RESUMO
Drought and cold stresses co-occur in nature, and significantly limited agricultural productivity in northwest China. Their effects on plant photosynthesis, physiology and biochemistry, and microstructure have been extensively studied, but their combined stress mechanisms remain unclear. Therefore, growth chamber experiments were conducted using the Annual bluegrass (Poa annua) with the 'HZ' and 'ZQ' germplasms, our goal was to clarify the responses of leaves to cold (C), drought (D), and combined (D&C) stresses in the morphology, photosynthesis, physiology and biochemistry, and microstructure. The results showed that three stress types significantly suppressed growth, reduced photosynthetic pigments, photosynthetic capacity, and photosynthetic enzyme activities, with 'ZQ' being more sensitive than 'HZ'. C stress (12/36 h) improved chlorophyll fluorescence parameters in 'HZ' (except for photochemical quenching coefficient), while D and D&C stresses diminished these parameters of both germplasms. Under single and combined stresses, leaf thickness and cuticle thickness generally increased then decreased with stress duration, except for 'ZQ' under D stress. D stress increased reactive oxygen species, relative conductivity, malondialdehyde content and stomatal density, while inhibited stomatal size. Conversely, C stress exacerbated the negative effects of drought on these traits. The D&C stress enhanced antioxidant enzyme activity and proline content, which were similar to the responses to D stress, except for 'ZQ' under D&C stress. After 24 h of stress relief, the recoveries degree of most traits in plant under D and D&C stresses were similar, with 'HZ' recovering more than 'ZQ'. This study indicated drought played a dominant role in combined stress. However, some unique responses could not be inferred from the superimposed effects of some single stresses.
RESUMO
Copper (Cu) is recognized as an essential trace elements for the body; However, excessive levels of Cu can lead to toxic effects. We investigated the effects of Cu2+(75⯵g/L, 150⯵g/L, and 300⯵g/L) on the rainbow trout liver. Combination of transcriptome and metabolome analyses, the regulatory mechanisms of the liver under Cu stress were elucidated. The results showed that Cu affected the antioxidant levels, leading to disruptions in the normal tissue structure of the liver. Combined transcriptome and metabolome analyses revealed significant enrichment of the insulin signaling pathway and the adipocytokine signaling pathway. Additionally, Cu2+ stress altered the amino acid metabolism in rainbow trout by reducing serine and arginine levels while increasing proline content. Apoptosis is inhibited and autophagy and lipid metabolism are suppressed; In summary, Cu2+ stress affects energy and lipid metabolism, and the reduction of serine and arginine represents a decrease in the antioxidant capacity, whereas the increase in proline and the promotion of apoptosis potentially serving as crucial strategies for Cu2+ resistance in rainbow trout. These findings provided insights into the regulatory mechanisms of rainbow trout under Cu2+ stress and informed the prevention of heavy metal pollution and the selection of biomarkers under Cu pollution.

Passiflora edulis Sims (2n = 18) is a perennial plant with high utilization values, but its spontaneous polyploidy in nature has yet to be seen. Thus, this study aims to enhance our understanding of polyploidy P. edulis and provide rudimentary knowledge for breeding new cultivars. In this study, colchicine-induced tetraploid P. edulis (2n = 36) was used as experimental material (T1, T2, and T3) to explore the variances between it and its diploid counterpart in morphology, physiology, and biochemical characteristics, and a comparison of their performance under cold stress was conducted. We measured and collected data on phenotype parameters, chlorophyll contents, chlorophyll fluorescence, photosynthesis, osmotic substances, and antioxidant enzymes. The results showed that tetraploid P. edulis exhibited a shorter phenotype, more giant leaves, darker leaf color, and longer and fewer roots. Moreover, the physiological and biochemical analysis indicated that the tetraploid P. edulis had better photosynthesis systems and higher chlorophyll fluorescence parameters than the diploid P. edulis. Additionally, the tetraploid P. edulis had higher activity of antioxidant enzymes (SOD, POD, CAT) and lower MDA content to maintain better resistance in low temperatures. Overall, we conclude that there were apparent differences in the morphological, physiological, and biochemical traits of the tetraploid and diploid P. edulis. The tetraploid plants showed better photosynthesis systems, higher osmotic substance content, and antioxidant enzyme activity than the diploid, even under cold stress. Our results suggest that tetraploids with more abundant phenotype variation and better physiological and biochemical traits may be used as a new genetic germplasm resource for producing new P. edulis cultivars.
