Resumo

This study evaluated the physicochemical and morphological properties of a marine sponge protein extract (PE) using scanning electron microscopy (SEM), Energy dispersive X-ray spectroscopy (EDS), analysis of mass loss and pH and in vitro and in vivo. Scanning electron microscopy showed that PE fibers present a granular aspect and irregular structure and the element carbon followed by oxygen was detected in the EDS analysis. Moreover, a 29% of mass loss was observed after 14 days and the pH slightly modified after 14 days. Cell viability of fibroblast cells (L929) of control and PE at a concentration of 25% demonstrated higher values compared to the groups. Osteoblast cell viability of PE at 25 and 50% was significantly higher. Comet assay on day 1 showed higher values for PE at 25%. In addition, in vivo experiments demonstrated that in the treated animals, the bone defects were filled with biomaterial particles, granulation tissue and some areas of newly formed bone. Furthermore, similar immunoexpression of Runx-2 and Cox-2 was observed. Taken together, all results suggest that PE is biocompatible, present non-citotoxicity in the in vitro studies (at the lower concentration) and in the in vivo studies and it can be considered as an alternative source of collagen for tissue engineering proposals.

Background: The most common conditions that compromise the stifle joint in dogs are medial patellar luxation (MPL) and cranial cruciate ligament (CCL) rupture. Surgical procedures are usually indicated for the treatment of these diseases. One ordinarytechnique for the treatment of MPL is the tibial tuberosity transposition, and one prominent technique for CCL rupture is the tibialtuberosity advancement. These techniques can be associated in one surgical procedure called tibial tuberosity transposition andadvancement (TTTA) for the simultaneous treatment of both stifle diseases. The aim of this study is to report the surgical treatmentof a dog with MPL and CCL rupture affecting the same joint by the TTTA technique with the use of a TTA-Maquet cage-only.Case: A 3-year-old Pitbull dog weighing 39 kg was attended at Veterinary Hospital with a history of marked lameness in theleft pelvic limb. The orthopedic examination showed positive results for cranial drawer motion and tibial compression tests,and a complete CCL rupture was diagnosed. The presence of patellar luxation was evaluated by manual pressure on the patella, and grade 3 of MPL was diagnosed. Both conditions were affecting the same stifle joint. In addition, survey radiographsof the affected joint were performed. Surgical treatment was indicated by the TTTA technique. Radiographic measurementswere taken to calculate the cranial tibial tuberosity advancement by the tibial plateau and the common tangent methods, and atitanium TTA-Maquet cage-only of 10.5 x 20.0 mm was selected. Linear osteotomy was performed on the tibial tuberosity withthe aid of an oscillating saw, based on the Maquet hole technique. The tibial tuberosity was carefully displaced cranially and thecage was inserted at the site of osteotomy. The cage ears were molded on the tibial surface and fixed with 2.4 mm self-tappingcortical screws...

Resumo

Com este estudo objetivou-se descrever os aspectos anatômicos e histológicos do coração do jacaré-do-pantanal (Caiman yacare), proveniente de zoocriadouro. Para tanto, estudou-se 13 exemplares da espécie, os quais foram perfundidos, conservados em solução de formaldeído a 10% e submetidos às técnicas anatômicas específicas. O coração foi separado e amostras foram colhidas e submetidas à avaliação histológica. Macroscopicamente o coração é tetracavitário, e além de dois átrios e dois ventrículos, apresenta uma estrutura denominada cone arterial, do qual emergem os vasos da base do coração. Foram identificadas duas aortas, direita e esquerda, sendo que a esquerda emerge do ventrículo direito e se comunica com o tronco sistêmico direito por meio do forame de Panizza. Histologicamente o coração possui epicárdio, miocárdio e endocárdio típicos. Concluímos que a histologia do coração, no jacaré-do-pantanal, é semelhante à de outras espécies de répteis. Contudo, anatomicamente apresenta particularidades importantes, as quais representam, possivelmente, adaptações que permitiram a perpetuação da espécie.(AU)


