Resumo

ABSTRACT: This study, evaluated the readability of electronic leg and ear tags in Saanen goats. Fifty-seven goats were identified with the electronic leg tags (ELT) and electronic ear tags (EET) from birth until the lactation period ends. Readability of ELT and EET was 96.30% and 90.55% respectively in static conditions at the end of 12 months. Foot and udder, with no infection rates for ELT and EET in calm and aggressive goats were 95.70% and 100%, respectively. No infection rates of foot and udder for ELT and EET in calm and aggressive goats were 95.70% and 100%, respectively. Tagging method and animal temperament was not statistically significant. As a result, low animal traceability with ear tags was determined by this study. Besides, it is suggested that smaller-sized tagging materials would be more accurate when the ankle was selected as a body area to place identification tags in goats. The resulting issue to be considered is that the leg tagging should not negatively affect the animal welfare and the foot and udder health. In the future, using a leg band in the identification of goats will become more widespread as it does not damage animals and has a high readability capacity.


RESUMO: Este estudo teve como objetivo avaliar a identificação eletrônica nos membros e orelhas de cabras da raça Saanen. Cinquenta e sete cabras foram identificadas eletronicamente nas pernas (ELT) e nas orelhas (EET) desde o nascimento até o término do período de lactação. A leitura de ELT e EET foi de 96,30% e 90,55%, respectivamente, em condições estáticas ao final de 12 meses. As taxas de infecção de pé e úbere, em cabras calmas e agressivas, foram de 95,70% e 100% para ELT e EET em animais sem infecção, respectivamente. Não foi encontrado efeito significativo do método de marcação e temperamento animal. Com o resultado, a baixa rastreabilidade dos animais com marcas de orelha foi determinada por este estudo. Além disso, sugere-se que materiais de etiquetagem de menor porte seriam mais precisos quando a região do metatarso do animal fosse selecionado como área corporal para a colocação de etiquetas de identificação em cabras. A questão resultante a ser considerada é que a marcação da perna não deve afetar negativamente o bem-estar animal e a saúde dos pés e do úbere. Futuramente, o uso de etiquetas na identificação de cabras será mais difundido, pois não agride os animais e tem alta capacidade de leitura.

The olive (Olea europaea L.) is a leading oil crop in the Mediterranean area. Limited information on the inheritance of agronomic significant traits hinders progress in olive breeding programs, which encourages the development of markers linked to the traits. In this study, we report on the development of 46 olive simple sequence repeat (SSR) markers, obtained from 577,025 expressed sequence tags (ESTs) in developing olive fruits generated in the framework of the Slovenian national olive transcriptome project. Sequences were de novo assembled into 98,924 unigenes, which were then used as a source for microsatellites searching. We identified 923 unigenes that contained 984 SSRs among which dinucleotide SSRs (36 %) were the most abundant, followed by tri- (33 %) and hexa- (21 %) nucleotides. Microsatellite repeat motif GA (37 %) was the most common among dinucleotides, while microsatellite repeat motif GAA was the most abundant trinucleotide SSR motif (16 %). Gene ontology annotations could be assigned to 27 % of the unigenes. A hundred and ten expressed sequence tag-derived-simple sequence repeats (EST-SSRs) with annotated genes were selected for primer designing and finally, 46 (42 %) polymorphic EST-SSRs were successfully amplified and used to validate genetic diversity among 24 olive varieties. The average number of alleles per locus, observed heterozygosity, expected heterozygosity, and polymorphic information content were 4.5, 0.649, 0.604 and 0.539, respectively. Twenty-seven EST-SSRs showed good diversity properties and were recommended for further olive genome investigation.

Only a limited number of simple sequence repeat (SSR) markers is available for the genome of garlic (Allium sativum L.) despite the fact that SSR markers have become one of the most preferred DNA marker systems. To develop new SSR markers for the garlic genome, garlic expressed sequence tags (ESTs) at the publicly available GarlicEST database were screened for SSR motifs and a total of 132 SSR motifs were identified. Primer pairs were designed for 50 SSR motifs and 24 of these primer pairs were selected as SSR markers based on their consistent amplification patterns and polymorphisms. In addition, two SSR markers were developed from the sequences of garlic cDNA-AFLP fragments. The use of 26 EST-SSR markers for the assessment of genetic relationship was tested using 31 garlic genotypes. Twenty six EST-SSR markers amplified 130 polymorphic DNA fragments and the number of polymorphic alleles per SSR marker ranged from 2 to 13 with an average of 5 alleles. Observed heterozygosity and polymorphism information content (PIC) of the SSR markers were between 0.23 and 0.88, and 0.20 and 0.87, respectively. Twenty one out of the 31 garlic genotypes were analyzed in a previous study using AFLP markers and the garlic genotypes clustered together with AFLP markers were also grouped together with EST-SSR markers demonstrating high concordance between AFLP and EST-SSR marker systems and possible immediate application of EST-SSR markers for fingerprinting of garlic clones. EST-SSR markers could be used in genetic studies such as genetic mapping, association mapping, genetic diversity and comparison of the genomes of Allium species.

