Resumo

The aim of this study was to perform the molecular characterization of conserved and variable regions of feline calicivirus capsid genome in order to investigate the molecular diversity of variants in Brazilian cat population. Twenty-six conjunctival samples from cats living in five public short-term animal shelters and three multicat life-long households were analyzed. Fifteen cats had conjunctivitis, three had oral ulceration, eight had respiratory signs (cough, sneeze and nasal discharge) and nine were asymptomatic. Feline calicivirus were isolated in CRFK cells and characterized by reverse transcription PCR target to both conserved and variable regions of open reading frame 2. The amplicons obtained were sequenced. A phylogenetic analysis along with most of the prototypes available in GenBank database and an amino acid analysis were performed. Phylogenetic analysis based on both conserved and variable region revealed two clusters with an aLTR value of 1.00 and 0.98 respectively and the variants from this study belong to feline calicivirus genogroup I. No association between geographical distribution and/or clinical signs and clustering in phylogenetic tree was observed. The variants circulating in public short-term animal shelter demonstrated a high variability because of the relatively rapid turnover of carrier cats constantly introduced of multiple viruses into this location over time.(AU)

Resumo

Rotaviruses are etiological agents of diarrhea both in humans and in several animal species. Data on avian Group D rotaviruses (RVD) are scarce, especially in Brazil. We detected RVD in 4 pools of intestinal contents of broilers, layer and broiler breeders out of a total of 111 pools from 8 Brazilian states, representing an occurrence of 3.6%, by a specific RVD RT-PCR targeting the VP6 gene. Phylogenetic tree confirmed that the Brazilian strains belong to group D and 3 of the sequences were identical in terms of amino acid whereas one showed 99.5% identity with the others. The sequences described in this study are similar to other sequences previously detected in Brazil, confirming the conserved nature of the VP6 protein.(AU)


Rotavírus são agentes etiológicos de diarreia tanto em humanos como em várias espécies animais. Dados sobre rotavírus do grupo D (RVD) em aves são escassos, especialmente no Brasil. Nós detectamos RVD em 4 pools de conteúdo intestinal de frango de corte, poedeiras e matrizes de um total de 111 pools originários de 8 estados brasileiros, representando uma ocorrência de 3,6% a partir de uma RT-PCR específica para RVD, tendo como alvo o gene VP6. A árvore filogenética confirmou que as amostras brasileiras pertencem ao grupo D e três das sequências obtidas foram idênticas em termos de aminoácidos enquanto uma apresentou 99,5% de identidade com as demais. As sequências aqui definidas são semelhantes a outras sequências previamente definidas no Brasil, confirmando a natureza conservada da proteína VP6.(AU)

Background: The platelet-derived growth factor (PDGF) is expressed in a wide variety of cell types, exerts a potent mitogenic role and acts on the growth, differentiation and cell chemotaxis. Studies have shown that during folliculogenesis, PDGF and their receptors are expressed in oocytes, granulosa cells and thecal cells of ovarian follicles at different developmental stages in several species. Although exist many information about its expression sites, as well as about its action in different cells types, the role of PDGF on ovarian folliculogenesis remains understudied. Thus, this article aims to review issues related to PDGF, suggesting the involvement of this mitogenic factor during follicular development. Review: Along this work, it was shown aspects related to structural characterization of PDGF and its receptors, as well as PDGF expression in different cells types, emphasizing its importance to follicular development. PDGF family is composed by four polypeptide chains (each encoded by a different gene), which are synthesized in the form of inactive pro-proteins. After a proteolytic processing, these chains undergo homo or heterodimerization, resulting in five isoforms (PDGF-AA, -BB, -AB, -CC e -DD). The cellular effects of these different PDGF isoforms are mediated by binding, with different specificities, to three transmembrane receptors isoforms of type tyrosine kinase generated by the association of subunits alpha e beta (PDGFR- alphaalpha, - alphabeta, - betabeta)...

