Resumo

Hepatitis C virus (HCV) genotypes vary greatly in different regions. The aim of this study is to investigate the distribution of HCV genotypes in HCV infected patients, in Ningxia Hui Autonomous Region. Nucleic acid extraction and amplification were performed with test kits on 153 HCV infected patients serum samples. The HCV viral load was measured using reverse transcriptase PCR (RT-PCR) and HCV genotypes were determined. Among the 153 HCV-infected patients, 56 had genotype (GT)1b (36.60%), 45 had GT2a (29.40%), 23 had GT3a (15.00%), 14 had GT3b (9.20%),13 had GT6a (8.50%), 1 had GT1g (0.70%), 1 had GT6xa (0.70%). In GT1b, 21.40% were female and 78.60% were male; in GT2a, 42.20% were female and 57.80% were male;Males were most prevalent in genotypes 1b(39.30%), while female were most prevalent in genotype 2a(46.30%). Rare GT1g and GT6xa were also detected in males. The 41-50 year age group had the highest HCV prevalence of 32.00%. HCV GT1b is the predominant HCV genotype in Ningxia Hui Autonomous Region.


Os genótipos do vírus da hepatite C (HCV) variam muito de acordo com a região. O objetivo deste estudo é investigar a distribuição do genótipo do vírus da hepatite C em pessoas infectadas pelo vírus na região autônoma de Ningxia Hui. A extração de ácido nucleico e a expansão de amostras séricas em 153 pacientes infectados com HCV foram realizadas utilizando kits de ensaio. A capacidade do vírus HCV foi medida pela reação em cadeia da polimerase retrotranscrição (RT-PCR) e o genótipo HCV foi determinado. Os genótipos (Gt) tiveram a seguinte distribuição entre os 153 casos de infecção HCV: 56 Gt 1-B (36, 60%), 45 Gt 2-A (29, 40%), 23 Gt 3-A (15, 00%), 14 Gt 3-B (9, 20%), 13 Gt 6-A (8, 50%), 1 Gt 1-G (0, 70%) e 1 Gt 6-Xa (0, 70%). Já o sexo dos indivíduos teve a seguinte distribuição: Gt 1-B, 21, 40% mulheres e 78, 60% homens; Gt 2-A, 42, 20% mulheres e 57, 80% homens. A presença de homens é mais comum no genótipo 1-B (39, 30%), enquanto as mulheres ocorrem mais comumente no genótipo 2-A (46, 30%). Gt 1-G e Gt 6-Xa raros também foram detectados em homens. A taxa de infecção com HCV para grupos etários de 41 a 50 anos é a mais alta, com 32,00%. HCV Gt 1-B é o genótipo dominante do HCV na região autônoma de Ningxia Hui.

Resumo

ABSTRACT: Malatya is an important production centre of apricots in Turkey, and the world. However, a major problem causing product loss, is late spring frosts. Cold damages the flowers and thus, fruit set and yield decrease. As a result, the farmer's income fluctuates from year to year. This study, stated apricot farmers demographics, and technical features of 52 interviewed apricot farms, in Malatya. Also, conjoint analysis (CA) is used to identify the farmers' preferences for agricultural insurance attributes. This method investigated the joint effect of a set of independent variables, on an ordinal scale of a measurement-dependent variable. Of the farmers, 76.9% have done agricultural insurance last year.


RESUMO: Malatya é um importante centro de produção de damascos na Turquia e no mundo. No entanto, um grande problema que causa a perda de produto são as geadas do final da primavera. O frio prejudica as flores e, com isso, a frutificação e a produção diminuem. Como resultado, a renda do agricultor varia de ano para ano. No estudo, objetiva-se constatar a demografia dos produtores de damasco e características técnicas de 52 fazendas de damasco entrevistadas, em Malatya. Além disso, a análise conjunta é usada para identificar as preferências dos agricultores por atributos de seguro agrícola. Este método investiga o efeito conjunto de um conjunto de variáveis independentes, em uma escala ordinal de uma variável dependente de medição. 76,9% dos agricultores fizeram seguro agrícola no ano passado.

Resumo

This study was conducted to evaluate the effect of feeding lupin kernel and faba bean as an alternative to soybean meal on the growth performance, blood profiles, relative organ weight, and hepatic fatty acid composition of broiler chicks. A total of 525, 1-day-old Ross 308 male chicks were randomly assigned into five groups with seven replicates. The treatments consisted of five experimental diets; corn-soybean meal without lupin or faba bean (as control), diets with lupin 5% or 10%, and diets with faba bean 5% or 10%. The body weight (BW) and average daily gain (ADG) were not significantly different among the groups during starter period. However, BW and ADG of chicks fed diets with 10% lupin and faba bean were significantly higher than those of 5% lupin and faba bean during grower period (p<0.01). The feed conversion ratio was significantly lower in the group fed diets with 10% lupin and faba bean than the control during total rearing periods (p<0.001). There were no significant differences in blood profiles among the groups. As for the hepatic fatty acid composition, the levels of total polyunsaturated fatty acids and total ω6 in chicks fed lupin and faba bean were significantly higher than the control. It was suggested that dietary lupin and faba bean could enhance the incorporation of the beneficial fatty acids into liver fraction. In conclusion, supplementation of lupin and faba bean up to 10% can be used as an alternative to soybean meal in broiler diets.(AU)

