The objective was to compare deoxynivalenol (DON) concentrations in feed ingredients and commercial swine diets measured by enzyme-linked immunosorbent assay (ELISA) and high-performance liquid chromatography (HPLC). Seventy feed ingredient samples consisted of corn, corn dried distillers grains with solubles, corn gluten feed, corn gluten meal, palm kernel expellers, rice bran, soy hulls, soybean meal, and wheat. Commercial swine diet samples (n = 92) were collected from 23 swine farms of varying regions in Korea and different growth stages of pigs. The DON concentration of all samples was determined in duplicate. Statistical comparisons were performed to compare the analytical methods (ELISA vs. HPLC), diet phases, and regions. The DON concentrations in most ingredients and all diets determined by ELISA method were greater than those determined by HPLC. The DON concentrations determined by the ELISA method were less than 1 mg/kg in all ingredients except corn dried distillers grains with solubles and corn gluten feed, and those determined by the HPLC were less than 0.5 mg/kg in all ingredients. The DON concentrations in complete diets did not vary by region or growth stages of pigs. The DON concentrations in most feed ingredients and commercial swine diets determined by ELISA method are greater than those determined by HPLC, but does not vary by regions in Republic of Korea or pig growth stages.

This study aimed to determine the efficacy of a finishing diet added with sugar beet pulp to reduce backfat skatole of entire male pigs, using the optimised high-performance liquid chromatographic (HPLC) method. The study comprised 72 males Pietrain (Large White × Landrace), divided into two groups of 36 animals each. Pigs in group A (treatment) were fed a supplemented formula (addition of 10 % beet pulp). while animals in group B (control) received a commercial feed, both for a period of 14 days before slaughter. The isocratic HPLC method achieved the chromatographic separation of indolic compounds in approximately 3 min. Skatole was significantly lower ( p = 0.002) in group A, showing that beet root supplementation reduced skatole levels in pig fat. In addition, the optimised HPLC method was reliable, less time-consuming, and showed a resolution suitable for small amounts of skatole.(AU)

ABSTRACT: Iron deficiency anemia (IDA) in humans is defined as the decrease of total hemoglobin concentration and the non-production of the adult hemoglobin subtype 2 - HbA2 (α2δ2 chains), which is considered a marker of IDA severity in humans, dosed together with the iron serum. This study aimed to determine the standard of hemoglobin types in piglets induced to experimentally IDA in the first 21 days of life (delivery to weaning). In the present study, 40 piglets born from four naïve gilts, were randomly and equally assigned among the gilts. On the third day after delivery, the groups were randomly distributed in different environments (cement and clay floors) and according to the iron supplementation (iron dextran and placebo). Erythrocyte parameters, serum iron, and hemoglobin trait were analyzed at four moments between birth and weaning days. The group of piglets that did not receive iron dextran supplementation on the third-day post-birth and were placed in the pen without soil did not present HbA2 from the seventh day onwards on the agarose electrophoretogram (pH 8.6) and this observation was correlated to decrease of serum iron (ρ: 0.156, p=0.003) when compared to the other groups' piglets that did not present iron deficiency. In the present study was possible to determine the swine hemoglobin pattern in IDA, since HbA2 was absent in piglets with IDA in comparison to the non-ferropenic groups and the correlation between the reduction of iron levels and the absence of HbA2.
RESUMO: A anemia por deficiência de ferro (ADF) em humanos é definida como a diminuição da concentração de hemoglobina total e a não produção da hemoglobina adulta subtipo 2 - HbA2 (cadeias α2δ2), que é considerada um marcador de gravidade de IDA em humanos, dosado em conjunto com o soro de ferro. Este estudo teve como objetivo determinar o padrão dos tipos de hemoglobina em leitões induzidos experimentalmente à IDA nos primeiros 21 dias de vida (parto ao desmame). Quarenta leitões, nascidos de quatro marrãs nulíparas, foram distribuídos aleatoriamente e igualmente entre as leitoas. No terceiro dia após o parto, os grupos foram distribuídos aleatoriamente em diferentes ambientes (piso de cimento e barro) e de acordo com a suplementação de ferro (ferro dextrano e placebo). Parâmetros eritrocitários, ferro sérico e traço de hemoglobina foram analisados em quatro momentos, entre o nascimento e o desmame. O grupo de leitões que não recebeu suplementação de ferro dextrano no terceiro dia pós-parto e foi colocado em baia sem solo não apresentou HbA2 a partir do sétimo dia no eletroforetograma de agarose (pH 8,6) e esta observação foi correlacionada com diminuição da concentração sérica ferro (ρ: 0,156, p=0,003) quando comparados aos demais grupos leitões que não apresentavam deficiência de ferro. No presente estudo foi possível determinar o padrão hemoglobinêmico suíno na IDA, uma vez que, a HbA2 estava ausente nos leitões com ADF em comparação aos grupos não ferropênicos e há correlação entre a redução dos níveis de ferro e a ausência de HbA2.

