RESUMO

Both Curcuma longa (CL) and Citrus junos Tanaka (CJ) have been used to treat various diseases due to their anti-inflammatory and antioxidative stress activities. In this study, we investigated the ameliorative effect of the combination of CL extract and CJ extract (CCC) against beta-amyloid (Aß) peptide-induced neurological damage. CCC prevented neurocytotoxicity in vitro. In addition, it was confirmed that abnormal alternation behavior and memory impairment caused by Aß peptide were reversed by treatment with CCC. Furthermore, CCC treatment led to recovery of the cholinergic system and reactive oxygen species (ROS) oxidative damage defense system. CCC induced expressions of cyclic-adenosine monophosphate response element-binding protein (CREB)-responsive element-binding protein and brain-derived neurotrophic factor were confirmed as was the significantly improved processing in the hippocampus of the mouse Aß peptides. Accordingly, these results suggest that CCC can prevent and/or reverse neurocytotoxicity and cognitive deficits in a mouse model of Alzheimer's disease.

Hypoxia frequently occurs in postharvest logistics, which greatly influences fruit storability. Here, we for the first time studied the dynamic variations of mitochondrial morphology in living citrus fruit cells, and revealed that waxing treatment-induced hypoxia strongly triggered mitochondrial fission and fragmentation. Correspondingly, hypoxia caused a decline in mitochondrial membrane potential and mobility. Besides, impairment of energetic and redox status was also found in waxed fruit. The proteomic changes of mitochondria after waxing treatment were also characterized. Using weighted gene co-expression network analysis (WGCNA), we identified 167 key hypoxia-responsive proteins, which were mainly involved in fatty acid, amino acid and organic acid metabolism. Metabolite analysis verified that waxing treatment promoted the accumulation of several hypoxic metabolites, such as ethanol, acetaldehyde, succinic acid and γ-aminobutyric acid (GABA). Taken together, our findings provide new insights into the cytological and proteomic responses of mitochondria to hypoxia during fruit storage.

Two Gram-positive, rod-shaped, motile, endospore-forming strains, FJAT-49780T and FJAT-49732T were isolated from a citrus rhizosphere soil sample. The optimal growth temperatures for strains FJAT-49780T and FJAT-49732T were 45 and 35-40 °C, respectively. The optimal growth pH for strains FJAT-49732T and FJAT-49780T were pH 8.0 and pH 6.0, respectively. The 16S rRNA gene sequence similarity between FJAT-49780T and FJAT-49732T was 98.6%. Strains FJAT-49780T and FJAT-49732T shared 97.9-98.4% 16S rRNA gene sequence similarities to the type strain of Lederbergia wuyishanensis. In phylogenetic trees (based on 16S rRNA gene sequence), strains FJAT-49732T and FJAT-49780T clade with Lederbergia members. Both strains contained meso-diaminopimelic acid in their cell-wall peptidoglycan and MK-7 was the only isoprenoid quinone detected. The major fatty acids of strains FJAT-49732T and FJAT-49780T were anteiso-C15:0 and iso-C15:0. The polar lipids of strain FJAT-49780T were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, an unidentified aminophospholipid, unidentified phospholipid and unidentified lipids while strain FJAT-49732T contained diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, an unidentified glycolipid, unidentified aminolipid and unidentified phospholipid. The genomic DNA G+C content of strains FJAT-49780T and FJAT-49732T were 37.0 and 36.7%, respectively. The digital DNA-DNA hybridization and average nucleotide identity values between strains FJAT-49780T and FJAT-49732T and with other members of the genus Lederbergia were below the cut-off level for species delineation. Thus, based on the above results, strains FJAT-49780T and FJAT-49732T represent two novel species of the genus Lederbergia, for which the names Lederbergia citri sp. nov., and Lederbergia citrisecundus sp. nov., are proposed. The type strains are FJAT-49780T (= CCTCC AB 2019242T = LMG 31583T) and FJAT-49732T (= CCTCC AB 2019246T = LMG 31584T).

