To effectively develop and utilize high-quality Tianfu broilers, this study evaluated the morphological and structural characteristics of the immune organs of such broilers with different strains (HS1 and HS2) at different developmental stages and analyzed the distribution of mast cells by toluidine blue staining. Moreover, the localization and expression of immunoglobulin, complement C3, C4 and CD3 in immune organs were also detected. The results showed that although there was no significant difference in the development of immune organs in the HS1 and HS2, the number of lymphatic follicles and capsule thickness in the spleen and bursa of Fabricius in HS1 were greater than those in HS2. Additionally, the number of mast cells in the spleen of HS1 was greater at Day 1 and Day 21 and was significantly higher than that of HS2 (p<0.05); the number of mast cells in the bursa of Fabricius reached 9.17 on Day 7, which was significantly higher than that of HS2 (p<0.05). Moreover, the serum IgA and IgM levels in HS1 were higher than those in HS2 on Day 14 and 21 (p<0.05). In addition, the complement C3 content in HS1 was significantly or extremely significantly higher than that in HS2 on Days 1, 14 and 21 (p<0.01, p<0.05), respectively, but significantly lower than in HS2 on Day 7 (p<0.05). These results indicated that the disease resistance of the HS1 line was stronger than that of the HS2 line, which lays a foundation for future disease- resistance breeding of Tianfu broilers.(AU)

Although propolis has been reported for having anti-inflammatory activities, its effects on complement system has not been much studied. This research was conducted to find out the effects of Indonesian propolis on the expression levels of C3, C1r/s, Bf, MBL, and C6 in zebrafish larvae which were induced by lipopolysaccharide (LPS). Counting of macrophages migrating to yolk sac and liver histology were carried out. Larvae were divided into four groups: CON (cultured in E3 medium only), LPS (cultured in a medium containing 0.5 μg/L LPS), LPSIBU (cultured in a medium containing LPS, and then treated with 100 μg/L ibuprofen for 24 hours), and LPSPRO (cultured in a medium containing LPS, and then immersed in 14,000 μg/L propolis for 24 hours) groups. The results showed that complement gene expression in larvae from the LPSIBU and LPSPRO groups were generally lower than in larvae from the LPS group. The number of macrophage migrations to the yolk in the LPSPRO group was also lower than in the LPS group. Histological structure of liver in all groups were considered normal. This study shows that Indonesian propolis has the potential to be used as an alternative to the substitution of NSAIDs.

The objective of this study was to determine the effects that breeder age has on digestive and immune system development; the transfer of immunoglobulins to egg yolk, yolk sac, and neonate chicks; and the immune response of chicks up to 35 days old. Three ages (32, 42, and 52 weeks) of Hubbard breeders were studied with ages as treatments. A total of 425 eggs were weighed for each of the three treatments and incubated. After hatching, a total of 300 1-day-old chicks were used in each treatment. We studied the development of the gastrointestinal tract and immune system of progeny and IgY transfer from breeder to progeny. Chicks from 52-week-old breeders had greater gastrointestinal tract growth up to seven days of life and greater body weight at 14 days. Older breeders (52 weeks) had higher amounts of IgY in serum and egg yolk. Chicks from the youngest breeders (32-weeks-old) had a better immune response at two weeks post-vaccination. It can be concluded that the older breeders have a greater capacity to immunize progeny up to 14 days. Strategies can be developed to increase IgY in the serum of young breeders and, consequently, increase the innate immunity of the newly-hatched chicks.(AU)

