Background: In dogs with bacterial cystitis that is resistant to multiple antibiotics, resulting from repeated infections andantimicrobial administration, especially if the dog has impaired renal function and the induction of systemic side effectsby intravenous or oral administration is a concern, intravesical instillation of antibiotics might represent an alternativetreatment option. In human and veterinary medicine, a number of studies showed intravesical instillation of antibiotics iseffective for the therapy multidrug-resistant bacterial urinary tract infection (UTI). This report firstly illustrates successfulintravesical meropenem treatment of a UTI caused by multidrug-resistant Escherichia coli with no systemic side effectsin dog with chronic kidney disease (CKD).Case: A 15-year-old spayed female Maltese was presented with recurrent bacterial cystitis. The risk factors for the recurrent UTI were spinal cord injury and CKD which had been managed for 1 year. Ultrasound-guided cystocentesis wasperformed to obtain a urine sample for urinalysis, bacteriologic culture, and antibiotic susceptibility testing. Bacterialcystitis caused by multidrug-resistant Escherichia coli was diagnosed on the basis of bacterial culture, and antimicrobialsusceptibility testing. Because the dog had CKD, reducing the clearance of meropenem, intravesical instillation of antibiotics was initiated. The intravesical instillation process consisted of the emptying of the urinary bladder, infusion of adiluted meropenem solution (8.5 mg/kg diluted in 20 mL of saline solution) into the bladder through a urethral catheter,and retention of the meropenem solution in the bladder for 1 h, and its removal. Materials, Methods & Results: A total of 306 samples were collected from the veterinary hospital environment (swabs from consultation tables, surgical tables, door handles, hospitalization cages, stethoscopes, thermometers, and muzzles). Aquaculture is one of the sectors of animal husbandry with the fastest growth rate. However, the increase in the sector's production chain without proper management can result in factors that favor the development of diseases, especially infectious diseases caused by bacteria. Many factors, such as agriculture or industry resides, improper use of antibiotics in animals or humans, have contributed to increased environmental pressure and the appearance of antibiotic-resistant bacteria, while residues from these drugs can remain in the carcasses and in water a risk to public and environmental health. From that, we identified the bacterial genus/species and their bacterial resistance to antibiotics from samples received from fish disease outbreaks for bacteriosis diagnosis between January 2017 and October 2020. Isolated bacteria were subjected to the Kirby and Bauer sensitivity test for five classes of antibiotics (penicillins, fluoroquinolones, aminoglycosides, amphenicols, and tetracyclines). Of the 181 analyzed outbreaks, 232 bacteria were isolated, including Streptococcus spp., Aeromonas spp., Edwardsiella spp., Plesiomonas shigelloides, Pseudomonas aeruginosa, Chromobacterium violaceum, Flavobacterium spp., Citrobacter spp., Enterococcus spp., Vibrio spp., Enterobacter spp., Chryseobacterium meningosepticum. ABSTRACT: The present study was aimed at subtyping of Stx1 and Stx2 genes and characterization of antimicrobial resistance in 106 Shiga toxin-producing Escherichia coli (STEC) strains isolated from cattle and sheep feces. PCR was used to determine the subtypes, and the disk-diffusion method was used to evaluate the antimicrobial resistance. Ten antibiotics from five different classes were tested. Among the isolates of bovine origin, two subtypes of Stx1 (Stx1a and Stx1c), and four subtypes of Stx2 (Stx2a, Stx2b, Stx2c, and Stx2d) were identified. In isolates of sheep origin, two subtypes of Stx1 (Stx1a and Stx1c), and four subtypes of Stx2 (Stx2a, Stx2b, Stx2c, and Stx2 g) were identified. The results obtained suggest the presence of high diversity in Stx1 and Stx2 genes. Further, 96.6% (57/59) of bovine fecal strains and 89.4% (42/47) of sheep fecal strains showed resistance to at least one tested antibiotic. In both animal species, most strains were multidrug-resistant (MDR) (67.8% in cattle and 59.6% in sheep), with no significant difference between host animals. Adult animals were eight times more likely to have STEC with greater pathogenic potential. STEC with the highest pathogenic potential were three times more likely to be multidrug-resistant than STEC with the lowest pathogenic potential. The data reported in this study suggests the occurrence of strains with high potential pathogenicity in the region studied. Therefore, the ruminants of this region are carriers of strains that can cause infections in humans.

