Resumo

Abstract Persea lingue Ness is a tree species that lives mainly in temperate forests of south-central Chile. Its leaves are used in ethnomedicine, the fruit is a drupe similar to that of the avocado and has not been studied. The aim of this study was to determine the cytotoxicity in leukemia cell and antibacterial activity, along with some chemical content characteristics of P. lingue fruit and leaf extracts. The antibacterial activity was determined by the inhibition of bacterial growth in liquid medium assay against Gram-positive and Gram-negative bacteria. The leukemia cell lines Kasumi-1 and Jurkat were used to evaluate the cytotoxic activity by using propidium iodide and AlamarBlue assays. Total phenolic, flavonoid, condensed tannin, alkaloid and lipid contents were evaluated in the fruit and in the leaf extracts. The antioxidant activity of both extracts were also elavaluated. Leaf extract presented the highest content of total phenols, condensed tannins and flavonoids, and also the highest antioxidant activity. While the fruit extract has a higher amount of lipids and alkaloids and the high antibacterial activity against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus megaterium and Micrococcus luteus. The leaf extract only showed activity against M. luteus. Concerning the cytotoxic activity, only the fruit extract showed cytotoxicity against the cell lines Jurkat and Kasumi-1. P. lingue fruit extract is a potential source of biologically active molecules for the development of new drugs to be used in some types of leukemia, as well as antibacterial agent.


Resumo Persea lingue Ness é uma árvore que vive principalmente na floresta temperada do centro-sul do Chile. As folhas são usadas na etnomedicina. O fruto é uma drupa similar ao abacate e que nunca foi pesquisada anteriormente. O objetivo deste estudo foi o de avaliar a citotoxicidade em células leucêmicas e as atividades antibacterianas, assim como algumas características químicas do extrato de fruto e da folha do P. lingue. As atividades antibacterianas foram determinadas pelo método da inibição do crescimento bacteriano em meio líquido empregando-se bactérias Gram-positivas e Gram-negativas. As linhagens celulares leucêmicas, Kasumi-1 e Jurkat foram usadas para avaliar a atividade citotóxica em ensaios empregando-se iodeto de propídio e AlamarBlue. Foram avaliados os teores totais de fenóis, flavonóides, taninos condensados, alcalóides e lipídeos presentes nos extratos das folhas e dos frutos. As atividades antioxidantes de ambos os extratos também foram avaliadas. O extrato das folhas foi o que apresentou o maior conteúdo de fenóis, taninos condensados e flavonóides totais e a maior atividade antioxidante. Já o extrato de fruto apresentou a maior quantidade de lipídios e alcaloides e a melhor atividade antibacteriana contra Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus megaterium e Micrococcus luteus. Já o extrato das folhas apresentou apenas atividade contra M. luteus. Em relação à atividade citotóxica, apenas o extrato do fruto apresentou citotoxicidade contra as linhagens celulares Jurkat e Kasumi-1. Em resumo, o extrato do fruto de P. lingue é uma potencial fonte de moléculas com atividade biológica para o desenvolvimento de novos fármacos a serem utilizados em alguns tipos de leucemia, bem como agente antibacteriano.

Abstract Transcription factors (TF) are a wide class of genes in plants, and these can regulate the expression of other genes in response to various environmental stresses (biotic and abiotic). In the current study, transcription factor activity in sugarcane was examined during cold stress. Initially, RNA transcript reads of two sugarcane cultivars (ROC22 and GT08-1108) under cold stress were downloaded from SRA NCBI database. The reads were aligned into a reference genome and the differential expression analyses were performed with the R/Bioconductor edgeR package. Based on our analyses in the ROC22 cultivar, 963 TF genes were significantly upregulated under cold stress among a total of 5649 upregulated genes, while 293 TF genes were downregulated among a total of 3,289 downregulated genes. In the GT08-1108 cultivar, 974 TF genes were identified among 5,649 upregulated genes and 283 TF genes were found among 3,289 downregulated genes. Most transcription factors were annotated with GO categories related to protein binding, transcription factor binding, DNA-sequence-specific binding, transcription factor complex, transcription factor activity in RNA polymerase II, the activity of nucleic acid binding transcription factor, transcription corepressor activity, sequence-specific regulatory region, the activity of transcription factor of RNA polymerase II, transcription factor cofactor activity, transcription factor activity from plastid promoter, transcription factor activity from RNA polymerase I promoter, polymerase II and RNA polymerase III. The findings of above results will help to identify differentially expressed transcription factors during cold stress. It also provides a comprehensive analysis of the regulation of the transcription activity of many genes. Therefore, this study provides the molecular basis for improving cold tolerance in sugarcane and other economically important grasses.


