Background: A new pit viper, Protobothrops kelomohy, has been recently discovered in northern and northwestern Thailand. Envenoming by the other Protobothrops species across several Asian countries has been a serious health problem since their venom is highly hematotoxic. However, the management of P. kelomohy bites is required as no specific antivenom is available. This study aimed to investigate the biochemical properties and proteomes of P. kelomohy venom (PKV), including the cross-neutralization to its lethality with antivenoms available in Thailand. Methods: PKV was evaluated for its neutralizing capacity (ER50), lethality (LD50), procoagulant and hemorrhagic effects with three monovalent antivenoms (TAAV, DSAV, and CRAV) and one polyvalent (HPAV) hematotoxic antivenom. The enzymatic activities were examined in comparison with venoms of Trimeresurus albolabris (TAV), Daboia siamensis (DSV), Calloselasma rhodostoma (CRV). Molecular mass was separated on SDS-PAGE, then the specific proteins were determined by western blotting. The venom protein classification was analyzed using mass spectrometry-based proteomics. Results: Intravenous LD50 of PKV was 0.67 µg/g. ER50 of HPAV, DSAV and TAAV neutralize PKV at 1.02, 0.36 and 0.12 mg/mL, respectively. PKV exhibited procoagulant effect with a minimal coagulation dose of 12.5 ± 0.016 µg/mL and hemorrhagic effect with a minimal hemorrhagic dose of 1.20 ± 0.71 µg/mouse. HPAV was significantly effective in neutralizing procoagulant and hemorrhagic effects of PKV than those of TAAV, DSAV and CRAV. All enzymatic activities among four venoms exhibited significant differences. PKV proteome revealed eleven classes of putative snake venom proteins, predominantly metalloproteinase (40.85%), serine protease (29.93%), and phospholipase A2 (15.49%). Conclusions: Enzymatic activities of PKV are similarly related to other viperid venoms in this study by quantitatively hematotoxic properties. Three major venom toxins were responsible for coagulopathy in PKV envenomation. The antivenom HPAV was considered effective in neutralizing the lethality, procoagulant and hemorrhagic effects of PKV.(AU)

This article reviews the natural history and sociocultural importance of King Cobra, cases of snakebite envenomation caused by this species, current practice of management (preclinical and clinical), and major toxinological studies of the venom with a focus on venom proteomics, toxicity and neutralization. Unfortunately, epidemiological data of King Cobra bite is scarce, and venom proteomes reported in various studies revealed marked discrepancies in details. Challenges, such as inconsistency in snake venom sampling, varying methodology of proteomic analysis, lack of mechanistic and antivenomic studies, and controversy surrounding antivenom use in treating King Cobra envenomation are herein discussed. Future directions are proposed, including the effort to establish a standard, comprehensive Pan-Asian proteomic database of King Cobra venom, from which the venom variation can be determined. Research should be undertaken to characterize the toxin antigenicity, and to develop an antivenom with improved efficacy and wider geographical utility. Objetivou-se investigar a presença do Vírus da Estomatite Vesicular (VEV) e seus fatores de risco para ocorrência e disseminação da enfermidade em equídeos das mesorregiões Leste e Oeste Potiguar do estado do Rio Grande do Norte, Brasil. Foram analisadas pela técnica de virusneutralização, 809 amostras sanguíneas de equídeos provenientes de noventa propriedades de dezesseis municípios Potiguares durante os meses de julho de 2018 a fevereiro de 2019. Os fatores de riscos associados ao VEV foram avaliados por meio de questionário epidemiológico e os dados submetidos a análise estatística no programa IBM SPSS Statistics versão 21.0 com nível de confiança de 95%. Posteriormente, todas as variáveis estatisticamente significantes foram submetidas a análise de regressão de Poisson. A soroprevalência de anticorpos anti-VEV foi 24,6% (199/809), sendo 3,2% (13/402) de soropositivos na mesorregião Leste e 45,7% (186/407) na do Oeste Potiguar. Com relação aos sorotipos, observou-se uma prevalência de 3,8% (31/809) e 24,5% (198/809) para Indiana 2 e 3 respectivamente, com 15,1% (30/198) de coinfecção. Equídeos criados na mesorregião Oeste, em propriedades que não realizam quarentena e onde os animais enfermos são mantidos no rebanho, foram consideradas fatores predisponentes a infecção pelo VEV. Esses resultados demonstram a circulação do VEV em equídeos no Rio Grande do Norte, com destaque ao Oeste Potiguar, e sendo necessário a aplicação de medidas sanitárias que impeçam introdução e disseminação do vírus ente as espécies susceptíveis, principalmente em condições climáticas favoráveis para a sua manutenção, no ambiente de criação e pastagens.

