RESUMO

Human dermal exposure to chlorinated paraffins (CPs) has not been well documented. Therefore, hand wipes were collected from four occupational populations to analyze short-chain CPs (SCCPs) and medium-chain CPs (MCCPs) in order to estimate dermal uptake and oral ingestion via hand-to-mouth contact. The total CP levels (∑SCCPs and ∑MCCPs) in wipes ranged from 71.4 to 2310 µg/m2 in security guards, 37.6 to 333 µg/m2 in taxi drivers, 20.8 to 559 µg/m2 in office workers, and 20.9 to 932 µg/m2 in undergraduates, respectively. Security guards exhibited the highest levels of ∑SCCPs among four populations (p < 0.01). In undergraduates engaged in outdoor activities, C13 emerged as the most dominant SCCPs homologue group, followed by C12, C11, and C10. The levels of ∑SCCPs and ∑MCCPs in males in light haze pollution were significantly higher than that in heavy haze pollution (p < 0.05). The median estimated dermal absorption dose of SCCPs and MCCPs via hand was 22.2 and 104 ng (kg of bw)-1 day-1, respectively, approximately 1.5 times the oral ingestion [12.3 and 74.4 ng (kg of bw)-1 day-1], suggesting that hand contact is a significant exposure source to humans.

The main organochlorinated compounds used on agricultural crops are often recalcitrant, affecting nontarget organisms and contaminating rivers or groundwater. Diuron (N-(3,4-dichlorophenyl)-N',N'-dimethylurea) is a chlorinated herbicide widely used in sugarcane plantations. Here, we evaluated the ability of 13 basidiomycete strains of growing in a contaminated culture medium and degrading the xenobiotic. Dissipation rates in culture medium with initial 25 mg/L of diuron ranged from 7.3 to 96.8%, being Pluteus cubensis SXS 320 the most efficient strain, leaving no detectable residues after diuron metabolism. Pycnoporus sanguineus MCA 16 removed 56% of diuron after 40 days of cultivation, producing three metabolites more polar than parental herbicide, two of them identified as being DCPU and DCPMU. Despite of the strong inductive effect of diuron upon laccase synthesis and secretion, the application of crude enzymatic extracts of P. sanguineus did not catalyzed the breakdown of the herbicide in vitro, indicating that diuron biodegradation was not related to this oxidative enzyme.

Metabolic rate parameters estimation using in vitro data is necessary due to numbers of chemicals for which data are needed, trend towards minimizing laboratory animal use, and limited opportunity to collect data in human subjects. We evaluated how well metabolic rate parameters derived from in vitro data predict overall in vivo metabolism for a set of environmental chemicals for which well validated and established methods exist. We compared values of VmaxC derived from in vivo vapor uptake studies with estimates of VmaxC scaled up from in vitro hepatic microsomal metabolism studies for VOCs for which data were available in male F344 rats. For 6 of 7 VOCs, differences between the in vivo and scaled up in vitro VmaxC estimates were less than 2.6-fold. For bromodichloromethane (BDCM), the in vivo derived VmaxC was approximately 4.4-fold higher than the in vitro derived and scaled up VmaxC. The more rapid rate of BDCM metabolism estimated based in vivo studies suggests other factors such as extrahepatic metabolism, binding or other non-specific losses making a significant contribution to overall clearance. Systematic and reliable utilization of scaled up in vitro biotransformation rate parameters in PBPK models will require development of methods to predict cases in which extrahepatic metabolism and binding as well as other factors are likely to be significant contributors.

