In bridging the knowledge gap on stress physiology of Nigerian indigenous chickens, this study investigated the effect of exogenous corticosterone (eCORT) as stress inducing agent on the testicular function and mating behavior of Nigerian indigenous cocks. Twenty-four (24) cocks and one hundred and forty four (144) hens (mating ratio of 1 cock: 6 hens) were grouped into four and assigned to each of the four eCORT treatments (0, 2, 4 and 6 mgeCORT/KgBW) daily for 14 days. Semen samples were collected on days 0, 7 and 14 and analyzed for semen volume (SV), progressive sperm motility (PSM), membrane integrity (MI) and sperm abnormality (SA). Mating behaviors were monitored on days 3, 5 and 8. Blood samples, for hormonal (Luteinizing Hormone (LH), Follicle Stimulating Hormone (FSH) Testosterone (TEST) and stress analysis (heterophil/lymphocyte ratio, H/L) were collected from brachial vein on days 7 and 14. On day 15, cocks were euthanized and testes harvested for histomorphometry. Data were analyzed using multivariate analysis, oneway ANOVA and Kruskal-Wallis tests all in SPSS 23. Administration of 4 mgeCORT/KgBW declined (P0.05) influence on the mating behaviors, H/L ratio, FSH and TEST. However, 2 mgeCORT/KgBW enhanced LH levels. Administration of eCORT did not affect the testicular epithelial height and seminiferous tubular diameter. In conclusion, optimal stress induced by eCORT impaired semen quality but with less impact on reproductive hormones, H/L and mating behaviors of intensively raised Nigerian indigenous cocks.(AU)

Common Palm Civet (Paradoxurus hermaphroditus) is a rare mammal appearing in South and Southeast Asia, first described in 1777. An expensive coffee is made of beans eaten and rejected by civets. In Vietnam, the animals are increasingly captured and kept captive to sell these beans, which has adversely affected their population. Because of the detrimental effects of stress, studying stress responses is important for this wildlife conservation. The stress response of the Common Palm Civet was investigated through the concentration of glucocorticoid hormones in feces under different conditions such as translocation and forced-pairing during the breeding season, among others. This is a non-invasive method for measuring glucocorticoid hormones in captive Common Palm Civet in Vietnam. Our results validated that fecal glucocorticoid metabolites (FGM) accurately reflect circulating glucocorticoid stress hormones in Common Palm Civet. FGM increased under adrenocorticotropic hormone stimulation and during the breeding season (April and NovemberDecember). FGM during estrus and parturition was high as well in female civets. Stress response was also observed in case of forced-pairing or residence change. These data could be useful for research and the conservation of this species.

The aim of this study was to compare the effects of beak trimming by hot blade or infrared radiation on production indicators and plasma levels of corticosterone in pullets. During the rearing phase, an entirely random delineation was used a 2 x 3 factorial design (two different techniques for beak trimming x 3 strains), a total of six treatments with six repetitions of 20 hens. In the growing phase, the previous treatments were divided into 12, with eight repetitions with seven hens. These treatments were then distributed in a factorial design of 2 x 3 x 2 (two methodologies for beak trimming during rearing phase x three strains x with or without beak trimming at ten weeks of age). In the rearing phase the birds treated by infrared radiation had a lower total consumption and a longer beak length. In the growing phase, the execution of beak trimming at ten weeks of age, led to a lower total consumption and a lower body weight, but a better food conversion, when compared to those hens that did not have beak treatment. Beak trimming at ten weeks did not change the plasmatic levels of corticosterone.(AU)