This study was conducted to investigate effects of vitamin A (VA) and vitamin K3 (VK3) on immune function and intestinal antioxidant capacity of aged laying hens. In a 3 × 3 factorial arrangement, the diets of 1080 Roman Pink laying hens (87 weeks old) was formulated with deficient, adequate and excess VA and VK3, including 0, 7000 and 14000 IU/kg VA and 0, 2.0 and 4.0 mg/kg VK3 for 8 weeks. Interactive effects between VA and VK3 were observed that VA and VK3 decreased the splenetic mRNA expression of inducible nitric oxide synthase (iNOS) and tumour necrosis factor α (TNF-α), but increased the plasma immunoglobulin G (IgG) content and jejunal mRNA expression of nuclear factor-like 2 (Nrf2). Hens fed adequate or excess VA had higher spleen index, mRNA expression of interleukin-10 (IL-10) in spleen, sIgA content, catalase (CAT), glutathione peroxidase and total dismutase (T-SOD) activity, and mRNA expression of polymeric immunoglobulin receptor (pIgR) in jejunum and lower mRNA expression of IL-1ß in jejunum and iNOS, TNF-α in spleen. Furthermore, adequate or excess VK3 significantly increased plasma IgG content, the CAT, T-SOD and total antioxidant capacity activities, up-regulated the mRNA expression of pIgR, Nrf2, SOD1 and CAT in jejunum and down-regulated the mRNA expression of iNOS and TNF-α in spleen.(AU)

For honey production, beekeepers add one or more supers to the hives to allow honeybees to store their products. However, the increase in hive space can affect the social and health organization in the colony, promoting stress. This study assessed the management of honey production, physicochemical honey properties, population development, and forages immune system gene expression patterns to be used as biomarker for monitoring beekeeping welfare. The treatments comprised 40 beehives divided in four treatments. Treatment 1 - control, supers added according to storage necessity. Treatments 2, 3, and 4 presented two, three, and four supers at the beginning of the experiment, respectively. T1 presented greater honey production (39.4 % increased). No difference in open brood area in the colonies was observed and honey properties and only T2 showed closed brood area higher than the other treatments. Foragers from T4 showed higher catalase and defensin gene expression at the middle-end experiment. Thus, the increasing internal space at the beginning of honey season can affect honey production and immune system of foragers. Catalase and defensin can be used as biomarkers for monitoring honey production welfare.

Two experiments were carried out to evaluate the effects of digestible valine supplementation in pre-starter and starter diets on the productivity, nutrient metabolizability coefficient (NMC), and immune response of broilers from breeders of different ages. Experiments I and II were conducted with broilers in the pre-starter (1 to 7 days of age) and starter (8 to 21 days of age) phases, respectively. Broilers were fed diets that differed in their digestible valine content. In each trial, 400 male Cobb 500® chicks were randomly housed in a 2 × 4 factorial arrangement with eight treatments and five replicates of 10 birds each. The main effects were breeder age (37 vs. 52 weeks) and the digestible valine level in pre-starter (9.2, 10.2, 11.2, and 12.2 g/kg) and starter (8.3, 9.3, 10.3, and 11.3 g/kg) diets. Productive performance, intestinal histology, and immune response of broilers were evaluated. Supplementation with 11.2 g/kg valine in pre-starter diets improved NMC and increased villus height and villus:crypt ratio in the duodenum, jejunum development, and lymphocyte proliferation in the spleen of broilers at seven days of age, without improving performance (body weight gain, feed intake, and feed conversion ratio). In the starter phase, valine supplementation reduced feed intake but did not affect NMC, intestinal development, or immune response. The present results suggest that the same level of digestible valine should be used in the diet of broilers born to breeders of different ages, and the use of 9.2 and 8.3 g/kg digestible valine in pre-starter and starter diets, respectively, is sufficient to ensure satisfactory broiler performance. However, to improve the duodenum and jejunum development and immune response of broilers in the pre-starter phase, higher digestible valine levels are required.

Insects can be found in numerous diverse environments, being exposed to pathogenic organisms like fungi and bacteria. Once these pathogens cross insect physical barriers, the innate immune system operates through cellular and humoral responses. Antimicrobial peptides are small molecules produced by immune signaling cascades that develop an important and generalist role in insect defenses against a variety of microorganisms. In the present work, a cecropin B-like peptide (AgCecropB) sequence was identified in the velvetbean caterpillar Anticarsia gemmatalis and cloned in a bacterial plasmid vector for further heterologous expression and antimicrobial tests. Methods AgCecropB sequence (without the signal peptide) was cloned in the plasmid vector pET-M30-MBP and expressed in the Escherichia coli BL21(DE3) expression host. Expression was induced with IPTG and a recombinant peptide was purified using two affinity chromatography steps with Histrap column. The purified peptide was submitted to high-resolution mass spectrometry (HRMS) and structural analyses. Antimicrobial tests were performed using gram-positive (Bacillus thuringiensis) and gram-negative (Burkholderia kururiensis and E. coli) bacteria. Results AgCecropB was expressed in E. coli BL21 (DE3) at 28°C with IPTG 0.5 mM. The recombinant peptide was purified and enriched after purification steps. HRMS confirmed AgCrecropB molecular mass (4.6 kDa) and circular dichroism assay showed α-helix structure in the presence of SDS. AgCrecropB inhibited almost 50% of gram-positive B. thuringiensis bacteria growth. Conclusions The first cecropin B-like peptide was described in A. gemmatalis and a recombinant peptide was expressed using a bacterial platform. Data confirmed tertiary structure as predicted for the cecropin peptide family. AgCecropB was capable to inhibit B. thuringiensis growth in vitro.(AU)

