RESUMO

This study was aimed to provide anatomical atlas of the ovine metacarpo/metatarsophalangeal joints using computed tomography (CT) scan and magnetic resonance imaging (MRI), as well as to investigate bone mineral density (BMD) and morphometric features of this joint. The limbs of twenty adult Sanjabi sheep were examined. Imaging was performed using a 16-slice multi-detector CT scanner and a 1.5 Tesla MRI scanner. The obtained images were correlated with corresponding anatomical sections. BMD was measured by Dual Energy X-ray Absorptiometry (DEXA) method. Also morphometric features included distance between metacarpal and metatarsal heads, width of metacarpal and metatarsal head, length, width and the height of the proximal sesamoid bones were measured. In MRI images, compact bones, ligaments and tendons showed less signal intensity (hypointense). The fatty tissue and bone marrow had more signal intensity (hyperintense), but articular cartilage and synovial fluid showed moderate signal intensity. BMD in the left hind limb was significantly higher than other three limbs (p ≤ 0.05). The length of proximal sesamoid bones in left forelimb was higher than right forelimb (p ≤ 0.05). The thickness of proximal sesamoid bones in left limbs was higher than those in their counterparts (p ≤ 0.05). The width of the medial head in the forelimbs was significantly greater than the hind limbs (p ≤ 0.05). The present results might be useful in managing the clinical techniques on this joint. Larger volume of the proximal sesamoid bones and wider medial head in the forelimbs compared to the hindlimbs impel this speculation that the centre of gravity is closer to the forelimbs.

The purpose of this study was to evaluate infrared beak trimming versus hot-blade beak trimming on performance, organ and bone development of laying pullets, using 240 birds, at 0-16 weeks of age, from two strains (white layer - Hy-Line W36 and brown layer - Hy-Line Brown). Infrared debeaking was performed on the first day of life in the hatchery, whereas hot-blade beak trimming was performed at eight days of age. Zootechnical performance at 6, 9, 12 and 15 weeks of age was analyzed. At the end of the trial, one Bird per experimental unit was euthanized for liver, gizzard, proventriculus, heart and small intestine relative weight evaluation. Both tibiotarsus were collected to measure bone breaking resistance, bone mineral content and bone densitometry. There was higher body weight and feed intake in both bird strains submitted to infrared beak trimming. Additionally, hot-blade white layers presented lower body development and feed intake. The debeaking methods did not influence relative organ weights or tibiotarsus parameters evaluated. We conclude that the infrared beak trimming method promotes better pullet body development than the conventional hot-blade method, improving feed intake, without compromising organ and bone development, being a more adequate debeaking method to be used in layer production.(AU)


Para avaliar a debicagem infravermelha versus a debicagem pelo método da lâmina quente no desempenho, no desenvolvimento de órgãos e na qualidade óssea de poedeiras em recria, foram utilizadas 240 aves, entre zero e 16 semanas de idade, em duas linhagens (branca - Hy-Line W36 e vermelha - Hy-Line Brown). A debicagem por infravermelho foi realizada no primeiro dia de vida no incubatório, enquanto a debicagem com lâmina quente foi realizada aos oito dias de idade. Analisou-se o desempenho zootécnico na sexta, nona, 12ª e 15ª semanas de idade. No final do estudo, uma ave por unidade experimental foi eutanasiada para avaliação do peso relativo do fígado, da moela, do proventrículo, do coração e do intestino delgado, e ossos do tibiotarso foram coletados para avaliação da resistência à ruptura, do conteúdo mineral e da densitometria óssea. Houve maior peso corporal e consumo alimentar nas aves pertencentes às duas linhagens quando submetidas à debicagem por infravermelho, sem haver influência sobre o peso relativo dos órgãos e a qualidade óssea. Conclui-se que o método de debicagem por radiação infravermelha promove melhor desenvolvimento corporal das frangas em relação à debicagem por lâmina quente, melhorando consumo de ração, sem comprometer o desenvolvimento de órgãos e ossos, sendo mais adequada para utilização na avicultura de postura comercial.(AU)