The occurrence of anti-VSV antibodies was 24.6% (199/809) with 3.2% (13/402) and 45.7% (186/407) of seropositivity in the Western and Eastern mesoregion, respectively. Regarding serotypes, there were an occurrence of 3.8% (31/809) and 24.5% (198/809) for Indiana 2 and 3, respectively, and 15.1% (30/198) of co-infection for both. Equines kept of the Western mesoregion, on properties that do not quarantine, and where sick animals are kept in the herd, were considered risk factors for LVV infection. These results demonstrate the presence of VSV in equines in Rio Grande do Norte, with emphasis on Oeste Potiguar, and that sanitary measures must be adopted to prevent the introduction and viral spreading among susceptible species, especially due to favorable climatic conditions for the maintenance of VSV in the breeding and pasture environment.

The Eastern Russell's viper, Daboia siamensis, is a WHO Category 1 medically important venomous snake. It has a wide but disjunct distribution in Southeast Asia. The specific antivenom, D. siamensis Monovalent Antivenom (DsMAV-Thailand) is produced in Thailand but not available in Indonesia, where a heterologous trivalent antivenom, Serum Anti Bisa Ular (SABU), is used instead. This study aimed to investigate the geographical venom variation of D. siamensis from Thailand (Ds-Thailand) and Indonesia (Ds-Indonesia), and the immunorecognition of the venom proteins by antivenoms. Methods: The venom proteins were decomplexed with reverse-phase high-performance liquid chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis, followed by in-solution tryptic digestion, nano-liquid chromatography-tandem mass spectrometry and protein identification. The efficacies of DsMAV-Thailand and SABU in binding the various venom fractions were assessed using an enzyme-linked immunosorbent assay optimized for immunorecognition profiling. Results: The two most abundant protein families in Ds-Thailand venom are phospholipase A2 (PLA2) and Kunitz-type serine protease inhibitor (KSPI). Those abundant in Ds-Indonesia venom are PLA2 and serine protease. KSPI and vascular endothelial growth factor were detected in Ds-Thailand venom, whereas L-amino acid oxidase and disintegrin were present in Ds-Indonesia venom. Common proteins shared between the two included snaclecs, serine proteases, metalloproteinases, phosphodiesterases, 5'nucleotidases and nerve growth factors at varying abundances. DsMAV-Thailand exhibited strong immunorecognition of the major protein fractions in both venoms, but low immunoreactivity toward the low molecular weight proteins e.g. KSPI and disintegrins. On the other hand, SABU was virtually ineffective in binding all fractionated venom proteins. Conclusion: D. siamensis venoms from Thailand and Indonesia varied geographically in the protein subtypes and abundances. The venoms, nevertheless, shared conserved antigenicity that allowed effective immunorecognition by DsMAV-Thailand but not by SABU, consistent with the neutralization efficacy of the antivenoms. A specific, appropriate antivenom is needed in Indonesia to treat Russell's viper envenomation.(AU)

