RESUMO

The Expert Panel for Cosmetic Ingredient Safety reviewed updated information that has become available since their original assessment from 2004, along with updated information regarding product types, and frequency and concentrations of use, and reaffirmed their original conclusion that Dioscorea Villosa (Wild Yam) Root Extract is safe as a cosmetic ingredient in the practices of use and concentration as described in this report.

There is ongoing research on various herbal bioactives and delivery systems which indicates that both lipid nanocarriers and herbal medicines will be fine tunned and integrated for future bio-medical applications. The current study was undertaken to systematically develop NLC-DSG-yam extract for the improved efficacy of herbal Diosgenin (DSG) in the management of anti-inflammatory disorders. NLCs were characterized regarding the mean size of the particles, morphological characteristics, physical stability in time, thermal behaviour, and entrapment efficiency of the herbal bioactive. Encapsulation efficiency and in vitro antioxidant activity measured the differences between the individual and dual co-loaded-NLC, the co-loaded one assuring a prolonged controlled release of DSG and a more emphasized ability of capturing short-life reactive oxygen species (ROS). NLCs safety properties were monitored following the in vitro MTS ((3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) tetrazolium reduction assay) and RTCA (Real-Time Cell Analysis) assays. Concentrations less than 50 µg/mL showed no cytotoxic effects during in vitro cytotoxicity assays. Besides, the NLC-DSG-yam extract revealed a great anti-inflammatory effect, as the production of pro-inflammatory cytokines (TNF-alpha, IL-6) was significantly inhibited at 50 µg/mL NLC (e.g., 98.2% ± 1.07 inhibition of TNF-α, while for IL-6 the inhibition percentage was of 62% ± 1.07). Concluding, using appropriate lipid nanocarriers, the most desirable properties of herbal bioactives could be improved.

BACKGROUND/AIM: Wild yam extract [Dioscorea villosa, (WYE)] is consistently lethal at low IC50s across diverse cancer-lines in vitro. Unlike traditional anti-cancer botanicals, WYE contains detergent saponins which reduce oil-water interfacial tensions causing disintegration of lipid membranes and causing cell lysis, creating an interfering variable. Here, we evaluate WYE at sub-lethal concentrations in MDA-MB-231 triple-negative breast cancer (TNBC) cells. MATERIALS AND METHODS: Quantification of saponins, membrane potential, lytic death and sub-lethal WYE changes in whole transcriptomic (WT) mRNA, miRNAs and biological parameters were evaluated. RESULTS: WYE caused 346 differentially expressed genes (DEGs) out of 48,226 transcripts tested; where up-regulated DEGS reflect immune stimulation, TNF signaling, COX2, cytokine release and cholesterol/steroid biosynthesis. Down-regulated DEGs reflect losses in cell division cycle (CDC), cyclins (CCN), cyclin-dependent kinases (CDKs), centromere proteins (CENP), kinesin family members (KIFs) and polo-like kinases (PLKs), which were in alignment with biological studies. CONCLUSION: Sub-lethal concentrations of WYE appear to evoke pro-inflammatory, steroid biosynthetic and cytostatic effects in TNBC cells.

Modern research is directed towards finding naturally-occurring antioxidants of plant origin for improving nutrition and health benefit. Wild yam species are used as indigenous food and medicine by various tribal groups of Koraput, India. However, detailed analysis of health promoting bioactive compounds and antioxidant properties are lacking. The present study aims to evaluate the flavonoid, total antioxidant capacity and in vitro antioxidant activity of different wild and cultivated Dioscorea species of Koraput. The extract of these tuber was screened for its potential antioxidant activities by various tests, such as DPPH, nitric oxide, superoxide and ABTS radical cation assay. The phenol, flavonoid and total antioxidant capacity of the samples were ranged from 2.19 to 9.62 mg g-1 dry weight, 0.62-0.85 mg g-1 dry weight and 1.63-5.59%, respectively. The IC50 values were 77.9-1164, 101.2-1031.6, 27.0-1022.6 and 47.1-690 µg ml-1 for DPPH, ABTS, superoxide and nitric oxide scavenging activity, respectively. Taken together, some wild yam species such as D. bulbifera, D. pubera and D. pentaphylla had significantly higher flavonoids content and antioxidant capacity along with the lower IC50 values than the standards in compared to the other species. The present studies confirmed that some wild yam tubers have superior potential for scavenging deleterious free radicals effectively. Results indicated that these wild yam species should be promoted as natural source of antioxidants.

Food and nutritional security are the major concerns in many countries of the world and may have potential to contribute to sustainable food systems under climate change. To address the food and nutritional insecurity, it has become extremely important to diversify the present-day agricultural system as well as to search for alternative food and feed ingredients. Some wild root and tuber crops occupy a remarkable position toward food security of the developing world due to their high calorific value and superior carbohydrate content. Yam (Dioscorea spp.) provides food and medicines to millions of people in the world especially in the tropics and sub tropics. It is recognized as the fourth most important tuber crop after potatoes, cassava, and sweet potatoes. It contributes about 10% of the total root and tubers production around the world. Yams also considered as famine food and plays a prime role in the food habit of small and marginal rural families and forest-dwelling communities during the food scarcity periods. These species are unique for their food, medicinal and economic values but their wider utilization is limited due to the presence of anti-nutritional compositions. This paper describes the ethnobotany of yam species in relation to their nutritional, anti-nutritional and pharmacological properties and highlights the potentiality for food and nutritional security for combating the "hidden hunger" caused by micronutrient deficiencies.