The objective of this study is to evaluate the influence of placental concentrations of some organochlorine pesticides on newborn's anthropometric measurements (height, weight, head circumference & ponderal index). For this study, we have collected 90 placental tissue samples for the measurement of Dichlorodiphenyltrichloroethane (DDT) & Hexachlocyclohexane (HCH) by Gas Chromatography equipped with Electron Capture Detector (GC-ECD). Regression analysis were performed between outcome variables such as birth weight, birth height, head circumference, ponderal index and independent variable i.e., organochlorine pesticide concentrations. In case of birth weight, there was a substantial decrease of 5.81, 1.94, 4.71, and 2.64 g for 1 ppb (µg/L) increase in placental α-HCH, ß-HCH, γ-HCH, and total HCH concentrations respectively. Significant decrease of 2.02 and 0.43 kg/m3 in ponderal index was found with 1 µg/L increase of total-HCH and total DDT concentrations in placenta. The results of this study support an association between environmental exposure to organochlorine pesticides and anthropometric development of the fetus leading to a significant reduction in its birth weight and ponderal index, slight and non-significant reduction in birth height and head circumference. Efforts should be made to reduce exposure of women of reproductive age in relation to long-term impact on health.

1,3-Dichloropropene (1,3-D) showed a statistically increased incidence of bronchioloalveolar adenomas in male B6C3F1 mice at 60 ppm air concentration during previous chronic inhalation testing. No tumors were observed in female mice, nor in either sex of F344 rats up to 60 ppm, the highest dose tested. Therefore, to understand if lung tumors observed in high dose male mice are due to saturation of metabolic clearance, the linearity of 1,3-D concentrations in mouse blood was investigated on day 15 of repeated nose-only inhalation exposure to 0, 10, 20, 40, 60, 90, and 120 ppm (6 h/d, 7 d/week). Additional groups were included at 20, 60, and 120 ppm for blood collection at 1.5 and 3 h of exposure and up to 25 or 40 min post-exposure to determine area-under-the-curve. The data provide multiple lines of evidence that systemic exposures to 1,3-D in the mouse become nonlinear at inhalation exposure levels of 30 ppm or above. A reduction in minute volume occurred at the highest exposure concentration. The glutathione (GSH)-dependent metabolism of 1,3-D results in significant depletion of GSH at repeated exposure levels of 30 ppm and above. This loss of GSH results in decreased metabolic clearance of this test material, with a concomitant increase of the 1,3-D isomers in circulating blood at exposure concentrations ≥30 ppm. Shifts in the ratio of cis- and trans-1,3-D also support nonlinear toxicokinetics well below 60 ppm. Based on this data, a kinetically derived maximum dose for 1,3-D in mice for repeated exposures should be at or below 30 ppm. These results support non-relevance of 1,3-D-induced benign pulmonary tumorigenicity in mice for human health risk assessment.

OBJECTIVE: Persistent organic pollutants (POPs) are lipophilic environmental toxicants that accumulate in adipose tissue. Weight loss leads to mobilization and increased redistribution of these toxicants. Many are obesogens and endocrine disruptors. Increased exposure could pose long-term health risks. The study objective was to measure the changes in serum concentrations of lipophilic POPs during significant weight loss. METHODS: This study enrolled 27 patients at a university hospital in a longitudinal, 6-month, observational study examining changes in POP blood levels after bariatric surgery. The primary outcome was the changes in the concentrations of 24 polychlorinated biphenyls (PCBs), 9 organochlorine pesticides (OCPs), 11 polybrominated diphenyl ethers, 2,2',4,4',5,5'-hexabromobiphenyl, and 4 perfluorochemicals (PFCs). RESULTS: Older adults (those born before 1976) had baseline levels of PCBs, OCPs, and PFCs that were two- to fivefold higher than younger adults (those born after 1976). Older adults had greater increases in PCBs, OCPs, and polybrominated diphenyl ethers associated with weight loss. Conversely, younger adults had greater increases in PFCs associated with weight loss. On average, blood POP levels increased as weight loss occurred. CONCLUSIONS: Although weight loss is considered beneficial, the release and redistribution of POPs to other lipid-rich organs such as the brain, kidneys, and liver warrant further investigation. Interventions should be considered to limit organ exposure to POPs when weight loss interventions are planned.

