Resumo

Background: Goat farming has been on the rise in Brazil in recent years. Overall, 93% of the national herd is concentrated in the Northeast, with the state of Paraíba being the largest goat milk producer in the country. Considering Mycobacterium avium subsp. paratuberculosis (MAP) as a sanitary issue for the development of animal farming with risks for human health and that is a notifiable disease, this research was structured with the objective of confirming the presence and performing a molecular characterization of MAP in goat milk destined for processing plants in the semiarid region of the Brazilian Northeast. Materials, Methods & Results: Samples from 179 production units and 5 collective bulk tanks and 4 samples of pasteurized goat milk were analyzed through Real-time polymerase chain reaction (qPCR). Genetic material (DNA) for MAP was found in the goat milk sample from 1 production unit (1/179). From this positive sample, 9 lactating goats were identified in the original property, 7 of which showed MAP DNA in milk samples (77.77%). The characterization of the nucleotide sequence detected in the positive sample has 99% identity with KJ173784. Discussion: One sample (1/179), from the production units, had MAP genetic material (DNA) detected using the molecular test. Samples from these production units represent the milk from all lactating goats from each producer. Therefore, it was possible to identify from which farm the samples originated, allowing individual animals to then be tested, with milk samples collected from 9 goats and MAP DNA detected in 7 of them (77.77%) via PCR. Control and/or prevention programs need this type of surveillance in reason that it allows the tracking of possible foci from milk samples collected from dairy products or cooling stations. The use of PCR to detect MAP foci via goat milk is thus advantageous because samples are obtained in a non-invasive manner, with faster results when compared to the culture technique. The low detection via PCR in goat milk may be related to factors such as the small amount of MAP eliminated and the intermittent excretion in asymptomatic animals, as also false-positive samples. Samples from the collective bulk tanks was negative. It is possible that the combination of milk from all the properties diluted the amount of MAP. This suggests that the sensitivity of the PCR can be improved if the samples are obtained from the pooled milk from the same property. In some regions of Brazil, for example, showed the frequency of Zona da Mata region of the state of Minas Gerais, Brazil, found 1.94% of positive samples (9/464) and 9.76% (4/41) of properties with at least 1 positive sample for MAP. Different results to what were found in the semiarid region of Paraíba, where climate and production characteristics are different. Goats are susceptible to 3 strains: type "S" (Sheep), "Bison type" and type "C" (Cattle). Previous contact with this species may explain the similarity between the strain found in goat milk and those detected from bovine samples. This must also be taken into consideration during diagnosis and upon implementation of control measures for paratuberculosis in goats. Mycobacterium avium subsp. paratuberculosis was recorded for the first time in goat milk in the semiarid region, which may reveal a potential biological risk to humans and suggests the need for active surveillance of the agent.

Background: Leptospirosis is an important infectious disease in goat farming, with a worldwide distribution. It is usually transmitted by rodents and the genital route, may cause reproductive losses, negatively impacting goat farming. The diagnosis lies on serological, molecular and isolation techniques. Considering the importance of this disease for small ruminants, this work aimed to evaluate the serological, molecular findings and isolation of pathogenic leptospires in the urinary tract (kidney and bladder tissues) of goats. Materials, Methods & Results: Thirty-four adult goats were used for slaughter. Renal samples (n = 34), bladder (n = 34), were collected for isolation of the agent and molecular detection of Leptospira sp. and blood samples (n = 34) for serological testing. The polymerase chain reaction (PCR) was used as a molecular test and the microscopic serum agglutination test (MAT) was used as a serological test. Samples with DNA amplification were subjected to genetic sequencing. The presence of Leptospira DNA was found in the tissues of 8 (23.4%) goats, and of these, only 2 were positive in PCR and MAT. There was a slight agreement between the PCR and MAT techniques (k = 0.150; P = 0.436). In 6 (17.6%) samples of renal tissue and 2 (5.8%) bladder samples, Leptospira DNA was detected. The genes in a kidney tissue sample were sequenced and demonstrated 99% similarity to Leptospira interrogans. Anti-Leptospira sp. were detected in 6 (17.6%) of the animals tested. Discussion: Serology identified 3 predominant serogroups: Icterohaemorrhagiae, Tarassovi and Autumnalis, serogroups that are related to the presence of rodents that coexist in rural environments. Autumnalis has been reported in small ruminants, raising the hypothesis that goats are adapted, becoming chronic carriers and possible maintenance hosts. The frequency obtained (17.6%) may be the result of the mixed breed pattern and rustic characteristics inherent to the goat species. Given the characteristics of the semi-arid region, such as low rainfall and high solar incidence, it is essential to use an adapted methodology, with a lower cut-off point (1:50), as the serological titer is an established relationship between the animal species, the level of exposure throughout its evolution and the region studied. Molecular findings and bacterial isolation reveal the agent's ability to colonize the urinary tract of goats. These data show the importance that urine has in the epidemiological chain, being able to transmit the agent through direct contact with this product or through contamination of soil and water. There was no statistical agreement between the diagnostic techniques used in this study, in this case, an association between PCR and MAT is recommended to obtain data with high sensitivity and specificity. A bladder sample was sequenced and showed 99% similarity to Leptospira interrogans. In the semiarid region, the most common form of leptospirosis spread is through the sale of animals in business fairs for breeding, rearing or slaughter, as well as sharing the same property with several breeders. The introduction of chronic and asymptomatic carriers on the properties represents a serious risk for the spread of the disease. The results show the presence of Leptospira spp. in semi-arid goat herds, having as risk factors the presence of rodents and intercropping. The association of MAT and PCR is necessary for a better diagnosis of the disease.

