Resumo
Bovine leukosis is caused by an oncogenic virus of the genus Deltaretrovirus, causing losses associated with decreased production indicators and restrictions on exports of cattle and cattle products. The disease has a prolonged incubation period of between 1-5 years and the antibodies can be detected 2-3 weeks post infection. The disease can present asymptomatically, and develop persistent lymphocytosis or lymphosarcoma. The objective of this study was to estimate the prevalence and risk factors associated with bovine leukosis in Villavicencio, Colombia. Blood samples were taken from 636 animals, and obtained randomly from 24 herds. The samples were analysed using a Competition ELISA kit for the detection of anti-gp51 antibodies. Information on possible risk factors was collected, then OR and X2 were calculated, and statistically significant with p < 0.05 variables were included in a linear regression multivariate analysis. The general seroprevalence was 24.6% and the herd seroprevalence was 83.3%. The seroprevalence was 21.3% in males and 25.0% in females. The risk factors identified were abortion, non-bearing cows, artificial insemination, and use of common needles, Creole breed and participation in cattle exhibitions. The study confirmed the presence of bovine leukosis associated with reproductive and management factors.(AU)

Sex steroid hormones are critical in gonadal differentiation in turtles. The gonads are not the only organs responsible for producing these hormones during this phase. Mesonephros play an important role in steroidogenesis. The present study aimed to investigate the presence of steroidogenic cells in mesonephros of Podocnemis expansa during gonadal differentiation and to evaluate their morphology and ultrastructure. Ten embryos of P. expansa were collected from 5 nests on day 36 of incubation, during spawning period on an artificial beach. Embryos were extracted from eggs by slicing the shell and euthanized. They were dissected under a stereoscopic microscope to collect the gonad-mesonephro complex, in which were fixed and subsequently processed for light microscopy, immunohistochemistry and transmission electron microscopy analysis. During histological analysis was observed mesonephros has typical morphological structure. Immunohistochemistry showed immunoreaction to aromatase in cells of intertubular space. Confirming these findings, it was possible to observe a type of intertubular cell in several regions of mesonephro, being more predominant in region close to blood vessels, distal and proximal tubules. In ultrastructural analysis these cells were characterized by having a clear, large, and rounded nucleus with evident nucleolus and cytoplasm rich in electron-dense droplets. This study demonstrated for the first time the presence of cells with morphological, immunohistochemical and ultrastructural characteristics similar to steroid-producing cells in P. expansa mesonephrons, suggesting that this organ may contribute to gonadal differentiation in this species.(AU)

Although the physiological principles that govern egg laying and incubation in birds are the same, we are faced with two very different realities with regard to these points when comparing domestic and wild birds. Both zoos and bird breeders (commercial and conservationist) usually hire multi-skilled professionals to take care of several issues ranging from health and general management to nutrition. The most apparent consequence of such a strategy is the difficulty these professionals and institutions have in accessing more recent methodologies for reproductive management and egg incubation. Additionally, employees from these institutions deal with an enormous diversity of avian groups and species, a situation that also hinders the implementation of certain management and incubation techniques. On the other hand, in poultry industrial poultry there is extremely specialized personnel with immense knowledge in both areas (posture and incubation), which, together with cutting-edge technology in its various industry sectors, provide optimization of results. In this context, the goal of this material is to show some concepts and methodologies that are routinely used in industrial poultry farming and that are already (in some breeders in other countries) or can be included in the daily management of wild birds.

This study aimed to characterize the development and gonadal differentiation of Podocnemis unifilis at a controlled temperature of 32° C in artificial incubator and with humidity control performed by an internal hygrometer. Macroscopic analyzes, light microscopy and transmission electron microscopy were performed. We grouped gonadal development into three stages: primordial germ cells (PGC), establishment of the undifferentiated gonad and gonadal differentiation. We only visualized the PGC from the 7th day of incubation, and in the caudal region of the embryo body; between days 9 and 11 of incubation we were able to observe the migration of the PGC from the viteline sac towards the ventromedial region of the mesonefro. At 20 days of development, the undifferentiated gonad was established and two distinct regions were identified: the external cortical region characterized by the presence of PGC, and the internal medullary region marked by primitive sexual cords. In this study, only gonadal differentiation occurred in the testicle. Gonadal differentiation began after 35 days of incubation with the beginning of the organization of sexual cords in the seminiferous tubules. The stages of gonadal differentiation were similar to those of other species of turtles with temperature determined sex, however in this study, using the pivotal temperature described for the species all the individuals analyzed were males, and it is there for necessary to carry out further studies using a higher incubation temperature.(AU)