ABSTRACT: Isoimmune thrombocytopenic purpura (ITP) is an immune-mediated disease that causes severe hemorrhagic lesions and high mortality in piglets. The disease can occur early in newborn piglets (EITP) or late in 2- to 3-week old piglets (LITP). In this study, we analysed the clinical, pathological, and hematological aspects of 391 ITP cases (312 with EITP and 79 with LITP). In LIPT cases, morbidity and mortality rates were higher, with rates of 60% (morbidity) and 53% (mortality). The main clinicopathological findings in ITP cases were different patterns of hemorrhages organs and tissues. In EITP, clinical signs were characterized by extensive subcutaneous hemorrhages and death occurred within a few days; however, in LITP, often sudden death occurred. In macroscopic analysis, hemorrhagic diathesis was observed in all affected animals. In EITP, the most severe hemorrhagic lesions were integumentary, mainly in the dermis and epidermis. In LITP, visceral lesions were predominant, mainly in the epicardium and intestines. Microscopic bone marrow analysis revealed mild cellular hyperplasia in EITP and bone marrow aplasia in LITP. hematological analyses revealed leucopenia, thrombocytopenia, and anemia in all ITP-affected animals. However, fostering by a different sow was only efficient in controlling EITP and had little effect in LITP-symptomatic piglets, due to more severe lesions. Further studies on the etiopathogenesis of LITP are required to improve our understanding of this disease form.
RESUMO: Púrpura trombocitopênica isoimune (PTI) é uma doença imunomediada que causa lesões hemorrágicas graves e alta mortalidade em leitões, que pode se apresentar através de uma forma precoce em leitões neonatos (PTIP) ou uma forma tardia em leitões com duas a três semanas de idade (PTIT). Neste trabalho analisamos aspectos clínicos, hematológicos e histopatológicos de 391 casos de PTI, sendo 312 de PTIP e 79 de PTIT. Observou-se maiores morbidade (60%) e mortalidade (53%) na PTIT. Os principais achados clínico-patológicos observado na PTI são hemorragias em diferentes graus de intensidade e nos diferentes órgãos e tecidos. Na PTIP observou-se predominantemente hemorragias subcutâneas extensas e morte em alguns dias, já na PTIT, observou-se além de grave hemorragia, morte súbita. Na análise macroscópica, observou-se diátese hemorrágica em todos os animais afetados. Na PTIP as lesões hemorrágicas mais graves foram tegumentares, principalmente em derme e epiderme, enquanto, na forma tardia, observou-se lesões predominantemente viscerais, em epicárdico e intestino. A análise microscópica de medula óssea revelou discreta hiperplasia celular na forma PTIP, enquanto, na PTIT observou-se aplasia medular. Na análise hematológica observou-se leucopenia, trombocitopenia e anemia em todos os animais com PTI. Os achados clínicos, histopatológicos e hematológicos para PTIP e PTIT da doença permitiram o diagnóstico de PTI. Entretanto, a troca de mãe se mostrou eficiente apenas para controle PTIP, uma vez que, esta estratégia apresenta pouco resultado para leitões sintomáticos com a PTIT, devido lesões mais severas. Estudos sobre a etiopatogênese da PTIT ainda são necessários para melhor entendimento desta forma da doença.
Resumo
Mycoplasma hyopneumoniae is one of the most challenging respiratory pathogens involved with swine pneumonia worldwide, responsible for a chronic infection with high morbidity, which predisposes secondary bacterial infections in growing and finishing pigs. Advances in diagnostic techniques allowed identification of genetic characteristics associated with high antigenic and proteomic variability among bacterial strains. This study aimed to evaluate the genetic diversity of M. hyopneumoniae strains in lungs with pneumonic lesions obtained from 52 pig farms located in Minas Gerais, one of the largest swine production states in Brazil. Genotyping was performed using multilocus variable number of tandem repeat (VNTR) analysis (MLVA), targeting two loci encoding P97 and P146 adhesins VNTR. The results showed that this agent is widely disseminated in pig farms and there is a high polymorphism of M. hyopneumoniae variants circulating in the state of Minas Gerais. Different M. hyopneumoniae genotypes are randomly distributed in several regions of the state, with no specific geographic population structure pattern. M. hyopneumoniae association with viral agents was sporadic (3.17% with Influenza A and 1.9% with PCV2).
Mycoplasma hyopneumoniae é um dos patógenos respiratórios mais desafiadores envolvidos com pneumonia suína em todo o mundo. É responsável por uma infecção crônica de alta morbidade, que predispõe a infecções bacterianas secundárias nas fases de crescimento e terminação. Avanços nas técnicas diagnósticas permitiram a identificação de características genéticas do agente, associadas à alta variabilidade antigênica e proteômica entre cepas. Este estudo teve como objetivo examinar a ocorrência e diversidade genética de cepas de M. hyopneumoniae em pulmões com lesões pneumônicas em 52 granjas de suínos no estado de Minas Gerais, um dos maiores estados produtores de suínos do Brasil. A genotipagem foi realizada utilizando a técnica de "multilocus variable number of tandem repeat analysis" (VTNR/MLVA), usando dois loci que codificam VNTR das adesinas P97 e P146. Os resultados mostraram que esse agente está amplamente disseminado em granjas de suínos e que existe alto polimorfismo das variantes de M. hyopneumoniae circulando no estado de Minas Gerais. Diferentes genótipos de M. hyopneumoniae estão distribuídos aleatoriamente em várias regiões do estado, sem um padrão de estrutura populacional e geográfica específico. Associação de M. hyopneumoniae e agentes virais foi esporádica (3,17% com Influenza A e 1,9% com PCV2).