RESUMO
Tartary buckwheat (Fagopyrum tataricum (L.) Gaertn) is a crop of significant interest due to its nutritional value and resilience to drought conditions. However, drought, particularly following flowering, is a major factor contributing to yield reduction. This research employed two distinct Tartary buckwheat genotypes to investigate the effects of post-anthesis drought on growth and physicochemical characteristics. The study aimed to elucidate the response of Tartary buckwheat to drought stress. The findings indicated that post-anthesis drought adversely impacted the growth, morphology, and biomass accumulation of Tartary buckwheat. Drought stress enhanced the maximum photosynthetic capacity (Fv/Fm) and light protection ability (NPQ) of the 'Xiqiao-2' genotype. In response to drought stress, 'Dingku-1' and 'Xiqiao-2' maintained osmotic balance by accumulating soluble sugars and proline, respectively. Notably, 'Xiqiao-2' exhibited elevated levels of flavonoids and polyphenols in its leaves, which helped mitigate oxidative damage caused by drought. Furthermore, rewatering after a brief drought period significantly improved plant height, stem diameter, and biomass accumulation in 'Dingku-1'. Overall, 'Xiqiao-2' demonstrated greater long-term tolerance to post-anthesis drought, while 'Dingku-1' was less adversely affected by short-term post-anthesis drought.
RESUMO
Coix lacryma-jobi L. is a traditional medicinal plant in east Asia and is an important crop in Guizhou province, southwest China, where there are elevated levels of soil mercury and arsenic (As). Exposure to multiple potentially toxic elements (PTEs) may affect plant accumulation of metal(loid)s and food safety in regions with high geological metal concentrations. Field experiments were conducted to study the effects of PTEs on metal(loid) accumulation and physiological response of C. lacryma in different plant parts at three pollution levels. Total root length, number of root tips, number of branches, and number of root crosses increased with increasing pollution level, with increases in highly polluted areas of 44.2, 57.0, 79.6, and 97.2%, respectively, compared to lightly polluted areas. Under multi-element stress the activity of C. lacryma antioxidant oxidase showed an increase at low and medium PTE concentrations and inhibition at high concentrations. The As contents were all below the maximum limit of cereal food contaminants in China (GB 2762-2022, As < 0.5 mg kg-1). The stems had high Tl bioconcentration factors but the translocation factors from stem to grain were very low, indicating that the stems may be a key plant part restricting Tl transport to the grains. C. lacryma increased root retention and reduced the transport effect, thus reducing metal accumulation in the grains. C. lacryma adapted to PTE stress through root remodeling and enhanced antioxidant enzyme activities.

To delve into the growth and physiological adaptations exhibited by the economically vital black wolfberry (Lycium ruthenicum) upon inoculation with arbuscular mycorrhizal fungi (AMF) under varying levels of saline-alkaline stress A series of pot experiments were conducted in a gradient saline-alkaline environment (0, 200, 400 mM NaCl: NaHCO3 = 1:1). One-year-old cuttings of black wolfberry, inoculated with two AMF species-Funneliformis mosseae (Fm) and Rhizophagus intraradices (Ri)-served as the experimental material, enabling a comprehensive analysis of seedling biomass, chlorophyll content, antioxidant enzyme activities, and other crucial physiological parameters. This study demonstrated that both Fm and Ri could form a symbiotic relationship with the root of Lycium ruthenicum. Notably, Fm inoculation significantly bolstered the growth of the underground parts, while exhibiting a remarkable capacity to scavenge reactive oxygen species (ROS), thereby effectively mitigating membrane oxidative damage induced by stress. Additionally, Fm promoted the accumulation of abscisic acid (ABA) in both leaves and roots, facilitating the exclusion of excess sodium ions from cells. Ri Inoculation primarily contributed to an enhancement in the chlorophyll b (Chlb) content, vital for sustaining photosynthesis processes. Furthermore, Ri's ability to enhance phosphorus (P) absorption under stressful conditions ensured a steady influx of essential nutrients. These findings point to different strategies employed for Fm and Ri inoculation. To holistically assess the saline-alkaline tolerance of each treatment group, a membership function analysis was employed, ultimately ranking the salt tolerance as Fm > Ri > non-mycorrhizal (NM) control. This finding holds paramount importance for the screening of highly resilient Lycium ruthenicum strains and offers invaluable theoretical underpinnings and technical guidance for the remediation of saline-alkaline soils, fostering sustainable agricultural practices in challenging environments.