Avaliou-se o efeito da utilização de níveis crescentes de vitamina A sobre os parâmetros ósseos de codornas de corte de 15 a 35 dias de idade. Foram utilizadas 1520 codornas, não sexadas, com peso médio inicial de 85±4,25g, distribuídas em um delineamento inteiramente ao acaso, totalizando oito tratamentos com cinco repetições e 38 codornas por unidade experimental. Os tratamentos consistiram de oito níveis de suplementação de vitamina A (0; 4.500; 6.000; 7.500; 9.000; 10.500; 12.000 e 13.500UI/kg da dieta). O período experimental foi de 15 a 35 dias de idade. Os ossos analisados apresentaram uma redução linear para o índice de Seedor, resistência óssea e teor de cinzas do fêmur e da tíbia de acordo com os níveis de suplementação de vitamina A. A suplementação de vitamina A para codornas de corte de 15 a 35 dias de idade mostrou resultados negativos para a qualidade óssea, com a necessidade de mais estudos sobre seu antagonismo com a vitamina D.(AU)


This study evaluated the effect of using increasing levels of vitamin A on bone parameters in meat-type quails from 15 to 35 days old. A total of 1520 unsexed quail with an average initial weight of 85g distributed in a completely randomized design, totaling eight treatments with five replicates and 38 quails per experimental unit were used. The treatments consisted of eight vitamin supplementation levels A (0; 4,500; 6,000; 7,500; 9,000; 10,500; 12,000 and 13,500IU / kg diet). The experimental period was 15 to 35 days of age. The bones analyzed showed a linear reduction to the Seedor index, bone strength and ash content of the femur and tibia according to vitamin supplementation levels Vitamin A supplementation for cutting quails from 15 to 35 days of age showed negative results for bone quality, with the need for more studies on its antagonism to vitamin D.(AU)

BACKGROUND: Densitometric quantitation using serum protein electrophoresis (SPE) is used to monitor monoclonal proteins (M-proteins) in human patients but has not been validated in the dog. Serum globulin concentrations, species-specific radial immunodiffusion (RID), and ELISAs are currently used in veterinary medicine. OBJECTIVE: We aimed to compare four methods that quantify M-proteins using densitometry and biuret protein (dM-protein) measurements. We also validated the best performing method and compared it with the RID and ELISA methods for measuring canine serum M-protein. METHOD: Serum from six normal dogs and 83 serum samples from 46 dogs with confirmed monoclonal gammopathies were used. A spike and recovery experiment with purified monoclonal IgG and IgM, inter-run and intra-run variability, linearity under dilution, and lower limit of detection were performed. Results of commercial canine RID and ELISA kits for total class-specific immunoglobulin were compared with dM-proteins. RESULTS: The corrected perpendicular drop gating method had <20% error for IgG/γ-globulin and IgM/ß-globulin M-protein quantifications. Linearity (r > .99), intra-run CV (1.1%-2.3%), and inter-run CVs (2.0%-3.5%) were acceptable. Correlation between the RID and densitometry results ranged from r = .25 to r = .88, depending on the class. The RID result was greater than that of the biuret total protein in 26/63 (41%) IgA cases. A panel of IgG, IgA, and IgM RIDs failed to correctly identify an IgM paraproteinemia in 6/6 (100%) cases. Densitometry was not comparable with any other tested method. CONCLUSION: Densitometric quantitation is a valid technique for measuring M-proteins in the ß- and γ-globulin regions. Immunotyping via RID using the tested kit does not appear to detect IgM. Densitometry is recommended for measuring M-proteins in canine patients.

The evaluation of radiodensity using quantitative computed tomography (QCT) has several indications in animals. It allows for standardization of reference values for bone densitometry and helps in the clinical diagnosis of diseases. Currently, there is a lack of such data in the literature for Boa constrictor constrictor. Therefore, the objective of this study was to estimate reference values for bone density in boas ( Boa constrictor constrictor). Computed tomography scans were performed on 10 adult, healthy, free-ranging boas. Results showed mean values of vertebral trabecular bone for free-ranging boas were 1,237.91 ± 54.61 mg/cm3. Because of the difficulty in obtaining clinical and radiographic diagnoses of metabolic bone diseases, especially in a timely manner, densitometry may become a helpful tool in preventive veterinary medicine.

