Three hundred hatching eggs from two layer breeder flocks (Hy-Line Brown and hy-Line W-36; 150 each) were obtained at three different periods of laying cycle (early, middle and late stages). The hatching eggs were randomly taken to assess the impact of chicken strain, age, and hatching time on some eggshell measurements and structures. Results indicate that egg weight, shell percentage and shell thickness were significantly affected by strain, breeder age, and incubation time. Increasing rate of calcium absorption and utilization by the embryos during the embryonic stage clearly reflects the decrease occurred in eggshell strength. The differences of eggshell breaking strength during incubation time periods were 0.41 and 0.55 kg/cm2, the differences due to the incubation time may be due to the higher demand for calcium utilization in the late incubation period (18 days), observed on the10th day of incubation. Brown eggs recorded significant higher calcium percentage in eggshell compared to the white ones. The brown breeder hens recorded significant higher total pores per egg compared to the white egg. Concerning eggshell ultrastructure, total thickness of the brown eggshell was significantly higher than those of the white eggshell by about 3.7%. All parameters, except for total thickness, were significantly affected by the interaction between strain and breeder age. Finally, we conclude that some traits including egg weight, shell percentage, shell thickness, calcium percentages in eggshell, and total pores per egg were significantly affected by strain, breeder age, and incubation time. Eggshell ultrastructure of brown egg was better compared to the white ones.(AU)

Abstract During a survey Myxozoa, four specimens of the sheepshead (18 ± 1.5 cm and 59 ± 2.5 g) (Archosargus probatocephalus) were collected in the Ipioquinha river (Maceió/AL). Transmission electron microscopy observations revealed erythrocyte agglutinations in gill capillaries located near spherical cysts containing myxospores of the genus Henneguya. This hemagglutination partially or totally obstructed the gill capillaries. Erythrocytes occurred in close adherence to each other, with a closed intercellular space. A few lysed erythrocytes were observed among agglutinated cells. The reduced lumen of the capillaries was partially filled with amorphous dense homogenous material adhering to the erythrocytes. In addition, heterogeneous masses of irregular lower electron density were observed in the reduced channel of the capillary. The agglutinated erythrocytes appeared dense and homogenous, lacking cytoplasmic organelles. The nuclei had the appearance of normal condensed chromatin masses, generally without visible nucleoli. This occurrence of hemagglutination only in the capillaries located in close proximity to the developing myxozoan cysts suggests that parasite development may be a factor triggering erythrocyte agglutination. This is supported by previous experimental studies that showed a probable correlation between parasitic infections and hemagglutination. Nonetheless, further studies are necessary in order to better understand the physicochemical processes involved in this phenomenon.

Background: Cryptoccocal meningitis continues to present high incidence among AIDS patients. The treatment of choice is the synergistic combination of flucytosine (5-FC) with amphotericin B deoxycholate (AmBd) or its lipid formulations. However, 5-FC is unavailable in many countries and AmB demands hospitalization. The combination of AmB with the fungistatic fluconazole (FLC) or the use of high FLC daily doses alone became the choice. Nonetheless, sterilization of cerebrospinal fluid is delayed with FLC monotherapy, mainly with high fungal burden. These findings suggest the search for new antifungal compounds, such as liriodenine. Methods: Liriodenine antifungal activity was evaluated by three procedures: determining the minimum inhibitory concentration (MIC) on 30 strains of the Cryptococcus neoformans (C. neoformans) complex and 30 of the Cryptococcus gattii (C. gattii) complex, using EUCAST methodology and amphotericin B deoxycholate as control; performing the time-kill methodology in two strains of the C. neoformans complex and one of the C. gattii complex; and injury to cryptococcal cells, evaluated by transmission electron microscopy (TEM). Liriodenine absorption and safety at 0.75 and 1.50 mg.kg-1 doses were evaluated in BALB/c mice. Results: Liriodenine MICs ranged from 3.9 to 62.5 µg.mL-1 for both species complexes, with no differences between them. Time-kill methodology confirmed its concentration-dependent fungicidal effect, killing all the strains below the limit of detection (33 CFU.mL-1) at the highest liriodenine concentration (32-fold MIC), with predominant activity during the first 48 hours. Liriodenine induced severe Cryptococcus alterations cytoplasm with intense rarefaction and/or degradation, injury of organelles, and presence of vacuoles. Liriodenine was better absorbed at lower doses, with no histopathological alterations on the digestive tract. Conclusion: The fungicidal activity confirmed by time-kill methodology, the intense Cryptococcus injury observed by TEM, the absorption after gavage administration, and the safety at the tested doses indicate that the liriodenine molecule is a promising drug lead for development of anticryptococcal agents.(AU)