PBDEs, HBCD, DBDPE, PBEB and HBB, dechloranes and OPFRs, as well as natural MeO-PBDEs were monitored in muscle tissue of three dolphin species from the southwestern Indian Ocean (Delphinus delphis, Sousa plumbea and Tursiops aduncus) collected between 2012 and 2015. The mean PBDE concentration was 416 ±â€¯333 ng g-1 lw. BDE-47 was found in all samples and was almost half the total PBDE contamination. BDE-209, BDE-100 and BDE-99 were present in ≥85% of the samples. HBCD was detected in just two samples at 20 and 330 ng g-1 lw. PBEB and HBB were not detected, while DBDPE was in all samples but always below its limit of quantification. Dec 602 was the only quantifiable dechlorane at 232 ±â€¯549 ng g-1 lw. Mean OPFR concentration was 10452 ±â€¯11301 ng g-1 lw. TBOEP was found in all samples making up most of the total OPFR contamination. MeO-PBDEs were detected in all samples at 114 ±â€¯137 ng g-1 lw. Data on flame retardants in biota and environmental samples from the southwestern Indian Ocean are scarce and, as a result, comparisons are difficult. However, data from other marine predators in the region, such as penguins, suggest that further studies are needed to determine if these concentrations are the consequence of a high local contamination or widespread thoughout the Indian Ocean.

Concentration of organochlorine pesticides (OCPs) (α-, ß-, γ- hexachlorocyclohexane (HCH), dichlorodiphenyltrichloroethane (DDT), dichlorodiphenyldichloroethane (DDD), dichlorodiphenyldichloroethylene (DDE)) in four species of Pacific salmon (pink, chum, chinook, and sockeye) are presented. OCPs in salmon organs increased in the following order: muscle < liver < eggs < male gonads. Concentrations of the OCP in salmon organs increased in following order: DDE < γ-HCH < α-HCH. The level of pollutants in salmon is compared with the sanitary and epidemiological norms of Russia and other countries. Cancer and noncancer hazard ratios through consumption of salmon in Russian Far East for both men and women also were summarized. Noncancer and cancer hazard ratio values were far below threshold values (<1.0).

Persistent organic pollutants (POPs) include polychlorinated biphenyls (PCBs) dichlorodiphenyltrichloroethane (DDT) and hexachlorobenzene (HCB), which are resistant to biodegradation and therefore accumulate in the marine environment. In Australia, POPs occur in high concentrations primarily in costal water near farming regions and urban centres. From contaminated sediments and biota, POPs are transferred and biomagnified in larger marine organisms. We quantified POPs concentrations in 57 individuals from ten species of sharks and rays caught in bather-protection gillnets deployed off northern New South Wales, Australia. Polychlorinated biphenyls, DDTs and HCB were detected in all species. For some individuals, concentrations were at levels known to have deleterious sub-lethal effects. Overall, the POP concentrations analysed in this study were comparable to those in similar species from more polluted regions, and may have negative impacts on longer-term health. Future research is warranted to investigate spatio-temporal patterns of species-specific contaminant loads and their implications.

Total 22 organochlorine pesticides (OCPs) compounds and 31 polychlorinated biphenyls (PCBs) congeners in mangrove sediments and tissues (leaf, branch, root and fruit) of nine species from three Mangrove Reserves of China were studied. The mean concentrations of total DDTs, HCHs, OCPs and PCBs in sediments were 2.84, 0.06, 3.84 and 0.17 ng g-1 dw, while those in tissues were 1.85, 0.22, 9.43 and 1.61 ng g-1 dw, respectively. The elevated OCPs and PCBs levels in mangrove leaves may be caused by atmospheric sedimentation. The biota sediment accumulation factor (BSAF) values of both OCPs (mean: 3.4) and PCBs (mean: 9.9) are generally larger than one, implying mangroves' bioaccumulation and their ability to intercept pollutants. The BSAFs of PCBs in mangrove tissues were negatively correlated with the PCB congener's octanol-water partition coefficients (KOW, R = 0.58, n = 31, p < 0.001), suggesting that lower chlorinated CB congeners are more bioaccumulative in mangroves. In order to better understanding the fate of these organochlorine compounds, the cycling (including the standing accumulation, the annual absorption, the annual net retention, the annual return, and the turnover period) of OCPs and PCBs in the Mangrove Reserves were estimated, and the results indicated that mangroves are playing important roles in retaining OCPs and PCBs.