The reflectance NIR spectroscopy and chemometric data treatment on mature intact lemons, Limone di Sorrento PGI (cv Ovale di Sorrento) and Limone Costa D'Amalfi PGI (cv Sfusato Amalfitano) from Campania region, collected in 2018 and 2019, were used to predict properties, and discriminate cultivar and geographical provenance. By PCA, lemon NIR spectra grouped for production years due to the year variation of lemon properties attributable to annual climatic differences, homogeneous in all sites. This agrees with lemon chemical and physical differences by production year. Consequently, the relationship of NIR spectra with lemon quality properties by MLR and the cultivar and provenances discrimination by LDA were affected by year climatic difference; therefore, better model reliability was for single production year. NIR detectability of lemon properties did not appear beyond lemon thick peels, therefore the measured properties of lemon juices could derive from measurable properties of peel correlating with pulp properties.

Penicillium oxalicum has been used as a biocontrol fungus in agriculture for many years, but the antimicrobial substances are still uncertain. Herein, we isolated a linear peptide named Sanxiapeptin in the culture broth of Penicillium oxalicum SG-4 collecting from the Three Gorges riparian zone. Sanxiapeptin exhibited potent inhibitory effect on citrus green mold Penicillium digitatum, the main fungi responsible for postharvest decay. Sanxiapeptin was elucidated as composing of five amino acids, which were ß-amino-α-methoxybutyric acid (Amoba), N-Me-l-Thr, d-Thr, N-Me-l-Val and l-Ser. By analyzing three chemically synthesized oligopeptides with similar structures, we found that the first amino acid of Amoba was crucial to the antifungal activity, as was the methylation of peptide bond. Sanxiapeptin may act as an antimicrobial agent by affecting the function of cell membranes or walls. The antimicrobial spectrum, safety and stability analysis supported that Sanxiapeptin was a promising antifungal agent for citrus preservation.

Continual input of neonicotinoid insecticides occurs in the citrus orchards from southern China. However, it is still unknown about the variations in the distribution and accumulation of neonicotinoids in soil profiles along a long-term chronosequence of cultivation and the driving factors contributing to these shifts. Here, changes of neonicotinoids in the 0-100 cm soil profiles with distinct orchard cultivation age (1, 10, and 20 years) were investigated, and their related factors were further determined. The results showed that the total levels of five target neonicotinoids (∑5NEOs) in the soil profiles were in the range of 0-25.76 ng/g dw. Imidacloprid was the most dominating neonicotinoid, followed by thiamethoxam. We observed higher neonicotinoid accumulations in the soil profiles from the citrus orchards after 10 and 20 years of cultivation. Neonicotinoids migrated deeper into the soil profiles in orchards with a longer time since cultivation. Imidacloprid, thiamethoxam, and the total amount of neonicotinoid (∑5NEOs) were mainly affected by the cultivation age of citrus orchards (accounting for 58.9% variance; P < 0.001); whereas clothianidin, acetamiprid, and thiacloprid were mainly influenced by soil depths (accounting for 66.9-85.2% variance; P < 0.05). Redundancy analyzes further indicated that the enhanced accumulation of neonicotinoids was mainly correlated with the increase of soil organic carbon (SOC) content and soil porosity, and the reduction of bulk density in the profiles of citrus orchards with increasing cultivation age. This study highlights the finding that we should give more concerns about the contamination and ecological risks of neonicotinoids in the orchards with a long cultivation age.

The Citrus genus is a good source of dietary flavonoids, which have many health benefits. As a representative citrus fruit, the flavonoids composition in Shatianyu (Citrus grandis L. Osbeck) pulp remains to be investigated. In the present study, 11 flavonoids were isolated and identified from Shatianyu pulp flavonoid extracts (SPFEs). Among them, 4 flavonoids were previously undescribed and 2 flavonoids were firstly isolated from pummelo. The cellular antioxidant activity (CAA) and oxygen radical absorbance capacity (ORAC) of isolated compounds were evaluated. Naringin and rhoifolin showed the highest ORAC activity, and the presence of a 3-hydroxy-3-methylglutaryl or a 4'-glucose decreased the ORAC activity of flavonoids. The contribution of isolated flavonoids to the holistic antioxidant activity of SPFEs was determined by an online knockout method. Melitidin, bergamjuicin and naringin contributed most to ORAC activity, while bergamjuicin, melitidin and apigenin-4'-O-ß-d-glucopyranosyl-7-O-α-l-rhamnopyranosyl-(1 â†’ 2)-[6″-O-(3- hydroxy-3-methylgltaryl)]-ß-d-glucopyranoside contributed most to CAA activity.