The present study was aimed at subtyping of Stx1 and Stx2 genes and characterization of antimicrobial resistance in 106 Shiga toxin-producing Escherichia coli (STEC) strains isolated from cattle and sheep feces. PCR was used to determine the subtypes, and the disk-diffusion method was used to evaluate the antimicrobial resistance. Ten antibiotics from five different classes were tested. Among the isolates of bovine origin, two subtypes of Stx1 (Stx1a and Stx1c), and four subtypes of Stx2 (Stx2a, Stx2b, Stx2c, and Stx2d) were identified. In isolates of sheep origin, two subtypes of Stx1 (Stx1a and Stx1c), and four subtypes of Stx2 (Stx2a, Stx2b, Stx2c, and Stx2 g) were identified. The results obtained suggest the presence of high diversity in Stx1 and Stx2 genes. Further, 96.6% (57/59) of bovine fecal strains and 89.4% (42/47) of sheep fecal strains showed resistance to at least one tested antibiotic. In both animal species, most strains were multidrug-resistant (MDR) (67.8% in cattle and 59.6% in sheep), with no significant difference between host animals. Adult animals were eight times more likely to have STEC with greater pathogenic potential. STEC with the highest pathogenic potential were three times more likely to be multidrug-resistant than STEC with the lowest pathogenic potential. The data reported in this study suggests the occurrence of strains with high potential pathogenicity in the region studied. Therefore, the ruminants of this region are carriers of strains that can cause infections in humans.(AU)
O presente estudo teve como objetivo subtipar os genes Stx1 e Stx2 e caracterizar a resistência antimicrobiana em 106 isolados de Escherichia coli produtoras de toxinas Shiga (STEC) isoladas de fezes de bovinos e ovinos. Em ambas as espécies animais, a maioria das cepas foi multirresistente (MDR) (67,8% em bovinos e 59,6% em ovinos), sem diferença significativa entre as espécies animais do reservatório. Os animais adultos tiveram oito vezes mais chances de apresentar STEC com maior potencial patogênico. STEC com o maior potencial patogênico teve três vezes mais chances de ser multirresistente do que o STEC com o menor potencial patogênico. Os dados relatados neste estudo sugerem a ocorrência de cepas com alto potencial de patogenicidade na região estudada. A proximidade dos primatas não humanos (PNH) com o ser humano pode ser considerada um fator de risco para transmissão de bactérias entre essas duas populações. Neste estudo, foi investigada a microbiota anfibiôntica aeróbica oral e retal de calitriquídeos em um fragmento de Mata Atlântica localizado no Rio de Janeiro, Brasil, e foram realizados testes fenotípicos para detecção de bactérias multirresistentes nos isolados encontrados. Foram capturados 14 calitriquídeos e coletadas 21 amostras (14 de cavidade oral e sete de cavidade retal) em dois pontos da mata próximos às habitações humanas. As espécies mais frequentes, na cavidade oral, foram Klebsiella oxytoca (50,0%), K. pneumoniae (28,6%), Kluyvera ascorbata (21,4%) e Stenotrophomonas maltophilia (21,4%) e, na cavidade retal, K. pneumoniae (85,7%), Escherichia coli (28,6%) e Enterobacter spp. (42,9%). Todos os 48 isolados da família Enterobacteriaceae foram negativos para ESBL (betalactamase de espectro ampliado), mostrando-se não produtores da enzima nos dois métodos utilizados: disco-aproximação e método de detecção automatizado. Na pesquisa de ERC (enterobactérias resistentes a carbapenêmicos), esses mesmos isolados não apresentaram resistência aos antibióticos imipenem, meropenem e ertapenem. Todas as bactérias isoladas apresentam um potencial zoonótico, o que representa um risco à saúde pública e à conservação das espécies.(AU)

Mastitis is a multifactorial disease and considered one of the most critical problems in the dairy industry worldwide. The condition is characterized by reduced milk and several abnormalities in the mammary gland. This study aimed to report an outbreak of gangrenous mastitis caused by multidrug-resistant Staphylococcus haemolyticus in a Santa Inês sheep herd. Eighteen sheep were affected, and five of them with severe clinical pictures were examined. The clinical and pathological picture were variable and characterized by apathy, anorexia, emaciation, opaque and brittle hair, apparent and congested episcleral vessels, and hyperthermia. These ewes had enlarged, firm, and painful mammary glands. Macroscopically, these lesions consisted of severe gangrenous mastitis, and microscopically, the primary lesions consisted of necrosis, thrombosis, and fibrosis of the mammary parenchyma. Milk samples from one of the five severely affected ewes were collected and cultured under aerobic or microaerophilic incubation at 37°C for 24 hours on sheep blood agar. The obtained colonies were then submitted to MALDI-TOF for speciation. The colonies were also submitted to an antimicrobial susceptibility test, genotyping of virulence factors and resistance genes were also performed. The isolates showed antimicrobial multiresistance since they were resistant to seven out of 13 tested antibiotics. The isolates were also positive for two staphylococcal enterotoxigenic genes (sec and see) and fibronectin-binding protein B (fnbB).(AU)

