Endophytic bacteria Bacillus safensis RS95 and Pseudomonas hibiscicola RS121 were evaluated for their ability to promote the growth of rice seedlings and produce indole-acetic acid (IAA) and siderophores and to solubilize phosphates. 'Guri' rice seeds were immersed in bacterial endophyte cell suspensions (separated and two-strain mixed), as well as in Escherichia coli DH5α, phosphate-buffered saline (PBS) and water treatments (negative controls). Seeds were sown on agar-water in Petri plates placed vertically at an angle of 65°. The ability of plant growth-promoting endophytic bacteria (PGPEB) to produce IAA and siderophores was determined by Salkowski colorimetric and chrome azurol S (CAS) assays, respectively. Mineral phosphate solubilization activity was calculated by inoculating the endophytes onto medium containing insoluble phosphate. PGPEB showed a positive effect on the growth of rice seedlings, causing a mean growth of shoots and primary-roots of 60 and 67%, respectively. Bacterial strains also showed positive traits for IAA and siderophore production, as well as phosphate-solubilization activity

Purpose: To evaluate the ameliorative effect of mesenchymal stem cells (MSCs) on acetic acid colitis model via Nrf2/HO-1 pathway in rats. Methods: In this study, 30 rats were divided into three groups. Acute colitis was induced by rectal administration of 4% solution of acetic acid. MSCs were injected intraperitoneally in the treatment group. Results: Increased levels of tumor necrosis factor-α (TNF-α), pentraxin-3, and malondialdehyde (MDA) in colitis group were revealed biochemically. Increased level of TNF-α and decreased levels of Nrf2 and interleukin-10 (IL-10) were observed in rectum tissues. Increased fibrous tissue proliferation, vascularization and inflammatory cell infiltration were described in the colitis group. Significant improvement was observed in MSCs treated group histopathologically. Increased immunopositivity of TNF-α, vascular endothelial growth factor (VEGF) and CD68 markers was observed in the colitis group cells, and decreased level of this positivity was observed in MSCs treated group. Conclusions: Biochemical, histopathological and immunohistochemical results strongly support the ameliorative effect of MSCs against acetic induced colitis model via Nrf2/HO-1 pathway in rats.

The objective was to evaluate the inclusion of chemical additives or bacterial inoculant in corn silage. The experimental design was completely randomized with five treatments: silage without additive; silage added with urea (3.0%); silage with limestone (3.0%); silage added with crystal sugar (3.0%); and silage with a bacterial inoculant. Five of them were opened during the fermentation process (15 days) to measure pH, and three were opened 60 days after ensiling to evaluate the ammonia nitrogen and organic acids content. For the pH at the end of the fermentation process, higher values were found for the limestone additive, followed by the silage with urea, which had higher participation of lactic acid, 19.06 ppm for limestone and 18.95 ppm for urea. Higher concentrations of acetic acid were observed in inoculant silages (18.49ppm) or silage without additive (18.46ppm). The ammonia nitrogen content was higher in the silage with urea (23.74mg dL-1), followed by the silage without additive (7.54mg dL-1), which also had the highest concentration of butyric acid (4.19ppm). The use of additives reduced the concentration of butyric acid in the silage. The bacterial inoculant was the most efficient in decreasing the pH of the ensiled material.
Objetivou-se avaliar a inclusão de aditivos químicos ou inoculante bacteriano na silagem de milho. O delineamento experimental foi inteiramente ao acaso, com cinco tratamentos: silagem sem aditivo; silagem aditivada com ureia (3,0%); silagem aditivada com calcário (3,0%); silagem aditivada com açúcar cristal (3,0%); e silagem com inoculante bacteriano. Cinco sacos de silagem foram abertos durante o processo fermentativo (15 dias) para mensuração de pH, e três 60 dias após a ensilagem, para avaliação do teor de nitrogênio amoniacal e de ácidos orgânicos. Para pH no final do processo fermentativo, maiores valores foram verificados para o aditivo calcário, seguido da silagem com ureia, os quais apresentaram maior participação de ácido lático, 19,06 ppm para calcário e 18,95 ppm para ureia. Maiores concentrações de ácido acético foram observadas nas silagens inoculante (18,49 ppm) ou sem aditivo (18,46 ppm). O teor de nitrogênio amoniacal foi superior na silagem com ureia (23,74mg dL '), seguida pela silagem sem aditivo (7,54mg dL), que também apresentou maior concentração de ácido butírico (4,19 ppm). O uso de aditivos reduziu a concentração de ácido butírico na silagem. O inoculante bacteriano foi mais eficiente que aditivos químicos no declínio do pH do material ensilado.