The objective of the present study was to investigate the influence of housing air temperature on the behavioural acts, physiological parameters and performance responses of fattening pigs. Animals were randomly allocated to 3 groups with 30 heads in each. During eight weeks of fattening, the animals of the two experimental groups were subjected to longterm technological temperature stress, provoked by a significant deviation of the microclimate parameters. The first control group of animals was kept following the Departmental Norms for Technological Design - Agro-Industrial Complex - 02.05 «Pig-breeding enterprises (complexes, farms, small farms)¼ at an air temperature of +17 +21 °C; pigs of the second experimental group were kept at a temperature of - +5 +8 °C, and the third experimental group - +28 +31 °C. During the experiment, the timing of behavioural acts and their index assessment, physiological parameters and productive characteristics of fattening pigs were studied. The experiment results showed that the thermoneutral zone for fattening pigs is + 17 + 21 °C. Animals 2nd experimental group showed less movement (P < 0.001), used huddling for decreasing body heat loss, more feed intake (P < 0.01), which increased its conversion, had reduced HR, RR, RT. Pigs of the 3rd experimental group showed increased movement (P < 0.001). This can be explained by their desire to find a cool place and rest lying on the side, consuming more water, having high HR, RR, and RT, showing signs of hyperthermia and were characterized by low-performance responses.

In pig farming, measurements of production parameters play a fundamental role in the success of the activity. Minimal differences in fertility between breeders can lead to less reproductive efficiency and, less productivity. However, assessing the fertility of each male and the early identification of subfertile males is a difficult task to be performed. Thus, the aim of this study was to evaluate the use of in vitro and in vivo parameters in the identification of subfertile males of the Landrace breed, aiming to collaborate with genetic improvement programs, routine optimization in the Genetic Diffusion Units (GDUs) and the results of performance. In experiment 1, an approach to identify males with subfertility was evaluated based on retrospective data. For this, the results (averages of birth rates, number of total births and average percentages of female and male piglets per litter) were evaluated for a total of 996 matings and 847 parturitions. The inseminations came from ejaculates of 32 males, who had at least 19 females inseminated with homospermic doses in the concentration of 2.5 x 109 total sperm from the same male. As for the birth rate (BR), an average of 85.47% ± 6.05 was observed with a group of median males, seven males that stood out and one individual (M32) with a performance of 58.06% ± 9.0. For the total number of piglets born (PB) the average was 13.41 ± 0.56, with three males with better performance and one (M32) with very poor performance (8.62 ± 0.59). In experiment 2, it was verified whether evaluations of inseminating doses (ID) of semen in vitro (motility and sperm morphology) after 96 hours of storage had correlations with fertility in vivo, which can be used to identify subfertile males. The evaluations were performed on 30 ejaculates regarding the means of BR and PB, considering only those who had at least 7 females inseminated. There were no correlations between the motility assessments and semen morphological changes and the reproductive parameters evaluated. The results obtained in vivo, referring to BR and PB, demonstrated that it was possible to identify differences between males, the individual (M32) had the worst results for the percentages of BR and PB. It is concluded that there are males of high and low fertility and that only the in vitro analyzes carried out in this study are not enough to categorize them, however, the evaluation of retrospective data was efficient for this purpose.(AU)
Na suinocultura moderna, as mensurações de parâmetros de produção têm papel fundamental para o sucesso da atividade. No entanto, a avaliação da fertilidade de cada macho e a identificação precoce de machos subférteis é uma tarefa difícil de ser realizada. O objetivo deste estudo foi avaliar a utilização de parâmetros in vivo e in vitro na identificação de machos subférteis da raça Landrace, visando colaborar com os programas de melhoramento genético, otimização da rotina nas Unidade de Difusão Genética (UDGs) e dos resultados a campo. No experimento 1, foi proposta uma abordagem de identificação dos machos subférteis tendo como base dados retrospectivos. Para isso, foram avaliados os resultados (médias das taxas de parto, número de nascidos totais e média das porcentagens de leitões fêmeas e machos por leitegada) de um total de 996 coberturas e 847 partos. As inseminações foram oriundas de ejaculados de 32 machos, que tiveram ao menos 19 fêmeas cobertas com doses homospérmicas na concentração de 2,5 x 109 de espermatozoides totais e obrigatoriamente do mesmo macho. Quanto a taxa de parto (TP) obtivemos uma média de 85,47% ± 6,05 e observou-se um grupo de machos medianos, sete machos que se destacaram positivamente e um indivíduo (M32) com um desempenho 58,06 ± 9,0. Para número de leitões nascidos totais (NT) obtivemos uma média de 13,41 ± 0,56 e notou-se três machos com melhor desempenho e um (M32) com péssimo desempenho (8,62 ± 0,59). No experimento 2, foi verificado se as avaliações das doses inseminantes (DI) de sêmen in vitro (motilidade e morfologia espermática) após 96 horas de armazenamento apresentaram correlação com a fertilidade in vivo. As avaliações foram realizadas em 30 ejaculados quanto às médias de TP e NT, considerando apenas ejaculados que tiveram ao menos 7 fêmeas inseminadas. Não foram verificadas correlações entre as avaliações de motilidade e alterações morfológicas do sêmen com os parâmetros produtivos avaliados. Os resultados obtidos in vivo, referentes a TP e NT, mostrou que foi possível identificar diferença entre os machos, onde o indivíduo (M32) apresentou os piores resultados para as porcentagens de TP e NT. Desta forma, pode-se concluir que existem machos de alta e baixa fertilidade e que somente as análises in vitro realizadas neste estudo não são suficientes para categorizá-los, no entanto, a avaliação de dados retrospectivos foi eficiente para esta finalidade.(AU)