The objective was to evaluate the use of the hot plate to replace the water bath in the thermo resistance test of ovine sperm after cryopreservation. Ten ejaculates were also collected from three adult sheep (n=30), by means of artificial sheep vagina. The collected samples were diluted in Tris-Egg Yolk, at a final concentration of 200 x106 sptz/mL and subjected to a cooling curve for subsequent cryopreservation in liquid nitrogen straws. Immediately after thawing, the samples were analyzed for membrane and acrosome integrity, and mitochondrial activity. The semen was then divided into two tubes and submitted to TTR, one in a water bath and the other in a hot plate, and evaluated every 30 minutes, from time zero (post-thaw) until 90 minutes of incubation at 37 °C, for sperm motility and progressive motility. The variables were subjected to analysis of variance and the means were compared using the Tukey test at 5% probability. After the freeze/thawing process, sperm showed a low percentage of membrane integrity, high percentage of mitochondrial activity, in addition to maintaining the integrity of acrossome. No difference was observed in sperm motility nor progressive motility, along the TTR, when comparing the sperm that were incubated in a water bath in relation to those incubated in a hot plate during the period of 90 minutes. The heating plate can be used as a means of incubating sheep sperm in replacement of the water bath.

The tegu (Salvator merianae) is the largest lizard found in Brazil and has a wide distribution, occurring in several biomes. The presence of fungi on reptilian eggs initially occurs on infertile eggs, which leads to a subsequent infection in fertile eggs, causing damage and death of embryos. The objective of this study was to identify the fungi that caused lesions in the eggs of tegu in captivity. Swabs were collected on infected and healthy eggshells, in an embryo that died and the substrate of the incubation box. Samples were submitted to saboraund dextrose agar. A total of 93.3% of positive samples were obtained for the fungi Fusarium (11/14), Trichoderma (1/15), Penicilium (1/15), Mucor (1/15), and yeasts of the genus Trichosporon. Therefore, Fusarium was the main agent that caused the lesion and death of embryos in the eggs of the tegu lizard. Sanitary control methods may be taken to avoid fungal infections in the eggs.(AU)

This study investigated in vitro the efficacy of four different extenders (TES-TRIS and TRIS with LDL low-density lipoprotein at concentrations of 10 or 5%) on the longevity of buffalo sperm in the refrigeration process at 5ºC. Sperm motility was assessed every 24 hours up to 72 hours of incubation using computer assisted sperm analysis and sperm membrane integrity was examined by the hypoosmotic test (HOST) at T1, T24, T48 and T72 hours. Eleven buffaloes (1 ejaculate per buffalo) of the Murrah breed were used, ranging in age from 4 to 5 years. Immediately after collection, each ejaculate was fractionated into 4 aliquots, and each aliquot was diluted in one of four diluents to obtain 50x106SPTZ/mL. The samples were packed in 0.5mL straws and refrigerated (-0.25°C/min) to 5°C and maintained at this temperature until evaluation. Prior to evaluation the samples were heated at 37°C for 30 seconds. The statistical package used for analysis was STATA 12.0 "Statistical Analysis Software" and means were compared by the Friedman test (P<0.05). The results of sperm kinetics and HOST indicate that the TRIS diluent with 10% LDL could be a promising alternative for semen refrigeration at 5ºC, to be used in conventional and fixed time artificial insemination.(AU)

The hatchery is one of the most important segments of the poultry chain, and generates an abundance of data, which, when analyzed, allow for identifying critical points of the process . The aim of this study was to evaluate the applicability of the data mining technique to databases of egg incubation of broiler breeders and laying hen breeders. The study uses a database recording egg incubation from broiler breeders housed in pens with shavings used for litters in natural mating, as well as laying hen breeders housed in cages using an artificial insemination mating system. The data mining technique (DM) was applied to analyses in a classification task, using the type of breeder and house system for delineating classes. The database was analyzed in three different ways: original database, attribute selection, and expert analysis. Models were selected on the basis of model precision and class accuracy. The data mining technique allowed for the classification of hatchery fertile eggs from different genetic groups, as well as hatching rates and the percentage of fertile eggs (the attributes with the greatest classification power). Broiler breeders showed higher fertility (> 95 %), but higher embryonic mortality between the third and seventh day post-hatching (> 0.5 %) when compared to laying hen breeders eggs. In conclusion, applying data mining to the hatchery process, selection of attributes and strategies based on the experience of experts can improve model performance.