Resumo Fatores de transcrição (FT) são uma ampla classe de genes em plantas e podem regular a expressão de outros genes em resposta a vários estresses ambientais (estresses bióticos e abióticos). No presente estudo, a atividade do fator de transcrição na cana-de-açúcar foi examinada durante o estresse pelo frio. Inicialmente, as leituras de transcrição de RNA de duas cultivares de cana-de-açúcar (ROC22 e GT08-1108) sob estresse frio foram baixadas do banco de dados SRA NCBI. As leituras foram alinhadas em um genoma de referência e as análises de expressão diferencial foram realizadas com o pacote R / Bioconductor edgeR. Com base em nossas análises no cultivar ROC22, 963 genes TF foram significativamente regulados positivamente sob estresse pelo frio entre um total de 5.649 genes regulados positivamente, enquanto 293 genes TF foram regulados negativamente entre um total de 3.289 genes regulados negativamente. No cultivar GT08-1108, 974 genes TF foram identificados entre 5.649 genes regulados positivamente e 283 genes TF foram encontrados entre 3.289 genes regulados negativamente. Os fatores de transcrição, em sua maioria, foram anotados com categorias GO relacionadas à ligação de proteína, ligação de fator de transcrição, ligação específica de sequência de DNA, complexo de fator de transcrição, atividade de fator de transcrição em RNA polimerase II, atividade de fator de transcrição de ligação de ácido nucleico, atividade de corepressor de transcrição, sequência específica da região reguladora, atividade do fator de transcrição da RNA polimerase II, atividade do cofator do fator de transcrição, atividade do fator de transcrição do promotor do plastídio, atividade do fator de transcrição do promotor da RNA polimerase I, polimerase II e RNA polimerase III. As descobertas dos resultados acima ajudarão a identificar fatores de transcrição expressos diferencialmente durante o estresse pelo frio. Ele também fornece uma análise abrangente da regulação da atividade de transcrição de muitos genes. Portanto, este estudo fornece base molecular para melhorar a tolerância ao frio em cana-de-açúcar e outras gramíneas economicamente importantes.

Resumo

Bromelain is a set of proteolytic enzymes usually obtained from pineapple (Ananas comosus). Although bromelain has distinguished therapeutic properties, little is known about its proteolytic potential against opportunistic pathogens related to wound healing complications, such as Staphylococcus aureus. This study aimed toinvestigate the antibiofilm and antibacterial activity of bromelain in 43 clinical strains of S. aureusisolated from chronic wounds and blood cultures. Bromelain's activity against S. aureusbiofilm in vitrowas assessed by analyzing biofilm formation in cultures grownin the presence of 1% bromelain and biofilm destruction after the addition of 1% bromelain to mature biofilms. Proteinase K and sodium metaperiodate were also added to mature biofilms in parallel to compare their activity with that of bromelain and, together with exopolysaccharide and protein production rate assays, to determine the chemical composition of the biofilm extracellular matrix of selected strains of S. aureus. Bromelain was also evaluated for its DNase activity and impact on cellular hydrophobicity and auto-aggregation. Mueller-Hinton agar dilution was used to determine bromelain minimal inhibitory concentration (MIC). Biofilm assays showed that 1% bromelain significantly inhibits S. aureusbiofilm formation (p= 0.0157) by up to 4-fold and destroys its mature biofilms (p < 0.0001) by up to 6.4-fold, both compared to the control grown without bromelain. Biofilms of methicillin-resistant S. aureusstrains isolated from chronic wounds were the most affected by bromelain treatment. No antibacterial activity was detected with bromelain MIC assays and the proteolytic activity of bromelain was identified as the main antibiofilm mechanism of the enzyme, though its DNase activity may also contribute. The epithelial therapeutic properties of bromelain combined with its antibiofilm activity against S. aureusmake it a promising alternative to compose the therapeutic arsenal for the control of S. aureusbiofilms in the context of wound care.(AU)

Abstract The aim of this study was to evaluate the oxidative stress in ovaries and corpus luteum (CL) of Bos taurus indicus females and the oxidant effect of CL in ovarian tissues in regions near, intermediate, or distant from it. Ovaries (n=12) of Nelore heifers (n=6) were collected from a slaughterhouse and fragmented. Experiment 1, each ovary was obtained from three fragments, resulting in 18 fragments of ovaries with CL (OV+CL) and another 18 fragments of ovaries without CL (OV-CL). Three fragments were generated from CL, totaling 18 CL fragments. In experiment 2, the ovarian fragments were removed from specific regions near, intermediate, or distant from the CL. All the fragments were placed in Eppendorf-type microtubes (1 mL), kept in a thermal container at 4 ºC, and then stored in a -80 ºC freezer for analysis of oxidative stress (TBARS and NBT) and antioxidant potential (FRAP and ABTS). In the antioxidant activity analysis, luteal tissues showed more antioxidant activity than ovarian tissue (FRAP = P < 0.0001; ABTS = P < 0.02). In the oxidative stress analysis, CL had lower levels of reactive oxygen species (ROS; TBARS = P < 0.03; NBT = P < 0.0001) than ovarian tissues. There was no difference in antioxidant activity and oxidative stress between the fragments obtained from different regions (OV+CL versus OV-CL; P > 0.05). The presence of CL in the ovaries of Bos taurus indicus females did not influence the oxidative stress or antioxidant potential of the gonad. Thus, the removal of ovarian fragments with or without the presence of CL indicates that biotechnologies such as in vitro follicle cultivation is possible.