In this study, tailored-made citrus pectin-derived compounds were produced through controlled enzymatic and/or chemical modifications of commercial citrus pectin with different degrees of methylesterification (DM) and similar average molecular weight (MW). In the first treatment, degradation of the citrus pectin (CP) materials by endo-polygalacturonase (EPG) yielded pectins with average Mw's (between 2 and 60 kDa). Separation and identification of the oligosaccharide fraction present in these samples, revealed the presence of non-methylesterified galacturonic acid oligomers with degree of polymerization (DP) 1-5. In the second treatment, exploiting the combined effect of EPG and pectin lyase, compounds with MW between 2 and 21 kDa, containing methylesterified and non-methylesterified polygalacturonans (DP 1-6), were generated. Finally, CP was sequentially modified by chemical saponification and the action of EPG. A sample of DM 11% and MW 2.7 kDa, containing POS (DP 1-5), was produced. Diverse pectin-derived compounds were successfully generated for further studies exploring their functionality.

This study investigated the effect of arginine (Arg) on the antagonistic activity of Metschnikowia citriensis against sour rot caused by Geotrichum citri-aurantii in postharvest citrus, and evaluated the possible mechanism therein. Arg treatment up-regulated the PUL genes expression, and significantly induced the pulcherriminic acid (PA) production of M. citriensis, which related to the capability of iron depletion of M. citriensis. By comparing the biocontrol effects of Arg-treated and untreated yeast cells, it was found that Arg treatment significantly enhanced the biocontrol efficacy of M. citriensis, and 5 mmol L-1 Arg exerted the best effect. Additionally, the biofilm formation ability of M. citriensis was greatly enhanced by Arg, and the higher population density of yeast cells in citrus wounds was also observed in Arg treatment groups stored both at 25 °C and 4 °C. Moreover, Arg was shown to function as a cell protectant to elevate antioxidant enzyme activity [including catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPX)] and intracellular trehalose content to resist oxidative stress damage, that directly helped to enhance colonization ability of yeasts in fruit wounds. These results suggest the application of Arg is a useful approach to improve the biocontrol performance of M. citriensis.

To maintain the viability of citrus farms in a scenario of scarce water supplies for irrigation purposes, such as in south-eastern Spain, it is necessary to detect the maximum threshold of crop water status and vegetative growth, both of which are vulnerable to the combined effect of water and salinity stress. With this aim, adult late mandarin trees were subjected to a gradual increase in salinity of the irrigation water for four consecutive growing seasons, accompanied by a water deficit for the first three seasons, while the fourth season counted as a recovery period. The treatments tested were: i) control (CTL), irrigated at 100% of the ETc in the first season followed by an additional 33% water leaching requirement in the second and third seasons; (ii) a regulated deficit irrigation (RDI100) treatment, irrigated as the CTL except in phase I and until mid-phase II, coinciding with the beginning of the slowdown in trunk growth, where irrigation was set at 65 and 50% CTL in the first and last two seasons, respectively, and 80% in phase III; (iii) RDI50, irrigated as the CTL during the first season, and from the second season onwards, at 50% RDI100 during the deficit period; (iv) FARMER, irrigated in accordance with the farmer's criteria. A moderate saline water stress combined with the application of RDI100 treatment did not reduce crop yield after one growing season, but did reduce plant growth. However, after two consecutive seasons (in both RDI strategies) water stress drastically reduced both yield and plant growth, mainly due to a high accumulation of sodium and chloride in the leaves. Gas exchange parameters were also greatly reduced. Furthermore, it was found that the water deficit could be prolonged until the trunk growth slowed down during the second fruit growth stage in mandarin trees, leading to a substantial increase in water productivity values.