Skin and cartilage have been the main source for the recovery of somatic cells to be used in conservation strategies in wild mammals. In this sense, an important step for the cryopreservation of these samples is to recognize the properties of the skin and cartilage. Thus, knowing that the skin may differ among species and aiming to contribute to the establishment of cryobanks, the study examined the differences in the ear skin and cartilage of wild rodents from South America, agouti (Dasyprocta leporina) and spix's yellow-toothed cavy (Galea spixii). Ultrastructural and quantitative methods were used to measure skin and cartilage thickness, density of collagen and elastic fibers, cell type number and distribution, and proliferative activity. Although ultrastructural analysis revealed a similar pattern between species, morphometric analysis of the skin and cartilage showed differences between agoutis and cavies regarding thickness of epidermis layers (corneum: 5.3±2.5μm vs. 3.9±0.6μm; intermediate: 16.4±6.2μm vs. 23.4±8.1μm; basal: 9.9±2.1μm vs. 4.8±0.5μm), dermis (183.1±44.0μm vs. 258.2±22.9μm), total skin (211.8±46.0μm vs. 290.3±23.7μm) and perichondrium (27.6±6.1μm vs. 10.5±1.8μm). A greater number of epidermal cells (61.7±15.2 vs. 24.8±7.6) and chondrocytes (32.7±9.0 vs. 27.5±4.7) were observed in agouti, while the cavy presented a greater number of melanocytes (12.6±4.7 vs. 29.9±6.2), keratinocytes (14.7±4.2 vs. 29.8±7.6), and fibroblasts (103.6±24.7 vs. 112.2±11.3). Moreover, a higher percentage of collagen fibers and proliferative activity was observed in the skin of cavies, when compared to the skin of agoutis. Therefore, there are differences between agouti and cavy for ear skin and cartilage, requiring the establishment of species-specific cryopreservation protocols.(AU)

This experiment aimed to verify if the proteins present in a 13th day conceptus induce changes in the equine endometrial ultra-structure, histology, and vascularization, two days after its infusion. Ten healthy cyclic mares were used. Once estrus was confirmed, mares were examined daily to detect ovulation (day 0). After ovulation, mares were examined daily until day seven by transrectal palpation and B-mode and Doppler ultrasonography. In this first cycle, intrauterine biopsies were collected at day seven after ovulation, constituting the Cyclic group (n = 10). In the second cycle, the same mares daily were examined until ovulation was detected. After ovulation, mares were examined daily by transrectal palpation and B-mode and Doppler ultrasonography until day 7. On day 5, after ovulation, fragments from previously collected 13-day-old concepti were infused into the uterus of each mare. Intrauterine biopsies were collected at day 7 in all mares (n = 10), constituting the Fragment group. The percentage of ciliated and flattened cells decreased in the Fragment group. Protruded cells, superficial and intraglandular secretion, glandular lumen and diameter, blood vessel diameter, endometrial vascularization, and immune cells were higher in the Fragment group than in the Cyclic group. In summary, proteins of 13th day equine conceptus fragments infused at day five after ovulation signaled histological and vascular changes in the endometrium at the 7th day after ovulation.

This study was conducted to examine the effect of gamma radiation on biological specimens. Thus, our concept is to clarify that exposure to accumulated dose of 0.2 Gy gamma rays (0.66 rad/Sec. dose rate) from Cs137 source induces cellular perturbations in the midgut epithelium of the F1 progeny of Blaps polycresta, therefore affecting nutrition and growth. Beetles were reared in laboratory conditions and the newly emerged adults were irradiated with the aforementioned dose. Histological and ultrastructure anomalies of midgut cells (digestive and regenerative cells) were observed by 72 h after radiation exposure to ensure that the cells will not return to control state. Retardation in the development of the F1 progeny was also noticed and beetles died through two weeks. In the light of these observations, biological tissue act as an indicator to the continuous exposure to environmental radiation.(AU)