The aim of this study was to describe anatomical and histological aspects of the heart of Caiman yacare from a crocodile breeding center. For this purpose, we have chosen and further studied 13 specimens which were perfused and preserved in a 10% formaldehyde solution and they were subjected to the specific anatomical techniques. The heart was separated and samples were collected and submitted to the specific histological procedures. Macroscopically, the heart is four-chambered and besides two atria and two ventricles, has a structure called arterial cone from which the large vessels of the heart emerge. Two aortas, left and right, were identified. The left aorta emerges from the right ventricle and communicates with the systemic trunk through the Foramen of Panizza. From a histological point of view, the heart is typically composed of epicardium, myocardium and endocardium. According to these observations, it is assumed that the histology of the heart of "Jacaré-do-Pantanal" is similar to other species of reptiles. However, there are some anatomic particularities, which possibly represent the adaptations allowing the perpetuation of the species.(AU)

Resumo

PURPOSE: To investigate hemostatic effects of supplementary factor XIII and desmopressin (DDAVP) in resuscitation of uncontrolled bleeding. METHODS: Fifty-four rabbits were randomized in nine groups: G1: Sham; G2: FXIII and normotensive resuscitation (NBP); G3: FXIII and permissive hypotension (PH) (MAP 60% baseline); G4: FXIII/DDAVP/NBP; G5: FXIII/DDAVP/PH; G6: NBP only; G7: FXIII no hemorrhage; G8: FXIII/DDAVP no hemorrhage; G9: PH only. Thromboelastometry and intra-abdominal blood loss were assessed. Scanning electron microscopy (EM) of the clots was performed. RESULTS: Compared to Sham, only G8 (FXIII/DDAVP w/o hemorrhage) showed clotting time (CT) significantly lower (p<0.05). NBP alone (G6) resulted in significantly prolonged CT compared to G2, G3 and G5 (p<0.05). Similarly, median alpha angle was significantly larger in G3,4,5, and 9 compared to G6 (p<0.05). Area under the curve was significantly greater in G5 than G2. Intra-abdominal blood loss was lower in G5 and G9 compared to G2 and G6. FXIII/DDAVP and PH resulted in more robust fibrin mesh by EM. CONCLUSIONS: Normotensive resuscitation provokes more bleeding and worsens coagulation compared to pH, that is partially reversed by factor XIII and desmopressin. FXIII and DDAVP can synergistically improve coagulation. Permissive hypotension reduces bleeding regardless of those agents.(AU)

O balanço de nitrogênio (BN) e os teores de uréia no soro (TUS) e urina (TUur) foram utilizados para avaliar o efeito da inclusão de níveis crescentes de uréia (0% - T1; 1% - T2; 2% - T3; e 3% - T4) sobre dietas contendo 40% de palma forrageira “in natura” e 60% de resíduo desidratado de uva de vitivinícolas. Para tanto, foram utilizados 16 ovinos machos não castrados, com média de 9 meses de idade e com peso médio de 37 kg de PV. Os animais foram distribuídos em um delineamento experimental de blocos, ao acaso. O período experimental total foi de 40 dias, divididos em dois blocos de 20 dias cada, e constou de 4 tratamentos e 8 repetições. Os valores do N ingerido e N absorvido, a perda de N pelas fezes e urina não foram alterados significativamente pela inclusão de uréia. Os valores médios para as relações entre N retido/N ingerido e N retido/N absorvido também não foram afetadas significativamente pelos níveis de uréia nas dietas. Os TUS e TUur apresentaram comportamento linear em relação aos níveis de uréia. Os resultados indicam a inclusão de 3% de uréia em dietas contendo resíduo desidratado de uva de vitivinícolas e palma forrageira “in natura,” havendo uma maior adequação ao metabolismo normal dos animais e um melhor desempenho dos animais.