Interrelationships between morphological, densitometric, and mechanical properties of deciduous mandibular teeth (incisors, canine, second premolar) were investigated. To perform morphometric, densitometric, and mechanical analyses, teeth were obtained from 5-month-old sheep. Measurements of mean volumetric tooth mineral density and total tooth volume were performed using quantitative computed tomography. Microcomputed tomography was used to measure total enamel volume, volumetric enamel mineral density, total dentin volume, and volumetric dentin mineral density. Maximum elastic strength and ultimate force of teeth were determined using 3-point bending and compression tests. Pearson correlation coefficients were determined between all investigated variables. Mutual dependence was observed between morphological and mechanical properties of the investigated teeth. The highest number of positive correlations of the investigated parameters was stated in first incisor indicating its superior predictive value of tooth quality and masticatory organ function in sheep. Positive correlations of the volumetric dentin mineral density in second premolar with final body weight may indicate predictive value of this parameter in relation with growth rate in sheep. Evaluation of deciduous tooth properties may prove helpful for breeding selection and further reproduction of sheep possessing favorable traits of teeth and better masticatory organ function, leading to improved performance and economic efficiency of the flock.

1. Sex-related differences of long pelvic limb bones and serum bone metabolism indices were evaluated in 14-month-old female (N = 7) and male (N = 7) ostriches of similar body weights. 2. Densitometric parameters of femur, tibia and tarsometatarsus were determined using quantitative computed tomography (volumetric bone mineral density, calcium hydroxyapatite density and mean volumetric bone mineral density) and dual energy X-ray absorptiometry (bone mineral density and bone mineral content) methods. Geometrical parameters such as cortical bone area, cross-sectional area, second moment of inertia, mean relative wall thickness and cortical index were determined in the midshaft of bones. Mechanical properties of bones (maximum elastic strength and ultimate strength) were evaluated using three-point bending test. Serum concentrations of free amino acids, osteocalcin, N-terminal propeptide of type I procollagen, C-terminal telopeptides of type II collagen and total antioxidative capacity were also determined. 3. Bone weight and relative bone weight of all bones were significantly higher in males than in females. Significantly lower values of trabecular bone mineral density and calcium hydroxyapatite density were found in the trabecular bone of tibia in males. The highest number of the sex-related differences was observed in the tarsometatarsus where bone length, bone mineral content, cortical bone area, cross-sectional area and ultimate strength were higher in males. Serum concentrations of taurine, hydroxyproline, valine and isoleucine were significantly higher in males. 4. Higher loading of the tarsometatarsus in comparison to femur and tibia may be an important factor interacting with sex hormones in regulation of bone formation and mineralisation processes. Sex-related differences of bone properties were associated with increased serum concentration of selected amino acids in males.

BACKGROUND: Lactate dehydrogenase (LD) exists as 5 isoenzymes (LD-1 through LD-5) that are expressed throughout the body and can be detected in both serum and cerebrospinal fluid (CSF). LD and its isoenzymes have been relatively unstudied in veterinary medicine, although studies in human medicine have demonstrated that changes in total LD activity and atypical isoenzyme patterns can indicate disease processes, including neurologic abnormalities. OBJECTIVES: The purpose of this study was to establish RIs for LD and its isoenzymes in the serum and CSF of clinically healthy dogs. By establishing a definitive RI for this enzyme in healthy canines, further study of the clinical and diagnostic usefulness of LD can be undertaken. METHODS: Serum and atlantoaxial CSF were collected from clinically healthy dogs. Total LD activity was measured spectrophotometrically immediately after collection. Isoenzyme distributions were also determined within 8 hours of collection using the QuickGel LD Isoenzyme technique and a densitometric scanner. RESULTS: The median serum total LD in healthy canines was 69.0 U/L (n = 41; range: 21.0-217.0 U/L), while the median CSF total LD was 10.0 U/L (n = 40; range: 6.0-19.3 U/L). LD-5 is the predominant isoenzyme in canine serum (n = 40), contributing over half of the total enzyme activity. Conversely, in canine CSF (n = 42), LD-1 is the predominant isoenzyme, followed by LD-2 and LD-3. CONCLUSIONS: Knowledge of the distribution and concentration of LD in the serum and CSF of healthy dogs will set the foundation for future studies of canine LD as a potentially clinically useful biomarker.

1. The aim of the study was to investigate the effects of caponisation on bone development of males of two native breeds in Poland. 2. The weight, length and cross-sectional area of tibiae and femora were measured, densitometric measurements and tests of strength were determined and dimensions were calculated. 3. Breed and caponisation did not influence bone weight and length. Higher mechanical strength of the femur was found in entire males, mainly in the Polbar breed. Tibial strength was reduced in capons of the Green Partridge breed. Maximum elastic strength was greater in the Polbar, irrespective of caponisation. Bone cross-sectional area was influenced by breed, while caponisation reduced femoral bone mineral density in both breeds. 4. Caponisation thus increased growth rate but had adverse effects on bone development. 5. Caponisation had fewer negative effects in the Polbar than in the Greenleg Partridge.

