Resumo

Dengue fever vectored by the mosquito Aedes aegypti is one of the most rapidly spreading insect-borne diseases. Current reliance of dengue vector control is mostly on chemical insecticides. Growing insecticide resistance in the primary mosquito vector, Aedes aegypti, limits the effectiveness of vector control through chemical insecticides. These chemical insecticides also have negative environmental impacts on animals, plants and human health. Myco-biocontrol agents are naturally occurring organisms and are found to be less damaging to the environment as compared to chemical insecticides. In the present study, entomopathogenic potential of local strains of fungi isolated from soil was assessed for the control of dengue vector. Local fungal isolates presents better alternative to introducing a foreign biocontrol strain, as they may be better adapted to environmental conditions of the area to survive and may have more entomopathogenic efficacy against target organism. Larvicidal efficacy of Fusarium equiseti and Fusarium proliferatum was evaluated against Aedes aegypti. Local strains of F. equiseti (MK371718) and F. proliferatum (MK371715) were isolated from the soil of Changa Manga Forest, Pakistan by using insect bait method. Larvicidal activity of two Fusarium spp. was tested against forth instar larvae of A. aegypti in the laboratory, using concentrations 105, 106, 107 and 108 conidia /ml. LC50 values for F. equiseti after 24h, 48h, 72h and 96h of exposure were recorded as 3.8x108, 2.9x107, 2.0x107, and 7.1x106 conidia /ml respectively while LC50 values for F. proliferatum were recorded as 1.21x108, 9.6x107, 4.2x107, 2.6x107 conidia /ml respectively after 24h, 48h, 72h and 96h of exposure. The results indicate that among two fungal strains F. equiseti was found to be more effective in terms of its larvicidal activity than F. proliferatum against larvae of A. aegypti.

By applying the in-silico method, resveratrol was docked on those proteins which are responsible for bone loss. The Molecular docking data between the resveratrol and Receptor activator of nuclear factor-kappa-Β ligand [RANKL] receptors proved that resveratrol binds tightly to the receptors, showed the highest binding affinities of −6.9, −7.6, −7.1, −6.9, −6.7, and −7.1 kcal/mol. According to in-vitro data, Resveratrol reduced the osteoclasts after treating Marrow-Derived Macrophages [BMM] with Macrophage colony-stimulating factor [MCSF] 20ng / ml and RANKL 50ng / ml, with different concentrations of resveratrol (2.5, 10 μg / ml) For 7 days, the cells were treated with MCSF (20 ng / ml) and RANKL (40 ng / ml) together with concentrated trimethyl ether and resveratrol (2.5, 10 μg / ml) within 12 hours. Which, not affect cell survival. After fixing osteoclast cells with formaldehyde fixative on glass coverslip followed by incubation with 0.1% Triton X-100 in PBS for 5 min and after that stain with rhodamine phalloidin staining for actin and Hoechst for nuclei. Fluorescence microscopy was performed to see the distribution of filaments actin [F.actin]. Finally, resveratrol reduced the actin ring formation. Resveratrol is the best bioactive compound for drug preparation against bone loss.

O resveratrol é o melhor composto bioativo para o preparo de medicamentos contra a perda óssea.

De acordo com dados in-vitro, o resveratrol reduziu os osteoclastos após o tratamento de macrófagos derivados da medula óssea [Bone Marrow-derived Macrophage (BMM)] com fator estimulador de colônias de macrófagos [Macrophage Colony-Stimulating Factor (MCSF)] 20ng / ml e RANKL 50ng / ml, com diferentes concentrações de resveratrol (2,5, 10 μg / ml). Durante sete dias, as células foram tratadas com MCSF (20 ng / ml) e RANKL (40 ng / ml) juntamente com éter trimetílico concentrado e resveratrol (2,5, 10 μg / ml) em 12 horas, processo que não afeta a sobrevivência celular. Após a fixação de células de osteoclastos com fixador de formaldeído em lamela de vidro seguido de incubação com 0,1% Triton X-100 em PBS por 5 min, foi realizado posteriormente o procedimento para corar com rodamina faloidina a actina e Hoechst os núcleos. A microscopia de fluorescência foi realizada para ver a distribuição dos filamentos de actina [F.actina]. Finalmente, o resveratrol reduziu a formação do anel de actina. O resveratrol é o melhor composto bioativo para o preparo de medicamentos contra a perda óssea.