RESUMO
As one of the most influential environmental factors, drought stress greatly impacts the development and production of plants. Triploid-induced Passiflora edulis Sims 'Mantianxing' is an important new cultivar for multi-resistance variety selective breeding, which is one of the P. edulis breeding essential targets. However, the performance of triploid 'Mantianxing' under drought stress is unknown. In order to study the drought resistance of triploid 'Mantianxing', our study compared drought-related indicators in diploids and triploids under natural drought experiments, including morphological, physiological, and biochemical characteristics. Results showed that triploid P. edulis 'Mantianxing' showed variable responses to drought treatment. Compared with diploids, triploids showed higher photosynthesis and chlorophyll fluorescence, osmotic adjustment substances, and antioxidant enzyme activity under drought stress and faster chlorophyll biosynthesis and growth recovery after rewatering. Generally speaking, these results indicate that the drought resistance of triploid P. edulis is superior to diploid. This study provides scientific information for breeding stress tolerance variety of P. edulis 'Mantianxing' new cultivar.
RESUMO
Plant-specific VQ proteins have crucial functions in the regulation of plant growth and development, as well as in plant abiotic stress responses. Their roles have been well established in the model plant Arabidopsis thaliana; however, the functions of the potato VQ proteins have not been adequately investigated. The VQ protein core region contains a short FxxhVQxhTG amino acid motif sequence. In this study, the VQ31 protein from potato was cloned and functionally characterized. The complete open reading frame (ORF) size of StVQ31 is 672 bp, encoding 223 amino acids. Subcellular localization analysis revealed that StVQ31 is located in the nucleus. Transgenic Arabidopsis plants overexpressing StVQ31 exhibited enhanced salt tolerance compared to wild-type (WT) plants, as evidenced by increased root length, germination rate, and chlorophyll content under salinity stress. The increased tolerance of transgenic plants was associated with increased osmotic potential (proline and soluble sugars), decreased MDA accumulation, decreased total protein content, and improved membrane integrity. These results implied that StVQ31 overexpression enhanced the osmotic potential of the plants to maintain normal cell growth. Compared to the WT, the transgenic plants exhibited a notable increase in antioxidant enzyme activities, reducing cell membrane damage. Furthermore, the real-time fluorescence quantitative PCR analysis demonstrated that StVQ31 regulated the expression of genes associated with the response to salt stress, including ERD, LEA4-5, At2g38905, and AtNCED3. These findings suggest that StVQ31 significantly impacts osmotic and antioxidant cellular homeostasis, thereby enhancing salt tolerance.
RESUMO
Saline and alkaline stress is one of the major abiotic stresses facing agricultural production, which severely inhibits the growth and yield of plant. The application of plant growth regulators can effectively prevent crop yield reduction caused by saline and alkaline stress. Exogenous melatonin (MT) can act as a signaling molecule involved in the regulation of a variety of physiological processes in plants, has been found to play a key role in enhancing the improvement of plant tolerance to abiotic stresses. However, the effects of exogenous MT on saline and alkaline tolerance of table grape seedlings and its mechanism have not been clarified. The aim of this study was to investigate the role of exogenous MT on morphological and physiological growth of table grape seedlings (Vitis vinifera L.) under saline and alkaline stress. The results showed that saline and alkaline stress resulted in yellowing and wilting of grape leaves and a decrease in chlorophyll content, whereas the application of exogenous MT alleviated the degradation of chlorophyll in grape seedling leaves caused by saline and alkaline stress and promoted the accumulation of soluble sugars and proline content. In addition, exogenous MT increased the activity of antioxidant enzymes, which resulted in the scavenging of reactive oxygen species (ROS) generated by saline and alkaline stress. In conclusion, exogenous MT was involved in the tolerance of grape seedlings to saline and alkaline stress, and enhanced the saline and alkaline resistance of grape seedlings to promote the growth and development of the grape industry in saline and alkaline areas.