We developed and evaluated a fast and simple method to obtain suitable bone samples for densitometry and chemical analysis through biopsies of the 12th rib of cattle. The postoperative recovery, dry matter intake (DMI) and average daily weight gain (ADG) was evaluated in 36 Nellore steers, nine of which were randomly selected for the control group and 27 others were rib biopsied. Every 30 days, rib biopsy was performed in nine steers, using a corded-electric pistol-grip drill coupled with a hole saw of a 3mm diameter pilot drill bit. This rib biopsy technique provided a suitable sample obtained in a fast way and allowed the surgeon to work alone with the animal slightly sedated and restrained in the crush. Dry matter intake (DMI) was not affected in biopsied animals. At the end of the experimental period (116 days) the average daily weight gain (ADG) was similar in the steers biopsied or not. The described method provided rib samples from cattle suitable for densitometry and chemical analysis of bone tissue without effects on health and performance. This information could greatly increase the accuracy for the diagnosis of phosphorus deficiency in cattle raised on pasture and allow the evaluation of bone metabolism in experimental animals.(AU)


No presente estudo foi desenvolvida uma técnica para se obter, de forma rápida e simples, amostras da 12a costela bovina por meio de biópsias. Avaliou-se a recuperação pós-operatória (durante 15 dias), o consumo de matéria seca (CMS) e o ganho de peso diário (GMD) de 36 novilhos Nelores, sendo nove animais controles e 27 submetidos à biópsia. A cada 30 dias biópsias de costelas foram realizadas em nove animais, usando uma furadeira elétrica acoplada a uma serra-copo com uma broca-guia de 3mm de diâmetro. Esta técnica de biópsia permitiu obter amostras ósseas de forma rápida, com o cirurgião operando sozinho e com os animais levemente sedados e em estação. Ao término do experimento (116 dias) o CMS e o GMD não foram afetados pelo procedimento cirúrgico. As amostras obtidas foram adequadas para exames radiográficos e subsequentes análises químicas. O conjunto de informações adquiridas com a técnica de biópsia de costelas permite aumentar a exatidão no diagnóstico da deficiência de fósforo em bovinos criados em pastagens e também realizar estudos sobre metabolismo ósseo em animais experimentais.(AU)

The study investigated in piglets the effect of replacing dietary inorganic P by addition of microbial phytase and its impact on performance, nutrient digestibility and on the geometrical characteristics and mineralisation of the femur. Sixteen pigs on day 58 of age were divided into two groups and fed either a diet free of additional inorganic phosphorus (P) and supplemented with phytase (Diet LP, 4.23 g total P/kg diet) or a diet with a mineral source of P and not supplemented with phytase (Diet SP, 5.38 g total P/kg diet). Performance data and the apparent total tract digestibility of nutrients were estimated between days 58 and 114, and 72 and 86 of age, respectively. On day 114 of age, the pigs were slaughtered, the femur was dissected and the mineral content and mineral density, maximum strength and maximum elastic strength, cortical wall thickness, cross-sectional area and cortical index were analysed. The growth performance and digestibility of nutrient fractions (with exception of P) did not differ between treatment groups. The P-digestibility was significantly higher in Group LP. The femur of pigs in Group LP had significantly greater cortical wall thickness, cortical index, bone mineral content, bone mineral density, maximum strength and maximum elastic strength than Group SP. Femur maximum strength and maximum elastic strength were correlated with cortical wall thickness and cortical index. Resulting from the different supply of digestible P, the femur geometrical, densitometric and mechanical properties of Group LP were better than those of Group SP. The mechanical properties of the femur of pigs depended more on its geometrical characteristics than on the degree of its mineralisation.