Resumo

By applying the in-silico method, resveratrol was docked on those proteins which are responsible for bone loss. The Molecular docking data between the resveratrol and Receptor activator of nuclear factor-kappa-Β ligand [RANKL] receptors proved that resveratrol binds tightly to the receptors, showed the highest binding affinities of −6.9, −7.6, −7.1, −6.9, −6.7, and −7.1 kcal/mol. According to in-vitro data, Resveratrol reduced the osteoclasts after treating Marrow-Derived Macrophages [BMM] with Macrophage colony-stimulating factor [MCSF] 20ng / ml and RANKL 50ng / ml, with different concentrations of resveratrol (2.5, 10 μg / ml) For 7 days, the cells were treated with MCSF (20 ng / ml) and RANKL (40 ng / ml) together with concentrated trimethyl ether and resveratrol (2.5, 10 μg / ml) within 12 hours. Which, not affect cell survival. After fixing osteoclast cells with formaldehyde fixative on glass coverslip followed by incubation with 0.1% Triton X-100 in PBS for 5 min and after that stain with rhodamine phalloidin staining for actin and Hoechst for nuclei. Fluorescence microscopy was performed to see the distribution of filaments actin [F.actin]. Finally, resveratrol reduced the actin ring formation. Resveratrol is the best bioactive compound for drug preparation against bone loss.(AU)

Resumo

Morphometric measurement and roosting ecology of Pteropus medius were aimed to find out in Mansehra district of KP, Pakistan. Total 3149 numbers of bats were found in eight biological spots visited; Baffa Doraha, Darband, Dadar, Jallu, Hazara University, Garhi Habibullah Chattar Plain and Jabori, in total 299 numbers of different species of trees including; Morus alba, Pinus raxburghi, Eucalyptus camaldulensis, Morus nigra, Grevillea robusta, Brousonetia papyrifera,Platanus orientalis, Ailanthus altissima, Hevea brasiliensis and Populus nigra. Morphometric features were measured and found vary according to sex of the bats. The average wing span, wing`s length from tip of wing to neck, from thumb to tip of wing and the body`s length from head and claws were recorded to be 102.98 cm, 49.07cm, 28.7 cm and 22.78 cm respectively in males while 93.67 cm, 44.83cm, 24.78cm and 22.78 cm respectively in female bats. Mean circumference of the body including wings and without wing were measured as 22.78 cm and 17.29 cm in males and that of female were 20.07 cm and 16.9 cm. Average length of thumb 3.64 cm, ear`s length 3.1 cm, snout 5.62cm, eye length were 1.07 cm for both sexes and length between the feet in extended position were16.3 cm. Generally different measurement of males bodies were found to be greater than female such as mean body surface area, mass, volume and pressure were found to be 2691.79 cm2, 855.7gm,1236.4 ml and 295.77 dyne/ c m 3for male and 2576.46 cm2, 852.71gm,1207 ml and 290.2 dyne/ c m 3 respectively for female. While weight and density for both males and females bats were same with mean of 8.59 newton and 0.701 g/m3. Findings of current reports can add valued information in literature about bats, which can be used for species identification and conservation.