This review focuses on the innate immune events modulated by conceptus signaling during early pregnancy in ruminants. Interferon-tau (IFN-) plays a role in the recognition of pregnancy in ruminants, which involves more than the inhibition of luteolytic pulses of PGF2 to maintain corpus luteum function. For successful pregnancy establishment, the allogenic conceptus needs to prevent rejection by the female. Therefore, IFN- exerts paracrine and endocrine actions to regulate the innate immune system and prevent conceptus rejection. Additionally, other immune regulators work in parallel with IFN-, such as the pattern recognition receptors (PRR). These receptors are activated during viral and bacterial infections and in early pregnancy, but it remains unknown whether PPR expression and function are controlled by IFN-. Therefore, this review focuses on the main components of the innate immune response that are involved with early pregnancy and their importance to avoid conceptus rejection.(AU)

In dairy cattle, supplementation with polyunsaturated fatty acids (PUFAs) is considered to be an important tool to decrease the negative energy balance of periparturient dairy cows and improve the reproductive and immune systems. The most common PUFAs added to ruminant diets are omega 3 (n-3 PUFA) as linolenic acid and omega 6 (n-6 PUFA) as linoleic acid. This paper aims to review the potential effects of n-3 PUFA. We consider the effects of n-3 PUFA on the bovine immune system, especially on immune cells, and on in vivo and in vitro reproductive parameters, emphasizing how n-3 PUFAs act as modulators through one or more molecular mechanisms. The incorporation of n-3 PUFA in the dairy cow diet has positive effects on animal fertility and immunity. Future research on n-3 PUFA should be more explored concerning reproduction and immune function, starting from the investigation of basic biology to their potential for application in the clinical and preventive medicine fields.(AU)

Stresses can be caused by multiple biotic and abiotic factors and their effects can affect both the biology and the immune system of insects. American cockroach - Periplaneta americana (Linnaeus, 1758) (Blattaria: Blattidae) -besides being an excellent model species, has great medical importance because it can act as a mechanical vector of several pathogens. This study aimed to evaluate the influence of starvation, dehydration and both stresses on weight, and total and differential haemocyte count in P. americana adults. Each specimen was isolated in glass flasks containing or not food and/or water. They were weighed periodically. Another group received water for 24 h after the end of stress period. In the immunologic bioassay, we counted their haemocytes after the final weighing. All stresses reduced the insect weight, especially when the stresses were combined. Females of the control group gained weight and males had it unaltered. Different stress conditions and time did not influence on total haemocyte count. Insects without food and water had the proportion of prohaemocytes increased and plasmatocytes decreased. This study can serve as a basis of further studies of bioecology, behaviour and the ability of resisting insecticides, besides serving as a model to studies in other insect species.(AU)

The objective of this study was to evaluate the effect of digestible methionine and cystine (Met + Cys) levels on the hematological and serum biochemical parameters of broiler chickens during the initial and growth stages. For this, 1,800 male chicks of the Coob 500 strain were used, with 900 chicks in the initial phase (1 to 21 days old) and 900 chicks in the growth phase (22 to 42 days old), distributed in a completely randomized design of five treatments with six replicates of 30 birds. The treatments consisted of 0.545, 0.616, 0.711, 0.782, and 0.853%; and 0.514, 0.571, 0.647, 0.704, and 0.761% digestible Met + Cys for “1 to 21” and “22 to 42” days of breeding, respectively. Results showed that digestible Met + Cys levels in broiler feed altered some hematological parameters (erythrocyte, hematocrit hemoglobin, total leukocytes, heterophile: lymphocyte) and serum biochemistry (uric acid, PST, total LDL, and TG). The digestible Met + Cys levels in the diet of broilers affected the hematological parameters and serum biochemistry, especially at higher levels. From the inclusion level 0.761 of Met + Cist in the broiler diet, red blood cells, hemoglobin and hematocrit changes begin to appear.(AU)