Resumo

Purpose: To investigate the active ingredients of walnut ointment (WO) and its mechanism in repairing wounds. Methods: The ingredients of WO were detected by gas chromatography­mass spectrometry. The effect of linoleic acid (LA) was tested by in vitro Alamar Blue (AB) reagent. Image J software, histological and immunohistochemical analysis were used to confirm the healing effect of LA in the porcine skin model. The animals were euthanized after the experiment by injection of pentobarbital sodium. Results: LA, 24% in WO, promotes keratinocytes and fibroblasts proliferation, which were 50.09% and 15.07% respectively higher than control (p < 0.05). The healing rate of the LA group (96.02% ± 2%, 98.58% ± 0.78%) was higher than the saline group (82.11% ± 3.37%, 88.72% ± 1.73%) at week 3 and week 4 (p < 0.05). The epidermal thickness of the LA was 0.16 ± 0.04 mm greater and the expression of the P63 and CK10 proteins was stronger in the LA group than the control (p < 0.05). Conclusions: LA, which is the main components in WO can promote full-thickness burning wounds (FBWs) by stimulating cell proliferation and differentiation.

ABSTRACT Purpose: Subarachnoid hemorrhage (SAH) is a common complication of cerebral vascular disease. Hydrogen has been reported to alleviate early brain injury (EBI) through oxidative stress injury, reactive oxygen species (ROS), and autophagy. Autophagy is a programmed cell death mechanism that plays a vital role in neuronal cell death after SAH. However, the precise role of autophagy in hydrogen-mediated neuroprotection following SAH has not been confirmed. Methods: In the present study, the objective was to investigate the neuroprotective effects and potential molecular mechanisms of hydrogen-rich saline in SAH-induced EBI by regulating neural autophagy in the C57BL/6 mice model. Mortality, neurological score, brain water content, ROS, malondialdehyde (MDA), and neuronal death were evaluated. Results: The results show that hydrogen-rich saline treatment markedly increased the survival rate and neurological score, increased neuron survival, downregulated the autophagy protein expression of Beclin-1 and LC3, and endoplasmic reticulum (ER) stress. That indicates that hydrogen-rich saline-mediated inhibition of autophagy and ER stress ameliorate neuronal death after SAH. The neuroprotective capacity of hydrogen-rich saline is partly dependent on the ROS/Nrf2/heme oxygenase-1 (HO-1) signaling pathway. Conclusions: The results of this study demonstrate that hydrogen-rich saline improves neurological outcomes in mice and reduces neuronal death by protecting against neural autophagy and ER stress.

ABSTRACT: This study aimed to examine the efficacy of magnesium chloride therapy in sheep experimentally poisoned by Palicourea marcgravii and describe the clinical and laboratory findings that may aid in early ante mortem diagnosis of this poisoning. P. marcgravii is an important plant within a group of 22 species in Brazil that cause sudden death. Its toxic component is sodium monofluoroacetate. It was observed that a time for evolution of poisoning was longer in our study compared to other studies that used the same dose of P. marcgravii in sheep. However, all animals that were poisoned and subsequently treated with magnesium chloride died. The sheep presented characteristic clinical signs before death in addition to other signs that have not been previously described for this type of poisoning, such as abdominal breathing, coughing, head pressing, and nystagmus. This is the first evaluation of hematological parameters of sheep poisoned by P. marcgravii. Leukocytosis with neutrophilia, hyperfibrinogenemia, and hyperglycemia, associated with hypophosphatemia and elevated levels of serum aspartato aminotransferase (AST), glutamate dehydrogenase (GLDH), urea, creatinine, creatine kinase (CK) and lactate dehydrogenase (LDH), were observed. Additionally, changes were noted in necropsy, such as engorged jugular, large vessel congestion, pulmonary edema. Histological examination revealed vacuolar hydropic degeneration in the distal twisted uriniferous tubules in the kidneys, similar to those reported in previous studies on P. marcgravii poisoning.