Triatoma vitticeps is a triatomine with geographic distribution restrict to Brazil, which exhibits high prevalence of Trypanosoma cruzi natural infection. Of special epidemiologic concern, this species often invades households in the states of Rio de Janeiro, Minas Gerais and Espírito Santo. The objective of this study was to evaluate morphological and ultrastructural parameters on three T. cruzi isolates obtained from wild T. vitticeps specimens. The growth and cell differentiation of the parasite was evaluated through epimastigote and trypomastigote forms obtained in the growth curves for three distinct isolates. The maximum growth showed differences at the 20th day of the curve. Our in vitro results show a heterogeneity, regarding these features for samples cultivated under the same conditions. Morphometric analyzes based on the shape of epimastigotes and trypomastigotes corroborated such differentiation. These results highlight the need of better understanding the meaning of this diversity under an eco-epidemiological perspective.(AU)

The aim of this study was to report a diffuse unilateral seminoma in a 4-year-old fighting rooster (Gallus gallus) by considering the anatomic, pathological and ultrastructural features. A significant decline in the viability of the bird's embryos was reported during the last year. The bird died with a marked abdominal distention. During necropsy, a firm ovoid neoplasm of approximately 9 cm long, 8 cm wide and 5 cm thick was found in the region adjacent to the pronephros, replacing the left testicle. The neoplasia was soft and encapsulated, and the cut surface showed extensive areas of haemorrhage interspersed with necrotic areas. Histologically, the neoplastic cells formed large mantles or cords with moderate and eosinophilic cytoplasm. The nuclei were pleomorphic, and some of them were central, while others were eccentric. Mitoses were scarce. Some important findings in the ultrastructural study were nuclei exhibited an elongated, electrodense, rope-like nucleoli and the presence of scarce intracytoplasmic glycogen. The anatomo-pathological and ultrastructural findings of the tumour were consistent with a seminoma. The ultrastructural study provided important support for the diagnosis, as other tumours were ruled out. The early diagnosis of malignant or benign testicular tumours in birds is the key for a timely treatment, as these tumours can metastasize or grow to such an extent that they compromise the lives of birds, as in the present case.

Infection by Helicobacter spp. has been associated with gastritis and ulcers in pigs and humans. Association between Helicobacter species and lesions can contribute to determine specific pathogenicity. The aim of this study was to describe ultrastructural aspects of Helicobacter spp. and identify Helicobacter species by PCR assay. Gastric samples from 13 naturally infected sows were analyzed. From these, 12 were positive for Helicobacter spp. 16S rRNA gene and seven were identified as H. suis. The species was not identified in five samples and all samples were negative for urease gene. The sequencing of rRNA gene of five samples showed similarity with H. suis and H. heilmannii type I. Seven samples positive for Helicobacter genus generated no sequenceable fragments. On ultrastructural study, three samples showed helical bacteria measuring 4 to 6 μm long, 0.5 to 0.8 μm width, 4 to 8 spirals and 2 to 6 bipolar flagella. Two samples showed bacteria measuring 9 to 10 μm in length, 0.5 μm width, 22 to 24 spirals and no flagella, characterizing Helicobacter non-H. pylori, but these samples were negative for H. suis. In conclusion, the results indicate that adult pigs are commonly infected by helical bacteria presenting different ultrastructural characteristics, suggesting that mixed infection is frequent.

Two samples showed bacteria measuring 9 to 10 μm in length, 0.5 μm width, 22 to 24 spirals and no flagella, characterizing Helicobacter non-H. pylori, but these samples were negative for H. suis. In conclusion, the results indicate that adult pigs are commonly infected by helical bacteria presenting different ultrastructural characteristics, suggesting that mixed infection is frequent.(AU)

The early diagnosis of malignant or benign testicular tumours in birds is the key for a timely treatment, as these tumours can metastasize or grow to such an extent that they compromise the lives of birds, as in the present case.(AU)

Purpose: To investigate the prophylactic and therapeutical effects of sildenafil in a model of acute radiation proctitis (ARP). Methods: All experimental procedures of this study was examined by histopathological, immunohistochemical and transmission electron microscopic analysis. Results: Our histopathological evaluations indicated significant increases in lesion severity, cryptic apsis, cryptitis, cryptic distortion, reactive atypia and infiltration depth of the control (proctitis) group. While the prophylaxis group and the treatment group had significantly lower scores. High-dose group showed similar results as prophylaxis group. Histopathological findings of the prophylaxis group was more significant than the treatment group. Immunoreactivities of IL-1β, FGF-2, TNF-α and HIF-1α increased in the control group especially in the epithelial and cryptic regions. On the contrary, sildenafil application caused significant decreases of inflammatory markers in all treatment groups, specifically better results in the prophylaxis group. Conclusion: The sildenafil has anti-inflammatory effects on ARP, as well as protective effects against ARP and the protective effect of sildenafil surpasses its therapeutic effect histopathologically.(AU)

