Resumo

The medicinal attributes of honey appears to overshadow its importance as a functional food. Consequently, several literatures are rife with ancient uses of honey as complementary and alternative medicine, with relevance to modern day health care, supported by evidence-based clinical data, with little attention given to honey’s nutritional functions. The moisture contents of honey extracted from University of Veterinary and Animal Sciences, Lahore honey bee farm was 12.19% while that of natural source was 9.03 ± 1.63%. Similarly, ash and protein contents of farmed honey recorded were 0.37% and 5.22%, respectively. Whereas ash and protein contents of natural honey were 1.70 ± 1.98% and 6.10 ± 0.79%. Likewise fat, dietary fiber and carbohydrates contents of farmed source documented were 0.14%, 1.99% and 62.26% respectively. Although fat, dietary fiber and carbohydrates contents of honey taken from natural resource were 0.54 ± 0.28%, 2.76 ± 1.07% and 55.32 ± 2.91% respectively. Glucose and fructose contents of honey taken out from honeybee farm were 27% and 34% but natural source were 22.50 ± 2.12% and 28.50 ± 3.54%. Glucose and fructose contents of honey taken out from honeybee farm were 27% and 34% but natural source were 22.50 ± 2.12% and 28.50 ± 3.54%. Similarly, sucrose and maltose contents of farmed honey were 2.5% and 12% while in natural honey were 1.35 ± 0.49% and 8.00 ± 1.41% respectively. The present study indicates that such as moisture, carbohydrates, sucrose and maltose contents were higher farmed honey as compared to the natural honey. In our recommendation natural honey is better than farmed honey.

Resumo

This study aimed to assess infestation levels of the mite Varroa destructor and microspore Nosema spp. in apiaries in the Picos microregion of Piauí state, Brazil, during the dry season. To that end, bees were collected (n=31) from 19 apiaries between September and October 2016. Infestation levels of the parasites were analyzed at the Laboratory of the Bee Technology Center, an institution affiliated with the Senador Helvídio Nunes de Barros Campus of the Federal University of Piauí. Varroa destructor was identified in 54% of the colonies, with infestation levels varying from 0 to 10.54%, and an average of 3.31% ± 4.12. Analysis of Nosema spp. revealed that all the colonies sampled exhibited microsporidia, albeit at low (1 to 1.4 spores/bee) and very low levels (0.35 to 0.95 spores/bee) of infestation. Given the relevance of beekeeping in the semiarid region of Piauí, it was concluded that although infestation levels were deemed low, there is a need to regularly monitor the health of colonies in the apiaries.(AU)


Objetivou-se avaliar o nível de infestação do ácaro Varroa destructor e do microsporídio Nosema spp em apiários da microrregião de Picos, Piauí, durante o período seco. Foram realizadas coletas de amostras de abelhas (n=31) em 19 apiários, entre setembro e outubro de 2016. As análises foram realizadas no Laboratório do Centro de Tecnologia Apícola. Observou-se a ocorrência de Varroa destructor em 54% das colônias, sendo que os níveis de infestação variaram de 0 a 10,54%, com média de 3,31% ± 4,12. A pesquisa referente à Nosema spp revelou que todas as colônias amostradas apresentavam o microsporídio, no entanto em níveis baixos (1 a 1,4 esporos/abelha) e muito baixos (0,35 a 0,95 esporos/abelha) de infestação. Considerando a relevância da apicultura para a região semiárida do Piauí, concluiu-se que embora os níveis de infestação tenham sido consi-derados baixos, ressalta-se a necessidade de monitoramentos periódicos quanto à sanidade das colônias nos apiários.(AU)

This study was designed to examine the protective effects of nano-selenium and nano-zinc oxide on queen and workers performance under heat stress condition and gene expression of heat shock protein 70 (hsp70) as an index of heat tolerance. Sixty colonies were randomly assigned to five treatments with 12 replicates from June until early September. Sugar syrup (50%) containing no supplement or nano-selenium at levels of 50 and 100 µg L-1 or nano-zinc at levels of 100 and 200 µg L-1 was fed to colonies. Nano-selenium supplementations had no effect, but nano-zinc at level of 100 µg L-1 significantly decreased body malondialdehyde concentration. The highest bee population was seen in nano-zinc at level of 100 µg L-1 and the lowest one in control group. The lowest and the highest body weight, fat and protein deposition was found in group received nano-zinc at level of 100 µg L-1 and control, respectively. The highest gene expression was for group received nano-zinc at level of 100 µg L-1 In group received nano-zinc at level of 100 µg L-1, an increase in hsp70 gene expression was found. In conclusion, nano-zinc oxide at level of 100 µg L-1 could increase queen and worker performance and heat resistance of bees in the hot climate condition.(AU)