Tibet's native fruit resources have not been explored and remain underutilized due to geographical isolation. "Medog lemon" (ML), an ethno-species of wild citron, is an important functional food and dietary resource for the local people in southeastern Tibet. Herein comprehensive characterization of components of ML were firstly performed with an integrated strategy based on UHPLC-QE Orbitrap MS. A total of 196 components were characterized, among which 33 were detected from Citrus genus for the first time, and 55 were identified as potential new phytochemicals. The chemical comparison of ML with cultivated citron and fingered citron based on partial least squares-discriminant analysis model and univariate analysis revealed the distinct chemical composition of ML and in which more than 30 differentiated components were identified. The distinct morphological and chemical characters, as well as its good antioxidant properties, indicated ML as a potential new food resource that can be beneficial for human health.

Citri reticulatae pericarpium (CRP) shows multiple bioactivities, including antioxidant, anti-tumor, and anti-inflammation. The folk proverb "CRP, the older, the better" means storing for longer time would improve its quality, which attributed to the influence of bioactive compounds. The aim of this work was to study which compounds are the factors that long storage would influence the quality of CRP. 161 compounds, including 65 flavonoids, 51 phenolic acids, 27 fatty acids, and 18 amino acids were identified through derivatization and non-derivatization liquid chromatography mass spectrometry approaches. Their dynamic changes indicated phenolic acids, which were reported to have various activities, were the main increased components. Furthermore, the representative phenolic acids were quantified and correlation analysis between their contents and antioxidant activity implicated they were the possible indicators that long storage would improve CRP quality. The results would provide basis for quality control of CRP during storage.

Citrus fruits are products of great market values, as used by the juice industry in huge quantities. The juice industry processes millions of tons of citrus fruits per year, but only the pulp is utilized, whereas peels, seeds, and membrane residues are mostly discarded. This generates vast amounts of byproducts (>100 million tons/year), since the peel can make up to 50% of the weight of the fresh fruit. Phytochemical investigations showed that citrus peels are great sources of bioactive compounds, e.g., phenolic compounds, carotenoids, and monoterpenes. These compounds could find numerous applications in the food, cosmetics, and pharmaceutical industries. The recovery of the phytochemicals would provide economic and environmental benefits. Researchers worldwide have developed innovative techniques to recover phytochemicals from the citrus waste, by endorsing the international waste-prevention policies. This chapter reviews the advances in the sector of food technology applied to citrus chemistry and describes the available green techniques that allow the recovery of phytochemicals from citrus byproducts.

Most perennial crops sensitive to water scarcity, such as citrus, can benefit from efficient water management, which allows for reduced water consumption while increasing crop production on a long-term basis. However, when implementing water-saving strategies, it is necessary to monitor soil and/or plant water status in order to determine crop water demand. A plethora of devices providing indirect measurements of volumetric soil water content, such as the "drill and drop" multi-sensors probes (Sentek, Inc., Stepney, Australia), have been developed over the last decade. The objective of the paper was to analyse time-series of soil water content profiles and meteorological data collected in an adult citrus orchard over three years of field observations (2017-2020) in order to estimate actual crop evapotranspiration and derive crop coefficients. Simultaneous measurements of sap fluxes also allowed for the estimation of the basal crop coefficient, Kcb, used as a control variable. The temporal dynamics of soil water content profiles following rainfall or irrigation events provided information on soil evaporation, root water uptake, and actual crop transpiration. After soil wetting events, in particular, it was possible to recognize patterns of actual crop evapotranspiration similar to those detected with sap flow sensors. The knowledge of actual crop evapotranspiration at the daily time-step, in conjunction with the corresponding reference crop evapotranspiration, allowed for appropriate estimations of the crop coefficient associated with the various development stages. The proposed method provided interesting insights into the dynamics of root water uptake and crop evapotranspiration of the studied citrus orchard, and it represents a promising tool for precise irrigation scheduling in other agroecosystems.

