Resumo

Cryptococcus gattii is often associated with pulmonary and systemic infections in humans and animals. In this research we report a case of nasopharyngeal cryptococoma caused by C. gatti in an equine. A 10-year-old mare presented a mass obstructing the oropharynx. Macroscopically the mass was asymmetric, and was attached to the ethmoidal sinuses and obstructed the oropharynx. Histopathological examination of the mass revealed multiple yeast cells ranging from spherical to oval, 4-8m in diameter, with some of them showing narrow base polar budding. Cryptococcus gattii growth in mycological culture (Sabouraud Dextrose Agar) and was L-canavanine-glycine-bromothymol blue Agar positive. The molecular identification confirmed the isolate as C. gattii by means of the amplification of universal primers. C. gattii is considered an emerging fungal agent, as it affects human and animals and does not respond efficiently to commonly established treatments.


Cryptococcus gattii é frequentemente associada a infecções pulmonares e sistêmicas em humanos e animais. Neste relato descreve-se um caso de criptococoma nasofaríngeo por C. gatti em um equino. Uma égua de 10 anos, apresentou uma massa obstruindo a orofaringe. Macroscopicamente a massa era assimétrica, e estava aderida aos seios etmoidais obstruindo a orofaringe. O exame histopatológico da massa revelou múltiplas células leveduriformes variando de esféricas a ovais, de 4-8m de diâmetro, com algumas delas apresentando brotação polar de base estreita. Cryptococcus gattii foi isolado na cultura micológica (Ágar sabouraud dextrose) e foi positivo no Ágar L-canavanina-glicina-azul de bromotimol. A identificação molecular confirmou o isolado como C. gattii por meio da amplificação de primers universais. O diagnóstico de criptococoma por C. gattii nasofaríngeo foi baseado nos sinais clínicos, achados macroscópicos, lesões histológicas, cultura micológica e Reação em Cadeia da Polimerase. O C. gattii é considerado um agente fúngico emergente, pois acometendo humanos e animais, não respondendo com eficiência aos tratamentos comumente estabelecidos.

Resumo

Cryptococcosis is an infection that affects humans and animals, the etiology is attributed to Cryptococcus neoformans variety neoformans, C. neoformans var. grubii and Cryptococcus gattii. The infection is common in dogs and cats, causing respiratory, neurological, cutaneous and ocular infections. Aiming to better understand the epidemiology of cryptococcosis in animals in the region, this paper describe the occurrence and characterization of the Cryptococcus species involved in this illness in pet animals at Mato Grosso State, Brazil. Clinical samples of four cases, two in cats and two dogs, were submitted for pathological, microbiological and molecular analysis. Microscopically, in three cases, tissue sections stained with hematoxylin and eosin had absence to severe granulomatous reaction composed by histiocytes, multinucleated cells and lymphocytes infiltration. In one case, citological imprint analysis showed similar inflammatory mainly mononuclear and lymphocyte cells infiltration. All cases had variable amounts of intracellular and extracellular fungal structures compatible with Cryptococcus sp. on Periodic Acid-Schiff (PAS) stain. All clinical samples were positive for culture on Sabouraud Dextrose Agar (SDA) and morphologically classified as Cryptococcus sp. The isolates were PCR positive for C. gatti, being confirmed by sequencing technique. The findings characterize the molecular species involved in animal infections in the region, and may contribute to future studies of the epidemiology of C. gattii.(AU)


A criptococose é uma infecção que afeta humanos e animais sendo a etiologia atribuída às espécies Cryptococcus neoformans variedade neoformans, C. neoformans var. grubii e C. gattii. A doença é comum em cães e gatos, causando infecções respiratórias, neurológicas, cutâneas e oculares. Com o objetivo de entender melhor a epidemiologia da criptococose em animais, este trabalho descreve a ocorrência e a caracterização de espécies de Cryptococcus em pequenos animais doentes no Estado de Mato Grosso, Brasil. Amostras clínicas de quatro casos, dois em felinos e dois em caninos, foram submetidas à análise patológica, microbiológica e molecular. Microscopicamente, em três casos, nos cortes de tecidos corados pela hematoxilina notou-se desde ausência até acentuada inflamação granulomatosa composta por histiócitos, células multinucleadas e infiltração linfocitária. Em um caso, na análise citológica de "imprint" observou-se infiltrado inflamatório similar composto principalmente por células mononucleares e linfócitos. Em todos os casos havia variável quantidade de estruturas fúngicas intra e extracelulares compatíveis com Cryptococcus spp pela coloração do ácido períodico de Schiff (PAS). Todas as amostras foram positivas para a cultura em Sabouraud Dextrose Agar (SDA), e morfologicamente classificadas como Cryptococcus sp. Os isolados foram positivos no PCR para C. gattii, sendo confirmados pelo seqüenciamento. Os resultados caracterizaram a espécie envolvida na região, e contribuem para futuros estudos sobre a epidemiologia de C. gattii.(AU)

Resumo

Cryptococcosis in animals is an important fungal disease caused by the encapsulated yeast Cryptococcus neoformans. This report describes the occurrence of Cryptococcus gattii and Cryptococcus albidus in domestic pigeon (Columba livia), living together with other birds in a breeding center. The animal presented a pinkish, vascularized mass with gelatinous aspect localized subcutaneously under the right lower eyelid, with approximately 2cm in diameter. At microbiological exam it was isolated Cryptococcus gatti from the eyelid mass, lungs and liver, C. albidus from the trachea and both Cryptococcus species from muscle and kidney.(AU)

A four-year-old male goat with a history of neurological disorder was euthanized. It presented uncommon nodules in the brain and lungs associated with multiple abscesses, predominantly in the spleen and liver. Histological examination of brain and lung sections revealed yeast forms confirmed to be Cryptococcus gattii after a combination of isolation and polymerase chain reaction (PCR) procedures. Moreover, Corynebacterium pseudotuberculosis infection was diagnosed by PCR of samples from the lung, spleen and liver. The present report highlights the rare concurrent infection of C. gatti and C. pseudotuberculosis in an adult goat from São Paulo state, Brazil, and indicates the necessity of surveillance in the treatment of goats with atypical pulmonary infections associated with neurological disorders.