Dexamethasone (DEX) is a corticoid hormone that is experimentally used to mimic the effects of increased levels of endogenous corticosterone observed during the stress response. Currently, stress is considered one of the major predisposing factors for diseases in the poultry industry. The aim of this study was to analyze the effects of DEX and/or of a 20-fold coccidial vaccine dose on leukocyte phenotypes in the spleen and cecal tonsils of chickens. Twenty specific-pathogen-free (SPF) Leghorn chickens were divided into four groups: a non-treated group (NT), a DEX-treated group (Dex), a vaccinated group (V) and a DEX-treated+vaccinated group (Dex+V). On experimental day (ED) 42, each bird in the vaccinated groups received a anti-coccidial vaccine. DEX was injected in the birds of the Dex and Dex+V groups (0.9 mg/kg) onED42 and ED45. The immunophenotyping was performed by flow cytometry analysis of splenocytes and cecal tonsils cells onED48. DEX treatment per se was unable to change CD4+CD8+, CD4+CD8+ and CD4-CD8+ populations with TCRgd or CD28 in the spleen, or macrophages and T lymphocytes in the cecal tonsils. V group birds presented higher numbers of splenic macrophages compared with those measured in the Dex+V group. The number of CD4+CD25+ cells in the spleen of birds of the V group was higher than those measured in the other experimental groups. Our data suggest that CD4+CD25+ cells and macrophages might be influenced by DEX treatment in spleen, but not in the cecal tonsils of chickens inoculated with Eimeria.

Embryo thermal stimulation has been studied as a means to promote epigenetic changes and to improve broiler health and performance. This study aimed at evaluating immunoglobulin (IgM, IgG, and IgA) titers, serum corticosterone levels, and organ integrity of day-old male Cobb(r) and Ross(r) broilers. Embryos were submitted to hot and cold stimuli (thermal stimulation) on days 14-18 of incubation. The thermal manipulation treatments did not affect immunoglobulin titers of 1 day-old Cobb(r) or Ross(r) chicks, and caused severe bursal lesions (scores 3 and 4) in both strains. However, serum corticosterone levels of Cobb(r) chicks were higher when embryos were submitted to hot stimulus (+1.39ºC) compared with cold stimulus (+36 ºC). Serum corticosterone levels of Ross(r) chicks were not affected by embryo temperature manipulation. The only effect of broiler breeder age was observed on IgM titers of Cobb(r) chicks, which were higher in chicks from 33-wk-old breeders. Further studies are needed to detect the optimal period of embryo thermal stimulation.

The Swiss Webster mice of the strain in animal facilities are individually and interesting and complex social behavior. The aggressiveness of adult male has varying intensity and is associated with hierarchical dominance. In approximately 5% of the animals we observed an exacerbation of aggressive behavior and the presence of severe damage to the other components of the group. Our goal in this work is to search to correlate the level of plasma corticosterone and their phenotypic behavioral categories: Har - the absence of aggressiveness; AgD - attacked and AgR animals - aggressive individuals, compared with the previous values to the regrouping (BfR). Our results demonstrate that the regrouping of adult male individuals is a stressor that promotes and decreased weight gain in categories and Har AgD and weight loss in individuals AgR. Furthermore, the values of plasma corticosterone rises in the first two categories, but is maintained in the category AgR. So, we believe that the exacerbation and / or uncontrolled aggression in some individuals is related to a Posttraumatic Stress Disorder (PTSD) characterized by sustained activation of the Hypothalamic-Pituitary-Adrenal (HPA) and the inability of the animal in dealing with new stress situations.

PURPOSE:To investigate the structural and functional changes induced by corticosterone (CORT) in the ventral prostrate (VP) of rats in order to study chronic stress effects in the prepubertal phase.METHODS:Wistar rats received daily saline or CORT injections during the pubertal period from the 5th to 25th day of postnatal life. The animals were distributed into four groups: 1 - Control (n=5); 2 - Control 99mTc-P (n=5); 3 - Treated with CORT (n=14); 4 - Treated with CORT and 99mTc-P (n=10). All rats were sacrificed at two months of age. Technical tissue uptakes of 99mTc-P were used to evaluate the functional and stereological methods for morphological analysis.RESULTS:Acini distribution in the group treated with CORT differed significantly (p 0.0001) from the control. The control group's epithelial average height (10.01±0.24 microns) was statistically significant (p 0.0001) from rats treated with CORT (19.27±0.73microns). The collagen distribution was lower in the treated group (2.79%) when compared to control (3.97%). The radioactivity percentage in the groups marked with 99mTc-P (%Ati/g) did not demonstrate a statistically significant difference (p=0.285897).CONCLUSION:Chronic administration of corticosterone in prepubertal rats causes changes in their acinar structure and their ventral prostate stroma, indicating possible deleterious effects of this hormone.

