Resumo

Purpose: The aim of this study was to determine the protective and antioxidative effects of intensive exercise on streptozotocin (STZ)-induced testicular damage, apoptotic spermatognial cells death, and oxidative stress. Methods: 36 male Sprague Dawley rats were divided into three groups: control, diabetes, and diabetes+intensive exercise (IE) groups. Testicular tissues were examined histopathologically and antioxidant enzymes, including catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and malondialdehyde (MDA) activity, as well as serum testosterone level, were measured. Results: Seminiferous tubules and germ cells were found to be better in the testis tissue of the intense exercise group than in the diabetes group. Diabetes suppressed antioxidant enzymes CAT, SOD, GPx and testosterone levels were significantly decreased, and increased MDA level in the diabetic group compared to diabetes+IE group (p < 0.001). Following four weeks of treatment, intensive exercise improved the antioxidant defense, significantly decreased MDA activity, and increased testosterone levels in testicular tissue in the diabetic group compared to diabetes+IE group (p < 0.01). Conclusion: STZ-induced diabetes causes damage to the testis tissue. In order to prevent these damages, exercise practice has become very popular nowadays. In present study, our intensive exercise protocol, histological, and biochemical analysis of the effect of diabetes on the testicular tissues is shown.

Resumo

Thai Mucuna pruriens (L.) DC. var pruriens (T-MP) seed containing levodopa (L-DOPA) and antioxidant capacity has been shown to improve sexual behavior and male reproductive parameters in rats treated with ethanol (Eth). However, its protective effect on testicular apoptotic germ cells has never been reported. This study aimed to investigate the potential effects of T-MP seed extract on expressions of caspase, proliferating cell nuclear antigen (PCNA), and dopamine D2 receptor (D2R) proteins in Eth rats. Thirty-six male Wistar rats were divided into four groups (9 animals/group), including control, Eth, T-MP150+Eth, and T-MP300+Eth, respectively. Control rats received distilled water, and Eth rats received Eth (3g/kg BW; 40%v/v). The T-MP groups were treated with T-MP seed extract at a dose of 150 or 300 mg/kg before Eth administration for 56 consecutive days. The results showed that the seminiferous tubule diameter and epithelial height were significantly increased in both T-MP treated groups compared to the Eth group. Additionally, the caspase-9 and -3, and PCNA expressions were decreased, but D2R expression was markedly increased in T-MP groups. It was concluded that T-MP seed extract could protect testicular apoptosis induced by Eth via changes in caspase, PCNA, and D2R protein expressions.


Thai Mucuna pruriens (L.) DC. var pruriens (T-MP) contendo levodopa (L-DOPA) e capacidade antioxidante demonstrou melhorar o comportamento sexual e os parâmetros reprodutivos masculinos em ratos tratados com etanol (Eth). No entanto, seu efeito protetor sobre células germinativas apoptóticas testiculares nunca foi relatado. Este estudo teve como objetivo investigar os efeitos potenciais do extrato de semente de T-MP na expressão de proteínas de caspase, antígeno nuclear de proliferação celular (PCNA) e receptor de dopamina D2 (D2R) em ratos Eth. Trinta e seis ratos Wistar machos foram divididos em quatro grupos (9 animais/grupo), incluindo controle, Eth, T-MP150+Eth e T-MP300+Eth, respectivamente. Ratos controle receberam água destilada e ratos Eth receberam Eth (3g/kg PC; 40%v/v). Os grupos T-MP foram tratados com extrato de semente de T-MP na dose de 150 ou 300 mg/kg antes da administração de Eth por 56 dias consecutivos. Os resultados mostraram que o diâmetro dos túbulos seminíferos e a altura epitelial foram significativamente aumentados em ambos os grupos tratados com T-MP em comparação com o grupo Eth. Além disso, as expressões de caspase-9 e -3 e de PCNA diminuíram, mas a expressão de D2R aumentou acentuadamente nos grupos T-MP. Concluiu-se que o extrato de semente de T-MP pode proteger a apoptose testicular induzida por Eth através de alterações na expressão de proteínas caspase, PCNA e D2R.

