Resumo

Listeriosis is an infectious disease caused by bacteria of the genus Listeria, the neurological form being more common in ruminants. There are many reports of listeriosis in small ruminants in the region that includes Brazil, Argentina and Uruguay. However, these diagnoses were mainly based on histological lesions in the central nervous system (CNS) without the isolation and characterization of the involved Listeria strains. The aim of this study was to report sheep and goats listeriosis cases from 2016 to 2021 in northwestern Uruguay. The diagnosis was made according to lesions observed at histopathology, plus Listeria isolation in CNS, identifying it at specie and serotype level. Nine animals (n=9) of three outbreaks and five sporadic cases of listeriosis were studied. Sheep was the species with more cases in relation to goats, and adults were the category most affected. Cases occurred in spring and less frequently in winter. All presented neurological clinical signs and the lesions in the CNS were consistent with suppurative meningoencephalitis and micro-abscesses in the brainstem. In eight of nine CNS samples, Listeria strains were isolated (seven L. monocytogenes and one L. innocua). All the L. monocytogenes isolates carried the inlA gene; serotyping showed that four strains belonged to serotype 1/2b, two isolates belonged to serotype 4b, and one to serotype 1/2a. Considering that listeriosis is a common disease in this region and the fact that isolates are scarcely recovered from small ruminants, it would be important to emphasize the need for Listeria isolation to better characterize the strains that affect animals. Not only to improve knowledge about the epidemiology of disease but also with the objective of developing serotype specific vaccines for animal use.


Listeriose uma doença bacteriana causada pelo gênero Listeria, a forma nervosa é a mais comum em ruminantes. No Brasil, Argentina e Uruguai há vários relatos de listeriose em pequenos ruminantes com diagnóstico baseado na histopatologia do sistema nervoso central (SNC), sem o isolamento e a caracterização do agente. O objetivo deste trabalho foi relatar uma série de casos diagnosticados em ovinos e caprinos no período 2016-2021 no noroeste do Uruguai. O diagnóstico foi feito basado nas lesões observadas na histopatologia, e caracterização das cepas de Listeria recuperadas do SNC quanto à espécie e sorotipo. Nove animais (n=9) do três surtos e cinco casos isolados de listeriose foram estudados. Os ovinos foram a espécie com o maior número de casos em relação aos caprinos, sendo os animais adultos a categoria mais afetada em ambas espécies. A doença ocorreu principalmente na primavera com alguns casos observados no inverno. Todos os casos apresentavam sinais clínicos nervosos e as lesões no SNC caracterizavam-se por meningoencefalite supurativa com presença de microabscessos no tronco encefálico. Em oito de nove amostras do SNC foram isoladas cepas de Listeria (sete L. monocytogenes e uma L. innocua). Todos os isolados de L. monocytogenes continham o gene inlA; a sorotipagem apresentou quatro cepas do serotipo 1/2b, duas cepas serotipo 4b e uma cepa 1/2a. Levando em consideração que nesta região a listeriose é uma doença frequente e que existem poucos isolados recuperados de casos clínicos em pequeño ruminantes, torna-se relevante o isolamento deste agente para caracterização das cepas que afetam os animais. Não só para melhorar o conhecimento sobre a epidemiologia da doença, mas também com o objetivo de desenvolver vacinas sorotipo-especificas para uso animal.