O presente estudo comparou, por meio do ensaio de flexão em quatro pontos, a resistência de segmentos de rádios de cães autoclavados e desvitalizados em nitrogênio líquido. Avaliaram-se 40 rádios, que foram divididos em quatro grupos: grupo autoclave (GA), grupo controle autoclave (GCA), grupo nitrogênio (GN) e grupo controle nitrogênio (GCN). Os corpos de prova dos grupos GCA e GCN pertenciam ao lado contralateral dos grupos GA e GN, respectivamente. Os ossos foram autoclavados a 121°C, por 20min a 2atm. A desvitalização em nitrogênio foi realizada por meio da imersão em nitrogênio líquido, por 20min, seguida de descongelamento à temperatura ambiente, sendo finalizada por imersão em solução de NaCL a 0,9%, por mais 15min. Após os testes biomecânicos, foram comparadas as variáveis força máxima, rigidez, altura máxima à secção transversa do corpo de prova, deslocamento do baricentro em relação ao eixo x, flecha, tensão-tração, tensão-compressão e tensão máxima. Não foram encontradas diferenças estatisticamente significantes entre os grupos GN e GCN. Foram encontradas diferenças estatisticamente significantes entre os grupos GA e GCA somente na comparação da variável força máxima. A análise das variáveis tensão-tração, tensão-compressão e tensão máxima evidenciou diferenças estatisticamente significantes entre os grupos GA e GN, sendo os valores maiores para o grupo GA. Também foi realizada comparação entre os deltas (GN menos GCN e GA menos GCA), em que não foram observadas diferenças estatísticas.(AU)


The study compared through the four points bending test, the strength of bone segments autoclaved and devitalized in liquid nitrogen radius segments. 40 radius were used, collected and divided into 4 groups, as follows: Autoclave Group (GA), Control Autoclave Group (GCA), Nitrogen Group (NG) and Control Nitrogen Group (GCN). Bone segments from groups GCA and GCN were contralateral to GA and NG. The segments were autoclaved at 121ºC and 2 atm for 20 minutes. Nitrogen devitalization of the bone segments was obtained by 20 minutes immersion, followed by thawing at room temperature and 15 minutes immersion in 0.9% NaCL solution. After biomechanical tests the variables compared were maximum strength, stiffness, maximum hight trough the transverse section of the bone segment, centers of mass dislocation in relation to x axe, arrow, stress-tension, stress-compression and maximum stress. No statistically significant differences between the GN and GCN groups were found. Statistically significant differences were found between GA and GCA groups only in the comparison of the maximum force variable. The analysis of variable stress-tension, stress-compression and maximum stress showed statistically significant differences between the GA and GN groups, with higher values for the GA group. Comparison of the deltas (GN least GCN and GA least GCA) where no statistical differences were observed was also performed.(AU)

Osteoporosis is a multifactorial disease of high prevalence and has great impact on quality of life, because the effects on bone structure increase the risk of fractures, what may be very debilitating. Based on the observation that patients with depression have lower bone mineral density than healthy individuals, many studies have indicated that stress could be an aggravating factor for bone loss. This study evaluates the effect of a protocol of chronic mild stress (CMS) on parameters of bone assessment in male and female rats. Five 5-monh-old rats of each sex underwent a schedule of stressor application for 28 days. Stressors included cold, heat, restraint, cage tilt, isolation, overnight illumination, and water and food deprivation. Five rats of each sex were kept under minimum intervention as control group. The animals were weighed at beginning and end of the period, and after euthanasia had their bones harvested. Femur, tibia and lumbar vertebrae were analyzed by bone densitometry. Biomechanical tests were performed in femoral head and diaphysis. Trabecular bone volume was obtained from histomorphometric analysis of femoral head and vertebral body, as well as of femoral midshaft cross-sectional measures. Not all parameters analyzed showed effect of CMS. However, tibial and L4 vertebral bone mineral density and cross-sectional cortical/medullar ratio of femoral shaft were lower in female rats submitted to the CMS protocol. Among male rats, the differences were significant for femoral trabecular bone volume and maximum load obtained by biomechanical test. Thus, it could be confirmed that CMS can affect the balance of bone homeostasis in rats, what may contribute to the establishment of osteopenia or osteoporosis.(AU)