Fecal scores are crucial for assessing the digestive and gastrointestinal status of animals. The Bristol fecal scoring system is a commonly used method for the subjective evaluation of host feces, there is limited research on fecal scoring standards for fattening Hu sheep. In this study, Hu sheep were collected for rumen, rectum, and colon contents for 16S rDNA sequencing. 514 Hu sheep feces were scored based on the Bristol fecal scoring system, and production performance at each stage was measured. Finally, we developed the scoring standard of the manure of Hu sheep in the fattening period (a total of five grades). The result shows that moisture content significantly increased with higher grades (p < 0.05). We analyzed the relationship between fecal scores and production traits, blood indices, muscle nutrients, and digestive tract microorganisms. The growth traits (body weight, body height, body length, average daily gain (ADG), and average daily feed intake (ADFI) during 80-180 days), body composition traits of the F3 group, and the carcass traits were found to be significantly higher (p < 0.05) than those of the F1 and F2 groups. There was no significant difference in gastrointestinal microflora diversity among all groups (p > 0.05). Significant differences were observed in Aspartate aminotransferase, Glucose, Total bilirubin, and Red Blood Cell Count between groups (p < 0.05). The mutton moisture content in group F4 was significantly higher than in the other groups, and the protein content was also the lowest (p < 0.05). The results of the correlation analysis demonstrated that Actinobacteria, Peptostreptococcaceae, Acidaminococcales, Gammaproteobacteria, and Proteobacteria were the significant bacteria affecting fecal scores. In addition, Muribaculaceae and Oscillospiraceae were identified as the noteworthy flora affecting growth performance and immunity. This study highlights the differences in production traits and blood indicators between fecal assessment groups and the complex relationship between intestinal microbiota and fecal characteristics in Hu sheep, suggesting potential impacts on animal performance and health, which suggest strategies for improved management.
RESUMO
High-temperature stress is the main environmental stress that restricts the growth and development of woody plants, and the growth and development of woody plants are affected by high-temperature stress. The influence of high temperature on woody plants varies with the degree and duration of the high temperature and the species of woody plants. Woody plants have the mechanism of adapting to high temperature, and the mechanism for activating tolerance in woody plants mainly counteracts the biochemical and physiological changes induced by stress by regulating osmotic adjustment substances, antioxidant enzyme activities and transcription control factors. Under high-temperature stress, woody plants ability to perceive high-temperature stimuli and initiate the appropriate physiological, biochemical and genomic changes is the key to determining the survival of woody plants. The gene expression induced by high-temperature stress also greatly improves tolerance. Changes in the morphological structure, physiology, biochemistry and genomics of woody plants are usually used as indicators of high-temperature tolerance. In this paper, the effects of high-temperature stress on seed germination, plant morphology and anatomical structure characteristics, physiological and biochemical indicators, genomics and other aspects of woody plants are reviewed, which provides a reference for the study of the heat-tolerance mechanism of woody plants.
