INTRODUCTION: Congenital deformities of the limbs occur sporadically in various species, but the cause is often unclear. The clinically healthy female Brown Swiss calf presented here showed a congenital peromelia of the left hind limb. The affected limb is twisted, disproportional and the bones distally of the metatarsus are missing. Karyotyping and genome sequencing did not indicate on a genetic cause of the anomaly. An infection with the Schmallenberg virus could not be ruled out. Furthermore, there was no evidence of further adverse environmental effects during pregnancy.


INTRODUCTION: Des malformations congénitales des membres, dont la cause est souvent peu claire, surviennent sporadiquement chez diverses espèces. Le veau Brown Swiss femelle présenté ici, tout en étant cliniquement sain, présentait une péromélie congénitale du postérieur gauche. Le membre concerné été en rotation interne, disproportionné et les os distalement au métatarse étaient absents. La détermination du caryotype et le séquençage de l'ensemble du génome n'ont apporté aucun élément parlant pour une cause génétique de l'anomalie. Il n'a pas été possible d'exclure une infection par le virus de ­Schmallenberg. D'autre part il n'y avait aucun élément évoquant d'autres influences environnementales néfastes durant la gestation.

RESUMO

RESUMEN: El objetivo del trabajo fue determinar los efectos del ciclamato de sodio en la placenta de ratas, administrado durante el periodo de la embriogénesis. Fue administrado por vía intraperitoneal, en las ratas del grupo tratado, ciclamato de sodio en una dosis de 60 mg/Kg, desde el 10 al 14 día de gestación, siendo inyectado un volumen equivalente de solución salina en el grupo de ratas control. En el 20 día de preñez, 10 fetos (5 de cada grupo) fueron escogidos al azar para su estudio. Fue utilizada la técnica de cariometría para la evaluación de los parámetros nucleares de las células de las capas decidua, esponjosa y de las vellosidades coriónicas de la placenta de las ratas. El peso de los fetos tratados y de sus placentas fue menor que el del grupo control, así como también el funículo umbilical del grupo tratado fue más corto que el de los fetos controles. No hubo alteraciones en la capa de la decidua. En la capa esponjosa placentaria ocurrieron alteraciones de los siguientes parámetros: diámetro mayor, diámetro medio, perímetro, área, volumen, relación volumen/área y excentricidad. Los parámetros alterados en las vellosidades coriónicas fueron: diámetro medio, perímetro, área, volumen y relación volumen/área. El estudio demostró alteración placentaria con el uso del ciclamato de sodio en la rata preñada y su repercusión en el peso fetal y largo del funículo umbilical.

By means of histological methods of histochemical nature we examined the qualitative alterations in neurosecretory cells of anterior (nucl. paraventricularis - NPV. nucl. preopticus medialis - NPM), medial (nucl. arcuatus s. infundibularis - NARC), and posterior (nucl. tuberomamillaris - NTM) hypothalamus of sheep with a simultaneous caryometric analysis of neurons of the same nuclei after administration of various PMSG doses. The examination was performed on the brain samples of 23 two- to three-year-old sheep, of the Slovak Merino breed, at the live weight of 30 to 40 kg. in the period of physiological anoestrus (June-July). The animals were divided into three experimental groups and were instilled agellin sponges (20 mg). On the 13th day the sponges were removed and the first group was applied 1000 i. u. PMSG, the second group 750 i. u. PMSG. The sheep were killed within 30 to 36 hours after the oestrus determination with a teaser ram. After bleeding of animals the brains were perfused with 2% buffered paraformaldehyde and after taking out the brains from the crania their fixation was finished in buffered picroformol. Paraffin slices were stained with haematoxylin-eosine and crotonaldehyde-fuchsin to neurosecretions. Caryometric analysis was carried out with a 1000 fold magnifying and with a measuring of 200 cells from one sample. We recorded the neurosecretion increase in all hypothalamic nuclei under study. The caryometric analysis has shown a moderate shift to the right, it means the volume increase of nuclei of neurosecretory cells, which demonstrates a stimulation of the function of hypothalamic structures.

RESUMO

After an incubation period of four days and colchicine pretreatment (air-cell method), clear metaphase figures were obtained from a cell suspension of the allantoic sacs. It was only by shortening the colchicine application to one hour that sufficiently elongated chromosomes suitable for karyological studies could be prepared. The total arm length and the arm length indices are given of the six pairs of macrochromosomes - including the Z chromosome - and of the three largest microchromosomes. Different from the previous findings as reported in the literature, the two largest macrochromosomes are described as submetacentric and not as metacentric. The findings on the seven chromosomes following agree with those given in the literature.