This study investigated the effects of dietary xylo-oligosaccharide (XOS) on growth performance, serum biochemical parameters, antioxidant function, and immunological function of nursery piglets. In total, three groups including 72 nursery piglets were designed and fed one of three diets: a control basal diet, basal diet supplemented with 0.2% ZnO, or basal diet supplemented with 0.04% XOS, for 28 days. Compared with the control group, the XOS group significantly increased the final body weight and average daily weight gain. No significant differences were found about these parameters between the control and ZnO groups. Compared with the control group, the ZnO group showed no changes in the serum content of total protein (TP), albumin (ALB), albumin:globulin (ALB:GLB), aspartate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen (BUN), glucose, triglyceride, total cholesterol (TC), or in the serum activity of amylase and alkaline phosphatase. However, in the XOS group, serum glucose content increased and blood urea nitrogen and triglyceride content decreased significantly. Compared with the control group, dietary supplementation with XOS significantly increased the serum activity of total antioxygenic capacity, superoxide dismutase, and catalase and decreased the serum activity of malondialdehyde. At the same time, serum IgG content in XOS group was significantly higher than that in control group. From the current study, supplementation of 0.04% XOS in the diet could improve the antioxidant and immune function of piglets, promotes nitrogen deposition, and accelerates lipid and glucose metabolism, thereby improving piglet growth performance.(AU)

The objective of this study was to evaluate the effects of injectable trace minerals (ITM) on growth, health, antioxidant enzyme activity, and immune system of newborn Boer kids. Newborn kids (n = 125) were assigned to one of two treatments: injection (0.1 mL 4.5 kg-1) of saline or ITM. Injectable trace minerals had 60, 10, 5, and 15 mg mL-1 of Zn, Mn, Se, and Cu, respectively. Kids were evaluated daily for the presence of diarrhea and weighted on d 0, 28, and 56. Blood samples were obtained on d 0, 3, 7, 14, 28, and 56. The ITM injection increased the plasma concentration of superoxide dismutase (d 14), glutathione peroxidase (d 3 and 7), and blood platelets (d 7) compared with saline injection. Kids receiving ITM showed greater amount of blood eosinophils and less mean corpuscular hemoglobin (MCH; d 3) and mean corpuscular hemoglobin concentration (MCHC) compared with kids receiving saline injection. The ITM injection did not affect other components of leukogram and erytogram. The ITM-injected kids tended to have less diarrhea incidence compared with saline-injected kids (20.7 vs. 34.8±7.10% respectively). The ITM injection did not affect mortality rate and growth. Therefore, a single ITM injection administered to newborn Boer kids increases the plasma concentration of antioxidant enzymes, platelets, and eosinophils, reduces MCH, MCHC, and tends to reduce the incidence of diarrhea, but does not affect mortality and growth.(AU)

The immune system is mainly responsible for protecting the organism against agents that may interfere in its homeostasis. Thus, understand how this system develops and operates is very important, for create new therapies to assist this system in its operation, such as its failure. In domestic dogs, few studies show how actually occurs the development, maturation and functioning of the immune system. Therefore, this study demonstrates the development and possible activation of it on dog fetus from late gestational period by in situ and microscopic analyzes.

Much of what we know about the involvement of the immune system in periovulatory follicle differentiation, ovulation and subsequent formation of the corpus luteum in cattle is drawn from the findings of studies in several mammalian livestock species. By integrating published histological data from cattle, sheep and pigs and referring back to the more comprehensive knowledge bank that exists for mouse and humans we can sketch out the key cells of the immune system and the cytokines and growth factors that they produce that are involved in follicle differentiation and luteinization, ovulation and early follicle development. These contributions are reviewed and the key findings, discussed.