Caprine arthritis encephalitis and Maedi-Visna are lentiviruses affecting goats and sheep, respectively. Despite the literature having studies about these diseases, there is a constant demand and the need to study the health status of flocks that exploit economically. Therefore, this study aimed to assess the frequency of small ruminant lentiviruses explored in regional locations of Chapadinha and Itapecuru Mirim, that compose the microregion of Low Parnaíba, Maranhão, Brazil, as well as analyze the spatial distribution of outbreaks in the studied regions. Therefore, 241 properties were visited, where blood was collected in 1150 sheep and 1260 goats and tested by agar gel immunodiffusion (AGID). Epidemiological questionnaire was applied and collected the geographic coordinates. There was a low frequency for lentivirus, with 0.39% (5/1260) of goats and 0.08% (1/1150) of sheep. Regarding the spatial analysis, the reagent flocks were distributed in strategic cities for commercialization throughout the microregion. There was a low occurrence of lentiviruses. The municipalities of Cantanhede and Pirapemas of the regional of Itapecuru Mirim and Brejo and Magalhães de Almeida had reagent flocks for CAE. Whereas the municipality of Matões do Norte presented flock reagent to Maedi-Visna, this belonging to the regional of Chapadinha.

Apesar da literatura apresentar estudos sobre essas doenças, existe uma demanda constante e a necessidade de se estudar o estado de saúde dos rebanhosexploradoseconomicamente. Portanto, este estudo teve como objetivo avaliar a frequência de lentivírus de pequenos ruminantes explorados nasregionais de Chapadinha e Itapecuru Mirim, que compõem a microrregião do Baixo Parnaíba, Maranhão, Brasil, bem como analisar a distribuição espacial dos surtos nas regiões estudadas. Para tanto, foram visitadas 241 propriedades, onde foi coletado sangue de 1150 ovelhas e 1260 cabras e testado por imunodifusão em gel de ágar (IDGA). Foi aplicado questionário epidemiológico e coletado as coordenadas geográficas. Houve baixa frequência para lentivírus, com 0,39% (5/1260) de caprinos e 0,08% (1/1150) de ovinos. Quanto à análise espacial, os rebanhos reagentes foram distribuídos em cidades estratégicas para comercialização em toda a microrregião. Houve baixa ocorrência de lentivírus. Os municípios de Catanhede e Pirapemas da regional de Itapecuru Mirime Brejo e Magalhães de Almeida possuíam rebanhos reagentes para CAE. Já o município de Matões do Norte apresentou reagente rebanho à Maedi-Visna, esta pertencente à regional de Chapadinha.

The objective of this work was to evaluate the efficiency technique of fixation of the caudal curvature of the sigmoid flexure with myectomy of the penis retractor muscle and the performance of teaser bulls after surgery, through the detection of females in estrus, without performing the copulation. 12 teaser buffaloes were used without defined breed, with an average age of 20 months, weighing on average 200 kg. The animals were subjected to food and water fasting for 12 hours to provide tranquilization in advance. The surgical procedure consisted of the exteriorization of the sigmoid flexure, myectomy of  penis retractor muscle and fixation of the caudal curvature of the penis sigmoid flexure. Of the 12 evaluated teaser buffaloes in the libido test, 100% performed  the  flehmen reflex, mount attempt, incomplete and complete jump  with total inability of  the penis exposure.    It was concluded that this technique can be used safely, having  simplicity in its execution, effective results and mainly low cost, not interfering in the animals libido. The animals when tested as teaser buffaloes kept the libido during the experimental period with total inability of exposure of the penis.