The aim of this study was to assess the addition of intestinal alkaline phosphatase (IAP) to diets on the count of bacterial populations, pH of digestive organ contents, histopathological description, proinflammatory markers, hepatic glycogen reserve, and diarrhoea incidence of piglets challenged with Escherichia coli. Sixty-four crossbred piglets (7.16±0.28 kg body weight, 25-days-old) were assigned to four treatments in a completely randomised block design: negative control (NC), NC + antibiotic (ANT), NC + 15 mg IAP, or NC + 30 mg IAP kg−1 of diet, eight replications of two piglets per experimental unit. All piglets were orally challenged with 6 mL of a solution containing enterotoxigenic Escherichia coli K88 at 106 CFU mL−1 at 15 days of experimentation. The study lasted for 19 days. At the end of the experimental period, the piglets were slaughtered (six animals per treatment). Enterobacteriaceae in caecum and colon was lower in piglets on 30 mg IAP than with ANT and NC, ANT or 15 mg IAP, respectively. Enterobacteriaceae adhered to the mesenteric lymph nodes (MLN) was greater in piglets fed ANT than the other treatments. Lactic acid bacteria (LAB) count in caecum was greater in piglets fed NC and ANT. In MLN, LAB count was greater in ANT and 30 mg IAP-fed piglets compared with 15 mg IAP. Piglets in 30 mg IAP in diet showed a tendency for lowering tissue necrosis compared with NC or ANT. Piglets fed 30 mg IAP showed a reduction in diarrhoea incidence in the pre- and post-challenge compared with 15 mg IAP and all other treatments, respectively. Based on the criteria, addition of 30 mg IAP to diet inhibits Enterobacteriaceae population and suggests a potential effect in mitigating intestinal injuries, as observed in piglets in the NC for some of the parameters investigated.(AU)

Two experiments were carried out: in the first (Experiment I), a metabolism assay was carried out to estimate the net energy (NE) of experimental diets. Twelve crossbred barrows, averaging 85.74 ± 6.80 kg initial body weight, were distributed in a randomized block design with two metabolizable energy (ME) levels (3100 and 3500 kcal kg¹) with six replicates and one animal per experimental unit. In the second (Experiment II), 45 castrated male pigs were used, averaging 70.10 ± 1.26 kg of initial body weight distributed in a randomized block design, with five NE levels (2345, 2425, 2505, 2585, 2665 kcal kg¹), nine replicates and one animal per experimental unit. The increase in diet NE levels provided a linear reduction (p ≤ 0.00039) in average daily feed intake (ADFI). There was a quadratic effect on NE efficiency (p ≤ 0.0027), average daily gain (ADG), (p ≤ 0.0352) and the feed:gain ratio (F:G), (p ≤ 0.0024), the optimal levels being estimated at 2485, 2493 and 2533 kcal kg¹, respectively. Drip loss (DL) decreased (p ≤ 0.0001) as NE levels increased. There was also a linear decrease (p ≤ 0.0462) in the Minolta color parameter (+a*), due to the NE levels. Plasma parameters were not affected (p > 0.05) by NE levels. The dietary net energy levels affected the performance and meat quality of finishing pigs and the level of 2493 kcal kg¹ provided the best ADG.