The present investigation was undertaken to highlight the histomorphometric changes of testicular tissues according to season and age in donkeys (Equus asinus) under northern Algeria conditions. The experiment was conducted from February 2019 to January 2020. A total of 21 sexually mature donkeys were selected randomly. The testis were immediately collected after slaughter for the histological observation. The analysis of the correlation coefficients shows negative correlations between DL and other parameters such as DST, GCEH, TTA and ITSA, ranged between -0.24 and -0.79. Also, there were a high negative correlations between season and the histomorphometric parameters, except DL was revealed positive (r=0.65). On the other hand, there were positive correlations between age and testicular histomorphometrics parameters. The results indicated that DST, GCEH, TTA and ITSA values were significantly higher in winter and autumn seasons than in spring and summer seasons. It is also noteworthy that DST, GCEH, TTA and ITSA values were significantly higher in adult and aged donkeys than in young donkeys. Our comparative analysis of histological parameters, suggests that the sexual activity usually occurs during winter and autumn in local donkeys of Algeria. In addition, our results of histomorphometric of testicular tissues are correlated with age of donkeys.(AU)

Resumo

This study was carried out to investigate the morphological and histological structures of the testis, epididymis and vas deferens of the Al-Ahsa native rooster (ANR). There were two types of ANR; the brown feather one with light yellow shank and the black feather one with grey or dark grey shank. Their body weight was 1840.88± 92.13 g and 1555.66± 82.83g, respectively. The morphology of the testes showed that the black rooster has larger testes than the brown rooster and there was asymmetry in size between the right and left testis in both. They were grey yellowish in color and oval-shaped, situated in the abdomen dorsal to the proventriculus, the liver and the gizzard, cranial to the lungs, caudal to the abdominal sac and ventral to the kidneys. The histology of the testes revealed the capsule, the different cells of the lining epithelium of the seminiferous tubules and the interstitial tissue. The morphology of the epididymis was revealed pseudostratified columnar epithelium, light brown in color with c to L-shaped, located cranial to the testis and extended caudally to continue with vas deferens. The latter has columnar epithelium, light grey in color, run caudally medial to the kidneys and opened in the cloaca.(AU)

Abstract Letrozole is used as a therapeutic agent in reproductive disorders caused by high estrogen levels. Letrozole inhibits cytochrome P450 aromatase and reduces estrogen levels. However, the effects of long-term use on reproductive traits are unknown. The aim of this study was to evaluate the prolonged use of letrozole in the gonads of rodents (Spix's yellow-toothed cavy; Galea spixii). Forty-eight rodents (24 males and 24 females) were randomly divided into the treated and control groups. Letrozole administration started at 15 days of age and continued weekly until 30, 45, 90, and 120 days of age. The body, testis, and ovary weights were analyzed, as well as the morphological progression of spermatogenesis and folliculogenesis. Macroscopically, body weight gain and gonads weight were increased in the letrozole group. Microscopically, the ovaries of treated females showed stratified epithelium and a cellular disorder of the tunica albuginea. In the testes of treated males, the development of seminiferous tubules was delayed and sperm was absent. The collective findings indicate that the prolonged use of letrozole alters secondary sexual characteristics, and causes weight gain, reproductive changes, and male infertility.

Leishmania infantum causes canine leishmaniasis. Using parasitological and molecular analyses, we identified L. infantum in the reproductive organs of male and female dogs. Using histochemistry, immunohistochemistry, and PCR, we examined tissue samples from the reproductive organs of 8 male dogs and 16 female dogs diagnosed with leishmaniasis. Despite the absence of macroscopic or microscopic lesions in these organs, we observed L. infantum amastigotes in tissue samples from the testis and the uterus. PCR and sequencing of these tissues revealed sequences that matched 100% with L. infantum DNA available at GenBank. The presence of L. infantum amastigotes and DNA in testicular and uterine tissue samples suggested that these organs can harbor the parasite without associated macroscopic or microscopic lesions, and this can be especially important in the vertical and venereal transmission of leishmaniasis in dogs.

Histopathological and spermatogenesis classification by Johnsen is widely used in the germinal epithelium maturation analysis, besides identifying pathological alterations able to cause subfertility and even infertility. The aim of this study was to analyze cell-differentiation histopathological data and to correlate them with expression of PRM-1, TNP-2, 17β-HSD3, LHR, generic MHC-I, MIC-B, NC1 and NC3 genes, involved in bovine spermatogenesis using qRT-PCR from testicular parenchyma. Based on Johnsen’s criteria, the results showed normal spermatogenic activity in these animals, classified at 6, 7 and 8 scores. The qRT-PCR analysis expression showed that MHC-I (generic) gene was less expressed than all the other genes in evaluated scores (p < 0.05) and, PRM-1 and TNP-2 were the most expressed genes (p < 0.05). The PRM-1 gene expression was significantly higher than TNP-2 (p < 0.05). Comparing scores, 17β-HSD3 gene expression was lower (p < 0.05) in score 6 when compared to scores 7 and 8 animals. It was also observed that PRM-1 expression was lower in score 6 when compared to 7, as well as TNP-2 gene was less expressed in the score 6 (p < 0.05) when compared to 7 and 8 scores. Our results demonstrated that MHC I (generic), MIC-B, NC1, NC3, and LHR genes are poorly expressed in bovine testis, suggesting their marginal action on spermatogenesis. Instead, PRM-1, TNP-2, and 17β-HSD3 expression were higher, supporting the notion that these genes can act directly on the germ cells differential development during bovine spermatogenesis.