O objetivo do presente trabalho foi avaliar a técnica de fixação da curvatura caudal da flexura sigmóide com miectomia do músculo retratordo pênis e o desempenho de rufiões bubalinos após cirurgia, através da detecção de fêmeas em estro, sem executar a cópula. Foram utilizados 12 bubalinos sem raça definida, com idade média de 20 meses, pesando em média 200 kg. Os animais foram submetidos ajejum alimentar e hídrico de 12 horas para tranquilização prévia. O procedimento cirúrgico constou naexteriorização da flexura sigmóide, miectomia do músculo retrator do pênis e fixação da curvatura caudal da flexura sigmóide do pênis. Dos 12 rufiões avaliados no teste de libido, 100% realizaram reflexode flehmen, tentativa de monta, salto incompleto e completo com total incapacidade de exposição do pênis. Conclui-se que esta técnica pode ser utilizada com segurança, apresentando simplicidade na sua execução, eficácia nos resultados e principalmente baixo custo, não interferindo na libidodos animais. Os animais quando testados como rufiões mantiveram a libido durante o período experimental com total incapacidade de exposição do pênis.

Leishmaniases are a group of diseases of zoonotic importance caused by over 20 species of protozoa of the genus Leishmania, in which domestic dogs are considered to be the main reservoir for the disease. However, the involvement of other vertebrates as reservoirs for these parasites has also been investigated. Therefore, the objective of the present study was to carry out a systematic review with meta-analysis on occurrences of leishmaniasis in equids. The case reports described animals with cutaneous symptoms of leishmaniasis (papules, nodules, ulcers or crusts) that regressed spontaneously, located mainly on the head and limbs, from which three species of protozoa were identified in the lesions: Leishmania braziliensis, Leishmania infantum and Leishmania siamensis. In turn, the meta-analysis showed a combined prevalence of 25%, although with high heterogeneity among the studies, which was attributed to the use of different methods for diagnosing the disease. Leishmaniasis in equids is a benign disease but it should be included in the differential diagnosis of cutaneous diseases among these species. Seroepidemiological studies are important in investigating and monitoring suspected exposure of these hosts to the parasite, especially in endemic areas. However, there is also a need to standardize diagnostic methods.(AU)


As leishmanioses são um grupo de doenças de importância zoonótica causadas por mais de 20 espécies de protozoários do gênero Leishmania, sendo o cão doméstico considerado o principal reservatório da doença. No entanto, diversas pesquisas têm investigado o envolvimento de outros vertebrados como reservatórios do parasita. Portanto, o objetivo do presente estudo foi realizar uma revisão sistemática com meta-análise da ocorrência de leishmaniose em equídeos. Os relatos de caso descreviam animais com sintomas cutâneos de leishmaniose (pápulas, nódulos, úlceras, crostas) que regrediam espontaneamente, localizadas principalmente na cabeça e membros, sendo identificadas três espécies do protozoário nas lesões: Leishmania braziliensis, Leishmania infantum e Leishmania siamensis. Por sua vez, a meta-análise evidenciou uma prevalência combinada de 25%, porém com alta heterogeneidade entre os estudos, atribuída às diferenças nos métodos utilizados no diagnóstico da doença. A leishmaniose em equídeos é uma doença benigna, porém deve ser incluída no diagnóstico diferencial de doenças cutâneas nessas espécies. Os estudos soroepidemiológicos são importantes para investigar e monitorar a suspeita de exposição desses hospedeiros ao parasita, principalmente em áreas endêmicas, porém há necessidade de padronização dos métodos de diagnóstico.(AU)

The objective of this research was to estimate the frequency, risk factors and mapping of outbreaks in ovine flocks in the State of Maranhao, Brazil, related to Leptospira spp. A total of 575 samples of blood serum from sheep were collected, from 116 flocks, and 20 municipalities. An epidemiological questionnaire was applied to verify the association of risk factors. Microscopic Agglutination Test (MAT) was used for 24 serovars of Leptospira spp. 100% of the flocks (116) and 94.43% of animals (543/575) were reagents for at least one of the 24 serovars tested. The most frequent serovars were Sentot (236, 41.04%), Hadjo (197, 34.26%), Adamana (178; 30.95%), Pomona (158, 27.47%) and Autumnallis (148, 25.73%). The presence of rodents, felines, goats, pigs and wild animals had a statistically significant association (p < 0.05) with the occurrence of Leptospira spp. It was concluded that the frequency of Leptospiraspp was high in sheep from the State of Maranhao. However, all municipalities had infected animals, therefore, it is considered necessary to implement sanitary measures for the control and prevention of this disease in ovine flocks in the state of Maranhao.