Helminths of the genus Oesophagostomum cause enteric diseases and affect domestic animals such as pigs. The aim of this study was to explore the species composition and genetic diversity of Oesophagostomum spp. infecting pigs in close contact with humans in the state of Piauí, Brazil. Eighty-seven fecal samples were collected for parasitological tests and molecular analysis. Through microscopy, the overall positivity rate for strongyliform eggs was 81.6% among the pigs studied. Forty-two strongyliform egg samples were subjected to PCR and six cox1 sequences (637 bp) were identified for the genus Oesophagostomum. The sequences were identified as Oesophagostomum dentatum, O. quadrispinulatum and O. columbianum. In the phylogenetic tree and haplotype network, 89 sequences were separated into seven clusters, which also included reference sequences from GenBank. Oesophagostomum dentatum and O. quadrispinulatum were seen to be closely related species and formed a monophyletic group related to O. aculeatum. Oesophagostomum columbianum showed similarity with sequences from parasites infecting small ruminants and the clade was positioned closer to O. bifurcum. High interspecific diversity was found and intraspecific diversity varied according to the species. This was the first study to characterize Oesophagostomum DNA sequences obtained from pigs in Brazil.(AU)
Parasitos do gênero Oesophagostomum causam doenças entéricas e podem afetar a criação de animais, como os suínos. O objetivo deste estudo foi identificar as espécies e explorar a diversidade genética de Oesophagostomum spp. infectando suínos em contato próximo com humanos, no estado do Piauí, Brasil. Oitenta e sete amostras fecais foram coletadas para testes parasitológicos, análise morfométrica dos ovos e análises moleculares. A taxa geral de positividade para ovos estrongiliformes foi de 81,6%. Quarenta e duas amostras de ovos estrongiliformes foram submetidas à PCR e seis sequências cox1 (637 bp) foram identificadas para o gênero Oesophagostomum. As sequências foram identificadas como Oesophagostomum dentatum, O. quadrispinulatum e O. columbianum. Na árvore filogenética e na rede haplotípica, 89 sequências foram separadas em sete clusters, incluindo sequências de referência do GenBank. Oesophagostomum dentatum e O. quadrispinulatum são espécies estreitamente relacionadas e formaram um grupo monofilético com O. aculeatum. Oesophagostomum columbianum apresentou semelhança com sequências de parasitas obtidos de pequenos ruminantes e o clado foi posicionado mais próximo de O. bifurcum. Alta diversidade interespecífica foi encontrada e a diversidade intraespecífica variou de acordo com as espécies. Esse foi o primeiro estudo a caracterizar sequências de DNA de Oesophagostomum isoladas de suínos no Brasil.(AU)

ABSTRACT The present study was carried out to evaluate the effect of net energy levels in diets for barrows, from 70 to 100 kg, on performance, plasma parameters and carcass characteristics. Two experiments were carried out: in the first (Experiment I), a metabolism assay was carried out to estimate the net energy (NE) of experimental diets. Twelve crossbred barrows, averaging 85.74 ± 6.80 kg initial body weight, were distributed in a randomized block design with two metabolizable energy (ME) levels (3100 and 3500 kcal kg1) with six replicates and one animal per experimental unit. In the second (Experiment II), 45 castrated male pigs were used, averaging 70.10 ± 1.26 kg of initial body weight distributed in a randomized block design, with five NE levels (2345, 2425, 2505, 2585, 2665 kcal kg1), nine replicates and one animal per experimental unit. The increase in diet NE levels provided a linear reduction (p 0.00039) in average daily feed intake (ADFI). There was a quadratic effect on NE efficiency (p 0.0027), average daily gain (ADG), (p 0.0352) and the feed:gain ratio (F:G), (p 0.0024), the optimal levels being estimated at 2485, 2493 and 2533 kcal kg1, respectively. Drip loss (DL) decreased (p 0.0001) as NE levels increased. There was also a linear decrease (p 0.0462) in the Minolta color parameter (+a*), due to the NE levels. Plasma parameters were not affected (p > 0.05) by NE levels. The dietary net energy levels affected the performance and meat quality of finishing pigs and the level of 2493 kcal kg1 provided the best ADG.