A medição morfométrica e a ecologia de poleiro de Pteropus medius foram realizadas no distrito de Mansehra de KP, Paquistão. Foram encontrados 3.149 morcegos em 8 pontos biológicos visitados; Baffa Doraha, Darband, Dadar, Jallu, Universidade Hazara, Garhi Habibullah Chattar Plain e Jabori, no total 299 números de diferentes espécies de árvores, incluindo: Morus alba, Pinus raxburghi, Eucalyptus camaldulensis, Morus nigra, Grevillea robusta, Brousonetia papyrifera, Platanus orientalis, Ailanthus altissima, Hevea brasiliensis e Populus nigra. As características morfométricas foram medidas e variam de acordo com o sexo dos morcegos. A envergadura média da asa, o comprimento da asa da ponta da asa ao pescoço, do polegar à ponta da asa e o comprimento do corpo da cabeça e garras foram registrados em 102,98 cm, 49,07 cm, 28,7 cm e 22,78 cm, respectivamente, nos machos. Enquanto 93,67 cm, 44,83 cm, 24,78 cm e 22,78 cm respectivamente, nas fêmeas. As circunferências médias do corpo incluindo asas e sem asas foram medidas de 22,78 cm e 17,29 cm nos machos e as das fêmeas foram 20,07 cm e 16,9 cm. O comprimento médio do polegar 3,64 cm, comprimento da orelha 3,1 cm, focinho 5,62 cm, comprimento dos olhos 1,07 cm para ambos os sexos e comprimento entre os pés em posição estendida foi de 16,3 cm. Geralmente, as medidas diferentes dos corpos masculinos foram maiores que as femininas, como a área de superfície corporal média, massa, volume e pressão foram encontrados em 2691,79 cm2, 855,7 gm,1236,4 ml e 295,77 dines para masculino e 2576,46 cm2, 852,71 gm, 1207 ml e 290,2 dines respectivamente para fêmeas. Enquanto o peso e a densidade para machos e fêmeas de morcegos foram os mesmos com média de 8,59 N e 0,701 g/m3. Os achados de relatórios atuais podem agregar informações valiosas na literatura sobre morcegos, que podem ser utilizadas para identificação e conservação de espécies.

The emergence of multi-drug resistant (MDR) bacterial strains, which are posing a global health threat has developed the interest of scientists to use bacteriophages instead of conventional antibiotics therapy. In light of an increased interest in the use of phage as a bacterial control agent, the study aimed to isolate and characterize lytic phages from sewage effluent. During the current study, bacteriophage AS1 was isolated from sewage effluent against E.coli S2. The lytic activity of phageAS1 was limited to E.coli S2 strain showing monovalent behavior. The calculated phage titer was 3.5×109 pfu/ml. PhageAS1 was stable at a wide range of pH and temperature. The maximum stability was recorded at 37ºC and pH 7.0, while showing its normal lytic activity at temperature 60ºC and from pH 5.0 to 11.0 respectively. At temperature 70ºC, phage activity was somewhat reduced whereas, further increase in temperature and decrease or increase in pH completely inactivated the phage. From the current study, it was concluded that waste water is a best source for finding bacteriophages against multi-drug resistant bacterial strains and can be used as bacterial control agent.


O surgimento de cepas bacterianas multirresistentes (MDR), que representam uma ameaça global à saúde, desenvolveu o interesse dos cientistas em usar bacteriófagos em vez da terapia convencional com antibióticos. Diante do crescente interesse no uso de fago como agente de controle bacteriano, o estudo visou isolar e caracterizar fagos líticos de efluente de esgoto. Durante o estudo atual, o bacteriófago AS1 foi isolado de efluente de esgoto contra E. coli S2. A atividade lítica de phageAS1 foi limitada à cepa E. coli S2, apresentando comportamento monovalente. O título de fago calculado foi de 3,5 x 109 ufp/ml. PhageAS1 foi estável em uma ampla faixa de pH e temperatura. A estabilidade máxima foi registrada a 37ºC e pH 7,0, enquanto mostrou atividade lítica normal em temperatura de 60ºC e pH 5,0 a 11,0, respectivamente. The present study aimed at evaluating the effects of dietary selenium (Se) supplementation on egg morphometric and quality traits in Aseel chicken. The results indicated greater egg length in Mushki and Mianwali, lower egg breadth in Lakha, the highest (p≤0.05) egg volume and surface area in Mianwali, and higher shape index in Lakha and Peshawari. Lakha had the highest Haugh unit and lower eggshell thickness with and without membrane than the other three varieties. Interaction of Se sources and Aseel varieties exhibited the highest (p≤0.05) egg volume and surface area in Mianwali variety when fed on organic Se supplemented diet. Similarly, the interaction showed the highest eggshell thickness with membrane in Peshawari and without membrane in Mianwali when fed on inorganic Se supplemented diet. From the findings, it can be concluded that egg physical characteristics of Mianwali variety hens can be improved by the supplementation of Se-enriched yeast, as an organic Se source, in the diet.(AU)

