RESUMO

Complex mixtures of plant derived triglycerol (TG) lipids are commonly used as feedstock components for the production of industrial polymers. However, there remains a need for the development of analytical strategies to investigate the intrinsic intermolecular cross-linking reactivity of individual TG molecules within these mixtures as a function of their structures and physicochemical properties, and for the characterization of the resultant products. Here, to address this need, we describe a novel multistage tandem mass spectrometry based method for intermolecular cross-linking and subsequent structural characterization of TG lipid ions in the gas phase. Cross-linking reactions were initiated using 266 nm ultraviolet photodissociation tandem mass spectrometry (UVPD-MS/MS) of saturated or unsaturated TG dimers introduced via electrospray ionization into a linear ion trap mass spectrometer as noncovalent complexes with protonated 3,4-, 2,4- or 3,5- diiodoaniline (diIA). UVPD resulted in the initial formation of an anilinyl biradical via the sequential loss of two iodine radicals, which underwent further reaction to yield multiple cross-linked TG products along with competing noncross-linking processes. These chemistries are proposed to occur via sequential combinations of hydrogen abstraction (H-abstraction), radical addition and radical recombination. Multistage collision induced dissociation tandem mass spectrometry (CID-MS(n)) was used to obtain evidence for the structures and mechanisms of formation for these products, as a function of both the TG lipid and diIA ion structures. The efficiency of the UVPD reaction was shown to be dependent on the number of unsaturation sites present within the TG lipids. However, when unsaturation sites were present, formation of the cross-linked and noncross-linked product ions via H-abstraction and radical addition mechanisms was found to be competitive. Finally, the identity of the anilinyl biradical (e.g., 3,4- versus 2,4-substituted) was found to significantly affect the distribution of these two types of product ions. Importantly, owing to the observed propensity for cross-linking to occur via H-abstraction-initiated processes, this novel gas-phase cross-linking reaction provides a convenient method to link two molecules covalently without the requirement of any specific functional group, and therefore could be applied to examine the gas-phase intermolecular interactions and cross-linking of a wide range of biomolecular classes.

RESUMO

The impact of intravenous laser irradiation of blood with green laser in patients with hyperlipidemia was investigated. The blood of patients was chosen as sample for analysis. The patients were divided in two groups: patients with atherosclerosis of various localization and patients with atherosclerosis associated with diabetes mellitus. The effectiveness of laser impact was evaluated according the blood biochemical indicators. The levels of crude cholesterol, triglycerides, low and very low density lipoproteins, apoproteins A and B, highly sensitive C-reactive protein, atherogenity indicator, glucose content, uric acid content were determined befor and after 1, 3 and 6 months after impact. The study results indicate the occurrence of hypolipedemic and hypoglycemic effects.

RESUMO

The present work aims at developing a new process to synthesize HMX from DAPT using ultrasound in ionic liquid. Reaction has been carried out in ultrasonic bath, effect of various parameters such as presence and absence of ultrasound, volume and type of solvent, temperature, concentration of nitrating agent has been investigated with an aim of obtaining the optimum conditions for the synthesis of HMX. It was observed that ultrasonically promoted nitroylsis of DAPT to HMX has exhibited significant enhancement in yield at ambient condition.

The aim of this study was to explore the relationship between local ischaemic preconditioning and the effectiveness of fractionated radiotherapy. The rat serum, bone marrow, and small intestine were examined for oxidative changes induced by total body irradiation with gamma rays with applied local ischaemic preconditioning immediately before irradiation. Serum concentrations of TBA-RS examined 12 hours after the last irradiation did not reveal any differences among the groups of animals analyzed. Twenty-four hours after the last dose of irradiation, the serum concentrations of TBA-RS varied in particular groups (P<0.0001). The concentration of triglycerides in the serum of local preconditioned ischaemia and irradiated animals showed a reversed shape similar to the TBA-RS fluctuation (P<0.003). The level of uric acid in the serum of animals treated only with radiation is slightly higher than the level of this acid in the serum of the local preconditioned ischaemia radiation group (P<0.58). The number of bone marrow polychromatic erythrocytes did not appear to differ substantially in both irradiated groups. At the first 12 hours after irradiation, the frequency of micronucleated polychromatic erythrocytes is significantly different in the bone marrow of both groups either in combination with ischaemic preconditioned radiation or with radiation alone (P<0.0002). In irradiated animals without ischaemic preconditioning, on the 3rd day after irradiation the number of crypts increased and in the next days decreased achieving the level of the control group on the 7th day. Irradiated rats with local ischaemic preconditioning did not reveal an increase in the number of crypts. The difference was statistically significant (P<0.05). These data indicate that the local ischaemic preconditioning modifies the radiation peroxidising effects through inhibition of free radical-dependent lipid peroxidation and, probably, other unrecognized mechanisms.

