Resumo
Background: Intravaginal devices containing progestins are widely used for oestrus synchronization in sheep. Progestins give economic benefits to farmers but may have some limitations and the efficacy strictly depends on farm management. There are different devices, with different molecules (progesterone, medroxyprogesterone acetate, fluorogestone acetate), different dosages and long (12-14 days) and short-term (5-7 days) protocols. Experimental studies often include a limited number of animals and are held at different latitudes and with different system of management. To our knowledge, there are few reports in the literature on field application of the recently licensed 20 mg fluorogestone acetate (FGA) sponges in large ewe flocks, excluding the registration trials. Materials, Methods & Results: This study was designed to evaluate oestrous synchronization in 2 different breeds. A total of 1100 Lacaune (L) and 618 Sarda (S) were assigned to 8 groups, comparing multiparous (M) and nulliparous (N) and those synchronized in November (n) and May (m). The groups were: LMm (n. 556), LNm (n. 180), SMm (n. 70), SNm (n. 32), LMn (n. 242), LNn (n. 222), SMn (n. 440) e SNn (n. 76). The intravaginal sponge was inserted for 14 days. At sponge withdrawal, eCG (400 IU, IM) was injected, and rams were joined into flocks 30 h later with a male/female ratio of 1:8. Transrectal ultrasonography was performed for pregnancy diagnosis after 30 days. Fertility parameters such as oestrus (OR), pregnancy (PR), lambing rates (LR) and prolificacy were calculated. Fisher exact test was used to compare parameters of each group with the corresponding and significance was set at P < 0.01. The use of intravaginal sponges in this study was easily performed in all animals. In nulliparous groups, digital insertion of the sponge, instead of the applicator, was preferred to avoid discomfort. Sponge loss was observed in 2% of treated ewes. At the removal of the sponge, mild vaginal contamination was observed in 90% of the ewes. Ultrasound finding of embryo resorption, pseudopregnancy and pyometra were found in rates under 2%. The loss of pregnancy ranged from 0 to 3.6% without significant differences among groups. The SMn group showed the best performances (0.91 OR, 0.88 PR, 0.85 LR), significantly higher than the other groups. Prolificacy was maximum in the LMn (1.68) and minimum in the SNm group (1.06), without significant differences among groups. Discussion: The 14-day regimen based on 20 mg FGA-releasing intravaginal devices is an easy and satisfactory synchronization regimen to improve the productivity and the fertility of sheep farm, and this can be managed and optimized in different breeding conditions. In this study, Sarda breed shows a good adaption to the environment with adult ewes giving the best results in reproductive season. However young Sarda ewes were less productive especially in non-breading season. Lacaune showed good reproductive potential, young and adult ewes responded to progestins in reproductive and non-reproductive season. It was confirmed that the reproductive performance is affected by season, age, and breed. This study encourages the breeding of indigenous breeds by implementing the management with modern technologies. The imported highly productive breeds, if well managed, can give a production less influenced by seasonal variables and age.

The combination of medroxyprogesterone acetate (MPA) and gonadotropin chorionic (eCG) has been widely used to synchronize oestrus cycle in sheep, but their effects on the gene expression in uterine tissue are yet to be elucidated. To evaluate the effect of MPA + eCG or prostaglandin analogue (PA) treatments on the rate of oestrus cycle synchronization, as well as further hormone production and gene expression profiles in uterine tissue, 14 Santa Inês ewes were randomly selected. The MPA + eCG group (n=7) received intravaginal insertion of MPA-impregnated sponges for 14 days and was administered 350 IU eCG on the day of sponge withdrawal. The PA group (n=7) was administered two doses of 100 µg of PA separated by 12 days. The ewes were assessed for the rate of oestrus cycle synchronization and the serum concentrations of progesterone (P4) and estradiol (E2). Additionally, the expression of estrogen receptor (ERα), progesterone receptor (P4R), and immunolocalization of interferon receptor (IFNAR1) in the uterine tissue samples collected 15th day post-mating were examined. The rate of oestrus cycle synchronization was 100% (n=7/7) and 57.14% (n=4/7) in the MPA + eCG and PA groups, respectively. Moreover, the MPA + eCG group exhibited higher serum concentration of P4 than the PA group (p < 0.05). However, the E2 serum concentration did not differ between the two groups (p > 0.05). The relative expression of P4R and ERα mRNA analyzed using real-time PCR and immunodetection of IFNAR1 were similar between the two groups tested (p > 0.05). Conclusively, MPA + eCG treatment improved the rate of oestrus cycle synchronization and endogenous P4 production; however, it did not affect the expression of sex steroid receptors and IFNAR1 in uterine ovine tissue.(AU)