The study was planned to evaluate the effect of different naked-neck (NN) chicken genotypes and breeder ages on the growth performance of their progenies. A total of 126 chicks, from two breeder ages and three NN varieties, were reared up to 20 weeks of age. A randomized complete block design in factorial arrangement, with six treatments of seven replicates of two females and one male each, was applied. Treatments consisted of three NN varieties (light brown, black, and dark brown) and two breeder age groups (45 and 55 weeks old). Growth performance (body weight gain, feed intake and feed conversion ratio) were evaluated. Higher feed intake was observed in both the male and female progenies of 55-week-old compared with 45-week-old breeders. Light brown males had higher weight gain than dark brown males, whereas in the female progeny, higher weight gain was obtained in the black variety than that in the light brown and dark brown varieties. Regarding breeder age groups, both male and female progenies of 55-week-oldbreeders exhibited higher weight gain than those of 45-week-old breeders. The male progeny of 55-week-old black breeders showed the highest body weight gain among all treatments. Regarding female progeny, higher weight gain was observed in the black variety compared with the light brown and dark brown variety, and in the progeny of 55-week-old breeders than in the progeny of 45-week-old breeders. The interaction between factors showed that females from 55-week-old black breeder has the highest weight gain, whereas the lowest weight gain was found in the progeny of 45-week-old light brown breeders. The male progeny of light brown breeders showed better FCR than those of black and dark brown breeders and of 55-week-old breeders compared with that of 45-week-old breeders. The best FCR was obtained in the progeny of 55-week-old light brown breeders. Resumo

The study was planned to evaluate the effect of different naked-neck (NN) chicken genotypes and breeder ages on the growth performance of their progenies. A total of 126 chicks, from two breeder ages and three NN varieties, were reared up to 20 weeks of age. A randomized complete block design in factorial arrangement, with six treatments of seven replicates of two females and one male each, was applied. Treatments consisted of three NN varieties (light brown, black, and dark brown) and two breeder age groups (45 and 55 weeks old). Growth performance (body weight gain, feed intake and feed conversion ratio) were evaluated. Higher feed intake was observed in both the male and female progenies of 55-week-old compared with 45-week-old breeders. Light brown males had higher weight gain than dark brown males, whereas in the female progeny, higher weight gain was obtained in the black variety than that in the light brown and dark brown varieties. Regarding breeder age groups, both male and female progenies of 55-week-oldbreeders exhibited higher weight gain than those of 45-week-old breeders. The male progeny of 55-week-old black breeders showed the highest body weight gain among all treatments. Regarding female progeny, higher weight gain was observed in the black variety compared with the light brown and dark brown variety, and in the progeny of 55-week-old breeders than in the progeny of 45-week-old breeders. The interaction between factors showed that females from 55-week-old black breeder has the highest weight gain, whereas the lowest weight gain was found in the progeny of 45-week-old light brown breeders. The male progeny of light brown breeders showed better FCR than those of black and dark brown breeders and of 55-week-old breeders compared with that of 45-week-old breeders. The best FCR was obtained in the progeny of 55-week-old light brown breeders. Obesity and related diseases represent greatest threats to human health. Nanoparticles (NPs) serve to reduce toxicity; reinforce bioactivity and improve targeting. This study was intended to investigate the antiobesity and antioxidant activities of selenium and zinc oxide nanoparticles. Methods: Twenty four adult male rats were divided into four groups, group1 control rats fed normal diet and the other three groups were fed high fat diet (HFD) for 10 weeks to induce obesity and injected intraperitoneally with saline, SeNPs (30µg/kg b.wt) and ZnONPs (5mg/kg b.wt) respectively on the last two weeks of feeding (9th and 10th). Results: HFD increased body weight, oxidative stress as indicated by elevated lipid peroxidation and decreased glutathione and catalase levels, increased significantly serum lipid fractions, leptin, liver enzymes, creatinine and uric acid. While causing a substantial decrease in HDL-C and thyroid hormone T4 levels. The results confirmed that treatment with SeNPs and ZnONPs significantly reduced body weight, MDA and improved liver and kidney functions, ameliorated serum lipid fractions level and significantly increased glutathione, catalase, HDL-C and thyroid hormone. Conclusion: SeNPs and ZnONPs significantly mitigate hyperlipidemia and oxidative stress. So, they might be potential candidate for obesity amelioration.