Pop-up satellite archival tags (PSATs) were deployed on four sailfish, Istiophorus platypterus, in the coastal waters of Rio de Janeiro State in southeast Brazil during January and February of 2009 (sailfish I and II) and between November 2010 and January 2011 (sailfish III and IV). The total number of days monitored (i.e., time that the tags remained attached) were 12 (sailfish I), 51 (sailfish II), 16 (sailfish III) and 43 days (sailfish IV). The results indicate a clear pattern of vertical habitat utilization with the majority of the time spent concentrated near the uniform sea surface layer occupying a relatively narrow temperature range. Despite the clear preference for epipelagic surface waters, sailfish regularly undertook vertical excursions into deeper waters (>50 m) within three to six hour intervals. "Most Probable Tracks" (estimated from raw geolocations using the state-space Kalman filter model) and linear displacements suggested that tagged sailfish did not move significant distances from the tagging site. In brief, our report provides information regarding the biology of sailfish in the southwestern Atlantic and how vertical distributions during the day and night are influenced by water temperature and how this information can improve sailfish stock assessments in southwestern Atlantic Ocean.(AU)


Quatro exemplares de agulhão-vela foram marcados com marcas eletrônicas monitoradas por satélite ('Pop-up satellite archival tags - PSATs') nas águas costeiras do Rio de Janeiro, sudeste do Brasil, durante janeiro e fevereiro de 2009 (agulhão-vela I e II) e entre novembro de 2010 e janeiro de 2011 (agulhão-vela III e IV). O número total de dias monitorados (ou seja, o tempo que as marcas permaneceram implantadas nos peixes) foram 12 (agulhão-vela I), 51 (agulhão-vela II), 16 (agulhão-vela III) e 43 dias (agulhão-vela IV). Os resultados demonstram um padrão claro de utilização do hábitat com a maior parte do tempo despendido predominantemente próximo à superfície do mar ocupando águas com uma faixa de temperatura restrita. Apesar da preferência por águas superficiais, os agulhões frequentemente realizaram mergulhos para águas mais profundas (ca. > 50 m) em intervalos de três a seis horas. A "rota mais provável" estimada a partir dos dados brutos de geolocalização e o modelo 'State-Space Kalman Filter' sugerem que os agulhões marcados não realizaram migrações significativas a partir do local de marcação. Em resumo, nossos resultados apresentam informações sobre a biologia da espécie no Atlântico Sudoeste e como as migrações verticais durante o dia e a noite são influenciadas pela temperatura da água e como essa informação pode auxiliar as avaliações de estoques de agulhão-vela no sudoeste do Atlântico.(AU)

Until the mid 1950s, boron was believed to play an important role in the transport of sugars in plants. However, boron actually depends on sugar alcohols to be taken up by the plant. In some cases, the main sugars involved in this process are sorbitol and mannitol which form stable complexes with boron. In this study, the sequences of the SugarCane EST Genome Project (SUCEST) database were searched for enzymes involved in the metabolism of these sugars by comparing them with enzymes from other organisms. Eighteen contigs from sugarcane (Saccharum sp.) presented high similarity with 11 enzymes involved in the putative biosynthetic pathway of sorbitol and mannitol from fructose in sugarcane. Seven of these contigs had high homology with sequences deposited in GenBank.


Até meados da década de 50 acreditava-se que o boro tinha uma importante função no transporte de açúcares em plantas. Na verdade, o boro depende de açúcares álcoois para serem mobilizados dentro da planta. Em alguns organismos os principais açúcares envolvidos neste processo são o sorbitol e o manitol, que formam complexos estáveis com o micronutriente. O objetivo deste estudo foi procurar seqüências no banco de dados SugarCane EST Genome Project (SUCEST) que codificam enzimas participantes na via metabólica destes açúcares através da comparação de enzimas de outros organismos. Dezoito "contigs" de cana-de-açúcar (Saccharum sp.) apresentaram similaridade com onze seqüências de enzimas que compõem a provável via metabólica de sorbitol e manitol a partir de frutose. Destes "contigs", sete apresentaram uma alta similaridade entre as seqüências depositadas no GenBank.