RESUMO: O presente estudo objetivou verificar a eficácia terapêutica do cloreto de magnésio em ovinos intoxicados experimentalmente por Palicourea marcgravii e descrever os achados clínicos e laboratoriais que possam auxiliar no diagnóstico precoce ante mortem dessa intoxicação. P. marcgravii é a principal planta dentro de um grupo de 22 espécies que causam morte súbita no Brasil e seu princípio tóxico é o monofluoracetato de sódio. Durante o procedimento, observou-se maior tempo de evolução em relação a outros trabalhos utilizando-se a mesma dose da P. marcgravii em ovinos, entretanto todos os animais intoxicados e posteriormente tratados com cloreto de magnésio vieram a óbito. Os ovinos apresentaram sinais clínicos característicos antes da morte, associados a sinais nunca antes descritos nesse tipo de intoxicação, como respiração abdominal, tosse, head pressing e nistagmo. Trata-se da primeira avaliação dos parâmetros hematológicos de ovinos intoxicados por P. marcgravi. Foi observado leucocitose com neutrofilia, hiperfibrinogemia e hiperglicemia, associado a elevação dos valores séricos de AST, GLDH, ureia, creatinina, CK, LDH e hipofosfatemia. À necropsia foram notadas alterações como jugular ingurgitada, congestão de grandes vasos, edema pulmonar. Histologicamente havia degeneração hidrópica vacuolar nos túbulos uriníferos contorcidos distais no rim, semelhantes às já relatadas em estudos anteriores, na intoxicação por P. marcgravii.

Purpose: The current study explored the involvement of neurogenic pathway-linked cholecystokinin (CCK) release in RIP-induced cardioprotection in rats. Methods: Male Wistar rats were subjected to four cycles of alternate episodes of ischemia and reperfusion (five min each) to induce RIP. Thereafter, the hearts were subjected to global ischemia and reperfusion ex vivo. The myocardial damage was assessed by quantifying the levels of heartspecific biochemicals i.e. LDH-1, CK-MB and cTnT. Apoptotic cell injury was assessed by measuring the levels of caspase-3 and Bcl-2. The levels of CCK were measured in the plasma following RIP. Results: Exposure to RIP significantly increased the plasma levels of CCK and attenuated IR-induced myocardial injury. Administration of CCK antagonist, proglumide significantly attenuated RIP-induced cardioprotection. Administration of hexamethonium, a ganglion blocker, abolished RIP-induced increase in plasma CCK levels and cardioprotective effects. Exogenous delivery of CCK-8 restored the effects of RIP in hexamethonium treated animals. Conclusion: RIP activates the neurogenic pathway that may increase the plasma levels of CCK, which may act on the heart-localized CCK receptors to produce cardioprotection against I/R injury.(AU)

ABSTRACT: This study aimed to evaluate the appropriate sites of abdominocentesis for peritoneal fluid collection in cattle and to investigate the time of cell viability in vitro, comparing three methods of sample conservation. Twenty-one healthy cattle (19 females and 2 males) were subjected to a laparocentesis procedure to obtain peritoneal fluid, with punctures in three defined sites: left cranial, right cranial, and right caudal. The total peritoneal fluid collected was divided into three aliquots and maintained under three preservation conditions: room temperature (26°C), refrigeration (4°C), and room temperature (26°C) with the addition of 1L of 10% formaldehyde per 1mL of peritoneal fluid. The peritoneal fluid analysis performed immediately after collection consisted of: physical examination (color, appearance, volume, and specific gravity), biochemical measures (pH, total protein, fibrinogen, creatinine, and glucose), and cellularity (total and differential counts). The determination of proteins and the examination of cells were repeated in each separate aliquot at two, four, six, and eight hours after harvest. Data were analyzed through repeated measures ANOVA or Friedman test. The harvest was productive in 67% of cattle. The left cranial and the right cranial puncture sites were the most appropriate. Peritoneal fluid analyzed after collection, the total protein concentration ranged from 1.4 to 3.6g/dL, and number of leukocytes ranged from 54 to 1,322 cells/L; 60 to 95% of leukocytes were lymphocytes. The protein concentration decreased, but the absolute values of leukocytes, lymphocytes, and segmented neutrophils did not change up to eight hours after collection, independent of the maintenance method. Cell lysis was delayed by cooling, and the addition of formaldehyde did not help preserve the integrity of cellular morphology. Laparocentesis is a safe and secure procedure in cattle and maybe more productive when performed in specific sites on the left or right sides of the cranial abdominal wall. Peritoneal fluid samples may be analyzed with reliable results for up to eight hours after collection when kept refrigerated and for up to six hours when kept at room temperature.