This study evaluated the micro-morphology as well as the chemical and biological features of Citrus lumia seeds. The cream-colored pyriform seed showed a woody coat covered by a thick layer of mucilage and an embryo with two large cotyledons rich in oil bodies. Hydroxycinnamic acid glycosides and flavonoids are the most abundant compounds in methanol and ethyl acetate extracts (ME and EAE), respectively. Conversely, fatty acids and α-tocopherol represent the main bioactive compounds in the hexane extract (HE). ME showed the most promising antioxidant and anti-inflammatory activities already in cell-free assays. These results were confirmed by experiments carried out on human primary cells. Indeed, ME showed the best inhibitory activity against heat-induced haemolysis and ROS formation in erythrocytes. Moreover, the same order of potency (ME > EAE > HE) was observed also on peripheral blood mononuclear cells, in which the seed extracts were able to decrease TNF-α and IL-6 release after LPS-induced inflammation.

RESUMO

Quantitative polymerase chain reaction (qPCR) and reverse transcription (RT)-qPCR have now become the gold standard for molecular diagnostics because of its sensitivity, specificity, and reproducibility. In addition, qPCR diagnostics are flexible because they can be scaled for high- or low-throughput applications. Here we describe an optimized assay and workflow for the universal detection of eight citrus viroid species and their variants by RT-qPCR. The assay allows for quick and efficient molecular detection of viroids without the need to run RT-qPCR for each individual viroid species.

BACKGROUND: Two edible coating (EC) emulsions based on potato starch (F6 and F10) alone or formulated with sodium benzoate (SB, 2% w/w) (F6/SB and F10/SB) were evaluated to maintain postharvest quality of cold-stored 'Fino' lemons and control sour rot on lemons artificially inoculated with Geotrichum citri-aurantii. Previous research showed the potential of these ECs to improve the storability of 'Orri' mandarins and reduce citrus green and blue molds caused by Penicillum digitatum and Penicillium italicum, respectively. RESULTS: The coatings F6/SB and F10/SB significantly reduced sour rot incidence and severity compared to uncoated control samples on lemons incubated at 28 °C for 4 and 7 days. The F6/SB coating reduced weight loss and gas exchange compared to uncoated fruit after 2 and 4 weeks of storage at 12 °C plus a shelf life of 1 week at 20 °C, without adversely affecting the lemon physicochemical quality. CONCLUSION: Overall, the F6/SB coating formulation, composed of pregelatinized potato starch, glyceryl monostearate, glycerol, emulsifiers and SB, with a total solid content of 5.5%, showed the best results in reducing citrus sour rot and maintaining the postharvest quality of cold-stored 'Fino' lemons. Therefore, it showed potential as a new cost-effective postharvest treatment suitable to be included in integrated disease management programs for citrus international markets with zero tolerance to chemical residues. © 2021 Society of Chemical Industry.

This work investigated the effects of feeding ensiled bergamot pulp to pigs on meat and salami quality. Eighteen pigs were assigned to two experimental treatments and fed a cereal-based concentrate diet (control) or the same diet in which ensiled bergamot pulp replaced 15% dry matter of the diet fed (BP). The BP treatment increased α-linolenic acid (+250%; P < 0.05), docosapentaenoic acid (+62%; P < 0.05), docosahexaenoic acid (+43%; P < 0.05) and consequently n-3 PUFA (+15%; P < 0.01) in meat. In salami, the content of α-linolenic acid, total PUFA and n-3 PUFA increased (+320%, +25% and + 258%, respectively) by feeding the BP diet (P < 0.001). The inclusion of bergamot pulp in the diet did not alter the oxidative stability in raw and cooked meat and colour descriptors. In salami, TBARS values were lower after 5 days of storage (P < 0.001) in BP group (1.54 vs 2.96). Finally, dietary supplementation with ensiled bergamot pulp to pigs improved the nutritional value of meat and meat products.
