Resumo

The most common form of psycho-social dysfunction is anxiety with depression being related closely without any age bar. They are present with combined state of sadness, confusion, stress, fear etc. Glyoxalase system contains enzyme named glyoxalase 1 (GLO1).It is a metabolic pathway which detoxifies alpha-oxo-aldehydes, particularly methylglyoxal (MG). Methylglyoxal is mainly made by the breakdown of the glycolytic intermediates, glyceraldehyde-3-phosphates and dihydroxyacetone phosphate. Glyoxylase-1 expression is also related with anxiety behavior. A casual role or GLO-1 in anxiety behavior by using viral vectors for over expression in the anterior cingulate cortex was found and it was found that local GLO-1 over expression increased anxiety behavior. The present study deals with the molecular mechanism of protective activity of eugenol against anxiolytic disorder. A pre-clinical animal study was performed on 42 BALB/c mice. Animals were given stress through conventional restrain model. The mRNA expression of GLO-1 was analyzed by real time RT-PCR. Moreover, the GLO-1 protein expression was also examined by immunohistochemistry in whole brain and mean density was calculated. The mRNA and protein expressions were found to be increased in animals given anxiety as compared to the normal control. Whereas, the expressions were decreased in the animals treated with eugenol and its liposome-based nanocarriers in a dose dependent manner. However, the results were better in animals treated with nanocarriers as compared to the compound alone. It is concluded that the eugenol and its liposome-based nanocarriers exert anxiolytic activity by down-regulating GLO-1 protein expression in mice.

Purpose: Simultaneous pancreas-kidney transplantation (SPKT) brings several benefits for insulin-dependent type-1 diabetic patients associated with end-stage renal disease (ESRD). However, data on psychological outcomes for the waiting list and the transplanted patients are still lacking. Methods: Using the psychological Beck inventories of anxiety (BAI) and depression (BDI), 39 patients on the waiting list were compared to 88 post-transplanted patients who had undergone SPKT. Results: Significant differences were found regarding depression (p = 0.003) but not anxiety (p = 0.161), being the pretransplant patients more vulnerable to psychological disorders. Remarkable differences were observed relative to the feeling of punishment (p < 0.001) and suicidal thoughts (p = 0.008) between the groups. It was observed that patients who waited a longer period for the transplant showed more post-transplant anxiety symptoms due to the long treatment burden (p = 0.002). Conclusions: These results demonstrated the positive impact of SPKT on psychological aspects related to depression when comparing the groups. The high number of stressors in the pretransplant stage impacts more severely the psychosocial condition of the patient.

Resumo

This study aimed to determine if the homeopathic formulation based on Natrum muriaticum, Matricaria, Chamomilla, and Ignatia amara was effective for stress control in dogs in order to prevent and/or reduce the frequency of unwanted behaviors. Ten male beagle dogs were divided into two groups, with five animals per group. The control group was animals receiving 0.5 mL/day of placebo via spray in the feed for 30 consecutive days as a preventive. The treated group received 0.5 mL/day of tested drug spray in the feed for 30 consecutive days as a preventive. Behavioral tests were performed at two time points, before and after receiving the tested drug. We performed the following behavioral tests: a) persons known and unknown in the internal and external environment; b) interaction with toys; c) interaction with an unknown dog, d) test with a leash; and e) firecracker test. There was more engagement in digging and scaling the fence in the control group than in the treatment group (P < 0.05). The animals that received homeopathic medicine sat 2.54 times longer than the control (P = 0.0269). Dogs that received homeopathic showed 4.49 times more interest in general at D30 than at D1 (P = 0.0214). Animals in the treated group showed an 83% less chance of urinating than the control group in the tests involving displacements. We conclude that the preventive addition of homeopathic medicine containing Natrum muriaticum, Chamomilla, and Ignatia amara reduced undesirable behaviors related to anxiety and increased the incidence of interest behavior.(AU)