Background: A listeriosis outbreak in a sheep fattening feedlot in the Southern Region of Rio Grande do Sul State, Brazilis described. This disease is caused by Listeria monocytogenes and represents a risk to public health since it affects notonly ruminants but also humans. This agent is widely spread in the environment, such as in the soil and water. It is alsofound in decaying vegetable matter and the feces and fluids of domestic animals. The aim of this study was to describe alisteriosis outbreak in sheep raised in feedlots, its epidemiology, and to establish the importance of this disease in this typeof sheep management system, evaluate the possible sources of infection, and suggest ways to control it.Cases: Sheep were kept in a 2-sector shed, one with east solar orientation and the other with west solar orientation, thelatter with free access to domestic birds. Sheep were fed silage and concentrate. Seven sheep were affected, 5 died and 2recovered. Clinically, the sheep displayed loss of balance, excessive drooling, and tremors; one exhibited circling, headdeviation, apathy, nystagmus, lateral recumbency, paddling, and labored breathing. At necropsy, macroscopic lesions werenot found, and histologically several micro-abscesses and perivascular cuffs with lymphocytes, macrophages, and neutrophils were present in the brain stem. Listeria monocytogenes suspected colonies were observed in the microbiologicalculture, and the bacteria was identified by biochemical analysis. Background: Nitric oxide is synthesized from L-arginine and catalyzed by a family of NOS. There are three different NOS isoforms: neuronal (nNOS), inducible (iNOS) and endothelial (eNOS). Nitric oxide is an important apoptosis regulator in mammalian system that can induce and prevent apoptosis depending on levels of NO production and environmental conditions of the cell. NOS expression and its relationship with apoptosis has not been well elucidated in listerial meningoencephalitis in sheep. The aim of this study was to investigate eNOS and iNOS expressions in the brain of sheep with natural listeriosis and to compare them with apoptosis which is shaped in the region.Materials, Methods & Results: In the study, formalin fixed and paraffin embedded brainstem tissue from 25 sheep naturally infected with LM were used from archives. Five μm-thick section was taken from each block. Histopathologically, sections were stained with H&E. Five normal sheep brain tissues were used as control. At the end of the study, Histopathologically in brainstem tissue infected with LM, multifocal microabscesses in different sizes mixed with neutrophils and macrophages were detected and perivascular mononuclear cell infiltration and meningitis characterized by mononuclear cell infiltration were found. All sections were also immunohistochemically stained with LM, eNOS and iNOS antibodies. In addition, TUNEL method was used to determine apoptosis in brain tissues. As a result of immunostaining, listeria immunoreactivity was observed in microabscesses. The Listeria antigens were detected mainly in the cytoplasm of the neutrophils and macrophages and located extracellulary in microabscesses. Both eNOS and iNOS immunoreactivity were observed in very few neurons and glial cells in normal control sheep. Neurons and glial cells in brain tissues of infected animals stained with eNOS and iNOS.[...][

Este trabalho teve como objetivo realizar a detecção de cepas de Listeria monocytogenes de cortes cárneos bovinos bem como no ambiente de abatedouros frigoríficos localizados no Distrito Federal, promover a sorotipificação pela reação em cadeia da polimerase (PCR), realizar antibiograma e submeter às cepas à eletroforese de campo pulsado (Pulsed-field gel electrophoresis - PFGE). Foram analisados um total de 125 cortes cárneos bovinos, 45 amostras de swabs de carcaças e 43 amostras de swabs em que foram detectados 13 cepas de Listeria monocytogenes, sendo 11 em cortes cárneos bovinos e 2 swabs de ambiente em um abatedouro frigorifico. Não foram isoladas cepas de swabs de carcaça. Dentre as 13 cepas de Listeria monocytogenes foram encontradas seis cepas do sorotipo 4b, cinco do sorotipo 1/2c e duas cepas do sorotipo 1/2a. Dentre as 11 cepas de L. monocytogenes encontradas em cortes cárneos bovino, uma (9,1%) cepa apresentou resistência a eritromicina, outra (9,1%) cepa a gentamicina e outra a ciprofloxacina (9,1%) e todas as cepas (100%) apresentaram resistência ao Ác. Nalidíxico. Das duas (2) cepas oriundas de ralos de abatedouro frigorífico, todas (100%) apresentaram resistência ao Ác. Nalidíxico e a sulfonamidas. A análise por eletroforese de campo pulsante (PFGE) demonstrou 13 diferentes pulsotipos, em que foram agrupados em 3 diferentes grupos clonais, que coincidentemente se correlacionavam com os 3 diferentes sorotipos encontrados sugerindo uma ampla disseminação desses perfis no Distrito Federal.(AU)

O relato objetivou descrever os achados epidemiológicos, clínico-patológicos, laboratoriais e microbiológicos em um surto de meningoencefalite em ovinos por Listeria monocytogenes associado ao consumo de silagem. A enfermidade acometeu oito ovinos pertencentes a um rebanho de 600 animais, alcançando letalidade de 100%. A única ovelha examinada apresentou apatia, febre, cegueira bilateral com midríase, hipotonia lingual, nistagmo e torneio à direita com quedas esporádicas. A hematologia revelou linfopenia absoluta com inversão na relação de linfócito / neutrófilo (0,16). A análise do líquido cefalorraquidiano (LCR) mostrou aspecto turvo e pleocitose neutrofílica (92 leucócitos/µL). L. monocytogenes foi isolada em quatro alíquotas de LCR submetidas à cultura microbiológica. Em quatro ovinos necropsiados, as lesões macroscópicas consistiram de congestão das meninges e turbidez do LCR. Microscopicamente, a principal característica foi meningoencefalite do troco encefálico, bulbo, ponte, mesencéfalo e medula oblonga, com microabscessos multifocais e gliose. Infiltrado neutrofílico e linfocítico multifocal nos fascículos do nervo trigêmeo foi também observado. Os autores reiteram a importância das medidas preventivas, a fim de reduzir a incidência da listeriose.(AU)

