Resumo

Hepatitis C virus (HCV) is the serious global public health burden of liver disease. Approximately 170 million people in the world are infected with (HCV). In Pakistan, where the disease has high occurrence rate. The present study envisages an up-to-date prevalence of HCV and genotypic distribution in the general population of Mardan District, Khyber Pakhtunkhwa (KP), Pakistan. The blood samples from 6,538 individuals including 3,263 males and 3,275 females were analyzed for hepatitis C surface antigen by Immuno-chromatographic test (ICT), Enzyme-linked immunosorbent assay (ELISA), and reverse transcription-polymerase chain reaction (PCR). It was found that 396 (12.13%) out of 3263 individuals contained antibodies in their blood against HCV, while among the different age groups, the highest incidences of HCV antibodies were found in the 31-40 age group (11.01%). The ICT positive samples were further screened by nested PCR to determine the existence of active HCV-RNA. It was identified that 7.11% (3263) of the total population (6538) tested was positive, among which the 461 (14.07%) females possessed antibodies in their blood against HCV. Our data showed total HCV infection in the investigated population was 5.78%. Higher percentage of HCV prevalence was detected in males than females in the age group 31-40 and 41-50. To compare the prevalence of HCV genotypes age-wise in male and female genotype 3a was found most prevalent genotype followed by 1a, 2a and 3b, respectively.

As amostras positivas para ICT foram posteriormente rastreadas por nested PCR para determinar a existência de HCV-RNA ativo. Identificou-se que 7,11% (3.263) do total da população (6.538) testada foram positivos, dentre os quais 461 (14,07%) mulheres possuíam anticorpos no sangue contra o HCV. Nossos dados mostraram que a infecção total pelo HCV na população investigada foi de 5,78%. It was observed that 169 (7.04%) out of (2399) male of the total population (4803) tested was positive. On the other hand, the female 463 (19.25%) possessed antibodies in their blood against HBV. Accumulatively, our results showed a higher percentage of HBV prevalence in males than females in the age group 21-30 years. The total HCV infected in Mardan general population was recorded at 5.7% comprising both male and female.

Observou-se que 169 (7,04%) de 2.399 homens do total da população (4803) testados foram positivos. Por outro lado, 463 mulheres (19,25%) possuíam anticorpos no sangue contra VHB. Acumulativamente, nossos resultados mostraram uma porcentagem maior de prevalência de HBV em homens do que em mulheres na faixa etária de 21 a 30 anos. O total de HCV infectados na população geral de Mardan foi registrado em 5,7%, incluindo homens e mulheres.(AU)

Resumo

This study investigated the effect of calcium (Ca) and phytase interaction on growth performance and bone quality in 1-42-day-old broiler chickens. A total of 624 female one-day-old Ross 308 broilers were allotted to 13 treatments with four replicates and 12 birds per replicate. A 2 × 6 factorial experiment was designed to test the combinations of 0.50% and 1.00% Ca with 0, 500, 1,000, 2,500, 5,000, and 10,000 FTU/kg phytase in the basal diet (0.25% non-phytate phosphorus, NPP). The control diet contained adequate Ca and phosphorus (P). Dietary Ca, phytase, and their interaction affected growth performance and bone mineralization of broilers at 1-42 days of age (p<0.05). The broilers fed with 1.00% Ca had lower body weight gain (BWG) and feed intake (FI) compared with the birds fed with 0.50% Ca (p<0.05). The BWG, FI, leg bone weight, and ash weight of the broilers fed with 0.25% NPP were lower than those of birds fed with the control diet (p<0.05). The addition of 500-10,000 FTU/kg phytase improved growth rate and leg bone quality, especially at 1.00% Ca (p<0.05). No differences were observed in growth performance and bone quality of 42-day-old broilers fed with 1.00% Ca + 2,500-10,000 FTU/kg phytase and the control diet (p>0.05). These data indicated that high doses of phytase (2,500-10,000 FTU/kg) alleviate the negative effects of Ca and P imbalance (Ca-to-NPP ratio = 4.0) on growth performance and bone mineralization of broiler chickens.(AU)

The current study was conducted to develop the correlations between muscle fiber characteristics and meat quality attributes in the biceps femoris muscle of Arbor Acres (AA) and Yellow-feathered chicken (YFC). A total of fourty pure breed birds of AA (n=20) and YFC (n=20) were used in the experiment. After slaughtering at their respective market age of slaughtering: AA 40 d and YFC 120 d, samples were collected for meat quality attributes and myosin ATPase staining for fiber types analyses. Meat quality attributes and muscle fiber characteristics i-e; diameter, cross-sectional area (CSA) and density of AA were significantly different from the YFC (p 0.05). Type I fibers number percentage was significantly higher in YFC than AA, whereas CSA and fiber diameter were higher in AA (p 0.05). Negative correlations were obtained between lightness (L*) and type I fiber number percentage in AA (p 0.05). In YFC fiber number percentage, CSA and diameter of type IIA were negatively correlated with Warner-Bratzler Shear Force. Taken together, muscle fibers characteristics of AA and YFC differ in both breeds and have influenced the meat quality attributes.(AU)