Abstract Acetylcholinesterase (AChE) activity levels can be used as an indicator for AChE inhibition due to pesticide poisoning in bird species. We assessed the comparative brain cholinesterase (AChE) activity level of five bird species inhabiting pesticide exposed croplands and Protected Area i.e. Deva Vatala National Park (DVNP), Bhimber by using a spectrophotometric method. AChE activity levels ranged from 56.3 to 85.9 µmol/min/g of brain tissue of birds representing DVNP. However, AChE activity levels ranged from 27.6 to 79.9 µmol/min/g of brain tissue of birds representing croplands. AChE activity levels observed in Jungle babbler, Common babbler, and Red-vented bulbul showed significant differences (P 0.05) at two sites. However, White wagtail and Black drongo demonstrated non-significant differences (P > 0.05). Maximum inhibition was recorded in Jungle babbler (53%) followed by Common babbler (35%), Red-vented bulbul (18%), White wagtail (15%), and Black drongo (7%). The brain cholinesterase inhibition levels under-protected ecosystems (DVNP, Bhimber) and agricultural landscape suggest insecticidal contamination and its impact on avifauna diversity. The study also emphasizes on the importance of pesticide-free zones to protect the biodiversity of birds.


Resumo Os níveis de atividade da acetilcolinesterase (AChE) podem ser usados como um indicador para a inibição da AChE devido ao envenenamento por pesticidas em espécies de aves. Avaliamos o nível de atividade comparativa da colinesterase cerebral (AChE) de cinco espécies de aves que habitam áreas cultivadas expostas a pesticidas e Área Protegida, ou seja, Deva Vatala National Park (DVNP), Bhimber, usando um método espectrofotométrico. Os níveis de atividade da AChE variaram de 56,3 a 85,9 µmol / min / g de tecido cerebral de aves representando DVNP. No entanto, os níveis de atividade da AChE variaram de 27,6 a 79,9 µmol / min / g de tecido cerebral de aves representando áreas de cultivo. Os níveis de atividade de AChE observados no tagarela da selva, tagarela comum e bulbul vermelho exalado mostraram diferenças significativas (P 0,05) em dois locais. No entanto, alvéola branca e drongo preto demonstraram diferenças não significativas (P > 0,05). A inibição máxima foi registrada no tagarela da selva (53%), seguido pelo tagarela comum (35%), bulbul vermelho (18%), alvéola branca (15%) e drongo preto (7%). Os níveis de inibição da colinesterase cerebral nos ecossistemas subprotegidos (DVNP, Bhimber) e na paisagem agrícola sugerem contaminação por inseticida e seu impacto na diversidade da avifauna. O estudo também enfatiza a importância das zonas livres de pesticidas para proteger a biodiversidade das aves.

Abstract This research aimed to investigate various mosquitocidal activities of Chenopodium botrys whole- plant n-hexane extract against Culex quinquefasciatus. The extract showed remarkable larvicidal, pupicidal, adulticidal, oviposition deterrent and adult emergence inhibitory activities against Cx. quinquefasciatus. During the larvicidal and pupicidal activities, the 24-hour lethal concentration (LC50) of extract against 2nd instar larvae, 4th instar larvae and pupae were 324.6, 495.6 and 950.8 ppm, respectively. During the CDC (Centers for Disease Control and Prevention) bottle bioassay for adulticidal activity, the median knockdown times (KDT50) at 1.25% concentration was 123.4 minutes. During the filter paper impregnation bioassay for adulticidal activity, the KDT50 value at 0.138 mg/cm2 concentration was 48.6 minutes. The extract was fractionated into 14 fractions through silica gel column chromatography which were then combined into six fractions on the basis of similar retention factor (Rf) value. These fractions were screened for adulticidal activity by applying CDC bottle bioassay. The fraction obtained through 60:40 to 50:50% n-hexanes-chloroform mobile phase with 0.5 Rf value showed 100% adulticidal activity at 0.2% concentration. During oviposition deterrent activity, the highest concentration (1000 ppm) showed 71.3 ± 4.4% effective repellence and 0.6 ± 0.1 oviposition activity index. During adult emergence inhibition activity, the median emergence inhibition (EI50) value was 312.3 ppm. From the outcome of the present investigation, it is concluded that the n-hexane extract of C. botrys whole- plant possesses strong larvicidal, pupicidal, adulticidal, oviposition deterrent and adult emergence inhibitory activities against Cx. quinquefasciatus.