RESUMO
The effects of different concentrations (100, 150, 200, 250 mg/L) and different particle sizes (0-75 µm, 75-120 µm, 120-150 µm, 150-500 µm) on the soluble protein content, superoxide dismutase (SOD) and catalase (CAT) activity, malondialdehyde (MDA) content, chlorophyll a (Chla) content, and photosynthetic parameters of Microcystis flos-aquae were studied, and the mechanism of the effect of suspended particulate matter on the physiology and biochemistry of Microcystis flos-aquae was discussed. The results showed that the soluble protein content of Microcystis flos-aquae did not change noticeably after being stressed by suspended particles of different concentrations/diameters. The SOD activity of Microcystis flos-aquae first increased and then decreased with increasing suspended particulate matter concentrations. The SOD activity of Microcystis flos-aquae reached 28.03 U/mL when the concentration of suspended particulate matter was 100 mg/L. The CAT activity of Microcystis flos-aquae increased with increasing concentrations of suspended particles and reached a maximum value of 12.45 U/mg prot in the 250 mg/L concentration group, showing a certain dose effect. Small particles had a more significant effect on SOD, CAT, and MDA in Microcystis flos-aquae than large particles. The larger the concentration was and the smaller the particle size was, the stronger the attenuation of light and the lower the content of Chla. Both the maximum quantum yield of PSII (Fv/Fm) and the potential photosynthetic activity of PSII (Fv/F0) of Microcystis flos-aquae increased at first and then decreased under different concentrations/sizes of suspended particles. The relative electron transfer rate gradually returned to a normal level over time. There was no significant difference in the initial slope (α) value between the treatment group and the control group, and the maximum photo synthetic rate (ETRmax) and the semilight saturation (Ik) decreased.

Background: To examine the effects of each dose of decoction of Angelica sinensis (Dang gui), Zingiberis Rhizoma Recens (Sheng jiang), and mutton (DAZM) on the physiological and biochemical indexes of female rats with spleen-kidney Yang deficiency (SKYD) through 30-day feeding of DAZM, and to evaluate the tonifying effect of DAZM combined with the system of benefit damage index-general score (BDI-GS). Methods: Sprague Dawley (SD) rats were administered adenine and senna water to establish a SKYD model. The rats were then allocated to 4 groups at random: Model group, and L group, 4.2 g/kg, M group, 8.4 g/kg and H group, 16.8 g/kg. In addition, the group of normal feeding with unlimited diet was set as N group. Blood samples were taken to detect the relevant physiological and biochemical indexes. For organ coefficient analysis, 10 kinds of organ tissues were dissected and weighed. The tonifying effect of DAZM was discussed according to the BDI-GS system. Results: During the modeling, the weight of rats in the normal group displayed a marked growth trend, and the weight of the model group was markedly lower than that of the normal group. After feeding the rats DAZM at a low, intermediate, and high dose, the anal temperature of rats in each group continued to rise, and finally remained basically the same as that of normal rats. Hematological and urine examinations revealed that the urea nitrogen and creatinine (CRE) of the model group and the experimental group were markedly higher than those of the normal group, and there were marked differences. After intragastric administration of DAZM, E2 increased markedly. The BDI-GS values of the liver, spleen, lung, kidney, brain, ovary, and adrenal gland of female rats in the 3 administration groups of DAZM were >1, and the total cumulative GS value of each organ of female rats was more than 10. Conclusions: The decoction of DAZM has no obvious effect on the growth, metabolism, and development of female rats with SKYD, causes no obvious damage to organs, and has a certain reparative effect on the kidney damage caused by SKYD.
RESUMO
Cadmium contamination in agricultural soils threatens food security and human health, and that has caused widespread concern worldwide. Willow and alfalfa are widely used for the phytoremediation of cadmium (Cd)-contaminated soil, and willow NJU513 is the promising plant for remediating Cd-contaminated soil. In order to discuss the effect of intercropping willow NJU513 with alfalfa on the phytoremediation of Cd-contaminated soil, a pot-culture experiment was conducted in the greenhouse. The result showed that the phytoremediation of Cd-contaminated soil was enhanced by this intercropping because of the 25.90 % increase in the available Cd content. In order to increase the phytoremediation efficiency of Cd in the intercropping treatment, a 24-epibrassinolide (Brs) treatment was designed in the current study. The results showed that the phytoremediation of Cd-contaminated soil by willow and alfalfa improved following a Brs treatment because of the 16.32-74.15 % and 16.91-44.48 % increases in the plant biomass and available Cd content, respectively. Additionally, the extracted Cd by plants in the intercropping treatments with and without Brs was 0.56 and 0.31 mg pot-1, respectively. Transcriptome analyses of willow leaves revealed that Brs up-regulated the expression of genes related to calcium channel activity, calcium and zinc transmembrane transport, photosynthesis, catalase/antioxidant activity, glutathione metabolic processes and detoxification, phagosomes, and vacuoles, and that these upregulated genes promoted plant remediation efficiency and resistance to Cd stress. Brs promoted the phosphate ion transporter activity in willow leaves, which may have enhanced the solubilization of insoluble phosphate minerals by bacterial species (e.g., Vicinamibacterales, Bacillus, and Gaiella) to release Cd, ultimately leading to increased phytoremediation efficiency. In addition, plants with and without Brs treatments induced the bacteria-mediated transformation of available Cd to stable Cd. The study findings may be useful for improving the phytoremediation of Cd-contaminated paddy soil.