The Chestnut-capped Blackbird, Chrysomus ruficapilus (Vieillot, 1819), is a common bird species in flooded areas of South America. Data on its reproductive parameters have been reported mainly for rice paddies from Uruguay and southern Brazil, where reproductive phenology might have been influenced by the chronology of agricultural activities. Here we provide reproductive data for a population in a natural marshland from São Paulo state, southeastern Brazil. A total of 45 active nests were monitored between December 2017 and April 2018. Clutch size was 2.8 ± 0.44. Incubation and nestling periods were respectively 11.8 ± 0.39, and 12.3 ± 0.75 days, and overall nesting success was 65%. The reproductive season lasted about five months, which is longer than that observed in rice paddies from southern Brazil. This suggests that the reproductive phenology has been underestimated before. Although clutch sizes were bigger in our study population than that from rice paddies from southern Brazil, nest survival was higher in the artificial habitat, suggesting that the Chestnut-capped Blackbird can obtain benefits from nesting in artificial habitats.

Boar spermatozoa are very susceptible to cryopreservation injuries and, for this reason, pig remains one of the few species in which fresh semen is still preferred to thawed one for routine artificial insemination (AI). The present work evaluated the effect of supplementing boar sperm thawing medium with Silvafeed SP (SSP), a mixture of Chestnut and Quebracho wood extracts (60/40 w/w) rich in polyphenols (92.4% tannin content) on in vitro fertilization (IVF) and on the following sperm parameters: sperm motility (assessed by CASA), viability, acrosome integrity, mitochondrial function and lipid peroxidation (assessed by flow cytometry) and capacitation status (immunolocalization of tyrosine phosphorylated proteins). Thawed spermatozoa were incubated 1 h at 37°C in BTS without (CTR) or with (5, 10, 20 µg/mL) SSP. After incubation sperm suspension was divided in three aliquots: one was used for IVF trials, one for sperm analysis, and the last one was capacitated for 1 h at 39°C 5% CO2 in IVF medium. Sperm motility parameters, viability, acrosome integrity, mitochondrial functionality, lipid peroxidation and tyrosine phosphorylated protein immunolocalization, used as capacitation parameter, were not influenced by SSP. However, oocytes inseminated with thawed spermatozoa pretreated with all the different SSP concentrations presented a significant (P < 0.01) increase in penetration rate compared to CTR. In addition, 5 µg/mL SSP exerted a positive effect (P<0.05) on the total efficiency of fertilization. These results encourage the use of SSP in the thawing medium since post-thawing fertility is a limit for the large-scale use of boar frozen semen.

Abstract The management and use of native species under laboratory conditions is the main difficult for species proposal for its use in bioassays. The present study showed the results about management under controlled conditions of Paratanytarsus grimmii (Diptera: Chironomidae), that is a parthenogenetic species with wide geographical distribution. It obtained its eggs from a pre Andean river from Araucania region (39° S), and it determined the conditions for larval rearing under artificial food, adults getting (manipulation), eggs (hatching sincronization) and larvae getting after hatching. The P.grimmii life cycle had larvae that late 23 days in get the pupa stage (stage I to IV), the adult within the next 48 hours deposits the eggs that hatched at 72 hours. It stablished laboratory conditions: incubation room temperature, water pH, and artificial feeding, photoperiod and larvae and eggs management techniques. In according to obtained results it managed all life cycles under laboratory conditions that can propose the use of this species as potential biological material for toxicity bioassays.

Abstract The tegu (Salvator merianae) is the largest lizard found in Brazil and has a wide distribution, occurring in several biomes. Sanitary control methods may be taken to avoid fungal infections in the eggs.