Resumo Esta pesquisa teve como objetivo investigar várias atividades mosquitocidas do extrato n-hexano de planta inteira de Chenopodium botrys contra Culex quinquefasciatus. O extrato mostrou atividades larvicida, pupicida, adulticida, dissuasora de oviposição e inibidora da emergência de adultos contra a Cx. quinquefasciatus. Durante as atividades larvicida e pupicida, a concentração letal de 24 horas (CL50) do extrato contra larvas de 2º estádio, larvas de 4º estádio e pupa foi de 324,6, 495,6 e 950,8 ppm, respectivamente. Durante o bioensaio com frasco do CDC (Centros para Controle e Prevenção de Doenças) para adulticida, o tempo médio de desativação (KDT50) na concentração de 1,25% foi de 123,4 minutos. Durante o bioensaio de impregnação com papel de filtro para a atividade adulticida do extrato, o valor KDT50 na concentração de 0,138 mg / cm2 foi de 48,6 minutos. O extrato foi fracionado em 14 frações através de cromatografia em coluna de gel de sílica que foram então combinadas em seis frações com base em um valor de fator de retenção (Rf) semelhante. Essas frações foram selecionadas quanto à atividade adulticida por meio da aplicação do bioensaio com garrafa do CDC. A fração obtida através da fase móvel de n-hexanos-clorofórmio 60:40% a 50:50% com valor de 0,5 Rf apresentou atividade adulticida de 100% na concentração de 0,2%. Durante a atividade de dissuasão da oviposição, a maior concentração de extrato (1000 ppm) apresentou repelência efetiva de 71,3 ± 4,4% e índice de atividade de oviposição de 0,6 ± 0,1. Durante a atividade de inibição da emergência de adultos, o valor médio de inibição da emergência (EI50) foi de 312,3 ppm. A partir do resultado da presente investigação, conclui-se que o extrato de n-hexano da planta inteira de C. botrys possui fortes atividades larvicida, pupicida, adulticida, dissuasora da oviposição e inibidora da emergência de adultos contra a Cx. quinquefasciatus.

Abstract Trees occurring on the margins of agricultural areas can mitigate damage from residual herbicides. Rhizospheric microbial activity associated with trees is one of the main remedial capacity indicators. The objective of this study was to evaluate the rhizospheric microbiological activity in tree species subjected to the herbicides atrazine and sulfentrazone via the rhizosphere. The experiment was designed in four blocks and a 6 × 3 factorial scheme. The first factor consisted of six tree species from Brazil and the second of atrazine, sulfentrazone, and water solutions. Four herbicide applications were performed via irrigation. The total dry mass of the plants, mycorrhizal colonization, number of spores, basal respiration of the rhizospheric soil, and survival rate of bioindicator plants after phytoremediation were determined. Trichilia hirta had higher biomass when treated with atrazine and sulfentrazone. Herbicides decreased the microbial activity in Triplaris americana and did not affect the microbiological indicators of Myrsine gardneriana, Schizolobium parahyba, and Toona ciliata. Fewer bioindicator plants survived in soil with Triplaris americana and sulfentrazone. Microbiological indicators were influenced in different ways between species by the presence of herbicides in the rhizosphere.


Resumo As árvores que ocorrem nas margens das áreas agrícolas podem mitigar os danos dos herbicidas residuais. A atividade microbiana rizosférica associada às árvores é um dos principais indicadores de capacidade corretiva. O objetivo deste trabalho foi avaliar a atividade microbiológica rizosférica em espécies arbóreas submetidas aos herbicidas atrazina e sulfentrazone via rizosfera. O experimento foi estruturado em quatro blocos e esquema fatorial 6 × 3. O primeiro fator consistiu em seis espécies de árvores do Brasil e o segundo em soluções de atrazine, sulfentrazone e água. Quatro aplicações de herbicidas foram realizadas via irrigação. Foram determinados a massa seca total das plantas, colonização micorrízica, número de esporos, respiração basal do solo rizosférico e taxa de sobrevivência de plantas bioindicadoras após fitorremediação. Trichilia hirta apresentou maior biomassa quando tratada com atrazina e sulfentrazone. Os herbicidas diminuíram a atividade microbiana em Triplaris americana e não afetaram os indicadores microbiológicos de Myrsine gardneriana, Schizolobium parahyba e Toona ciliata. Menos plantas bioindicadoras sobreviveram no solo com Triplaris americana e sulfentrazone. Os indicadores microbiológicos foram influenciados de formas distintas entre as espécies pela presença dos herbicidas na rizosfera.

Abstract A ninety days nutritional trial was directed to explore the effects of dietary chromium on body composition, gut enzyme activity and physiological status of Cirrhinus mrigala by using G & NG corn. Six experimental diets were prepared by using different levels of chromium chloride hexahydrate (0, 0.2, 0.4 mg/kg, each with G & NG corn). For this experimental trial, 480 fingerlings, irrespective of sex were distributed in six aquariums each with replicate. Results revealed that gelatinized corn along with increasing level of Cr2Cl3.6H2O have a positive impact upon body composition of fish. Hematology was positively correlated with chromium chloride hexahydrate supplementation in gelatinized corn. Amylase gut enzyme also showed significant (P 0.05) increase in group fed with chromium chloride hexahydrate supplemented diet (G corn). However, corn with chromium chloride hexahydrate supplementation did not revealed any significant impact on gut protease enzyme activity. From these results it can be concluded that both chromium chloride hexahydrate and gelatinized corn in fish feed are very beneficial to improve body composition, enzymes activity and physiological health status of fish.