Trophodynamics and chiral signatures of organochlorine pesticides (OCPs), including dichlorodiphenyltrichloroethanes (DDTs), hexachlorocyclohexanes (HCHs), and chlordanes in a food web from Zhoushan Fishing Ground, China, were studied. Residues of OCPs in all teleost fishes were within food safety levels. Strong positive correlations were found between trophic levels (TLs) and wet weight concentrations of target chemicals, with trophic magnification factors (TMFs) from 4.17 to 9.77. Lipid contents and TLs significantly correlated, which indirectly affect the bioaccumulation processes of OCPs. The consistently racemic EF values of α-HCH, as well as invariability of the relative proportions of HCH isomers in different marine species implied that HCHs in animals originate directly from the surrounding environment. However, in vivo biotransformation and/or elimination of o,p'-DDT cannot be precluded. TMFs of the individual enantiomers further suggest that the influence of achiral biotransformation is too minor to induce enantioselective biomagnification of chiral OCPs through the studied food web.

Environmental chemicals originating from human activities, such as persistent organic pollutants (POPs), may interfere with the endocrine system of aquatic organisms. The effect of these chemicals on biota and human populations is of high public concern but remains poorly understood, especially in aquatic environments of South America. The aim of this study was to investigate the bioavailability of POPs and the related effects in caged male tilapia (Oreochromis niloticus) in four cascading reservoirs of the Iguaçu River, Southern Brazil. POPs including organochlorine pesticides (OCPs), polychlorinated biphenyl (PCBs), and polybrominated diphenyl ethers (PBDEs) were determined in the reservoir water and tissue samples of tilapia after two months of exposure. The PCB levels in water (14.7 ng L-1) were 14 times higher than the limits permitted by the Brazilian legislation in the Salto Santiago (SS) reservoir. Similarly, concentrations of aldrin and its metabolites (6.05 ng L-1) detected in the water sample of the Salto Osório (SO) reservoir were also above the permitted limits. RT-qPCR analysis revealed different transcript levels of cytochrome P450 enzymes (CYP1A and CYP3A) in the liver among the four groups, with induced activity in tilapia from the SS reservoir. Quantification of the CYP3A mRNA expression and catalytic activity showed higher values for fish caged at the SS reservoir. The fish from this site also had a higher number of eosinophils observed in the testes. Although overt measurements of endocrine disruption were not observed in caged fish, alteration of CYP enzymes with co-occurrence of organochlorine contaminants in water may suggest bioavailability of contaminants from agricultural sources to biota. Additional studies with feral or caged animals for a longer duration may be necessary to evaluate the risks of the waterways to humans and wildlife.

Acetofenate (AF) is a chiral organochlorine pesticide used for controlling hygiene pests. In this study, the metabolism of AF in rabbits in vivo and in vitro was investigated and the primary chiral metabolite acetofenate-alcohol (AF-A) was analyzed. The cytotoxicity of AF and AF-A was also determined. AF in rabbits in vivo was eliminated so rapidly that AF could not be detected within 10min after intravenous administration at 20mg/kg (body weight), and AF-A was quickly formed. In vitro metabolism assay, using plasma and liver microsomes, showed that AF was also quickly metabolized to AF-A and the metabolic process was significantly enantioselective with preferential degradation of (-)-AF and formation of (-)-AF-A. The cytotoxicity of AF and AF-A were investigated by assessing cell proliferation, apoptosis and generation of reactive oxygen species. The results showed that AF and AF-A induce enantioselective cytotoxicity. In addition to eating contaminated prey, sea turtles may be exposed to persistent organic pollutants (POPs) from ingesting plastic debris that has absorbed these chemicals. Given the limited knowledge about POPs in pelagic sea turtles and how plastic ingestion influences POP exposure, our objectives were to: 1) provide baseline contaminant levels of three species of pelagic Pacific sea turtles; and 2) assess trends of contaminant levels in relation to species, sex, length, body condition and capture location. In addition, we hypothesized that if ingesting plastic is a significant source of POP exposure, then the amount of ingested plastic may be correlated to POP concentrations accumulated in fat. To address our objectives we compared POP concentrations in fat samples to previously described amounts of ingested plastic from the same turtles. Fat samples from 25 Pacific pelagic sea turtles [2 loggerhead (Caretta caretta), 6 green (Chelonia mydas) and 17 olive ridley (Lepidochelys olivacea) turtles] were analyzed for 81 polychlorinated biphenyls (PCBs), 20 organochlorine pesticides, and 35 brominated flame-retardants. The olive ridley and loggerhead turtles had higher ΣDDTs (dichlorodiphenyltrichloroethane and metabolites) than ΣPCBs, at a ratio similar to biota measured in the South China Sea and southern California. Green turtles had a ratio close to 1:1. These pelagic turtles had lower POP levels than previously reported in nearshore turtles. POP concentrations were unrelated to the amounts of ingested plastic in olive ridleys, suggesting that their exposure to POPs is mainly through prey. In green turtles, concentrations of ΣPCBs were positively correlated with the number of plastic pieces ingested, but these findings were confounded by covariance with body condition index (BCI). Green turtles with a higher BCI had eaten more plastic and also had higher POPs. Taken together, our findings suggest that sea turtles accumulate most POPs through their prey rather than marine debris.

