Resumo

The objective of this study was to evaluate the effectiveness of common antibiotics against different microorganisms in apparently healthy cattle in Shandong province and its suburb. A total of 220 nasal swab samples were collected and cultured for bacteriological evaluation. All the bacteria isolates after preliminary identification were subjected to antibiogram studies following disc diffusion method. It was found in the study that E. coli is the most commonly associated isolate (21%), followed by Klebsiella spp. (18%), Pseudomonas aeruginosa (13%), Salmonella spp. (15%), Shigella spp (12%), and Proteus spp (11%). While the antibiogram studies reveled that highest number of bacterial isolates showed resistance to Ampicillin (95%), followed by Augmentin (91%), Cefuroxime (85%) and Tetracycline (95%) of (Escherichia coli and Klebsiella spp). In the case of pseudomonas spp. and Salmonella the highest resistance was showed by Ampicillin (90%) followed by Amoxicillin + Clavulanic Acid (80%), Cefixime (90%), and Erythromycin (80%). In Shigella spp and Salmonella spp highest resistance was showed by Amoxicillin, Ceftazidime, Augmentin (60%), and Amoxicillin + Clavulanic Acid (50%). It is concluded that in vitro antibiogram studies of bacterial isolates revealed higher resistance for Ampicillin, Augmentin, Cefuroxime, Cefixime, Tetracycline, Erythromycin, and Amoxicillin + Clavulanic Acid. The high multiple Antibiotics resistance indexes (MARI) observed in all the isolates in this study ranging from 0.6 to 0.9. MARI value of >0.2 is suggests multiple antibiotic resistant bacteria and indicate presence of highly resistant bacteria.


O objetivo deste estudo foi avaliar a eficácia dos antibióticos comuns contra diferentes microrganismos em bovinos aparentemente saudáveis na província de Shandong e seus subúrbios. Um total de 220 amostras de esfregaço nasal foi coletado e cultivado para avaliação bacteriológica. Todos os isolados de bactérias após identificação preliminar foram submetidos a estudos de antibiograma seguindo o método de difusão em disco. Os altos índices de resistência a antibióticos múltiplos (MARI) observados em todos os isolados neste estudo variaram de 0,6 a 0,9. O valor MARI de > 0,2 sugere várias bactérias resistentes a antibióticos e indica a presença de bactérias altamente resistentes.

Resumo

This experiment was conducted to study the effects of arginine supplementation on organ development, egg quality, blood parameters, and immune status of laying hens. A total of 360 25-week-old brown Leghorn laying hens were randomly divided into three groups with six replicates of 20 birds each and fed diets supplemented with 0, 8.5, or 17 mg of L-arginine/kg for 42 days. Results showed that the weight of proventriculus and duodenum in the treatment supplemented with 17 mg/kg L-arginine was heavier than that of 0 mg L-arginine/kg treatment (p 0.05). The weight of oviduct in the treatments supplemented with 17 mg/kg L-arginine was smaller than that of 0 and 8.5 mg L-arginine/kg treatment (p 0.05), and the small yellow follicle amount in the treatment supplemented with 17 mg/kg L-arginine was less than that of 0 mg L-arginine/kg treatment (p 0.05), while its yolk color was deeper than that of 0 mg L-arginine/kg treatment (p 0.05), and the IgY content showed the same tendency. Total cholesterol and triglyceride levels in the treatment supplemented with 8.5 mg/kg L-arginine were lower than that of 0 mg L-arginine/kg treatment (p 0.05). The concentration of IL-2 in the treatment supplemented with 17 mg/kg L-arginine were more than that of 0 mg L-arginine/kg treatment (p 0.05). The findings of this study show that 17 mg/kg L-arginine supplementation has beneficial effects onlayers' immune status and yolk IgY content, as well as on proventriculus and duodenum weight while no adverse effects were observed on laying performance, egg quality, or blood parameters.

Peptide toxins are usually highly bridged proteins with multipairs of intrachain disulfide bonds. Analysis of disulfide connectivity is an important facet of protein structure determination. In this paper, we successfully assigned the disulfide linkage of two novel peptide toxins, called HNTX-III and HNTX-IV, isolated from the venom of Ornithoctonus hainana spider. Both peptides are useful inhibitors of TTX-sensitive voltage-gated sodium channels and are composed of six cysteine residues that form three disulfide bonds, respectively. Firstly, the peptides were partially reduced by tris(2-carboxyethyl)-phosphine (TCEP) in 0.1 M citrate buffer containing 6 M guanidine-HCl at 40° C for ten minutes. Subsequently, the partially reduced intermediates containing free thiols were separated by reversed-phase high-performance liquid chromatography (RP-HPLC) and alkylated by rapid carboxamidomethylation. Then, the disulfide bonds of the intermediates were analyzed by Edman degradation. By using the strategy above, disulfide linkages of HNTX-III and HNTX-IV were determined as I-IV, II-V and III-VI pattern. In addition, this study also showed that this method may have a great potential for determining the disulfide bonds of spider peptide toxins.(AU)