Pharmacological and cytogenetic evaluations of the protective effects of polyethoxylated castor oil cremophor-EL (cremophor) against hepato, renal and bone marrow toxicity induced by gamma irradiation in normal rats were carried out. A single dose of irradiation (6 Gy) caused hepatic and renal damage manifested biochemically as an elevation in levels of serum alanine and aspartate aminotransferase as well as an increase in blood urea. Cremophor administration at a dose level of 50 microl kg-1 intravenously 1 day before exposure to irradiation (6 Gy) protected the liver and kidney as indicated by the recovery of levels of hepatic aminotransferase, urea and lipid profiles to normal values. Gamma irradiation of male rats caused a decrease in reduced glutathione and an increase in the oxidized form in rat-liver homogenate. A highly significant increase in the incidence of micronucleated normochromatic erythrocytes and micronucleated polychromatic erythrocytes was observed after irradiation exposure. The induced genotoxicity in the bone marrow cells was corrected by pretreatment with cremophor. The findings of this study suggest that cremophor pretreatment can potentially be used clinically to prevent irradiation-induced hepato, renal and bone marrow toxicity without interference with its cytotoxic activity.

Primary radicals were generated by UV photolysis of samples of trilinolein, at 77 K and under a controlled atmosphere. The resulting EPR spectra clearly show that the amount of radicals is dependent on the purity of the lipid, the exposure to visible light in the presence of a photosensitizer and oxygen, and, finally, the presence of an antioxidant. These solid state EPR experiments indicate that if all of the elements for the production of singlet oxygen (Rose Bengal, molecular oxygen, and visible light) are not present, primary radicals are practically not generated. They also point out the various steps of the oxidation mechanism: formation of singlet oxygen, which reacts with the lipid to form a hydroperoxide; and photolytic formation of the hydroxyl radical, which reacts with the frozen lipid to generate primary lipidic radicals. This constitutes a new method for investigating lipid oxidation and studying the influence of photosensitizers and molecules that are likely to react with singlet oxygen.

The aim of this study was to design a convenient, specific, sensitive, and continuous lipase activity assay using natural long-chain triacylglycerols (TAGs). Oil was extracted from Parinari glaberrimum seed kernels and the purified TAGs were used as a substrate for detecting low levels of lipase activities. The purified TAGs are naturally fluorescent because more than half of the fatty acids from Parinari oil are known to contain 9,11,13, 15-octadecatetraenoic acid (parinaric acid) in its esterified form. The presence of detergents (sodium taurodeoxycholate, CHAPS, Sulfobetaine SB12, Tween 20, Brij 35, Dobanol, n-dodecylglucoside) above their critical micellar concentration dramatically increases the fluorescence of the parinaric acid released by various lipases. This increase in the fluorescence intensity is linear with time and proportional to the amount of lipase added. This new method, performed under non-oxidative conditions, was applied successfully to detecting low lipase levels in crude protein extracts from plant seeds and could be scaled down to microtiterplate measurements. Quantities as low as 0.1 ng of pure pancreatic lipase could be detected under standard conditions (pH 8). Lipase activity can also be assayed in acidic media (pH 5) using human gastric lipase. This simple and continuous assay is compatible with a high sample throughput and might be applied to detecting true lipase activities in various biological samples.

Continuous irradiation of experimental animals is an appropriate model for the research in space radiobiology. The onset and recovery of radiation injury can be estimated on the basis of the concentration/content of glycogen in liver, the phospholipid content in thymus and other radiosensitive organs and the triacylglycerol concentration in bone marrow. Further, the picture of the metabolism in irradiated organism may be completed by the analysis of serum glucocorticoid and thyroid hormone levels.

RESUMO

A study was made of the influence of gamma-irradiation of rats (14.4, 9.6, 7.2 and 4.8 Gy) on the number of nucleate cells, and the concentration of triacylglycerides (TG) and phospholipids (PL). Cellularity and concentration of PL decreased while that of TG increased under the effect of radiation. The degree of the changes in the above parameters was a function of dose and time after irradiation.

The method of low-temperature ESR-spectroscopy was used to study a modifying effect of cystamine on the yield of radiation-induced free radicals in different biomolecules of liver and spleen tissues of mice. Intraperitoneal administration of cystamine (150 mg/kg) 15 min before isolation and freezing of the tissues was shown to reduce by 11 per cent the yield of radicals of H-adducts of thymine DNA bases, to decrease by 23 per cent the yield of radicals of triacyglycerol and phospholipid radiolysis, and to increase by 24 per cent the yield of radicals of lipid fatty acid residues in splenic tissue. According to the criterion used, cystamine has no modifying action on the yield of free-radical damages to liver biomolecules.