The disease affected eight sheep from a 600 sheep flock, achieving 100% lethality. The only ewe examined presented apathy, fever, bilateral blindness with mydriasis, tongue hypotony, nystagmus and right circling with sporadic falls. Hematology revealed absolute lymphopenia with inversion in the lymphocyte/neutrophil ratio (0.16). Cerebrospinal fluid (CSF) analysis showed turbid aspect and neutrophilic pleocytosis (92leukocytes/µL). L. monocytogenes was isolated from four CSF samples submitted to microbiological assays. In four necropsied sheep, gross lesions included meningeal congestion and CSF clouding. Microscopically, the main feature was meningoencephalitis of the brainstem, bulb, pons, mesencephalus and medulla oblongata, with multifocal microabscesses and gliosis. Multifocal lymphocyticand neutrophilic infiltrate in the trigeminal nerve fascicles was also observed. The authors reiterate the importance of adequate preventive measures to reduce incidence of listeriosis.(AU)

In addition to listeriosis which is relatively common in ruminants, there are three other uncommon suppurative intracranial processes (SIP) identifiable in adult ungulates as brain abscess, basilar empyema and suppurative meningitis. The present paper reports the epidemiological, clinical, laboratorial, pathological and microbiological findings of 15 domestic ruminants with SIP. A total of 15 animals were selected (eight sheep, four cattle and three goats); with the definitive diagnoses of basilar empyema (n=3), brain abscess (n=1), listeriosis (n=5) and suppurative meningitis (n=6). Hematology revealed leukocytosis with inversion of the lymphocyte/ neutrophil ratio in 4 cases. In the majority of animals, cerebrospinal fluid (CSF) presented light yellow coloration and cloudy aspect due to neutrophilic pleocytosis (15 - 997 leukocytes/µL). Microbiological culture of CSF or central nervous system (CNS) fragments resulted on isolation of Trueperella (Arcanobacterium) pyogenes,Listeria monocytogenes,Escherichia coli and Stenotrophomonas sp. In a goat with thalamic abscess, microbiological assay was not performed, but Gram positive bacilli type bacteria were observed in histology. The diagnosis of these outbreaks was based on the association of epidemiological, clinical, pathological and bacteriological findings; reiterating that the infectious component remains an important cause of CNS disease in domestic ruminants and also shows the need for dissemination of information about the most effective preventive measures for the ranchers.(AU)


Além da listeriose, que é relativamente comum em ruminantes, existem outros três processos supurativos intracranianos (PSI) identificáveis em ungulados adultos, que não são comuns, como: abscesso cerebral, empiema basilar e meningite supurativa. O presente trabalho tem como objetivo relatar os achados epidemiológicos, clínicos, laboratoriais, patológicos e microbiológicos de 15 ruminantes domésticos com PSI. O total de 15 animais foi selecionado (oito ovinos, quatro bovinos e três caprinos), com o diagnóstico definitivo de empiema basilar (n=3), abscesso cerebral (n=1), listeriose (n=5) e meningite supurativa (n=6). A hematologia revelou leucocitose com inversão da relação linfócito/ neutrófilo em quatro animais. Na maioria dos animais, a análise do líquido cefalorraquidiano (LCR) revelou coloração amarelado clara e aspecto turvo devido à pleocitose neutrofílica (15 - 997 leucócitos/µL). A cultura microbiológica de LCR ou de fragmentos do sistema nervoso central (SNC), permitiu o isolamento de Trueperella (Arcanobacterium) pyogenes,Listeria monocytogenes,Escherichia coli e Stenotrophomonas sp. No caprino com abscesso de tálamo, a cultura microbiológica não foi realizada, mas bactérias Gram positivas tipo bacilos foram observadas na histologia. O diagnóstico desses surtos foi baseado na associação dos achados epidemiológicos, clínicos, patológicos e bacteriológicos; reiterando que o componente infeccioso continua sendo uma causa importante de doença do SNC em ruminantes domésticos e também mostra a necessidade de disseminação da informação sobre as medidas preventivas mais eficazes para os criadores.(AU)

