Gelatina de tilápia (Oreochromis niloticus) e astaxantina de camarão branco (Litopenaeus vannamei) na rota de valorização de resíduos do pescado: obtenção de filmes antioxidantes e antimicrobianos

RESUMO
O crescimento da produção aquícola causa aumento da quantidade de resíduos gerados a partir do beneficiamento dessa matéria-prima. O que pode gerar problemas ambientais, caso não seja dado um destino adequado. Além disso, indústrias de alimentos e embalagens têm se esforçado para reduzir a quantidade de materiais não biodegradáveis em embalagem, que são uma fonte de poluição e podem causar problemas ambientais relacionados ao seu tratamento após descarte. Paralelamente, a indústria procura novas estratégias para aumentar o tempo de prateleira dos alimentos. Nesse contexto, tem-se a produção de filmes comestíveis e/ou biodegradáveis a partir de compostos naturais. O presente estudo teve por objetivos extrair e caracterizar gelatina a partir da pele de tilápia do Nilo (O. niloticus), astaxantina extraída a partir do cefalotórax do camarão branco do Pacífico (L. vannamei) e aplicar em filmes comestíveis. A gelatina tipo A foi extraída por meio do pré-tratamento ácido e caracterizada quanto ao rendimento, distribuição do peso molecular, FTIR, força do gel e TGA. O extrato contendo astaxantina foi extraído com acetona e caracterizada quanto ao rendimento, teor de astaxantina, composição de ácidos graxos, atividade antioxidante e FTIR. Os filmes foram feitos utilizando a técnica casting. Foram elaborados quatro filmes, Gel (Controle), GQ, GQ0,5Ast e GQ1,0Ast. Para isso, as soluções filmogênicas foram preparadas com 3% de gelatina (mv), 1% de quitosana (mv) e 0,5 e 1,0% de extrato (mv). Os filmes foram caracterizados quanto a espessura, cor, FTIR, MEV, atividade antioxidante, atividade antimicrobiana, PVA e TGA. Os dados foram submetidos à análise de variância (ANOVA) e teste de Tukey com o auxílio do software BioEstat 5.0 e nível de significância 5%. A gelatina apresentou elevado peso molecular e força do gel igual a 400,97 ± 10 g. O extrato apresentou teor de astaxantina igual a 294,3 µg AST g -1 extrato e atividade antioxidante (PI) 50,19% para uma concentração de 5,87 µg AST mL -1 . Os filmes GQ0,5Ast e GQ1,0Ast apresentaram cor amarelada devido à cor do extrato, comprovadamente pelos maiores valores de a* e b* (p<0,05). De acordo com as imagens MEV, os filmes GQ0,5Ast e GQ1,0Ast apresentaram superfície irregular, com bastante poros e protuberâncias. E seção transversal heterogênea, rugosa e com poros ou fissuras principalmente na região próxima a superfície, caracterizando uma separação de fases. Os filmes GQ0,5Ast e GQ1,0Ast apresentaram maior atividade antioxidante (PI) (p<0,05), sendo 33,15 ± 2,21% e 41,57 ± 2,07%, respectivamente. De acordo com o método de difusão em ágar e técnica do poço, nenhum filme apresentou atividade antimicrobiana. O PVA dos filmes GQ0,5Ast e GQ1,0Ast não apresentaram diferenças significativas entre si e o Gel (Controle) (p>0,05), indicando que os mesmos não apresentaram uma melhor propriedade de barreira ao vapor de água. Com relação à TGA, a adição de extrato teve pouca influência na estabilidade térmica dos filmes. Portanto, conclui-se que os filmes comestíveis de gelatina de tilápia (O. niloticus) e quitosana incorporados com extrato de astaxantina de camarão branco do Pacífico (L. vannamei) possuem potencial como filmes antioxidantes para aplicação e aumento da vida de prateleira de alimentos.
ABSTRACT
The growth of aquaculture production causes an increase in the amount of waste generated from the processing of this raw material. What can cause problems environmental hazards, if an appropriate destination is not given. In addition, food and packaging have strived to reduce the amount of non-biodegradable materials in packaging, which are a source of pollution and can cause environmental problems related to their treatment after disposal. At the same time, the industry is looking for new strategies to increase food shelf life. In this context, there is the production of edible and/or biodegradable films from natural compounds. The present study aimed to extract and characterize gelatin from the skin of Nile tilapia (O. niloticus), astaxanthin extracted from the cephalothorax of Pacific white shrimp (L. vannamei) and apply in edible films. Type A gelatin was extracted by acid pretreatment and characterized for yield, molecular weight distribution, FTIR, gel strength and TGA. The extract containing astaxanthin was extracted with acetone and characterized for yield, astaxanthin content, fatty acid composition, antioxidant activity and FTIR. The films were made using the casting technique. Four films were made, Gel (Control), GQ, GQ0.5Ast and GQ1.0Ast. For this, the film-forming solutions were prepared with 3% gelatin (mv), 1% chitosan (mv) and 0.5 and 1.0% extract (mv). The films were characterized for thickness, color, FTIR, SEM, antioxidant activity, antimicrobial activity, PVA and TGA. Data were submitted to analysis of variance (ANOVA) and Tukey's test with the help of BioEstat 5.0 software and a significance level of 5%. Gelatin had a high molecular weight and gel strength equal to 400.97 ± 10 g. The extract showed astaxanthin content equal to 294.3 µg AST g -1 extract and antioxidant activity (PI) 50.19% for a concentration of 5.87 µg AST mL -1 . The movies GQ0.5Ast and GQ1.0Ast showed a yellowish color due to the color of the extract, which has been proven by the highest values of a* and b* (p<0.05). According to the SEM images, the films GQ0,5Ast and GQ1,0Ast had an irregular surface, with many pores and protrusions. And cross section heterogeneous, rough and with pores or cracks mainly in the region close to the surface, characterizing a separation of phases. The films GQ0.5Ast and GQ1.0Ast showed higher antioxidant activity (PI) (p<0.05), being 33.15 ± 2.21% and 41.57 ± 2.07%, respectively. According to the agar diffusion method and well technique, none of the films showed antimicrobial activity. The PVA of the GQ0.5Ast and GQ1.0Ast films did not show significant differences between them and the Gel (Control) (p>0.05), indicating that they did not show a better barrier property to water vapor. Regarding TGA, the addition of extract had little influence on the thermal stability of the films. Therefore, it is concluded that edible films of gelatine tilapia (O. niloticus) and chitosan incorporated with astaxanthin extract from Pacific white shrimp (L. vannamei) have potential as antioxidant films for application and increase in the shelf life of foods.