A osteoporose é uma doença multifatorial, de alta prevalência e que tem um grande impacto na qualidade de vida, principalmente porque os efeitos sobre a estrutura do osso aumentam o risco de fraturas, que podem ser muito debilitantes. Com base na observação de que pacientes com depressão têm menor densidade mineral óssea que indivíduos saudáveis​​, muitos estudos têm indicado que o estresse pode ser um fator agravante para a perda óssea. Este estudo avalia o efeito de um protocolo de estresse moderado crônico (EMC) em parâmetros de avaliação óssea em ratos machos e fêmeas. Cinco animais de cada sexo, com cinco meses de idade, foram submetidos a um cronograma de aplicação de estressores durante 28 dias. Os estressores incluídos foram: frio, calor, contenção, inclinação da gaiola, isolamento, iluminação durante a noite e privação de água e ração. Cinco animais de cada sexo foram mantidos com um mínimo de intervenção como grupo controle. Os animais foram pesador no início e no final do período, e após eutanásia tiveram seus ossos coletados. Fêmur, tíbia e vértebra lombar foram analisados por densitometria óssea. Testes biomecânicos foram realizados na cabeça e na diáfise do fêmur. Volume trabecular ósseo foi obtido a partir de análise histomorfométricas da cabeça do fêmur e do corpo vertebral, bem como medidas da seção transversal diáfise femoral. Nem todos os parâmetros avaliados sofreram efeito do protocolo de EMC. No entanto, a densidade mineral óssea da tíbia e da vértebra L4 e a razão osso cortical/medula da seção transversal da diáfise femoral foram menores nas fêmeas submetidas ao protocolo. Entre os ratos machos, as diferenças foram significativas no volume trabecular ósseo da cabeça femoral e na carga máxima obtida no teste biomecânico. Assim, confirma-se que o protocolo de EMC pode afetar o equilíbrio da homeostase óssea em ratos, o que pode contribuir para o estabelecimento de osteopenia ou osteoporose.(AU)

: Infidum similis Travassos, 1916 (Dicrocoeliidae: Leipertrematinae) was found in the gall bladder of Leptophis diplotropis Günther, 1872 from El Podrido, Acapulco, Guerrero, Mexico. A phylogenetic analysis based on partial sequences of the 28S ribosomal RNA using maximum likelihood (ML) and Bayesian inference (BI) analyses was carried out to assess its phylogenetic position within suborder Xiphidiata, alongside members of the superfamilies Gorgoderoidea and Plagiorchoidea. The phylogenetic trees showed that the genus is most-closely related to the Plagiorchoidea rather than to the Gorgoderoidea, in keeping with previous taxonomic designations. Phylogenies obtained from ML and BI analysis of the 28S rDNA gene revealed a well supported clade in which Choledocystus hepaticus (Lutz, 1928) Sullivan, 1977 is sister to I. similis. On the other hand, a tree obtained using a partial sequence of the cytochrome c oxidase subunit 1 (cox1) mtDNA gene (ML and BI analysis), with species supposed to be closely related to I. similis according to 28S, does not support this relatedness. Based on the independence of Infidum from the subfamily Leipertrematinae Yamaguti, 1958 , our results clearly demonstrated that the genus corresponds to a different family and with species closely related to C. hepaticus within Plagiorchoidea. New data are presented about the tegumental surface of I. similis by scanning electron microscopy as well as the estimation of its haploid genome size using Feulgen Image Analysis Densitometry of sperm nuclei as part of the characterization of this species. This is the first genome size estimated for a member of Plagiorchiida, and these data will provide a new source of knowledge on helminth diversity and evolutionary studies. This constitutes the first host record, and new geographical distribution, for this species in Mexico.

Este trabalho objetivou verificar se o ultrassom de baixa potência (US) previne a ocorrência de osteopenia em tíbias de ratos sob ausência de carga. Foram utilizados 45 Rattus novergicus albinus, Wistar adultos, machos, distribuídos em cinco grupos iguais: C - animais-controle livres em gaiolas por 21 dias; S - animais suspensos pela cauda por 21 dias; ST - suspensos pela cauda por 21 dias e concomitantemente tratados com US; S→C - suspensos por 21 dias e depois permanecendo livres em gaiolas por mais 21 dias; S→CT - suspensos por 21 dias e depois permanecendo livres em gaiolas por mais 21 dias e concomitantemente tratados com US. O tratamento foi realizado com US de 1,5MHz, ciclo de trabalho 1:4, 30mW/cm², na tíbia direita, por 15 sessões de 20 minutos cada, cinco sessões por semana. Ainda vivos, os animais foram submetidos a exame de densitometria óssea para verificação da densidade mineral óssea (DMO) e do conteúdo mineral ósseo (CMO). Após a eutanásia dos animais, as tíbias foram desarticuladas, dissecadas e submetidas a ensaio mecânico destrutivo para análise da força máxima (Fmáx) e da rigidez (R). Foram avaliados também o comprimento (L) e o diâmetro (D) no ponto médio da tíbia. O grupo S apresentou valores de DMO, CMO, Fmáx, R, L e D menores em relação ao grupo C, demonstrando que a suspensão pela cauda é prejudicial a estas variáveis. O tratamento dos animais suspensos com o US, grupo ST, elevou os valores de CMO e DMO em relação aos do grupo S, igualando-os aos do grupo C...