A obesidade e doenças relacionadas representam as maiores ameaças à saúde humana. As nanopartículas (NPs) servem para reduzir a toxicidade, reforçar a bioatividade e melhorar o direcionamento. Este estudo teve como objetivo investigar as atividades antiobesidade e antioxidante de nanopartículas de selênio e óxido de zinco. Métodos: Vinte e quatro ratos machos adultos foram divididos em quatro grupos, grupo 1 ratos controle alimentados com dieta normal e os outros três grupos foram alimentados com dieta hiperlipídica (HFD) por 10 semanas para induzir obesidade e injetados intraperitonealmente com soro fisiológico, SeNPs (30µg/kg b.wt) e ZnONPs (5mg/kg b.wt) respectivamente nas duas últimas semanas de alimentação (9ª e 10ª). Resultados: HFD aumentou o peso corporal, estresse oxidativo indicado pela peroxidação lipídica elevada e diminuição dos níveis de glutationa e catalase, aumentou significativamente as frações lipídicas séricas, leptina, enzimas hepáticas, creatinina e ácido úrico. Enquanto causa uma diminuição substancial nos níveis de HDL-C e hormônio tireoidiano T4. Os resultados confirmaram que o tratamento com SeNPs e ZnONPs reduziu significativamente o peso corporal, MDA e melhorou as funções hepáticas e renais, melhorou o nível das frações lipídicas séricas e aumentou significativamente a glutationa, catalase, HDL-C e hormônio tireoidiano. Conclusão: SeNPs e ZnONPs atenuam significativamente a hiperlipidemia e o estresse oxidativo, então eles podem ser candidatos em potencial para a melhora da obesidade.

Resumo

Acute myocardiopathy in alloxan treated experimental dogs and rabbits was induced by subcutaneous (SQ) injection of scorpion venom from Mesobuthus tamulus concanesis, Pocock. Envenoming resulted in an initial transient hypertension (180-320 mm Hg.) followed by hypotension. Simultaneous administration of venom and species-specific scorpion antivenom (SAV) prevented hypertension and hypotension. Hypotension did not occur when SAV was given 60 min after envenoming. Blood glucose, triglycerides, cholesterol, amylase, insulin, glucagon, cortisol, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), platelet count, red blood cell (RBC) count, hemoglobin (Hb), 2,3-diphosphoglycerate (2,3-DPG), and glutathione levels were increased 60 and 90 min after envenoming. Total white blood cell (WBC) count was reduced 60 min and increased 90 min after envenoming. Simultaneous administration of venom and SAV did not alter Hb, MCHC, and packed cell volume (PCV) levels, or ECG, and cardiovascular, biochemical, metabolic, and hormonal changes. Hematological parameters were reversed when SAV was given 30 and 60 min after envenoming. PCV, Hb, and MCHC values returned to normal 120 min after SAV. Alloxan-treated dogs showed increased blood glucose, cholesterol, glucagon, cortisol levels; reduced glycogen content of liver, cardiac and skeletal muscles; and reduced insulin levels and insulin/ glucagon ratio (I/G ratio). Envenoming in the alloxan pre-treated dogs further increased these levels and reduced tissue glycogen content, insulin levels, and I/G ratio. Administration of 4 U of insulin to alloxan pre-treated envenomed rabbits caused a biochemical and clinical improvement and increased glycogen content of all tissues in comparison with the values from those administered with SAV to alloxan pre-treated envenomed animals. SAV administration to envenomed alloxan pre-treated rabbits did not cause clinical or biochemical improvement. Severe scorpion envenoming causes an autonomic storm with a massive release of catecholamines and other counter-regulatory hormones; changes in insulin secretion resulting in fuel energy deficits producing multi-system-organ-failure (MSOF); and death. Administration of either insulin or SAV (through the release of insulin) appears to be the physiological basis for the control of the metabolic support to control the adverse effects triggered by counter-regulatory hormones.
