The demand of the global population for aquatic products is increasing while the production of extractive fisheries is reducing, and, in many cases, even reaching its maximum productive potential. As a consequence, it will not be possible in the short term to sustain the supply of aquatic products aimed at a population in constant growth and in demand for fish. The productive sector follows the current trend of other animal production systems, which have been seeking to sustainably increase productivity from an economic and environmental point of view, mainly requiring the contribution of nutrition to this trend. Studies with additives to the diet of captive animals is a strategy that has shown high potential for inclusion in aquaculture, with the possibility of an increase in production rates and/or improvement in the quality of the product for the consumer. Ractopamine is an additive classified as a beta-adrenergic agonist and its mechanism of action is associated with effects on fish metabolism that reduce the accumulation of fats through the inhibition of lipogenesis and stimulation of lipolysis, and by mechanisms that favor the synthesis of muscle protein. The studies carried out so far prove that there are metabolic changes in fish, although no significant differences have been found in zootechnical indexes in order to establish its use in the industry. Further research is required for a better understanding of ractopamine in fish nutrition, especially in understanding the receptors and mechanisms of action in fish.(AU)

This study evaluated chromium supplements and energy restriction as substitutes for ractopamine in the diets of late finishing gilts. Sixty gilts were used, with initial weights of 98.87 ± 0.25 kg and final weights of 122.69 ± 10.97 kg, distributed in a randomized block design with five diets: control; yeast chromium (0.8 ppm); chromium picolinate (0.48 ppm); ractopamine (20 ppm) and energy restriction (reduction of 150 kcal of EM kg-1 of feed), with six replicates and two animals per experimental unit. Feeding diet containing ractopamine enabled better (P<0.05) feed conversion and greater weights of the hot carcass of the gilts. The diet containing ractopamine exhibited a lower (P<0.05) cost of feed per kilogram of gain and higher economic efficiency, when compared to diets containing chromium and energy restriction; these exhibited better outcomes, compared to the control diet. Diets supplemented with chromium and ractopamine resulted in a higher (P<0.05) percentage and quantity of lean meat and a higher rate of carcass bonus, when compared to the control diet and energy restriction. The energy reduction did not harm the gilts' responses, compared to the control diet. Ractopamine supplementation allowed for a better feed conversion, lower feed cost per kilogram of gain, and a higher economic efficiency index. Supplements of chromium and ractopamine increased the percentage and quantity of lean meat and the bonus index of the carcasses. Therefore, chromium picolinate and chromium yeast are potential substitutes for ractopamine for optimizing the gilts carcass characteristics.

The objective of this study was to evaluate the effects of combining immunocastration and ß-adrenergic agonists (ß-AA) on blood metabolites and their correlations with performance and carcass traits of Nellore cattle. Ninety-six Nellore males were distributed in a randomized block design with a 2×3 factorial arrangement. The factors were two sexual conditions (immunocastrated and non-castrated), and three diets (control, with no inclusion of ß-AA; RH, with 300 mg of ractopamine hydrochloride/d; and ZH, with 80 mg of zilpaterol hydrochloride/animal/d). The trial was conducted during 100 d, during which animals were fed ZH and RH for the last 30 and 33 d, respectively. Blood metabolites related to lipid and protein metabolism were assessed at the baseline (0 d of ß-AA supplementation) and after 13 and 30 d of ß-AA supplementation. No effect of sexual condition × ß-AA supplementation × time on feed interaction was observed. Combining immunocastration with ß-AA supplementation modified cholesterol and non-esterified fatty acids concentrations without affecting protein metabolism. Immunocastration enhances lipogenesis and reduces skeletal muscle accretion by increasing high-density lipoprotein and triglycerides concentrations and decreasing creatinine and creatine kinase concentrations, respectively. Zilpaterol hydrochloride enhances skeletal muscle accretion by decreasing urea and total protein concentrations and increases creatinine and creatine kinase concentrations without modifying lipid metabolism.(AU)