Este estudo visou determinar se a formulação homeopática baseada em Natrum muriaticum, Matricaria, Chamomilla, e Ignatia amara foi eficaz para o controle de stress em cães, a fim de prevenir e/ou reduzir a frequência de comportamentos indesejados. Dez cães beagle machos foram divididos em dois grupos, com cinco animais por grupo. O grupo controle recebeu 0,5 mL/dia de placebo via spray na ração durante 30 dias consecutivos, como preventivo. O grupo tratado recebeu 0,5 mL/dia de homeopático via spray na ração durante 30 dias consecutivos, como preventivo. Os testes comportamentais foram realiza-dos em dois momentos, antes e depois de receber o fármaco testado. Foram realizados os seguintes testes comportamentais: a) pessoas conhecidas e desconhecidas no ambiente interno e externo; b) interação com brinquedos; c) interação com um cão desconhecido, d) teste com a guia; e teste com fogos de artifício. Houve um aumento nos comportamentos cavar e escalar a grade no controle (P < 0,05). Os animais que receberam o homeopático sentaram-se 2,54 vezes mais do que o grupo con-trole (P = 0,0269). Os cães que receberam homeopático mostraram 4,49 vezes mais interesse em geral no D30 do que no D1 (P = 0,0214). Os animais do grupo tratado mostraram uma probabilidade 83% menor de urinar do que o grupo controle nos testes envolvendo deslocamento. Concluímos que a adição preventiva de medicamentos homeopáticos contendo Natrum muriaticum, Chamomilla, e Ignatia amara reduziu os comportamentos indesejáveis relacionados com a ansiedade e aumentou a incidência de comportamentos de interesse.(AU)

Resumo

Medicinal and aromatic plants have very substantial emotional effects on rats, which is part of the current study. Decoction products of three Moroccan plants (Lavandula angustifolia L., Laurus nobilis L., and Artemisia herba-alba) were used to be tested on Wistar rats in the laboratory. The goal was to check if they had an anti-depressant and/or anti-anxiety action on the animals' tests. Wistar Rats were born and bred in the pet store of the Faculty of Science, Kenitra. The antidepressant and anti-anxiety effects were assessed according to three animal models: Open Field, Elevated Plus-maze for anxiety, and the Forced Swimming animal model for depression. The results showed that drinking water containing plant extracts has anti-depressant and anti-anxiety effects. Rats have overcome depression by reducing downtime during forced swimming. Concerning anti-anxiety, the open-field test showed an increase in the rat's locomotor activity, which indicates unhesitating exploratory behavior in the form of confident movement through the number of visits to the central part and the total distance crossed. The anxiety reduction was also corroborated by the elevated Plus-maze test, where the rats displayed no aversion to open spaces on account of the prolonged time they spent in the open arms and platform of the raised cross maze. In light of the findings from the experiments conducted in the study, the decoction products of the three plants appear to be viable candidates for the treatment of depression and anxiety.

Resumo

O aumento progressivo no número de casos de depressão profunda e suicídio na Medicina Veterinária é alarmante. A amplitude de atuação da prática veterinária, cujas demandas comumente levam a sobrecargas que resultam em desordens mentais, como a Síndrome de Burnout (SB), e que muitas vezes culminam no suicídio, é um problema atual sério e crescente. Como o debate acerca da temática no Brasil ainda é incipiente, conduziu-se esta revisão de literatura.


The progressive increase in the number of cases of severe depression and suicide in Veterinary Medicine is alarming. The range of action of veterinary practice, whose demands commonly lead to overloads that result in mental disorders, such as Burnout Syndrome (BS), and that often culminate in suicide, is a serious and growing current problem. As the debate on the subject in Brazil is still incipient, this literature review was conducted.

Resumo

This study aims to evaluate the effect of fenugreek on biochemical parameters, body weight, and the behavior of male Wistar rats. The purpose of administering the aqueous extract of fenugreek seeds (Trigonella foenum-graecum L) for 30 successive days by oral gavage (force-feeding) at a dose of 250g / l of water is intended to assess, on the one hand, the neurobehavioural effects and on the other hand its effect on some biochemical parameters (glycemia and lipid balance) as well as weight. The behavioral tests performed are the Light/Dark Box tests (LDB) and the Elevated Plus Maze test (EPM). This work indicated that the consumption of the aqueous extract of the seeds of fenugreek is beneficial to stimulate appetite and gain weight, thus decreasing the level of total cholesterol, triglycerides, LDL-C, and a significant increase in HDL-C, also a reduction in blood sugar and a decrease in anxiety.(AU)