O objetivo deste estudo foi relatar um surto de mastite gangrenosa causada por Staphylococcus haemolyticus multirresistente em um rebanho ovino Santa Inês. Dezoito ovelhas foram afetadas e cinco delas com quadro clínico severo foram examinadas. O quadro clínico-patológico era variável quanto a severidade e consistia em apatia, anorexia, magreza, pelos opacos e quebradiços e vasos episclerais aparentes e ingurgitados. As ovelhas apresentavam glândulas aumentadas, firmes e dolorosas. Macroscopicamente, as principais lesões consistiam em mastite gangrenosa e microscopicamente havia necrose do parênquima glandular, trombose e fibrose. Amostras de leite de uma das cinco ovelhas severamente afetadas foram coletadas e cultivadas sob incubação aeróbica ou microaerofílica a 37°C por 24 horas em ágar sangue de ovelha. As colônias obtidas foram então submetidas ao MALDI-TOF para especiação. Além disso, as colônias foram submetidas a um teste de suscetibilidade antimicrobiana e foi realizada a genotipagem de fatores de virulência e genes de resistência. Background: During the last years, there is a global concern about the increasing levels of antimicrobial resistance in human and veterinary medicine. Klebsiella oxytoca isolates are ubiquitous in nature, including surface water, soils, sewage, and plants. Klebsiella oxytoca is a nosocomial pathogen in humans but few studies reported as a pathogen in dogs and cats. Antimicrobial resistance represents a serious problem due to the increasing prevalence of extended-spectrum β-lactamaseproducing K. oxytoca isolates. The aim of the present study is describe the first clinical case of disseminated dermatitis in a dog caused by K. oxytoca multidrug resistant and the importance of correct protocol treatment.Case: A 4 year-old dog, Shih-tzu, female showed a disseminated dermatitis displayed generalized alopecia, with seborrheic aspect and pruritus, in non-nosocomial conditions in the city of São Paulo. Skin samples were collected and sent to a veterinary microbiology laboratory for bacterial identification and antimicrobial susceptibility testing (AST). The tests demonstrated a Gram negative bacilli, oxidase negative, catalase positive, non motile, DNase negative, glicose fermentative, lactose positive in MacConkey agar. In all biochemical complementary tests the results were indole positive. The strain of K. oxytoca was tested for resistance to the following antibiotics: amikacin, amoxicillin-clavulanic acid, ampicillin, cephalexin, cephalosporin, chloramphenicol, ciprofloxacin, doxycycline, enrofloxacin, gentamicin, imipenem, levofloxacin, marbofloxacin, meropenem, neomycin, sulfamethoxazole/trimethoprim and tetracycline. The AST result demonstrated a resistant Klebsiella oxytoca for 76,4% of tested antibiotics and sensitivity to few antibiotics such as meropenem, imipenem, neomycin, amikacin, sulfamethoxazole and trimethoprim. Proteus spp. are opportunistic multidrug resistant enterobacteria associated with diverse clinical diseases in domestic animals. However, Proteus infections in domestic animals are often misdiagnosed or considered contaminants in microbiological cultures rather than a primary agent of disease. Descriptions of Proteus infections in domestic animals are typically restricted to case reports, retrospective studies, or surveillance of other microorganisms. The present study investigated multiple antibiotic resistance indices, minimum inhibitory concentrations (MICs), and ESBL production in 73 strains of Proteus mirabilis (n = 69) and Proteus vulgaris (n = 4) isolated from domestic animals with various clinical manifestations. In dogs, the pathogen was most commonly associated with cystitis (48.21), enteritis (21.42%), otitis (14.29%), and conjunctivitis (3.57%). In bovines, the microorganism was predominant in cases of enteritis (22.22%), abscess (11.11%), otitis (11.11%), omphalitis (11.11%), and peritonitis (11.11%), and in organ fragments (11.11%). In equines (50.0%) and cats (100.0%), diarrhea was the main clinical sign. In vitro standard disk diffusion assay showed that the most effective antimicrobials against the isolates were imipenem (98.63), norfloxacin (95.89), amikacin (95.89), levofloxacin (90.41), ceftriaxone (87.64), and florfenicol (87.67). In contrast, the isolates commonly showed resistance to novobiocin (95.89), azithromycin (57.53), and trimethropim/sulfamethoxazole (39.73). Among the 73 isolates, the efficacy of amoxicillin/clavulanic acid, gentamicin, ceftriaxone, and ciprofloxacin according to MICs was 87.67%, 86.30%, 84.93%, and 82.19%, respectively. The MIC50 values of amoxicillin/clavulanic acid, ceftriaxone, ciprofloxacin, and gentamicin were, respectively, 1.0, 0.004, 0.03, and 1.0 µg/mL. [...](AU)