RESUMO: O estudo teve como objetivo avaliar os locais adequados de laparocentese para a colheita de fluido peritoneal de bovinos e estabelecer o tempo de viabilidade celular in vitro, comparando três métodos de conservação. Vinte e um bovinos hígidos (19 fêmeas e 2 machos) foram submetidos ao procedimento de laparocentese para obtenção de fluido peritoneal, com punção em três pontos definidos: cranial esquerdo, cranial direito e caudal direito. O volume total do líquido peritoneal foi dividido em três alíquotas mantidas sob três métodos de conservação: temperatura ambiente (26°C); refrigeração (4°C); e temperatura ambiente (26°C) com adição de 1L de formol 10% para cada 1mL de líquido peritonial. A análise do líquido peritoneal realizada imediatamente após sua obtenção consistiu em: exames físico (cor, aspecto, volume e densidade); bioquímicos (pH, proteína total, fibrinogênio, creatinina e glicose); e da celularidade (contagens total e diferencial). A determinação de proteínas e o exame da celularidade foram repetidos, em cada alíquota separada, as duas, quatro, seis e oito horas após a colheita. Análise de variâncias de medidas repetidas ou teste de Friedman foram empregados para avaliação ao longo do tempo. A colheita foi produtiva em 67% dos bovinos e os locais de punção craniais esquerdo e direito foram os mais adequados. A concentração de proteína total variou de 1,4 a 3,6g/dL e o número de leucócitos de 54 a 1.322 células/L, com predomínio de linfócitos (60 a 95% das células) no fluido peritoneal analisado logo após a colheita. A concentração de proteínas diminuiu, mas os valores absolutos de leucócitos, de linfócitos e de neutrófilos segmentados não se modificaram até oito horas após a colheita, independente do método de manutenção das amostras. A lise celular foi retardada pela refrigeração e a adição de formol não contribuiu para preservar a integridade da morfologia celular. A laparocentese é um procedimento seguro e de execução fácil em bovinos sendo mais produtiva quando realizada em locais específicos à esquerda ou à direita craniais da parede abdominal. Amostras de fluido peritoneal podem ser analisadas com resultados confiáveis quando mantidas refrigeradas por até oito horas após a colheita e quando mantidas à temperatura ambiente por até seis horas.

Two experiments were conducted to evaluate the effects of -mannanase on egg production performance, egg quality, intestinal microbiota, viscosity, and ammonia concentration in laying hens. In Exp. 1, two hundred and seventy 30-wk-old Hy-Line Brown laying hens were assigned to 6 diets arranged in a 3 × 2 factorial of three levels of MEn and CP [(a corn-soybean meal based diet (HEHP), a diet containing 50 kcal of MEn/kg and 1.0% less energy and CP than the HEHP (MEMP), and a diet containing 80 kcal of MEn/kg and 1.5% less energy and CP than HEHP (LELP)], and -mannanase supplementation (0 or 0.04%). In Exp. 2, A total of two hundred and sixteen62-wk-old Hy-Line Brown laying hens were assigned to 6 dietary treatments in a 3 × 2 factorial arrangement. In Exp. 1, -mannanase supplementation increased egg production rate in hens fed LELP diet, but not in those fed HEHP or MEMP diet (interaction, p 0.01), and the interaction was significant (p 0.01) for egg mass. -mannanase supplementation decreased (p 0.05) ammonia concentration. In Exp. 2, the supplementation of -mannanase increased egg production rate and egg mass in hens fed LELP diet, whereas no differences were found in those fed HEHP or MELP diet (interaction, p 0.01). The supplementation of -mannanase in a lower energy and protein diet resulted in similar production performance when compared to high-energy and high-protein diet during early and late stages of egg production.(AU)

ABSTRACT: The objective of this study was to evaluate the enzymatic activity of plasma cholinesterase in Chelonia mydas marine turtles belonging to two populations, according to their capture sites, under the absence and probable influence of anthropic effects. A total of 74 animals were used and later divided into two groups, based on the capture site. Blood samples were collected from all captured animals, which were then released into the sea at the site of capture. A descriptive statistical analysis of the plasma cholinesterase activity values and an analysis comparing these values based on the capture site were performed. Samples of heparinized plasma from animals captured at the two different sites were analyzed. Plasma cholinesterase activity ranged from 121 to 248U/L, with a mean and standard deviation of 186.1±30.68U/L. When comparing plasma cholinesterase activity values in individuals based on the capture site, a significant difference was observed. Establishing reference values for different sea turtle populations is necessary to interpret future sampling results and to allow sea turtles to be used as sentinels of ecosystem health. Future studies are needed to evaluate other populations and the activity of plasma cholinesterase in juvenile marine turtles, in relation to environmental contamination.


RESUMO: O objetivo desse estudo foi avaliar a atividade enzimática da colinesterase plasmática em tartarugas marinhas da espécie Chelonia mydas em duas populações de acordo com o local de captura, sob ausência e provável influência de efeito antrópico. Foi utilizado um total de 74 animais e posteriormente divididos em dois grupos de acordo com o local de captura. Foram coletadas amostras de sangue de todos os animais capturados e em seguida liberados ao mar no mesmo local. Foi realizada uma análise estatística descritiva dos valores da atividade plasmática de colinesterase do total de animais e análise comparando os valores de acordo com o local de captura. Foram analisadas amostras de plasma heparinizado de animais capturados em dois locais distintos. Os valores da atividade plasmática de colinesterase variaram de 121 a 248U/L, com média e desvio padrão de 186.1±30.7U/L. Quando comparados os valores de atividade plasmática da colinesterase nos indivíduos de acordo com o local de captura, foi observada diferença significativa. O estabelecimento de valores de referência para diferentes populações de tartarugas marinhas são necessários para interpretar os futuros resultados amostrais e permitir que as tartarugas marinhas sejam usadas como sentinelas da saúde do ecossistema. Estudos futuros são necessários para avaliar outras populações e a atividade da colinesterase plasmática de tartarugas marinhas juvenis em relação à contaminação ambiental.