Abstract Purpose To evaluate the analgesic effect of esmolol in patients submitted to laparoscopic gastroplasty. Methods Forty patients aged between 18 and 50 years with American Society of Anesthesiologists (ASA) physical status scores of II and III who underwent gastric bypass were allocated to two groups. Group 1 patients received a 0.5-mg/kg bolus of esmolol in 30 mL of saline before induction of anesthesia, followed by an infusion at 15 µg/kg/min until the end of surgery. Group 2 patients received 30 mL of saline as a bolus and then an infusion of saline. Anesthesia included fentanyl (3 µg/kg), propofol (2-4 mg/kg), rocuronium (0.6 mg/kg), and 2% sevoflurane, with remifentanil if necessary. The following parameters were evaluated: pain intensity over 24h, remifentanil consumption, the first analgesic request, morphine consumption, and side effects. Results Pain intensity was lower in the esmolol group except at T0 (after extubation) and 12h postoperatively. Remifentanil supplementation, recovery time, and postoperative morphine supplementation were lower in the esmolol group. No differences in the time to the first analgesic request or side effects were found between the groups. Conclusion Intraoperative esmolol promotes reductions in pain intensity and the need for analgesic supplementation without adverse effects, thus representing an effective drug for multimodal analgesia in gastroplasty.

Beta-cardiotoxin (ß-CTX), the three-finger toxin isolated from king cobra (Ophiophagus hannah) venom, possesses ß-blocker activity as indicated by its negative chronotropy and its binding property to both ß-1 and ß-2 adrenergic receptors and has been proposed as a novel ß-blocker candidate. Previously, ß-CTX was isolated and purified by FPLC. Here, we present an alternative method to purify this toxin. In addition, we tested its cytotoxicity against different mammalian muscle cell types and determined the impact on cardiac function in isolated cardiac myocyte so as to provide insights into the pharmacological action of this protein. Methods: ß-CTX was isolated from the crude venom of the Thai king cobra using reverse-phased and cation exchange HPLC. In vitro cellular viability MTT assays were performed on mouse myoblast (C2C12), rat smooth muscle (A7r5), and rat cardiac myoblast (H9c2) cells. Cell shortening and calcium transient dynamics were recorded on isolated rat cardiac myocytes over a range of ß-CTX concentration. Results: Purified ß-CTX was recovered from crude venom (0.53% w/w). MTT assays revealed 50% cytotoxicity on A7r5 cells at 9.41 ± 1.14 µM (n = 3), but no cytotoxicity on C2C12 and H9c2 cells up to 114.09 µM. ß-CTX suppressed the extend of rat cardiac cell shortening in a dose-dependent manner; the half-maximal inhibition concentration was 95.97 ± 50.10 nM (n = 3). In addition, the rates of cell shortening and re-lengthening were decreased in ß-CTX treated myocytes concomitant with a prolongation of the intracellular calcium transient decay, indicating depression of cardiac contractility secondary to altered cardiac calcium homeostasis. Conclusion: We present an alternative purification method for ß-CTX from king cobra venom. We reveal cytotoxicity towards smooth muscle and depression of cardiac contractility by this protein. These data are useful to aid future development of pharmacological agents derived from ß-CTX.(AU)

Abstract Purpose: To evaluate the cardioprotective response of the pharmacological modulation of β-adrenergic receptors (β-AR) in animal model of cardiac ischemia and reperfusion (CIR), in spontaneously hypertensive (SHR) and normotensive (NWR) rats. Methods: CIR was induced by the occlusion of left anterior descendent coronary artery (10 min) and reperfusion (75 min). The SHR was treated with β-AR antagonist atenolol (AT, 10 mg/kg, IV) 5 min before CIR, and NWR were treated with β-AR agonist isoproterenol (ISO, 0.5 mg/kg, IV) 5 min before CIR. Results: The treatment with AT increased the incidence of VA, AVB and LET in SHR, suggesting that spontaneous cardioprotection in hypertensive animals was abolished by blockade of β-AR. In contrast, the treatment with ISO significantly reduced the incidence of ventricular arrhythmia, atrioventricular blockade and lethality in NWR (30%, 20% and 20%, respectively), suggesting that the activation of β-AR stimulate cardioprotection in normotensive animals. Serum CK-MB were higher in SHR/CIR and NWR/CIR compared to respective SHAM group (not altered by treatment with AT or ISO). Conclusion: The pharmacological modulation of β-AR could be a new cardioprotective strategy for the therapy of myocardial dysfunctions induced by CIR related to cardiac surgery and cardiovascular diseases.