The aim of this study was to evaluate alterations to the physiological profile of cortisol in pro-metamorphose phase tadpoles of Lithobates catesbeianus exposed to hypoxia stressor in a capture experiment and in a crowding experiment. The capture study was performed by the treatments: stress due to individual capture with a hand net, stress due to batch capture with a hand net and stress due to capture by emptying. Three simultaneous replicates was done witch 12 animals were sampled (6 normoxia - immediately blood collection) and 6 hypoxia - blood collection after 15 min of air exposition) in two collection times with 5 days by intervals. The crowding study was performed by the treatments 1 tadpole L-1, 5 tadpoles L-1 and 10 tadpoles L-1. Three simultaneous replicates was done witch 8 animals (4 normoxia and 4 hypoxia) were sampled in the zero moment (ZM) - blood collection before the experiment, 6 animals/treatment (3 normoxia and 3 hypoxia) to 4 and 8 days and 18 animals/treatment (9 normoxia and 9 hypoxia) to 12 days. The average values to plasmatic cortisol varying from 1.7 to 5.1ng mL-1 (capture study) and 1.0 to 4.2ng mL-1 (crowding study). It concludes that the biomarker tested (cortisol) showed no alterations front of the stressor used. Alternatively, a larger response pattern to these stimuli may have been expressed in another level of an unmeasured hormone (corticosterone). And the bullfrog has great ability to adapt to different management compared to other aquatic organisms, which demonstrates the plasticity of these animals.(AU)

The objectives of the present study were to evaluate fecal concentrations of metabolites of glucocorticoids, measured by enzyme immunoassay with a cortisol antibody and by radioimmunoassay with a corticosterone antibody, and progesterone by radioimmunoassay with a progesterone antibody in blue-fronted parrot (Amazona aestiva) after ACTH challenge. The adrenal stimulation with ACTH (25 UI/animal) resulted in an increase of fecal glucocorticoids metabolites concentration, but it did not affect the concentrations of fecal progesterone metabolites. Although there were no synchronized peaks of glucocorticoid metabolites excretion measured by enzyme immunoassay and radioimmunoassay, there were two peaks of excretion, one at 2-4 hours and other at 8-10 hours. Despite the occurrence of peaks, the analysis of fecal glucocorticoids metabolites and progesterone metabolites showed no effect of group (control and treatment), moment (hours of sampling) and sex.

The mice used in animal facilities belong to the family Muridae, subfamily Murinae, order Rodentia, genus Mus and species Mus musculus. They are social and territorial animals. Several lineages developed in the laboratory show aggressive behavior similar to that of wild mice. The aim of this study was characterize the hierarchical structure, body characteristics and hormonal levels in dominant, subordinate and non-aggressive mice. The model established for spontaneous aggression (MSA) evaluates the pattern of aggressive behavior (PBA) and categorizes the animals as follows: LAg - individuals with low aggressive behavior, HAg – dominant (highly aggressive) and Sb - subordinate. The regrouping of adult male mice produces substantial stress and influences the formation of their hierarchical structure. Behavioral comparisons before regrouping (BfR) and after regrouping showed that the percentage of body weight lost in mice was dependent of the specific categories as follows: LAg - 68.1%, HAg - 86.9% and Sb - 90.5%; the average corticosterone levels by category were BfR: 43.5±17.5, LAg: 177.0±40.4, HAg: 72.8±23.8 and Sb: 136.4±51.2 ng/mL. Dominant mice showed differences in body characteristics (primarily the body/tail relationship) relative to subordinate mice. Two additional hierarchical positions were observed: “neutral individual” (without aggressive behavior) and “subordinate target” (an animal that receives approximately 30% of the total number of attacks made by dominant animals).