Resumo

Aedes aegypti is the vector of several viral diseases. The main way to control these diseases is to fight the vector. Thus, it is necessary to breed mosquitoes in the laboratory in order to develop strategies to control these insects. In laboratories, different carbohydrates are used for feeding mosquitoes. The aim of this study is to evaluate the longevity and the weight of Ae. aegypti fed with different carbohydrates diets. As methods, 120 mosquitoes were distributed in insectaries and each group received a different diet, based on honey, dextrose or maltodextrin. To assess the longevity, survival analysis was performed using the Long Rank test and chi square test. To assess the weight, the dead insects were frozen and weighed at the end of the experiment. As results it was observed that mosquitoes fed with the honey, maltodextrin and dextrose diet lived on average 33, 35 and 47 days respectively. When weight was assessed, mosquitoes fed with honey weighed 125 ± (35.3) µg, while those fed with dextrose and maltodextrin weighed 225 ± (35.3) µg and 275 ± (35.3) µg respectively. The results show that the intake of dextrose and maltodextrin by Ae. aegypti adults increases their survival and their weight.(AU)


O Aedes aegypti é vetor de várias doenças virais. A principal maneira de controlar essas doenças é combatendo o seu vetor. Nesse sentido, é necessário criar esses mosquitos em laboratório, visando desenvolver estratégias de controle. Nos laboratórios, diferentes carboidratos são utilizados na alimentação de mosquitos. O objetivo deste estudo é avaliar longevidade e peso de Ae. aegypti alimentados com diferentes fontes de carboidratos. Como método, distribuíram-se 120 mosquitos insetários. Cada grupo recebeu uma dieta diferente à base de mel, dextrose ou maltodextrina. Para avaliar a longevidade, a análise de sobrevida foi realizada pelo teste de Logrank e pelo teste de qui quadrado. Para avaliar o peso, os insetos mortos foram congelados e pesados ​​no final do experimento. Como resultado, observou-se que os mosquitos alimentados com a dieta à base de mel, maltodextrina e dextrose viveram em média 33, 35 e 47 dias, respectivamente. Com relação ao peso, os mosquitos alimentados com mel pesavam 125 ± (35,3)µg, enquanto os alimentados com dextrose e maltodextrina pesavam 225 ± (35,3)µg e 275 ± (35,3)µg, respectivamente. Os resultados mostram que a ingestão de dieta à base de dextrose e maltodextrina por Ae. aegypti adultos aumenta sua sobrevivência e seu peso.(AU)

Resumo

The Mercosur Technical Regulation for honey determines microbiological parameters for total coliforms and molds and/ yeasts, and absence of physical contaminants, of any nature. The purpose of this study was to verify the biological and physical contamin of Apis mellifera ligustica and Melipona fasciculata, commercialized in the northeastern region of the State of Pará, Brazil. The fourteen samples from informal trade of the municipalities of Bragança, Capanema, Nova Timboteua, São João de Pirabas, Salinas and Tracauateua were analyzed for the Most Probable Number of colifroms at 35ºC, Fungus and Yeast counts, besides the detection of dirt by Macro and Microscopy techniques. The results obtained from the microbiological analyzes were compared to the standard required by MERCOSUR, for coliforms. 57.14% of the samples were non-standard and ranged from <3.0 NMP/g to 20 NMP/g. The molds and/ yeast counts ranged from 1x10¹ CFU/g to 8.6x10² CFU/g, with atotal of 21.42% of the samples being in disagreement with the legislation. From the macroscopic analysis, it was observed that, although 50% of the samples did not present any apparent dirt, microscopically it was possible to observe dirt in 100% of the samples of honey analyzed, which is out of the standard required by the Brazilian legislation. This shows the low sanitary quality of honey commercialized in this region of the state of Pará.