In the present work, we sought to determine the effects of an individual application of prenatal stress and postnatal stress and their combination on the anxious and locomotor behavior of first-time mother rats. Indeed, three batches of rats were exposed to three types of daily repeated stress: one batch exposed to prenatal restraint stress (SC): 01h a day from day 11 to day 19 of gestation, one batch exposed to postnatal mother prenatal stress (SS): 03h a day from the third to the fourteenth postnatal day (JPN), and one batch exposed to combined restraint stress: 01h from day 11 to day 19 of gestation followed by a postnatal mother separation of 03h a day from the third to fourteenth JPN (CSS); a fourth control batch (T) of female rats that did not undergo any type of stress was also part of our study. At postnatal days 15 and 20, the behavior of the female rats in the four batches was assessed using the Open field Test along with the Plus Maze, and at JPN 30, the female rats were decapitated, the blood of which was collected for plasma adrenocorticotropic hormone (ACTH) level evaluation. Our findings show that all studied behaviors were very highly affected relative to the results of the control batch after individually applying prenatal restraint stress and mother separation. We noticed an intensified anxious behavior in CSS with a significant increase in plasma levels of ACTH.(AU)

Resumo

The study we conducted aimed to evaluate the acute toxicity of abamectin (ABM) (pesticide) on liver function and neurobehavioral by using the open field test. As a therapy, we have opted for a treatment based on natural antioxidants ginger extract (Zingiber officinale) mixed with olive oil to see the restorative effects during the experiment. The study was conducted on female Wistar rats. The latter was given an inhalation exposure of abamectin (ABM) at a dose CL50 > 5.04mg/L/1hr each morning for one week. The effects of this exposure showed a drop in body weight gain (g), a significant decrease in the relative and absolute weights liver (gm/100 gm body weight) accompanied by hepatic impairment indicating a significant disturbance of transaminases (AST/ALT), alkaline phosphatase (ALP) activity and significant behavioral changes. Administration of ginger (Zingiber officinale) extract at a dose of 3 ml mixed with olive oil at a dose of 1 ml/kg for 7 days orally (gavages) after inhalation indicates that ginger has a more remarkable and rapid effect on physical and mental health, it acts as a major means of defense and hepato-protective against toxicity.(AU)

In the present work, we sought to determine the effects of an individual application of prenatal stress and postnatal stress and their combination on the anxious and locomotor behavior of first-time mother rats. Indeed, three batches of rats were exposed to three types of daily repeated stress: one batch exposed to prenatal restraint stress (SC): 01h a day from day 11 to day 19 of gestation, one batch exposed to postnatal mother prenatal stress (SS): 03h a day from the third to the fourteenth postnatal day (JPN), and one batch exposed to combined restraint stress: 01h from day 11 to day 19 of gestation followed by a postnatal mother separation of 03h a day from the third to fourteenth JPN (CSS); a fourth control batch (T) of female rats that did not undergo any type of stress was also part of our study. At postnatal days 15 and 20, the behavior of the female rats in the four batches was assessed using the Open field Test along with the Plus Maze, and at JPN 30, the female rats were decapitated, the blood of which was collected for plasma adrenocorticotropic hormone (ACTH) level evaluation. Our findings show that all studied behaviors were very highly affected relative to the results of the control batch after individually applying prenatal restraint stress and mother separation. We noticed an intensified anxious behavior in CSS with a significant increase in plasma levels of ACTH.

Resumo

This study aims to evaluate the effect of fenugreek on biochemical parameters, body weight, and the behavior of male Wistar rats. The purpose of administering the aqueous extract of fenugreek seeds (Trigonella foenum-graecum L) for 30 successive days by oral gavage (force-feeding) at a dose of 250g / l of water is intended to assess, on the one hand, the neurobehavioural effects and on the other hand its effect on some biochemical parameters (glycemia and lipid balance) as well as weight. The behavioral tests performed are the Light/Dark Box tests (LDB) and the Elevated Plus Maze test (EPM). This work indicated that the consumption of the aqueous extract of the seeds of fenugreek is beneficial to stimulate appetite and gain weight, thus decreasing the level of total cholesterol, triglycerides, LDL-C, and a significant increase in HDL-C, also a reduction in blood sugar and a decrease in anxiety.