The environment contributes to production diseases that in turn badly affect cow performance, fertility and culling. Oestrus intensity is lower in lame cows, and in all cows 26% potential oestrus events are not expressed (to avoid getting pregnant). To understand these trade-offs, we need to know how animals react to their environment and how the environment influences hypothalamus-pituitary-adrenal axis (HPA) interactions with the hypothalamus-pituitary-ovarian axis (HPO). Neurotransmitters control secretion of GnRH into hypophyseal portal blood. GnRH/LH pulse amplitude and frequency drive oestradiol production, culminating in oestrus behaviour and a precisely-timed GnRH/LH surge, all of which are disrupted by poor environments. Responses to peripheral neuronal agents give clues about mechanisms, but do these drugs alter perception of stimuli, or suppress consequent responses? In vitro studies confirm some neuronal interactions between the HPA and HPO; and immuno-histochemistry clarifies the location and sequence of inter-neurone activity within the brain. In both species, exogenous corticoids, ACTH and/or CRH act at the pituitary (reduce LH release by GnRH), and hypothalamus (lower GnRH pulse frequency and delay surge release). This requires inter-neurones as GnRH cells do not have receptors for HPA compounds. There are two (simultaneous, therefore fail-safe?) pathways for CRH suppression of GnRH release via CRH-Receptors: one being the regulation of kisspeptin/dynorphin and other cell types in the hypothalamus, and the other being the direct contact between CRH and GnRH cell terminals in the median eminence. When we domesticate animals, we must provide the best possible environment otherwise animals trade-off with lower production, less intense oestrus behaviour, and impaired fertility. Avoiding life-time peri-parturient problems by managing persistent lactations in cows may be a worthy trade-off on both welfare and economic terms better than the camouflage use of drugs/hormones/feed additives/intricate technologies? In the long term, getting animals and environment in a more harmonious balance is the ultimate strategy.

A neuroendocrine reflex attributed to the mechanical stimulation of sensory receptors during copulation induces ovulation in cats. This stimulation can be simulated with a vaginal swab, triggering pseudopregnancy. The efficiency of ovulation induction with swab was evaluated in order to establish a natural contraceptive treatment for domestic cats as well as to analyze the effect on uterus of the repeated use of this technique. At the first phase of the study, 12 animals were evaluated in three consecutive estrous cycles. In the first cycle (T1), vaginal swab stimulation was applied. In the second cycle (T2), a vasectomized male was used. In the third cycle (T3), ovulation was accompanied unstimulated (control). At the second phase of the study, 13 female cats were induced to successive pseudopregnancy states to check the effects on the uterus. Confirmation of ovulation in the two phases of the study was carried out by measuring progesterone serum levels. The vaginal swab stimulation showed a similar response to that obtained by natural mating (P>0.05). Some cats showed mild uterine changes without pathological relevance. Thus, ovulation induction with swab is secure and without significant side effects.

In order to correlate the beginning of estrus to vaginal and cervical morphologies after synchronizationof estrus in ewes. Forty crossbred females of Dorper and Santa Inês were submitted to estrous synchronizationusing CIDR and eCG. After hormonal treatment, we recorded oestrus, morphology of vagina and cervix. Thedata were analyzed using the Analysis of Variance and the Tukey and Chi-square tests. All sheep presentedestrus, beginning at 32.30 ± 0.99 h after removal of the CIDR. There was a predominance of ewes with vaginaland cervical epithelium with pink coloration. At 0 h and 24 h after CIDR removal, 90% and 75% of the ewespresented incomplete crystallization respectively. The maximum crystallization of the cervical mucus occurred at48 hours after withdrawal of the CIDR. Therefore, the histomorphology of the mucosa and cervical mucus variesaccording to the time of estrus of sheep.(AU)