In recent years, heavy metals-induced soil pollution has increased due to the widespread usage of chromium (Cr) in chemical industries. The release of Cr into the environment has reached its peak causing hazardous environmental pollution. Heavy metal-induced soil pollution is one of the most important abiotic stress affecting the dynamic stages of plant growth and development. In severe cases, it can kill the plants and their derivatives and thereby pose a potential threat to human food safety. The chromium ion effect on plants varies and depends upon its severity range. It mainly impacts the numerous regular activities of the plant's life cycle, by hindering the germination of plant seeds, inhibiting the growth of hypocotyl and epicotyl parts of the plants, as well as damaging the chloroplast cell structures. In this review article, we tried to summarize the possible effects of chromium-induced stress on plant growth, developmental physiology, biochemistry, and molecular regulation and provided the important theoretical basis for selecting remedial plants in chromium-induced contaminated soils, breeding of low toxicity tolerant varieties, and analyzing the mechanism of plant resistance mechanisms in response to heavy metal stress.
RESUMO
Pinus massoniana Lamb. is the timber species with the widest distribution and the largest afforestation area in China, providing a large amount of timber, turpentine and ecological products. but low temperature limits its growth and geographical distribution. Physiological and molecular studies can well explain the mechanism of P. massoniana response to low temperature. In this study, physiological and biochemical indexes, cell morphology, lignin content, gene regulatory networks, and gene expression patterns of different P. massoniana varieties (cold-tolerant and cold-sensitive) were studied from physiological, biochemical, and molecular perspectives. The results indicated that under low-temperature stress, the cold-tolerant cultivar maintained high contents of osmoregulatory substances, and the root morphology and structure remained intact. In the initial stage of low-temperature stress, the number of differentially expressed genes was 7148, and with the extension of stress time, the number of differentially expressed genes decreased to 1991. P. massoniana might direct its responses to low temperature by regulating phenylpropane metabolism, starch and sucrose metabolism, hormone signaling pathways, and transcription factors. BAM, 4CL, CCoAOMT, PRX5, WRKYs, and hormone synthesis related genes play important roles. P. massoniana cultivars may vary in response mechanisms. In this study, physiological and analytical techniques were used to study the root tip response mechanism of Masson's pine to low temperature stress. The results of this study lay a foundation for in-depth research on the molecular functions of P. massoniana under low-temperature stress conditions.
RESUMO
We aimed to elucidate the physiological and biochemical mechanism by which exogenous hydrogen peroxide (H2O2) alleviates salt stress toxicity in Tartary buckwheat (Fagopyrum tataricum (L.) Gaertn). Tartary buckwheat "Chuanqiao-2" under 150 mmol·L-1 salt (NaCl) stress was treated with 5 or 10 mmol·L-1 H2O2, and seedling growth, physiology and biochemistry, and related gene expression were studied. Treatment with 5 mmol·L-1 H2O2 significantly increased plant height (PH), fresh and dry weights of shoots (SFWs/SDWs) and roots (RFWs/RDWs), leaf length (LL) and area (LA), and relative water content (LRWC); increased chlorophyll a (Chl a) and b (Chl b) contents; improved fluorescence parameters; enhanced antioxidant enzyme activity and content; and reduced malondialdehyde (MDA) content. Expressions of all stress-related and enzyme-related genes were up-regulated. The F3'H gene (flavonoid synthesis pathway) exhibited similar up-regulation under 10 mmol·L-1 H2O2 treatment. Correlation and principal component analyses showed that 5 mmol·L-1 H2O2 could significantly alleviate the toxic effect of salt stress on Tartary buckwheat. Our results show that exogenous 5 mmol·L-1 H2O2 can alleviate the inhibitory or toxic effects of 150 mmol·L-1 NaCl stress on Tartary buckwheat by promoting growth, enhancing photosynthesis, improving enzymatic reactions, reducing membrane lipid peroxidation, and inducing the expression of related genes.