ABSTRACT: Monofluoroacetate (MFA)-containing plants cause sudden death associated with exercise in ruminants, and are responsible for significant losses in Brazilian livestock, estimated at 500,000 bovine deaths annually. Most of the times, the control and treatment of this type of poisoning are not efficient, because disease evolution is superacute, usually causing the death of the animal. Due to the difficulty in controlling this intoxication, several studies have suggested alternatives to prevent it, mainly by making animals resistant to the MFA present in these plants or by avoiding their consumption. This literature review addresses the techniques used experimentally to control the poisoning of ruminants by plants containing MFA. The first studies carried out in Brazil demonstrated that goats and sheep that continuously receive non-toxic doses of plant containing MFA show greater resistance to poisoning than untreated animals, and that this resistance can be transmitted by ruminal fluid transfaunation, suggesting that poisoning occurs due to the presence of bacteria that hydrolyze MFA in the rumen. Based on this hypothesis, several MFA-hydrolyzing bacteria were isolated (Enterococcus faecalis, Bacillus sp., Paenibacillus sp., Burkholderia sp., Cupriavidus sp., Staphylococcus sp., Ancylobacter sp., Ralstonia sp., Stenotrophomonas sp., Pigmentiphaga kullae, and Ancylobacter dichloromethanicus). When some of these bacteria were administered intraruminally, they provided the animal with a different level of protection against poisoning. However, it was observed that protection is gradually lost when the bacterium administration is interrupted. Consequently, to obtain more efficient protection, these bacteria should be administered continuously, probably in the form of probiotics. In another assay, MFA was administered to sheep at non-toxic doses to test the hypothesis that this substance could induce the multiplication of bacteria that hydrolyze it in the rumen. There was no increase in resistance to poisoning after administration of MFA; however, no signs of poisoning were observed when animals received trifluoroacetate and no clinical signs were verified when they were challenged with toxic doses of MFA; in contrast, all control animals presented clinical signs. These results suggest that trifluoroacetate induces the proliferation of MFA-degrading bacteria, and can be used in intoxication prophylaxis. The conditioned food aversion technique, using lithium chloride, has been successfully used experimentally to prevent ruminants from ingesting plants that contain MFA. Another alternative tested was the spraying of Amorimia septentrionalis with the endophytic bacterium Herbaspirillum seropedicae, which degrades MFA, resulting in decreased concentration of this compound in the plants. In conclusion, several experimental techniques have been proved efficient in the control and prophylaxis of MFA-containing plant poisoning; however, none of these techniques are available commercially. Further experiments, mainly in the field, should be carried out to adapt some of these techniques to the conditions of extensive breeding in the numerous areas where MFA-containing plants occur.


RESUMO: As plantas que contém monofluoroacetato (MFA) causam morte súbita associada ao exercício em ruminantes, e são responsáveis por grandes perdas na pecuária brasileira, estimadas em 500.000 mortes de bovinos anualmente. O controle e tratamento desse tipo de intoxicação, na maioria das vezes, não apresenta eficiência, visto que a evolução da doença é superaguda, e geralmente ocasiona a morte do animal. Devido à dificuldade no controle dessa intoxicação, diversos estudos sugerem alternativas para preveni-la, principalmente tornando os animais resistentes ao MFA presente nessas plantas ou evitando seu consumo. O objetivo do presente trabalho é fazer uma revisão bibliográfica das técnicas utilizadas experimentalmente para controlar a intoxicação de ruminantes por plantas que contém MFA. Nos primeiros trabalhos realizados no Brasil, foi determinado que caprinos e ovinos que recebem continuadamente doses não tóxicas de planta que contém MFA apresentam maior resistência a intoxicação que animais não tratados e que essa resistência pode ser transmitida por transfaunação de fluído ruminal, sugerindo que a mesma ocorre devido a presença de bactérias que hidrolisam MFA no rúmen. Com base nessa hipótese foram isoladas diversas bactérias que hidrolisam MFA (Enterococcus faecalis, Bacillus sp., Paenibacillus sp., Burkholderia sp., Cupriavidus sp., Staphylococcus sp., Ancylobactersp., Ralstoniasp., Stenotrophomonas sp., Pigmentiphaga kullae e Ancylobacter dichloromethanicus). Quando algumas dessas bactérias foram administradas intraruminalmente conferiram diferentes graus de proteção contra a intoxicação. No entanto foi observado que a proteção se perde gradualmente quando se deixa de administrar a(s) bactéria(s). Em consequência, para obter uma proteção mais eficiente essas bactérias deveriam ser administradas continuadamente, provavelmente na forma de probiótico. Em outro ensaio administrou-se MFA a ovinos em doses não tóxicas para testar a hipótese de que esta substância poderia induzir a multiplicação de bactérias que hidrolisam o mesmo no rúmen. Não houve um aumento da resistência a intoxicação após a administração de MFA; no entanto quando foi administrado trifluoroacetato, os animais não desenvolveram nenhum sinal de intoxicação e quando desafiados com doses tóxicas de MFA não apresentaram sinais clínicos, pelo contrário todos os animais controles apresentaram sinais clínicos. Esses resultados sugerem que o trifluoroacetato induz a proliferação de bactérias que degradam MFA e pode ser utilizado para a profilaxia da intoxicação. A técnica da aversão alimentar condicionada, utilizando cloreto de lítio, tem sido empregada experimentalmente, com sucesso, para evitar que ruminantes ingiram plantas que contém MFA. Outra alternativa testada foi a pulverização de Amorimia septentrionalis com a bactéria endofítica Herbaspirullum seropedicae, que degrada MFA, resultando na diminuição da concentração deste composto na planta. Conclui-se que há diversas técnicas que experimentalmente tem demonstrado eficiência no controle e profilaxia das intoxicações por plantas que contém MFA; no entanto, nenhuma dessas técnicas está disponível comercialmente. Futuros experimentos, principalmente, a campo, deverão ser realizados para adaptar alguma(s) dessas técnicas as condições de criação extensiva nas numerosas áreas onde ocorrem plantas que contém MFA.

