Background: Staphylococcus spp. are the most frequently isolated microorganisms in mastitis cases of small ruminants. The virulence factors of Staphylococcus spp. are critical in the treatment. Therefore, preventive medicine and mastitis control programs, especially herd vaccinations are of great importance in the prevention of mastitis. However, it is not always easy to obtain these vaccines under field conditions. This study, it was aimed to compare the effects of different commercial Staphylococcus spp. vaccines licensed for bovines and species-specific mastitis vaccines on mastitis rates and somatic cell count (SCC) on Saanen goats on field conditions. Materials, Methods & Results: The animal material consisted of 115 (230 udder halves) nulliparous Saanen goats. Goats were randomly grouped as bovine vaccine 1 (BV1 , n = 58), bovine vaccine 2 (BV2 , n = 58), small ruminant vaccine (SRV, n = 56), and control (n = 56). Vaccines were administered to goats in 2 doses according to the label regimen. First milk samples were collected between 0-5 days in milk (DIM) for microbiological analysis and 25-35 DIM for SCC. The other milk samples were collected at 25-35 (1st month) DIM, 60-65 (2nd month) DIM, 85-95 (3rd month) DIM, 115-125 (4th month) DIM, 145-155 (5th month) DIM for microbiological analysis and SCC. Non-aureus staphylococci (NAS) and Staphylococcus aureus were the most frequently isolated microorganism. It was found that the total mastitis rate decreased in vaccine groups compared to the control group. A significant difference was found only in the BV2 and SRV groups. The significant difference in S. aureus infection was found only in the SRV group. Mastitis vaccines used in this study decreased the NAS mastitis rate, but no significant difference was observed. It was found that the clinical mastitis incidence decreased in all vaccine groups compared to the control group, and a significant difference was found between the BV2 and SRV groups compared to the control group (P < 0.05). Somatic cell count was lower in the SRV and BV2 groups compared to the control group (P < 0.05). Discussion: In this study, compatible with the previous reports NAS and S. aureus were the most frequently isolated microorganism. The diversity of virulence factors of Staphylococcus spp. also plays an important role in its high incidence. In some countries, mastitis vaccines used in cows are also administered to small ruminants for reducing infection rates. Similarly, in this study, it was found that the mastitis rate decreased in all vaccine groups compared to the control group. A significant difference was found only in the BV2 and SRV groups. It is thought that the reason for the statistical difference may be due to the biofilm antigen in the BV2 and SRV. In addition, J5 strain in the BV2 is estimated to be effective in reducing the prevalence of gram-negative mastitis. It was observed that the infection rates decreased in the vaccine groups, especially due to S. aureus and NAS. Spontaneous treatment rates were very close to each other between the groups. The reason for the high rate of spontaneous treatment in this study can be explained by the fact that the animals were young and in their 1st lactation. SCC was lower in all vaccine groups compared to the control group. This situation is associated with the decrease in infection rates related to the use of vaccines. It was observed that SCC was lower in the vaccine groups. In addition, SCC was found to be lower in this study compared to similar studies. However, it is evident that the use of species-specific vaccines in the SRV group significantly reduced the rates of total S. aureus mastitis, subclinical NAS mastitis, and new infections by NAS compared to other vaccines. Furthermore, the species-specific vaccine significantly increased the rate of spontaneous treatment for S. aureus mastitis.

Mastitis causes significant economic losses to the dairy cattle industry. The present study aimed to evaluate the antibacterial properties of 39 heterocyclic derivatives (1,3-thiazoles and 4-thiazolidinones) against clinical mastitis isolates from dairy cows. Milk samples were collected from cows with clinical mastitis and the bacterial species were identified by PCR. Antibacterial activity was assessed using the broth microdilution method. First, 39 heterocyclic compounds were tested against four bacterial isolates (Staphylococcus aureus, Streptococcus agalactiae, Corynebacterium bovis and Escherichia coli) randomly chosen from those recovered from the milk samples (Study 1). Subsequently, the compounds with the strongest antibacterial activity were tested against all the bacterial isolates recovered from the milk samples (Study 2). 1,3-thiazoles showed the strongest antibacterial activity, specially compounds 30 and 38, which also showed bactericidal properties according to their minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values. Corynebacterium spp. and Enterobacteriaceae isolates were the most susceptible to compounds 30 and 38. Compounds 30 and 38 are promising targets for new antimicrobial agents.(AU)