Twenty-one organochlorine pesticides (OCPs) were measured in the muscle of six predominant waterbird species from Jianghan Plain, Hubei Province, Central China. Among OCPs, DDTs were the most prevalent compounds, with average concentration ranging from 31.1 to 1445 ng/g lipid weight. Little egrets (Egretta garzetta) and Chinese pond herons (Ardeola bacchus) showed significantly higher concentrations of OCPs (p < 0.05) due to their dietary habits and migratory patterns. There were no statistically significant differences (p > 0.05) for most OCPs between sex and age groups. The accumulation profiles of HCHs and DDTs suggested that these OCPs in Jianghan Plain were largely derived from historical usage. Risk assessment indicated that heptachlor could be likely to pose adverse health effects on people consuming ducks in Jianghan Plain.

We examined the degree of exposure of lead (Pb), mercury (Hg), and several organohalogen contaminants and its potential impact on survival of bald eagles in Ontario from 1991 to 2008. Overall, results for 43 dead or dying bald eagles collected in the province indicate that 23% (10/43) of birds died of Pb poisoning and 9% (4/43) died of suspected Hg poisoning. Pb poisoning was diagnosed based on exceedances of toxicity thresholds in liver and kidney and supported by clinical observations, necropsy results, and histology findings when available. Evidence for Hg poisoning in eagles was limited; however, Hg concentrations exceeded the toxicity threshold in kidney. Pb concentrations ranged widely in liver and kidney. Total Hg concentrations were relatively higher in kidney compared with liver and were significantly correlated with selenium (Se) concentrations in both tissues. Concentrations of p,p'-DDE and sum PCBs in livers of 12 bald eagles collected from 2001 to 2004 were likely below concentrations associated with adverse effects. Hepatic concentrations of total polybrominated diphenyl ethers were generally higher in birds collected from southern Ontario compared with northern Ontario. Potential impacts of exposure to these flame retardants and others are not known. Elevated metal exposure appears to influence survivorship and may affect the recovery of bald eagles in the province, particularly in southern Ontario and along the Great Lakes where a disproportionate number of poisoned eagles were collected. Increased efforts are needed to identify sources of exposure and develop measures to reduce metal exposure in this top predator.