The curcumin is part of the curcuminoids family. Present in the spice turmeric is a vegetable alkaloid obtained from Curcuma longa. Many studies have been carried out to describe the multiple biological actions of curcumin. The positive antidepressant and anti-anxiety effects of curcumin are most likely due to its ability to normalize specific physiological mechanisms, in addition to being a potent antioxidant and anti-inflammatory agent. Curcumin also protects the brain and simultaneously acts on several mechanisms related to depression. It is considered to be the most promising therapeutic target for treating depressive behaviors. Through this work, we will prove the protective effect of curcumin on the anxious behavior of Wistar rats and the complications induced by the administration of a toxic product that is formaldehyde. To this fact, 25 white rats of Wistar strains (250±50 g) were separated into five experimental batches: control batch (T; n = 5),vehicle control batch (CV; n = 5), Curcumin control batch (CC; n = 5), one batch formaldehyde vehicle (FV; n = 5) and one batch formaldehyde Curcumin (FC; n = 5). A single intraperitoneal injection carried out the formaldehyde administration diluted in distilled water at a dose of 10 mg/kg. Turmeric dissolved in olive oil was administered by gastric gavage at a dose of (60 mg/kg). Our results show the therapeutic benefits of curcumin against the deficits caused by formaldehyde reflected in a disturbance in rat behavior and degree of anxiety using the open-field test and the light and dark box test.(AU)

This study aimed was to evaluate Elevated Plus-Maze and Hole Board test as an analytical methodology for quality control of anxiolytic medicines based on Passiflora incarnata. The efficacy of three brands commercial differents of Passiflora incarnata extracts was tested in the Elevated Plus-maze (for anxiolytic effect) and the Hole Board test (for locomotor and neophilia activity) in Wistar rats. Treatment with the P. incarnata brands revealed a significant 3-fold increase in the length of stay in the open arms in the LCE. In the ambulation, animals from bands 1 and 3 presented a significant increase, being brand 2 the only one that did not present this relevance. In the neophilia, Passiflora brand 1 presented a significant increase of 3 times and groups 2 and 3 an increase of approximately 2 times. The results showed that different brands of Passiflora have a satisfactory quality standard related to biological activity and showed that the use of Elevated plus-maze/Hole board test can be an effective tool in the quality control of herbal medicines with anxiolytic activity associated with the phytochemical analysis.(AU)


Na busca pela redução dos efeitos colaterais causados pela alopatia, os fitoterápicos já se encontram disponíveis no mercado veterinário. O objetivo deste estudo foi avaliar os testes neurocomportamentais Labirinto em Cruz Elevado e Campo Vazado como metodologias analíticas para o controle de qualidade de medicamentos ansiolíticos a base de Passiflora incarnata. A eficácia de três diferentes marcas comerciais de extrato de Passiflora incarnata foi testada no Labirinto em Cruz Elevado (para efeitos ansiolíticos) e no Campo Vazado (para atividade locomotora e busca por novidade) em ratos Wistar. O tratamento com as marcas de P. incarnata demonstrou aumento significativo de 3 vezes no tempo de permanência nos braços abertos no Labirinto em Cruz Elevado. Na deambulação, animais das marcas 1 e 3 apresentaram aumento significativo, sendo a marca 2 a única que não apresentou relevância para este parâmetro. Na neofilia, a marca 1 de Passiflora apresentou aumento significativo de 3 vezes e as marcas 2 e 3 aumento de aproximadamente 2 vezes. Os resultados mostraram que diferentes marcas de Passiflora apresentam um padrão de qualidade satisfatório relacionado à atividade biológica e mostraram que a utilização dos testes Labirinto em Cruz Elevado e Campo Vazado podem ser ferramentas eficazes no controle de qualidade de fitoterápicos com atividade ansiolítica associadas à análises fitoquímicas.(AU)

Resumo

We aim was to evaluate the protective effects of the antioxidants cinnamon and quercetin on neurobehavioral alterations and complications, besides biochemical parameters of induced-diabetics Wistar rats. Diabetes was induced by asingle intraperitoneal injection of streptozotocin at a dose of (45 mg/kg). The administration of streptozotocin was considered acting on anxiety behaviors and biochemical parameters in adult Wistar rats. On the other hand, the protective role of antioxidants (cinnamon and quercetin) on streptozotocin-induced disorders was also evaluated. Behavioral tests in the open field (OF) revealed that diabetic animals exhibited an anxious behavior and an alteration in thelocomotive and exploratory activities when compared to control. The administration of the cinnamon (2g/kg) and Quercetin (0.5g/kg) by gastric gavage reduces anxiety and decreases hyperglycemia-related harm. However, antioxidants cinnamon and quercetin administration significantly alleviated anxious and depressive behaviors.(AU)