O presente estudo teve como objetivo avaliar as propriedades antibacterianas de 39 derivados heterocíclicos (1,3-tiazóis e 4-tiazolidinonas) contra isolados clínicos de mastite em vacas leiteiras. Amostras de leite foram coletadas de vacas com mastite clínica e as espécies bacterianas isoladas foram identificadas por PCR. A atividade antibacteriana foi avaliada pelo método de microdiluição em caldo. Primeiramente, os 39 compostos heterocíclicos foram testados contra quatro isolados bacterianos (Staphylococcus aureus, Streptococcus agalactiae, Corynebacterium bovis e Escherichia coli) escolhidos aleatoriamente dentre os recuperados das amostras de leite (Estudo 1). Posteriormente, compostos com atividade antibacteriana mais forte foram testados contra todos os isolados bacterianos recuperados das amostras de leite (Estudo 2). Os compostos 1,3-tiazóis apresentaram a maior atividade antibacteriana, principalmente os compostos 30 e 38, que também apresentaram propriedades bactericidas de acordo com seus valores de concentração inibitória mínima (CIM) e concentração bactericida mínima (CBM). Os isolados Corynebacterium spp. e Enterobacteriaceae foram os mais suscetíveis aos compostos 30 e 38. Os compostos 30 e 38 mostraram-se promissores como novos agentes antimicrobianos.(AU)

O objetivo deste trabalho foi avaliar a transferência de imunidade passiva de cabras, que pariram com mastite, para seus respectivos cabritos. Os animais foram distribuídos em dois grupos, a saber: grupo 1 (GI), constituído por cabritos, filhos de cabras sem isolamento microbiológico em ambas as glândulas mamárias, e grupo 2 (GII), composto por cabritos, filhos de cabras com resultado positivo à lactocultura, em pelo menos uma das glândulas mamárias. Foram coletadas amostras de colostro e sangue à parição, bem como às 24 e às 48 horas após o parto/nascimento. O diagnóstico e o monitoramento da mastite nos animais foram realizados por meio do California Mastitis Test (CMT), contagem de células somáticas e isolamento microbiológico. A proteína total foi mensurada pelo método do biureto, e as concentrações de imunoglobulina A (IgA), imunoglobulina G (IgG), transferrina, albumina e haptoglobina por meio da eletrofoerese em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE). Os agentes mais isolados na cultura microbiológica foram os Staphylococcus coagulase negativa. Não houve diferença significativa (P<0,05) entre os valores médios de imunoglobulina G (IgG) nos cabritos provenientes de cabras com mastite quando comparados aos recém-nascidos oriundos de cabras livres de infecções intramamárias. Da mesma forma, a atividade de gamaglutamiltransferase (GGT) não mostrou diferença entre os grupos em todos os momentos avaliados. A ingestão de colostro decorrente de cabras com mastite não causou falha na transferência de imunidade passiva nos respectivos conceptos.(AU)
The aim of this study was to evaluate the transfer of passive immunity goats kidded with mastitis to their kids. The animals were divided into two groups, namely: Group 1 (GI) containing kids, sons of goats without microbiological isolation in both mammary glands, and Group 2 (GII), composed of kids, sons of goats with positive result to lactoculture in at least one of mammary glands. Colostrum samples and blood were collected after delivery, 24 and 48 hours after delivery / birth. The diagnosis and monitoring of mastitis in animals were performed using the California Mastitis Test (CMT), somatic cell count and microbiological isolation. Total protein was measured by the biuret method, and the concentrations of immunoglobulin A (IgA), immunoglobulin G (IgG), transferrin, albumin and haptoglobin through eletrofoerese polyacrylamide gel containing sodium dodecyl sulfate (SDS-PAGE). The agents most isolated in microbiological culture were coagulase-negative Staphylococcus. There was no significant difference (p <0.05) between the acquisition of immunoglobulin G (IgG) in goats from goats with mastitis compared to infants originating free goat mammary infections. Similarly the gamma glutamyl transferase (GGT) was equal in the comparison between groups in all evaluated moments. The colostrum intake resulting from goats with mastitis caused no failure in the passive transfer of immunity in their fetuses.(AU)