Resumo Um ensaio nutricional de 90 dias foi dirigido para explorar os efeitos do cromo dietético na composição corporal, atividade enzimática intestinal e estado fisiológico de Cirrhinus mrigala usando milho G & NG. Seis dietas experimentais foram preparadas usando diferentes níveis de cloreto de cromo hexa-hidratado (0, 0,2, 0,4 mg/kg, cada um com milho G e NG). Para este ensaio experimental, 480 alevinos, independentemente do sexo, foram distribuídos em seis aquários, cada um com réplicas. Os resultados revelaram que o milho gelatinizado, juntamente com o aumento do nível de Cr2Cl3.6H2O, tem um impacto positivo na composição corporal dos peixes. A hematologia foi positivamente correlacionada com a suplementação de cloreto de cromo hexa-hidratado em milho gelatinizado. A enzima amilase intestinal também apresentou aumento significativo (P 0,05) no grupo alimentado com dieta suplementada com cloreto de cromo hexa-hidratado (milho G). No entanto, o milho com suplementação de hexahidrato de cloreto de cromo não revelou nenhum impacto significativo na atividade da enzima protease intestinal. Desses resultados, pode-se concluir que tanto o cloreto de cromo hexa-hidratado quanto o milho gelatinizado na alimentação de peixes são muito benéficos para melhorar a composição corporal, a atividade de enzimas e o estado fisiológico de saúde dos peixes.

Abstract The present study was aimed to manifest the antibacterial and antifungal activity of methanolic extracts of Salix alba L. against seven Gram-positive and Gram-negative bacterial pathogens e.g. Streptococcus pyogenes, Staphylococcus aureus (1), S. aureus (2), Shigella sonnei, Escherichia coli (1), E. coli (2) and Neisseria gonorrhoeae and three fungal isolates from the air such as Aspergillus terreus, A. ornatus, and Rhizopus stolonifer. Two different serotypes of S. aureus and E. coli were used. The agar well-diffusion method results showed the dose-dependent response of plant extracts against bacterial and fungal strains while some organisms were found resistant e.g. E. coli (1), S. sonnei, A. terreus and R. stolonifer. The highest antibacterial activity was recorded at 17.000±1.732 mm from 100 mg/mL of leaves methanolic extracts against S. pyogenes while the activity of most of the pathogens decreased after 24 h of incubation. The highest antifungal activity was reported at 11.833±1.0 mm against A. ornatus at 50 mg/mL after 48 h of the incubation period. These experimental findings endorse the use of S. alba in ethnopharmacological formulations and suggest the use of methanolic extracts of the said plant to develop drugs to control the proliferation of resistant disease causing pathogenic microbes.


Resumo O presente estudo teve como objetivo manifestar a atividade antibacteriana e antifúngica de extratos metanólicos de Salix alba L. contra sete patógenos bacterianos Gram-positivos e Gram-negativos. Streptococcus pyogenes, Staphylococcus aureus (1), S. aureus (2), Shigella sonnei, Escherichia coli (1), E. coli (2) e Neisseria gonorrhoeae e três isolados de fungos do ar, como Aspergillus terreus, A. ornatus, e Rhizopus stolonifer. Dois sorotipos diferentes de S. aureus e E. coli foram usados. Os resultados do método de difusão em ágar mostraram a resposta dependente da dose de extratos de plantas contra cepas de bactérias e fungos, enquanto alguns organismos foram considerados resistentes, e.g. E. coli (1), S. sonnei, A. terreus e R. stolonifer. A maior atividade antibacteriana foi registrada em 17.000 ± 1.732 de 100 mg/mL de extratos metanólicos de folhas contra S. pyogenes, enquanto a atividade da maioria dos patógenos diminuiu após 24 h de incubação. A maior atividade antifúngica foi relatada em 11,833 ± 1,0 contra A. ornatus a 50 mg/mL após 48 h do período de incubação. Esses achados experimentais endossam o uso de S. alba em formulações etnofarmacológicas e sugerem o uso de extratos metanólicos da referida planta para o desenvolvimento de fármacos que controlem a proliferação de doenças resistentes que causam micróbios patogênicos.

ABSTRACT: This research determined the nutritional composition, antioxidant activity and total phenolic compound content of sweet potato flour from different cultivars: Beauregard cv. sweet potato (BF) and the common sweet potato Brazlândia Rosada cv. (CF). Total lipids, proteins, moisture, carbohydrates, pH, titratable acidity, fiber, ash, iron, zinc, beta-carotene, vitamin A and determination of caloric value were analyzed. Antioxidant activity and total phenolic compounds were also determined by the DPPH (2,2-diphenyl-1-picryl-hydrazyl), ABTS (2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) and FRAP (Ferric Reducing Antioxidant Power) methods. CF showed higher levels of carbohydrates, ash, fiber, pH and antioxidant activity by the ABTS method. Protein and zinc contents were similar between the two flours. BF had higher moisture content, caloric value, lipids, iron, DPPH radical scavenging capacity and antioxidant activity by the FRAP method, as well as total phenolic compounds, beta-carotene and vitamin A content. The data revealed good nutritional composition for both analyzed flours. The Beauregard flour can be highlightedregarding its higher beta-carotene and iron content. Moreover, the Beauregard sweet potato flour is a good source of provitamin A, in addition to being a source of fiber.