This study aimed to evaluate the effects of increasing levels of chitosan (CHI) on sugarcane fermentation profile and losses, chemical composition, and in situ degradation. Treatments were: 0, 1, 2, 4, and 8 g of CHI/kg of dry matter (DM). Twenty experimental silos (PVC tubing with diameter 28 cm and height 25 cm) were used. Sand (2 kg) was placed at the bottom of each silo to evaluate effluent losses, and silos were weighed 60 d after ensiling to calculate gas losses. Samples were collected from the center of the silo mass to evaluate silage chemical composition, in situdegradation, fermentation profile, and mold and yeast count. Data were analyzed as a completely randomized design, and the treatment effect was decomposed using polynomial regression. Chitosan linearly increased acetic acid and NH3-N concentration, while yeast and mold count, and ethanol concentration decreased. Intermediary levels of CHI (from 4.47 to 6.34 g/kg DM) showed the lower values of effluent, gas, and total losses. There was a quadratic effect of CHI on the content of non-fiber carbohydrates, neutral and acid detergent, and in situ DM degradation. The lowest fiber content was observed with levels between 7.01 and 7.47 g/kg DM, whereas the highest non-fiber carbohydrate content and in situ DM degradation were found with 6.30 and 7.17 g/kg DM of CHI, respectively. Chitosan linearly increased acetic acid and NH3-N concentration, whereas it linearly reduced ethanol concentration and count of yeast and mold. Thus, intermediary levels of CHI, between 4.47 and 7.47 g/kg of DM, decrease fermentation losses and improve the nutritional value of sugarcane silage.(AU)
Foram avaliados os efeitos do aumento dos níveis de quitosana (CHI) sobre o perfil e as perdas fermentativas, a composição química e degradação in situ da silagem de cana-de-açúcar. Os tratamentos foram: 0, 1, 2, 4 e 8 g de CHI / kg de matéria seca (MS). Foram utilizados vinte silos experimentais (tubos de PVC com 28 cm de diâmetro e 25 cm de altura). Areia (2 kg) foi adicionada na porção inferior de cada silo para avaliar as perdas por efluentes e os silos foram pesados 60 dias após a ensilagem para calcular as perdas por gases. Amostras foram coletadas do centro da massa do silo para avaliar a composição química, degradação in situ, perfil fermentativo e a contagem de fungos e leveduras da silagem. Os dados foram analisados como um delineamento inteiramente casualizado e o efeito do tratamento foi decomposto usando regressão polinomial. A CHI aumentou linearmente a concentração de ácido acético e N-NH3, enquanto diminuiu a contagem de leveduras e bolores e a concentração de etanol. The aim of this study was to evaluate diets supplemented with probiotic (Bacillus amyloliquefaciens) and organic acids (lactic, acetic and butyric acid) in attempt to replace the antibiotics and anticoccidial (avilamycin + sodium monensin) growth promoters on performance and economic viability of broilers challenged by Eimeria acervulina, E. maxima and E. tenella. A total of 900 male Cobb® chickens, with a mean weight of 39.90g, were distributed in a completely randomized design in a 2 × 2 + 1 factorial arrangement: supplementation or not of probiotic and organic acids and a treatment with inclusion of antibiotics, comprising five treatments with six replicates. For the studied performance variables, there was no effect of the isolated additives and no interaction between them (p>0.05). Only the antibiotics promoted better results for weight gain (WG), feed intake (FI) and feed conversion rate (FCR). Therefore, the use of organic acids and probiotic, isolated or associated, provided lower performance to those receiving antibiotics, not improving the performance of chickens under the imposed challenge conditions. The highest revenues were generated with the use of antibiotics, providing greater profits.(AU)