Wild yams (Dioscorea spp.) are important tuber crops used both as vegetable and medicine by the tribal people of Koraput, India. There is deficiency of documented information on genetic structure and diversity of wild yams and its genetic assessment is necessary for crop improvement program. The present study assessed the level of genetic diversity of eight wild and one cultivated yam species of Koraput by using different morphological and molecular markers. Significant variation in different yield and morphological traits was observed among the studied yam species. The major morphological traits such as branch number, stem thickness, tuber depth, tuber length, number of tubers per plant and yield showed high genetic heritability accompanied with high genetic advance and major determinants of phenotypic diversity. Molecular profiling was carried out by taking five simple sequence repeat markers. A total of 10 polymorphic bands with an average of two were detected at the loci of the five markers across the nine yam species. Genetic similarity analysis revealed that some wild yam species such as D. oppositifolia, D. hamiltonii and D. pubera showed higher genetic similarity with cultivated (D. alata) species. The knowledge of the extent of genetic variations of wild yam species is important for planning of the genetic conservation and the utilization of this resource especially for genetic improvement.

OBJECTIVES: Central African small-scale foragers subsist primarily on hunting game activities and wild plant-food gathering. Starch-rich tubers are underground storage organs (USOs) and staple food resources in savanna and tropical rainforests. However, little is known about the effect of USO consumption on tooth wear development in living hunter-gatherers. We report age- and sex-dependent tooth wear rates in forest-dwelling Baka Pygmies with well-documented wild-yam-tuber-based diet to explore the long-term impact of USO mechanical hardness and abrasiveness on the wearing down of the teeth. MATERIALS AND METHODS: Percentages of dentine exposure (PDEs) of permanent left mandibular first molars (M1 ) were recorded using in vivo high-resolution replicas of Baka individuals (aged 8-33 years), inhabiting Le Bosquet district in Cameroon (Western Africa). Regression and covariance analyses were used to test the effect of individual aging by sex on PDE rates. RESULTS: We found a strong increase of PDE by age among Baka individuals. No evidence of sexual dimorphism in wear patterns suggests similar sex-related dietary and masticatory demands during growth. Overall, greatest dentine exposure values ≈4% denote unexpected slow wear down rates for foraging diets relying on USO consumption. DISCUSSION: The low molar wear rates with age found in Baka Pygmies contrast with extensive wear rates in savanna-dwelling foragers, reflecting differences in thermal processing techniques affecting fracture toughness and grittiness of mechanically challenging foods. Our findings reveal that culture-specific dietary proclivities influence tooth wear among foraging behaviors with important implications in hominin dietary versatility and abrasive stress on chewing surfaces.

Wild yam (Dioscorea spp.) tubers, an indigenous food makes a significant contribution to the diets of tribal people of Koraput, India. However, there is dearth of documented information of their mineral bioavailability and heavy metal content. To know their quality and safety concerns about their use, minerals and heavy metal concentrations were evaluated in eight wild and one cultivated yam species from Koraput. The samples were further investigated for their antinutrients to determine bioavailability of minerals. The majority of the wild yam tubers were rich in some of the essential minerals like calcium (18.08-74.79 mg/100 g), iron (11.15-28.61 mg/100 g), zinc (2.11-6.21 mg/100 g) and phosphorous (179-248 mg/100 g). The heavy metals concentration (mg/100 g) ranged from: cobalt (1.06-1.98), nickel (0.30-0.89), chromium (2.10-4.53) and lead (0.11-0.93) among the studied yam species. These values were lower than the recommended tolerable levels proposed by WHO Expert Committee on Food Additives. Based on these results of molar ratio between phytate and Fe, Zn and Ca were below the suggested critical values indicating the bioavailability of Fe, Zn and Ca to be high. The study also suggests these wild yam species as the safe food sources for mass consumption and can be beneficial for health.

Hyperproduced prostaglandin E2 by cyclooxygenase-2 and microsomal prostaglandin E synthase-1 evokes several pathophysiological responses such as inflammation and carcinogenesis. Our recent study demonstrated that Dioscorea japonica extract suppressed the expression of cyclooxygenase-2 and microsomal prostaglandin E synthase-1 and induced apoptosis in lung carcinoma A549 cells. In the present study, we investigated the effects of Dioscorea japonica on squamous cell carcinoma of mouse skin. Dioscorea japonica feeding and Dioscorea japonica extract topical application suppressed the expression of cyclooxygenase-2, microsomal prostaglandin E synthase-1, interleukin-1ß and interleukin-6 and inhibited tumor formation, hyperplasia and inflammatory cell infiltration. Immunohistochemical analyses showed the immunoreactivities of cyclooxygenase-2 and microsomal prostaglandin E synthase-1 in tumor keratinocytes and stronger immunoreactivities of cyclooxygenase-2 and hematopoietic prostaglandin D synthase in epidermal dendritic cells (Langerhans cells). Treatment with Dioscorea japonica decreased the immunoreactivity of cyclooxygenase-2 and microsomal prostaglandin E synthase-1. These results indicate that Dioscorea japonica may have inhibitory effects on inflammation and carcinogenesis via suppression of the prostaglandin E2 synthetic pathway.