RESUMO: O objetivo deste trabalho foi determinar a composição nutricional, atividade antioxidante e teor de compostos fenólicos totais de farinhas de batata-doce de diferentes cultivares: batata-doce Beauregard (FB) e batata-doce comum, cv. Brazlândia Rosada (FC). Foram realizadas as análises de lipídeos, proteínas, umidade, carboidratos, pH, acidez titulável, fibras, cinzas, ferro, zinco, betacaroteno, vitamina A e determinação do valor calórico. Também foram determinadas a atividade antioxidante, pelos métodos DPPH (2,2-difenil-1-picril-hidrazil), ABTS (2,2-azinobis (3-etilbenzotiazolina-6-ácido sulfônico) e FRAP (Ferric Reducing Antioxidant Power), e compostos fenólicos totais. A FC apresentou maiores teores de carboidratos, cinzas, fibras, pH e atividade antioxidante pelo método ABTS. Os teores de proteínas e zinco foram semelhantes entre as duas farinhas. A FB apresentou maiores teores de umidade, valor calórico, lipídeos, ferro, capacidade sequestradora de radicais DPPH e atividade antioxidante pelo método FRAP, compostos fenólicos totais, conteúdo de betacaroteno e vitamina A. Os dados revelaram uma boa composição nutricional para ambas as farinhas analisadas. O destaque para a farinha da batata-doce Beauregard é maior em relação ao seu teor de betacaroteno e ferro. Sendo a farinha obtida pela batata-doce Beauregard uma boa fonte de pró-vitamina A, além de ser fonte de fibras.

ABSTRACT Aiming to the red mite Oligonychus yothersi control in yerba-mate, the systemic action of an azadirachtin-based product (12 g of azadirachtin.L-1) was evaluated in the laboratory. To assess the activity on oviposition and mortality of the mite (1), three weekly applications of the product in an aqueous solution (30, 48, and 66 mg i.a..L-1) were performed. The applications were carried out in the soil, next to the base of yerba mate seedlings. After seven days of each application, leaves were removed for infestation with 15 adult mite females. To evaluate the activity in the development, survival, and reproduction of the progeny (2), yerba mate seedlings were irrigated with azadirachtin solution of 30 mg i.a..L-1 (recommended concentration). After seven days, leaves were removed and infested with adult females for oviposition and observation of the progeny. Reduction in fecundity (from 23.7 to 44.2%), fertility (27.4 to 48.1%), and an increase in mortality from 14.6 to 47.5% were observed in females fed on azadirachtin-treated plants. Continuous feeding on plants treated with azadirachtin solution significantly reduced oviposition, pre-oviposition period, and longevity of males. In addition, the viability of the egg-adult, and the duration of the egg-adult period increased. The potential of azadirachtin for the control of O. yothersi and its action from the irrigation of yerba mate plants was confirmed.

Soil enzymes play a fundamental role in nutrient cycling in forest systems. The stoichiometry of C, N, and P­acquiring enzymes has been used to indicate nutrient limitation in the soil. However, the enzymatic stoichiometry remains poorly understood in pure and mixed eucalypt plantations. Thus, this study aims to assess the activity of enzymes in the soil to address the hypothesis that the introduction of N2-fixing trees could influence the enzymatic stoichiometry on C, N, and P cycling. The activity of ß-glucosidase (BG), urease (U), and acid phosphatase (AP) was assessed in soil (0-20 cm depth) of pure Eucalyptus grandis without (E) and with N fertilization (E+N), and a mixed system with E. grandis and Acacia mangium (E+A), and a pure A. mangium (A) plantation at 27 and 39 months after planting. The activities of BG/U, BG/AP, and U/AP were used to calculate the enzyme C/N, C/P, and N/P ratios, respectively. Rates of N­acquiring enzymes were higher in E and E+N, while soil microorganisms invested in P­acquiring enzymes in A and E+A. The vector length and angle demonstrated that C demand by microorganisms does not change in relation to N and P, regardless of the treatment. However, N demand decreased in relation to P in A and E+A (mainly at 27 months). Our results suggest that enzymes activity in pure eucalypt systems is limited for their soil-litter nutrient contents. At the same time while acacia and mixed plantation seem to invest in P­acquiring enzymes to improve biological N2 fixation promoted by diazotrophic bacteria associated to acacia.

Resumo

Top-quality pellets can significantly increase density and durability of agricultural residues, reducing logistic costs. However, these pellets depend on numerous parameters, including feedstock properties and production conditions. To ensure high-quality pellets, a single-factor experiment and the response surface methodology were used to investigate the effects of particle size, moisture content, molding pressure, pelleting time, ultrasonic power, and interaction effects between variables on density and durability of pellets for ultrasonic vibration-assisted pelleting of corn stover. The response surface models between variables and response were established. The results showed that all variables affect the density and durability of pellets. An optimal condition for density and durability was obtained, and a further experiment was conducted to validate the values. The results suggested that desirability (0.999) under optimal conditions confirmed the validation of models. The optimal combination of process parameters included particle size of 1.5 mm, moisture content of 10 %, molding pressure of 379 kPa, pelleting time of 80 s, ultrasonic power of 250 W, with values of 1,381.14 kg m-3 and 97.58 % for density and durability of pellets, respectively.

