This study was conducted to compare Central Anatolian Merino (CAM) and Malya sheep under intensive conditions in terms of reproductive traits, birth and weaning weights, fattening performance, and some fleece characteristics. In this study, 840 CAM and 194 Malya ewes were used for the comparison of reproductive performance, whereas 740 and 211 lambs were used to compare growth traits between the two breeds. Additionally, the fattening performance of 61 CAM and 68 Malya lambs were compared. Finally, for the comparison of wool quality traits, 94 CAM and 90 Malya lambs were used. The lambs were kept together with the sheep that gave birth to them for 10 days. After this period, lambs were given lamb starter feed, alfalfa hay, and vetch hay in addition to milk. When lambs reached 40 days old, they were taken to the pasture with the sheep. In this period, pasture grass, growing lamb concentrate feed, alfalfa hay, and vetch hay were given in addition to milk. Lambs were weaned when they reached an average age of 90 days, and their live weights were determined. A determined number of lambs was selected from weaned male lambs. For these animals, in the fattening period, barley, alfalfa hay, vetch hay, meadow hay, and lentil straw were provided ad libitum in addition to 400 g/day/head of fattening feed. This study indicated that birth rate was better for CAM sheep than for Malya sheep. However, Malya lambs may have more advantages than CAM lambs in terms of survival and fattening performance. Although birth weight was higher for Malya sheep than for CAM sheep, the opposite result was observed for the weaning weight. The beginning weights of fattened male lambs were higher for CAM lambs than for Malya lambs, but the ending weights of both fattened groups were similar.(AU)

Background: Epithelial cell adhesion molecule (EpCAM) is a glycoprotein responsible for multiple cellular functions that includes cell-to-cell adhesion and signal transduction. Studies in human breast cancer demonstrated that its overexpression has been linked to a more aggressive histological phenotype. However, in canine mammary tumors, its use is still scarce and its function, remains unknown. The main goal of the present study was to characterize the expression of EpCAM in canine mammary malignant tumors and associate its expression with clinicopathological features. Materials, Methods & Results: Forty-eight canine mammary samples were analyzed in the present study and went through the Hematoxylin-Eosin (HE) routine technique for histopathological and immunohistochemical analysis. EpCAM expression was evaluated considering the total immunolabelling (TI) which consists of the addition of 2 parameters: proportion score (PS) and intensity score (IS). Thirteen (27.1%) cases showed weak expression, 9 (18.7%) were characterized by a moderate expression and 27 (54.2%) were classified as intense. EpCAM overexpression was described in 36 (72.9%) primary tumors and in 5 lymph node metastases (71.4%). Immunoreactivity was, mainly, characterized by a staining in the lateral cell membrane of luminal epithelial cells, both in primary tumors and lymph node metastases. The statistical analysis was performed using the SPSS Statistics, version 24. Clinicopathological features, such as histological grade (P = 0.027), intravascular emboli (P = 0.004) and lymph node metastases (P = 0.016) demonstrated to be associated with a more aggressive histological type. Intravascular emboli and lymph node metastases were also linked to a higher histological grade. Discussion: EpCAM overexpression has been described in the literature both in canine mammary tumors and breast cancer. While the results in breast cancer could be ambiguous, depending on the tumor subtype, in the present study our findings demonstrated that overexpression was present in more than 50% of the total cases evaluated. In addition, immunoreactivity was also in accordance with the literature that describes membranous staining as the most prevalent pattern type. It was not possible to correlate overexpression with histological type and histological grade. However, those clinicopathological features are essential to evaluate tumor prognosis. For instance, our results demonstrated that histological type was statistically significant with histological grade (P = 0.027), intravascular emboli (P = 0.004) and lymph node metastases (P = 0.016). Additionally, the Nottingham system was statistically significant with intravascular emboli (P = 0.033) and lymph node metastases (P = 0.006). These findings have also been described in similar studies using canine mammary tissues in bitches. When it comes to pattern of expression in primary tumors and lymph nodes metastases, our findings described that neoplastic cell presented the same phenotype in these 2 different locations. In addition, loss of expression was also described in cluster cells within the subcapsular area which reflects the idea that the expression of EpCAM molecule changes according to time, which could be translated into the multistep process of metastases. Finally, it demonstrates that the development of neoplasia is not something static, but rather dynamic.

