Leptospirosis is an endemic zoonotic disease that is distributed worldwide, which has the potential to have health and economic impacts. Leptospira spp. is spiral-shaped and capable of infecting mammals, including horses, which may result in asymptomatic or clinical forms. Therefore, the current study aimed to analyze the frequency of anti-Leptospira antibodies in serum samples from horses from Santarém, Pará, Brazil. For that purpose, 88 blood samples from horses without a history of leptospirosis vaccination were serologically evaluated through the microscopic agglutination technique (MAT) with a 13 serovars, belonging to ten different serogroups. There were 58 samples that were seropositive (65.90%), which included 28 samples seropositive for Pyrogenes (48.3%), 24 for Autumnalis (41.4%), 18 for Icterohaemorrhagiae (31.0%), and 16 for Grippotyphosa (27.6%). Even without clinical suspicion of leptospirosis or a history of vaccination, the horses showed different frequency of seropositivity. Considering the well-known impact of leptospirosis in human and animal health, our results are important to establish preventive measures to reduce the economic loss in equine production as well as a reduction in public health risk.

Fungal dermatitis is classified into superficial, cutaneous and subcutaneous mycoses. However, the incidence of fungal dermatitis in small mammals is relatively low. Among rodents guinea pig the most affected specie, however it usually has an asymptomatic pattern. The present text reports an unusual case of fungal dermatitis in Caviaporcelluscaused by Scopulariopsis brevicaulis. The animal was received with complaint of alteration of coat around the neck, as well as report of incorrect feeding management. During the physical examination the presence of a dry and crusty dermatitis, negative on fluorescence test of wood. Samples of blood, skin, scabs and hair were collected for parasitic and mycological analysis. The hematological evaluation showed only a discrete eosinophilia; no ectoparasites were observed, but it was noticed the presence of Scopulariopsis brevicaulis. The animal underwent terbinafine and griseofulvin protocol, with total regression of the lesions after 30 days. S. brevicaulis is an unusual geophilic fungus causing animal mycosis, presenting more pathological reports in human mycoses, thus, representing a zoonotic potential. According to the clinical findings and results obtained from mycology assay, it was diagnosed dermatitis by Scopulariopsis brevicaulis, secondary to nutritional deficiency. The combined use of topical terbinafine with systemic griseofulvin was efficient in the treatment. Finally, it was recommended a dietary correction.

The occurrence of leptospirosis is significantly higher in the tropical climate than in temperate regions, especially in periods with high rainfall levels. In horses, leptospirosis can range from subclinical to asymptomatic but maintains an important role in the transmission of leptospirosis by eliminating the agent in the environment. Regarding horses, the Amazonas herd increased by 30% between 2004 and 2013. Thus, the study aimed to determine the serological prevalence of leptospirosis in horses in Manaus and metropolitan regions, in the State of Amazonas, Brazil as well as to assess the main serogroups involved in the infections of these animals. For this purpose, from August 2018 to July 2019, serum from 198 horses was evaluated through the microscopic agglutination test with a panel of 10 serogroups. As a result, 92 horses (44.46%) were found to be positive for one or more Leptospira serovars, with the highest prevalence of serogroups Icterohaemorrhagiae and Pyrogenes. Therefore, with the increase in the equine herd in the region and, consequently, greater interaction between breeders and animals, the identification of reagents to at least one serovar of Leptospira spp. signals the possible existence of reservoirs of pathogenic strains for other animals and man.(AU)

Modern swine production employs a high degree of technology and is organized in various production stages, in which reproduction is one of the most significant. However, reproductive losses associated to fetal death are still high. Fetal losses, including fetal mummification, may occur at a rate below 1.5% in a sound herd. The causes of fetal death can be very diverse, and include various infectious agents. Therefore, investigating the causes of fetal losses is important in the decision-making process related to the control and prophylaxis of the herd. The goal of this study was to carry out a molecular survey, analyzing porcine circovirus 2 (PCV2), porcine parvovirus (PPV), and pathogenic Leptospira as potential agents related to fetal death in swine farms in Southern Brazil. We collected samples from three farms with a mummified index ≥ 2.5%. Fragments of brain, lung, kidney, liver, and heart were sampled for PCR analysis. Out of 100 samples examined, 87, 68, and 22 were positive for PCV2, PPV, and Leptospira spp., respectively. Moreover, we also identified coinfections with two and three pathogens in the same sample. Our findings contribute to the spreading of scientific knowledge related to infectious causes (PPV, PCV2, and pathogenic leptospires) of fetal losses in swine, as evidenced by the high frequencies of the investigated agents.(AU)