Heat stress induces oxidative stress, and reduces body antioxidant metabolite levels, which can affect poultry production performance. Dietary antioxidants protect birds against the adverse effects of heat stress. The effects of increasing concentrations of dietary curcumin on the antioxidant parameters of layers maintained under high-temperature conditions for nine weeks were evaluated. Roman laying hens (n = 336, 22 weeks old, 1420 g BW) were divided into three treatment groups. The first group served as a thermoneutral control (kept at 25 ± 1 °C). The second group was exposed to high temperatures (32 ± 1 °C, 6 h/d), given a basal diet. The third group was further divided into five treatment groups (100, 150, 200, 250, 300 mg/kg Curcumin) fed a basal diet (treatments H1, H2, H3, H4, H5) under high temperatures conditions (32 ± 1 °C, 6 hours/day). As a result of this study, total superoxide dismutase activity was significantly higher in H2 and H3 groups, and total antioxidant capacity was higher in H2, H3, and H5 groups. Catalase and glutathione peroxidase activity was significantly higher in the H3 group. Malondialdehyde concentration was lowered in curcumin supplemented hens compared with control groups hens. Laying hens in all curcumin treatment groups had slightly higher activities of CAT, SOD, GSH-Px, and T-AOC in the liver, heart, and lungs, compared with heat stressed control group. It was concluded that dietary curcumin given to laying hens under heat stress may enhance their antioxidant status, and alleviate the detrimental effects of stressful environmental conditions.(AU)

Purpose:To investigate the effect of mesenteric lymph drainage on the spleen injury and the expressions of inflammatory cytokines in splenic tissue in mice following hemorrhagic shock.Methods:Male C57 mice were randomly divided into the sham shock, shock and shock+drainage groups. The mice in both shock and shock+drainage groups suffered femoral artery bleeding, maintained mean arterial pressure (MAP) of 40±2 mmHg for 90 min, and were resuscitated. And mesenteric lymph drainage was performed in the shock+drainage group at the time of resuscitation. After three hours of resuscitation, the splenic tissues were harvested for the histological observation and protein and mRNA expression analysis of cytokines.Results:The spleen in the shock group revealed a significantly structural damage and increased mRNA expressions of MyD88 and TRAF6 and protein expressions of TIPE2, MyD88, TRIF and TRAF3 compared to the sham group. By contrast, the splenic pathological injury in the shock+drainage group was alleviated significantly, and the mRNA and protein expressions of TIPE2, MyD88, TRIF, TRAF3 and TRAF6 were significantly lower than those in the shock group.Conclusion:These results indicate that post-hemorrhagic shock mesenteric lymph drainage alleviates hemorrhagic shock-induced spleen injury and the expressions of inflammatory cytokines.(AU)