This experiment aimed to investigate whether Escherichia coli (E. coli) infection could affect the TGF-/smads signaling pathway in the jejunal tissue of chickens. One-day-old Cobb 500 broilers were randomly divided into 2 groups and treated with intraperitoneal E. coli or broth injection. Clinical signs of the birds were assessed every day. Spleen and bursa of Fabricius of the birds, post-infection (pi), were collected to evaluate immune organ index. Jejunal tissues were collected to ascertain the expression of TGF-s, TRs, and Smads. The results showed that the infected birds had significantly higher index of the spleen (24hrs and 48hrs pi) compared with birds in the control group (p 0.05). The relative gene expression of TGF-4 increased (p 0.05), while the expression of Smad7 down-regulated in the E. coli group (p 0.01). There was no significant difference in TGF-2, TGF-3, TR I, TR II, Smad2, Smad3 expression (p>0.05). In conclusion, TGF-/Smad signaling pathway was associated with the immune response of broilers in E. coli infection and TGF-4 was the main subtype interacting with E. coli infection.(AU)

This study aims to investigate whether resveratrol (RES) could inhibit NF-κB activity and protect in vitro cultured chicken lymphocytes from apoptosis induced by continuous LPS stimulation. Blood lymphocytes of chickens were collected and cultured. LPS was added to the samples of treatment group, while the same volume of normal saline (NS) was supplemented to those of the control group. In the treatment groups, four different concentrations of RES (0, 20, 40, and 80 µg/ml) were admixed. Then, the following indicators were tested: lymphocyte apoptotic rate, lymphocyte viability, NF-κB activity, TNF-a level, reactive oxygen species (ROS), and expression of apoptosis-related proteins (Fas/FasL and Caspase-8). The results indicated that the application of different concentrations of RES could significantly increase the viability of lymphocytes and decrease the apoptotic rate (p<0.05) after continuous stimulations by LPS. The activity of NF-κB levels of TNF-a and ROS in the RES treatment subgroups was significantly decreased in comparison with that in the RES-blank subgroup (p<0.05). The cells in the treatment group that had been treated with 20, 40 and 80 μl/ml of RES exhibited a significantly lower Fas and Caspase-8 expression level than that in the RES-blank subgroup (p<0.05). These findings revealed that RES could substantially diminish the concentrations of ROS and TNF-a, down-regulate NF-κB activity, decrease the expression of Fas and Caspase-8, stimulate the activity of peripheral lymphocytes, and lower the rate of their apoptosis, induced by continuous LPS stimulation in chicken lymphocyte cultures.(AU)

This study has the objective of investigating the effects of traditional Chinese medicine prescriptions (TCM) on serum lipid, abdominal and hepatic fat percentage, cholesterol content in eggs, and mRNA expression of genes apoA I and apoB100. One hundred and thirty five healthy (300-day-old) layers were randomly assigned to three treatments. The hens in control group were fed with the basal diet. The hens in the experimental groups (TCM 1 and TCM 2) were fed with the basal diet supplemented with 1% TCM 1 and 1% TCM 2 respectively over a period of 60 days. Laying performance and the serum parameters relevant to fat metabolism were measured. The results showed that no significant differences were found in average daily feed intake and egg weight among three treatments. Average daily laying rate in TCM treatments was increased, and the cholesterol content in eggs was decreased. The serum triglyceride (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) levels in experimental treatments were decreased (p<0.05), while the serum high-density lipoprotein cholesterol (HDL-C) level was increased (p<0.05) compared to the control group. Additionally, abdominal fat percentage decreased in TCM 1 treatment (p<0.05), and hepatic fat percentage decreased in both TCM treatments (p<0.05). The expression of apolipoproteinA I (apoA I) and apolipoproteinB100 (apoB100) mRNA in the liver increased in both TCM treatments (p<0.05). These results suggest that the diet supplemented with TCM could increase the expression of apoA I and apoB100 mRNA in the liver, and decrease lipid content in the serum, and reduce egg cholesterol in layers.(AU)

Orphan nuclear receptors, those without known ligands, were discovered because of their structural similarity to the ligand-driven steroid and thyroid receptors. Since their characterization, many of the orphan receptors have been adopted, i.e., ligands, usually lipids or derived lipids, have been discovered. The orphan receptors are transcriptional regulators, functioning in the reproductive context to upregulate or suppress gene expression. By this means, the orphan receptors regulate a plethora of reproductive events. In the majority of cases, the effects are stimulatory, indeed, members of the NR2 family promote Leydig cell differentiation and testicular steroidogenesis, while those of the NR4 family regulate early gestation and placental formation. The NR5 family has two members, steroidogenic factor-1 (SF-1, NR5A1) and liver receptor homolog-1 (LRH-1, NR5A2). These receptors interact with the same DNA sequence and are believed to be constitutive transcription factors. Their effects are modulated by the repressive effects of the NR0 family of orphan receptors that comprise the short heterodimeric partner (SHP, NR0B2) and dosage-sensitive sex reversal adrenal hypoplasia congenital region on the X chromosome, gene 1 (DAX1, NROB1). SHP and DAX1 inhibit the interaction of LRH-1 and SF-1 with coactivators, thereby reducing their constitutive transcriptional effects. Overall, the orphan nuclear receptors are essential regulators of reproductive function in mammals.