Proteus spp. são enterobactérias oportunistas, multirresistentes aos antimicrobianos, associadas a diversas infecções em animais domésticos. No entanto, as infecções por Proteus em animais de produção e de companhia são negligenciadas ou, por vezes, o patógeno é considerado contaminante, ainda que em infecções como agente primário. Os registros de infecções por Proteus sp. em animais domésticos estão restritos aos relatos de casos, estudos retrospectivos ou compondo estudos com outros microorganismos. O presente estudo investigou o índice de resistência múltipla (IRMA) e a concentração inibitória mínima (CIM) de 73 Proteus mirabilis (n=69) e Proteus vulgaris (n=4) a diferentes antimicrobianos, bem com a produção fenotípica de ESBL, em isolados obtidos de várias manifestações clínicas em animais domésticos. Em cães, o micro-organismo foi identificado predominantemente em casos de cistite (48,21%), enterite (21,42%), otite (14,29%) e conjuntivite (3,57%). Nos bovinos, o agente foi isolado predominantemente de casos enterite (22,22%), abscesso (11,11%), otite (11,11%), onfalite (11,11%), peritonite (11,11%), metrite (11,11%) e em fragmento de órgão (11,11%). Salmonellosis is a foodborne disease caused by bacteria of the genus Salmonella, being pigs and pork-products potentially important for its occurrence. In recent decades, some serovars of Salmonella have shown increase of resistance to conventional antimicrobials used in human and animal therapy, with serious risks for public health. The aim of this study was to evaluate feces (n=50), mediastinal (n=50), mesenteric (n=50) and mandibular (n=50) lymph nodes obtained from slaughter houses for Salmonella spp. Positive samples were serotyped and subjected to an in vitro antimicrobial susceptibility test, including the extended-spectrum beta-lactamase (ESBL) production. Salmonella species were identified in 10% (20/200) of total samples. From these, 20% (10/50) were identified in the submandibular lymph nodes, 18% (9/50) in the mesenteric lymph nodes, 2% (1/50) in feces and 0% (0/50) in the mediastinal lymph nodes. The serotypes found were Salonella Typhimurium (55%), S. enterica subsp. enterica 4,5,12: i: - (35%), S. Brandenburg and S. Derby with 5% (5% each). All strains showed resistance to at least one antimicrobial; 90% were resistant to four or more antimicrobials, and 15% were multidrug-resistant. Resistance to ciprofloxacin, tetracycline and nalidixic acid was particularly prevalent amongst the tested serovars. Here, we highlighted the impact of pigs in the epidemiological chain of salmonellosis in domestic animals and humans, as well as the high antimicrobial resistance rates of Salmonella strains, reinforcing the necessity for responsible use of antimicrobials for animals as an emergent One Health issue, and to keep these drugs for human therapy approaches.(AU)

Deste modo, o objetivo do presente trabalho foi pesquisar a presença de Salmonella a partir de fezes (n=50), linfonodos mediastinos (n=50), mesentéricos (n=50) e submandibular (n=50) oriundos de um abatedouro suíno. As cepas isoladas foram sorotipadas e testadas quanto a resistência antimicrobiana. A presença de Salmonella isolada foram em 10% (20/200) do total de amostras, sendo 20% dos linfonodos submandibulares, 18% dos linfonodos mesentéricos e 2% das fezes. Os sorotipos encontrados foram S. Typhimurium (55%), S. enterica subsp. enterica 4,5,12: i: - (35%), S. Brandenburg (5%) e S. Derby (5%). Todas a cepas apresentaram resistência a pelo menos um antimicrobiano testado, sendo 90% resistente pelo menos quatro antimicrobianos. Destes, 15% foram classificadas como multidrogas resistentes. Os antimicrobianos mais resistentes entre os sorovares isolados foram a ciprofloxacina, tetraciclina e o ácido nalidixico. A presença de cepas de Salmonella resistente a antimicrobianos na espécie suína tem gerado um grande impacto epidemiológico entre homem e animal, reforçando cada vez mais a necessidade do uso adequado de drogas principalmente relacionado com o tema "One Health".(AU)