Background: Leptospirosis remains the most widespread zoonotic disease in the world. As a clinical entity it is strongly associated with regional occupational and environmental exposures. While the exact global disease burden remains unknown, recent estimates by the leptospirosis Burden Epidemiology Reference Group (LERG) at the World Health Organization have set the number of human cases of severe leptospirosis to over 500,000 per year. This number almost certainly represents an under-representation due to poor surveillance and difficult diagnosis. Leptospira spp. belong to the order Spirochaetales, family Leptospiraceae, composed by 13 pathogenic Leptospira species with more than 260 serovars. Wildlife species are commonly considered to be important epidemiological carriers, mainly because of their frequent reactivity to Leptospira serovars native to their habitat. Capybara (Hydrochaeris hydrochaeris), a known leptospirosis host, is a widespread species in South America. However, reports regarding the importance of this animal in the epidemiology of leptospirosis are rare. Therefore, the objective of this study was to report the results of leptospirosis serological survey of capybaras from a residential park area in southeastern Brazil. Vesicular stomatitis virus (VSV) is the agent of a vesicular disease that affects many animal species and may be clinically confounded with foot-and-mouth disease in ruminant and swine. Horses are especially susceptible to VSV and may serve as sentinels for virus circulation. The present study investigated the presence of neutralizing antibodies against VSV Indiana III (VSIV-3) in serum samples of 3,626 horses from six states in three Brazilian regions: Southern (RS, n = 1,011), Midwest (GO/DF, n = 1,767) and Northeast (PB, PE, RN and CE, n = 848) collected between 2013 and 2014. Neutralizing antibodies against VSIV-3 (titers ≥40) were detected in 641 samples (positivity of 17.7%; CI95%:16.5-19.0%), being 317 samples from CE (87.3%; CI95%: 83.4-90.5 %); 109 from RN (65.7%; CI95%: 57.8 -72.7%); 124 from PB (45.4%; CI95%: 39.4-51.5%); 78 from GO/DF (4.4%; CI95%: 3.5-5.5%) and nine samples of RS (0.9%; CI95%: 0.4-1.7%). Several samples from the Northeast and Midwest harbored high neutralizing titers, indicating a recent exposure to the virus. In contrast, samples from RS had low titers, possibly due to a past remote exposure. Several positive samples presented neutralizing activity against other VSV serotypes (Indiana I and New Jersey), yet in lower titers, indicating the specificity of the response to VSIV-3. These results demonstrated a relatively recent circulation of VSIV-3 in northeastern Brazilian States, confirming clinical findings and demonstrating the sanitary importance of this infection.(AU)

This study aimed to identify the parasitical etiologic agents of visceral cysts in pigs from the central/northern region of Rio Grande do Sul State, Brazil. Fifty-eight cysts were found in livers during veterinary inspection of swine slaughtered from January 2008 to 2012. Collected samples were submitted to macroscopic and molecular analyzes. Polymerase chain reaction (PCR), DNA sequencing and BLAST alignment of sequences was used to molecular characterization of the samples. By PCR 10.3% (6/58) of tested samples were positive for Echinococcus granulosus sensu lato and 56.9% (33/58) for Cysticercus tenuicollis. In this study, it was verified the occurrence of larval forms of E. granulosus sensu lato and Taenia hydatigena in pig herds from the central/northern region of Rio Grande do Sul State. The presence of both parasites is relevant due to the economic losses for the meat industry. Additionally, E. granulosus sensu lato has zoonotic importance and may be infecting pig herds in southern Brazil.(AU)