Canola (Brassica napus L. var. oleifera) is cultivated in the Southern Brazil as an alternative winter crop in the rotation system. No-tillage and hydromorphic soils favor the formation of aliphatic organic acids with short chain and low molecular weight, as acetic, butyric, and propionic acids. These acids may cause negative effects on the canola production. Thus, this study aimed to evaluate the response of five canola hybrid genotypes under the effect of acetic, butyric and propionic acids on the germination and initial phase of seedling growth. The results showed the characters evaluated respond differently to treatments applied within the same genotype. Therefore, the selection of a genotype with a resilient performance for the characters evaluated under stress is complex. Nevertheless, a sensitive profile response was observed in Hyola 433 genotype under different organic acids. Furthermore, acetic acid showed no negative impact on germination and initial growth of the genotypes.

O sistema de plantio direto e a ocorrência de solos hidromórficos favorece a formação de ácidos orgânicos alifáticos com cadeia curta e baixo peso molecular, como os ácidos acético, butírico e propiônico. Estes ácidos podem causar efeitos negativos na produção de canola. Assim, este estudo teve por objetivo avaliar a resposta de cinco genótipos híbridos de canola sob o efeito dos ácidos acético, butírico e propiônico na germinação e na fase inicial do crescimento da plântula. Os resultados demonstraram que os caracteres avaliados respondem de forma diferente em relação aos tratamentos aplicados dentro do mesmo genótipo. Dessa forma, a seleção de um genótipo que manteve um comportamento padrão nos caracteres avaliados sob estresse é dificultado. Entretanto, em uma visão geral, o genótipo Hyola 433 manteve respostas características de sensibilidade nos caracteres avaliados em relação aos ácidos utilizados. Ainda, foi detectado que o ácido acético teve menor efeito na germinação e na fase inicial de crescimento dos genótipos estudados.

The aim of this study was to study the effect of the auxin naphthalene acetic acid (NAA) and the brassinosteroid 28-homocastasterone (28-HCS) applied every 15 days (harvest 2015/16) or every 21 days (harvest 2016/17) after full bloom, on the physical, chemical and biochemical attributes of the Galaxy apples. The study was performed at a commercial orchard at Vacaria county, Rio Grande do Sul State, Brazil. The following treatments were applied: water (control), NAA (0.1%), 28-HCS (10‒6 M) and NAA (0.1%) + 28-HCS (10‒6 M). For this purpose, 300 μL of the plant growth regulator solutions were applied to the fruit peduncles. Fruits treated with NAA every 21 days presented higher starch-iodine index and lower total soluble solids (TSS), titratable acidity (TA), lightness index (L) and hue angle (h°) than 28-HCS-treated fruits. Fruits treated every 15 days with NAA presented reduced skin rupture force (SRF), total antioxidant activities (TAA) of the skin and flesh, and hydrogen peroxide content compared to the control fruits. In addition, NAA application every 15 and 21 days resulted in increased anthocyanin content in the skin. Application of NAA + 28-HCS reduced superoxide dismutase activity. 28-HCS applications increased TSS, color attributes (C and h°) of the skin, total antioxidant activity of the skin and flesh and peroxidase activity compared to control fruits. These results are expected to help to understand how plant growth regulators affect apple quality. In addition, results described here are also expected to help on the development of strategies to reduce post harvest losses and to increase fruit shelf life.(AU)
O objetivo deste estudo foi avaliar a ação do ácido naftaleno acético (ANA) e do brassinosteroide (catasterona), aplicados a cada 15 (safra 2015/16) ou 21 (safra 2016/17) dias a partir dos 40 dias após a plena floração, nos atributos físico-químicos e bioquímicos de maçãs Galaxy. O estudo foi realizado em um pomar comercial de macieiras Galaxy no município de Vacaria, RS. Os tratamentos avaliados foram: água (controle), ANA (0,1%), catasterona (10‒6 M) e ANA (0,1%) + catasterona (10‒6 M). As aplicações foram realizadas no pedúnculo dos frutos, utilizando 300 µL de solução por pedúnculo. Frutos tratados com ANA a cada 21 dias tiveram maior índice de iodo-amido e menores valores de sólidos solúveis (SS), acidez titulável e dos atributos de cor L e ângulo hue (h°) do que os frutos tratados com catasterona. Aplicações de ANA a cada 15 dias reduziu a força de ruptura da casca (FRC), atividade antioxidante (AAT) da casca, da polpa e conteúdo de peróxido de hidrogênio. Além disso, ANA aplicada a cada 15 ou 21 dias aumentaram o teor de antocianinas na casca. ANA + catasterona reduziu a atividade da enzima superóxido dismutase. Entretanto, aplicações de catasterona a cada 15 dias aumentou o teor de SS, atributos de cor (C e h°) na casca, atividade antioxidante da casca e da polpa, compostos fenólicos totais (CFT) e atividade da enzima peroxidase dos frutos de maçãs. Portanto, estudos adicionais devem ser realizados para confirmar a ação desses fitorreguladores na manutenção da qualidade e redução das perdas pós-colheitas de maçãs Galaxy.(AU)