This study was conducted to investigate the effects of dietary oxidized oil and natural polyphenol antioxidants on laying performances and egg quality of laying hens. A total of two hundred, 61-week-old Lohmann Brown laying hens were divided into five groups, consisting in four replicates of 10 birds each. They were fed one of the five experimental diets (fresh oil only, oxidized oil without antioxidants, oxidized oil with vitamin E 200 ppm, oxidized oil with vitamin E 100 ppm + Cabanin CSD 100 ppm, oxidized oil with Cabanin CSD 500 ppm)respectively, for 6 weeks. There were no significant differences in laying performances and egg weight among the treatments. However, egg mass in group fed oxidized oil with vitamin E 100 ppm + Cabanin CSD 100 ppmwas significantly higher than group fed oxidized oil only (p 0.05). Eggshell thickness and eggshell strength was not affected by feeding of oxidized oil and antioxidants. Haugh unit checked after storage of 14 d from the end date of the trial showed significant difference (p 0.05). Serum superoxide dismutase (SOD) like activity was significantly higher in antioxidant treated groups. The level of serum glutamic oxaloacetic transaminase (GOT) was significantly lower in groups fed vitamin E 100 ppm + Cabanin CSD 100 ppm or Cabanin CSD 500 ppm compared to groups fed oxidized oil without antioxidant. The number of total microbes, lactobacilli, and Enterocossus faecium showed significant difference favored to antioxidants groups. This study indicates that addition of oxidized oil to layer diet may have harmful effect on oxidative status and egg quality, but concurrent addition of vitamin E and natural polyphenol can alleviate the toxic effect of oxidized oil.(AU)

Resumo

Purpose: To investigate the relations of neuropeptide Y (NPY) and heme oxygenase-1 (HO-1) expressions with fetal brain injury in rats with intrahepatic cholestasis of pregnancy (ICP). Methods: Sixty rats pregnant for 15 days were randomly divided into experimental and control groups. The ICP model was established in experimental group. On the 21st day, the blood biochemical test, histopathological examination of pregnant rat liver and fetal brain tissues and immunohistochemical analysis of fetal rat brain tissues were performed. Results: On the 21st day, the alanineaminotransferase, aspartate aminotransferase and total bile acid levels in experimental group were significantly higher than control group (P<0.01). Compared with control group, there was obvious vacuolar degeneration in pregnant rat liver tissue and fetal brain tissue in experimental group. NPY expression in fetal brain tissue was negative in control group and positive in experimental group. HO-1 expression in fetal brain tissue was strongly positive in control group and positive in experimental group. There was significant difference of immunohistochemical staining optical density between two groups (P<0.01). Conclusion: In fetal brain of ICP rats, the NPY expression is increased, and the HO-1 expression is decreased, which may be related to the fetal brain injury.(AU)

ABSTRACT: Herbaspirillum seropedicae is a nitrogen-fixing bacterium capable of using toxic compounds as a source of carbon. Bacteria with this capacity can be used to make animals resistant to plant poisoning containing monofluoroacetate (MFA), such as Amorimia septentrionalis. The aim of this study was to evaluate if H. seropedicae is efficient in the degradation of MFA present in A. septentrionalis and if the inoculation of this bacterium in goats confers protection to A. septentrionalis intoxication. Two experiments were performed: in the first experiment 12 goats were divided into 2 groups. Goats in Group 1 were orally administered a solution containing the H. seropedicae bacterium for 10 days. From day 10 onwards, they received a daily dose of 5g/kg of A. septentrionalis with the bacteriauntil clinical signs of intoxication were observed. Group 2 goats received only the plant at the same dose, also until the observation of clinical signs of intoxication. The amount of MFA found in A. septentrionalis used in the experiment with goats was 1.6±0.058g/mg. The total plant dose ingested by all goats in Group 1 was 80.83±12.81g/kg (129.33±20.50mg/kg MFA), which were significantly greater (p 0.05) than those of Group 2 goats (39.16±19.08g/kg plant and 62.66±30.53mg/kg MFA). Group 1 goats took an average of 16.16±2.56 days to develop clinical signs of intoxication, significantly longer (p=0.0012) than Group 2 goats (7.83±3.81 days). Two Group 2 goats died on the same day that they developed clinical signs of intoxication. At necropsy of these two animals, no significant changes were observed. In the second experiment, samples of A. septentrionalis were sprayed with a solution containing H. seropedicae. Before and eight days after spraying, the samples were pressed and dried for quantitation of MFA. The amount of MFA present in samples of A. septentrionalis 8 days after spraying with H. seropedicae was significantly lower (p=0.017) than that found prior to spraying. It can be concluded that administration of H. seropedicae in goats is capable of causing greater resistance to A. septentrionalis intoxication, and spraying the plant with this bacterium significantly reduces the amount of MFA in the plant.