The objective of this study was to evaluate the accuracy of the following six body weight (BW) estimation methods in Campolina (CAM) horses: A - weight tape placed at three different positions on the animal's thorax; B - Crevat and Quetelet's formula; C - Hall's formula; D - Hintz and Griffiths's table; E - Santos's table; and F - Cintra's formula. A total of 380 CAM horses were separated according to sex, age class, and gestational stage and evaluated. To determine their accuracy, weights measured on a scale and weight estimates of the six methods were compared by paired t-test, mean prediction error (MPE), and coefficient of determination (R²), using R software. The predictive capacity of method F was lower in the 6-12 months age class, so this formula is not indicated. The BW was overestimated compared with the actual weight by methods A (with weight tape placed in position 3) and B and underestimated by method C. Methods D and E accurately estimated BW of CAM horses. Correction factors are required to accurately estimate BW in this breed using methods B and C. Method A with the weight tape placed in position 2 is the most accurate for predicting BW, including pregnant female horses, and can, therefore, be considered the most suitable method for estimating BW of CAM horses.

PnPa11 and PnPa13 are synthetic peptides derived from Phoneutria nigriventer spider venom, which display antinociceptive and neuroprotective properties. In this work, we evaluated the safety of intravitreal use and the neuroprotective effect of these peptides. Methods: The cytotoxicity and the antiangiogenic activity of these peptides were evaluated by the sulforhodamine-B method and chicken chorioallantoic membrane (CAM) assay, respectively. The in vivo safety was analyzed in Wistar rats that were intravitreally injected with different doses (0.50; 1.25; 2.50; 3.75 and 5.00 µg/mL) of these peptides (right eye, n = 6). The retinal function was assessed by electroretinography exams (ERG), intraocular pressure (IOP), and histological analyzes. In order to investigate the neuroprotective effect, Wistar rats received intravitreal injections (right eye, n = 6) of peptides at 1.25 µg/mL and then were exposed to blue LED light. In addition, the visual function and the retinal microstructure were verified. Results: Cytotoxicity analyses demonstrated that the peptides did not present any toxicity over ARPE-19 (adult retinal pigmented epithelial) cell line and the antiangiogenic study highlighted that the peptides promoted the reduction of blood vessels. The intravitreal injection did not cause major changes, neither induced any irreversible damage. In the retinal degeneration assay, the ERG records demonstrated that the prior treatment with PnPa11 and PnPa13 protected the retina from damage. Morphological analyses confirmed the ERG findings. Immunoblotting analyses revealed that PnPa11 increased Erk1/2, NR2A, and NR2B retinal expression after the light stress model, but did not cause Akt1 activation, while PnPa13 prevented Erk1/2 and Akt1 dephosphorylation. Conclusions: The intraocular administration of these peptides was well tolerated and presented protective activity against retinal degeneration, suggesting the potential use of these peptides as neuroprotectors in the ophthalmological field.(AU)

ABSTRACT: This study aimed to determine and evaluate whether there are differences in values of MAC in wildlife captured guan (Penelope obscura) under different conditions of social stress. This study used 27 bird species guan (P. obscura), divided into two groups: animals kept in the same enclosure (Collective Group) and animals kept in individual cages (Individual Group). The research was conducted at the Advanced Research Base of IBAMA, Painel/SC, and at the Clinical Veterinary Hospital of the Universidade do Estado de Santa Catarina, Lages/SC. The birds were captured using network trap with manual disarmament and transported to the Veterinary Hospital in cages. The animals were fasted for 2 to 6 hours before the procedure; anesthetic induction was performed with isoflurane for instrumentation. The isoflurane CAM was placed in the target value 1.3v% in the first animal of each group, and waited 15 minutes for the nociceptive (electric) stimulus, in the value of 50 hertz and 50mA, held in faradic form (3 consecutive simple stimuli, followed by 2 continuous stimuli). The stimulus was performed on the lateral side of the left pelvic limb in the tibiotarsal region, and the electrodes were fixed with a 22G needle at a distance of 5cm between them. The birds responses to the stimulus were considered positive (wing movements, head or vocalization) or negative (not presented movements of wings, head or vocalization) and the MAC value of the animal was recorded. Negative responses reduced next birds MAC value by about 10%, and MAC positive responses increased by about 10%. Statistical analysis was done by methods up and down and analyze quantal for MAC and paired t-test for equivalent or t-test for variances not equal variances for the physiological variables. At the end of the experiment, the birds were reintroduced in the same capture area. The MAC value of isoflurane in the Collective Group was 1.4v% and the Individual Group 1.9v% to 0.903atm. It is observed that guan (P. obscura) in the Collective Group showed lesser anesthetic resistance to isoflurane than the birds in the Individual Group, showing that some levels of social stress can influence the MAC values of the isoflurane.