Resumo

This study was conducted to evaluate the antibacterial effect of Carum copticum essential oil (Ajowan EO) against Listeria monocytogenes in fish model system. Ajowan EO chemical composition was determined by gas chromatography/mass spectral analysis and the highest concentration of Carum copticum essential oil without any significant changes on sensory properties of kutum fish (Rutilus frisii kutum) was assigned. Then the inhibitory effect of Ajowan EO at different concentrations in presence of salt and smoke component was tested on L. monocytogenes growth in fish peptone broth (FPB), kutum broth and cold smoked kutum broth at 4 ºC for 12 days. Ajowan EO completely decreased the number of L. monocytogenes in FPB after 12 days of storage, however, antimicrobial effect of EO significantly reduced in kutum and cold smoked kutum broth. Addition of 4% NaCl and smoke component improved the anti-listerial activity of Ajowan EO in all fish model broths.(AU)

Resumo

A retrospective study of the goat necropsies performed over a period of 48 years (1964 to 2011) at the Laboratório de Patologia Veterinária, Universidade Federal de Santa Maria, Rio Grande do Sul (RS) state, Brazil, was performed. A total of 114 reports of goat necropsies were analyzed. Ninety five necropsies (83.33%) had a conclusive diagnosis and 19 (19.66%) had inconclusive diagnosis. Out of the conclusive cases, infectious and parasitary diseases were the most prevalent, followed, in decreasing order of prevalence, by metabolic and nutritional diseases, poisonings and toxi-infections, and developmental diseases. Other conditions or lesions that did not fit any of the above groups of diseases affected about 10% of the necropsied goats. Hemonchosis was the most prevalent cause of death in this study. Eimeriosis and listeriosis were also important causes of goat deaths. Among the metabolic and nutritional diseases, urolithiasis, osteoporosis, pregnancy toxemia, malnutrition, and white muscle disease were the most prevalent. Mostly the infectious and parasitary diseases and the metabolic and nutritional diseases occurred many times as outbreaks, causing even more important economic losses.(AU)


Foi realizado um estudo retrospectivo dos arquivos do LPV-UFSM e examinados os laudos de necropsias de caprinos realizadas num período de 48 anos (1964 a 2011). Foram analisados 114 laudos de necropsias de caprinos. Deste total, 95 (83,33%) tinham diagnóstico conclusivo e 19 (19,66%) tinham diagnóstico inconclusivo. Dos 95 casos conclusivos, as doenças infecciosas e parasitárias foram as mais prevalentes, seguidas em ordem decrescente de prevalência, pelas doenças metabólicas e nutricionais, intoxicações e toxi-infecções e alterações do desenvolvimento. Outras alterações de diferentes naturezas e etiologias que não se enquadravam nos grupos de doenças acima afetaram cerca de 10% dos caprinos examinados. A hemoncose foi a principal causa de morte de caprinos na área de abrangência do LPV-UFSM. Eimeriose e listeriose também foram causas importantes de morte. Dentre as doenças metabólicas e nutricionais, urolitíase, osteoporose, toxemia da prenhez, desnutrição e doença dos músculos brancos foram as mais prevalentes. Principalmente as doenças infecciosas e parasitárias e as metabólicas e nutricionais ocorreram muitas vezes na forma de surtos, acarretando maiores perdas econômicas associadas.(AU)

Resumo

São descritos sete casos de doença neurológica em ovinos por Listeria monocytogenes no Rio Grande do Sul e Paraná entre 2000 e 2007. Foram afetados ovinos com idades entre 12-24 meses. Os casos ocorreram no verão e início da primavera e os índices gerais de morbidade e letalidade foram de 3,15% e 100%, respectivamente. Quando essa informação estava disponível, nenhum dos ovinos afetados era alimentado com silagem. Em três propriedades havia contato próximo dos ovinos afetados com outras espécies. A evolução do quadro clínico foi de 12 horas a três dias e os sinais clínicos foram caracterizados por decúbito (7/7), desvio da cabeça (4/7), incoordenação (3/7), depressão (3/7), andar em círculos (2/7), cegueira unilateral, emagrecimento progressivo, febre, midríase, movimentos de pedalagem, nistagmo lateral, opistótono, paralisia flácida dos membros pélvicos ou dos quatro membros, salivação excessiva e tremores (1/7 cada). Histologicamente observou-se encefalite com microabscessos, predominantemente unilateral com variáveis graus de gliose e alterações degenerativas como esferóides axonais e infiltração de células Gitter. As lesões se estendiam desde a medula oblonga até o mesencéfalo. Antígenos de Listeria monocytogenes foram detectados por imuno-histoquímica em seções de tronco encefálico de todos os ovinos afetados. O diagnóstico foi realizado com base nos achados epidemiológicos e clinico-patológicos, e confirmado pela imuno-histoquímica (IHQ) utilizando anticorpo policlonal anti-L. monocytogenes.(AU)