The aim of this study was to access the efficacy of four disinfectants to inactivate influenza A [H1N1] 0 hour and 72 hours after disinfectant dilution. A pandemic H1N1 influenza virus isolated from a pig with respiratory disease was used to obtain inoculums containing 6.4log10 EID50/mL; 5.4log10 EID50/mL; 4.4log10 EID50/mL and 3.4log10 EID50/mL. Suspension test was composed of 400µL of viral inoculum, 100µL of organic load and 500µL of each individually diluted disinfectant and incubated for ten minutes of contact time. After a neutralizing step, each mixture was filtered on a 0.22µm membrane and 0.2mL was inoculated in six 9-day-old embryo chicken egg through allantoic route. The allantoic fluid from eggs was harvest for RT-PCR and hemagglutination test. The experiment was repeated 72 hours after disinfectant dilution. On the first assessment with fresh disinfectant, influenza virus was inactivated by oxidizing compost disinfectant and phenolic disinfectant in all virus concentrations, the quaternary ammonium compound (QAC) and glutaraldehyde association inactivated the virus up to a concentration of 5.4log10 EID50/mL. QAC disinfectant did not eliminate virus viability. Seventy-two hours after disinfectants were diluted, oxidizing compost disinfectant and QAC and glutaraldehyde association disinfectant demonstrated the same result as the evaluation with fresh disinfectant solution. Phenolic disinfectant inactivated viral inoculum up to a concentration of 5.4log10 EID50/mL. QAC had no effect on inactivating 3.4log10 EID50/ mL of influenza virus. In conclusion, three of the four disinfectants tested were effective to inactivate pandemic H1N1 influenza virus in the presence of organic load. Test result performed 72hours after disinfectant dilution suggest a decrease in the effectiveness of one disinfectant.
O objetivo deste trabalho foi avaliar a eficácia de quatro desinfetantes em inativar o vírus da influenza A [H1N1] 0-hora e 72-horas após a diluição dos produtos. Um vírus H1N1 pandêmico isolado previamente de um suíno com doença respiratória foi utilizado e foram obtidas quatro concentrações de inóculo contendo 6,4log10 EID50/mL; 5,4log10 EID50/mL; 4,4log10 EID50/mL and 3,4log10 EID50/mL. Para compor o teste em suspensão foram adicionados 400µL de inóculo viral, 100µL de matéria orgânica e 500µL de cada desinfetante diluído individualmente e a mesma foi incubada por 10 minutos. Após a etapa neutralizante, a suspensão foi filtrada em membrana 0,22µm e 0,2mL foi inoculado em seis ovos de galinha embrionados de nove dias de incubação, via rota alantóide. O fluido alantóide foi colhido após 72 horas para testes de hemaglutinação e RTPCR. O mesmo protocolo experimental foi repetido usando as soluções desinfetantes 72 horas após a diluição. O vírus da influenza foi inativado pelo composto oxidante e também pelo desinfetante fenólico em todas as concentrações virais testadas 0-hora após diluição. O desinfetante com associação de amônia quaternária e glutaraldeído inativou o vírus na concentração de até 5,4log10 EID50/mL. O desinfetante à base de amônia quaternária não inativou o vírus. Os resultados 72-horas após a diluição não diferiram quando comparado com 0-hora, exceto o desinfetante fenólico, o qual inativou o vírus da influenza somente até a concentração 5,4log10 EID50/ mL. Concluindo, três dos quatro desinfetantes testados foram efetivos ao inativar o vírus da influenza [H1N1] pandêmico na presença de matéria orgânica. Os resultados do teste com produtos diluídos após 72 horas sugerem redução da efetividade em, pelo menos, um desinfetante.

Streptococcus suis is a Gram-positive pathogen that inhabits the upper respiratory tract and can cause severe systemic inflammatory disease in pigs, mainly during the nursery phase. Streptococcus suis is a reemergent pathogen, and outbreaks of its inducing disease represent significant economic losses for the pig industry worldwide. In this study, we described the clinical, pathological, and molecular aspects of an outbreak of S. suis infection with atypically high mortality. The outbreak occurred in nursery farms integrated into a cooperative in the state of Paraná, Brazil. Of the 30 nurseries, 10 were severely affected by the pathogen and had high economic losses. Clinical signs usually started approximately 10 days after weaning and were mainly characterized by acute nervous and locomotor disorders. The mortality of the affected batches usually ranged between 8% and 10%, but in some cases, it reached 18%. Nine piglets were submitted to post mortem examination. Macroscopically, the synovial joints were enlarged and contained fibrinous exudates. In the central nervous system, there was hyperemia of the leptomeningeal vessels associated with deposition of fibrin and purulent exudate in the leptomeninges. In three piglets, there was thickening of the choroid plexus associated with dilation of the lateral ventricles. Microscopic lesions were characterized mainly by fibrinosuppurative inflammation, which involved the synovial membranes, leptomeninges of the brain, and spinal cord. Furthermore, it also affects the choroid plexus, ependyma, nerve roots, and central canal of the spinal cord. S. suis was isolated from the cerebrospinal fluid, meningeal swabs, and/or synovial fluid of 8/9 piglets, and typified as serotype 9 by multiplex PCR.
Streptococcus suis é um patógeno Gram positivo que habita o trato respiratório superior e pode causar doença inflamatória sistêmica grave em suínos, principalmente durante a fase de creche. Streptococcus suis é um patógeno reemergente e surtos representam perdas econômicas significativas a suinocultura mundial. Neste estudo descrevemos os aspectos clínicos, patológicos e moleculares de um surto de infecção por S. suis com mortalidade atipicamente alta. O surto ocorreu em creches integradas a uma cooperativa do estado do Paraná, Brasil. Das 30 creches, 10 foram severamente afetadas pelo patógeno e apresentavam elevadas perdas econômicas. Os sinais clínicos iniciavam em torno de 10 dias após o desmame e eram caracterizados principalmente por sinais clínicos nervosos e locomotores agudos. A mortalidade dos lotes afetados variava entre 8% e 10%, mas em alguns casos ultrapassava 18%. Nove leitões foram submetidos ao exame post mortem. Macroscopicamente, as articulações sinoviais estavam aumentadas e continham exsudato fibrinoso. No sistema nervoso central havia hiperemia dos vasos leptomeníngeos associada a deposição de fibrina e exsudato purulento nas leptomeninges. Em três leitões havia espessamento do plexo coroide associado a dilatação dos ventrículos laterais. As lesões microscópicas eram caracterizadas principalmente por inflamação fibrinossupurativa que envolvia as membranas sinoviais, as leptomeninges do cérebro e medula espinhal. Além disso, também afetava o plexo coroide, epêndima, raízes nervosas e canal central da medula espinhal. S. suis foi isolado do líquido cefalorraquidiano, suabe de meninge e/ou líquido sinovial de 8/9 leitões e tipificado como sorotipo 9 por PCR multiplex.