Neosporosis is an infectious disease caused by the parasite Neospora caninum. Knowledge regarding neosporosis in goats is still quite limited, especially in the state of Santa Catarina (SC), southern Brazil. Therefore, this study aimed to assess the seroprevalence and risk factors for N. caninum in goats in the western and mountain regions of SC. Blood samples were collected from 654 goats in 57 municipalities. The indirect immunofluorescence test was used for antibody detection against N. caninum. Thirty samples (4.58%) were seropositive, with titers ranging from 1:50 to 1:6400. An epidemiological survey was also conducted in order to identify risk factors for neosporosis in goats. It was found that reproductive problems on the farms, as well as the diet and direct contact with dogs were casual risks for neosporosis. These results indicate that N. caninum infects goats in these regions, which may lead to reproductive problems.
Background: Trypanosoma evansi is a flagellate which belongs to the salivate section, commonly observed parasitizing blood of equines, ruminants, pigs, dogs and wild animals in different regions of the world. It causes many losses to farmers due to death of animals and drug spending in endemic areas. The treatment of this disease in Brazil is only performed with diminazene aceturate; however it has been ineffective for many animals. During the last years many studies have been carried out with natural products such as the essential oils. Copaiba oil stands out due some properties described as anti-inflammatory, healing, antiedematogenic, antitumor, parasitic and antibacterial. Therefore, this study aimed to test, in vitro and in vivo, the susceptibility of T. evansi to copaiba oil. Materials, Methods & Results: The oils used in this study were obtained from Copaifera reticulata and Copaifera duckei trees, commonly found in the Tapajos National Forest. The procedure received authorization of IBAMA due the scientific purposes. This study identified three oils identified as copaiba 4-C (C. reticulata), copaiba 5-C (C. duckei) and copaiba 8-C (C. reticulata). The bioassay was performed in vitro using specific culture medium for T. evansi, previously described by Baltz, 1985. Copaiba oil was dissolved in dimethyl sulfoxide (DMSO) and tested in three concentrations (0.5, 1.0 and 2.0%) in culture medium containing the parasite. To the control test (without oil) the same volume of DMSO (10 µL) was added. Dimnazene aceturate was also used as a positive control at 0.5% of concentration. The counting of trypanosomes was performed in triplicate in a Neubauer chamber after 1, 3, and 6 hours after the experiment onset. For the tests in vivo, mice were infected (n = 40) and divided in 5 groups of 8 animals each. Group A consisted of healthy animals and Group B comprised animals infected with T. evansi and untreated. The other groups were infected and treated orally with oil of copaíba 4-C (Group C), copaiba 5-C (Group D) and copaiba 8-C (Group E), using a dose of 0.63 mL/kg/day for five consecutive days at intervals of 24 hours. In vitro tests with copaiba oil showed a reduction in the number of alive trypanosomes for the three tested concentrations, when compared to the control test after 1 and 3 h, similar to what occurred with testing aceturate. At 6 hours, it was not observed alive parasites in the test groups, differently from the control group which had an increase of trypanosomes compared to the time zero. The trypanocidal activity had a dose-dependent effect. In the in vivo experiment, the oil of copaiba administered orally had no curative efficacy for any of the groups; however group D treated with C. duckei showed prolonged longevity of the mice when compared to the groups B and C. Discussion: Trypanosoma cruzi and Leishmania amazonensis have been challenged on their susceptibility to oil of copaiba, with obtention of trypanocidal and leishmanicidal effects, similar process thet occurred in this study with T. evansi. According to scientific literature the copaíba oil increases the membrane permeability, as well as provides the depolarization of the mitochondrial membrane in parasite cells. A group of mice in this study showed prolonged longevity, showing that the variation of their compositions influence the trypanocidal effect. Based on these results it was concluded that the T. evansi may is susceptible to the oil of copaiba. Therefore, it can be natural product used as a new alternative and supplementar activity in the treatment of this protozoan, as have been suggested for leishmaniasis.