A classificação histopatológica e espermatogênica segundo Johnsen é amplamente utilizada na análise da matu-ração do epitélio germinativo, além de identificar alterações patológicas capazes de causar subfertilidade e até infertilidade. O objetivo deste estudo foi analisar dados histopatológicos de diferenciação celular e correlacioná-los à expressão dos genes PRM-1, TNP-2, 17β-HSD3, LHR, MHC-I genérico, MIC-B, NC1 e NC3, envolvidos na espermatogênese bovina usando qRT-PCR do parênquima testicular. Com base nos critérios de Johnsen, os resultados demonstraram atividade espermatogê-nica normal nesses animais, classificados nos escores 6, 7 e 8. A análise de expressão gênica (qRT-PCR) demonstrou que o gene MHC-I (genérico) foi menos expresso do que os demais nos escores avaliados (p < 0,05) e, PRM-1 e TNP-2 foram os mais expressos (p < 0,05). A expressão do PRM-1 foi significativamente maior do que TNP-2 (p < 0,05). Comparando os escores, a expressão do 17β-HSD3 foi menor (p < 0,05) no escore 6 quando comparado aos escores 7 e 8. Observou-se também que a expressão do PRM-1 foi menor no escore 6 quando comparado ao 7, bem como o TNP-2 foi menos expresso no escore 6 (p < 0,05) quando comparado aos escores 7 e 8. Nossos resultados demonstraram que os genes MHC-I (genérico), MIC-B, NC1, NC3 e LHR são pouco expressos em testículos bovinos, sugerindo sua ação indireta na espermatogênese. Em contrapartida, a expressão de PRM-1, TNP-2 e 17β-HSD3 foi alta, corroborando com a hipótese de que esses genes podem atuar diretamente no desenvolvimento diferencial de células germinativas durante a espermatogênese bovina.

ABSTRACT: The morphology of the male reproductive tract of Tomodon dorsatus was described in the austral seasons of the year considering macroscopic and microscopic variables. For this purpose, 56 specimens from the herpetological collection of the Instituto Butantan were used. Fragments of the testes, kidneys and ductus deferens were collected and submitted to histological routine. The peak of the testicular volume was observed in the summer and the epithelium of the seminiferous tubules had higher height in the summer (p=0.001). The testes were active throughout the year, however, the spermiogenesis peaked in the summer. There were spermatozoa in the lumen of the ductus deferens in all seasons of the year. Renal length was higher in autumn (p=0.027), and renal width did not show a significant increase (p=0.237). The diameter and epithelial height of the sexual segment of the kidney (SSK) showed hypertrophy in winter and spring, coinciding with the mating period. Based on findings of this study, we can suggest that, at the population level, the reproductive cycle of T. dorsatus can be considered seasonal semi-synchronous, due to the peak of spermiogenic activity in the hot season, and discontinuous at the individual level.


RESUMO: A morfologia do trato reprodutivo do macho de Tomodon dorsatus foi descrita nas estações climáticas do ano com base em variáveis macroscópicas e microscópicas. Para isto, foram usados 56 espécimes oriundos da coleção herpetológica do Instituto Butantan. Fragmentos dos testículos, rins e ductos deferentes foram coletados e submetidos à rotina histológica. O volume testicular foi maior no verão e o epitélio dos túbulos seminíferos mostrou uma maior altura no verão (p=0.001). Os testículos estavam ativos durante todo o ano, contudo, a espermiogênese foi maior no verão. Espermatozoides foram encontrados no lúmen do ducto deferente em todas as estações do ano. O comprimento renal foi maior no outono (p=0.027), e a largura renal não mostrou um aumento significativo (p=0.237). O diâmetro e a altura epitelial do segmento sexual do rim (SSR) mostrou hipertrofia nas estações inverno e primavera, coincidindo com o período reprodutivo. Com base nestes resultados, pode-se sugerir que, em nível populacional, o ciclo reprodutivo de T. dorsatus possa ser considerado semi-sincrônico sazonal, devido à atividade espermiogênica na estação quente, e descontínuo em nível individual.