The four fish species, Trichogaster pectoralis, Barbodes schwanenfeldii, Osphronemus gouramy, and Wallago leeri have the same body size, representing different habitats. Therefore, this study aimed to determine the four species swimming abilities and speeds, including sustained, maximum sustained, prolonged swimming speed, and burst speed. The four species were brought using oxygenated plastic bags to the Aquatic Resources Utilization Laboratory at Riau University. Then it was acclimated in an aquarium measuring 100×50×50 cm with an aeration and filtering system to maintain water quality. The four groups of species were forced to swim in the swimming channel of the flume tank, with different current speeds for each individual tested. The swimming activity of the fish was recorded using a high-speed video camera (Casio HS Exlim, EX-ZR400, Casio Computer Co. LTD, Japan). The results showed that the swimming speed of B. schwanenfeldii was higher than O. gouramy, T. pectoralis, and W. leeri. The maximum sustained swimming speed of B. schwanenfeldii is about 5.1 BL/s, and 14.2 BL/s for the burst speed. Meanwhile, the sustained swimming speed of O. gouramy (1.6 BL/s) and the burst speed is 7.2 BL/s. The maximum sustained swimming speed of T. pectoralis was 1.6 BL/s, and the burst speed was 5.7 BL/s. In addition, the maximum sustained swimming speed of W. leeri is only 0.5 BL/s and 2.4 BL/s for the burst speed. The ability of these four fish to swim was influenced by the morphology of the tail fin, body shape, and the habitat where they live.

ABSTRACT: The production of bioactive food is one of the goals of agriculture. Sprouts used for human consumption are an efficient way to increase the intake of essential nutrients, such as minerals, vitamins, and other bioactive compounds. The use of elicitors can increase the nutritional value of these products. Biomass production, phenolic compound content, and antioxidant activity of mung bean sprouts (Vigna radiata L.) after elicitation were examined in this study. Mung bean sprouts were treated with salicylic acid (SA), ascorbic acid (AA), and tocopherol (TOC) at different concentrations and numbers of applications. Shoot and root lengths and dry mass were measured on five-day-old sprouts. Total phenol content and antioxidant activity were determined using the DPPH assay on dried ethanolic extracts. Total soluble protein content and superoxide dismutase activity were measured in frozen hypocotyls. All elicitors stimulated the antioxidant functions of sprouts and, at the highest concentrations, reduced the biometric parameters; therefore, lower concentrations were better. For the first time, a balance between elicitor concentration and application in instalments was achieved to maintain the production of sprouts with enhanced nutritional characteristics.


RESUMO: A produção de alimentos bioativos é um dos objetivos da agricultura. O uso de brotos para consumo humano é uma forma eficiente de aumentar a ingestão de nutrientes essenciais, como minerais, vitaminas e outros compostos bioativos. O uso de elicitores pode aumentar o valor nutricional desses produtos. A produção de biomassa, o conteúdo de compostos fenólicos e a atividade antioxidante de brotos de feijão (Vigna radiata L.) após a elicitação foram examinados neste estudo. Brotos de feijão foram tratados com ácido salicílico (SA), ácido ascórbico (AA) e tocoferol (TOC) em diferentes concentrações e número de aplicações. O comprimento da parte aérea e da raiz e a massa seca foram medidos em brotos com cinco dias de idade. O teor de fenol total e a atividade antioxidante foram determinados usando o ensaio DPPH em extratos etanólicos secos. O conteúdo total de proteína solúvel e atividade da superóxido dismutase foram medidos em hipocótilos congelados. Todos os elicitores estimularam as funções antioxidantes dos brotos e, nas maiores concentrações, reduziram os parâmetros biométricos; assim, concentrações mais baixas foram mais eficientes. Pela primeira vez, foi alcançado um equilíbrio entre a concentração do elicitor e a aplicação em parcelas para incrementar a produção de brotos e aprimorar características nutricionais aprimoradas.

Evaluating the effects of N and K supply on grapevines ( Vitis vinifera L.) and the techniques for nutritional diagnosis is of great importance for fertigation management of this crop. This study evaluated the effects of N and K fertigation on the soluble concentrations of NO 3 - and K + in the petiole sap and on the leaf chlorophyll index in drip irrigated 'Syrah' grapevine (from 17 June 2013 to 25 Nov 2014). The treatments consisted of five levels of N (0, 15, 30, 60 and 120 kg ha -1 ) and K 2 O (0, 15, 30, 60 and 120 kg ha -1 ), combined in an incomplete 5 2 factorial scheme in 13 combinations and arranged in randomized blocks with four replications. We determined NO 3 - and K + concentrations in petiole sap, leaf chlorophyll index, grapevine cluster mass and number per plant, mean grapevine cluster mass, and phenolic composition. High NO 3 - concentrations contributed positively to grapevine yield; however, increased K+ concentrations caused a negative response of sap. For 120 kg N ha -1 rate, NO 3 - in the sap and chlorophyll index showed higher values at the flowering stage, while high values for K + were observed during the grape-ripening stage.(AU)