Abstract Purpose: To evaluate the effects of Dexmedetomidine (Dex) on spinal pathology and inflammatory factor in a rat model of Diabetic neuropathic pain (DNP). Methods: The rats were divided into 3 groups (eight in each group): normal group (N group), diabetic neuropathic pain model group (DNP group), and DNP model with dexmedetomidine (Dex group). The rat model of diabetes was established with intraperitoneal streptozotocin (STZ) injections. Nerve cell ultrastructure was evaluated with transmission electron microscopy (TEM). The mechanical withdrawal threshold (MWT) and motor nerve conduction velocity (MNCV) tests documented that DNP rat model was characterized by a decreased pain threshold and nerve conduction velocity. Results: Dex restored the phenotype of neurocytes, reduced the extent of demyelination and improved MWT and MNCV of DNP-treated rats (P=0.01, P=0.038, respectively). The expression of three pain-and inflammation-associated factors (P2X4, NLRP3, and IL-IP) was significantly upregulated at the protein level in DNP rats, and this change was reversed by Dex administration (P=0.0022, P=0.0092, P=0.0028, respectively). Conclusion: The P2X4/NLRP3 signaling pathway is implicated in the development and presence of DNP in vivo, and Dex protects from this disorder.

Increasing the amount of meat in the pig carcass has been the goal not only of the industry but also of the pig producer as it improves profitability and reduces production costs. In this sense, the objective of this research was to evaluate the performance and carcass characteristics of finishing pigs supplemented with different levels of ractopamine in the diet. Sixty 60 pigs (30 castrated males and 30 females) were used, with a mean initial weight of 75.0 kg housed in partially slatted stalls. The experimental design was a randomized block with three ractopamine levels (0, 5 and 10 ppm), totaling three treatments and ten replicates, with two animals (one male and one female) per pen (experimental plot). There were analyzed animals performance (final weight, average daily weight gain, daily feed intake and feed conversion) and carcass quality (carcass yield, carcass meat yield, backfat thickness and loin depth). The supplementation of 10 ppm of ractopamine in the finishing pig diet increased the meat yield in the carcass and reduced the backfat thickness. Thus, it is concluded that the supplementation of 10 ppm of ractopamine in the finishing pig diet improves the carcass traits.

An experiment was conducted during 60 days with forty pacu males fed diets supplemented with increasing concentrations of ractopamine (0.00, 11.25, 22.50, 33.75, and 45.00 mg kg−1). Eight fish were evaluated for each experimental diet. Performance and survival rate of the fish were measured. At the end of the experiment, blood was collected to determine the levels of cortisol, triacylglycerol, and protein. Moreover, the liver was collected to determine the activities of glucose-6-phosphate dehydrogenase and malic enzymes. The fillets were collected to determine chemical composition, and histologic cuts were analyzed to verify muscle growth and deposition of adipose tissue between muscle fibers. Increasing concentrations of dietary ractopamine did not change feed intake, fillet yield, fillet content of protein and ash, and frequency of relative distribution of muscle fibers. By increasing the dietary ractopamine concentration, the serum cortisol level was elevated. Ractopamine supplementation (45.00 mg kg−1) increased serum levels of triacylglycerol and protein and reduced the activity of hepatic lipogenic enzymes and the survival rate of the fish, probably in response to the high concentration of circulating cortisol. In addition, the higher level of ractopamine supplementation evaluated in this research impaired the weight gain and feed conversion. However, 11.25 mg kg−1 ractopamine reduced the ether extract level determined in the fillet and the fat deposition between muscle fibers, improving the nutritional quality of meat.(AU)

Abstract Purpose: To investigate the biochemical, histopathologic, and spermatogenetic changes in the detorsionated testicle after experimental torsion and to study the antioxidant effects of pheniramine maleate and nebivolol. Methods: Twenty-four Sprague-Dawley male rats were divided into 4 groups: Group 1: Sham; Group 2: Torsion/Detorsion (T/D); Group 3: T/D + Pheniramine maleate (PM); Group 4: T/D + Nebivolol (NB) group. Paroxanase (PON), total antioxidant status (TAS), total oxidant status (TOS), and oxidative stres index (OSI) were measured, and spermatogenetic and histopathologic evaluation was performed in tissue and blood samples. Results: The evaluation of tissue TAS indicated no statistically significant difference in Group 3 compared to Group 2. A statistically significant increase was detected in Group 4 compared to Group 2. Serum PON levels revealed a statistically significant increase in Groups 3 and 4 compared to Groups 1 and 2. The Johnsen testicular biopsy score decreased in Groups 3 and 4, but the decrease was not statistically significant. Conclusions: Pheniramine maleate and nebivolol have antioxidant effects against ischemia-reperfusion damage. They also support tissue recovery, which is more significantly observed by nebivolol.