Considering the great economic and sanitarian importance of Haematobia irritans − the development of resistance of this species to the main chemical insecticides used in its control, in several other localities of the world; as well as that different strains of the same species frequently present different types and distribution patterns −, the present aim study was to investigate the typology and distribution of different types of sensillae of H. irritans adults, from populations of this fly present in Brazilian Midwest, with emphasis on olfactory sensillae. This study provides new data on the typology and sensillary distribution of antennal sensilla of this fly. In the antennal segments were found non-innervated spinules and ten subtypes of sensilla: long bristles sensillum, long basiconic sensillum, blunt coeloconic sensillum, clavate coeloconic sensilla − single-tip and double-tip subtypes −, grooved coeloconic sensillum, long grooved coeloconic sensillum, trichoid sensillum, coeloconic sensillum, and smaller basiconic sensillum. A slight sexual dimorphism was observed in the antennal sensillae of H. irritans. These results provide a morphological basis for future investigations on olfactory-mediated behavior of this species, and could assist future studies for the development of alternative measures to the monitoring and control of this fly populations, with less environmental impact.(AU)

This report introduced the description of two different species of digenean parasites isolated from the intestine of Rhinopoma hardwickii with new host and locality records in Egypt. The recovered helminthes were studied morphologically and morphometrically by light microscopy and the surface topography of the two species was elucidated by scanning electron microscopy (SEM). Urotrema scabridum had an elongated body, testes were tandem, ovaries were pretesticular, and vitelline follicles were observed in 2 lateral fields. SEM showed that the anterior half was covered with random and backwardly directed tegument spines. The lumen of the oral sucker was as a longitudinal slit encircled with type I dome-shaped papillae. The ventral sucker was wrinkled and covered by tongue-shaped tegument spines and several scattered papillae. Renschetrema indicum had a fusiform body with minute spines densely distributed in the anterior part of the body; testes sub-triangular, ovary fusiform; vitellaria were randomly distributed around the ceca and genital organs. SEM showed that the fore-body was ventrally concave and surrounded by cytoplasmic ridges equipped with numerous closely packed claw-shaped spines. The oral sucker was externally surrounded by two circles of papillae while the lip of the ventral sucker was rounded and surrounded by three papillae located in its upper end and anterolaterally.(AU)

The aim of this study was to characterize the preantral ovarian follicular population in agoutis (D. leporina) by estimating the number of follicles at each developmental category, and also describe the morphometry and the specific features of the follicle and the oocyte by using light and transmission electron microscopy. The length of each ovary was measured using a caliper rule, longitudinally sectioned into two halves and both were immediately fixed to perform the estimation of follicular population and ultrastructural analysis. The mean (±S.E.M.) population of follicular per pair of ovary was estimated at 4419.8±532.26 and 5397.52±574.91 for right and left ovaries, respectively, but no differences were observed between them. The diameters for follicles, oocyte and nuclei were: 18.62±3.40µm, 12.28±2.37µm and 6.10±0.93µm for primordial, 23.75±5.70µm, 14.22±3.00µm and 6.70±1.24µm for primary and 88.55±17.61µm, 52.85±17.56µm and 22.33±17.61µm for secondary follicles, respectively. The most of the follicles found belonged to the primordial category (86.63%), followed by primary (13.01%) and secondary (0.35%) one. Additionally, polyovular follicles were observed in all the animals and they represented 7.51% of the total follicles counted. The ultrastructural analysis showed that the oocyte presented a central and regular nuclei, displaying a homogenous mass. Among the organelles, the mitochondria were the most abundant and the oocyte Golgi apparatus was rarely observed. In conclusion, this work shows for the first time the characterization of the population of preantral follicles in the ovary of Dasyprocta leporina. Those information will be useful for further development and adaptation of biotechniques such as germplasm cryopreservation and in vitro gametes manipulation.(AU)