Equine alphaherpesvirus is a major pathogen that causes respiratory disorders, abortion, neurological syndromes, and perinatal mortality in horse populations worldwide. The aim of this study was to determine whether the occurrence of anti-alphaherpesvirus antibodies is correlated to reproductive abnormalities in horses in the state of Pernambuco, Brazil. The virus neutralization was carried out in 322 serological samples from unvaccinated horses from 42 farms located in 16 municipalities in the Microregion of the Vale do Ipojuca, Pernambuco, Brazil. Of these horses, 23.3% (75/322; 95% CI: 18.9-28.4%) were found to be reactive to anti-alphaherpesvirus antibodies. In addition, 61.9% (26/42) of farms had at least one reactive animal, with an inter-herd variation of 5% to 100%. With regard to reproductive rates, herpetic infection was associated with the use of biological reproductive techniques such as artificial insemination (AI) and embryo transfer (ET), which uses natural mating (p = 0.003); history of perinatal mortality (p = 0.004); birth interval greater than two years (p = 0.003); and reproductive seasonality of mares with irregular cycles in the herd (p = 0.041). This is the first report on the identification of horses reactive to anti-alphaherpesvirus antibody in the state of Pernambuco, Brazil. From the results obtained, the adoption of prophylactic measures such as vaccination is suggested in order to prevent infection in susceptible animals and to reduce the viral load excreted by animals with quiescent infection.
O alfaherpesvírus equino é um importante agente patogênico causador de distúrbios respiratórios, aborto, síndrome neurológica e mortalidade perinatal em populações de equinos em todo o mundo. O objetivo deste trabalho foi determinar a ocorrência de anticorpos anti-alfaherpesvírus e relacionar às alterações reprodutivas em equinos no estado de Pernambuco, Brasil. A vírus neutralização foi realizada em 322 amostras sorológicas de equinos não vacinados, provenientes de 42 propriedades de 16 municípios pertencentes à Microrregião do Vale do Ipojuca, Pernambuco, Brasil. Observou-se uma ocorrência de 23,3% (75/322; IC 95%:18,9-28,4%) de equinos reagentes à pesquisa de anticorpos anti-alfaherpesvírus. Além disso, 61,9% (26/42) das propriedades possuíam pelo menos um animal reagente, com variação inter-rebanho de 5 a 100%. Em relação aos índices reprodutivos, associou-se à infecção herpética a utilização de biotécnicas reprodutivas, como a inseminação artificial (IA) e a transferência de embriões (TE), com a monta natural (p = 0,003); histórico de mortalidade perinatal (p = 0,004); intervalo entre partos superior a dois anos (p = 0,003); e sazonalidade reprodutiva de éguas com ciclos irregulares no rebanho (p = 0,041). Este é o primeiro relato de detecção de equinos reagentes à pesquisa de anticorpos anti-alfaherpesvírus no estado de Pernambuco, Brasil. A partir dos resultados obtidos, sugere-se a adoção de medidas profiláticas como a vacinação, a fim de prevenir a infecção nos animais suscetíveis e reduzir a carga viral excretada nos animais com infecção quiescente.