Salmonellosis is a foodborne disease caused by bacteria of the genus Salmonella, being pigs and pork-products potentially important for its occurrence. In recent decades, some serovars of Salmonella have shown increase of resistance to conventional antimicrobials used in human and animal therapy, with serious risks for public health. The aim of this study was to evaluate feces (n=50), mediastinal (n=50), mesenteric (n=50) and mandibular (n=50) lymph nodes obtained from slaughter houses for Salmonella spp. Positive samples were serotyped and subjected to an in vitro antimicrobial susceptibility test, including the extended-spectrum beta-lactamase (ESBL) production. Salmonella species were identified in 10% (20/200) of total samples. Os primatas não-humanos (PNH) podem atuar como reservatórios de agentes patogênicos com implicação para a saúde pública. O Campus Fiocruz Mata Atlântica (CFMA) está inserido em uma região marcada pela expansão urbana, o que favorece a interação entre animais de vida livre, como os PNH, animais domésticos e o ser humano. Este estudo investigou na população de calitriquídeos (Callithrix sp.) do CFMA a circulação de agentes virais da família Flaviviridae, especificamente, o zika vírus (ZIKV); protozoários do gênero Trypanosoma; identificou os achados histopatológicos em fígado e coração possivelmente, relacionados a esses dois agentes infecciosos e identificou as bactérias aeróbicas da cavidade oral e retal, realizando testes fenotípicos para detecção de bactérias multirresistentes. Para tal, foram capturados 37 calitriquídeos, por meio de armadilhas Tomahawk (TW) suspensas em plataformas dentro da área do CFMA. Três animais (8,1% n=37) apresentaram sorologia positiva para flavivírus (ELISA de bloqueio), indicando a circulação de agentes virais da família Flaviviridae nos calitriquídeos. Foi realizada a técnica de reação em cadeia pela polimerase em tempo real (RT-PCR) para ZIKV, sendo todas as amostras negativas. Devido a um período curto de viremia do ZIKV, a sua pesquisa no soro dos PNH pode apresentar resultados negativos, principalmente fora de um período de epidemia de ZIKV. Cinco animais (13,5% n=37) apresentaram hemocultura positivas para pesquisa de Trypanosoma sp. e pela técnica de nested PCR, dois calitriquídeos (5,4% n=37) foram identificados como Trypanosoma. minasense. As informações sobre o ciclo epidemiológico do T. minasense ainda são escassas. Todas as bactérias isoladas apresentam potencial patogênico, entretanto não foram detectadas caraterísticas de resistência antimicrobiana através dos testes realizados, betalactamase de espectro ampliado e pesquisa de enterobactérias resistentes a carbapenêmicos. O estudo visa colaborar para um melhor entendimento da interação entre esses agentes infecciosos, os seres humanos e os calitriquídeos, destacando-se dessa forma, a relevância desta pesquisa no âmbito da saúde pública e da biodiversidade.
Non-human primates (NHP) can be reservoirs for infectious agents with consequences to public health. The Fiocruz Mata Atlântica Campus (CFMA) is located at an expanding urban region which enables a closer contact of free-living animals, such as NHP with domestic animals and human beings. This study investigated the circulation of viral agents of the family Flaviviridae, specifically Zika virus (ZIKV); protozoa of the genus Trypanosoma; identified the histopathological findings in liver and heart possibly related to these two infectious agents and, identified the oral and rectal aerobic bacteria and performed phenotypic detections tests for multidrug-resistant bacteria population of callitrichids from CFMA. For that, thirty-seven callitrichids were captured through Tomahawk (TW) traps suspended on platforms in the geographic region of the CFMA. Three animals (8.1% n=37) presented positive serology for flavivirus (blocking ELISA), indicating the circulation of Flaviviridae viral agents among callitrichids. The real time polymerase chain reaction (RT-PCR) for ZIKV was made and all samples were negative. Due to a short period of ZIKV viremia, RT-PCR may have negative results, especially outside a period of ZIKV epidemic. Five animals (13.5% n=37) had positive haemoculture for Trypanosoma sp. and by nested PCR, two callitrichids (5.4% n =37) were identified as Trypanosoma minasense. The Information about the T. minasense epidemiological cycle is scanty. All the isolated bacteria have pathogenic potential, however there was not multidrug-resistant characteristics detected through the tests performed: extended spectrum -lactamase and carbapenem-resistant enterobacteriaceae research. This study aims to contribute for a better understanding of the interaction between these infectious agents, human beings and callitrichids, highlighting its relevance for the public health area and biodiversity as well.