Clinical signs associated with parasitism were wheezing, sneezing followed by catarrhal nasal secretion, some restlessness, excessive head movement and walking in circles. The breasts and turbinates were hyperemic, with mucosal edema and presence of O. ovis larvae. All larvae collected from the turbinates ranged from the first to the third stage of development. Some L3 larvae collected at necropsy were incubated and the gray colored Imago with dark abdomen obtained from the pupae measured about 10mm. A descriptive analysis of the climatic conditions was carried out; in the year of investigation the incidence of O. ovis infection has grown (p<0.001), and the lowest mean minimum temperature (p<0.001) caused the development the O. ovis fly, so that there was an introduction of an increased number of these flies into the sheep and goat flocks in state of Bahia with the ideal climatic conditions for their perpetuation.(AU)

Between January 2007 and September 2013, 71 sheep belonging to 12 farms in the Umuarama microregion, State of Paraná, were evaluated regarding presence of Oestrus ovis larvae, during necropsies. The farms from which these animals originated were visited and the owners and employees were interviewed. Occurrences of O. ovis parasitism in sheep were diagnosed for the first time on this microregion. Of the 71 animals, 12 (16.9%) were parasitized by O. ovis, with mean intensity of 2.25 larvae per infested head (1 to 8 larvae/infested head). There was a high correlation (0.81, p=0.0346) between the number of larvae and the macroscopic lesions observed in these animals nasal cavities, such that sheep with more than 3 larvae may contain mucupurulent secretions or epistaxis. From the interviews conducted, it was found that all the farm owners were applying chemical parasite control methods (helminths and/or O. ovis), administered to all animals in the herds every 30 days (91.6% of the producers), using derivatives of macrocyclic lactones and/or benzimidazoles/imidazothiazoles. Further studies need to be conducted in this particular region, in an attempt to elucidate the prevalence of O. ovis parasitism in herds.(AU)

Further studies need to be conducted in this particular region, in an attempt to elucidate the prevalence of O. ovis parasitism in herds.

Oestrus ovis is a botfly whose larvae cause nasal myiasis, an environmental-dependent disease in small ruminants, generating acute and chronic injuries in the cranial cavities of sheep. Chile is a country of the southernmost worldwide distribution of this parasite, and there is few information about. Whence, the objective of this study was to approximate the epidemiological situation of O. ovis infection in Chilean sheep. From December 2009 to March 2010, a total of 87 samples were obtained by necropsy for skull inspection. The larvae were collected, and microscopically identified. The prevalence in the sampled sheep was 60.9%. From those that were infected, 85.7% (18/21) of sheep were 1 to 3 years old, constituting the stratum with the highest prevalence. The difference of infection in females and males was not significant. The high risk of infection seems to be dependent upon the environmental conditions of this southern region, especially during summer when the first larval stage (L1) could be found as the evidence. Therefore, the disease should be considered as a significant problem for this kind of livestock production.(AU)

Nasal cavity diseases that affect small ruminants can cause losses to sheep and goat herds in Central Brazil. A retrospective study of the University of Brasilia´s Veterinary Pathology Laboratory autopsy reports from 2003 to 2014 was conducted to verify the occurrence of small ruminants nasal cavity diseases. Six necropsied sheep (6/463 1.29%) showed mycotic or oomicotic granulomatous rhinitis and 22 animals (22/538, 4.08%) presented oestrosis diagnosis, affecting 86.36% of sheep and 13.64% of goats. The pyogranulomatous rhinitis in sheep occurred in flooded areas with abundant plant material decomposing. Rhinofacial pythiosis infection in animals showed as major changes swelling in the nasal region due to extensive granulomatous lesions associated with tissue necrosis, characterized by numerous macrophages and polymorphonuclear cells surrounding necrotic centers containing the agent surrounded by Splendore-Hoeppli reaction. Sheep with conidiobolomycosis showed extensive areas of necrosis and pyogranulomatous inflammation associated with fungal hyphae, localized in the nasopharynx and also in peribulbar region and exophthalmia. Most animals with oestrosis showed no significant clinical and pathological changes, even with the presence of larvae mainly in the sinuses and nasal turbinates, trachea and paranasal sinus. The importance of such diseases is still unknown in the region, and the knowledge of the clinical-pathological and epidemiological conditions is of great relevance for the diagnosis, control and prevention to avoid the expansion and losses to livestock.(AU)