Walnut is one of the world's four largest nuts. Currently, the bottleneck in walnut breeding is the production of resistant variants. Soil salinization is a global problem, and the use of salt-tolerant rootstocks is a basic strategy to overcome the challenge of sustained walnut production. Providing a scientific basis for the selection of walnut salt-tolerant rootstocks is possible by studying the physiological and biochemical response characteristics and salt tolerance variations of different walnut genotypes under salt stress. In the present study, seedlings of four genotypes of walnut rootstocks, including J1 (Juglans hindsii), J2 (J. mandshurica), J3 (J. regia × J. mandshurica), and J4 (J. regia × J. hindsii), were employed as test materials to conduct a 28-day pot experiment under NaCl stress with five NaCl concentrations (0, 50, 100, 200, and 300 mmol/L). Under different NaCl treatment concentrations, seedling morphology, growth indices, chlorophyll content, photosynthetic parameters, relative electrical conductivity (REC), malondialdehyde (MDA), proline (Pro), soluble sugar (SS), and the activity of superoxide dismutase (SOD) and peroxidase (POD) in the leaves were examined. Salt stress altered the morphological characteristics and growth indices of seedlings from four genotypes to varying degrees. In addition, according to the analysis of physiological and biochemical data, salt stress had a considerable impact on both the physiological and biochemical processes of seedlings. Salt stress decreased the chlorophyll content and photosynthetic parameters of four genotypes, the REC, MDA content, Pro content, and SS content of each genotype increased by different degrees, and the enzymatic activities showed different trends. The salt tolerance of rootstocks was evaluated thoroughly using principal component analysis and membership function analysis based on the 16 parameters. The results of a comprehensive evaluation of salt tolerance showed that the order of salt tolerance of the four genotypes was J4 > J1 > J3 > J2, which corresponded to the order of the morphological symptoms of salt injury. In conclusion, J4 has strong salt tolerance and is an important germplasm resource for walnut salt-tolerant rootstock breeding.
RESUMO
Hypoxic environments have an adverse effect on the growth and development of P. praecox, and this is accompanied by the production of reducing substances such as Fe and Mn. In this study, the effect of hypoxic stress and Mn concentrations on leaf chlorophyll contents, root morphology, root activity, element absorption, antioxidant enzymes, and respiratory enzyme system of P. praecox were evaluated in a hydroponics environment. The results revealed that application of Mn2+ during hypoxic stress enhanced leaf chlorophyll contents and boosted up the indexes of the root system. The root activity of P. praecox was reduced with stresses of hypoxia. The treatment of Mn2+ initially improved and then decreased the root activity of P. praecox, and attained its maximum with application of 300 µmol/L Mn2+ compared with control. The indexes of antioxidant enzymes of P. praecox were higher than that of 8 mg/L oxygen concentrations except for variable superoxide dismutase (SOD) in the treatment of 300 µmol/L Mn2+ with hypoxia stress. The application of Mn had inhibited the absorption of mineral elements in P. praecox. The activities of pyruvate decarboxylase (PDC), alcohol dehydrogenase (ADH), and lactic dehydrogenase (LDH) were initially improved and then diminished with hypoxia stress. It is concluded that hypoxia is a key factor affecting the growth and degradation of P. praecox, while combining it with the increase of Mn concentration enhances the damage to Phyllostachys pubescens. Our research is helpful for the sustainable management and scientific fertilization management of Phyllostachys praecox.