Tubérculos del género Dioscorea comercializados con fines medicinales, fueron recolectados con el propósito de lograr su establecimiento a condiciones in vitro. Previamente se lograron identificar taxonómicamente las especies y por medio de análisis fitoquímicos demostrar su potencial farmacéutico. El material recolectado fue identificado como Dioscorea coriacea, D. lehmannii, D. meridensis, D. polygonoides y una especie comestible D. trifida. Tubérculos recolectados de centros de acopio y traídos de campo fueron lavados, desinfectados, asperjados con Ácido Giberélico (AG3) y sembrados en sustrato BM-2®, en invernadero a 18°C día y 10°C noche. Los tubérculos completos o por secciones fueron almacenados en bolsas herméticas a temperatura ambiente. Posteriormente se desinfectó material vegetal de las especies D. coriacea, D. lehmannii, D. meridensis y D polygonoides, seleccionando explantes de brotes sanos (D. coriacea / laboratorio) para su establecimiento. Se evaluaron tres medios de cultivo para establecimiento, el que presentó los mejores resultados fue Medio Murashige & Skoog (1962) suplementado con BAP 1 mL/L, AG3 1 mL/L y Putrescina 2 mL/L. Para la extracción y análisis de metabolitos secundarios se utilizaron tubérculos de D. coriacea, D. lehmannii y D. polygonoides, empleando como solvente de extracción metanol. Se encontró mayor concentración de extracto vegetal en D. coriacea (54%), y mediante cromatografía en capa delgada (CCD), se confirmó la presencia de saponinas, que resultó mayor en comparación con D. polygonoides especie reconocida por su alto contenido de saponinas. Estos resultados permitirán realizar análisis más avanzados de los compuestos presentes y plantear su propagación masiva en condiciones in vitro.


Wild tubers of the genus Dioscorea sold for medicinal use were collected for the purpose of achieving its establishment under in vitro conditions. First we taxonomically identified the species and through phytochemical analysis demonstrated pharmaceutical potential. The material collected was identified as Dioscorea coriacea, D. lehmannii, D. meridensis, D. polygonoides and the edible species D. trifida. Tubers collected from wholesale distributors and from the field were washed, disinfected, sprayed with Gibberellic Acid (GA3) and planted in substrate BM-2®, in a greenhouse at 18 ° C during the day and 10 ° C overnight. Whole tubers or sections thereof were stored in sealed bags at room temperature. Subsequently plant material of the species D. coriacea, D. lehmannii, D. meridensis and D. polygonoides was disinfected and healthy buds (D. coriacea / laboratory) were selected for in vitro establishment. Three different culture media were evaluated for establishment; that which presented the best results was the Murashige & Skoog (1962) medium, supplemented with BAP 1 mL / L, GA3 1 mL / L and Putrescin 2 mL / L. For the collection and analysis of secondary metabolites, tubers of D. coriacea, D. lehmannii and D. polygonoides were used, using methanol as the extraction solvent. The highest concentration of plant extract, 54%, was found in D. coriacea, a higher value than that of D. polygonoides, which had been reported previously; the presence of saponins was confirmed by thin layer chromatography (TLC). These results will enable more advanced analysis of the present compounds and enhance their mass propagation under in vitro conditions.

Two groups of lipidated steroid saponins including seven new compounds (2, 3, 5, and 7-10) were isolated from the widely used botanical, wild yam (Dioscorea villosa), employing a fractionation protocol of metabolomic mining. This methodology recently led to the isolation of 14 diarylheptanoids from the same plant. Together with these lipidated steroid saponins, they establish additional new markers for D. villosa. The lipidation of steroids with analog long-chain fatty acids containing different degrees of unsaturation generates an entire series of compounds which are difficult to purify and analyze. The structures of the two series of lipidated steroid saponins (series A and B) were established by a combination of 1D and 2D NMR as well as GC-MS after chemical modification. Series A was determined to be a mixture of lipidated spirostanol glycosides (1-5), while series B (6-10) was proved to be a mixture of five lipidated clionasterol glucosides. The latter group represents the first derivatives of clionasterol to be found in D. villosa. The discovery of this specific structural type of aliphatic esters of steroid saponins expands the characterization of the secondary metabolome of D. villosa. It may also inspire biological studies which take into account the lipophilic character and significantly altered physiochemical characteristics of these otherwise relatively polar phytoconstituents.
