MnO2) with varying thicknesses of the MnO2 layer is reported, and the characterization and evaluation of the resulting materials as SERS and SEF substrate. The MnO2 layer over the AuNR was prepared by reducing potassium permanganate by sodium oxalate in a basic medium. The AuNR@MnO2 hybrid material was characterized by UV-Vis spectroscopy, transmission electron microscopy, X-ray powder diffraction, and cyclic voltammetry. It was studied the SEF effect of the cyanine dye IR-820 excited at 785 nm with high performance for several thicknesses of the MnO2 ultrathin film. The enhancement factor increased for thicker oxide layers. The SERS effect of the IR-820 dye excited at 633 nm showed the most significant enhancement factor for thinner layers. The seemly opposite behavior of the two plasmonic effects may be assigned to the distance dependence of the electromagnetic field generated in the AuNR, which results in decreasing SERS performance. For SEF, the thinner layers resulted in the Au nanoparticles' emission quenching, so a more significant distance was necessary to observe enhancement.

Candida albicans filamentation, the ability to convert from oval yeast cells to elongated hyphal cells, is a key factor in its pathogenesis. Previous work has shown that the integral membrane protein Dfi1 is required for filamentation in cells grown in contact with a semisolid surface. Investigations into the downstream targets of the Dfi1 pathway revealed potential links to two transcription factors, Sef1 and Czf1. Sef1 regulates iron uptake and iron utilization genes under low-iron conditions, leading us to hypothesize that there exists a link between iron availability and contact-dependent invasive filamentation. In this study, we showed that Sef1 was not required for contact-dependent filamentation, but it was required for wild-type (WT) expression levels of a number of genes during growth under contact conditions. Czf1 is required for contact-dependent filamentation and for WT levels of expression of several genes. Constitutive expression and activation of either Sef1 or Czf1 individually in a dfi1 null strain resulted in a complete rescue of the dfi1 null filamentation defect. Because Sef1 is normally activated in low-iron environments, we embedded WT and dfi1 null cells in iron-free agar medium supplemented with various concentrations of ferrous ammonium sulfate (FAS). dfi1 null cells embedded in media with a low concentration of iron (20 µM FAS) showed increased filamentation in comparison to mutant cells embedded in higher concentrations of iron (50 to 500 µM). WT cells produced filamentous colonies in all concentrations. Together, the data indicate that Dfi1, Czf1, Sef1, and environmental iron regulate C. albicans contact-dependent filamentation. IMPORTANCE Candida albicans is an opportunistic pathogen responsible for a larger proportion of candidiasis and candidemia cases than any other Candida species. The ability of C. albicans cells to invade and cause disease is linked to their ability to filament. Despite this, there are gaps in our knowledge of the environmental cues and intracellular signaling that triggers the switch from commensal organism to filamentous pathogen. In this study, we identified a link between contact-dependent filamentation and iron availability. Over the course of tissue invasion, C. albicans cells encounter a number of different iron microenvironments, from the iron-rich gut to iron-poor tissues. Increased expression of Sef1-dependent iron uptake genes as a result of contact-dependent signaling will promote the adaptation of C. albicans cells to a low-iron-availability environment.