Background: hypothyroidism is a disease of clinical importance that causes multisystem disorders, which can be confused with other endocrinopathies. Rapid and accurate diagnosis is necessary in order to avoid worsening of the clinical manifestation. The use of drugs in a wrong way can directly imply the difficulty of the diagnostic approach, since they may cause changes in the biochemical profiles, which are of great importance as markers in diseases of animals with low thyroid function.Case: The case reports a German Spitz male, one year old and six months old, neutered, who presented bilateral do not cause itching alopecia, being treated by another professional for endocrine and fungal affections. In this same medical appointment were neglected biochemical and hematological profile exams. The exams that had been performed were skin scrapings, which showed growth of fungi and bacteria, besides the measurement of thyroid hormones as TSH, T4L (T4 free) e T3, and also the low-dose dexamethasone suppression test. Based on the findings the therapy instituted by such professional was the administration of itraconazole (ITL®), levothyroxine sodium (compounded drugs) and trilostane (compounded drugs). With absence of clinical improvement, the guardian came to the hospital veterinary, in search of a second diagnostic opinion, where during the collection of information at the time of the anamnesis the patient presented lethargy, drowsiness and absence of hair on both sides. During the clinical examination, the animal presented mild bradycardia and a slightly diminished rectal temperature; in the dermatological evaluation the presence of pup pelt, cutaneous hyperpigmentation and hair thinning in the abdominal area were noted. From this evaluation, hematological exams were requested, which had altered the presence of lymphocytosis, and biochemical tests, where changes in the cholesterol and triglyceride levels were expected, but they were within the normal range.[...]

This study evaluated the effects that gradual ractopamine supplementation in diets with nutritional adjustments on pig meat. Were used 80 finishing crossbred barrows in a randomized block design with a 2×5 factorial arrangement (two diets: with and without nutritional adjustment; five levels of ractopamine supplementation: 5-5, 10-10, 20-20, 5-10 and 10-20 ppm) in the 14 initial and 14 final study days, four replicates with two animals by experimental unit. Higher shear force values (P<0.05) were obtained using the 5-5 and 10-20 ppm ractopamine supplementation plans in the diets without nutritional adjustment. With nutritionally adjusted diets, the 5-10 ppm of ractopamine supplementation plan yielded higher shear force values (P<0.05). Water retention capacity was higher (P<0.05) for animals fed adjusted diets and 5-5 and 10-20 ppm of ractopamine plans. In the 10-20 ppm of ractopamine supplementation plan, meat pH was higher (P<0.05) for diets without nutritional adjustment, whereas in the 20-20 ppm of supplementation plan, pH was higher for adjusted diets.

The objective of this study was to evaluate the effects of compensatory gain associated with the use of 10ppm ractopamine after a period of feed restriction in finishing pigs on performance, carcass and meat quality. Twenty castrated males and 20 females, at 110 days of age and 66.137±6.13kg live weight, were submitted to four treatments using a 2 x 2 factorial design (fed ad libitum or with 20% restriction between 0(21 days of age and fed with or without 10ppm ractopamine for 22(42 days of experimentation), with 10 replicates (animals). There was no interaction between the factors for any of the evaluated parameters. Animals treated with ractopamine presented better weight gain (1.083 versus 1.259kg), feed conversion (2.910 versus 2.577), warm and cold carcass weight (86.08 versus 89.00 and 83.46 versus 87.20kg, respectively), loin depth (63.02 versus 68.40mm), loin eye area (41.43 versus 46.59mm2) and muscle fiber diameter (27.48 versus 35.85µm). Animals submitted to feed restriction followed by ad libitum feed presented compensatory gain without losses to carcass and meat characteristics, but with a reduction in the ethereal extract (2.19 versus 1.64%) and lower water loss due to thawing in the meat (11.35 versus 9.42%). The effects of compensatory gain after food restriction and ractopamine are independent of the parameters evaluated.(AU)

Abstract Purpose: To determine if the combination of lidocaine with epinephrine or gamma globulin would decrease the rate or reduce the amount of local absorption of lidocaine through the airway. Methods: Twenty adult male cats were randomly and evenly distributed into four groups: 1) Group LG: lidocaine administered with gamma globulin; 2) Group LS: lidocaine administered with physiological saline); 3) Group LE: lidocaine administered with epinephrine; 4) Group C: control group. Invasive blood pressure, heart rate, and concentration of lidocaine were recorded before and after administration. Results: The peak of plasma concentrations appeared difference (Group LG: 1.39 ± 0.23 mg/L; Group LS: 1.47 ± 0.29 mg/L and Group LE: 0.99 ± 0.08 mg/L). Compared to Group C, there were significant differences in the average heart rate of Groups LG, LS, and LE (P < 0.05). The average systolic blood pressures were significantly different when each group was compared to Group C (P < 0.05). The biological half-life, AUC0-120, peak time, and half-life of absorption among the three groups have not presented statistically significant differences (P > 0.05). Conclusion: Administering lidocaine in combination with gamma globulin through airway causes significant decrease the rate and reduce the amount of local absorption of lidocaine in cats.