It is important to identify the best inspired fraction of oxygen in a variety of situations, including sevoflurane or isoflurane anesthesia, in spontaneously breathing rabbits. For this, 64 rabbits were assigned to eight groups: GI100 (FiO2= 1,0 + isoflurane), GS100 (FiO2= 1,0 + sevoflurane), GI80 (FiO2= 0,8 + isoflurane), GS80 (FiO2= 0,8 + sevoflurane), GI60 (FiO2= 0,6 + isoflurane), GS60 (FiO2= 0,6 + sevoflurane), GI21 (FiO2= 0,21 + isoflurane), GS21 (FiO2= 0,21 + sevoflurane). The induction was performed with (2.5MAC) of the anesthetic. The vaporizer was setted at 1.5 MAC and FiO2 as attributed for each group. After the induction, the concentration was changed to 1 MAC. Measurements of parameters were performed 30 minutes after induction (T0), and then at 15 minute intervals (from T15 to T60). The arterial partial pressures of oxygen (PaO2), alveolar oxygen partial pressure (PAO2) and alveolar-arterial oxygen gradient [P(A-a)O2] were higher with the use of high FiO2. The GI80 showed higher levels of PaO2 FiO2 ratio and respiratory index (RI). In conclusion, the FiO2 of 0.21 is not indicated, because it causes hypoxemia. The isoflurane determines better ventilation when compared to sevoflurane, but isoflurane associated with 80% of oxygen promotes intrapulmonary shunt increase.(AU)

The great diversity of molecules found in spider venoms include amino acids, polyamines, proteins and peptides, among others. Some of these compounds can interact with different neuronal receptors and ion channels including those present in the ocular system. To study potential toxicity and safety of intravitreal injection in rabbits of LyeTx I b, a synthetic peptide derived from the toxin LyeTx I found in venom from the spider Lycosa eritrognatha and to evaluate the angiogenic activity on a CAM model. Methods: ARPE-19 cells were treated with LyeTx I b (0.36; 0.54; 0.72; 2.89; 4.34 or 9.06 µM). In this study, New Zealand rabbits were used. LyeTx I b (2.89 µM) labeled with FITC dissolved in PBS, or only PBS, were injected into vitreous humor. Electroretinogram (ERG) was recorded 1 day before injection and at 7,14 and 28 days post-injection. Clinical examination of the retina was conducted through tonometer and eye fundus after ERG. Eyes were enucleated and retinas were prepared for histology in order to assess retinal structure. CAMs were exposed to LyeTx I b (0.54; 0.72; 2.17 or 2.89 µM). Results: ARPE-19 cells exposed to LyeTx I b showed cell viability at the same levels of the control. The fluorescence of LyeTx I b labeled with FITC indicated its retinal localization. Our findings indicate ERG responses from rats injected in the eye with LyeTx I b were very similar to the corresponding responses of those animals injected only with vehicle. Clinical examination found no alterations of intraocular pressure or retinal integrity. No histological damage in retinal layers was observed. CAM presented reduced neovascularization when exposed to LyeTx I b. Conclusions: Intravitreal injection of LyeTx I b is safe for use in the rabbit eye and prevents neovascularization in the CAM model, at Bevacizumab levels. These findings support intravitreal LyeTx l b as a good candidate to develop future alternative treatment for the retina in neovascularization diseases.(AU)

The aim of this study was evaluate the effects of Bracken fern (BF) (Pteridium aquilinum (L.) Kuhn.) on biological systems. When consumed by animals can cause acute intoxication, hematuria, biochemistry alterations and cancer. To humans the toxicity is associated with its intake on contaminated ground water or milk and inhalation of its spores. In order to check the BF aqueous extract (AEB) deleterious effects on animals blood vessels system, chick embryo chorioallantoic membrane (CAM) was used. It were applying on CAM 0.1, 0.5, 1, 5, 10 e 15 µg/mL of AEB and saline as control. The angiogenesis was analyzed and the vascular density index (VDI) calculated. The CAM samples were prepared and stained with H&E to evaluation of microvessels, Massons trichrome to characterize collagen and fibrin deposition and Picro-sirius used to evaluate collagen using polarized light. Also the morphological aspects of embryos were analysed. We observe on the results of neovascularization that AEB did not change significantly the number of vessels/mm², however, membranes treated with AEB (5 or 10 µg/ mL) exhibit opacity and tissue fibrosis, both signs of inflammation. Histological analysis with Massons trichrome and picro-sirius on tissues exposed to AEB respectively has shown increased collagen fibers and presence of fibrilar collagen. The embryos exposed to concentrations of 5 or 10 μg/mL AEB, showed changes as poor face formation and poor closing of abdominal wall. The highest concentration of AEB (15 μg/mL) was lethal to embryos. Although significant effects on the CAMs vasculature has not observed, tissue aggression was detected, a desmoplasia (an extensive inflammatory signal triggered by tissue injury), changes caused on embryos as well as the presence of toxic substances in the AEB show us an important and deleterious pathway of this bracken fern extract on its intoxicants effects on humans and animals, and even cancer or the death of animals.