Vector-borne diseases have increased pesticide use in urban areas (UA) and agricultural areas (AA) in Mexico. Breast milk can be contaminated by pesticide exposure. The objective of the study was to measure breast milk intake by deuterium oxide dilution as well as organochlorine and pyrethroid transfer from mother to infant in AA and UA of Sonora, Mexico. Human milk intake was determined by the 'dose-to-mother' technique using deuterium oxide (D2O) dilution. Mothers' body composition was also assessed by this technique and the intercept method. Pyrethroids (deltamethrin, cypermethrin and cyhalothrin) and organochlorine pesticide residues (p,p'- DDT, p,p'- DDE, p,p'- DDD) in breast milk samples were measured by gas chromatography. Sixty-two lactating women and their infants participated in the study, 32 lived in the UA and 30 lived in the AA. Breast milk intake was approximately 100 mL higher in the AA than in the UA 799 ± 193 and 707 ± 201 mL/day, respectively (p < 0.05). The concentrations of p,p'- DDT and cypermethrin levels in breast milk were higher in the UA than in the AA (p < 0.05 and p = 0.001, respectively). None of the pyrethroids and organochlorine pesticides studied surpassed the Acceptable Daily Intake (ADI) in milk for humans according to EPA and FAO/WHO. In conclusion, breast milk intake was higher in the AA compared to the UA. The p,p'- DDT and cypermethrin levels in breast milk were higher in the UA compared to the AA. Since pesticide levels in human milk did not exceed the ADI, breastfeeding is still a safe practice and should be encouraged.

Organochlorines were determined in fish and sediment collected from Izmir and Çandarli Bays. The results indicated that ΣCyclodiens were generally predominant contaminants. In all samples, p,p'-DDE was the predominant DDT congener. Aroclors were found in noticeably higher levels than OCPs in sediment and the highest levels of Aroclors, OCPs were found in Nemrut which can be attributed to industrial activities. According to Sediment Quality Guidelines, DDTs were lower than the values that may cause adverse biological risk in sediment samples. Aroclor 1254 in sediments only exceeded the TEL value at Nemrut site. The maximum values of ΣOCPs were found in fish collected from Gülbahçe, while Aroclors were measured in Aliaga. According to related indices, results indicate no recent influxes of DDT in the sampling areas. The estimated daily intake of DDTs, Aroclor1254 were below the acceptable daily intake level recommended by FAO/WHO.

Organochlorines and carbamates are common pesticides predominantly employed in agriculture. Large amounts of pesticides make their way into rivers and marine habitats. They accumulate in aquatic organisms through different exposure routes and gradually move up the food chain. Since contaminant bioaccumulation in animals is affected by several factors, this work harnessed several different approaches to explore the persistence of lindane, a long banned organochlorine pesticide, and carbaryl, a newer generation pesticide, in common sole (Solea solea), a major commercial species in Adriatic fisheries. Lindane was not only more accumulated than carbaryl in sole liver, but it was also detected in greater amount in muscle tissue, the edible part (lindane, 7 ± 4 ng/g; carbaryl, <0.004 ng/g w/w). Additional assays documented a greater accumulation of lindane in adults compared with juveniles and in specimens caught offshore than in those collected close to the coast. The present findings demonstrate the different accumulation dynamics of the two pesticides to confirm the benefits derived from the replacement of organochlorine pesticides with carbamate compounds.

Background. Many individuals have been exposed to organochlorinated pesticides (OCPs) through food, water, air, dermal exposure, and/or vertical transmission. Due to enterohepatic reabsorption and affinity to adipose tissue, OCPs are not efficiently eliminated from the human body and may accrue in tissues. Many epidemiological studies demonstrate significant exposure-disease relationships suggesting OCPs can alter metabolic function and potentially lead to illness. There is limited study of interventions to facilitate OCP elimination from the human body. This study explored the efficacy of induced perspiration as a means to eliminate OCPs. Methods. Blood, urine, and sweat (BUS) were collected from 20 individuals. Analysis of 23 OCPs was performed using dual-column gas chromatography with electron-capture detectors. Results. Various OCPs and metabolites, including DDT, DDE, methoxychlor, endrin, and endosulfan sulfate, were excreted into perspiration. Generally, sweat samples showed more frequent OCP detection than serum or urine analysis. Many OCPs were not readily detected in blood testing while still being excreted and identified in sweat. No direct correlation was found among OCP concentrations in the blood, urine, or sweat compartments. Conclusions. Sweat analysis may be useful in detecting some accrued OCPs not found in regular serum testing. Induced perspiration may be a viable clinical tool for eliminating some OCPs.