RESUMO: Herbaspirillum seropedicae é uma bactéria fixadora de nitrogênio, capaz de utilizar compostos tóxicos como fonte de carbono. Bactérias com essa capacidade podem ser utilizadas para tornar os animais resistentes à intoxicação por plantas que contém monofluoroacetato (MFA), como Amorimia septentrionalis. O objetivo do presente estudo é avaliar se H. seropedicae é eficiente na degradação do MFA presente em A. septentrionalis e se a inoculação dessa bactéria, em caprinos, confere proteção à intoxicação por A. septentrionalis. Foram realizados dois experimentos: no primeiro experimento foram utilizados 12 caprinos, divididos em dois grupos. Os caprinos do Grupo 1 receberam diariamente, oralmente, uma solução contendo a bactéria H. seropedicae durante 10 dias. A partir do décimo dia passaram a receber, diariamente, além da solução com a bactéria 5g/kg de A. septentrionalis até a observação de sinal clínico de intoxicação. Os caprinos do Grupo 2 receberam apenas a planta na mesma dose, também até que a observação de sinais clínicos de intoxicação. A quantidade de MFA encontrada em A. septentrionalis utilizada no experimento com caprinos foi de 1,6± 0,058µg/mg de planta em média. A dose total de planta ingerida por todos os caprinos do Grupo 1 foi de 80,83±12,81g/kg (129,33±20,50mg/kg de MFA), valores significativamente maiores (p 0,05) do que os dos caprinos do Grupo 2 (39,16±19,08g/kg de planta e 62,66± 30,53mg/Kg de MFA). Os caprinos do Grupo 1 demoraram em média 16,16 ±2,56 dias para desenvolver sinais clínicos da intoxicação, período significativamente maior (p=0,0012) que os caprinos do Grupo 2 (7,83±3,81dias). Dois caprinos do Grupo 2 morreram no mesmo dia que desenvolveram sinais clínicos da intoxicação. Na necropsia desses dois animais não foram observadas alterações significativas. No segundo experimento, amostras de A. septentrionalis foram pulverizadas com uma solução contendo a bactéria H. seropedicae. Antes e oito dias após a pulverização, as amostras foram prensadas e secas para posterior quantificação do MFA. A quantidade de MFA presente nas amostras de A. septentrionalis oito dias após a pulverização com H. seropedicae foi significativamente menor (p=0,017) do que a encontrada antes da pulverização. Pode-se concluir que a administração de H. seropedicae em caprinos é capaz de causar uma maior resistência à intoxicação por A. septentrionalis, e a pulverização da planta com esta bactéria reduz significativamente a quantidade de MFA na planta.

ABSTRACT: Clinical and metabolic evaluation is necessary for the monitoring of pregnant and lactating mares, as they reflect the health of the animal. The body condition of the mare is an indicator of reproductive efficiency. The study aimed to determine the possible variations in body and metabolic condition in Mangalarga Marchador mares during the transition period. Forty-eight mares distributed in two groups were used: Maintenance Group (MG), composed of non-pregnant and non-lactating mares, and Transition Group (TG), formed by pregnant mares and who after delivery became lactating. Analyzes were performed in the times T-60, T-30 and T-15 before delivery, first six hours (T0) after delivery and T15, T30 and T60 days after delivery. MG was evaluated only at one time (T-60). Body weight and fat-free mass differed (P 0.05) between the groups. The percentage of fat was lower in MG. Mares had a higher fat percentage in TG at T-60 and T-30 times. There was a difference (P 0.05) in the amount of cholesterol between MG and TG (T0, T15 and T30). Triglycerides were different between the groups. TG showed higher concentrations of non-esterified fatty acids (P 0.05). There was a higher amount of glucose in TG (delivery and lactation) when compared to mares in MG. Changes in body condition and metabolic constituents occurred in the animals resulting from physiological adaptations of the transition period. The energetic components are the most affected from the transition period, with intense fat mobilization to supply the body demands.


RESUMO: Avaliações clínicas e metabólicas são necessárias para o monitoramento de éguas gestantes e lactantes, pois refletem a saúde do animal. A condição corporal da égua pode ser um indicador da eficiência reprodutiva. O estudo objetivou determinar as possíveis variações na condição corporal e metabólica em éguas Mangalarga Marchador durante o período de transição. Foram utilizadas 48 éguas distribuídas em dois grupos: Grupo em Manutenção (GM) composto por éguas não gestantes e não lactantes; Grupo em Transição (GT) formado por éguas gestantes e que após o parto tornaram-se lactantes. As análises foram realizadas nos tempos T-60, T-30 e T-15 antes do parto, primeiras seis horas (T0) após o parto e T15, T30 e T60 dias após o parto. O GM foi avaliado apenas uma vez (T-60). O peso corporal e a massa livre de gordura diferiram (P 0,05) entre os grupos. O percentual de gordura foi menor em GM. As éguas apresentaram maior porcentagem de gordura no GT no T-60 e no T-30. Houve diferença (P 0,05) na quantidade de colesterol entre GM e GT (T0, T15 e T30). Triglicérides foram diferentes entre os grupos. GT apresentou maiores concentrações de ácidos graxos não esterificados (P 0,05). Houve maior quantidade de glicose no GT (parto e lactação) quando comparada às éguas no GM. Mudanças na condição corporal e nos constituintes metabólicos ocorreram nos animais resultantes de adaptações fisiológicas do período de transição. Os componentes energéticos são os mais afetados neste período, existindo intensa mobilização de gordura para suprir demandas corporais.