Background: Perinephric pseudocyst is a rare disease that affects animals and humans. It is characterized by an accumulation of serous fluid around of one or both kidneys, and in cats the manifestation as sub-capsular, containing liquid with transudate characteristics are most frequently observed. The etiology is not well understood yet, but it is known that it is associated with chronic renal injury. While it has no direct relation to race and gender, it shows a higher occurrence in elderly patients above 10 years. According to the scientific literature, surgery is the treatment of choice for this condition. Therefore, the objective of this paper is to report the first case of bilateral perinephric pseudocyst in a young cat in Brazil, treated with palliative maneuver associated with a therapeutic feeding balanced. Case: A 1-year-old Siamese cat, 3 kg, female non-castrated was referred for clinical investigation in the state of Rio de Janeiro, Brazil. It presented a 30 days history of progressive increase of abdominal volume, followed also by weight loss, hyporexia, and apathy. The patient underwent to an abdominal ultrasound, showing a large cyst in both kidneys, suggestive of perinephric pseudocyst. It was found that the surgical therapy would the choice for such alterations; however, the animal owners we do not accepted surgical treatment, were oriented about the risks of recurrence a treatment palliative. Nevertheless, recognizing the risks, they preferred by choose for a minimally invasive treatment. With this clinical challenge, we established a supportive treatment, with fluid therapy and commercial therapeutic feeding. At day of diagnosis of perinephric pseudocyst, which we consider as day 0, blood sample was collected for evaluation hemogram and biochemical serum, tests revealed normochromic normocytic anemia and azotemia. Elapsed 10 days of supportive therapy, the liquid was aseptically drained, being classified as a transudate. It was performed only two collection of perirenal liquid within a interval of 10 days between them. After the second perinephric pseudocyst draining the animal did not return to the veterinary clinic, since the owners had reported, by phone call, the improvement of the animal. The reports were maintained during three months and two years after the diagnostic, with no longer observation of abdominal enlargement. Discussion: The abdominal ultrasound proved to be an effective diagnostic method for the differentiation and confirmation of PNP e the liquid analysis revealed pure transudate. These results are similar to those described by other authors. Laboratory tests showed normocytic normochromic anemia and azotemia. The azotemia may be prerenal, by dehydration, as observed in this report; however, as shown in other studies of conical renal injury, obstruction by calculus can lead to this azotemic condition. Thus, it was possible to observe in this study, corroborating the literature, that the prognosis is related to the degree of renal dysfunction. Since this animal was a young cat, when the dehydration was, corrected along with the percutaneous puncture of liquid, the animal's response was immediate and satisfactory. The levels of urea and creatinine declined, with consequent decrease in accumulation of liquid in cysts. Another fundamental process in the therapeutic care, was the combination of mechanical therapy with therapeutic feeding, providing lower renal overload of minerals and a greater supply of proteins with high digestibility to the animal. It is important to emphasize that it is not recommended to replace the surgery by this protocol, but it is expected with this report demonstrate the importance of this rare disease in our country, highlighting that a palliative maneuver in association with changes in diet are therapies that can be employed with favorable prognosis.

Background: The mite Ornithonyssus bursa is a bloodsucking parasite of domestic and wild birds and it is typical of tropical regions. On the host it can cause considerable loss of blood and lesions with intense itching. Cases of parasitism in human beings have been reported causing dermatitis, and most of these cases are due to the presence of nests of synanthropic birds near of the residences. When in large infestations or lack of hosts, the mites leave the nests to perform the parasitism in humans. Human infestation occurs mainly in people who handle infected birds and nests, or when happen the dispersion of these mites, through some path which lead to the interior of homes, factories and hospitals. Case: The present case occurred in November of 2008, when one of the dwellers of an apartment on the fourth floor of a building downtown in Santa Maria, woke up with pruritic lesions over the arms and legs. Upon inspection of the bedroom, several mites were seen, measuring almost a millimeter long, coming out of a pipe of a portable air conditioner installed on site. They were already scattered by the inside walls, floor and bed. The mites were collected and maintained in 70% alcohol for later identification. After the clarification the slides mounting were performed, and the mites with measure between 0.48 mm wide by 0.74 mm long were classified by the position of the anal opening, formed by the dorsal plate and three pairs of bristle on the sternal plate, as belonging to the species Ornithonyssus bursa. On external outlet of the air portable conditioner, a nest with a couple of birds and three cubs was found. The birds were identified as belonging to the species Myiodynastes maculatus, popularly known as brindle Bem-Te-Vi. The birds were removed from the place and treated with antiparasitic macrolactones to control the infestation. Discussion: In the present case report the O. bursa was found inside of an apartment in the downtown area, parasitizing birds which had their nest sheltered in the outlet air of a portable air conditioner. The presence of synanthropic bird brindle Bem-Te-Vi and the ease of access to the interior of the residence led to a home infestation of this ectoparasite. This fact may be happend due to the nesting of birds in urban buildings, seeking for a favorable environment for their survival, specially near of local that providing heat, like heaters, furnaces and hot water pipes. When the young birds parasitized by hematophagous leave the nests, the mites can cause the parasitism in humans mainly by food shortage. In the present study the occurrence of accidental home infestation and accidental parasitism in humans is related to the presence of many specimens of birds in the same nest, with offering of food and favorable environmental conditions for reproduction of the mites and their dissemination. Mite infestations in humans are characterized by erythematous papulovesicles distributed throughout the body, mainly on the limbs and chest. In this case report the parasitism caused erythematous lesions, especially in the arms with intense itching. The dermatitis produced by mesostigmatas hematophagous mites are not specific impeding the diagnosis, and may be confusing with other ectoparasites. For this reason the present diagnosis was performed by isolation and identification of the mite. Based on the description above it was possible to conclude that the bird Myiodynastes maculatus are parasitized by the mite Ornithonyssus bursa and its synanthropic habits allowed the infestation of mites in this residence.