This work intended to verify if the low power ultrasound (US) prevents the occurrence of osteopenia on the tibia of rats under the absence of load. 45 adult male Wistar Rattus novergicus albinus were used, separated in five equal groups: C - control free animals in cages for 21 days; S - animals suspended by the tail for 21 days; ST - animals suspended by the tail for 21 days and concurrently treated with US; S→C - animals suspended for 21 days and then free in cages for another 21 days; S→CT - animals suspended for 21 days and then free in cages for another 21 days and concurrently treated with US. The treatment was performed with US with 1.5 MHz, work cycle 1:4, 30 mW/cm², on tibia, for 15 sessions of 20 minutes each, 5 sessions per week. While still alive, the animals underwent bone densitometry examination to verify the Bone Mineral Density (BMD) and the Bone Mineral Content (BMC). After the euthanasia of the animals, the tibias were disjointed, dissected and underwent destructive mechanical testing for the analysis of the maximum force (Fmax) and Rigidity (R). The length (L) and the diameter (D) of the middle point of the tibia were also measured. In group S, BMD, BMC, Fmax, R, L and D were smaller than in group C, demonstrating that the suspension by the tail is detrimental to these variables. The treatment of the suspended animals with the US, in group ST, increased the BMD and BMC compared to group S, leveling them to group C...

The bone mineral density BMD and content BMC were analysed in the tibiotarsal bones of ducks in the postnatal development as influenced by age and sex. One hundred birds from the nesting till the slaughtering maturity were included in the experiment. The analysis was conducted using a densitometer, Norland - Excell Plus and pQCT computed tomography, XCT Research SA Plus. The statistical analysis was conducted using the Kruskal-Wallis one-way analysis of variance (age) and the U-Mann -Withney test (sex). All calculations were performed in Statistica 9.0 (StatSoft, Inc. Tulsa, USA), at P < or = 0.05. Two-factor analysis of variance ANOVA was applied and Pearson's correlation coefficients were calculated. The densitometer research showed that BMD and BMC increased in the postnatal development for both sexes. The volumetric bone mineral density vBMD analysis using computed tomography showed that volumetric bone mineral density vBMD of the middle of the diaphyses in situ gradually attenuated during the postnatal development both in males and females, i.e. from 620 mg/cm3 (2 wk) to 500 mg/cm3 (8 wk). The biggest vBMD loss was observed in the diaphyses of females in 4 and 6 wk (r = -0.63 and r = -0.79; P > or = 0.05). The BMC decrease was observed in the proximal metaphyses between 4 and 6 wk for both sexes, r = -0.52 (males), r = -0.53 (females); P < or = 0.05. The gradual loss may be the cause of deformities and fractures of the tibiotarsal bones observed from 4 wk in particular bone sections of both sexes. The achieved results may constitute a helpful source of information for water poultry breeders to prevent limb diseases.

BACKGROUND: Proximal phalanges in horses are among bones that are most prone to injuries. So far, the detailed analysis of densitometric and geometric parameters of both front legs proximal phalanges in horses has not been investigated. The aim of this study was to compare the densitometric and geometric parameters between proximal phalanges in equine both front legs with the use of peripheral quantitative computed tomography (pQCT). METHODS: The study material comprised isolated both front legs proximal phalanges derived from 22 horses. The structure analysis of the proximal phalanges was conducted with the pQCT. The following bone parameters were determined: bone mineral content, volumetric bone mineral density, total bone area, trabecular area, cortical area, cortical thickness, periosteal circumference, endocortical circumference, Strength Strain Index. Tomographic analysis of proximal phalanges was conducted at three levels: at 15%, 50% and 85% of the bone length. RESULTS: The statistical analysis showed that both the densitometric and geometric parameters of the bone at 50% and 85% of its length, did not present any statistically significant differences for the left or right proximal phalanges of the forelimb. At the same time, all examined parameters measured at 15% of the bone length, in the vicinity of the proximal metaphysis revealed significant statistical differences between both front legs proximal phalanges. CONCLUSIONS: The proximal phalanx parameters in the forelimbs are significantly different for the left and right proximal phalanx at 15% of the length and they indicate higher Strength Strain Index of the left bone in this location. The densitometric and geometric parameters of the bone at 50% and 85% of its length, did not present any statistically significant differences for the left or right proximal phalanges of the left and right forelimbs. The most serious changes caused by asymmetrical load of the thoracic limbs in horses occur near the proximal metaphysis, where the spongious substance is most abundant. This may happen because the metabolism of the spongious bone tissue is eight times faster compared to the compact bone tissue. Thus, any changes, including those caused by asymmetrical strain exerted on the right and left thoracic limbs, are the earliest to be observed.