Equine alphaherpesvirus is a major pathogen that causes respiratory disorders, abortion, neurological syndromes, and perinatal mortality in horse populations worldwide. The aim of this study was to determine whether the occurrence of anti-alphaherpesvirus antibodies is correlated to reproductive abnormalities in horses in the state of Pernambuco, Brazil. The virus neutralization was carried out in 322 serological samples from unvaccinated horses from 42 farms located in 16 municipalities in the Microregion of the Vale do Ipojuca, Pernambuco, Brazil. Of these horses, 23.3% (75/322; 95% CI: 18.9-28.4%) were found to be reactive to anti-alphaherpesvirus antibodies. In addition, 61.9% (26/42) of farms had at least one reactive animal, with an inter-herd variation of 5% to 100%. With regard to reproductive rates, herpetic infection was associated with the use of biological reproductive techniques such as artificial insemination (AI) and embryo transfer (ET), which uses natural mating (p = 0.003); history of perinatal mortality (p = 0.004); birth interval greater than two years (p = 0.003); and reproductive seasonality of mares with irregular cycles in the herd (p = 0.041). This is the first report on the identification of horses reactive to anti-alphaherpesvirus antibody in the state of Pernambuco, Brazil. From the results obtained, the adoption of prophylactic measures such as vaccination is suggested in order to prevent infection in susceptible animals and to reduce the viral load excreted by animals with quiescent infection.(AU)
O alfaherpesvírus equino é um importante agente patogênico causador de distúrbios respiratórios, aborto, síndrome neurológica e mortalidade perinatal em populações de equinos em todo o mundo. O objetivo deste trabalho foi determinar a ocorrência de anticorpos anti-alfaherpesvírus e relacionar às alterações reprodutivas em equinos no estado de Pernambuco, Brasil. A vírus neutralização foi realizada em 322 amostras sorológicas de equinos não vacinados, provenientes de 42 propriedades de 16 municípios pertencentes à Microrregião do Vale do Ipojuca, Pernambuco, Brasil. Observou-se uma ocorrência de 23,3% (75/322; IC 95%:18,9-28,4%) de equinos reagentes à pesquisa de anticorpos anti-alfaherpesvírus. Além disso, 61,9% (26/42) das propriedades possuíam pelo menos um animal reagente, com variação inter-rebanho de 5 a 100%. Em relação aos índices reprodutivos, associou-se à infecção herpética a utilização de biotécnicas reprodutivas, como a inseminação artificial (IA) e a transferência de embriões (TE), com a monta natural (p = 0,003); histórico de mortalidade perinatal (p = 0,004); intervalo entre partos superior a dois anos (p = 0,003); e sazonalidade reprodutiva de éguas com ciclos irregulares no rebanho (p = 0,041). Este é o primeiro relato de detecção de equinos reagentes à pesquisa de anticorpos anti-alfaherpesvírus no estado de Pernambuco, Brasil. Background: Equine Herpesvirus type 1 (EHV-1) and Equine Arteritis Virus (EAV) have a high capacity for dissemination, causing losses in horse breeding due to reproductive, respiratory and neurological problems. In the State of Rio Grande do Norte there is only one survey, in the municipality of Mossoró, about these diseases. The purpose of this study was therefore to determine the prevalence and assess the risk factors associated with EHV-1 and EAV infections in unvaccinated horses raised in the mesoregions of East and West Potiguar in the State of Rio Grande do Norte, Brazil. Materials, Methods & Results: Between August 2018 and February 2019, blood samples were collected from equines on farms located in sixteen municipalities in the mesoregions of East and West Potiguar for the serological diagnosis of EAV (809 samples from 90 farms) and of EHV (778 samples from 88 farms), being that, only municipalities that had at least five hundred equines were included in the study. An investigative epidemiological questionnaire was applied to each property visited, containing questions related to the breeder, property, animal and the sanitary, reproductive and nutritional management. A logistic regression was performed to assess the risk factors associated with EVH, considering the serological test (positive or negative) as the dependent variable. EAV and EVH antibodies were detected through the serum virus neutralization assay. No evidence was found of EAV [0% (0/809)], but EHV showed a prevalence rate of 32% (249/778), with 80.6% (71/88) of the farms having at least one animal that tested positive, and EHV was present in all the municipalities covered in this study. The prevalence of seropositivity for BoHV-1 was 61.1%, ranging from 43.3 to 86.2% among the analyzed properties. The variable absence of veterinary assistance showed statistically significant association (odds ratio OR = 2.10; p < 0.001) with alphaherpesvirus infection. Este estudo teve como objetivo avaliar variáveis de manejo que possam constituir fatores de risco da infecção por BoHV-1 e/ou por BVDV em rebanhos leiteiros com histórico de problemas da reprodução em vacas mestiças em manejo extensivo e sem histórico de vacinação prévia para o controle de IBR e BVD. Anticorpos neutralizantes anti-BoHV-1, anti-BVDV e para ambos os vírus simultaneamente foram identificados em 62,5% (165/264), 45,1% (119/264) e 31,4% (83/264), respectivamente, das amostras analisadas. Os fatores de risco associados à infecção por BoHV-1 foram rebanhos com número total de fêmeas superior a 100, presença de ordenha mecânica, não utilização de inseminação artificial na reprodução e a compra infrequente de animais. Para BVDV, os fatores de risco foram aptidão mista (leite/corte) do rebanho, presença de ordenha mecânica, ausência de quarentena para os animais recém-adquiridos, presença de piquete de parição e a não utilização de inseminação artificial. Para a infecção simultânea (BoHV-1/BVDV), a presença de ordenha mecânica aumentou o risco em 3,36 vezes, e o uso de inseminação artificial reduziu em 56% o risco de infecção nos rebanhos avaliados.(AU)

The aim of this study was to verify managerial practices that may constitute risk factors associated with infection by BoHV-1 and/or BVDV in dairy herds with a history of reproductive disease in extensively reared dairy cows without a previous history of vaccination against IBR and BVD. Neutralizing antibodies anti-BoHV-1, anti-BVDV or both were detected in 62.5% (165/264), 45.1% (119/264), and 31.4% (83/264), respectively, in the samples analyzed. The risk factors associated with infection by BoHV-1 were herds with more than 100 cows, the presence of mechanical milking, the non-utilization of artificial insemination, and the infrequent acquisition of animals. Risk factors associated with BVDV were dual-purpose herds (milk and beef), these include the utilization of mechanical milking, absence of quarantine for newly acquired animals, the presence of picket calving, and the absence of artificial insemination. For simultaneous infections by both viruses (BoHV-1 and BVDV) the use of mechanical milking increased the chance of infection 3.36-fold while the use of artificial insemination reduced the risk of infection by 56% in these herds.(AU)

Livestock is one of the main activities of the gross domestic Product (GDP), exercising a fundamental role in the land use and occupation process in Brazil. Infectious diseases, bacterial, viral or parasitic origin, may affect the reproductive system of both males and females, causing a series of problems of infertility with different levels of occurrence. One of the most important diseases that affect the reproductive sphere of viral origin and that have already been identified in cattle in Brazil the infectious bovine rhinotracheitis (IBR). The aim of this work was to carry out an epidemiological study in the State of Goiás, specifically in the Vale do Rio dos Bois microregion (municipalities of Cezarina, Mairipotaba and Pontalina), detecting the presence of antibodies to the IBR, by vírusneutralização (VN). The properties studied were found a seroprevalence of 96.4% for neutralizing antibodies against IBR in bovine females over 24 months. The results found in this study showed that there is a high prevalence of infection for BoHV-1 at Vale do Rio dos Bois microregion. The data for these studies can serve as a base to plot strategies for the control of the disease, with the guidance of producers.