The present study evaluated the minimum alveolar concentration of isoflurane (ISOMAC) in twenty three dogs premedicated with acepromazine (0.02mgkg-1) and morphine (0.5mgkg-1) and administered racemic (RK) or S(+)-ketamine (SK). Dogs randomly received a single dose (3mgkg-1, IM) of either RK or SK 15minutes after anesthetic induction with propofol. The ISOMAC was determined by the up-and-down method. Approximately 20 minutes after administration of RK or SK, a surgical noxious stimulus was applied and the response evaluated. The ISOMAC was 0.50±0.01% in the RK group (n=10) and 0.31±0.04% in the SK group (n=13). The ISOMAC was 38% lower in the SK group compared to the RK group. Results of the present study revealed that in dogs premedicated with acepromazine and morphine, IM administration of 3mgkg-1 ketamine approximately 20 minutes before the noxious stimulus produced clinically important reduction in the ISOMAC and the MAC-sparing effect was significantly greater with SK compared to RK.

The objective was to relate the need for analgesic postoperative recovery through Visual Analogue Scale (VAS), Glasgow, Colorado and Melbourne, by an experienced assessor (AE) and other non-experienced ones (ANE), in bitches undergoing total unilateral mastectomy. Otherwise healthy bitches, a total of 24, were admitted 24 hours before the surgical procedure for assessment of behavior with the help of the above scales to determine the baseline (M0) moment. They were pre-medicated with morphine and acepromazine (0,02 and 0,5 mg/kg) and general anesthesia was induced with propofol (4 mg/kg) and maintained with isoflurane at 1% MAC. The analgesic during surgery was maintained with ketamine and fentanyl (10 µg/kg/min and 10 µg/kg/h). The other evaluations were performed at 1, 2, 4, 6, 8, 12 and 24 hours postoperatively, and redemptions made with intramuscular morphine (0,5 mg/kg) when a greater than or equal score of 50, 6, 2 and 9 points was observed respectively for the described ranges, as observed by the AE and when at least two scales demonstrated these values. There was an increase of M1 to M12 pain scores for AE and the ANE for VAS. In Colorado analyses, the highest painful scores occurred in relation to M0 between M2 and M8 to the AE and M1 to M12 for ANE. In Glasgow analyses, higher scores were detected between M1 to M12 for AE and M1 to M24 for ANE. And for the Melbourne highest values were observed in M1 and M24 for AE and ANE. The best correlation between the scales was 0,775 between Glasgow and Colorado and of the evaluators of 0,925 to Glasgow. The Glasgow scale was shown to be more sensitive to detect painkiller redemptions in dogs undergoing total unilateral mastectomy, the inexperience of the appraiser does not compromise the quality of painful reviews, and it is suggested to reduce the score VAS and Melbourne to increase it is ability to detect rescue postoperative analgesics.(AU)

The objective of this study was to compare the influence of continuous intravenous infusion of tramadol alone, or tramadol combined with lidocaine and ketamine, on minimum alveolar concentration of sevoflurane (MACsevo) of dogs undergoing an ovariohysterectomy (OHE). We used 28 healthy dogs of various breeds and age, randomly divided into two groups according to the infusion given: TRA (tramadol alone) or TLK (tramadol, lidocaine and ketamine). The patients were premedicated with acepromazine and midazolam, and then anesthesia was induced with propofol and maintained with sevoflurane. Fifteen minutes after induction, the patients received their loading dose of treatment. Then, the continuous infusion was then set to 1.3mg/kg/hour of tramadol with or without 3mg/kg/hour of lidocaine and 0.6mg/kg/hour of ketamine, diluted in a 500mL bag of saline solution at an infusion rate of 10mL/kg/hour. The Dixon method was chosen to determine the MACsevo and a skin incision was used as a noxious stimulus. An unpaired Student's t-test was used to identify statistically significant differences between the treatments. These differences were considered significant when p<0.05. The MACsevo of the TRA group was 1.22±0.15 vol% and the MACsevo of the TLK group was 0.85±0.22 vol%. We conclude that TLK infusion decreased the MACsevo by 30.22% compared to tramadol alone, demonstrating that the combination of drugs was effective in reducing MACsevo in dogs.(AU)