Abstract Bacterial endophytes are considered to have a beneficial effect on host plants, improving their growth by different mechanisms. The objective of this study was to investigate the capacity of four endophytic Bacillus strains to solubilize iron phosphate (Fe-P), produce siderophores and indole-acetic acid (IAA) in vitro, and to evaluate their plant growth promotion ability in greenhouse conditions by inoculation into pearl millet cultivated in a P-deficient soils without P fertilization, with Araxá rock phosphate or soluble triple superphosphate. All strains solubilized Fe-P and three of them produced carboxylate-type siderophores and high levels of IAA in the presence of tryptophan. Positive effect of inoculation of some of these strains on shoot and root dry weight and the N P K content of plants cultivated in soil with no P fertilization might result from the synergistic combination of multiple plant growth promoting (PGP) traits. Specifically, while B1923 enhanced shoot and root dry weight and root N P content of plants cultivated with no P added, B2084 and B2088 strains showed positive performance on biomass production and accumulation of N P K in the shoot, indicating that they have higher potential to be microbial biofertilizer candidates for commercial applications in the absence of fertilization.

Although growth regulators of the auxin group are normally used to induce rooting of cuttings, in recent years, research has been of interest to obtain alternative methods for the induction of rhizogenesis in cuttings of plants with difficult vegetative propagation, such as Peach tree cv. Chimarrita. This makes the Cyperus rotundus L. a large-scale plant found in much of the world, an alternative for the induction of rooting in cuttings, because it has phenolic compounds that have a synergistic effect with the endogenous auxin. The objective of this study was to evaluate the rooting capacity of hardwood peach cuttings with the application of different concentrations of aqueous extract of C. rotundus L. in comparison with the application of the plant growth regulator indole- 3-acetic acid. The treatments consisted of different concentrations of the aqueous extracts of roots and shoots of C. rotundus L. and the growth regulator indole-3-acetic acid. The percentage of live cuttings, rooted cuttings, callus, and the length of roots and dry matter of the root system were evaluated. The aqueous extract of shoot and root system of C. rotundus L. does not promote the rooting of hardwood peach cuttings cv. Chimarrita within 90 days. The 500 mg L-1 concentration of indole-3-acetic acid promotes greater rooting of the hardwood peach cuttings.
Embora os reguladores de crescimento do grupo das auxinas sejam comumente utilizados para induzir o enraizamento de estacas, nos últimos anos, as pesquisas têm voltado interesse para obtenção de métodos alternativos de indução da rizogênese em estacas de plantas de difícil propagação vegetativa, como a do pessegueiro cv. Chimarrita. Isso faz com que a tiririca, planta encontrada em larga escala em grande parte do mundo, apresente-se como uma alternativa para a promoção de enraizamento de estacas, por possuir compostos fenólicos que apresentam efeito sinergístico com a auxina endógena. Diante do exposto, o objetivo deste trabalho foi avaliar a capacidade de enraizamento de estacas lenhosas de pessegueiro cv. Chimarrita com a aplicação de diferentes concentrações do extrato aquoso de Cyperus rotundus L. em comparação com a aplicação do regulador de crescimento ácido indol-3-acético. Os tratamentos consistiram em diferentes concentrações de extratos aquosos do sistema radicular e de parte aérea de C. rotundus L. e do regulador de crescimento ácido indol-3-acético. Avaliou-se a percentagem de estacas vivas, de estacas enraizadas, de calos, o comprimento da raiz primária e a matéria seca do sistema radicular. O extrato aquoso de parte aérea e sistema radicular de C. rotundus L. não promove o enraizamento de estacas lenhosas de pessegueiro cv. Chimarrita no período de 90 dias.