O Regulamento Técnico do Mercosul para mel, determina parâmetros microbiológicos para coliformes totais e fungos e/leveduras, e ausência de contaminantes físicos, de qualquer natureza. A proposta do presente estudo foi verificar a contaminação biológica e física em méis de abelha das espécies Apis mellifera ligustica e Melipona fasciculata, comercializados na região Nordeste do Estado do Pará, Brasil. As amostras provenientes do comercio informal dos de municípios de, Bragança, Capanema, Nova Timboteua, São João de Pirabas, Salinas e Tracauateua, foram analisadas quanto a determinação do Número Mais Provável de coliformes a 35ºC, Contagem de Fungos e/leveduras, além da detecção de sujidades pelas técnicas de Macro e Microscopia. Os resultados obtidos a partir das análises microbiológicas foram comparadas ao padrão exigido pelo MERCOSUL, para coliformes, 57,14% das amostras encontravam-se fora do padrão variaram de <3,0 NMP/g a 20NMP/g. Já a contagem de fungos e/leveduras variou de 1x10¹ UFC/g a 8,6x10² UFC/g, no total de 21,42 % das amostras apresentaram-se em desacordo com a legislação. A partir da análise macroscópica observou-se que, embora 50% das amostras não apresentassem sujidades aparentes, porém microscopicamente foi possível observar a presença sujidades em 100% das amostras de méis analisadas encontrando-se fora do padrão exigido pela legislação Brasileira. Demostrando a baixa qualidade sanitária do mel comercializado nessa região do estado do Pará.

Africanized honey bee populations (Apis mellifera L.) have been decreasing mainly due to the intense use of synthetic insecticides associated with pollution and climate change. To minimize these impacts on the environment and bee populations, the use of biological control agents has been intensified. These products are generally safer for non-target insects, such as bees, which are important pollinating insects. Thus, the objective of this study was to evaluate the impact of entomopathogenic nematodes on the longevity of the Africanized honey bee A. mellifera workers. Seven treatments were used: Heterorhabditis amazonensis, Heterorhabditis bacteriophora, Heterorhabditis indica, Steinernema carpocapsae, Steinernema feltiae, and Steinernema rarum, at a concentration of 40 infective juveniles per cm² (IJs/cm²), and a control in which autoclaved distilled water was used. Two bioassays were performed: 1) spraying nematodes on the workers and 2) spraying nematodes on glass plates, in which the bees remained for two hours. Each treatment consisted of five replicates with 20 bees each. Bees were kept in cages of PVC (20 × 10 cm) covered with a voile fabric and provided pieces of cotton soaked in water and Candy paste. The cages were kept in a climatized room (27 ± 2 °C temperature, 60 ± 10% relative humidity, and 12 h photophase) and the mortality was evaluated from 12 to 240 hours. In bioassay 1, the three treatments with nematodes of the genus Steinernema reduced the longevity of the workers (103.9, 96.3, and 99.6 h) when compared to treatments with Heterorhabditis (149.7, 126.8, and 134.7 h), of which, only H. amazonensis (149.7 h) did not differ from the control (166.0 h). As populações de abelhas africanizadas (Apis mellifera L.) vêm diminuindo devido, principalmente, ao intenso uso de inseticidas sintéticos. Desde modo, o emprego de agentes de controle biológico tem-se intensificado de forma a minimizar este impacto no ambiente. Estes produtos são, de modo geral, mais seguros aos insetos não-alvos, como as abelhas que são importantes insetos polinizadores. Desta forma, o objetivo deste trabalho foi avaliar os efeitos de nematoides entomopatogênicos utilizados para controle de insetos pragas, sobre a longevidade de operárias de A. mellifera africanizada. A adoção de práticas fraudulentas é comum na cadeia produtiva do mel e está associada à popularidade desse alimento e a dificuldade de detecção da adulteração do produto a olho nu ou por degustação. O objetivo deste trabalho foi verificar se práticas fraudulentas são adotadas em méis comercializados no estado do Pará. Para a realização da pesquisa, 14 amostras comerciais de méis provenientes de 06 (seis) municípios, no estado do Pará foram submetidas às análises Lund, Fiehe, Lugol e melissopalinologia. Os resultados obtidos demostraram que 57,14% das amostras apresentaram alteração do precipitado para reação de Lund, indicando alteração no teor proteico. A coloração o azul intenso na reação de Lugol foi observada em 64,28% das amostras, indicando possível adição de amido. Ainda, 85,71% das amostras analisadas apresentaram mudança de coloração para a reação de Fiehe, indicando alteração no teor de hidroximetilfurfural. O conteúdo polínico foi observado em somente 35,71% das amostras e apenas 2,42% do total das amostras analisadas foram consideradas autenticas mediante as análises realizadas. Concluímos que os testes de autenticidade, quando aplicados em conjunto, foram capazes de detectar fraudes nas mostras de mel comercializadas no estado do Pará e que adulterações nos méis comercialmente disponíveis na região alvo do estudo são uma realidade.