Two consecutive experiments were conducted using 480 pigs each with six replicate pens per treatment and 10 pigs per pen to evaluate the effect of standardized ileal digestible (SID) Lys (0.81, 0.91, 1.01, and 1.11%) in diets containing ractopamine (RAC; 10 or 20 mg kg-1) on growth and carcass traits of barrows in a commercial production system. There were no effects of either RAC levels or the interaction between RAC and SID Lys levels on any of the performance and carcass variables. No lack of fit was detected in the response surface analysis. From d 0 to 21, there was no effect of SID Lys levels on ADFI. However, as SID Lys increased, there was a linear increase in ADG and G:F. As SID Lys increased, the ADG, G:F, loin depth, and lean percentage improved linearly from d 0 to 28, with no effect on ADFI and backfat thickness. In conclusion, performance and carcass traits of pigs fed RAC (10 or 20 mg kg-1) in a commercial finishing facility are improved by up to 1.11% SID Lys supplementation. There is no additional benefit of feeding 20 mg kg-1 RAC in comparison with 10 mg kg-1 RAC.

The use of adrenergic agonists such as ractopamine leads to lean carcasses due to an increase in muscle deposition. The effects of ractopamine on performance and carcass traits in pigs may be varied depending on the level of agonist and amino acids, especially lysine, on diet, sex and age of the animals at the beginning of the additive supply. These factors, as well as the feed cost and the adoption or not the carcass classification influence the economic viability of the use of ractopamine. The economic analysis, therefore, is necessary to evaluate the feasibility of ractopamine inclusion in diets of finishing gilts. This study aimed to evaluate the addition of ractopamine at 0, 5, 10 and 15 mg/kg of diets for gilts slaughtered with 110 kg, on feed cost, gross and net incomes, considering or not the carcass rewarding. A total of 468 gilts were used and fed for 28 days. There was an increase in feed cost and linear decrease in net income without carcass rewarding with raising level of additive. For net income with carcass rewarding, there was a linear effect with higher income for the level of 15 mg/kg. Considering the carcass classification, the inclusion of ractopamine at the level of 15 mg/kg promotes higher income.

ABSTRACT PURPOSE: To evaluate the effects of dobutamine (DB), noradrenaline (NA), and their combination (NADB), on volume retention in rabbits submitted to hemorrhage. METHODS: Thirty six rabbits were randomly divided into 6 groups: SHAM, Control, Saline, DB, NA, DB+NA. All the animals, except for SHAM, were subjected to hemorrhage of 25% of the calculated blood volume. Control animals were replaced with their own blood. The other groups received NSS 3 times the volume withdrawn. The intravascular retention, hematocrit, diuresis, central venous pressure, mean arterial pressure, NGAL, dry-to-wet lung weight ratio (DTWR) and the lung and kidney histology were analyzed. RESULTS: Replacement with NSS and NA, DB or NA+DB did not produce differences in the intravascular retention. After hemorrhage, the animals presented a significant decrease in the MAP and CVP, which were maintained until volume replacement. Regarding NGAL, dry-to-wet-lung-weight ratio, lung and kidney histology, there were no statistical differences between the groups. CONCLUSION: The use of noradrenaline, dobutamine or their combination did not increase the intravascular retention of volume after normal saline infusion.

PURPOSE: To evaluate the effect of propranolol on capsular architecture around silicone implants by measuring the inflammation, capsular thickness, and collagen fiber density, using a guinea pig experimental model. METHODS: Thirty six adult male guinea pigs randomly divided into two groups (n=18) were used. Each one received a silicone implant with textured-surface. The capsular tissue around implants from untreated or treated animals with the beta-adrenoceptor antagonist propranolol (10 mg/kg, dissolved in daily water) were analyzed for inflammation by histological scoring, capsular thickness by computerized histometry, and collagen fibers type I and Type III density by picrosirius polarization at different time points (7, 14 or 21 days after silicone implantation). RESULTS: Propranolol treatment reduced inflammation and impaired capsular thickness and delayed collagen maturation around the textured implant. CONCLUSION: Propranolol reduces the risk of developing capsular contracture around silicone implants with textured surface. .