PURPOSE:To investigate the effect of medical ozone treatment on the experimental acute distal colitis in rats.METHODS:Eighteen rats were randomly distributed into three equal groups; control, acute distal colitis (ADC) without and with medical ozone treatment. Rats in the control group were taken saline. ADC was performed by rectal way with 4% acetic acid in groups 2 and 3, and the group 3 was treated with medical ozone for three weeks both rectally and intraperitoneally. At the twenty second day the distal colons samples were obtained for malondialdehyde and myeloperoxidase, blood samples were obtained to measure the levels of TNF-α and IL-1β levels. Histolopatological examination was evaluated with Ki-67, IL-1β and VEGF immunostaining densities.RESULTS:There was significant increase in tissue MDA, MPO activity, TNF-α and IL-1β after ozone administration. There was also a significant difference at immunostaining densities of histopathological examination.CONCLUSIONS:Medical ozone treatment ameliorated the experimental acute distal colitis induced by acetic acid in rats. Its possible effect is by means of decreasing inflammation, edema, and affecting the proliferation and the vascularization.(AU)

A high concentration of histamine, one of the biogenic amines (BAs) usually found in fermented foods, can cause undesirable physiological side effects in sensitive humans. The objective of this study is to isolate indigenous Acetobacter strains from naturally fermented Bokbunja vinegar in Korea with reduced histamine production during starter fermentation. Further, we examined its physiological and biochemical properties, including BA synthesis. The obtained strain MBA-77, identified as Acetobacter aceti by 16S rDNA homology and biochemical analysis and named A. aceti MBA-77. A. aceti MBA-77 showed optimal acidity % production at pH 5; the optimal temperature was 25 °C. When we prepared and examined the BAs synthesis spectrum during the fermentation process, Bokbunja wine fermented with Saccharomyces cerevisiae showed that the histamine concentration increased from 2.72 of Bokbunja extract to 5.29 mg/L and cadaverine and dopamine was decreased to 2.6 and 10.12 mg/L, respectively. Bokbunja vinegar prepared by A. aceti MBA-77 as the starter, the histamine concentration of the vinegar preparation step was decreased up to 3.66 mg/L from 5.29 mg/L in the wine preparation step. To our knowledge, this is the first report to demonstrate acetic acid bacteria isolated from Bokbunja seed vinegar with low spectrum BA and would be useful for wellbeing vinegar preparation.(AU)