Transferrin receptor 1 (TfR1) expression was measured by immunohistochemistry in 78 archived cases of canine B-cell and T-cell lymphoma with an anti-human TfR1 monoclonal antibody that was validated in the dog by immunohistochemistry and immunoblotting. The canine lymphoma cases were initially classified on the basis of World Health Organization criteria and then subdivided into the following 4 subgroups: low-grade B-cell (LGB), high-grade B-cell (HGB), low-grade T-cell (LGT), and high-grade T-cell (HGT). A visual scoring system and densitometric analysis of the proportion and intensity of positive staining were used to quantify TfR1 expression. TfR1 expression was also correlated to mitotic rate. TfR1 expression was significantly lower in the LGT tumors compared to all other lymphoma subgroups (LGB, HGB, and HGT). LGB tumors showed a TfR1 expression similar to those of the high-grade tumors (HGB and HGT). Significant correlations were found between mitotic rate and densitometric TfR1 variables in the T-cell tumors but not in the B-cell tumors. Further studies are needed to investigate the underlying molecular basis of the high TfR1 expression in LGB lymphomas and its pathological relevance. The anti-human TfR1 monoclonal antibody is a useful tool for measurement of total cellular transferrin receptor expression in the dog; however, an antibody with specificity for the canine TfR1 ectodomain is needed to investigate the potential of this receptor as an oncolytic target.

BACKGROUND: Disease-specific biomarkers hold diagnostic promise in both human and veterinary medicine, but serum biomarkers in low concentrations may be masked by the presence of abundant proteins, mostly albumin and IgG. Methods to deplete albumin and IgG exist, but efficacy of these methods for depleting equine serum of these proteins has not been established. OBJECTIVE: The aim of this study was to determine if albumin and IgG could be depleted from equine serum using several commercially available kits and procedures. METHODS: One-dimensional gel electrophoresis followed by densitometry was used to determine percent of albumin, IgG, and both in pooled serum from 3 horses before and after application of 7 depletion methods. Repeatability was determined by applying the 2 best methods to serum samples from 6 grade horses. RESULTS: For pooled serum, depletion rates varied from 35-90% for albumin and 0-94% for IgG. In the repeatability study, the ProteoExtract method combined with protein G Sepharose beads to remove additional IgG provided the best overall performance with 66% albumin depletion and 100% IgG depletion. A protocol using protein G Sepharose beads to remove IgG followed by ethanol precipitation of nonalbumin proteins with albumin remaining in the supernatant was the second most effective, with 85% albumin depletion and 55% IgG depletion. Although a multiprotein immunodepletion column effectively removed 90% of the albumin, the method was ineffective at removing IgG. CONCLUSION: Albumin and IgG removal kits optimized for human use have variable efficacy for equine serum. Combined use of the ProteoExtract kit and manual incubation with protein G Sepharose beads provided the most effective depletion.

PURPOSE: The present study was carried out to identify and assess the expression levels of toll-like receptors (TLRs) 1-10 mRNA in corneal epithelial cells of buffalo, goat, sheep and bull. MATERIALS AND METHODS: The globes from the respective species were collected and the corneal epithelium was denuded. The expression levels of the different TLR mRNAs were assessed by densitometry of the band intensities following reverse transcriptase polymerase chain reaction (RT-PCR). RESULTS: In all species TLR5 and TLR7 were highly expressed with lower levels of TLR10. The TLR6 mRNA levels were the lowest in the cornea of bull. The TLR2 expression was high in sheep and bulls, while TLR4 mRNA was higher in sheep alone. The level of the other TLRs varied across other species. CONCLUSION: Based on these observations it may be inferred that these animals would be resistant to flagellin (TLR5) and single-stranded RNA viruses (TLR7), while sheep alone may show more resistance to lipo-polysaccharide (TLR4) and peptidoglycan (TLR2)-mediated injury of the cornea. This work reports for the first time the expression of TLR mRNAs in the corneal epithelial cells of farm animals. The relationship of the TLR mRNA levels to corneal disease susceptibilities and pathogenicity remains to be explored further.