The morphology of the male reproductive tract of Tomodon dorsatus was described in the austral seasons of the year considering macroscopic and microscopic variables. For this purpose, 56 specimens from the herpetological collection of the "Instituto Butantan" were used. Fragments of the testes, kidneys and ductus deferens were collected and submitted to histological routine. The peak of the testicular volume was observed in the summer and the epithelium of the seminiferous tubules had higher height in the summer (p=0.001). The testes were active throughout the year, however, the spermiogenesis peaked in the summer. There were spermatozoa in the lumen of the ductus deferens in all seasons of the year. Renal length was higher in autumn (p=0.027), and renal width did not show a significant increase (p=0.237). The diameter and epithelial height of the sexual segment of the kidney (SSK) showed hypertrophy in winter and spring, coinciding with the mating period. Based on findings of this study, we can suggest that, at the population level, the reproductive cycle of T. dorsatus can be considered seasonal semi-synchronous, due to the peak of spermiogenic activity in the hot season, and discontinuous at the individual level.(AU)

In non-neutered dogs, the testis is the second most common site for development of neoplasms, frequently occurring in elderly animals and with orchiectomy as main treatment. Regarding the great relevance of testicular disorders and the importance of accurate diagnoses, this study aims to report a case of canine leydigocytoma at the Veterinary Hospital in the State University of Ceará. The 10-years-old non-neutered Pitbull-breed animal was attended presenting hematuria and, physical examination showed a bilateral nodular formation in testicles in addition to an enlarged prostate. In ultrasonographic examination, the prostate was enlarged, presenting irregular shape and heterogeneous echotexture. These changes indicate the presence of prostatic hyperplasia. Both testicles presented well-defined hypoechogenic regions with evident marginal and internal vascularization, which indicate neoplastic neoformation. In cytology, a hypercellular epithelial sample was observed with cellular modifications suggestive of interstitial cell neoplasia. In the lab, macroscopically, the testis showed multiple soft, well-defined, white-colored to brownish nodules. Histopathology revealed neoplastic proliferation of Leydig cells, sustained by moderate fibrovascular stroma, with degenerated adjacent parenchyma and without signs of malignancy. Four months after orchiectomy, the prostate turned to normal dimensions, with no further ultrasonographic changes. This case reinforces the importance of bilateral orchiectomy and ultrasound for prostatic abnormalities and testicular neoplasms. The association of these conditions is not commonly described in the literature, being an important contribution in this regard.

Our objective was to investigate whether the pulp and paper mill industry effluent could affect the testis and Sertoli cells in a fast exposure period. For this, the present study was carried out in immature rats at 10-day-old. Testis treated in vitro with 4% effluent for 1 h presented changes in energy metabolism in terms of a decrease in lactate content and glucose uptake. Elevation in GSH content, as an antioxidant defense mechanism, was also detected. Sertoli cells treated with 4% effluent for 1 hour showed alterations in the mitochondrial metabolism that favor the decoupling of oxidative phosphorylation and the generation of oxygen reactive species and also a time and concentration-dependent delay secretion of acidic vesicles. Our results showed that pollutants present in the pulp and paper mill effluents, in a short time of exposure, are capable of inducing alterations in important metabolic functions in the testis and in Sertoli cells that are crucial for the correct progression of spermatogenesis and fertility.

Cryptorchidism is a genital alteration wherein one or both testicles fail to descend into the scrotum and has multifactorial causes. A free-range adult male was captured twice in the Pantanal of Nhecolândia to put a GPS collar and semen collection. Pharmacological semen collection, andrological examination and semen analysis were performed. At the first capture and during the andrological examination only the left testis was found, and the male qualified as cryptorchid. The penis had no penile spines at either procedure. The semen volume obtained at first and second capture was 435 and 160 L, respectively, with a concentration of 618 and 100 x 106 sperm/mL, progressive motility of ~ 5% and ~ 1% and total morphological sperm abnormalities of 74% and 86%. The male was monitored by a GPS collar, but the signal was lost, making it difficult to re-captures and perform new seminal and ultrasound evaluations to discard monorchidism exceedingly rare in felids. Genetic studies to assess the individual's homozygosity are necessary to verify whether cryptorchidism in this individual has a genetic factor.