Abstract A serological, molecular and histopathological study was carried out in order to investigate occurrences of Toxoplasma gondii in pigs slaughtered with and without inspection service. Serum samples were collected from 60 pigs to detect anti-T. gondii antibody by indirect fluorescent antibody (IFAT). Tongue, masseter and diaphragm fragments were also collected for parasite DNA detection by means of the polymerase chain reaction (PCR) and histopathological analysis. The serological results showed that 77% (44/60) of the pigs were positive. Regarding PCR, 66.67% (40/60) were positive for T. gondii. Among the tissues evaluated, the diaphragm was the one with the highest frequency of positivity (40%; 24/60), followed by the masseter (38.33%; 23/60) and tongue (33.3%; 20/60). Histopathological changes were only observed in the diaphragm, which presented inflammatory infiltrates of lymphohistiocytic and neutrophilic types. These results not only show the potential threat of T. gondii to human health, but also demonstrate the dynamic epidemiological situation of toxoplasmosis in pigs in the city of São Luís, providing support for food security regarding pigs and for T. gondii control programs in Brazil.
Resumo Realizou-se um estudo sorológico, molecular e histopatológico com o objetivo de verificar a ocorrência de Toxoplasma gondii em suínos abatidos com e sem serviço de inspeção. Foram coletados soros de 60 suínos para a pesquisa de anticorpos anti-T. gondii pela reação de imunofluorescência indireta (RIFI). Também foram coletados fragmentos de língua, masseter e diafragma para a detecção do DNA do parasito por meio da reação em cadeia da polimerase (PCR) e análise histopatológica. A análise sorológica demonstrou que 77% (44/60) dos suínos apresentaram anticorpos anti-T. gondii. Com relação ao PCR, 66,67% (40/60) foram positivos para T. gondii. Dentre os tecidos avaliados, o diafragma foi o que obteve maior frequência de positividade (40%; 24/60), seguidos de masseter (38,33%; 23/60) e língua (33,3%; 20/60). Alterações histopatológicas foram observadas apenas no diafragma, que apresentou infiltrado inflamatório do tipo linfohistiocitário e neutrofílico. Esses resultados não evidenciam apenas a ameaça potencial de T. gondii à saúde humana, mas também demonstram a dinâmica situação epidemiológica da toxoplasmose em suínos na região da cidade de São Luís, fornecendo suporte para a segurança alimentar de suínos e programas de controle de T. gondii no país.

The objective of this study was to evaluate the cholecalciferol and 1,25-dihydroxycholecalciferol glycoside supplementation in piglet diet on organ biometry and physicochemical composition, pH of the digestive tract contents, and bone traits. A total of 128 entire male piglets (21 days-old, 6.82±0.38 kg body weight) were distributed in a completely randomized block design with eight replications and four animals per experimental unit. The treatments were composed of a diet 100/0 = 100% of the vitamin D supplemented with cholecalciferol; 50/50 = 50% supplemented with cholecalciferol + 0.25 µg of 1,25(OH)2 D3 glycoside; 25/75 = 25% supplemented with cholecalciferol + 0.375 µg of 1,25(OH)2 D3 glycoside; and 0/100 = 0.50 µg of 1,25(OH)2 D3 glycoside. The results indicated that piglets fed 100/0 showed an increase in spleen weight compared with those fed 0/100 and greater heart dry matter than piglets fed the 50/50 diet. Phosphorus concentration in the spleen was higher in piglets that received 25/75 when compared with those that received the 100/0 and 0/100 diets. Piglets that received the 0/100 diet showed higher pH of the stomach contents, but showed reduced pH of the contents of the jejunum and ileum. The width of the epiphysis and diaphysis was greater when piglets consumed the 50/50 treatment; however, a lower epiphysis height was observed. The use of 1,25(OH)2 D3 glycoside alone in piglet diets does not negatively influence the pH of the gastrointestinal tract and organ physicochemical composition. In addition, the use of diets with 1,25(OH)2 D3 glycoside as a replacement for cholecalciferol reduces spleen weight and promotes improvements in bone development.