The intensification of animal production systems presents a potential impact on the welfare of animals. The objective of this work was to assess the welfare of quail Coturnixcoturnix japonicain two maintenance systems: battery cages (BC),and enriched aviary (EA),with saw-dust bedding, sand-bathing area and nests. The experiment procedure involved eight animals per holding area and four repetitions per treatment, an overall of 64 quails. Welfare was assessed through behavioral freedom, sanitary freedom (feather condition and injuries), blood analyses and glicocorticoid metabolites measurement in droppings. Results are presented in the BC order, followed by EA. Water drinking behavior and agonistic behavior were different between treatments (P<0.05). Feather condition was adequate in both treatments, except for the head in BC quails. Blood data were statistically different forred blood cells, hematocrit, hemoglobin, blood proteins, eosinophils, heterophils, lymphocytes and heterophil: lymphocyte ratio. Glicocorticoid metabolites levels were significantly different between maintenance systems. The behavioral and physiological welfare indicators showed higher welfare degree for quails in enriched aviary as compared to battery cages system.(AU)

The effects of feeding levels of Valeriana officinalis on behavior and physiologic parameters of female quails in the rearing periods were evaluated. In the rearing phase, 192 quails, from 28 to 42 days old, average weight 121,25±6,96g, were lodged to cages and allotted to randomized blocks design, with four treatments (control; 250; 500 and 750mg of valeriana/kg of ration) with eight replicates and six birds per cage. Behavior (body injury, tonic immobility and focal observation) and physiological parameters (heterophil/lymphocyte ratio and corticosterone blood plasma) was evaluated. In the rearing periods the tested valeriana levels did not affect behavior and physiological parameters of quails.(AU)

Stress is a reality in the world poultry production. Environmental stressors impair both welfare, performance parameters and immunity in broiler chickens. Heat stress decreases macrophage activity in broiler chickens and many environmental stressors that impact animal production increases animal's susceptibility to diseases. Samonella spp is one of the most endemic zoonotic diseases of the world, inducing more than 1 billion infection cases per year. In this way, we studied the effects of 31±1°C heat stress on performance parameters, immunity, bacteria invasion and intestinal integrity in broiler chickens experimentally infected with Salmonella enteritidis; the data were discussed under a neuroimmune perspective. The broiler chickens were divided into four different groups: 1) Control group (C); 2) 31±1 °C heat stressed group (HS31°C); 3) Control group infected with Salmonella enteritidis Positive control (PC) and; 4) 31±1 °C heat stressed and Salmonella enteritidis infected group (PHS31°C). We showed the heat stress applied in the course of Salmonella enteritidis infection (PHS31°C group) decreased poultry performance parameters; specifically, it decreased the body weight gain, the feed intake and the food conversion; 2) decresead INF-γ and IgA plasmatic levels; 3) decreased the mRNA expression of IL-6 and IL- 12 in spleen and the mRNA expression of IL1-β, IL-10 and TGF-β in cecal tonsil; 4) decreased the mRNA expression of AvBD-4 and AvBD-6 in cecal tonsil and; 5) decreased the mRNA expression of TLR-2 in spleen and cecal tonsil. We also observed 6) an increase in corticosterone serum levels in the animals of the HS31°C and the PHS31°C groups and, 7) more severe intestinal inflamation produced by Salmonella enteritidis in heat stressed chickens, characterized by a moderate enteritis throughout all the small intestine mucosa (PHS31°C group). Finally, 8) we showed that the heat stress increased splenic Salmonella enteritides invasion in PHS31°C broiler chickens; we also observed the presence of Salmonella in the bone marrow of stressed and infected broiler chickens. These data suggest that heat stress and Salmonella infection working together impair chicken's performance parameters, intestinal integrity and immunity (specially the macrophage activity), increasing ate the same time splenic and bone marrow Salmonella enteritidis invasion. Thus, heat stress could have impared the intestinal immunity barrier quality, via HPA axis activation and corticosterone serum levels release, decreasing the inate immunity and, providing pathogenic bacteria migration through the intestinal mucosa for spleen and bone marrow of the heat stressed chickens.