Resumo

The aim of this study was to assess the occurrence of Cryptosporidium in domestic animals in rural properties surrounding rain forest fragments within the municipality of Teodoro Sampaio, southeastern Brazil. Conventional sucrose lotation method followed by molecular characterization of the parasites by sequencing PCR products amplified from SSU rRNA gene were used. Stool samples were collected from domestic animals raised as pets and livestock in all rural properties surrounding three forest fragments. Samples from cattle (197), equine (63), pigs (25), sheep (11), and dogs (28) were collected from 98 rural properties. he frequency of occurrence of Cryptosporidium within each animal species was 3.0% (6/197) among cattle and 10.7% (3/28) among dogs. Cryptosporidium was not detected in stool samples from equine, sheep, and pigs. All sequences obtained from the six samples of calves showed molecular identity with Cryptosporidium andersoni while all sequences from dog samples were similar to C. canis. The frequency of occurrence of Cryptosporidium in these domestic animal species was low. The absence of C. parvum in the present study suggests that the zoonotic cycle of cryptosporidiosis may not be relevant in the region studied. he presence of Cryptosporidium species seldom described in humans may be, otherwise, important for the wild fauna as these animals are a source of infection and dissemination of this protozoan to other animal species. The impact and magnitude of infection by C. andersoni in wild ruminants and C. canis in wild canids have to be assessed in future studies to better understand the actual importance of these species in this region. O objetivo deste estudo foi avaliar a ocorrência de Cryptosporidium, em animais domésticos de propriedades rurais ao redor de fragmentos de mata Atlântica de interior no município de Teodoro Sampaio, por exame convencional de flutuação em solução de sacarose, seguido de caracterização molecular dos parasitas através do sequenciamento dos produtos amplificados na PCR do gene SSU rRNA. Foram coletadas amostras fecais de animais domésticos criados para subsistência e estimação nas propriedades rurais do entorno de três fragmentos florestais. Amostras de bovinos (197), equinos (63), suínos (25), ovinos (11) e cães (28) foram coletadas de 98 propriedades rurais. A ocorrência de Cryptosporidium para a espécie bovina foi de 3,0% (6/197); para os cães, de 10,7% (3/28); e para os demais animais os resultados foram negativos. Todas as sequências obtidas das seis amostras de bovinos apresentaram identidade molecular com Cryptosporidium andersoni, enquanto as sequências oriundas de amostras de fezes de cães revelaram-se similares ao C. canis. A ocorrência do Cryptosporidium entre os animais estudados foi baixa. Diante dos resultados do presente estudo, o ciclo zoonótico da criptosporidiose parece ter menos importância nesta região. A presença de espécies de Cryptosporidium ainda pouco relatadas em humanos pode ser, por outro lado, importante para a fauna silvestre, uma vez que estes animais podem ser considerados como uma fonte de infecção e disseminação deste protozoário. O impacto e a magnitude da infecção de C. andersoni, em ruminantes selvagens, e de C. canis, em cães silvestres, deve ser avaliado em estudos futuros, com intuito de verificar a real importância dessas espécies nesta região.(AU)

Resumo

The occurrence of Cryptosporidium in fecal samples of lambs was determined in order to test correlations among the type of animal raising, feces consistence, index of oocysts elimination, animalage, and molecular characterization of species and genotypes involved in this parasitism. A total of 460 fecal samples were collected from lambs during the first 30 days of life, in various screeds of 21 farms.All samples were analyzed using malachite green negative coloration stain under microscopy. For molecular characterization of Cryptosporidium, on positive samples under microscopy, a nested PCR protocol was used for amplification of fragmentts from 18S rRNA gene or actin gene. It was found 6.7%of animals expeling oocysts of Cryptosporidium in feces of eight farms, without statistics relation among the analyzed indexes. The species and genotype involved were C. parvum type A 18S rRNA, C. parvum type B 18S rRNA and cervid genotype, which present a zoonotic potential of disease.(AU)

Resumo

A região p17-p24 do gene gag de 10 amostras do vírus da imunodeficiência felina detectadas no estado de Minas Gerais (Brasil) foi seqüenciada com o objetivo de determinar a sua classificação molecular e a sua relação com seqüências de amostras previamente descritas. As amostras pertenciam ao subtipo B, entretanto foi possível observar que a maioria delas encontra-se em um subgrupo dentro do subtipo B, o que indica presença de um possível ancestral comum entre elas.(AU)