Mycobacterium tuberculosis var. bovis is the etiologic agent of animal tuberculosis (aTB), a neglected zoonotic disease. Animal tuberculosis can affect many species, including swine. aTB-consistent granulomas in these animals lead to carcass disposal, generating economic losses and posing risks to human health. In the present study, an aTB outbreak was identified at an intensive swine farming operation in Southern Brazil. Inspection during swine slaughter revealed aTB-suspected lesions, which were collected for diagnosis by histology, PCR, and bacterial isolation. The animals had no clinical signs of tuberculosis. Granulomatous lesions were identified in 0.73% (59/8,071) of the slaughtered swine, and were confirmed by histology. Nine samples were further examined by PCR and bacterial isolation, with 44.4% and 55.5% positive results, respectively. Data from abattoirs subjected to federal surveillance show an aTB prevalence in Brazil of <0.001%. The present data thus indicate a swine aTB outbreak in intensive breeding. Swine infection can be related to exposure to infected animals or to contaminated food or environment. Biosecurity measures must be taken to avoid aTB transmission. Although certified swine breeding farms adopt such measures, this report indicates that constant monitoring is crucial, and greater control in swine breeding and finishing units is required to prevent outbreaks and spread of tuberculosis.

Nove amostras foram posteriormente examinadas por PCR e isolamento bacteriano, com 44,4% e 55,5% de resultados positivos, respectivamente. Dados de frigoríficos submetidos à vigilância federal demostram prevalência da aTB de < 0,001% no Brasil. Os dados presentes indicam, portanto, um surto de aTB em suínos de criação intensiva. A infecção em suínos pode estar relacionada à exposição a animais infectados, alimentos ou ambientes contaminados. Medidas de biossegurança devem ser tomadas para evitar a transmissão da aTB. Embora as granjas suinícolas certificadas adotem tais medidas, este relato indica que o monitoramento constante é crucial, e maior controle nas unidades de criação e terminação de suínos é necessário para prevenir surtos e disseminação da tuberculose.

This study evaluated the effect of the three inulin levels (0.1%, 0.2%, 0.4%) supplemented as a substitute for an antibiotic growth promoter (AGP, zinc bacitracin) and control in guinea pigs raised for human consumption. Fifty 14-day-old male guinea pigs were used. Productive parameters (weight gain, total dry matter intake, and feed conversion ratio (FCR)) and intestinal morphology of the duodenum, jejunum, and ileum at slaughter (70 days of age) were evaluated. An inverse relationship was observed between inulin levels and FCR (linear effect; P = 0.006). There was no statistically significant effect of the treatments on total dry matter intake and weight gain (P > 0.05). A linear effect of the inulin level on the villis length (VL), villis width (VW), and length/depth ratio (VL/DC) in the duodenum; VW in the jejunum; and VL in the ileum (P <0 .05) was reported. In conclusion, a linear effect of the increasing doses of inulin was found on the FCR and the morphological parameters of the duodenums integrity, and no differences in the effects of the inulin added to the diet and the treatment with AGP were found.
O objetivo do estudo foi avaliar o efeito da suplementação na dieta de cobaias com inulina, em níveis crescentes (0,1%, 0,2%, 0,4%) como substituto para um antibiótico promotor de crescimento (AGP, bacitracina de zinco) além do grupo controle (dieta padrão). Foram utilizados 50 porquinhos-da-índia machos com 14 dias de idade. Os parâmetros produtivos foram avaliados do desmame aos 70 dias de idade e os parâmetros morfológicos intestinais foram avaliados no duodeno, jejuno e íleo no momento do abate. Foi encontrado um efeito linear do nível de inulina sobre na taxa de conversão alimentar (FCR; P = 0,006), indicando que em níveis mais elevados de inulina o FCR diminui. Não houve diferença significativa entre os grupos quando avaliado o efeito dos diferentes tratamentos sobre o consumo de ração e ganho de peso corporal (P > 0,05). Um efeito linear do nível de inulina foi encontrado no comprimento das vilosidades (VL), na largura das vilosidades (VW) e na relação comprimento / profundidade (VL/DC) no duodeno, sobre a VW no jejuno; e no VL no íleo (P < 0,05). Em conclusão, um efeito linear do aumento do nível de inulina foi encontrado na taxa de conversão alimentar e nos parâmetros morfológicos da integridade do duodeno, além disso, não houve diferença entre a adição de inulina na dieta e o tratamento com um antibiótico promotor de crescimento.