The results obtained showed 57.14% of the samples showed a change in the precipitate for Lund's reaction, indicating a change in protein content. The intense blue staining in the Lugol reaction was observed in 64.28% of the samples, indicating a possible addition of starch. In addition, 85.71% of the samples showed a color change for the Fiehe reaction, indicating a change in the hydroxymethylfurfural content. Pollen content was observed in only 35.71% of the samples and only 21.42% of the total samples analyzed were considered authentic through the analyzes performe. We conclude that the authenticity tests, when applied together, were able to detect fraud in honey samples marketed in the state of Pará and that adulterations in the honey commercially available in the target region of the study are a reality.

Resumo

The objective of this research was to estimate the correlations between the biometric and reproductive parameters of Melipona scutelaris. Four collections were performed with 30-day intervals, consisting of the evaluation of each colony, considering the following parameters: honey pot size and volume (HPS and VHP), estimated honey production (EHP), number of combs (NC), mean diameter of the combs (MDC), height of the combs (HC), height of rearing cells (HRC), number of pups (NP) population (POP) and wax envelope in pups. The data were analyzed using the SAS software (2004), and the Pearson correlation coefficient was used for the correlation calculations. The correlations of the studied variables of production and biometrics of Melipona scutellaris colonies made it possible to perceive that the variable number of honey pots (NHP) present a higher correlation with HPS, EPH, DMC, HRC, NP and POP, followed by size of pots of honey (HPS) with: VHP, EHP, MDC, HC and POP. For future studies using the productive and biometric characteristics of the bees Melipona scutellaris can be considered for EPH, VHP, EPH, NC and POP, as variable responses.(AU)

Bees are fundamental in several aspects, especially in relation to plant biodiversity and pollination. Recently, immense losses are being faced in the number of Brazilian colonies, mainly in southern states of the country, which has a strong beekeeping activity. There are indications that, among the reasons for the losses, pathogens that affect the health of bees may be involved. Among them, the microsporidium Nosema and the black queen cell virus (BQCV) stand out for their prevalence. In this study, 92 colonies of 17 apiaries from southern Brazil were evaluated for infection by Nosema ceranae, Nosema apis and BQCV. Nucleic acid extractions and cDNA synthesis were performed from adult bee samples, followed by Reverse Transcription Polymerase Chain Reaction (RT-PCR) and multiplex PCR. Eight BQCV positive samples were subjected to sequencing. The results showed that N. ceranae and BQCV are circulating in the Southern region of the country, which may be the reason for the loss of colonies. N. apis was not found. N. ceranae was found in 57.6% (53/92) of the colonies and BQCV in 32.6% (30/92). Co-infection was found in 25% (23/92) of the colonies studied, a factor that is suggested to be reducing the hosts' longevity due to the synergistic action of the pathogens. The samples submitted to sequencing indicated similarity of 96.8 to 100% between them, in addition to strong similarity with sequences from Asia, United States, Germany and Peru. This study reports the circulation of N. ceranae and BQCV in apiaries in southern Brazil, in addition to being the first phylogenetic analysis of the Brazilian BQCV sequence.(AU)


As abelhas mostram-se fundamentais em diversos aspectos, especialmente com relação à biodiversidade de plantas e polinização. Recentemente, estão sendo enfrentadas imensas perdas no número de colônias brasileiras, principalmente nos estados do sul do país, com forte atividade apícola. Há indicativos de que, dentre as razões para as perdas, possam estar envolvidos patógenos que afetam a saúde das abelhas. Dentre eles, o microsporídio Nosema e o vírus da realeira negra (BQCV) destacam-se pela prevalência. Neste estudo, foram avaliadas 92 colônias, de 17 apiários do sul do Brasil, a respeito da infecção por Nosema ceranae, Nosema apis e BQCV. Foram realizadas extrações de ácidos nucleicos e síntese de cDNA a partir de amostras de abelhas adultas, seguidos de Reação em Cadeia da Polimerase-Transcriptase Reversa (RT-PCR). Oito amostras positivas para BQCV foram submetidas a sequenciamento. Os resultados mostraram que N. ceranae e BQCV estão circulando na região sul do país, podendo ser a razão para as perdas de colônias. N. apis não foi encontrado. N. ceranae foi encontrado em 57.6% (53/92) das colônias e BQCV em 32.6% (30/92). Foi encontrada coinfecção por ambos em 25% (23/92) das colônias estudadas, fator que sugere a diminuição da longevidade do hospedeiro por ação sinérgica dos patógenos. As amostras submetidas ao sequenciamento indicaram similaridade de 96.8 a 100% entre elas, além de forte similaridade com sequências da Ásia, Estados Unidos, Alemanha e Peru. Este estudo relata a circulação de N. ceranae e BQCV nos apiários do sul do Brasil, além de ser a primeira análise filogenética da sequência do BQCV brasileiro.(AU)