Abstract Croton argyrophylloides Muell. Arg., from the Euphorbiaceae family, popularly known as marmeleiro prateado or sacatinga, is a plant from the Caatinga biome commonly found in Brazils northeastern region. The present study aimed to evaluate the antioxidant activity of the species. The phytochemical study was performed through qualitative analysis of chemical constituents and quantitative determination of the total phenol content through the Folin-Ciocalteu test. The qualitative and quantitative antioxidant tests were performed using the DPPH method (2.2 diphenyl-1-picryl hydrazil) and ferric reducing antioxidant power (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The ethanolic extract of the leaves of C. argyrophylloides manifested antioxidant action in the quantitative DPPH test with a significant bioactivity of 84.70 AAO% in 500 µg/mL, with an EC50 of 236.79. The content of total phenolic compounds was 946.06 mg of gallic acid equivalents/g of sample, and total flavonoids was 58.11 mg of quercetin equivalents/g of sample, the result obtained for FRAP was 15294.44 µM Trolox/g of sample and ABTS was 718 M Trolox of sample. The prospecting of the chemical constituents of the leaves of C. argyrophylloides revealed the presence of the main compounds that manifests the antioxidant activity and it was proven by the DPPH method that there is antioxidant activity in the analyzed sample, in addition to demonstrating a significant content of phenolic compounds and total flavonoid content in the species, which corroborates the antioxidant activity of the plant sample. The leaf extracts presented growth inhibition halos of 10 and 12 mm upon Staphylococcus aureus ATCC 25923.


Resumo Croton argyrophylloides Muell. Arg., pertencente à família Euphorbiaceae, conhecida popularmente como marmeleiro prateado e sacatinga, é um vegetal do bioma caatinga comumente encontrado no Nordeste do Brasil. O presente trabalho teve como objetivo avaliar a atividade antioxidante da espécie. O estudo fitoquímico foi realizado por meio de análise qualitativa dos constituintes químicos e determinação quantitativa do teor de fenóis totais pelo teste de Folin-Ciocalteu. Os testes antioxidantes qualitativos e quantitativos foram realizados pelo método do DPPH (2,2 difenil-1- picril-hidrazila) e redução do ferro (FRAP). A concentração inibitória mínima (CIM) foi determinada por microdiluição em placas de 96 poços. O extrato etanólico das folhas de C. argyrophylloides apresentou ação antioxidante no teste DPPH quantitativo com uma significativa bioatividade de 84.70 AAO% em 500 µg/mL, apresentando um CE50 de 236.79. O teor de compostos fenólicos totais, foi de 946,06 mg equivalentes de ácido gálico/g de amostra, e de flavonoides totais de 58,11 mg equivalentes de quercetina/g da amostra, o valor encontrado para FRAP foi de 15294,44 µM Trolox/g da amostra e de ABTS foi 718 M Trolox da amostra. A prospecção dos constituintes químicos das folhas de C. argyrophylloides revelou a presença dos principais compostos que caracterizam a atividade antioxidante e foi possível comprovar pelo método de DPPH que há atividade antioxidante na amostra analisada, além de demonstrar um resultado significativo de teor de compostos fenólicos e teor de flavonoides totais na espécie e o que corrobora com a atividade antioxidante da amostra vegetal. Os extratos das folhas apresentaram halos de inibição de crescimento de 10 e 12mm frente a Staphylococcus aureus ATCC 25923.

The objective of this review is to bring information about innovations and technologies that, through genetic improvement, are being used to improve the sustainability and productivity of agricultural crops, improve human nutrition, as well as conservation and decontamination of soils. Bioremediation consists of using microorganisms that have the ability to modify or decompose certain pollutants, with the possibility of increasing their activity through genetic engineering, building new strains for the transformation of pollutants into inert substances. Genetic improvement is seeking to develop cultivars that are more tolerant to periods of water deficit. Plant biofortification consists of varieties of improved plants that have a higher content of vitamins and minerals, which are obtained through genetic improvement. Thus, biotechnology is once again essential for world agricultural production and can bring a series of other benefits to society.(AU)

Transcription factors (TF) are a wide class of genes in plants, and these can regulate the expression of other genes in response to various environmental stresses (biotic and abiotic). In the current study, transcription factor activity in sugarcane was examined during cold stress. Initially, RNA transcript reads of two sugarcane cultivars (ROC22 and GT08-1108) under cold stress were downloaded from SRA NCBI database. The reads were aligned into a reference genome and the differential expression analyses were performed with the R/Bioconductor edgeR package. Based on our analyses in the ROC22 cultivar, 963 TF genes were significantly upregulated under cold stress among a total of 5649 upregulated genes, while 293 TF genes were downregulated among a total of 3,289 downregulated genes. In the GT08-1108 cultivar, 974 TF genes were identified among 5,649 upregulated genes and 283 TF genes were found among 3,289 downregulated genes. Most transcription factors were annotated with GO categories related to protein binding, transcription factor binding, DNA-sequence-specific binding, transcription factor complex, transcription factor activity in RNA polymerase II, the activity of nucleic acid binding transcription factor, transcription corepressor activity, sequence-specific regulatory region, the activity of transcription factor of RNA polymerase II, transcription factor cofactor activity, transcription factor activity from plastid promoter, transcription factor activity from RNA polymerase I promoter, polymerase II and RNA polymerase III. The findings of above results will help to identify differentially expressed transcription factors during cold stress. It also provides a comprehensive analysis of the regulation of the transcription activity of many genes. Therefore, this study provides the molecular basis for improving cold tolerance in sugarcane and other economically important grasses.