This study aimed to test an alternative protocol with human plasma to control Trypanosoma evansi infection in mice. Plasma from an apparently 27-year-old healthy male, blood type A+, was used in the study. A concentration of 100 mg.dL-1 apolipoprotein L1 (APOL1) was detected in the plasma. Forty mice were divided into four groups with 10 animals each. Group A comprised uninfected animals. Mice from groups B, C and D were inoculated with a T. evansi isolate. Group B was used as a positive control. At three days post-infection (DPI), the mice were administered intraperitoneally with human plasma. A single dose of 0.2 mL plasma was given to those in group C. The mice from group D were administered five doses of 0.2 mL plasma with a 24 hours interval between the doses. Group B showed high increasing parasitemia that led to their death within 5 DPI. Both treatments eliminated parasites from the blood and increased the longevity of animals. An efficacy of 50 (group C) and 80% (group D) of human plasma trypanocidal activity was found using PCR. This therapeutic success was likely achieved in the group D due to their higher levels of APOL1 compared with group C.(AU)

Background: Leptospirosis, a spirochetal zoonotic disease caused by different serovars of Leptospira interrogans, is increasingly recognized as an important cause of hemorrhagic fever. Although the haemorrhagic potential of leptospirosis was noted by Weil (1886) as early as 1886, its pathophysiology is still not clearly elucidated, particularly regarding the cause and mechanisms of bleeding. Studies with ectonucleoside triphosphate diphosphohydrolase (NTPDase; EC 3.6.1.5; CD39), 5'-nucleotidase (EC 3.1.3.5; CD73) and adenosine deaminase (ADA) have demonstrated the involvement of these enzymes in thromboregulation mechanisms, and altered enzymatic activities have been reported in many diseases. Since leptospirosis is a disease increasingly recognized as an important cause of hemorrhagic fever, the aim of this study was to evaluate these enzymes activities and parameters of platelet aggregation in platelets from rats experimentally infected with Leptospira interrogans serovar icterohaemorrhagiae during different periods of experimental infection. Materials, Methods & Results: For this purpose, thirty-six adult male rats were divided into two groups: A, as uninfected control (subgroups A1, A2 and A3); and B, infected (subgroups B1, B2 and B3). Group B was inoculated intraperitoneally (Day 0) with 2 x 108 organisms per rat. Blood samples were collected on days 05 (A1 and B1), 10 (A2 and B2) and 20 (A3 and B3) post-inoculation (PI). In the infected group, platelet count had a decrease on day 10 PI and prothrombin time (PT) had an increase on day 5 PI. In the same group, platelet aggregation decreased (P < 0.01) day 10 PI. The hydrolysis of ATP in platelets was also decreased (P < 0.05) on day 10 PI, when compared to the control group. By the other side, ADP hydrolysis was increased (P < 0.05) on days 5 and 10 PI. 5'-nucleotidase activity was significantly increased on day 5 (P < 0.01) and 20 (P < 0.05) PI. Results of adenosine deamination into inosine by ADA in platelets showed a signifi cant (P < 0.01) increase on days 5 and 10 PI in the infected group. Discussion: Studies with NTPDase, 5'nucleotidase and ADA have demonstrated the involvement of these enzymes in the thromboregulation mechanisms, and altered enzymatic activities have been reported in many diseases. It has been established that extracellular adenosine nucleotides and adenosine are versatile signaling molecules known to participate in an array of platelet functions. For example, the nucleotide ADP is the main promoter of platelet aggregation, while adenosine can act as a vasodilator and an inhibitor of platelet aggregation. In addition, high concentrations of ATP have been shown to inhibit ADP-induced aggregation in vitro, while low concentrations of ATP can significantly enhance platelet aggregation. In our experimental study the coagulation cascade was activated, since when the activities of NTPDase, 5'-nucleotidase and ADA were analyzed is possible to suggest that levels of ATP were decreased, unlike of ADP and AMP levels, supposedly increased during determinate periods of our experiment. Adenosine levels were also enhanced due to the higher levels of its precursors. This cascade activation may be a mechanism of bleeding prevention front to leptospires infection, especially the ones caused by serovar icterohaemorrhagiae.