Seven cases of neurological disease in sheep caused by Listeria monocytogenes in Rio Grande do Sul and Paraná state, southern Brazil are described. The cases occurred between 2000 and 2007 and 12-24-month-old sheep were affected. Overall morbidity and lethality rates were 3.15% and 100%, respectively. Cases occurred in the summer and early spring. When this information was available, affected sheep had not been fed with silage. In three farms there were close contact among affected sheep and other species. Clinical signs were characterized by recumbency (7/7), head tilt (4/7), incoordination (3/7), depression (3/7), circling (2/7), unilateral blindness, wasting, fever, midriasis, paddling, opisthotonus, hind or hind and fore limb paralysis, drooling, and muscle tremors (1/7 each). Clinical evolution varied from 12 hours to three days. Histological findings consisted of predominantly unilateral, microabscedative encephalitis with variable degrees of gliosis and degenerative lesions characterized by axonal spheroids and infiltration by Gitter cells. These lesions were observed extending from medulla oblongata to mesencephalon. Listeria monocytogenes antigen was showed by imunohistochemistry in routinely processed sections of brainstem from all seven affected sheep. The diagnostic was based on epidemiological, clinical, and pathological findings and confirmed by immunohistochemistry (IHQ) using polyclonal anti-L. monocytogenes antibody.(AU)

Em um rebanho de 100 caprinos, foram observados casos de uma doença neurológica em três animais adultos jovens (identificados como A-C) entre outubro e dezembro de 2004. Os sinais clínicos incluíam desvio da cabeça, torcicolo, nistagmo, andar desequilibrado e quedas que acabavam em decúbito, durante o qual o caprino executava movimentos de pedalagem com os membros rígidos. Tratamento com antibióticos no Caprino C resultou em remissão temporária dos sinais. A evolução clínica foi de 5, 10 e 30 dias, respectivamente, para os Caprinos A, B e C. O Caprino A morreu espontaneamente e os outros dois foram eutanasiados na fase terminal da doença. Não foram observadas alterações macroscópicas. Lesões histológicas eram predominantemente unilaterais e consistiam de alterações inflamatórias e degenerativas que se estendiam do bulbo ao tálamo. Em todos os casos havia manguitos perivasculares de um ou mais tipos de células mononucleares (linfócitos, plasmócitos, macrófagos ativados) e ocasionalmente neutrófilos associados a áreas de malacia onde células Gitter preenchiam os espaços de perda do parênquima. Adicionalmente, o Caprino B tinha microabscessos no bulbo, ponte, e mesencéfalo e infiltrado neutrofílico e linfocítico multifocal nos fascículos do nervo trigêmeo, próximo ao gânglio de Gasser, e nas leptomeninges do cerebelo. No tronco encefálico do Caprino C, a reação era multifocal e com característica granulomatosa e incluía células epitelióides e, ocasionalmente, células gigantes multinucleadas. Listeria sp foi detectada por imunoisto-química em cortes de mesencéfalo rotineiramente processado dos Caprinos A e C e de ponte do Caprino B. (AU)


A neurologic disease was observed in three young adult goats (indentified as A-C) from a herd of 100 goats during October-December, 2004. Clinical signs included head tilt, torticollis, nystagmus, staggering, falls and eventually recumbency and paddling, with stiff limbs. Treatment of Goat C with antibiotics resulted in a temporary remission of the clinical signs. The clinical courses where 5, 10 and 30 days respectively for Goats A, B and C. Goat A died spontaneously and the other two where euthanatized in extremis. No gross changes were observed. Histologic lesions were predominantly unilateral, with inflammatory and degenerative changes, extending from the medulla oblongata to the thalamus. In all 3 cases there were perivascular cuffings of one or more types of mononuclear cells (lymphocytes, plasma cells, activated macrophages) and occasionally neutrophils associated with areas of malacia where Gitter cells filled spaces of parenchymal loss. Additionally, Goat B had microabscesses in the medulla, pons, and mesencephalon and multifocal neutrophilic and lymphocytic infiltrates within the fascicles of the trigeminal nerve and in the cerebellar leptomeninges. In the brainstem of Goat C, there was multifocal granulomatous inflammation which included epithelioid macrophages and occasional multinucleated giant cells. Listeria sp antigen was detected by imunohistochemistry in routinely processed sections of mesencephalon from Goats A and C and of pons from Goat B. (AU)