Doenças infecciosas de origem bacteriana, viral ou parasitária podem afetar o sistema reprodutivo, tanto dos machos como das fêmeas, causando uma série de problemas de infertilidade nos rebanhos, com diferentes níveis de gravidade. Dentre as doenças mais importantes de origem viral que afetam a natureza reprodutiva e que já foram identificadas em bovinos no Brasil, destaca-se a Rinotraqueíte Infecciosa Bovina (IBR). O objetivo deste trabalho foi realizar um estudo epidemiológico no estado de Goiás, especificamente na Microrregião do Vale do Rio dos Bois (municípios de Cezarina, Mairipotaba e Pontalina), detectando a presença de anticorpos contra a IBR pelo teste de vírus-neutralização (VN). Das propriedades estudadas, foi encontrada uma soroprevalência de 96,4% para anticorpos neutralizantes contra a IBR em fêmeas bovinas acima de 24 meses. Os resultados encontrados no presente estudo demonstraram que há uma elevada taxa de prevalência da infecção por BoHV-1 na Microrregião do Vale do Rio dos Bois. Os dados deste estudo poderão servir de base para se traçarem estratégias para o controle da doença, com orientação aos produtores.

O botulismo é uma zoonose causada pelas neurotoxinas produzidas por Clostridium botulinum. Existem oito variedades de toxinas (A-H), as toxinas C e D são as principais responsáveis por prejuízos na bovinocultura, causando óbito. Dentre as medidas preventivas eficientes, destaca-se a vacinação dos animais. Uma metodologia padrão para medir a eficiência de uma vacina é a soroneutralização in vitro tendo camundongos como sistema indicador. O objetivo do trabalho foi verificar a interferência do uso de anestésicos em Cavia porcellus vacinados sobre o efeito protetor dos soros, quando realizada a coleta de sangue para realizar a soroneutralização frente às toxinas padrões C e D. Neste trabalho foram realizados procedimentos de vacinação subcutânea, na região laterodorsal de Cavia porcellus, na dose de 5 ml no primeiro dia e repetição após 21 dias, e sangria após mais 21 dias. Foram formados dois grupos de 10 animais para cada amostra vacinal, um para sangria com o uso de associação anestésica (cloridrato de xilazina 2%, 5mg/Kg/IM e cloridrato de quetamina 10%, 25mg/Kg/IM) e outro sem uso, após isso submeteu-se o pool de soro coletado de cada grupo à soroneutralização in vitro com toxinas padrões C e D. Esses procedimentos de validação possibilitarão o seu emprego visando o bem-estar animal nos testes de controle de qualidade de vacinas. Foi observado que a aplicação de anestésicos para a punção cardíaca não interferiu no nível de proteção dos soros dos animais frente ao desafio de toxinas C e D, comparado aos não anestesiados.
Botulism is a zoonosis caused by neurotoxins produced by Clostridium botulinum. There are eight varieties of toxins (A-H), toxins C and D are main responsible for illness in cattle, causing death. Among the effective preventive measures, the vaccination of the animals stands out. To ensure adequate protection of animals, vaccines must be efficient. A standard methodology for measuring the efficiency of a vaccine is in vitro serum neutralization having mice as the indicator system. The objective of the study was to verify the interference of anesthetic use in vaccinated guinea pigs on the protective effect of sera when blood collection was performed to perform serum neutralization against standard C and D toxins. In this work, subcutaneous vaccination procedures were performed in the later dorsal region of guinea pigs at the dose of 5 ml on the first day and repetition after 21 days and bleeding after another 21 days. Two groups of 10 animals were formed for each sample de vaccine, one for bleeding with the use of an anesthetic association (xylazine hydrochloride 2%, 5mg/Kg/IM and ketamine hydrochloride 10%, 25mg/Kg/IM) and the other with no use, after which the serum mixture collected from each group were submitted to in vitro serum neutralization with standard C and D toxins. This validation study will make it possible to introduce the use this procedures aimed at animal welfare in the quality control tests of vaccines. It was observed that the application of anesthetics to cardiac puncture did not interfere in the level of protection of the sera of the animals against the challenge of toxins C and D compared to those obtained of not anesthetized animals.