Certain nodulating nitrogen-fixing bacteria in legumes and other nodule endophytes perform different plant-growth promoting processes. The objective of this study was to evaluate 26 bacterial strains isolated from cowpea nodules grown in floodplain soils in the Brazilian savannas, regarding performance of plant-growth promoting processes and ability to enhance cowpea growth. We also identified these strains by 16S rRNA sequencing. The following processes were evaluated: free-living biological nitrogen fixation (BNF), solubilization of calcium, aluminum and iron phosphates and production of indole-3-acetic acid (IAA). The abilities to nodulate and promote cowpea growth were evaluated in Leonard jars. Partial sequencing of the 16S rRNA gene identified 60 % of the strains as belonging to genus Paenibacillus. The following four genera were also identified: Bacillus, Bradyrhizobium, Enterobacter and Pseudomonas. None of the strains fixed N2 free-living. Among the strains, 80 % solubilized Ca phosphate and one solubilized Al phosphate and none solubilized Fe phosphate. The highest IAA concentrations (52.37, 51.52 and 51.00 g mL1) were obtained in the 79 medium with tryptophan by Enterobacter strains UFPI B5-7A, UFPI B5-4 and UFPI B5-6, respectively. Only eight strains nodulated cowpea, however, all increased production of total dry matter. The fact that the strains evaluated perform different biological processes to promote plant growth indicates that these strains have potential use in agricultural crops to increase production and environmental sustainability.

The main objective of the present study was to isolate phytohormone-producing, phosphate-solubilizing strains of Azospirillum from wheat to be used as inoculants for plant growth promotion. Five Azospirillum strains were isolated from the rhizosphere of field-grown wheat (Triticum aestivum L.), and it was confirmed by BOX-polymerase chain reaction (PCR) that the isolates were different and not re-isolates of the same strain. Sequence analysis of the PCR-amplified 16S rRNA gene indicated that four isolates showed maximum similarity to Azospirillum brasilense and one isolate showed maximum similarity to Azospirillum zeae. This is the first report indicating the presence of an A. zeae like isolate in the wheat rhizosphere in Pakistan. The bacterial isolates were characterized for their plant growth-promoting traits, phosphate solubilization, and indole-3-acetic acid (IAA) production. None of the isolates showed phosphate solubilization activity in the commonly used Pikovskaya medium. However, all strains (except AzoK4) exhibited ability to solubilize tricalcium phosphate (TCP) in modified Pikovskaya medium in which sucrose was replaced by Na-malate, as well as in TCP-supplemented Luria-Bertani (LB) medium. Organic acids, such as acetic, citric, lactic, malic, and succinic acids, were detected in culture supernatants of the tested Azospirillum strains. All strains exhibited ability to produce IAA in the growth medium, except Azospirillum sp. AzoK1. Among the strains tested, the maximum IAA production (30.49 ± 1.04 mg L-1) and phosphate solubilization (105.50 ± 4.93 mg L-1) were shown by a pure culture of Azospirillum sp. AzoK2. In pot experiments, single-strain inocula of Azospirillum sp. AzoK1 and AzoK2 improved wheat plant growth.(AU)

Common pepper (Capsicum annuum L.) is one of the most important vegetables in the world, and extensive breeding efforts are being made to develop new improved strains of this species. In this regard, in vitro culture of immature embryos may help breeders accelerate breeding cycles and overcome interspecific barriers, among other applications. In this study, we have optimized a protocol for in vitro culture of immature embryos of C. annuum. Levels of indole-3-acetic acid (IAA) and zeatin have been tested to improve the efficiency (germination rates) of this technique in C. annuum embryos at the four main immature stages (i.e. globular, heart, torpedo, and early cotyledonary) from four varietal types of this species (California Wonder, Piquillo, Guindilla, and Bola). The effect of 5-day initial incubation in the dark was also tested on the most efficient hormone formulation. On average, relatively low levels of both IAA and zeatin (0.01 mg L¹ each) (M1) provided the highest germination rates, particularly in the advanced stages (torpedo and cotyledonary). To a lesser extent, the lack of these growth regulators (M0) or high IAA (0.2 mg L¹)/low zeatin (0.01 mg L¹) (M2) combination also had a positive response. On the contrary, high zeatin levels (0.2 mg L¹) produced very low germination rates or callus development (efficiency 0-7 %). Different responses were also found between genotypes. Thus, considering the best media (M0, M1, M2), Bola embryos had the highest rates. M1 plus 5-days of initial dark incubation (M1-D) improved the efficiency rates at all embryo stages, particularly in the earliest (globular) embryos which increased from 3 % to > 20 %.
